The implementation of green credits has become an important engine for China's high-quality energy development (HQED). On the basis of constructing an index of HQED and the panel data of thirty provinces in China from 2008 to 2019, this study empirically investigated the effects of green credits on HQED and the action mechanisms behind it in a multi-dimensional manner using a panel fixed-effects model, mediating-effects model, and spatial Durbin model. The results indicated that green credits had significantly contributed to China's HQED, and that conclusion still held true after a series of robustness tests were conducted. It was found that industrial structures and human capital were important channels through which green credits influenced China’s HQED. Moreover, the spatial spillover effects of green credits on HQED were also confirmed. Finally, in terms of temporal heterogeneity, the positive effects of green credits on HQED were found to have increased significantly after 2012. Also, in terms of regional heterogeneity, this study observed that the positive influence of green credits on HQED was more significantly in central and western China than in eastern China, and in southern China than in northern China. The results obtained in this research investigation will potentially provide some important insights for energy planners and policymakers to further the understanding of the drivers of HQED, and the corresponding transmission mechanisms and regional differences.

The anticoagulation and hemostatic properties of blood-contacting materials are opposite lines of research, but their realization mechanisms are inspired by each other. Contact between blood and implantable biomaterials is a classic problem in tribological research, as both antithrombotic and hemostatic materials are closely associated with this problem. Thrombus formation on the surfaces of blood-contacting biomedical devices can detrimentally affect their performance and patient life, so specific surface functionalization is required. Currently, intensive research has focused on the development of super-lubricated or super-hydrophobic coatings, as well as coatings that deliver antithrombotic drugs. In addition, hemostatic biomaterials with porous structures, biochemical substances, and strongly adhesive hydrogels can be used to achieve rapid and effective hemostasis via physical or biochemical mechanisms. This article reviews methods of preparing anticoagulant coatings on material surfaces and the current status of rapid hemostatic materials. It also summarizes fundamental concepts for the design and synthesis of anticoagulant and hemostatic materials by discussing thrombosis and hemostasis mechanisms in biomedical devices and normal organisms. Because there are relatively few reports reviewing the progress in surface-functionalized design for anticoagulation and hemostasis, it is anticipated that this review can provide a useful summary of the applications of both bio-adhesion and bio-lubrication techniques in the field of biomedical engineering.

Cultural relic conservation capability is an important issue in cultural relic conservation research, and it is critical to decrease the vulnerability of immovable cultural relics to rainfall hazards. Commonly used vulnerability assessment methods are subjective, are mostly applied to regional conditions, and cannot accurately assess the vulnerability of cultural relics. In addition, it is impossible to predict the future vulnerability of cultural relics. Therefore, this study proposed a machine learning-based vulnerability assessment method that not only can assess cultural relics individually but also predict the vulnerability of cultural relics under different rainfall hazard intensities. An extreme rainfall event in Henan Province in 2021 was selected as an example, with a survey report on the damage to cultural relics as a database. The results imply that the back propagation (BP) neural network-based method of assessing the vulnerability of immovable cultural relics is reliable, with an accuracy rate higher than 92%. Based on this model to predict the vulnerability of Zhengzhou City’s cultural relics, the vulnerability levels of cultural relics under different recurrence periods of heavy rainfall were obtained. Among them, the vulnerability of ancient sites is higher than those of other cultural relic types. The assessment model used in this study is suitable for predicting the vulnerability of immovable cultural relics to heavy rainfall hazards and can provide a technical means for cultural relic conservation studies.

Recombinant protein drugs, which are typically produced by mammalian host cells, have been approved for the treatment of a range of diseases. Accordingly, systems for selecting recombinant cell lines with efficient protein expression and for testing the content of recombinant proteins in vivo are crucial to the large-scale production and application of protein-based therapeutic drugs. In this study, we designed three aptamer beacons to detect His-tag, a common label of recombinant proteins. We found that all three beacons could specifically and quantitatively measure the His-tagged recombinant proteins with a short reaction time. Among these three beacons, the 6H5-MU beacon had the highest sensitivity for His polypeptides with a detection limit of 250 ng/mL and the shortest detection time within 1 min. Furthermore, we established a rapid and highly effective recombinant cell line construction system, which could obtain monoclonal cell lines with high yields of target proteins within 21 days, by combining 6H5-MU with pSB, a novel plasmid composed of a Sleeping Beauty transposase and a transposon. Finally, 6H5-MU also discriminately tested the serum concentration of His-tagged recombinant proteins in vivo, with consistent results compared to enzyme-linked immunosorbent assay (ELISA). We thus established a rapid and high-throughput method for generating recombinant cell lines and in vivo monitoring of recombinant protein levels, thereby providing a new platform for the development and preparation of recombinant protein drugs. KEY POINTS: • The 6H5-MU aptamer beacon rapidly and accurately binds to His-tagged recombinant proteins. • A system for rapid and high-throughput generation of recombinant cell lines is established using 6H5-MU and pSB. • 6H5-MU allows in vivo monitoring of recombinant protein levels.

