Green space could influence adult cognition and childhood neurodevelopment , and is hypothesized to be partly driven by epigenetic modifications. However, it remains unknown whether some of these associations are already evident during foetal development. Similar biological signals shape the developmental processes in the foetal brain and placenta.Therefore, we hypothesize that green space can modify epigenetic processes of cognition-related pathways in placental tissue, such as DNA-methylation of the serotonin receptor HTR2A. HTR2A- methylation was determined within 327 placentas from the ENVIR ON AGE (ENVIRonmental influence ON early AGEing) birth cohort using bisulphite-PCR-pyrosequencing. Total green space exposure was calculated using high-resolution land cover data derived from the Green Map of Flanders in seven buffers (50 m-3 km) and stratified into low (<3 m) and high (≥3 m) vegetation. Residential nature was calculated using the Land use Map of Flanders. We performed multivariate regression models adjusted for several a priori chosen covariables. For an IQR increment in total green space within a 1,000 m, 2,000 m and 3,000 m buffer the methylation of HTR2A increased with 1.47% (95%CI:0.17;2.78), 1.52% (95%CI:0.21;2.83) and 1.42% (95%CI:0.15;2.69), respectively. Additionally,, we found 3.00% (95%CI:1.09;4.91) and 1.98% (95%CI:0.28;3.68) higher HTR2A- methylation when comparing residences with and without the presence of nature in a 50 m and 100 m buffer, respectively. The methylation status of HTR2A in placental tissue is positively associated with maternal green space exposure. Future research is needed to understand better how these epigenetic changes are related to functional modifications in the placenta and the consequent implications for foetal development.

Purpose: To explore the relation between exercise load, physical activity intensity, and movement disorders during powered wheelchair (PW) mobility in people with severe dyskinetic cerebral palsy (DCP)., Methods: Ten participants with DCP, 6-21 years old, users of a head/foot steering system were included. Dystonia and choreoathetosis were assessed using the Dyskinesia Impairment Mobility Scale (DIMS), heart rate (HR) was used to assess the exercise load of the tasks on the participants, and the accelerometry-based activity index (AI) to measure the physical activity intensity and energy expenditure during mobility task performance., Results: Neck- and distal arm dystonia showed significant correlations with HR (0.64 <  r s  < 0.77; 0.009 <  p  < 0.048), whereas neck- and proximal arm choreoathetosis with AI (0.64 <  r s  < 0.76, 0.011 <  p  < 0.044). Total-body AI was strongly correlated to the AI of the arms (0.66 <  r s  < 0.90,  < 0.001 <  p  < 0.038), but not to the AI of the head., Conclusions: During PW mobility tasks, dystonia is associated to exercise load and choreoathetosis to physical activity intensity and energy expenditure. Findings highlight the difficulties in measuring exercise load and activity intensity in PW users with DCP due to the involuntary hypertonic and/or hyperkinetic hallmark of the movement disorders. Nevertheless, a relaxed surrounding with minimal distractions during PW training may increase learning efficiency. Future studies with a bigger sample size are highly recommended to fully establish the relationship between the variables and to allow generalizability of results.Implications for rehabilitationDystonia is positively related to heart rate during powered mobility, which may be explained by the hypertonic hallmark of dystonia causing an increase in exercise load.Choreoathetosis is positively related to the physical activity index during powered mobility where the hyperkinetic hallmark of choreoathetosis may lead to an increase in physical activity intensity and energy expenditure.Arm overflow movements are the component which contribute the most to total-body activity index, thus, minimizing these movements may lower the overall energy expenditure during powered mobility.Mobility training in a relaxed surrounding with minimal distractions and minimized arm overflow movements may lead to a less-demanding powered wheelchair mobility experience and increased learning efficiency.

Introduction: Telemonitoring applications are expected to become a key component in future healthcare. Despite the frequent use of SpO 2 measurements in telemonitoring of patients with chronic obstructive pulmonary disease (COPD), no profound overview is available about these measurements. Areas covered: A systematic search identified 71 articles that performed SpO 2 measurements in COPD telemonitoring. The results indicate that long-term follow-up of COPD patients using daily SpO 2 spot checks is practically feasible. Very few studies specified protocols for performing these measurements. In many studies, deviating SpO 2 values were used to raise alerts that led to immediate action from healthcare professionals. However, little information was available about the exact implementation and performance of these alerts. Therefore, no firm conclusions can be drawn about the real value of SpO 2 measurements. Future research could optimize performance of alerts using individualized, time-dependent thresholds or predictive algorithms to account for individual differences and SpO 2 baseline changes. Additionally, the value of performing continuous measurements should be examined. Expert commentary: Standardization of the measurements, data science techniques and advancing technology can still boost performance of telemonitoring applications. All these opportunities should be thoroughly explored to assess the real value of SpO 2 in COPD telemonitoring.

