We simulate and analyse three types of two-dimensional networked polymers which have been predicted to exhibit on-axis auxetic behaviour (negative Poisson's ratio), namely (1) polyphenylacetylene networks that behave like flexing re-entrant honeycombs, commonly referred to as 'reflexynes', (2) polyphenylacetylene networks that mimic the behaviour of rotating triangles, commonly referred to as 'polytriangles' and (3) networked polymers built from calix[4]arene units. More specifically, we compute and compare their in-plane off-axis mechanical behaviour, in particular their off-axis Poisson's ratios and show that in some cases, the sign and magnitude of the Poisson's ratio are dependent on the direction of loading. We propose two functions that can provide a measure for the extent of auxeticity for such anisotropic materials and show that the polytriangles are predicted as the most auxetic when compared with the other networks with the reflexyne re-entrant networks being the least auxetic. Social workers often need a larger range of assessment scales to be used in education, research, and practice. The Higher Power Relationship Scale (HPRS) is a new brief instrument designed for easy administration and scoring that may be of use to social work educators, researchers, and practitioners, particularly those who work in the field of addictions. This paper describes the HPRS; provides information about its administration, scoring, and interpretation; and describes the initial research conducted to validate the scale. The results clearly indicate that the scale ( I )has good reliability, (2) is strong enough in terms of its measurement error traits to recommend it for use in educational, research, and clinical settings, and (3) appears to be acceptable in terms of its content and construct validity. Preliminary use in chemically dependent settings illustrated the value of the scale in understanding respondents' relationships with a higher power and Alcoholics Anonymous affiliation. Rapid growth rates and versatility have made hybrid poplar (Populus L.) a common choice to fulfill timber and pulp needs in temperate regions with adequate precipitation. Hybrid poplar production in drier regions has recently received interest as an irrigated source of cut logs and shredded fiber. Ten hybrid poplar clones were selected from nurseries in Washington and Oregon to determine their growth potential and water-use requirements on a calcareous soil at the NMSU Agricultural Science Center in Farmington, NM. Planted in May 2002, the clones were evaluated for survival growth characteristics over two sea- sons. Clones 50-194 and 184-411 had poor survival and were not continued during the 2003 season. Clone OP-367 had low chlorosis ratings, the greatest height (4.16 m), basal diameter (9.16 cm), diameter at breast height (4.65 cm) and wood volume (2.87 m³ ha-1) during both seasons. A range of responses to environmental conditions of the Four Corners region was exhibited. Mid-season leaf greenness, as determined with a SPAD-502 chlorophyll meter, was a good predictor of growth characteristics. TAF4b is a gonadal-enriched subunit of the TFIID complex that is required for mouse fertility. TAF4b-deficient male mice undergo a complex series of developmental defects that result in the inability to maintain long-term spermatogenesis. To decipher the transcriptional mechanisms upon which TAF4b functions in spermatogenesis, we used two-hybrid screening to identify a novel TAF4b-interacting transcriptional cofactor, ZFP628. Deletion analysis of both proteins reveals discrete and novel domains of ZFP628 and TAF4b protein that function to bridge their direct interaction in vitro. Moreover, co-immunoprecipitation of ZFP628 and TAF4b proteins in testis-derived protein extracts supports their endogenous association. Using CRISPR-Cas9, we disrupted the expression of ZFP628 in the mouse and uncovered a post-meiotic germ cell arrest at the round spermatid stage in the seminiferous tubules of the testis in ZFP628-deficient mice that results in male infertility. Coincident with round spermatid arrest, we find reduced mRNA expression of transition protein (Tnp1 and Tnp2) and protamine (Prm1 and Prm2) genes, which are critical for the specialized maturation of haploid male germ cells called spermiogenesis. These data delineate a novel association of two transcription factors, TAF4b and ZFP628, and identify ZFP628 as a novel transcriptional regulator of stage-specific spermiogenesis. In a model, such as a logit regression model, we wish to compare the relative importance of two groups of predictors for various objectives. In the example that motivated this work, the model predicts patient outcomes during hospital stays, and we wish to measure the relative contribution of patient and hospital characteristics to the variation in outcomes among patients and among hospitals. This is done using the relative dispersion of patient and hospital contributions to the fitted outcomes. As is seen, this question is distinct from other common questions, including the quality of the overall fit, the degree to which the outcome is accurately predicted, the statistical significance of groups of predictors, and the correlations among and between groups of predictors. Relevant point estimates, confidence intervals, and hypothesis tests are developed. In the