Abstract

Ocean acidification (OA) is beginning to have noticeable negative impact on calcification rate, shell structure and physiological energy budgeting of several marine organisms; these alter the growth of many economically important shellfish including oysters. Early life stages of oysters may be particularly vulnerable to OA-driven low pH conditions because their shell is made up of the highly soluble form of calcium carbonate (CaCO3) mineral, aragonite. Our long-term CO2 perturbation experiment showed that larval shell growth rate of the oyster species Crassostrea hongkongensis was significantly reduced at pH < 7.9 compared to the control (8.2). To gain new insights into the underlying mechanisms of low-pH-induced delays in larval growth, we have examined the effect of pH on the protein expression pattern, including protein phosphorylation status at the pediveliger larval stage. Using two-dimensional electrophoresis and mass spectrometry, we demonstrated that the larval proteome was significantly altered by the two low pH treatments (7.9 and 7.6) compared to the control pH (8.2). Generally, the number of expressed proteins and their phosphorylation level decreased with low pH. Proteins involved in larval energy metabolism and calcification appeared to be down-regulated in response to low pH, whereas cell motility and production of cytoskeletal proteins were increased. This study on larval growth coupled with proteome change is the first step toward the search for novel Protein Expression Signatures indicative of low pH, which may help in understanding the mechanisms involved in low pH tolerance.

Abstract

The Vitreoscilla hemoglobin (VHb) gene (vgb) was integrated into the chromosome of Bacillus thuringiensis BMB171 using integrative vector pEG491. The production of VHb was confirmed by CO-difference spectra analysis. Fermentation experiments results showed that with the production of VHb, the critical oxygen concentration (COC) of the host strain was reduced from 18 to 12%. The maximum viable cell counts of the VHb+ strain in high, middle, and low aeration/agitation fermentations were 0.94-, 1.23-, and 1.59-fold of those of the VHb- strain, respectively. Under the same conditions, the yields of insecticidal crystal proteins (ICP) by VHb+ strain were 1.22-, 1.63-, and 3.13-fold of those of the VHb- strain. The production of VHb also accelerated the formation of ICP and spores. These results indicated that the production of VHb could improve the cell density and ICP yield of B. thuringiensis, especially under low aeration/agitation condition.

Abstract

A microcalorimetric technique was used to investigate the influence of plasmid copy number and various promoters on the green fluorescent protein expression in Bacillus thuringiensis. The power-time curves of the growth metabolism of B. thuringiensis have been determined by using an LKB-2277 bioactivity monitor at 28°C. For evaluation of the results, the maximum peak-heat output power ( P max ), the generation time ( t G ), the growth rate constants ( k ) in the log phase, the total heat effect ( Q total ) for the growth metabolism of Bacillus thuringiensis were determined. The results firstly indicated that the more plasmid copy number per cell the more protein synthesization, and promoter BtI-BtII had a stronger impact on the gene expression than promoter 3A investigated by the method of microcalorimetry.

The analysis of the thermogenic curves indicated for the first time that the more plasmid copy number per cell the more protein synthetization. Promoter BtI-BtII had a stronger impact on the gene expression than promoter 3A investigated by the method of microcalorimetry.

Abstract

By using an LKB-2277 Bioactivity Monitor, cycle-flow method, the thermogenic curves of aerobic growth for Bacillus thuringiensis cry II strain at 28°C have been obtained. The metabolic thermogenic curves of B. thuringiensis cry II contained two distinct patterns: the first reflects the changes during the bacterial growth phase and the second corresponds to the sporulation phase. From these thermogenic curves in the absence and presence of Sm3+ ions, the thermokinetic parameters such as the growth rate constants k, the interval time τI, the maximum power Pmax 1 and heat-output Qlog for log phase, the maximum power Pmax 2 and heat-output Qstat for stationary phase, the heat-output Qspor for sporulation phase and total heat effects QT are calculated. Sm3+ ion has promoting action on the growth of B. thuringiensis cry II in its lower concentration range; on the other hand, this ion has inhibitory action on the sporulation of B. thuringiensis in its higher concentration range. We also found that the effects of Sm3+ ion on B. thuringiensis during the sporulation phase were far greater than that during the bacterial phase. It is concluded that the application of B. thruringiensis of controlling insecticides is not affected by the presence of the rare-earth elements in the environmental ecosystem.

Abstract

By using an LKB-2277 Bioactivity Monitor, ampoule mode, the heat output of Bacillus thuringiensis growth metabolism is determined at 28°C and the effect of Cu2+ on B. thuringiensis growth is studied. Copper is regarded as an essential trace element for life. Its deficiency may be the cause of diseases. Cu2+ at different concentrations has different effects on B. thuringiensis growth metabolism: a low concentration (0–30 μg/ml) of Cu2+ can promote the growth of B. thuringiensis, a high concentration (40–120 μg/ml) can inhibit growth of the bacteria, and a concentration of Cu2+ of up to 130 μg/ml completely inhibits B. thuringiensis growth.
