In recent years, nanotechnology has emerged as cutting-edge technology with multifarious applications in a wide array of fields. Green synthesis of iron nanoparticles (FeNP) are an upcoming cost effective and eco-friendly technique and recently gained significant importance. In the present study, green FeNPs were prepared using leaf litter which is one of the major seasonal waste contributors in urban built-up areas. Shedding trees during winter months (January – March) were selected. Most abundant trees were Pongamia pinnata (Indian beech), Morus alba (mulberry), Prosopis juliflora (mesquite) and Kigelia africana (sausage tree). Synthesized FeNPs were further used for degrading two commercial dyes, eosin yellow and fuchsin basic, via Fenton's mechanism. The study showed that the prepared nanoparticles were of iron oxides, but also reported presence of polyphenols as a capping agent. Dye degradation efficiency of nanoparticles synthesized by P. pinnata leaf litter was recorded to be highest, whereas the efficiency of nanoparticles synthesized by K. africana leaf litter was lowest. Chances of iron leaching during dye degradation process was also tested and observed that Fe was present in treated water below the standard guidelines. Thus, FeNPs can serve as a low-cost solution to remediate water pollutants with a green approach. Implications: Nanoparticles prepared in the study were showed as a promising adsorbent and demonstrating high surface area and well-developed porosity. The prepared adsorbent will have a great impact on wastewater treatment technology and possible applications at a large scale. There are several applications of nanoparticles in pollution remediation and at the same time it can solve solid waste issues as it required to prepare nanoparticles. With the advancement of nanotechnology, many different forms of nanoparticles (NPs) are created, which specifically enhance anticancer drug delivery to tumour cells. Albumin bio-macromolecule is a flexible protein carrier for the delivery of drugs that is biodegradable, biocompatible, and non-toxic. As a result, it presents itself as an ideal material for developing nanoparticles for anticancer drug delivery. Toxicological investigations demonstrated that this novel drug delivery technique is safe for use in the human population. Furthermore, drug compatibility with the albumin nanoparticle is remarkable. The robust structure of the nanoparticle, high drug encapsulation, and customisable drug release make it a promising carrier option for the treatment of lung cancer. In the present work, Ni0.6Cd0.4DyxFe2-xO4 (x = 0.0, 0.05, 0.10, 0.15, 0.20) nanoparticles (NPs) were synthesized by using sol–gel method. X-ray diffraction (XRD) patterns confirmed that the pure and dysprosium substituted Ni–Cd ferrites are in single-phase spinel structures, while a trace of DyFeO3 appears as a minor phase for higher concentrations (x = 0.10, 0.15 and 0.20). The Debye–Scherrer's method and Williamson–Hall (W-H) method were used to evaluate the crystallite sizes and lattice strain. Scanning electron microscope (SEM) images show that reduction of grain size with Dy3+ content. Elemental composition features of samples were examined by Energy-dispersive X-ray spectroscopy (EDS). The average particles size estimated from transmission electron microscopy (TEM) analysis is in good agreement with results obtained from XRD. The results showed that saturation magnetization (Ms) decreases and coercivity (Hc) increases with an increase in Dy3+ concentrations. Temozolomide is a drug approved for treating glioblastomas, which has 100% oral bioavailability but gets degraded at physiological pH thus having very short half-life and only 20–30% brain bioavailability. Due to its amphiphilic nature, reported nanoformulations exhibits poor drug loading. The objective of this work was to formulate lipid-based drug delivery systems to enhance the brain bioavailability by prolonging the drug release and circulation time of the drug to overcome the limitations of the existing therapies and possible reduction of side effects. The size of the nanocarriers obtained was less than 300 nm and the PDI obtained was less than 0.3. The designed formulation showed higher entrapment efficiency as compared to the other reported nanocarriers of temozolomide. The designed formulations showed prolonged drug release from 12 to 20 h compared to 6 h for the pure drug. About 95% of the pure drug was degraded at plasma pH at the end of 12 h, whereas only 68% and 77% was degraded when entrapped inside the lipid crystal nanoparticles and proliposomes respectively. The purpose of the present study was to optimise diclofenac diethylamine-loaded