Osteoporosis is closely related to iron metabolism. This study aimed to investigate whether hops extract (HLE) and its active component xanthohumol (XAN) could ameliorate bone loss caused by iron overload, and explored its potential mechanism.Iron overload mice induced by iron dextran (ID) were used in vivo, and were treated with HLE and XAN for 3 months. Bone micro-structure and bone morphology parameters were determined by Micro-CT and TRAP staining. Bone metabolism markers and oxidation indexes in serum and bone tissue were evaluated. For in vitro experiment, bone formation indexes were determined. Moreover, the expression of key proteins in protein kinase B (Akt)/glycogen synthetase kinase 3β (GSK3β)/nuclear factor E2-related (Nrf2) pathway was evaluated by Western blotting.HLE and XAN effectively improved the bone micro-structure of the femur in mice, altered bone metabolism biomarkers, and regulated the expression of proteins related to bone metabolism. Additionally, they significantly promoted cell proliferation, runt-related gene 2 (Runx2) expression, and increased ALP activity in ID-induced osteoblasts. Moreover, HLE and XAN markedly inhibited the increase of oxidative stress caused by iron overload in vivo and in vitro. Further studies showed that they significantly up-regulated the expression of p-Akt, p-GSK3β, nuclear-Nrf2, NAD(P)H: quinone oxidoreductase 1 (NQO1), and heme oxygenase-1 (HO-1) in ID-induced osteoblasts.These findings indicated hops and xanthohumol could ameliorate bone loss induced by iron overload via activating Akt/GSK3β/Nrf2 pathway, which brought up a novel sight for senile osteoporosis therapy.

Amphotericin B (AmB) is a powerful but toxic fungicide that operates via enigmatic small molecule–small molecule interactions. This mechanism has challenged the frontiers of structural biology for half a century. We recently showed AmB primarily forms extramembranous aggregates that kill yeast by extracting ergosterol from membranes. Here, we report key structural features of these antifungal ‘sponges’ illuminated by high-resolution magic-angle spinning solid-state NMR, in concert with simulated annealing and molecular dynamics computations. The minimal unit of assembly is an asymmetric head-to-tail homodimer: one molecule adopts an all-trans C1–C13 motif, the other a C6–C7-gauche conformation. These homodimers are staggered in a clathrate-like lattice with large void volumes similar to the size of sterols. These results illuminate the atomistic interactions that underlie fungicidal assemblies of AmB and suggest this natural product may form biologically active clathrates that host sterol guests.

Fungal infections cause more than 1.5 million deaths annually. With an increase in immune-deficient susceptible populations and the emergence of antifungal drug resistance, there is an urgent need for novel strategies to combat these life-threatening infections. Here, we use a combinatorial screening approach to identify an imidazopyrazoindole, NPD827, that synergizes with fluconazole against azole-sensitive and -resistant isolates of Candida albicans. NPD827 interacts with sterols, resulting in profound effects on fungal membrane homeostasis and induction of membrane-associated stress responses. The compound impairs virulence in a Caenorhabditis elegans model of candidiasis, blocks C. albicans filamentation in vitro, and prevents biofilm formation in a rat model of catheter infection by C. albicans. Collectively, this work identifies an imidazopyrazoindole scaffold with a non-protein-targeted mode of action that re-sensitizes the leading human fungal pathogen, C. albicans, to azole antifungals.

BackgroundMicroorganisms are known to be important drivers of biogeochemical cycling in soil and hence could act as a proxy informing on soil conditions in ecosystems. Identifying microbiomes indicative for soil fertility and crop production is important for the development of the next generation of sustainable agriculture. Earlier researches based on one-time sampling have revealed various indicator microbiomes for distinct agroecosystems and agricultural practices as well as their importance in supporting sustainable productivity. However, these microbiomes were based on a mere snapshot of a dynamic microbial community which is subject to significant changes over time. Currently true indicator microbiomes based on long-term, multi-annual monitoring are not available.ResultsHere, using samples from a continuous 20-year field study encompassing seven fertilization strategies, we identified the indicator microbiomes ecophysiologically informing on soil fertility and crop production in the main agricultural production base in China. Among a total of 29,184 phylotypes in 588 samples, we retrieved a streamlined consortium including 2% of phylotypes that were ubiquitously present in alkaline soils while contributing up to half of the whole community; many of them were associated with carbon and nutrient cycling. Furthermore, these phylotypes formed two opposite microbiomes. One indicator microbiome dominated byBacillus asahii, characterized by specific functional traits related to organic matter decomposition, was mainly observed in organic farming and closely associated with higher soil fertility and crop production. The counter microbiome, characterized by known nitrifiers (e.g.,Nitrosospira multiformis) as well as plant pathogens (e.g.,Bacillus anthracis) was observed in nutrient-deficit chemical fertilizations. Both microbiomes are expected to be valuable indictors in informing crop yield and soil fertility, regulated by agricultural management.ConclusionsOur findings based on this more than 2-decade long field study demonstrate the exciting potential of employing microorganisms and maximizing their functions in future agroecosystems. Our results report a “most-wanted” or “most-unwanted” list of microbial phylotypes that are ready candidates to guide the development of sustainable agriculture in alkaline soils.

