Background Anaplastic lymphoma kinase (ALK) overexpression and gene alterations have been detected in several mesenchymal tumors, with significant implications for diagnosis, therapy and prognosis. However, few studies have investigated the correlation between ALK expression status and clinicopathological characteristics in patients with gastrointestinal stromal tumors (GISTs). Methods A total of 506 GIST patients were enrolled. Sanger sequencing was employed to detect c-KIT and PDGFRA gene mutations. The tissue microarray (TMA) technique and immunohistochemistry were employed to identify the ALK (clone: 1A4 and D5F3) expression status in the tumor tissues. The ALK gene variants of IHC-positive cases were analyzed by fluorescence in situ hybridization (FISH) and next-generation sequencing (NGS). The clinicopathological data were analyzed using SPSS Statistics 26.0. Results Among the 506 GIST patients, the c-KIT mutation accounted for 84.2% (426/506), followed by PDGFRA mutation (10.3%, 52/506), while the wild-type accounted for the least (5.5%, 28/506). ALK-positive expression was detected in PDGFRA-mutant GISTs (7.7%, 4/52) but negative for c-KIT-mutant or wild-type GISTs by IHC. Four ALK IHC-positive patients were all male. The tumors all occurred outside the stomach. The predominant patterns of growth were epithelioid (2/4), spindle (1/4), and mixed type (1/4). They were all identified as high-risk classification according to the National Institutes of Health (NIH) classification. Aberrant ALK mutations were not identified by DNA-based NGS except in one of the 4 cases with amplification by FISH. Conclusion Our study revealed 7.7% (4/52) of ALK expression in PDGFRA-mutant GISTs, indicating that molecular tests were required to rule out the possibility of PDGFRA-mutant GISTs when encountering ALK-positive mesenchymal tumors with CD117-negative or weakly positive in immunohistochemical staining.

Background Macro-autophagy/Autophagy is an evolutionarily well-conserved recycling process to maintain the balance through precise spatiotemporal regulation. However, the regulatory mechanisms of biomolecular condensates by the key adaptor protein p62 via liquid-liquid phase separation (LLPS) remain obscure. Results In this study, we showed that E3 ligase Smurf1 enhanced Nrf2 activation and promoted autophagy by increasing p62 phase separation capability. Specifically, the Smurf1/p62 interaction improved the formation and material exchange of liquid droplets compared with p62 single puncta. Additionally, Smurf1 promoted the competitive binding of p62 with Keap1 to increase Nrf2 nuclear translocation in p62 Ser349 phosphorylation-dependent manner. Mechanistically, overexpressed Smurf1 increased the activation of mTORC1 (mechanistic target of rapamycin complex 1), in turn leading to p62 Ser349 phosphorylation. Nrf2 activation increased the mRNA levels of Smurf1, p62, and NBR1, further promoting the droplet liquidity to enhance oxidative stress response. Importantly, we showed that Smurf1 maintained cellular homeostasis by promoting cargo degradation through the p62/LC3 autophagic pathway. Conclusions These findings revealed the complex interconnected role among Smurf1, p62/Nrf2/NBR1, and p62/LC3 axis in determining Nrf2 activation and subsequent clearance of condensates through LLPS mechanism.

Convolutional neural networks (CNN) based object detection usually assumes that training and test data have the same distribution, which, however, does not always hold in real-world applications. In autonomous vehicles, the driving scene (target domain) consists of unconstrained road environments which cannot all possibly be observed in training data (source domain) and this will lead to a sharp drop in the accuracy of the detector. In this paper, we propose a domain adaptation framework based on pseudo-labels to solve the domain shift. First, the pseudo-labels of the target domain images are generated by the baseline detector (BD) and optimized by our data optimization module to correct the errors. Then, the hard samples in a single image are labeled based on the optimization results of pseudo-labels. The adaptive sampling module is approached to sample target domain data according to the number of hard samples per image to select more effective data. Finally, a modified knowledge distillation loss is applied in the retraining module, and we investigate two ways of assigning soft-labels to the training examples from the target domain to retrain the detector. We evaluate the average precision of our approach in various source/target domain pairs and demonstrate that the framework improves over 10% average precision of BD on multiple domain adaptation scenarios on the Cityscapes, KITTI, and Apollo datasets.

