Resident tissue macrophages (RTMs) are differentiated immune cells that populate distinct niches and exert important tissue-supportive functions. RTM maintenance is thought to rely either on differentiation from monocytes or on RTM self-renewal. Here, we used a mouse model of inducible lung interstitial macrophage (IM) niche depletion and refilling to investigate the development of IMs in vivo. Using time-course single-cell RNA-sequencing analyses, bone marrow chimeras and gene targeting, we found that engrafted Ly6C+ classical monocytes proliferated locally in a Csf1 receptor-dependent manner before differentiating into IMs. The transition from monocyte proliferation toward IM subset specification was controlled by the transcription factor MafB, while c-Maf specifically regulated the identity of the CD206+ IM subset. Our data provide evidence that, in the mononuclear phagocyte system, the ability to proliferate is not merely restricted to myeloid progenitor cells and mature RTMs but is also a tightly regulated capability of monocytes developing into RTMs in vivo.

The strips with good flatness after finish rolling will have flatness defects such as edge wave after run-out table cooling. These defects are caused by the residual stress due to inhomogeneous thermal and microstructural behaviors during the cooling stage. The traditional residual stress control methods rely too much on experience, resulting in poor control effect and poor universality. Therefore, this study proposed two residual stress control methods based on model calculation: water flow central crown, and center wave compensation control. The former is mainly based on controlling the uniformity of strip transverse temperature and microstructure. The latter is mainly controlled by mutual compensation of internal stresses. In this paper, the mathematical models of water flow central crown cooling and critical center wave compensation stress were established respectively. Meanwhile, a post-buckling model of hot-rolled strip was established to calculate the aim flatness value at exit of finishing mill under critical compensation stress. Furthermore, a fully coupled finite element model based on temperature, phase transitions, and stress was developed and it was verified using experimental data. Finally, the above two residual stress control methods were realized in the finite element model. The results showed that the two methods can reduce the residual stress and improve the flatness, and the application of center wave compensation control is more effective.

SMARCE1 gene, encoding a core subunit of SWI/SNF chromatin remodeling complex, is situated on chromosome 17q21-ter region that is frequently gained in neuroblastoma. However, its role in the tumorigenesis remains unknown. Here, we showed that high expression of SMARCE1 was associated with poor prognosis of patients with neuroblastoma, especially those with MYCN amplification. Knockdown of SMARCE1 reduced proliferation, colony formation, and tumorigenicity of neuroblastoma cells. Mechanistically, SMARCE1 directly interacted with MYCN, which was necessary for MYCN-mediated transcriptional activation of downstream target genes including PLK1, ODC1, and E2F2. Overexpression of PLK1, ODC1 or E2F2 significantly reversed the inhibiting effect of SMARCE1 knockdown on the proliferation, colony formation, and tumorigenicity of MYCN-amplified neuroblastoma cells. Moreover, we revealed that MYCN directly regulated SMARCE1 transcription through binding to a non-canonical E-box of SMARCE1 promoter, thus enhancing SMARCE1-MYCN cooperativity. These findings establish SMARCE1 is a critical oncogenic factor in neuroblastoma and provide a new potential target for treatment of neuroblastoma with 17q21-ter gain and MYCN amplification.

The study aimed to investigate the cognitive processing of prospective memory (PM) in patients with schizophrenia spectrum disorders (SSDs) by using an eye-tracking paradigm. In addition, the facilitating effects of prosocial intention (the desire to help others) on PM in SSDs were also examined. In phase 1, 26 patients (group1) and 25 healthy controls (HCs) were compared in an eye-tracking PM paradigm in terms of the PM accuracy and eye-tracking indices. In phase 2, 21 more patients (group2) were recruited, and a prosocial intention was introduced in the eye-tracking PM paradigm. Their PM accuracy and eye-tracking indices were compared with those in group1. The PM cue monitoring was indicated by the total fixation counts and fixation time on distractor words. In phase 1, group1 showed lower PM accuracy, fewer fixation counts and less fixation time on distractor words than HCs. In phase 2, group2 (with prosocial intention) performed significantly better than group1 (with typical instruction) on both PM accuracy and fixation time on distractor words. In both groups of SSDs, the PM accuracy was significantly correlated with both the fixation counts and the fixation time of distractor words. After controlling for the cue monitoring indices, the difference in PM accuracy remained significant between group1 and HCs but disappeared between group1 and group2. The cue monitoring deficit contributes to the PM impairment in SSDs. The facilitating effect of prosocial intention disappears after the control of cue monitoring, also indicating its critical role in PM.

