Increasing evidences show that circRNAs are associated with some autoimmunity diseases either as a biomarker or therapeutic target. Exosomes containing nucleic acids and proteins are found in sera of series diseases and could serve as either diagnostic or therapeutic target. ANA serves as first common diagnostic test for autoimmunity disease, different ANA staining reflecting different types of autoimmunity disease. Till now, whether different ANA sera exosomes express different circRNAs and relevant ceRNA networks are still shortage of investigation. This study analyzed circRNAs, miRNAs, and their interaction networks in different ANA sera exosomes by high-throughput sequencing. It found no significant difference of total circRNAs and miRNAs amount across different ANA sera exosomes. However, significant differences were found of circRNAs, miRNA constituents, function analysis by KEGG and GO, and their ceRNA networks including miRNA-circRNA and miRNA-mRNA among different ANA sera exosomes, suggesting sera exosome circRNAs as either biomarker or mechanism of autoimmunity diseases.

As the predominant modification in RNA, N6-methyladenosine (m6A) has attracted increasing attention in the past few years since it plays vital roles in many biological processes. This chemical modification is dynamic, reversible and regulated by several methyltransferases, demethylases and proteins that recognize m6A modification. M6A modification exists in messenger RNA and affects their splicing, nuclear export, stability, decay, and translation, thereby modulating gene expression. Besides, the existence of m6A in noncoding RNAs (ncRNAs) could also directly or indirectly regulated gene expression. Colorectal cancer (CRC) is a common cancer around the world and of high mortality. Increasing evidence have shown that the changes of m6A level and the dysregulation of m6A regulatory proteins have been implicated in CRC carcinogenesis and progression. However, the underlying regulation laws of m6A modification to CRC remain elusive and better understanding of these mechanisms will benefit the diagnosis and therapy. In the present review, the latest studies about the dysregulation of m6A and its regulators in CRC have been summarized. We will focus on the crucial roles of m6A modification in the carcinogenesis and development of CRC. Moreover, we will also discuss the potential applications of m6A modification in CRC diagnosis and therapeutics.

The use of the anthelmintic drug pyrvinium pamoate (PP) in cancer therapy has been extensively investigated in the last decade. PP has been shown to have an inhibitory effect in colorectal cancer (CRC), but the underlying mechanism remains elusive. We aimed to investigate the antitumor activity and mechanisms of PP in CRC. In the present study, we used CCK-8 assays, colony formation assays, and western blotting to reveal that PP effectively suppressed CRC cell proliferation and the AKT-dependent signaling pathway in a concentration-dependent and time-dependent manner. Flow cytometric analysis and fluorescence microscopy demonstrated that PP increased intracellular reactive oxygen species (ROS) accumulation. We found that the inhibitory effect of PP on cell proliferation and AKT protein expression induced by PP could be partially reversed by N-acetyl-l-cysteine (NAC), an ROS scavenger. In addition, the results also demonstrated that PP inhibited cell migration by modulating epithelial-to-mesenchymal transition (EMT)-related proteins, including E-cadherin and vimentin. In conclusion, our data suggested that PP effectively inhibited cell proliferation through the ROS-mediated AKT-dependent signaling pathway in CRC, further providing evidence for the use of PP as an antitumor agent.

The existing depth video coding algorithms are generally based on in-loop depth filters, whose performance are unstable and easily affected by the outliers. In this paper, we design a joint weighted sparse representation-based median filter as the in-loop filter in depth video codec. It constructs depth candidate set which contains relevant neighboring depth pixel based on depth and intensity similarity weighted sparse coding, then the median operation is performed on this set to select a neighboring depth pixel as the result of the filtering. The experimental results indicate that the depth bitrate is reduced by about 9% compared with anchor method. It is confirmed that the proposed method is more effective in reducing the required depth bitrates for a given synthesis quality level.

In the era of big data, the gap between the fast increasing size of the surveillance video data and the relatively stable video compression rate has become prominent. Existing data compression methods based on the known redundancies are not able to eliminate the dominant redundancy embedded in the big surveillance video data (BSVD) of multiple sources in large time span. In this study, a new approach based on global redundancy of the BSVD has been explored. We first analyze the compositions of the global redundancy based on characteristics of BSVD. A coding framework is proposed to eliminate global redundancy. Simulated experiments have been performed to examine the performance of the approach in comparison with the high efficiency video coding. The experimental result showed that the proposed approach can reach a compression rate of 1/400 for a huge dataset of surveillance videos.

Sparse representation based face hallucination methods only utilize generic sparsity prior irrespective of the fact that the near bases in dictionary do make a greater contribution to patch reconstruction. Consequently, they cannot provide satisfactory hallucinated results in terms of accuracy and noise robustness. In this paper, instead of modifying the existing l 1 norm regularization, we propose a completely novel trend-constrained regularization method by exploiting a priori known as shape trend regarding coefficient and similarity curves. In particular, this method explicitly encourages the unknown coefficient curve to follow the shape variation trend of a predetermined similarity curve by means of maximizing their correlation. Owning to the differentiability of the underlying model, both the solution and regularization parameter can be analytically tractable, thus avoiding complex numerical optimization. Based on the observation on distance statistics, we further devise two optimal similarity functions in the form of power and exponential kernel functions. The proposed method boosts face hallucination performance considerably compared with the state-of-the-art methods. Experiments on commonly used face database demonstrate its effectiveness.