Background During cataract phacoemulsification surgery, an Intrepid® balanced (IB) tip can achieve a larger amplitude, which may lead to higher energy efficiency than a Kelman (K) tip when paired with a torsional phaco platform. In this retrospective cohort study, we compared their energy efficiency and damage to the cornea under a new energy setting. Methods The medical records of 104 eyes of 79 patients were reviewed, with 47 eyes belonging to the IB group and 57 eyes belonging to the K group. All surgeries were performed on an Alcon Centurion® platform with gravity infiltration. Surgical parameters, visual outcome, central corneal thickness (CCT) changes, and endothelial cell density (ECD) loss rate were recorded and calculated. Results No significant differences in postoperative best corrected visual acuity (BCVA), intraocular pressure (IOP), total ultrasound time, estimated fluid aspirated, CCT changes, or ECD loss rate were observed between the two groups. We divided the included eyes into soft nucleus and hard nucleus subgroups and found lower cumulative dissipated energy (CDE, 8.15 ± 8.02 vs 14.82 ± 14.16, P = 0.023), cumulative torsional energy (CTE, 8.06 ± 7.87 vs 14.13 ± 13.02, P = 0.027), and cumulative longitudinal energy (CLE, 0.09 ± 0.17 vs 0.69 ± 1.37, P = 0.017) in the IB group than in the K group, implying less energy used and higher energy efficiency of the IB tip. Conclusion Lower CLE in the IB group indicates fewer phaco tip obstructions and a significantly higher capability to conquer hard nuclei with IB tips with statistical significance. With an ultra-perfusion cannula, the balanced tip does not cause more corneal damage.

Evasion of growth suppression is among the prominent hallmarks of cancer. Phosphatase and tensin homolog (PTEN) and p53 tumor-suppressive pathways are compromised in most human cancers, including glioblastoma (GB). Hence, these signaling pathways are an ideal point of focus for novel cancer therapeutics. Recombinant viruses can selectivity kill cancer cells and carry therapeutic genes to tumors. Specifically, oncolytic viruses (OV) have been successfully employed for gene delivery in GB animal models and showed potential to neutralize immunosuppression at the tumor site. However, the associated systemic immunogenicity, inefficient transduction of GB cells, and inadequate distribution to metastatic tumors have been the major bottlenecks in clinical studies. Mesenchymal stem cells (MSCs), with tumor-tropic properties and immune privilege, can improve OVs targeting. Remarkably, combining the two approaches can address their individual issues. Herein, we summarize findings to advocate the reactivation of tumor suppressors p53 and PTEN in GB treatment and use MSCs as a "Trojan horse" to carry oncolytic viral cargo to disseminated tumor beds. The integration of MSCs and OVs can emerge as the new paradigm in cancer treatment.

Heat stress (HS) results in health problems in animals. This study was conducted to investigate the effect and the underlying mechanism of HS on the proliferation and differentiation process of 3T3-L1 preadipocytes. 3T3-L1 preadipocytes were treated at 37 °C or 41.5 °C. HS up-regulated the mRNA and protein expression level of heat shock protein 70 (HSP70). Furthermore, the proliferation of 3T3-L1 preadipocytes were significantly inhibited after HS treatment for 2 days. A large number of accumulated lipid droplets were observed under the microscope after HS treatment for 8 days. Notably, the result of oil red O staining showed that the number of lipid droplets increased significantly and the differentiation ability of the cells was enhanced after HS. Moreover, after 2 and 8 d of differentiation, HS increased the transcription levels of fat synthesis genes including peroxisome proliferators activated receptor γ (PPARγ), fatty acid binding protein 2 (AP2), fatty acid synthase (FAS) and CCAAT enhancer binding protein α (CEBPα) genes, while decreasing the transcription levels of lipid decomposition genes including ATGL and HSL genes. In addition, HS reduced the expression of AMPK and PGC-1α, as well as the dephosphorylation of AMPK. 5-Aminoimidazole-4-carboxamide ribonucleotide (AICAR) can eliminate HS induced lipogenesis by activating AMPK. These results indicated that HS inhibited the proliferation of 3T3-L1 preadipocytes and promoted lipid accumulation by inhibiting the AMPK-PGC-1α signaling pathway in 3T3-L1 preadipocytes. This work lays a theoretical foundation for improving the effect of HS on meat quality of livestock and provides a new direction for the prevention of obesity caused by HS.