Purpose This study aimed to investigate the biomechanical characteristics of the external spinal fixation for treating lumbar fracture through finite element analysis (FEA) and provide a theoretical basis for its further application. Methods Two different models of L3 fracture fixed with the external spinal fixation and the internal fixation system respectively were constructed. The ROM, maximum stresses at L3, and the screws of the two models were measured under load control. Subsequently, the applied torque, the maximum stressed at L3, L1/2, L2/3, L3/4, L4/5 discs and the screws were analyzed under displacement control. Results Under load control, the external fixation model reserved more ROM than the internal fixation model (40.4–48.0% vs 30.5–41.0%). Compared to the internal fixation model, the maximum stresses at L3 and screws in the external fixation model were increased. Under displacement control, the external fixation model required fewer moments (N·mm) than the internal fixation model (flexion: 7500 vs 12,294; extension: 7500 vs 9027). Further, the maximum stresses at L3 and the screws in the external fixation model were greater than those of the internal fixation model, while the maximum stresses at the upper and lower adjacent discs of fixed segments were less than the internal fixation model. Conclusion Compared to the internal fixation system, the external fixation has a better stress distribution with the greater overall mobility. It theoretically reduces the stress concentration of the adjacent discs and the stress shielding of the fractured vertebral body.

Extracellular vesicles (EVs) contain thousands of proteins and nucleic acids, playing an important role in cell-cell communications. Sertoli cells have been essential in the testis as a "nurse cell". However, EVs derived from human Sertoli cells (HSerCs) have not been well investigated.EVs were isolated from HSerCs via ultracentrifugation and characterized by transmission electron microscopy, tunable resistive pulse sensing, and Western blotting. The cargo carried by HSerCs-EVs was measured via liquid chromatography-mass spectrometry and GeneChip miRNA Arrays. Bioinformatic analysis was performed to reveal potential functions of HSerCs-EVs.A total of 860 proteins with no less than 2 unique peptides and 88 microRNAs with high signal values were identified in HSerCs-EVs. Biological processes related to molecular binding, enzyme activity, and regulation of cell cycle were significantly enriched. Specifically, many proteins in HSerCs-EVs were associated with spermatogenesis and regulation of immune system, including Septins, Large proline-rich protein BAG6, Clusterin, and Galectin-1. Moreover, abundant microRNAs within HSerCs-EVs (miR-638, miR-149-3p, miR-1246, etc.) had a possible impact on male reproductive disorders such as asthenozoospermia and oligozoospermia.Our study has shown that HSerCs-EVs contain diverse components such as proteins and microRNAs. Further research is required to evaluate HSerCs-EVs in spermatogenesis, which are underutilized but highly potent resources with particular promise for male infertility.

Astrocytes are critical regulators of the immune/inflammatory response in several human central nervous system (CNS) diseases. Emerging evidence suggests that dysfunctional astrocytes are crucial players in seizures. The objective of this study was to investigate the role of transient receptor potential vanilloid 4 (TRPV4) in 4-aminopyridine (4-AP)-induced seizures and the underlying mechanism. We also provide evidence for the role of Yes-associated protein (YAP) in seizures. 4-AP was administered to mice or primary cultured astrocytes. YAP-specific small interfering RNA (siRNA) was administered to primary cultured astrocytes. Mouse brain tissue and surgical specimens from epileptic patient brains were examined, and the results showed that TRPV4 was upregulated, while astrocytes were activated and polarized to the A1 phenotype. The levels of glial fibrillary acidic protein (GFAP), cytokine production, YAP, signal transducer activator of transcription 3 (STAT3), intracellular Ca(2+)([Ca(2+)](i)) and the third component of complement (C3) were increased in 4-AP-induced mice and astrocytes. Perturbations in the immune microenvironment in the brain were balanced by TRPV4 inhibition or the manipulation of [Ca(2+)](i) in astrocytes. Knocking down YAP with siRNA significantly inhibited 4-AP-induced pathological changes in astrocytes. Our study demonstrated that astrocytic TRPV4 activation promoted neuroinflammation through the TRPV4/Ca(2+)/YAP/STAT3 signaling pathway in mice with seizures. Astrocyte TRPV4 inhibition attenuated neuroinflammation, reduced neuronal injury, and improved neurobehavioral function. Targeting astrocytic TRPV4 activation may provide a promising therapeutic approach for managing epilepsy. Background Intravascular papillary endothelial hyperplasia (IPEH) is a vascular tumor characterized by the proliferation of endothelial cells with papillary formation. It is a rare benign condition affecting the head and neck. Currently, no cases of IPEH of the spleen have been reported. Here, we report a case of IPEH of the spleen in a child and discuss its clinical manifestations, imaging features, and surgical treatment. Case presentation A 5-year-old female presented with a 4-month-old tumor in the left upper abdomen, abdominal pain, and constipation. She underwent radiography, barium enema, US, and MRI. A solid space-occupying mass was found in the left abdominal cavity on preoperative imaging, and it was diagnosed as angiosarcoma. The lesion was surgically resected. Histopathological analysis was consistent with IPEH. Conclusion Clinicians should consider the possibility of IPEH in patients presenting with tumors in the spleen, which is curable by surgical resection. Malignant vascular tumors must be excluded in the differential diagnosis of IPEH to prevent misdiagnosis and inappropriate overtreatment.

