Highly asymmetric bimetal nanostructures, such as Au-Pt nanorings and Au-Ag nanoplates possess superior plasmonic properties owing to various synergistic effects between different components and diversified morphologies. Herein, we report a controllable growth of asymmetric Pt-Au nanoplates and Au@Pt nanorings with excellent photothermal conversion efficiency. Hexagonal Au nanoplates are used as templates, which is achieved through a transformation reaction from Au nanotriangles. Pt-Au nanoplates is prepared by a site-selective growth of Pt on the rim of obtained hexagonal Au nanoplates. Subsequently, Pt nanorings is obtained by a selective etching of Au, and a regrowth of Au on the Pt nanorings leads to bimetallic Au@Pt nanorings. The evolution of extinction spectra during the whole process is carefully studied. Under irradiation by 1064 nm laser located in the second near-infrared bio-window, the Pt-Au nanoplates exhibit excellent photothermal conversion, better than that of initial Au nanotriangles and hexagonal Au nanoplates with same mass concentration. The improvement of photothermal effect can be ascribed to the strong surface plasmon resonances and coupling between Au and Pt.

The optimization of positive end-expiratory pressure (PEEP) according to respiratory mechanics [driving pressure or respiratory system compliance (Crs)] is a simple and straightforward strategy. However, its validity to prevent postoperative pulmonary complications (PPCs) remains unclear. Here, we performed a meta-analysis to assess such efficacy. We searched PubMed, Embase, and the Cochrane Library to identify randomized controlled trials (RCTs) that compared personalized PEEP based on respiratory mechanics and constant PEEP to prevent PPCs in adults. The primary outcome was PPCs. Fourteen studies with 1105 patients were included. Compared with those who received constant PEEP, patients who received optimized PEEP exhibited a significant reduction in the incidence of PPCs (RR = 0.54, 95% CI 0.42 to 0.69). The results of commonly happened PPCs (pulmonary infections, hypoxemia, and atelectasis but not pleural effusion) also supported individualized PEEP group. Moreover, the application of PEEP based on respiratory mechanics improved intraoperative respiratory mechanics (driving pressure and Crs) and oxygenation. The PEEP titration method based on respiratory mechanics seems to work positively for lung protection in surgical patients undergoing general anesthesia.

Several scoring systems are currently used to predict prognosis of hepatocellular carcinoma (HCC), but none of them integrates liver function, systemic inflammation, and tumor characteristics in a unified model. The current study aimed to develop and validate a novel prognostic score that integrates liver function, systemic inflammation, and tumor characteristics in a unified model to predict the prognosis of HCC after curative resection.Patients with HCC who underwent curative liver resection were included in a training set (n = 1027). Multivariate Cox regression was performed to determine the risk factors for a poor prognosis. A prognostic score was developed by assigning points for risk factors in proportion to beta coefficients in a Cox multivariable model. Predictive performance and distinction ability of the prognostic score were further evaluated in two independent validation cohorts treated with either curative resection (n = 281) or transarterial chemoembolization (TACE) (n = 404) and compared with 16 other models.The prognostic predictive system, named the function-inflammation-burden-alpha-fetoprotein (FIBA) score, was derived by assigning points for six independent predictors including albumin, total bilirubin, lymphocyte count, diameter of the largest tumor, number of tumors, and alpha-fetoprotein (AFP). The FIBA score showed an outperformed predictive value compared with other systems in both training and validation cohorts by giving the highest C-index, likelihood ratio chi-square values, and Wald test values as well as the lowest Akaike information criterion.The FIBA score can be used to stratify HCC patients treated with curative resection. Meanwhile, the FIBA score performs well against other prognostic scoring systems and is potentially broadly applicable to a TACE-treated patient cohort.

Goldfish (Carassius auratus) have long fascinated evolutionary biologists and geneticists because of their diverse morphological and color variations. Recent genome-wide association studies have provided a clue to uncover genomic basis underlying these phenotypic variations, but the causality between phenotypic and genotypic variations have not yet been confirmed. Here, we edited proposed candidate genes to recreate phenotypic traits and developed a rapid biotechnology approach which combines gene editing with high-efficiency breeding, artificial gynogenesis, and temperature-induced sex reversal to establish homozygous mutants within two generations (approximately eight months). We first verified that low-density lipoprotein receptor-related protein 2B (lrp2aB) is the causal gene for the dragon-eye variation and recreated the dragon-eye phenotype in side-view Pleated-skirt Lion-head goldfish. Subsequently, we demonstrated that the albino phenotype was determined by both homeologs of oculocutaneous albinism type II (oca2), which has subfunctionalized to differentially govern melanogenesis in the goldfish body surface and pupils. Overall, we determined two causal genes for dragon-eye and albino phenotypes, and created four stable homozygous strains and more appealing goldfish with desirable traits. The developed biotechnology approach facilitates precise genetic breeding, which will accelerate re-domestication and recreation of phenotypically desirable goldfish.