To investigate the effects of long-term fertilization on microbial decomposition of residues and priming effect (PE), 13C-labeled maize (Zea mays L.) residues were supplied to arable soils with a 20-year application of compost (COM), mineral NPK fertilizer (NPK), or without any treatments, the no-fertilizer control (NF). The soils that had been stored for 6 years were used in the present incubation experiment. The release of CO2–C and the microbial incorporation of residue-derived C determined by phospholipid fatty acids (PLFAs) analysis were monitored over a 90-day incubation period. Residue additions significantly increased cumulative CO2–C emission and induced positive PE. Cumulative residue-derived CO2-C emission and PE mainly occurred within the first 15 days. The COM soil had significantly higher cumulative residue-derived CO2–C emission but lower PE than the NF and NPK soils. Residue additions significantly increased microbial abundance and changed the composition of main microbial groups. The COM soil showed a significantly lower relative fungal abundance (mol%) but a higher relative actinomycetes abundance than the NF and NPK soils. The incorporation of residue-derived C within fungi was the highest among all the main microbial groups and decreased from 15 to 45 days, while the incorporation of residue-derived C within actinomycetes increased with time in three soils. The incorporation of residue-derived C within fungi was the highest in the COM soil over the course of incubation. The long-term compost input promoted fungal use of residue C and stimulated residue decomposition.

Field decomposition of wheat and maize straws was monitored for 20 months using litterbag method in Calcaric Fluvisol soils with three different textures (sand, sandy loam, and silty clay). Residual straw samples were collected at 0 or after 4, 6, 10, and 20 months of decomposition. The chemical structure of straw was analyzed by solid-state 13C nuclear magnetic resonance (13C-NMR) spectroscopy, and the composition of main microbial groups was evaluated by phospholipid fatty acid analysis. Regardless of the straw type and soil texture, the straw biomass and C loss increased steadily in the first 10 months and then leveled off in the following 10 months; both the chemical structure of straw and the composition of main microbial groups differed during incubation. During the first 4 months of wheat straw degradation, the decrease in di-O-alkyl and O-alkyl C and the increase in alkyl and N-alkyl/methoxyl C contents were related to the enrichment of fungi (18:1ω9c) and Gram-negative bacteria (18:1ω7c and 16:1ω7c), while the degradation of maize straw was associated with the decrease in the fungal (18:1ω9c) abundance and the increase in the abundance of Gram-negative (18:1ω7c and 16:1ω7c) and Gram-positive (a15:0) bacteria. During the 6–10-month period, the decrease in di-O-alkyl and O-alkyl C and the increase in alkyl, aryl, and carboxyl/amide C contents were related to the enrichment of arbuscular mycorrhizal fungi (AMF) (16:1ω5c) and Gram-negative (cy19:0ω8c) bacteria and the decrease in fungal (18:2ω6,9c) abundance, with AMF playing an important role in the degradation of both straw types. The altered chemical structure of wheat and maize straws had opposite links with fungal abundance during the first 4 months, while alterations occurring during the 6–10-month period mainly depended on the variation in the AMF abundance.

