The hypothesis postulating that the antioxidant and immunological effects of dietary methionine (Met) in young turkeys (1-8 weeks of age) can be differentiated by level and source of Met was investigated in this study. A total of 544 female Hybrid Converter turkeys were divided into four groups and fed diets in which Met content was tailored through supplementation with dl-methionine (dl-Met) or dl-methionine hydroxy analogue (MHA) to levels recommended by NRC (1994) (Groups dl-Met L and MHA L ) and exceeding them by 50% (Groups dl-Met H and MHA H ). Regardless of its source, the increased dietary Met content led to significantly higher body weight gains but had no effect on feed conversion rates. Moreover, an increased Met content lowered lipid peroxide concentrations in breast meat and increased selected indicators of the plasma antioxidant status like uric acid levels, activity of superoxide dismutase (SOD), glutathione (GSH) concentrations, the ferric-reducing ability of plasma (FRAP), increased immunoglobulin A (IgA) plasma levels and decreased interleukin 6 levels. In comparison with dl-Met, MHA decreased the activities of SOD and catalase, and GSH concentrations in plasma. A dosage by source interaction revealed that the lower MHA content was associated with the lowest plasma GSH concentrations, FRAP values and activities of SOD and catalase. The higher dietary MHA level resulted for most parameters similar values, except for a decrease in lipid peroxide concentrations and an increase in plasma IgA levels. It can be concluded that an increased dietary dl-Met and MHA content (about 150% of the recommendations given by NRC 1994) not only increased the growth rate of young turkeys but also improved their antioxidant status. MHA appears to be a less desirable source of dietary Met for young turkeys when the inclusion level of Met matches the current recommendations. Therefore, a further debate is needed to establish the dietary requirements for Met in poultry.

(1) Two hundred and twenty-seven adult turkeys of both sexes, of two varieties (104 Black and 123 White) were used to evaluate their immunocompetence status and body weights. (2) Response to sheep red blood cells (SRBC) (humoral immunity) was measured by Haemagglutination (HA) test 5 days post immunisation (dpi) and expressed as log2 values. Mercaptoethanol resistant (MER) antibodies representing IgG were determined by Mercaptoethanol HA test and Mercaptoethanol sensitive (MES) antibodies, representing IgM as the difference in total HA titre and IgG. Serum lysozyme concentrations were estimated by 'Lysoplate assay' and expressed in log2 values. (3) Least squares analysis of variance revealed that the White variety had higher adult body weight (4.788 +/- 0.040 kg) than the Black (3.774 +/- 0.044 kg). Sexual dimorphism was apparent and meals were heavier than females in both varieties. The interaction effect of variety and sex on body weight was also significant. (4) Least squares means for immunological traits, namely, total anti-SRBC antibodies, MER, MES titres and serum lysozyme were 7.161 +/- 0.189, 0.801 +/- 0.071, 6.362 +/- 0.160 and 1.766 +/- 0.043 microg/ml, respectively. The Black variety had a higher MES antibody titre than the White. (5) Sex had an effect on all the immunological traits except on MER titres. Females generally had higher anti-SRBC, MER and MES titres and serum lysozyme. The variety x sex interaction effect was significant for MES titres and serum lysozyme. White males had the lowest MES titres.

In this study we investigated a panel of 20 polyclonal and monoclonal antibodies specific for chicken cells and tissues for cross-reactivity with other avian species, i.e. turkey, duck and quail, using immunoperoxidase staining on cryostat sections. The number of cross-reacting antibodies was highest for turkey (12/20) and quail (10/20) and lowest for duck (5/20), reflecting the phylogenetic distance between these birds. In ducks only antibodies specific for mononuclear phagocytes and for stromal molecules in mesenchym, muscle cells, endothelial, and epithelial cells cross-reacted. In turkeys and quails cross-reaction was seen with antibodies against T-helper lymphocytes, IgM- and IgG-positive B cells and plasma cells, and mononuclear phagocytes. In addition, most antibodies specific for stromal molecules reacted against turkey or quail molecules. The panel of reacting antibodies can be used for research into the natural and specific defence mechanisms of turkeys and quails.

1. This study examined the effect of increasing amounts of dietary polyunsaturated fatty acids on the fatty acid composition in serum and antibody production following a standard vaccination programme in growing turkeys. Turkey poults were fed on 5 diets containing 75g/kg added fat made up of different proportions of palm and soyabean oils, and were vaccinated against Newcastle disease, infectious bronchitis and necrotic enteritis according to a standard vaccination programme. Blood samples were taken before and one week after each vaccination. 2. Fatty acid composition in serum reflected the composition of the diets although arachidonic acid concentration was not changed by dietary fatty acid content. Growth, erythrocyte and leukocyte parameters were not affected by the respective diets. 3. Specific antibody production was related quadratically to serum linoleic and total n-6 polyunsaturated fatty acid concentrations. No correlation was found with linolenic or arachidonic acids. 4. It is concluded that dietary fatty acid composition can augment the specific anti-vaccine immune response in turkey poults.

1. The effect of dietary vitamin A on antibody production and T-cell proliferative response was determined in poults from 21 to 41 d old. Poults were fed on soyabean meal-sorghum-based diets with concentrations of supplemented vitamin A from 0 to 13.2 micrograms/g retinol equivalents from hatching and were immunised with Newcastle disease virus (NDV) and turkey pox vaccines. T-cell proliferation response to concanavlin A was determined in vitro at 31 d old. Antibodies to NDV and turkey pox in serum were determined at 10 and 20 d after inoculation. 2. Poults receiving the diet with no added dietary vitamin A died by 22 d and had very low concentrations of plasma and liver vitamin A. 3. Increasing dietary concentrations of vitamin A enhanced the proliferative response until the diet contained 6.0 micrograms/g, above which the response began to decrease. The antibody titres to NDV and turkey pox increased as dietary vitamin A increased, with maximal values found 10 d after inoculation with 6.0 micrograms/g. At 20 d after inoculation low antibody titres were found with low vitamin A intake. 4. These data suggest that maximal immune responses in the poult may be achieved at dietary intakes of vitamin A at or higher than those recommended by NRC (1984, 1994).

1. White blood cell responses of broilers, turkeys and ducks were examined at regular intervals after being subjected to various degrees of food restriction. 2. Restricted-fed broilers showed increases in heterophil and basophil numbers, together with a corresponding decrease in lymphocytes. The heterophil/lymphocyte ratio was raised. There were no differences between broiler strains. 3. After only one week of feeding restricted diets, heterophils were significantly raised in selected and unselected 2-week-old ducks. At 21 weeks of age, those ducks receiving 50% of food required to achieve their ad libitum-fed body weight had raised heterophils. 4. Ducks receiving food to achieve 25% of ad libitum-fed birds produced a marked basophilia, but no heterophilia. 5. After two weeks of food restriction, turkeys responded with significant heterophil/lymphocyte ratios following two degrees of restricted feeding. 6. It was concluded that in some poultry, a heterophilia may be the response to mild to moderate stress but a basophilia may result after severely stressing birds.

1. Bovine serum albumin (BSA) in saline applied to the cloacal "lips" of turkeys was sucked in by quick movements (cloacal drinking). 2. BSA-stimulated antibody production was limited to lymphoid cells in the bursa of Fabricius and in the caecal tonsils. No serum antibody was detected after cloacal drinking of BSA. 3. This response is probably of significance in the local defence mechanism of the intestine.