To build and validate a multi-parametric MRI (mpMRI)-based radiomics nomogram for early prediction of treatment response to neoadjuvant chemotherapy (NAC) in locally advanced gastric cancer.Baseline MRI were retrospectively enrolled from 141 patients with gastric adenocarcinoma who received NAC followed by radical gastrectomy. According to pathologic response of tumor regression grading (TRG), patients were labeled as responders (TRG = 0 + 1) and non-responders (TRG = 2 + 3) and further divided into a training (n = 85) and validation dataset (n = 56). Radiomics score (Radscore) were built from T2WI, ADC, and venous phase of dynamic-contrasted-enhanced MR imaging. Clinical information, laboratory indicators, MRI parameters, and follow-up data were also recorded. According to multivariable regression analysis, an mpMRI radiomics nomogram was built and its predictive ability was evaluated by ROC analysis. Decision curve analysis was applied to evaluate the clinical usefulness. Kaplan-Meier survival curves based on the nomogram were used to estimate the progression-free survival (PFS) and overall survival (OS) in the validation dataset.Both single sequence-based Radscores and mpMRI radiomics nomogram were associated with pathologic response (p0.001). The nomogram achieved the highest diagnostic ability with area under ROC curve of 0.844 (95% CI, 0.749-0.914) and 0.820 (95% CI, 0.695-0.910) in the training and validation datasets. The hazard ratio of the nomogram for PFS and OS prediction was 2.597 (95% CI: 1.046-6.451, log-rank p = 0.023) and 2.570 (95% CI: 1.166-5.666, log-rank p = 0.011).The mpMRI-based radiomics nomogram showed preferable performance in predicting pathologic response to NAC in LAGC.• This study investigated the value of multi-parametric MRI-based radiomics in predicting pathologic response to neoadjuvant chemotherapy in locally advanced gastric cancer. • The nomogram incorporating T2WI Radscore, ADC Radscore, and DCE Radscore as well as Borrmann classification outperformed the single sequence-based Radscore. • The nomogram also exhibited a promising prognostic ability for patient survival and enriched radiomics studies in gastric cancer.

Gastric cancer is heterogeneous cancer and the causes of this disease are complex. New diagnostic and therapeutic targets are urgently needed to explore. Huntingtin-associated protein 1 (HAP1) is directly related to Huntington's disease (HD). However, patients with Huntington's disease have a lower incidence of cancer. Therefore, we are committed to studying the correlation between HAP1 and gastric carcinogenesis and development.Immunohistochemical staining, western blot analysis, and RT-qPCR were conducted to explore the localization and expression of HAP1 in gastric cancer. To study the biological significance of HAP1, we overexpressed HAP1 in both MKN28 and AGS cell lines by lentivirus infection. To explore the role of HAP1 in cell proliferation, the cells counting assay, EdU incorporation assay, and colony formation assay were carried out. We performed the wound healing assay and transwell assay to study the cell migration and invasion. To further investigate whether HAP1 could regulate gastric cancer cell death during glucose deprivation, Annexin V-FITC/PI staining was performed. In our study, we elucidated that HAP1 was downregulated in gastric cancer. What's more, overexpressing HAP1 inhibited cell proliferation, cell migration and invasion, and triggered apoptosis during glucose deprivation. More importantly, the antitumor properties and mechanisms of HAP1 have been elucidated further in gastric cancer.Taken together, the available evidence implies that HAP1 may serve as a potential tumor suppressor, making it a significant target in preventing and treating gastric cancer. This research provides a theoretical basis for the early diagnosis, clinical targeted therapy, and prognosis evaluation of gastric cancer.