This study aims to investigate the presence of aggregate hierarchy during the decomposition of rice straw in a subtropical soil (Ultisol) with large quantities of Fe and Al oxides and in two temperate soils with a large amount of montmorillonite (Vertisol) or with less organic matter but a large content of CaCO3 (Inceptisol) and to explore the relationship between relative contributions of biotic and abiotic processes in soil aggregation and the presence of aggregate hierarchy. Elevated CO2 and nitrogen addition can increase rice yield and stimulate rice to absorb more nutrients from the soil. The aim of this study was to investigate how soil organic carbon and aggregates change under such high-output conditions. Rice cultivars IIY 084 and WYJ 21 (strong and weak response to elevated CO2, respectively) were planted in free-air CO2 enrichment (FACE) platforms. Treatments included: 1) elevated CO2 without nitrogen addition (EC); 2) nitrogen addition and ambient CO2 (EN); 3) combined elevated CO2 and nitrogen addition (ECEN); and 4): ambient CO2 without N addition (CK). Soil organic carbon (SOC), readily-oxidizable organic carbon (ROC) and soil aggregate components were measured. Treatment EC increased SOC and ROC, while EN decreased SOC and ROC compared to CK in soil under either rice cultivar. EC and EN decreased the percentage of large macroaggregates (LM) and the mean weight diameter (MWD) of aggregates compared to CK in ‘WYJ 21’ soil, while they increased LM and MWD in ‘IIY 084’ soil. ECEN increased LM and MWD compared to CK in soil under either rice cultivar. High productivity of ‘IIY 084’ through long-term cultivation under current N fertilization in future climatic conditions of elevated CO2 could be achieved due to increased soil aggregate stability and SOC.

A 2-year field experiment was conducted on the North China Plain to assess the effects of two straw management practices and different nitrogen fertilizer addition levels on soil organic matter (SOM) fractions and microbial properties under a winter wheat-summer maize cropping system. Eight treatments (two straw management practices and four nitrogen fertilizer input levels) were established using a randomized complete block design. The straw management practices were no straw return and straw return. Each straw management practice received nitrogen fertilizer rates at 0 (N0), 270 (N270), 360 (N360), or 450 (N450) kg N ha−1 year−1. The soil properties measured included SOM fractionation, enzyme activities, community level physiology profile (CLPP), and microbial community abundance and composition, which were represented by phospholipid fatty acid (PLFA). Compared to no straw return, the addition of straw significantly increased soil total organic carbon and nitrogen, particulate organic carbon and nitrogen, and occluded particulate organic carbon and hot water extractable organic carbon by 7–29%. Invertase, protease, urease, and dehydrogenase activities increased by 22–40%. The biomasses of Gram-positive bacteria and fungi also increased. The increase in SOM fractions may be attributed to the decomposition of the added straw by more active enzymes, which may result from the shift of microbial community composition. In addition, hot-water extractable organic carbon, NO3-N, urease activity, and fungal biomass in the N270, N360, and N450 treatments significantly increased, independent of the nitrogen application rate, which indicated that N input level had a non-linear effect on soil properties in this area. Short-term straw return combined with a modest nitrogen fertilizer input (270 kg N ha−1 year−1) can increase SOM contents and microbial activity, which may help improve soil fertility in this region.

Humic substances are recalcitrant and might act as persistent binding agents to form macroaggregates. The focus of this study is in investigating the contribution of humic carbon (HC) to soil aggregation in response to various tillage and residue managements. Arable soils following 8-year contrasting managements were collected to determine aggregate size distribution and stability and HC fractions including humic acid (HA) and fulvic acid (FA). The contribution of HC to aggregation was divided into three special effects including positive effect (PE), negative effect (NE), and combined effect (CE), and these effects were measured using aggregate fractionation techniques. As well as to promote structural stability, HC bounds predominantly with the silt + clay fraction and secondarily with microaggregates to form larger aggregates. The PE increased with increasing aggregate size, whereas the NE followed the opposite pattern. A positive CE was observed for large and small macroaggregates, whereas the CE for microaggregates and the silt + clay fraction was negative. Compared to continuous tillage, reduced- and no-tillage decreased the PE for large and small macroaggregates by 1.58–30.98% at the 0–20 cm depth, and straw returning also slightly decreased the corresponding PE relative to straw removing. By contrast, a significantly higher NE for small macroaggregates at the 0–10 cm depth while 6.33–81.11% decreases in CE for large and small macroaggregates at the 0–10 cm depth as well as for large macroaggregates at the 10–20 cm depth, were observed under reduced- and no-tillage. The extraction of HC significantly reduced the aggregate stability and reduced- and no-tillage effectively limited its decrease magnitude. Small macroaggregates and microaggregates made larger contributions to soil HC accumulation than did other fractions. An averagely increased contribution from large or small macroaggregates was observed under both reduced-/no-tillage and straw returning at the 0–20 cm depth. A significant and positive relationship was found between the mass proportion of macroaggregates and the HC accumulation in 0–20 cm soil. Large macroaggregates had significantly higher HA/FA ratios than small macroaggregates, and reduced- and no-tillage significantly increased these ratios both in large and in small macroaggregates. The CE for large or small macroaggregates was also significantly negatively correlated with their HA/FA ratios. Overall, the HC accumulation in soil is likely to play a key role in macroaggregation, but conservation tillage might decrease the contribution magnitude of HC to large or small macroaggregation through increasing the corresponding HA/FA ratios.