liquid crystal nanoparticles (LCNPs) using the principles of quality by design. Based on risk assessment, the effect of various formulation variables on the critical quality attributes was investigated. A three-level Box-Behnken design with 14 runs was utilised for optimisation. The LCNPs were evaluated for size, polydispersity index, zeta potential, entrapment efficiency, morphology, solid-state characterisation, and drug release. The LCNPs were found to show prolonged drug release up to 12 h as compared to the free drug which showed a 100% release in less than 3 h. The optimised formulation was further investigated for scale-up studies, incorporated into carbopol gel and characterised for rheological parameters, skin permeation, and skin accumulation. Ex-vivo skin permeation studies revealed 1.55 times more permeation as compared to the marketed formulation. Sol-gel (SG) and direct precipitation (DP) routes were employed to synthesize zinc oxide (ZnO) nanoparticles. Al doping and Al-Cu dual doping in ZnO nanoparticles were performed. The structural, morphological, optical, wetting properties and antifungal activity of the undoped and doped nanoparticles were studied. The Al doping transformed the ZnO to zinc aluminate nanostructure, while Al-Cu dual doping produced zincite and tenorite phases of ZnO. FESEM revealed the morphology of undoped and doped nanoparticles. The doping of ZnO increased the band gap compared to the band gap of the ZnO nanoparticles. Photoluminescence (PL) spectroscopy endorsed the formation of zinc aluminate, zincite and tenorite phases. The hydrophilic SG-derived ZnO nanoparticles transformed to the hydrophobic due to Al doping which is attributed to the variation in the roughness of the doped nanostructures. A novel one-pot synthetic method to produce crystalline tri-octylphosphine (TOP) capped iron phosphide nanoparticles is reported here. Standard method of synthesizing FeP includes preparation of a precursor, sodium phosphide, which is finally reacted with ferric chloride (FeCl3).The methods for synthesizing iron phosphide (FeP), reported so far, rely on the use of toxic red or yellow phosphorus to generate the precursor, Sodium phosphide (Na3P). In present investigation, instead of red or yellow phosphorus, a relatively less toxic substance TOP and sodium metal (Na) have been used to yield Na3P. The synthesized nanoparticles were fully characterized by X-ray diffraction pattern (XRD), Infrared spectroscopy (IR), X-ray photoelectron spectroscopy (XPS), Transmission electron microscopy (TEM) and superconducting quantum interference device (SQUID) analyses. This study focuses on the electrical characteristics of a strongly polar nematic liquid crystal, Hexyloxy-cyanobiphenyl (6OCB), doped with a low concentration (2% by weight) of citrate buffer stabilised gold nanoparticles (GNPs) at low frequencies between 20 Hz and 35 MHz. The doped samples have lower values of nematic-isotropic transition temperature, permittivity (both parallel and perpendicular to the field direction) and dielectric anisotropy; however, relaxation time and activation energy were increased. The observed results could be explained on the basis of weakly anisotropic nature of GNPs and a local rearrangement of liquid crystal molecules surrounding the nanoparticles. Moreover, a complimentary suggestion on a possible change in the dipole-dipole correlation is made to explain the difference in changes (qualitative and quantitative) observed for permittivity of the host nematic liquid crystal doped with GNP. Temperature dependent dielectric relaxation studies indicate an increase in viscosity and potential barrier; and hence a change in strength of inter-molecular and intra-molecular interactions is suggested. The purpose of this study was to develop PEGylated nanoparticles of bendamustine (BM) to improve therapeutic efficiency of drug and reduce the side-effects. The nanoparticles were prepared by a modified diffusion–emulsification method. The particle size and zeta potential of optimized BM-loaded PEGylated NPs were found to be 256 nm and −29.1 mV. Thein vitrorelease showed biphasic behavior, with initial burst release followed by slow sustained delivery. The anti-tumor activity was determined using the A- 549 cell line, by the MTT assay. Four transition metal nanoparticles (TMNs) of 3d series (Cu, Co, Ni, and Fe) were prepared by hydrazine reduction of metal chloride in ethylene glycol at 60°C and characterized by X-ray diffraction (XRD). The XRD pattern showed average particle sizes for Cu, Ni, Co, and Fe of 16.7, 40.5, 27.4, and 35.0 nm, respectively. The activity of these TMN accelerants on the thermal decomposition of ammonium perchlorate (AP) was investigated using thermogravimetry (TG), differential scanning calorimetry (DSC), and ignition delay studies. Isothermal TG data were used to evaluate the kinetic parameters by model fitting as well as an isoconversional methods. The activation energy for thermal decomposition of AP was found to be 66.8, 68.7, 78.5, and 85.4 kJmol−1, respectively, for Co, Cu, Ni, and Fe, when they were mixed with AP. Hence, the order of activity was found to be Co > Cu > Ni > Fe. The accelerant effect of nanoparticles of TMNs was found to be better than their respective nano-oxides. [ABSTRACT FROM PUBLISHER]

