The development of nervous system (NS) in the non-feeding vestibula larva of the sea urchin, Holopneustes purpurescens, and the feeding echinopluteus larva of Hemicentrotus pulcherrimus was examined by focusing on fate during metamorphosis. In H. purpurescens, the serotonergic NS (SerNS) appeared simultaneously and independently in larval tissue and adult rudiment, respectively, from 3-day post-fertilization. In 4-day vestibulae, an expansive aboral ganglion (450 x 100 mum) was present in the larval mid region that extended axons toward the oral ectoderm. These axons diverged near the base of the primary podia. An axonal bundle connected with the primary podia and the rim of vestopore on the oral side. Thus, the SerNS of the larva innervated the rudiment at early stage of development of the primary podia. This innervation was short-lived, and immediately before metamorphosis, it disappeared from the larval and adult tissue domains, whereas non-SerNS marked by synaptotagmin remained. The NS of 1-month post-fertilization plutei of H. pulcherrimus comprised an apical ganglion (50 x 17 mum) and axons that extended to the ciliary bands and the adult rudiment (AR). A major basal nerve of serotonergic and non-serotonergic axons and a minor non-serotonergic nerve comprised the ciliary band nerve. In 3-month plutei, axonal connection among the primary podia in the neural folds completed. The SerNS never developed in the AR. Thus, there was distinctive difference between feeding- and non-feeding larvae of the above sea urchins with respect to SerNS and the AR.

Recently, a novel type of neurogenic placode was described in anurans. These hypobranchial placodes were recognized as ectodermal thickenings situated ventral to the second and third pharyngeal pouch that give rise to neurons of unknown fate. Here, the development of hypobranchial placodes in Xenopus laevis is described in more detail using in situ hybridization and immunohistochemistry for various placodal ( Six1, Eya1) and neurogenic ( NGNR-1, NeuroD, Delta-1, Hu, acetylated tubulin) markers. Moreover, the fate of hypobranchial placodes was determined by analyzing tadpoles that had received orthotopic grafts of ventral branchial arch ectoderm at embryonic stages from donor embryos injected with the lineage tracer green fluorescent protein. The neurogenic epibranchial and hypobranchial placodes are shown to develop in certain subregions of a broader branchial placodal area as defined by Six1 and Eya1 expression, viz., adjacent to the dorsal and ventral tip of the pharyngeal pouches, respectively. Grafting experiments show that each of the two hypobranchial placodes gives rise to a small and previously undescribed hypobranchial ganglion (identified by its immunoreactivity for the neuron-specific Hu protein) of unknown function located in the ventral branchial arch region. No contributions of hypobranchial placodes to any other ganglia (including cardiac ganglia and the ganglia of branchiomeric nerves located dorsal to pharyngeal pouches) were found.

Developing submandibular, trigeminal and superior cervical ganglia, which provide innervation to the submandibular glands, were studied for substance P (SP)- and neurokinin A (NKA)-immunoreactive (IR) ganglion cells and nerve fibres in rat. These ganglia were examined by using an indirect immunofluorescence technique at daily intervals from the 16th day in utero (i.u.) until birth, and subsequently on the 2nd, 5th, 7th, 12th, 16th, 30th, 42nd postnatal day and in the adult (3 months). In the submandibular ganglion SP- and NKA-IR cells and fibres first appeared in considerable numbers on the 19th day i.u. (in one sample out of five on the 18th day i.u.), when more than 90% of the ganglion cells were immunoreactive to SP and NKA. The number stayed at more than 90% to the 7th postnatal day and then slowly decreased to the levels of adult animals (18% SP, 17% NKA). The first SP- and NKA-IR ganglion cells and fibres appeared in the trigeminal ganglion on the 18th day i.u. when they represented 7% (SP) and 4% (NKA) of the ganglion cells. The number of SP- and NKA-IR cells increased steadily, reaching a maximum at the time of birth when 68% (SP) and 74% (NKA) of the ganglion cells were immunoreactive. Thereafter they began to decrease toward the level of an adult rat (10% SP, 11% NKA). In the superior cervical ganglion only a few SP- and NKA-IR ganglion cells were detected from the 19th day i.u. to the fifth postnatal day. Positive ganglion cells were also occasionally found in the nerve trunks outside the superior cervical ganglion.(ABSTRACT TRUNCATED AT 250 WORDS)

The development of the hypogastric ganglion of normal and testosterone-treated rats was studied using formaldehyde-induced fluorescence (FIF) method. The fluorescence intensities were recorded microspectrofluorimetrically. In normally developing rats cytoplasmic FIF decreases and cell size increases with age. In normally developing rats cytoplasmic FIF decreases and cell size increases with age. In testosterone-treated animals FIF increases during 2--6 weeks compared to the controls. The differences between control and experimental rats were significant. The diameters were significantly longer in treated animals in three and four week old groups. Vacuolated neurons were seen earlier in testosterone-treated rats. No changes in FIF or in cell size were noticed in the superior cervical ganglion. The male sex steroid, testosterone evidently influences the catecholamine turnover and cellular growth during development in the male pelvic ganglion.

The frequency of pineal ganglia associated with the pineal tract, and the numbers of acetylcholinesterase-positive neurons in these ganglia were studied in the domestic fowl during the post-hatching period by means of the acetylcholinesterase method. Furthermore, the degeneration of nerve cells in pineal ganglia of 40-day-old domestic fowl was investigated in detail at the electron-microscopic level. The rate of pineal organs containing one or more ganglia was 50% in 2- to 13-day-old, 38% in 40-day-old, and only 10% in 1-year-old domestic fowl. In parallel, the number of acetylcholinesterase-reactive nerve cells that constitute individual pineal ganglia decreased after hatching. Various degrees of neuronal degeneration were found in the pineal ganglia: swelling of the endoplasmic reticulum, electron-dense degeneration of the cytoplasm, and pyknosis of the nerve cell nucleus. Clusters of macrophages containing numerous lysosomes filled with debris-like material were scattered in the ganglion. In addition, plasma cells were observed in association with degenerating nerve cells. These results confirm the suggestion that the loss of acetylcholinesterase-positive nerve cells in the pineal ganglia of the domestic fowl is due to naturally occurring, programmed neuronal cell death. This process is discussed with reference to phenomena of cell death observed in other components of central nervous system.

The synthesis in vitro of the putative dipteran juvenile hormone (JHB3) by ring glands isolated from third instar Drosophila melanogaster larvae was quantified by a radiochemical assay. The data indicate that JHB3 synthesis is developmentally regulated during the period prior to wandering until after puparium formation. The highest level of basal production occurred during the postfeeding stage, and synthesis declined after pupariation. Similar relative profiles of synthesis were obtained upon the addition of the JHB3 precursor, farnesoic acid, although the absolute levels of production were elevated considerably. Basal JHB3 production by brain-ring gland complexes in vitro was also highest during the postfeeding stage, although the synthetic rates were much lower than displayed by isolated ring glands. Further analysis revealed that methyl farnesoate, a JHB3 precursor, was synthesized by brain-ring gland complexes in significant quantity. A dual mechanism of brain-centered control of JHB3 biosynthesis is proposed.