To investigate the occurrence of mosaicism in epilepsy probands and their parents using amplicon-based deep sequencing (ADS).Patients were recruited from the outpatient of Peking University First Hospital. Two hundred and sixty-four probands with pathogenic variants tested by next-generation sequencing (NGS) were enrolled.Mosaic variants were detected in seventeen disease-associated genes from 20 probands, 5 paternal, and 6 maternal parents. The frequency of mosaicism was 11.74% (31/264). Mosaicism in 11 genes was identified from 20 probands with the mutant allelic fractions (MAFs) of 12.95-38.00% in autosomal dominant genes. Five paternal mosaicisms were identified in genes with a MAF of 6.30-20.99%, and six maternal mosaic individuals with a MAF of 2.07-21.90%. Only four mosaic parents had milder seizure history. The affected sibling had the same phenotype consistent with that of the proband, who inherited the variant of SLC1A2 or STXBP1 from their unaffected mosaic mothers, respectively.Mosaic phenomenon is not rare in families with epilepsy. Phenotypes of mosaic parents were milder or normal. Mosaicism detection is helpful to identify the mutation origin and it provides a theoretical basis for prenatal diagnosis of family reproduction. ADS is a reliable way of mosaicism detection for clinical application.

We investigated whether glycolytic heterogeneity correlated with histopathology, and further stratified the survival outcomes pertaining to resectable lung adenocarcinoma.We retrospectively analyzed theEntropy significantly correlated with histopathology, including tumor grades, lympho-vascular invasion, and visceral pleural invasion. Furthermore, entropy was an independent predictor of unfavorable DFS (p = 0.031) and OS (p = 0.004), while pathological nodal metastasis independently predicted DFS (p = 0.009). Our entropy-based models outperformed the traditional staging system (c-index = 0.694 versus 0.636, p = 0.010 for DFS; c-index = 0.704 versus 0.630, p = 0.233 for OS). The models provided further survival stratification in subgroups comprising different tumor grades (DFS: HR = 2.065, 1.315, and 1.408 for grade 1-3, p = 0.004, 0.001, and 0.039, respectively; OS: HR = 25.557, 6.484, and 2.570, for grade 1-3, p = 0.006, 0.001, and = 0.224, respectively).The glycolytic heterogeneity portrayed by entropy is associated with aggressive histopathological characteristics. The proposed entropy-based models may provide more sophisticated survival stratification in addition to histopathology and may enable personalized treatment strategies for resectable lung cancer.

A macro continuum model of the traffic flow is derived from a micro car-following model that considers both the upslope and downslope by using the transformation relationship between macro- and micro variables. The perturbation propagation characteristics and stability conditions of the macroscopic continuum equation are discussed. For uniform flow in the initial equilibrium state, the stability conditions reveal that as the slope angle increased under the action of a small disturbance, the upslope stability increases and downslope stability decreases. Moreover, under a large disturbance, the global stability analysis is carried out by using the wavefront expansion technique for uniform flow in the initial equilibrium state. For the initial nonuniform flow, nonlinear bifurcation analysis is conducted at the equilibrium point. Subcritical Hopf bifurcation exists when the traffic flow state changes,thus the limit cycle formed by the Hopf bifurcation is unstable. Simulation results verify the stability conditions of the model and determine the critical density range. The numerical simulation results show the existence of critical Hopf bifurcation in phase space, and the spiral saddle point varies with the slope angle. Furthermore, the impact of the angle of both the upslope and downslope on the evolution of density waves is investigated.

Membrane bioreactor (MBR) is an attractive option method for treating azo dye wastewater under extreme conditions. The present study assessed the effect of salinity on the performance of anaerobic MBR in treating azo dye wastewater. Increased salinity showed adverse effects on the decolorization efficiency and chemical oxygen demand (COD) removal efficiency. The decolorization efficiency decreased from 95.8% to 82.3% and 73.1% with a stepwise increasing of salinity from 0 to 3% and 5%, respectively. The COD removal efficiency decreased from 80.7% to 71.3% when the salinity increased from 0 to 3% and then decreased to 58.6% at 5% salinity. The volatile fatty acids (VFAs) concentration also increased as the salinity increased. Furthermore, increased salinity led to the elevated production of soluble microbial products (SMP) and extracellular polymeric substances (EPS), which can provide a protective barrier against harsh environments. More serious membrane fouling was observed as the SMP and EPS concentrations increased. The concentration of loosely bound EPS (LB-EPS), tightly bound EPS (TB-EPS), and the polysaccharide/protein (PS/PN) ratios in LB-EPS and TB-EPS all increased when the salinity was elevated. The production of SMP and EPS was caused by the generation of PS in response to the saline environment. Lactobacillus, Lactococcus, Anaerosporobacter, and Pectinatus were the dominant bacteria, and Lactobacillus and Lactococcus were the decolorization bacteria in the MBR. The lack of halophilic bacteria was the main reason for the decreased decolorization efficiency in the salinity environment.