Improvements to the dynamic characteristics and stiffness of composite girder bridges are necessary to expand their applications and capabilities. The natural frequency of a bridge directly reflects the dynamic characteristics of a structure. From the calculation principle for the natural frequency, the natural frequencies of a bridge can be significantly increased if the load increases slightly while the structural stiffness increases significantly. Based on this, a new type of concrete-steel composite truss bridge is proposed. The new bridge has a reasonable new arrangement for the position of the concrete and steel, according to the bending moment law for continuous girders. The mechanical analysis of the new bridge shows that it has the characteristic of adding a small amount of load at a reasonable position, which can significantly improve the stiffness. Therefore, it is possible to achieve the purpose of improving the dynamic characteristics so that they are far better than those of a conventional composite truss bridge. To further study the dynamic characteristics of the new composite truss bridge, the finite element (FE) method was used to analyse the new bridges, and the results are compared with those of conventional composite truss bridges. Furthermore, several parameters that influence the dynamic characteristics of the new composite truss bridge were studied. Finally, the dynamic characteristics of the small-span new bridge were studied by finite element analysis (FEA) and experiment. The results show that the new composite truss bridges with different span arrangements and different second-stage dead loads always have better natural frequencies and stiffness advantages. Therefore, the new composite truss bridge is suitable for girder bridges with strict requirements for their stiffness and dynamic characteristics, especially for super-long span high-speed railway girder bridges.

Endometriosis is a common disease in women, which impairs the quality of life in patients. Recently, accumulating evidences reported that miRNAs play an essential role in diagnosis and treatment of endometriosis. However, the regulatory mechanism of miRNAs has not been fully explored. The expression of miR-17-5p and VEGFA was detected using qRT-PCR. The protein level of VEGFA was measured via Western blot. Cell proliferation was determined by CCK-8 assay. Cell migration and invasion were measured via transwell assay. The relationship of miR-17-5p and VEGFA were verified via luciferase reporter assay. Then miR-17-5p was remarkably down-regulated in endometriosis tissues, serums and cells, and overexpression of miR-17-5p inhibited cell proliferation, migration and invasion in endometriosis. Results showed that VEGFA was significantly up-regulated in endometriosis tissues and cells and acted as a target of miR-17-5p. Moreover, miR- 17-5p negatively regulated VEGFA expression in endometriosis. Otherwise, up-regulation of VEGFA improved cell proliferative, migrated and invasive ability in ECSCs with transfection of miR-17-5p mimics group. Our data showed miR-17-5p inhibits cell proliferation, migration and invasion in endometriosis by directly repressing VEGFA expression.

Intelligent vehicle greatly benefits traffic safety, efficiency and driving comfortable. With the development of intelligent driving technology and its application, it is becoming increasingly important to do effective and comprehensive tests before putting on the market. Comprehensively considering the cost of test, an automatic generation method of test scenarios is proposed to ensure both coverage and effectiveness. Based on the analyzed key infuence factors of an intelligent driving system, the analytic hierarchy process (AHP) is used to determine their importance and accordingly an complex index is defined, based on which an improved test case generation algorithm based on the pairwise independent combinatorial testing tool (PICT) is proposed to ensuring both combinational coverage and complexity of test cases. Finally, the test scenario is generated by clustering these discrete test cases considering similarity and complexity. The high complex test cases are preferred to be combined together and conducted preferentially to increase the test efficiency. The effectiveness of this method is validated by applying it on a lane departure warning system (LDW).

Direct contact membrane distillation technology was considered as a promising and efficient technology for the treatment of textile wastewater. In this study, hydrophobic polytetrafluoroethylene and polyvinylidene fluoride membranes for the treatment of selected model compounds of textile wastewater (e.g., phenol, aniline, and sulfanilic acid) were explored comparatively in a bench-scale direct contact membrane distillation technology test unit. The effect of various operational parameters including temperature, flow rate, and concentration on the rejection performance was investigated systematically. The results indicated that an increased feed temperature and a faster cross flow velocity contributed positively to the direct contact membrane distillation performance. Limited rejection for phenol and aniline was witnessed, which can be due to their relatively lower boiling point. A > 99% of sulfanilic acid rejection was obtained under the same conditions. Furthermore, the polytetrafluoroethylene membrane always presented enhanced performance compared with the polyvinylidene fluoride samples. In brief, the direct contact membrane distillation process could be potentially used as a promising technique for the treatment of textile wastewater.

To verify the pre-earthquake ionospheric anomaly (PEIA), statistical analyses are implemented on the relationship between the total electron content (TEC) of global ionosphere map (GIM) and 62 M ≥ 6.0 earthquakes in China during 1998 - 2015. A median-based method together with z test is employed to determine the criteria and/or characteristics of TEC anomalies related to earthquakes. It is found that the GIM TEC significantly decreases at 18:00 - 22:00 UT (universal time, post-midnight to pre-dawn) 4 - 5 days before 37 6.0 ≤ M < 6.5 earthquakes, at 01:00 - 04:00 UT (morning) 3 - 6 days before 18 6.5 ≤ M < 7.0 earthquakes, and 04:00 - 10:00 UT (pre-noon to afternoon) 3 - 5 days; but increases 08:00 - 12:00 UT (late afternoon to early evening), 18 - 20 days before 7 M ≥ 7.0 earthquakes. The receiver operating characteristic (ROC) curve is used to compare the TEC anomaly-based method with some competitive alternatives for predicting the earthquakes under study. We found, based on possible TEC anomalies, that the observed PEIAs are significantly earthquake-related. Moreover, the results of regression analyses show that the PEIA strength is associated with the magnitude of earthquakes.

