Objective of this study was to evaluate the efficacy and adverse drug reactions (ADR) of palipeddone sustained release tablet in the treatment of schizophrenia in outpatients. Outpatients with schizophrenia treated with palipeddone sustained release tablet (observation group)in our hospital from Apr. to Sep. in 2009 were followed up and compared with those treated with risperidone (control group) during the same period. This study examined the formulation variables and optimization of a sustained-release metformin hydrochloride 750 mg tablet using a combination of hydroxypropyl methylcellulose (HPMC), polyvinylpyrrolidone (PVP K30), and glyceryl behenate with a computer optimization technique based on a response surface methodology (RSM) utilizing a polynomial equation. A three-factor, three-level Box–Behnken design was used for the optimization procedure with the contents of HPMC (X 1), PVP K30 (X 2), and glyceryl behenate (X 3) as independent variables. The drug release percentages at 1, 3, and 10 h were the target responses and were restricted to 25–35 % (Y 1), 55–65 % (Y 2), and 85–100 % (Y 3), respectively. The quadratic model fit the data well and the resulting equation was used to predict the responses in the optimal region. The optimized formulation was achieved with 165 mg HPMC, 40 mg PVP K30, and 22.5 mg glyceryl behenate, and the observed responses of the optimized formulation were very close to the values predicted using the RSM optimization technique. Furthermore, the drug release rate and dissolution curve shape of the optimized formulation were similar to those of the commercial product.

【The objectives of this study were to evaluate the effect of formulation ingredients on the drug release and to optimize the novel sustained release matrix tablet formulations of bupropion hydrochloride. A three factor, three-level Box-Behnken design was used for the optimization procedure, with the amounts of PEO ( $X_1$ ), citric acid ( $X_2$ ) and Compritol 888 ATO ( $X_3$ ) as the independent variables. The selected dependent variables were the cumulative percentage values of bupropion hydrochloride that had dissolved after 1, 4 and 8 hr. Various dissolution profiles of the drug from sustained release matrix tablets were obtained. Optimization was performed for $X_1$ , $X_2$ and $X_3$ using the following target ranges; $30%{\leq}Y_1{\leq}45%$ ; $70{\leq}Y_2{\leq}85%$ ; $85%{\leq}Y_3{\leq}100%$ . The optimized formulation for bupropion hydrochloride was achieved with 12.5% PEO ( $X_1$ ), 2.5% citric acid ( $X_2$ ) and 10% Compritol 888 ATO ( $X_3$ ). The sustained release matrix tablets with the optimized formulation provided a release profile that was close to predicted values. In addition, the dissolution profiles of the sustained release matrix tablet with the optimized formulation were similar to those of the commercial product Wellbutrin $^{(R)}$ SR tablets ( $f_2$ =79.8 79.83).】

The objective of this study was to compare a novel sustained release tablet formulation that has the potential to be used for drugs of different physicochemical properties using a binary mixture of polymethacrylate polymers in their salt forms with the polymethacrylate interpolyelectrolyte complex (IPEC) tablets in terms of drug release and compactness. Also, we aimed to compare this formulation with an IPEC tablet in terms of drug release. Tablets prepared using Eudragit E-Citrate and Eudragit L-Sodium were more convenient, easier to prepare, and showed better sustained release and compactness characteristics compared to IPEC tablets of similar concentrations and preparation methods.

To develop a sustained-release tablet which had the similar dissolution to commercial ibudilast-loaded sustained-release capsule, the tablets were prepared using hydroxypropylmethylcellulose phthalate (HPMCP), ethylcellulose (EC) and hydroxypropylcellulose (HPC), and dissolution test were carried out with paddle method in KP. The tablet prepared only with HPMCP and EC showed sno similar dissolution pattern to the commercial product. As the ratio of HPMCP/HPC in tablet decreased, the dissolution rate of drug decreased in pH 1.2 but increased in pH 6.8. Furthermore, an ibudilast-loaded sustained-release tablet composed of [ibudilast/EC/HPMCP/HPC (10/10/170/10 mg/tab)] gave similar dissolution to commercial product in pH 1.2 for 3 h and in pH 6.8 for 10 h. Thus, it could be a potential candidate for the substitute of commercial capsule.

Adopting prospective, multi-center, random, single-blind and equal rank-control methods, 226 patients with rheumatoid arthritis (RA) were divided into two groups. The 114 patients of the test group were treated with oral sustained release tablets of Tripterygium Wilfordii (TW-SR), 2 tablets twice a day for 4 weeks, 112 patients of the control group received tablets of Tripterygium Wilfordii (TW) orally 2 tablets 3 times per day for 4 weeks. Testing results showed the total effective rates of the two groups were 92.11 % and 90.65 %, respectively (P>0.05).