This paper was primarily devoted to reveal the composition of soil organic carbon (C) at a molecular level and to compare the differences existing among tillage practices after 8 years of contrasting managements, aiming to identify the effects of conservation tillage on the accumulation process of soil organic C and its association with macroaggregation. A sandy loam soil following 8-year conservation tillage was collected to identify the dynamics of organic components and their associations with macroaggregation. The content of particulate organic matter within macroaggregates was determined using aggregate fractionation techniques. Conservation tillage improved the accumulation efficiency of different C functional groups in large and small macroaggregates, and resulted in a higher soil organic C level. The content of C functional groups was significantly and positively correlated with the C amount of subfractions in macroaggregates. The accumulation of organic components, particularly the methoxyl/N-alkyl C in the 0–5 cm layer and the O-alkyl C in the 5–10 cm layer, in turn, accelerated soil macroaggregation. Overall, organic component accumulation and soil macroaggregation were interacted on each other. Our results suggested that the enrichment of methoxyl/N-alkyl C and O-alkyl C in the test soil following conservation tillage could effectively promote organic C accumulation and macroaggregation.

Determining soil pore size distribution is difficult and time-consuming using traditional methods. Additionally, for swelling and shrinking soil, the specific volume of soil changes with soil moisture conditions. Nuclear magnetic resonance (NMR) relaxometry allows observation of pore size distribution changes during the process of dehydration and soil shrinkage. Naturally structured soil cores of a Vertisol with characteristic swelling and shrinking behavior were collected from the Huang-Huai-Hai Plain of China. The samples were saturated with water and dehydrated gradually at room temperature and relaxometry tests conducted at different moisture contents as they air-dried. Then, the soil cores were oven-dried at 105 °C and saturated with n-octane, which prevented clay swelling. The pore size distribution of soil cores at the end of the shrinking process was determined through the description of n-octane-filled pores. The shrinkage characteristic curves were determined as naturally structured soil cores were air-dried from moisture content at full expansion to constant volume, indicating the relationship between the volume of bulk soil and moisture content. The transverse relaxation time (T 2) distribution deduced from NMR relaxometry gives a good description of the size distribution of pores filled with protons (contained in water or n-octane). The T 2 distribution curves of soil cores at saturation were trimodal, due to the presence of interlayer, interparticular, and interaggregate pores. Combined with the shrinkage characteristic curves, it was deduced that the structural pores were evacuated during the structural shrinkage period. The normal and residual shrinkage was accompanied by the narrowing and closure of the interlayer spaces. During the residual shrinkage period, the frame structure of the soil particles prevented further shrinkage of the bulk soil. The shrinkage process was accompanied by the closure of interlayer spaces and the formation of large cracks between aggregates. 1H NMR relaxometry was especially suited to studying the changing pore size distribution of swelling and shrinking soils. When the soil cores began shrinking, almost all remaining water was retained in interlayer spaces. The volume change of the interlayer space was the main cause of swelling and shrinking. The swelling limit could be estimated from the T 2 distributions of soils at full expansion.

Atmospheric nitrogen (N) deposition has caused concern due to its effects on litter decomposition in subtropical regions where N-fixing tree species are widespread. However, the effect of N deposition on litter decomposition in N-fixing plantations remains unclear. We investigated the effects of a 2-year N deposition treatment on litter decomposition, microbial activity, and nutrient release in two subtropical forests containing Alnus cremastogyne (AC, N-fixing) and Liquidambar formosana (LF, non-N-fixing). The decomposition rate in AC was faster than in LF when there was no experimental N deposition. In AC, the initial decomposition rate was faster when additional N was applied and was strongly linked to higher cellulose-degrading enzyme activities during the early decomposition stage. However, N deposition reduced litter decomposition and inhibited lignin-degrading enzyme activities during the later decomposition stage. Nitrogen deposition enhanced carbohydrate and alcohol utilization, but suppressed amino acid and carboxylic acid uptake in the AC plantation. However, it did not significantly affect litter decomposition and microbial activity in the LF plantation. In conclusion, N deposition could inhibit litter decomposition by changing microbial enzyme and metabolic activities during the decomposition process and would increase carbon accumulation and nitrogen retention in subtropical forests with N-fixing tree species.