example, we examine three outcome measures and find that nearly all of the predictable variation in patient outcomes and most of the predictable variation in outcomes among hospitals reflects variation in patient characteristics rather than hospital characteristics; however, this is true in varying degrees for the three outcomes. In an emergency department (ED) sample, we investigated the concordance between identification of suicide-related visits through standardized comprehensive chart review versus a subset of 3 specific chart elements: ICD-9-CM codes, free-text presenting complaints, and free-text physician discharge diagnoses. The method for this study was review of medical records for adults (≥18 years) at 8 EDs across the United States. A total of 3,776 charts were reviewed. A combination of the 3 chart elements (ICD-9-CM, presenting complaints, and discharge diagnoses) provided the most robust data with 85% sensitivity, 96% specificity, 92% PPV, and 92% NPV. These findings highlight the use of key discrete fields in the medical record that can be extracted to facilitate identification of whether an ED visit was suicide-related. [ABSTRACT FROM AUTHOR]

The objective of this article was to conduct a treatment development study to examine the feasibility, acceptability, and preliminary efficacy of treating depressed, overweight/obese adolescents using both an exercise regimen and a Cognitive Behavioral Therapy (CBT) protocol modified to address aspects of healthy living and nutrition (CBT plus healthy lifestyle; CBT-HL). A randomized controlled repeated measures design was used to test the hypothesis that CBT-HL would lead to greater reductions in depressed mood and weight compared to CBT for Depression Only (CBT). Participants (n=33; 24 in CBT-HL condition) included 33 adolescents (median age 15, 73% female, 61% white, 36% Hispanic) who met DSM-IV criteria for Current Major Depressive Episode (MDE) and had BMI ≥ 85th percentile. CBT-HL was found to be feasible to implement with most adolescents. Both conditions resulted in improvement in depressed mood. The CBT-HL protocol wasmore effective in stabilizing weight status as assessed by BMI. Percent time spent in MVPAwas increased at 12 weeks for adolescents in CBT-HL compared to those in CBT. The CBT-HL protocol was acceptable to most, but not all, adolescents, and resulted in an improvement in depressed mood as well as stabilization of weight status. A larger study to test efficacy and moderators of treatment outcome is necessary to better understand which adolescents would benefit most from the increased demands of exercise and adhering to nutrition recommendations in addition to standard CBT for depression. The case of a female runner presenting with previously undiagnosed chronic hip pain is reported. Initial X-ray and MRI of her affected hip were reported as normal; a subsequent review of these films by a specialist musculoskeletal radiologist revealed this was incorrect. A repeat MRI scan was performed using dedicated hip views which revealed a large, intra-articular, ganglion cyst within the acetabulum and an acetabular labral tear. This case raises awareness of acetabular ganglions as a possible cause of hip pain. It also reinforces the need to be alert to potential operator variability when interpreting radiology reports. Background/Study Context: Conceptual frameworks are analytic models at a high level of abstraction. Their operationalization can inform randomized trial design and sample size considerations. Methods: The Advanced Cognitive Training for Independent and Vital Elderly (ACTIVE) conceptual framework was empirically tested using structural equation modeling (N=2,802). ACTIVE was guided by a conceptual framework for cognitive training in which proximal cognitive abilities (memory, inductive reasoning, speed of processing) mediate treatment-related improvement in primary outcomes (everyday problem-solving, difficulty with activities of daily living, everyday speed, driving difficulty), which in turn lead to improved secondary outcomes (health-related quality of life, health service utilization, mobility). Measurement models for each proximal, primary, and secondary outcome were developed and tested using baseline data. Each construct was then combined in one model to evaluate fit (RMSEA, CFI, normalized residuals of each indicator). To expand the conceptual model and potentially inform future trials, evidence of modification of structural model parameters was evaluated by age, years of education, sex, race, and self-rated health status. Results: Preconceived measurement models for memory, reasoning, speed of processing, everyday problem-solving, instrumental activities of daily living (IADL) difficulty, everyday speed, driving difficulty, and health-related quality of life each fit well to the data (all RMSEA < .05; all CFI > .95). Fit of the full model was excellent (RMSEA = .038; CFI = .924). In contrast with previous findings from ACTIVE regarding who benefits from training, interaction testing revealed associations between proximal abilities and primary outcomes are stronger on average by nonwhite race, worse health, older age, and less education (p < .005). Conclusions: Empirical data confirm the hypothesized ACTIVE conceptual model. Findings suggest that the types of people who show intervention effects on cognitive