In order to further improve the control accuracy of thickness, crown and tension in hot strip finishing mill, a multivariable optimizing strategy based on inverse linear quadratic (ILQ) control theory is proposed in this paper. First, the state space model of hot rolling process is established based on the basic dynamic mathematical model of rolling process and measured data collected from a certain plant. Then, the new ILQ control strategy is designed for the thickness, crown and tension control system in the complex rolling process, and the response performance and anti-interference performance of ILQ controller and PI controller were tested. The results show that the proposed ILQ control strategy has excellent control performance with the maximal overshoot 24.13% smaller than that of PI controller when strip crown, thickness and tension step signals are added simultaneously, and the ILQ control strategy has certain guiding significance for the actual production of hot rolling.

High sensitivity of sensors is extremely significant for precisely monitoring imperceptible changes of motion in real-time, which cannot be achieved by traditional piezoelectric devices. Herein, a hierarchical polyvinylidene fluoride hexafluoropropylene (PVDF-HFP)/ZnO composite nanaofiber piezoelectric sensor with highly sensitivity has been prepared through epitaxial growing ZnO nanosheets on the surface of electrospun PVDF-HFP nanofibers. Systematic investigations have shown that their optimum pressure sensing performance with a sensitivity of 1.9 V kPa−1 and a short response time of 20 ms are achieved for forces from 0.02 to 0.5 N with excellent durability and stability up to 5000 cycles. Moreover, this sensor can precisely detect the imperceptible changes in player’s motions to avoid injury from overtraining. Additionally, a Bluetooth-low-energy that tracks player’s workout and transmits the output signals wirelessly to a smartphone app is utilized. The study provides a feasible approach for high-precision detecting and safety monitoring in the fields of medical, rehabilitation medicine, and workout security.

An experimental study on the traveling crossflow instability over a 60 $$^\circ $$ swept flat plate was conducted. The Mach number is 6, the angle of attack of the model is 5 $$^\circ $$ . The traveling crossflow waves and the secondary instability of the traveling crossflow waves were visualized by nano-tracer-based planar laser scattering (NPLS) technique. In the spanwise NPLS images, the traveling crossflow waves appeared as regular strikes, and the secondary instability appeared as small eddies attached to strikes. The wavelet transform was used to study the wavelength contents of the traveling crossflow waves. The most amplified wavelength is stable before the secondary instability happening, which is around 12 mm at $$Re_\infty $$ = 3.45 $$\times $$ 10 $$^6$$ m $$^{-1}$$ . Besides, the Reynolds number effects on the boundary layer transition and traveling crossflow instability were discussed.

To make up the poor quality defects of traditional control methods and meet the growing requirements of accuracy for strip crown, an optimized model based on support vector machine (SVM) is put forward firstly to enhance the quality of product in hot strip rolling. Meanwhile, for enriching data information and ensuring data quality, experimental data were collected from a hot-rolled plant to set up prediction models, as well as the prediction performance of models was evaluated by calculating multiple indicators. Furthermore, the traditional SVM model and the combined prediction models with particle swarm optimization (PSO) algorithm and the principal component analysis combined with cuckoo search (PCA-CS) optimization strategies are presented to make a comparison. Besides, the prediction performance comparisons of the three models are discussed. Finally, the experimental results revealed that the PCA-CS-SVM model has the highest prediction accuracy and the fastest convergence speed. Furthermore, the root mean squared error (RMSE) of PCA-CS-SVM model is 2.04 µm, and 98.15% of prediction data have an absolute error of less than 4.5 µm. Especially, the results also proved that PCA-CS-SVM model not only satisfies precision requirement but also has certain guiding significance for the actual production of hot strip rolling.

To study the transverse temperature uniformity with transverse flux induction heating (TFIH) during endless strip production (ESP) rolling, a coupled electromagnetic-thermal finite element model of transverse flux induction heating is established. The initial temperature distribution at the TFIH inlet is taken into consideration, which is the basis of subsequent numerical studies. Magnetic screens are designed to weaken the influence of edge overheating, and the influence of magnetic shielding amounts on temperature uniformity is discussed. Moreover, an evaluation criterion is provided to determine the temperature uniformity. Furthermore, the effect of different shielding amounts on the edge temperature is studied. An ideal temperature distribution at the TFIH outlet is obtained based on a model combining various shielding amounts, and the temperature standard deviation of this method is 2.01% lower than the traditional arrangement of magnetic shielding amounts. Actual data are collected to verify the results of the proposed model. The temperature deviation between the middle and edge is approximately 12°C, and the temperature uniformity is enhanced with the optimized combination of magnetic shielding amounts.