Erenumab, an anti-calcitonin gene-related peptide (CGRP) receptor monoclonal antibody (mAb), was approved by the US Food and Drug Administration in May 2018. Constipation with serious complications was added to the Warning and Precautions section in the erenumab Prescribing Information in October 2019 after events were observed during post-marketing surveillance. We aimed to assess and compare the risk of inpatient constipation, and, separately, inpatient constipation with serious complications, among patients with migraine treated with CGRP mAbs and standard of care antiepileptic drugs (AEDs).Within Optum's Electronic Health Record Research Database, patients with migraine who initiated erenumab, other CGRP mAbs, and AEDs were identified from May 2018 through March 2020. Erenumab initiators were propensity score-matched separately to initiators of other CGRP mAbs and AEDs. Incident inpatient constipation events, and serious complications, were identified using multiple risk windows for outcome assessment (30-, 60-, 90-day risk windows, and all available follow-up). Odds ratios (ORs) were calculated comparing inpatient constipation risk among matched erenumab initiators relative to comparators.We identified 17,902 erenumab, 13,404 other CGRP mAb, and 49,497 AED initiators who met study criteria. Among matched initiators, the risk of inpatient constipation was 0.46% (95% confidence interval (CI) 0.35-0.60) for erenumab and 0.44% (95% CI 0.33-0.58) for other CGRP mAbs within the 90-day risk window, with a corresponding OR of 1.06 (95% CI 0.72-1.55). Among matched erenumab and AED initiators, inpatient constipation risk was 0.53% (95% CI 0.42-0.66) and 0.76% (95% CI 0.62-0.92), respectively, and the OR was 0.69 (95% CI 0.51-0.94). Few serious complications were observed.Patients initiating erenumab had similar risk of inpatient constipation within 90 days of treatment initiation versus patients initiating other CGRP mAbs, and lower risk versus patients initiating AEDs. These findings provide context to events observed during post-marketing surveillance.

In this study, the pollution status of polychlorinated biphenyls (PCBs) and organochlorine pesticides (OCPs) was investigated in indoor and outdoor dust from three different functional areas of Hefei, China. The relationship between the concentrations of PCBs and OCPs and different influencing factors in dwellings was studied. The results showed that the concentrations of PCBs and OCPs were higher in samples from dwellings with higher smoking frequency, lower cleaning frequency, higher floors and smaller household size. The results of Spearman's correlation coefficient analysis indicated that PCBs and OCPs were not consistently associated with each other, while sources of low-chlorinated PCBs and high-chlorinated PCBs were different. Scanning electron microscopy (SEM) shows the shape of indoor dust was a mixture of blocky, flocculated, spherical structures, and irregular shapes. Few studies have evaluated the association between air pollutants and neural tube defects (NTDs). Moreover, the existing research ignores the lag effect of air pollution on health and provides inconsistent epidemiological evidence. We aim to estimate the association between air pollution and NTDs during the first trimester of pregnancy and identify specific susceptible windows. Birth data was collected from the Birth Defects Surveillance Network in Lanzhou from September 1, 2014, to December 31, 2019. Air quality and meteorological data were collected from ambient air monitoring stations and China Meteorological Data Network. The log connection function of the Poisson distribution function is used to establish a DLNM model to estimate the exposure-effect relationship and exposure-lag relationship association between air pollutants levels and NTDs. As one of the five great lakes in China, the Chaohu Lake Basin is the main water source for regional economic and ecological development in Hefei city and is considered a source of drinking water. The spatial-temporal distributions and occurrence of soluble trace elements were studied in the surrounding ten rivers in the Chaohu Lake Basin as well as water quality and risk assessment during the normal and wet seasons. Principal component analysis (PCA) showed that the main source of river pollution during the two seasons was the mining industry. High values of most elements were found in the northwestern rivers in the two seasons. The temporal changes in the elements showed that the distributions of As, Mn, Cd, and Cu in the two seasons were very different, but the trends of Ni, Co, and V were basically the same. The sodium adsorption ratio (SAR) showed that almost all river samples needed to be treated before irrigation, and the water quality index (WQI) showed that most samples were of excellent water quality for drinking. The ecological risk assessment results showed that the risks in the two seasons were all slight. The results of the health risk evaluation suggested that no noncarcinogenic risks were found in the normal season and that the carcinogenic risks from Cr and As reached their highest levels in the normal and wet seasons, respectively. This research can provide vital data for rational water control and water quality conservation, offer a scientific basis for ecological environment safety, and offer a reference for carcinogenic and noncarcinogenic health risks to regional residents.

Triploids are rare in nature because of difficulties in meiotic and gametogenic processes, especially in vertebrates. The Carassius complex of cyprinid teleosts contains sexual tetraploid crucian carp/goldfish (C. auratus) and unisexual hexaploid gibel carp/Prussian carp (C. gibelio) lineages, providing a valuable model for studying the evolution and maintenance mechanism of unisexual polyploids in vertebrates. Here we sequence the genomes of the two species and assemble their haplotypes, which contain two subgenomes (A and B), to the chromosome level. Sequencing coverage analysis reveals that C. gibelio is an amphitriploid (AAABBB) with two triploid sets of chromosomes; each set is derived from a different ancestor. Resequencing data from different strains of C. gibelio show that unisexual reproduction has been maintained for over 0.82 million years. Comparative genomics show intensive expansion and alterations of meiotic cell cycle-related genes and an oocyte-specific histone variant. Cytological assays indicate that C. gibelio produces unreduced oocytes by an alternative ameiotic pathway; however, sporadic homologous recombination and a high rate of gene conversion also exist in C. gibelio. These genomic changes might have facilitated purging deleterious mutations and maintaining genome stability in this unisexual amphitriploid fish. Overall, the current results provide novel insights into the evolutionary mechanisms of the reproductive success in unisexual polyploid vertebrates.