performance potentially may be different from those with the greatest chance of transfer to realworld activities. There is considerable interest in the role of sleep in weight regulation, yet few studies have examined this relationship in overweight/obese (OW/OB) adults. Using a within-subject, counterbalanced design, 12 OW/OB women were studied in lab with two nights of short (5 hr time in bed [TIB]) and two nights of long (9 hr TIB) sleep. Hunger, consumption at a buffet, and fasting hormone levels were obtained. Significant polysomnographic differences occurred between conditions in total sleep time and sleep architecture (ps < .001). Percent energy from protein at the buffet increased following short sleep. No differences were observed for total energy intake or measured hormones. Further research is needed to determine how lengthening sleep impacts weight regulation in OW/OB adults. [ABSTRACT FROM AUTHOR]

Seven acidophilic/acidotolerant fungal strains were characterized from samples of process waters (raffinate) at one of Australia's largest uranium mines, the Ranger Mine in Northern Territory. They were isolated from raffinate, which typically were very acidic (pH 1.7-1.8) and contained high concentrations of total dissolved/colloidal salts (> 100 g/L). Five of the isolates correspond to two new acidotolerant Ascomycota fungi. The first is a member of a new genus, here described as Fodinomyces (Teratosphaeriaceae, Capnodiales, Dothideomycetes) and does not show clear close affiliation with any other described fungus in the scientific literature. The second belongs to the genus Coniochaeta (Coniochaetaceae, Coniochaetales, Sordariomycetes) and is closely related to Coniochaeta hansenii. [ABSTRACT FROM AUTHOR]

Purpose: The objectives of this study were to develop numerical models of interstitial ultrasound ablation of tumours within or adjacent to bone, to evaluate model performance through theoretical analysis, and to validate the models and approximations used through comparison to experiments. Methods: 3D transient biothermal and acoustic finite element models were developed, employing four approximations of 7-MHz ultrasound propagation at bone/soft tissue interfaces. The various approximations considered or excluded reflection, refraction, angle-dependence of transmission coefficients, shear mode conversion, and volumetric heat deposition. Simulations were performed for parametric and comparative studies. Experiments within e x vivo tissues and phantoms were performed to validate the models by comparison to simulations. Temperature measurements were conducted using needle thermocouples or magnetic resonance temperature imaging (MRTI). Finite element models representing heterogeneous tissue geometries were created based on segmented MR images. Results: High ultrasound absorption at bone/soft tissue interfaces increased the volumes of target tissue that could be ablated. Models using simplified approximations produced temperature profiles closely matching both more comprehensive models and experimental results, with good agreement between 3D calculations and MRTI. The correlation coefficients between simulated and measured temperature profiles in phantoms ranged from 0.852 to 0.967 (p-value < 0.01) for the four models. Conclusions: Models using approximations of interstitial ultrasound energy deposition around bone/soft tissue interfaces produced temperature distributions in close agreement with comprehensive simulations and experimental measurements. These models may be applied to accurately predict temperatures produced by interstitial ultrasound ablation of tumours near and within bone, with applications toward treatment planning. Background and aims. Primary sclerosing cholangitis (PSC) is a chronic cholestatic liver disease characterized by progressive inflammation and fibrosis of the bile ducts eventually leading to biliary cirrhosis. Recent genetic studies in PSC have identified associations at 2q13, 2q35, 3p21, 4q27, 13q31 and suggestive association at 10p15. The aim of this study was to further characterize and refine the genetic architecture of PSC. Methods. We analyzed previously reported associated SNPs at four of these non-HLA loci and 59 SNPs tagging the IL-2/IL-21 (4q27) and IL2RA (10p15) loci in 992 UK PSC cases and 5162 healthy UK controls. Results. The most associated SNPs identified were rs3197999 (3p21 ( MST1), p = 1.9 × 10-6, ORA vs G = 1.28, 95% CI (1.16-1.42)); rs4147359 (10p15 ( IL2RA), p = 2.6 × 10-4, ORA vs G = 1.20, 95% CI (1.09-1.33)) and rs12511287 (4q27 ( IL-2/IL-21), p = 3.0 × 10-4, ORA vs T = 1.21, 95% CI (1.09-1.35)). In addition, we performed a meta-analysis for selected SNPs using published summary statistics from recent studies. We observed genome-wide significance for rs3197999 (3p21 ( MST1), Pcombined = 3.8 × 10-12) and rs4147359 (10p15 ( IL2RA), Pcombined = 1.5 × 10-8). Conclusion. We have for the first time confirmed the association of PSC with genetic variants at 10p15 ( IL2RA) locus at genome-wide significance and replicated the associations at MST1 and IL-2/IL-21 loci in a large homogeneous UK population. The technique of H(D) atom photofragment translation spectroscopy has been used to investigate the dissociation of jet-cooled HOD molecules following excitation to individual rovibrational levels of its [image omitted](1B1) Rydberg state