The reaction of 5,5'-(pyridine-2,6-diyl)bis(4-amino-3-mercapto-1,2,4-triazole) with various aromatic aldehydes in acetic acid yielded the corresponding 5,5'-(pyridine-2,6-diyl)bis(4-arylideneamino-3-mercapto-1,2,4-triazole) derivatives. The structures of the synthesized compounds as well as their intermediates were confirmed by elemental analysis, infrared spectra, (1)H NMR spectra and mass spectra studies. All the synthesized title compounds were screened for their antibacterial activities, and the preliminary results revealed that some of them showed good activities against Escherichia coli and Pseudomonas aeruginosa.

The structures of the newly synthesized compounds were confirmed by elemental analysis, 1H NMR and 13C NMR spectral, mass spectral, and Infrared spectral studies. All the synthesized target compounds have been investigated for their in vitro antibacterial activity, and the preliminary results revealed that the compounds 6b and 6k exhibited very promising activity against Escherichia coli and Pseudomonas aeruginosa.

In the applications of Free View TV, pre-estimated depth information is available to synthesize the intermediate views as well as to assist multi-view video coding. Existing view synthesis prediction schemes generate virtual view picture only from interview pictures. However, there are many types of signal mismatches caused by depth errors, camera heterogeneity or illumination difference across views and these mismatches decrease the prediction capability of virtual view picture. In this paper, we propose an adaptive learning based view synthesis prediction algorithm to enhance the prediction capability of virtual view picture. This algorithm integrates least square prediction with backward warping to synthesize the virtual view picture, which not only utilizes the adjacent views information but also the temporal decoded information to adaptively learn the prediction coefficients. Experiments show that the proposed method reduces the bitrates by up to 18 % relative to the multi-view video coding standard, and about 11 % relative to the conventional view synthesis prediction method.

The molecular processes underlying epidemic waves of methicillin-resistant Staphylococcus aureus (MRSA) infection are poorly understood(1). Although a major role has been attributed to the acquisition of virulence determinants by horizontal gene transfer(2), there are insufficient epidemiological and functional data supporting that concept. We here report the spread of clones containing a previously extremely rare(3,4) mobile genetic element–encoded gene, sasX. We demonstrate that sasX has a key role in MRSA colonization and pathogenesis, substantially enhancing nasal colonization, lung disease and abscess formation and promoting mechanisms of immune evasion. Moreover, we observed the recent spread of sasX from sequence type 239 (ST239) to invasive clones belonging to other sequence types. Our study identifies sasX as a quickly spreading crucial determinant of MRSA pathogenic success and a promising target for therapeutic interference. Our results provide proof of principle that horizontal gene transfer of key virulence determinants drives MRSA epidemic waves.

Background The Th17 subset and IL-17 have been found in increased frequencies within certain tumors. However, their relevance in cancer biology remains controversial. This study aimed to clarify the biological action of IL-17 on hepatocellular carcinoma (HCC). Methods Effects and underlying molecular mechanisms of IL-17 on human HCC were explored in vitro using exogenous IL-17 stimulation and in nude mice by implanting IL-17 overexpressed HCC cells. The clinical significance of IL-17 was investigated in tissue microarrays containing HCC tissues from 323 patients following hepatectomy using immunohistochemistry. Results Although exogenous IL-17 showed no direct effect on the growth rate of HCC cells in vitro, PCR and ELISA showed that IL-17 selectively augmented the secretion of diverse proinvasive factors and transwell showed a direct promotion of invasion of HCC cells by IL-17. Furthermore, transfection of IL-17 into HCC cells significantly promoted neoangiogenesis, neutrophil recruitment and tumor growth in vivo. Using siRNA mediated knockdown of AKT and STAT3, we suggested that the effects of IL-17 were operated through activation of the AKT signaling in HCC, which resulted in IL-6 production. Then, IL-6 in turn activated JAK2/STAT3 signaling and subsequently up-regulated its downstream targets IL-8, MMP2, and VEGF. Supporting these findings, in human HCC tissues, immunostaining indicated that IL-17 expression was significantly and positively associated with STAT3 phosphorylation, neutrophil infiltration and increased tumor vascularity. The clinical significance of IL-17 was authenticated by revealing that the combination of intratumoral IL-17+ cells and phospho-STAT3 served as a better prognosticator for postoperative tumor recurrence than either marker alone. Conclusions IL-17 mediated tumor-promoting role involves a direct effect on HCC cells through IL-6/JAK2/STAT3 induction by activating the AKT pathway.