The North China Plain (NCP) is a strategic grain production base in China with a wild distribution of fertile soils. During the past 20 years, high-input intensive agriculture with excess chemical fertilizer application has sustained high grain yields, but may have resulted in contamination of some elements in farmland. In this study, the accumulation and transfer of arsenic (As) and mercury (Hg) in typical Calcaric Fluvisols with long-term different fertilization practices were investigated. Field experiments with seven treatments were launched in 1989, and soil and plant samples were collected and analyzed periodically. The treatments include OM (organic manure), OM + NPK (50 % organic manure + 50 % chemical fertilizer), NPK, NP, PK, NK, and CK (the control experiment with no fertilizer). With over 20 years (1989–2009) of cultivation, various extents of As and Hg accumulations were really observed in the soil. The higher As and Hg contents were found in P fertilizers than those in N, K, or OM fertilizers. As a result, the long-term P fertilization slightly promoted Hg accumulations with decreased soil Hg concentrations in the order of NPK ≈ NP ≈ PK ≈ OM + NPK > OM > NK ≈ CK, which was similar to the order of crop yields. At the tillage layer (0–20 cm), Hg accumulation in the soil was enhanced by crop production, due to the highly accumulated Hg in plant roots finally remained in the soil. However, no significant differences of soil As concentrations can be found between treatments with and without P fertilizers probably due to water leaching and plant uptake. Soil As and Hg were mainly contributed by fertilizers, irrigation, and atmospheric deposition in recent years, but they did not exhibit in significant accumulations in the soil. The quick assessment approaches for accurately measuring water-use efficiency (WUE) in maize under water stress are important to water-saving agriculture. We investigated how and whether carbon isotope composition (δ 13C) and specific leaf area (SLA) could be used to assess WUE for maize as influenced by water stress. A pot experiment was conducted twice during six typical maize (Zea mays) growth stages of seedling, jointing, booting, tasseling, filling and maturity, respectively. The ratio between the activities of ribulose 1,5-bisphosphate carboxylase (Rubisco) and phosphoenolpyruvate carboxylase decreased bundle sheath leakiness (φ) under water deficiency, caused more 13C to be assimilated, and resulted in increased δ 13C in leaves. Water stress increased the fractionation of 13C when assimilates were transported from leaf to stem, indicating that water stress affected leaf expansion and translocation of assimilates from leaf to stem, and resulted in thicker leaves and lower SLA. WUE showed significant positive correlations with leaf δ 13C and SLA, implying that leaf δ 13C and SLA could effectively reflect the drought adaptation and high WUE under different water conditions.

Gaseous nitrogen (N) losses are mainly responsible for low N use efficiency of applied fertilizers and their derivative environmental hazards, while N application rates and soil water regime are important factors regulating these N processes. In this study, gaseous N losses via ammonia volatilization (AV) and denitrification, determined by sponge trapping-KCl extraction and acetylene inhibition-soil core incubation, respectively, were investigated under different coupled water-N management practices with a winter wheat (Triticum aestivum L.) and summer maize (Zea mays L.) rotation system in the Huang-Huai-Hai Plain. Ammonia volatilization after urea application, particularly after topdressing during maize growing seasons, was the dominant gaseous N loss, amounting up to 53.2 kg N ha−1 (38.6 % of the applied N). Ammonia volatilization generally decreased by increasing irrigation rate applied immediately after fertilization. Denitrification N losses were lower than ammonia-N losses, accounting for 1.3 to 18.7 kg N ha−1 year−1, with more than 90 % occurring during the maize growing seasons. Soil aggregation is one of the key properties affecting the productivity of soils and the environmental side effects of agricultural soils. In this study, we aimed to identify whether biochar could be used to improve aggregate stability. A 2-year field experiment was conducted to investigate the effect of biochar application (0, 2.5, 5, 10, 20, 30 and 40 t ha−1) on aggregate characteristics of upland red soil under a rapeseed–sweet potato rotation in subtropical China. Percentage of aggregate destruction (PAD0.25), mean weight diameter (MWD), geometric mean diameter (GMD) and fractal characteristics of soil aggregates were measured using both wet and dry sieving methods. Results showed that applying biochar significantly decreased the percentage of aggregate destruction and soil fractal dimension and increased the MWD and GMD. The optimal amelioration was observed when biochar was applied at a rate of 40 t ha−1. The decline of the fractal dimension of dry aggregates was 2–9 times as much as that of water-stable aggregates in the 0–15 soil layer and 1–4 times in the 15–30 cm soil layer. These results suggested that biochar could improve the resistance of aggregates to stresses and provide scientific strategies for the agricultural production.