Alterations in cellular ubiquitin (Ub) homeostasis, known as Ub stress, feature and affect cellular responses in multiple conditions, yet the underlying mechanisms are incompletely understood. Here we report that autophagy receptor p62/sequestosome-1 interacts with E2 Ub conjugating enzymes, UBE2D2 and UBE2D3. Endogenous p62 undergoes E2-dependent ubiquitylation during upregulation of Ub homeostasis, a condition termed as Ub+ stress, that is intrinsic to Ub overexpression, heat shock or prolonged proteasomal inhibition by bortezomib, a chemotherapeutic drug. Ubiquitylation of p62 disrupts dimerization of the UBA domain of p62, liberating its ability to recognize polyubiquitylated cargoes for selective autophagy. We further demonstrate that this mechanism might be critical for autophagy activation upon Ub+ stress conditions. Delineation of the mechanism and regulatory roles of p62 in sensing Ub stress and controlling selective autophagy could help to understand and modulate cellular responses to a variety of endogenous and environmental challenges, potentially opening a new avenue for the development of therapeutic strategies against autophagy-related maladies.

Adsorption and desorption of hydrogen on/from single-vacancy and double-vacancy graphenes were studied by means of first-principles calculations. The structure and stability of continuous hydrogenation in single vacancy were investigated. Several new stable structures were found, along with their corresponding energy barriers. In double-vacancy graphene, the preferred sites of H atoms were identified, and H2 molecule desorption and adsorption of from/on were calculated from the energy barriers. This work provides a systematic and comprehensive understanding of hydrogen behavior on defected graphene.

In the penumbra after focal cerebral ischemia, an increase of protease Omi is linked to a decrease of Hs1-associated protein X-1 (Hax-1), a protein belonging to the Bcl-2 family. In this study we investigated the mechanisms underlying the regulation of Hax-1 by protease Omi in cerebral ischemia/reperfusion (I/R) injury. Mouse neuroblastoma N2a cells were subjected to oxygen-glucose deprivation and reoxygenation (OGD/R); cell viability was assessed with MTT assay. Mice underwent 2-h middle cerebral artery occlusion (MCAO) and reperfusion, and the infarct volume was determined with TTC staining. The expression of Omi and Hax-1 was detected using immunoblot and immunofluorescence assays. The mitochondrial membrane potential was measured using TMRM staining. In the brains of MCAO mice, the protein level of Omi was significantly increased, while the protein level of Hax-1 was decreased. Similar changes were observed in OGD/R-treated N2a cells, but the mRNA level of Hax-1 was not changed. Furthermore, in OGD/R-treated N2a cells, knockdown of Omi significantly increased Hax-1 protein level. Immunofluorescence assay showed that Omi and Hax-1 were co-localized in mitochondria of N2a cells. OGD/R caused marked mitochondrial damage and apoptosis in N2a cells, while inhibition of Omi protease activity with UCF-101 (10 μmol/L) or overexpression of Hax-1 could restore the mitochondrial membrane potential and attenuate cell apoptosis. Moreover, pretreatment of MCAO mice with UCF-101 (7.15 mg/kg, ip) could restore Hax-1 expression, inhibit caspase activation, and significantly reduce the infarct volume. Protease Omi impairs mitochondrial function by cleaving Hax-1, which induces apoptosis in OGD/R-treated N2a cells and causes I/R injury in MCAO mice.

The grass carp is an important farmed fish, accounting for similar to 16% of global freshwater aquaculture, and has a vegetarian diet. Here we report a 0.9-Gb draft genome of a gynogenetic female adult and a 1.07-Gb genome of a wild male adult. Genome annotation identified 27,263 protein-coding gene models in the female genome. A total of 114 scaffolds consisting of 573 Mb are anchored on 24 linkage groups. Divergence between grass carp and zebrafish is estimated to have occurred 49-54 million years ago. We identify a chromosome fusion in grass carp relative to zebrafish and report frequent crossovers between the grass carp X and Y chromosomes. We find that transcriptional activation of the mevalonate pathway and steroid biosynthesis in liver is associated with the grass carp's adaptation from a carnivorous to an herbivorous diet. We believe that the grass carp genome could serve as an initial platform for breeding better-quality fish using a genomic approach.