In recent years, the lysosome has emerged as a highly dynamic, transcriptionally regulated organelle that is integral to nutrient-sensing and metabolic rewiring. This is coordinated by a lysosome-to-nucleus signaling nexus in which MTORC1 controls the subcellular distribution of the microphthalmia-transcription factor E (MiT/TFE) family of "master lysosomal regulators". Yet, despite the importance of the lysosome in cellular metabolism, the impact of traditional in vitro culture media on lysosomal dynamics and/or MiT/TFE localization has not been fully appreciated. Here, we identify HEPES, a chemical buffering agent that is broadly applied in cell culture, as a potent inducer of lysosome biogenesis. Supplementation of HEPES to cell growth media is sufficient to decouple the MiT/TFE family members-TFEB, TFE3 and MITF-from regulatory mechanisms that control their cytosolic retention. Increased MiT/TFE nuclear import in turn drives the expression of a global network of lysosomal-autophagic and innate host-immune response genes, altering lysosomal dynamics, proteolytic capacity, autophagic flux, and inflammatory signaling. In addition, siRNA-mediated MiT/TFE knockdown effectively blunted HEPES-induced lysosome biogenesis and gene expression profiles. Mechanistically, we show that MiT/TFE activation in response to HEPES requires its macropinocytic ingestion and aberrant lysosomal storage/pH, but is independent of MTORC1 signaling. Altogether, our data underscore the cautionary use of chemical buffering agents in cell culture media due to their potentially confounding effects on experimental results.

Importance of the Field: Inherited lysosomal storage diseases often cause severe disability and have a devastating effect on quality of life. Enzyme replacement therapy (ERT) forms a cornerstone in the treatment of lysosomal enzyme deficiencies. Although for some lysosomal disorders ERT is lifesaving, important intrinsic restrictions of the approach are limited access of infused enzyme to less accessible body compartments such as the CNS, the burden of frequent intravenous administration, the emergence of antibodies and the high associated costs. Pharmacological small molecules may overcome these limitations., Areas Covered in This Review: Several novel therapeutic approaches using small molecules are emerging: substrate reduction therapy, pharmacological chaperone therapy, premature nonsense mutation suppressors and proteostasis regulators., What the Reader Will Gain: Based on an extensive literature search up until June 2010, we here review the various therapeutic approaches with small compounds, including those currently in clinical use and those that have entered clinical trials. Compounds that are still in the preclinical phase are also briefly discussed., Take Home Message: pharmacological small molecules are a new class of agents that show great promise for the treatment of lysosomal storage disorders.

1. Two different methods, categorised as input-output and single output models, were evaluated for slaughter weight prediction of broiler chickens. The input-output models included linear and non-linear recursive modelling with a time-varying model structure, whereas the output models consisted only of empirical growth equations and several growth curve fitting techniques. 2. The results suggested that a simple linear growth curve fitting method gives the greatest accuracy in a prediction horizon of 4 d or less. Error is minimised to an average of 0.14% when 4 d of past information is used to fit a line to predict the end weight one day ahead.

1. In previous studies it was found that automatic broiler weighing systems can have accuracy problems. Researchers reported poor agreement between automatic and manual mean weighing used as a reference. The difference was observed especially after 4 to 5 weeks and this was explained by assuming that heavier birds visited the weighing system less at the end of the growing period. 2. Our study tested this hypothesis under practical production conditions. 3. In total, 193,536 digital images of broilers on and in the neighbourhood of the platforms were collected over three growing periods of 6 weeks. The area (upper view) of the broilers was calculated using image analysis techniques. 4. The hypothesis that the weighing system was used less frequently by heavier animals was confirmed. More specifically, statistical differences between the area of broilers on and in the neighbourhood of the weighing system could be demonstrated for week 5.

1. The objective was to investigate the possibility of detecting daily body weight changes of broiler chickens with computer-assisted image analysis. 2. The experiment included 50 broiler chickens reared under commercial conditions. Ten out of 50 chickens were randomly selected and video recorded (upper view) 18 times during the 42-d growing period. The number of surface and periphery pixels from the images was used to derive a relationship between body dimension and live weight. 3. The relative error in weight estimation, expressed in terms of the standard deviation of the residuals from image surface data was 10%, while it was found to be 15% for the image periphery data. 4. Image-processing systems could be developed to assist the farmer in making important management and marketing decisions.

1. The objective of this work was to explore the possibilities of modelling the static and dynamic responses of total heat production of broiler chickens to step changes in temperature and light intensity (light-dark alterations) using compact dynamic model structures. 2. Seventy-seven experiments were performed in an open-circuit respiration chamber to measure the dynamic response of heat production to step variations in temperature and light (on/off). The animal responses were modelled using transfer function model structures. 3. It was demonstrated that the complex process of the dynamic response of heat production of broiler chickens to step changes in air temperature and light-dark alterations can be modelled assuming 1st order dynamics. The coefficient of determination between measured and simulated heat production was on average 0.83 for responses to air temperature and 0.93 for responses to light-dark alterations.

1. A new open-circuit respiration unit consisting of 6 respiration chambers, gas analysis unit and data-acquisition system is briefly described. 2. Energy and protein metabolism in broiler lines selected for improved food efficiency (FC) or for growth rate (GL) were measured weekly from 3 to 6 weeks of age. 3. Gross and apparent metabolisable energy intake per kg W0.75 was on average higher for GL than for FC chickens without differences in metabolisability. Fed and fasted heat production per kg W0.75 did not differ between the lines. FC chickens retained less energy per kg W0.75 than GL chickens. 4. FC chickens deposited much less of the retained energy as fat than their GL counterparts and also showed greater protein conversion efficiency. The leaner composition of the body weight gain in FC chickens was confirmed by the estimated lower fat deposition per kg W0.75 and by the lower fat: protein ratio.