Returning crop straw into fields is a typical agricultural practice to resolve an oversupply of straw and improve soil fertility. Soil microorganisms, especially eukaryotic microorganisms, play a critical role in straw decomposition. To date, microbial communities in response to straw amendment at different moisture levels in Chinese fluvo-aquic soil are poorly understood. The aim of this study was to explore the effects of straw amendment and moisture on microbial communities in Chinese fluvo-aquic soil. Two soils (one was applied with organic manure, and the other was not applied with any fertilizer) from a long-term field experiment in the North China Plain were collected. Soils with and without straw amendment at 25 and 55 % of the average water-holding capacities of the two soils were incubated at 25 °C for 80 days. All treatments were sampled 20 and 80 days after the start of incubation. Microbial biomass and community structure were analyzed by phospholipid fatty acids (PLFA) assay, and the eukaryotic diversity and community composition were assessed via barcoded pyrosequencing of the 18S ribosomal RNA (rRNA) gene amplicons. PLFA analysis showed that straw amendment increased the biomass of Gram-positive bacteria, Gram-negative bacteria, actinobacteria, and fungi and shifted microbial community structure. The varied straw availability resulted in a large variation in microbial community structure. In the presence of straw, actinobacterial and fungal biomass both decreased under high moisture content. 18S rRNA gene pyrosequencing indicated that straw amendment decreased eukaryotic diversity and richness and probably restructured the eukaryotic community. Under identical moisture content, long-term organic manure-fertilized soil had higher eukaryotic diversity and richness than the unfertilized soil. In the amended soils under high moisture content, the relative abundance of dominant fungal taxa (Dikarya subkingdom, Ascomycota phylum, and Pezizomycotina subphylum) decreased. Straw amendment increases microbial biomass, shifts microbial community structure, and decreases eukaryotic diversity and richness. High moisture content probably has a negative effect on fungal growth in the amended soils. In conclusion, microbial communities in Chinese fluvo-aquic soil are significantly affected by straw amendment at different moisture levels.

The backward Lagrangian stochastic (BLS) model and open-path tunable diode laser (OPTDL) analyzer were used to monitor ammonia (NH3) emissions from urea applied to winter wheat in the North China Plain. The high-temporal resolution measurements of ammonia concentrations provided an opportunity for estimating the diel patterns of ammonia emissions, as well as valuable information about the factors that influence NH3 emissions. The results showed both large diel variability and daily variability in NH3 volatilization, with NH3 emissions highest during the daytime. The diel pattern of ammonia volatilization depended mainly on the diel variation of wind speed and soil temperature, while the overall pattern of NH3 loss was strongly affected by soil moisture content, soil NH4 +-N concentration, wind speed and soil temperature. At the end of the measurement period, the cumulative NH3 loss was 12.21–16.43 kg N ha−1, calculated based on different time scale average Q BLS. Due to sensitivity of the OPTDL analyzer, the estimated total ammonia loss was still doubtful in this study.

Continuous application of organic and inorganic fertilizers can affect soil and food quality with respect to heavy metal concentrations. The risk of cadmium (Cd) contamination in a long-term (over 20 years) experimental field in North China with an annual crop rotation of winter wheat and summer maize was investigated. The long-term experiment had a complete randomized block design with seven fertilizer treatments and four replications. The seven fertilizer treatments were (1) organic compost (OM), (2) half organic compost plus half chemical fertilizer (OM + NPK), (3) NPK fertilizer (NPK), (4–6) chemical fertilizers without one of the major nutrients (NP, PK, and NK), and (7) an unamended control (CK). Soil samples from 0 to 20 cm were collected in 1989, 1999, and 2009 to characterize Cd and other soil properties. During the past 20 years, various extents of Cd accumulation were observed in the soil, and the accumulation was mainly affected by atmospheric dry and wet deposition and fertilization. In 2009, the average Cd concentration in the soil was 148 ± 15 μg kg−1 and decreased in the order of NPK ≈ OM + NKP ≈ PK > NP ≈ NK > OM ≈ CK. Sequential extraction of Cd showed that the acid-soluble fraction (F1, 32 ± 7 %) and the residual fraction (F4, 31 ± 5 %) were the dominant fractions of Cd in the soil, followed by the reducible fraction (F2, 22 ± 5 %) and oxidizable fraction (F3, 15 ± 6 %). The acid-soluble Cd fraction in the soil and Cd accumulation in the crops increased with soil plant available K. Fraction F3 was increased by soil organic C (SOC) and crop yields, but SOC reduced the uptake of soil Cd by crops. The long-term P fertilization resulted in more Cd buildup in the soil than other treatments, but the uptake of Cd by crops was inhibited by the precipitation of Cd with phosphate in the soil. Although soil Cd was slightly increased over the 20 years of intensive crop production, both soil and grain/kernel Cd concentrations were still below the national standards for environmental and food safety.