Objective To observe the effect of acupoint massage in relieving pain after ureteroscopic holmium laser lithotripsy.

Long-term treatment with an agonist of peroxisome proliferator-activated receptor (PPAR)-γ is associated with bone fractures in the clinical practice. However, the mechanisms underlying the fractures are not fully understood. This study was aimed to examine the effect of rosiglitazone (an agonist of PPAR-γ) of different doses on the proliferation, differentiation, and transforming growth factor beta 1 (TGF-β1)-induced expression of connective tissue growth factor (CTGF) in primary rat osteoblasts in vitro. Osteoblasts were isolated from newly born SD rats and treated with different doses of rosiglitazone (0-20 μmol/L). The proliferation and differentiation of osteoblasts were measured by MTT assay and NPP assay, respectively. The expression of CTGF was determined by RT-PCR and Western blotting. The results showed that most isolated osteoblasts displayed strong alkaline phosphatase (ALP) activity and treatment with different doses of rosiglitazone did not affect their proliferation, but significantly inhibited the differentiation of osteoblasts in a dose-dependent manner. Moreover, treatment with different doses of rosiglitazone significantly reduced the TGF-β1-induced CTGF mRNA transcription and protein expression in a dose-dependent manner in rat osteoblasts. It was concluded that the activation of PPAR-γ may inhibit the differentiation of osteoblasts by reducing the TGF-β1-induced CTGF expression in vitro.

The K2O–B2O3–SiO2, K2O–B2O3–SiO2–2 %Al2O3, K2O–B2O3–SiO2–4 %Al2O3 glasses with different Al2O3 content were prepared. Different proportions (50, 55, 60, 65, 70 %) of the three glasses were respectively mixed with alumina ceramic-filler, then the mechanical and dielectric properties were investigated. The results showed K2O–B2O3–SiO2–2 %Al2O3 glass/alumina filler (glass:alumina = 60:40) had the excellent comprehensive properties, so further study was continued with part of alumina ceramic-filler replaced by the silica ceramic-filler on this composite. Then the X-ray diffraction analysis revealed that the alumina and silica fillers existed as the crystal phase, and the densification was seriously damaged when the silica content reached to three quarters of the fillers. With the increase of the silica-filler, the composites’ density and dielectric constant exhibited uniform decrease, but thermal expansion coefficient (TEC) uniformly increased. When the glass:alumina:silica was equal to 60:30:10, a best composite property was presented as a bulk density of 2.582 (g cm−1), a dielectric constant of 6.1 and a dielectric loss of 2 × 10−3 at 1 MHz, a flexural strength of 168 MPa, and a TEC of 8.62 × 10−6 °C−1.

The gut-associated lymphoid tissue, connected with liver via bile and blood, constructs a local immune environment of both defense and tolerance. The gut-liver immunity has been well-studied in mammals, yet in fish remains largely unknown, even though enteritis as well as liver and gallbladder syndrome emerged as a limitation in aquaculture. In this study, we performed integrative bioinformatic analysis for both transcriptomic (gut and liver) and proteomic (intestinal mucus and bile) data, in both healthy and infected tilapias. We found more categories of immune transcripts in gut than liver, as well as more adaptive immune in gut meanwhile more innate in liver. Interestingly reduced differential immune transcripts between gut and liver upon inflammation were also revealed. In addition, more immune proteins in bile than intestinal mucus were identified. And bile probably providing immune effectors to intestinal mucus upon inflammation was deduced. Specifically, many key immune transcripts in gut or liver as well as key immune proteins in mucus or bile were demonstrated. Accordingly, we proposed a hypothesized profile of fish gut-liver immunity, during either homeostasis or inflammation. Current data suggested that fish gut and liver may collaborate immunologically while keep homeostasis using own strategies, including potential unique mechanisms.

Many coronaviruses are capable of interspecies transmission. Some of them have caused worldwide panic as emerging human pathogens in recent years, e.g., severe acute respiratory syndrome coronavirus (SARS-CoV) and Middle East respiratory syndrome coronavirus (MERS-CoV). In order to assess their threat to humans, we explored to infer the potential hosts of coronaviruses using a dual-model approach based on nineteen parameters computed from spike genes of coronaviruses. Both the support vector machine (SVM) model and the Mahalanobis distance (MD) discriminant model achieved high accuracies in leave-one-out cross-validation of training data consisting of 730 representative coronaviruses (99.86% and 98.08% respectively). Predictions on 47 additional coronaviruses precisely conformed to conclusions or speculations by other researchers.