Purpose: Neuroendocrine tumors often present a diagnostic and therapeutic challenge. We have aimed to synthesize and develop biodegradable nanoparticles of somatostatin analogue, octreotide for targeted therapy of human neuroendocrine pancreatic tumor. Methods: Direct solid phase peptide synthesis of octreotide was done. Octreotide loaded PCL/PEG nanoparticles were prepared by solvent evaporation method and characterized for transmission electron microscopy, differential scanning calorimetery (DSC), Zeta potential measurement studies. The nanoparticles were evaluated in vitro for release studies and peptide content. For biological evaluations, receptor binding & cytotoxicity studies were done on BON-1 neuroendocrine tumor cell line. Biodistribution of radiolabeled peptide and nanoparticles, tumor regression studies were performed on tumor-bearing mouse models. Results: We have synthesized and purified octreotide with the purity of 99.96% in our laboratory. PEG/PCL nanoparticles with an average diameter of 130-195 nm having peptide loading efficiency of 66-84% with a negative surface charge were obtained with the formulation procedure. Octreotide nanoparticles have a negative action on the proliferation of BON-1 cells. In vivo biodistribution studies exhibited major accumulation of octreotide nanoparticles in tumor as compared to plain octreotide. Octreotide nanoparticles inhibited tumor growth more efficiently than free octreotide. Conclusions: Thus, it was concluded that the PCL/PEG nanoformulation of octreotide showed high tumor uptake due to the enhanced permeation and retention (EPR) effect and then peptide ligand imparts targetability to the sst2 receptor and there by showing increase tumor growth inhibition. The present study evaluates the potential of surface functionalized gelatin nanoparticles ( f-GNPs) for efficient macrophage-specific delivery of amphotericin B (AmB) in the treatment of visceral leishmaniasis (VL). Further, the effect of spacer for macrophage targeting was also evaluated. Gelatin was functionalized either through conjugation to mannose via direct coupling (mGelatin) or via PEG spacer (m-Gelatin), and the synthesis was confirmed by FTIR. AmB-loaded f-GNPs, that is, mGNPs and m-GNPs prepared from mGelatin and m-Gelatin conjugates, respectively, were characterized. In vitro concanavalin A (Con-A) agglutination assay confirmed the availability of mannose on the surface of these f-GNPs. Kinetics of cellular uptake of AmB-loaded f-GNPs by J774A.1 macrophage cells assessed through flow cytometry demonstrated a steady increase and maximum cell-associated fluorescence was observed at 4h for m-GNPs and 6 h for m-GNPs. Measurement of cytotoxicity using Annexin-V–FITC/PI apoptosis assay delineated marginal changes (7–9%) in treated macrophages following 48 h incubation, establishing the safety of f-GNPs. m-GNPs showed a 5.4-fold reduction in IC50 in comparison with plain AmB suggesting significant enhancement of antileishmanial activity. Our results indicate that f-GNPs could be a promising carrier for specific delivery of AmB to macrophages for effective treatment of VL. Hydrophilic drugs/peptides have poor cross Blood–brain permeability. Various drug delivery systems with diverse surfacial characteristics have been reported for effective translocation of drugs across Blood–brain barrier. In present investigation, the potential of engineered albumin nanoparticles was evaluated for brain specific delivery after intravenous administration. Long circulatory PEGylated albumin nanoparticles encapsulating water-soluble antiviral drug azidothymidine (AZT) were prepared by ultra-emulsification method using chemical cross-linking by glutaraldehyde. Surface of the PEGylated nanoparticles was modified by anchoring transferrin as a ligand for brain targeting. Nanoparticles were characterized for their size, polydispersity, surfacial charge, drug loading and in vitro drug release. Fluorescence studies revealed the enhanced uptake of transferrin-anchored nanoparticles in the brain tissues when compared with unmodified nanoparticles.