A viable method—open-path tunable diode laser absorption spectroscopy (OPTDL) in conjunction with a backward Lagrangian stochastic (bLS) dispersion model—has been used for micrometeorological monitoring of ammonia fluxes. In this technique, the gas concentration measured with the OPTDL sensor is used to infer the surface emission rate with the aid of dispersion model calculations. On the basis of numerous assessment experiments and field trials, several beneficial strategies for using the OPTDL technique properly to monitor atmospheric NH3 concentrations in the field have been summarized. Theoretically, the location of the concentration measurement can be anywhere in the emission plume, but in practice, the concentration measurement position must be carefully selected to avoid making measurements which are on the periphery of the downwind plume or are affected by obstructions. To obtain accurate estimates, periods with low friction velocity or extreme atmospheric stability, where Monin–Obukhov similarity theory-based relationships are invalid, or unrepresentative estimates due to unsuitable wind direction, should be excluded. A validation experiment showed that there was no significant difference between the ammonia emission rates obtained by the micrometeorological mass balance method and those obtained by the bLS model combined with the OPTDL technique. This study also indicated the potential of the bLS and OPTDL technique for investigation of diurnal emission patterns and environmental influences.

Differences in the composition of a bacterial community engaged in decomposing wheat straw in a fluvo-aquic soil at 15 °C, 25 °C and 35 °C were identified using barcode pyrosequencing. Functional carbon groups in the decomposing wheat straw were evaluated by 13C-NMR (nuclear magnetic resonance). Actinobacteria and Firmicutes were more abundant, whereas Alphaproteobacteria and Bacteroidetes were less abundant, at higher temperatures during the later stages of decomposition. Differences in the chemical properties of straw accounted for 19.3% of the variation in the community composition, whereas soil properties accounted for more (24.0%) and temperature, for less (7.4%). Carbon content of the soil microbial biomass and nitrogen content of straw were significantly correlated with the abundance of Alphaproteobacteria, Actinobacteria and Bacteroidetes. The chemical properties of straw, especially the NCH/OCH3, alkyl O-C-O and O-alkyl functional groups, exercised a significant effect on the composition of the bacterial community at different temperatures during decomposition—results that extend our understanding of bacterial communities associated with the decomposition of straw in agro-ecosystems and of the effects of temperature and chemical properties of the decomposing straw and soil on such communities.

Arbuscular mycorrhizal (AM) fungi are crucial for ecosystem functioning and can contribute to the formation and maintenance of soil aggregates through the exudation of glomalin by extraradical hyphae. Monitoring fertilization effects on AM fungi may help us to develop sound management strategies. The objectives of this study were to investigate the impacts of long-term fertilization on AM fungal parameters and to find out the key factor that affects the diversity and function of AM fungi. In this study, a regional irrigation schedule optimization method was proposed and applied in Fengqiu County in the North China Plain, which often suffers serious soil water drainage and nitrogen (N) leaching problems caused by excessive irrigation. The irrigation scheduling method was established by integrating the ‘checkbook irrigation method’ into a GIS-coupled soil water and nitrogen management model (WNMM) as an extension. The soil water and crop information required by the checkbook method, and previously collected from field observations, was estimated by the WNMM. By replacing manually observed data with simulated data from WNMM, the application range of the checkbook method could be extended from field scale to regional scale. The WNMM and the checkbook irrigation method were both validated by field experiments in the study region. The irrigation experiment in fluvo–aquic soil showed that the checkbook method had excellent performance; soil water drainage and N leaching were reduced by 83.1 and 85.6%, respectively, when compared with local farmers’ flood irrigation. Using the validated WNMM, the performance of checkbook irrigation in an entire winter wheat and summer maize rotation was also validated: the average soil water drainage and N leaching in four types of soils decreased from 331 to 75 mm year−1 and 47.7 to 9.3 kg ha−1 year−1, respectively; and average irrigation water use efficiency increased from 26.5 to 57.2 kg ha−1 mm−1. The regional irrigation schedule optimization method based on WNMM was applied in Fengqiu County. Knowledge about shifts of microbial community structure and diversity following different agricultural management practices could improve our understanding of soil processes and thus help us to develop sound management strategies. A long-term fertilization experiment was established in 1989 at Fengqiu (35°00′N, 114°24′E) in northern China. The soil (sandy loam) is classified as aquic inceptisols and has received continuous fertilization treatments since then. The fertilization treatments included control (CK, no fertilizer), chemical fertilizers nitrogen (N) and potassium (K) (NK), phosphorous (P) and K (PK), NP, NPK, organic manure (OM), and half chemical fertilizers NPK plus half organic manure (1/2NPKOM). The objective of this study was to examine if the microbial community structure and diversity were affected by the long-term fertilization regimes. Soil samples were collected from the long-term experimental plots with seven treatments and four replications in April 2006. Microbial DNAs were extracted from the soil samples and the 16S rRNA genes were PCR amplified. The PCR products were analyzed by DGGE, cloning and sequencing. The bacterial community structures and diversity were assessed using the DGGE profiles and the clone libraries constructed from the excised DGGE bands. The bacterial community structure of the OM and PK treatments were significantly different from those of all other treatments. The bacterial community structures of the four Ncontaining treatments (NK, NP, NPK and 1/2NPKOM), as well as CK, were more similar to each other. The changes in bacterial community structures of the OM and PK treatments showed higher richness and diversity. Phylogenetic analyses indicated that Proteobacteria (30.5%) was the dominant taxonomic group of the soil, followed by Acidobacteria (15.3%), Gemmatimonadetes (12.7%), etc. Irrespective of the two fertilization treatments of OM and PK, the cluster analysis showed that bacterial communities of the remaining five treatments of CK, NK, NP, NPK and 1/2NPKOM seemed to be more similar to each other, which indicated the relatively weak effects of the four N-containing treatments on soil bacterial communities. N fertilizer may be considered as a key factor to counteract the effects of other fertilizers on microbial communities. Our results show that long-term fertilization regimes can affect bacterial community structure and diversity of the agricultural soil. The OM and PK treatments showed a trend towards distinct community structures, higher richness and diversity when compared to the other treatments. Contrasting to the positive effects of OM and PK treatments on the bacterial communities, N fertilizer could be considered as a key factor in the soil to counteract the effects of other fertilizers on soil microbial communities. Because of the extremely high abundance and diversity of microorganisms in soil and the high heterogeneity of the soil, it is necessary to further examine the effects of fertilization regimes on microbial community and diversity in different type soils for comprehensively understanding their effects through the appropriate combination of molecular approaches.

Goal, Scope and Background Andisols are widespread in Japan and have some special properties such as high anion exchange capacity, low bulk density, and high organic matter content, which might influence the accumulation or chemical fractionation of heavy metals. However, few such data exist in Japanese andisols. The primary objective of this study was to investigate the distribution and chemical fractions of Cu, Zn, Ni, and Cr in the soil profiles and subsequently to assess their potential environmental hazard.

A pot experiment was conducted in a glasshouse to clarify and quantify the effect of plant part, water regime, growth period, and cultivar on carbon isotope discrimination (CID), and to analyze the relationship between CID, stomatal behavior and water-use efficiency (WUE). The experiment was comprised of two upland rice (Oryza sativa L.) cultivars and three water regimes (100, 70, and 40% of saturation moisture) in a completely randomized design. Plants were harvested at tillering, flowering, and maturity. No significant cultivar differences in above-ground dry matter-based WUE (WUEA) and total dry matter-based WUE (WUET) were observed. WUEA (and WUET) increased with water stress up to tillering, but decreased with water stress after tillering. Significant cultivar differences in CID in all the analyzed plant parts were observed at all harvest times. Reduction in CID with water stress was greatest at tillering, and the effect was less pronounced at flowering and at maturity. At each harvest, the effect was most pronounced in newly developed plant parts. Root and grain tended to have the lowest CID values, and stem the highest, at all harvest times. A negative relationship was observed between CID measured at tillering and WUEA (and WUET) measured over the period from seedling to tillering, whereas a reverse relationship was obtained between CID measured at flowering and WUEA (and WUET) measured over the period from tillering to flowering, and an unclear relationship between CID measured at maturity and WUEA (and WUET) measured over the period from flowering to maturity. The ratio of the intercellular and atmospheric concentration of CO2 (Ci/Ca) were closely associated with CID throughout the water regimes when one cultivar was considered, however, cultivar differences in CID were not related to variations in Ci/Ca. The results indicate that significant cultivar difference existed in CID in all the analyzed plant parts at all harvest times, while corresponding difference in WUEA (and WUET) between the cultivars was not necessarily consistent.