near 124 nm. Spectra have been recorded for both the D + OH and H + OD dissociation channels. The branching ratios between OH/OD(X [image omitted], high v, low N), OH/OD(X [image omitted], low v, high N), and OH/OD(A [image omitted]) channels vary between different parent rotational levels, and between the OH and OD products. The variation between the OH and OD channels can be attributed to an isotopic mass effect on the rotational axes which influences non-adiabatic coupling strengths. In addition, there is a population alternation with product rotational quantum number N for the OH/OD(X, high v, low N) product, most striking for the OH case, the sense of which is correlated with the value of the parent pseudo-quantum number [image omitted]. It is shown that this is a consequence of recoil forces which lead to a closing of the HOD angle to near 90° accompanying non-adiabatic transfer from the [image omitted] state to the [image omitted](1B1) state. The oscillation in population then derives from the symmetry properties around this angle of the product rotational wavefunctions. This suggests that the symmetry-induced population oscillation can only occur for a narrowly defined angular dissociation path. Model calculations also explain the different structure of the spin-orbit doubled low N band heads for each v(OH/OD) which also alternates for even and odd [image omitted]. Lactobacillus plantarum 299v is a probiotic bacterium effective in treating symptoms of irritable bowel syndrome. A mouse model of irritable bowel syndrome (IBS) can be produced by rectal administration of 1% allyl isothiocyanate (oil of mustard) in 30% ethanol. This study examined the effect of L. plantarum 299v on colonic inflammation and motility in the oil of mustard model. L. plantarum 299v (1×109 cfu) was gavaged for up to 28 days, beginning either 7 days before (pretreatment) or 8 days after oil of mustard administration (post-treatment). Colonic interferon gamma (IFNγ) release as an assessment of the inflammatory response was measured at day 4 and day 20 following oil of mustard administration. Small intestinal transit was assessed via the dye-transit score technique at day 20. L. plantarum 299v reduced inflammation at both day 4 and day 20 and normalized intestinal transit rates in the oil of mustard murine IBS model. [ABSTRACT FROM AUTHOR]

Background: Although the body mass index (BMI, kg m-2) is widely used as a measure of adiposity, it is a measure of excess weight, rather than excess body fat. It has been suggested that skinfold thicknesses be measured among overweight children to confirm the presence of excess adiposity. Objective: The present study examined the additional information provided by skinfold thicknesses on body fatness, beyond that conveyed by BMI-for-age, among healthy 5- to 18-years old (n = 1196). Methods and procedures: Total body dual-energy X-ray absorptiometry (DXA) provided estimates of % body fat, and the sum of two skinfolds (triceps and subscapular) was used as an indicator of the overall skinfold thickness. Results: As assessed by the multiple R2s and the residuals of various regression models, information on the skinfold sum significantly ( p < 0.001) improved the prediction of body fatness beyond that obtained with BMI-for-age. For example, the use of the skinfold sum, in addition to BMI-for-age, increased the multiple R2s for predicting % body fat from 0.81 to 0.90 (boys), and from 0.82 to 0.89 (girls). The use of the skinfold sum also reduced the overall prediction errors (absolute value of the residuals) for % body fat by 20-30%, but these reductions varied substantially by BMI-for-age. Among overweight children, defined by a BMI-for-age ≥95th percentile, the skinfold sum reduced the predication errors for % body fat by only 7-9%. Conclusions: Although skinfold thicknesses, when used in addition to BMI-for-age, can substantially improve the estimation of body fatness, the improvement among overweight children is small. Abstract: Although it is known to be a rich source of the putative anti-cancer chemicals isothiocyanates, watercress has not been extensively studied for its cancer preventing properties. The aim of this study was to investigate the potential chemoprotective effects of crude watercress extract toward three important stages in the carcinogenic process, namely initiation, proliferation, and metastasis (invasion) using established in vitro models. HT29 cells were used to investigate the protective effects of the extract on DNA damage and the cell cycle. The extract was not genotoxic but inhibited DNA damage induced by two of the three genotoxins used, namely hydrogen peroxide and fecal water, indicating the potential to inhibit initiation. It also caused an accumulation of cells in the S phase of the cell cycle indicating (possible) cell cycle delay at this stage. The extract was shown to significantly inhibit invasion of HT115 cells through matrigel. Component analysis was also carried out in an attempt to determine the major phytochemicals present in both watercress leaves and the crude extract. In conclusion, the watercress extract proved to be significantly protective against the three stages of the carcinogenesis process investigated. [ABSTRACT FROM AUTHOR]

Molecular chlorine, methane and helium are co-expanded into the extraction region of a Wiley–McLaren time-of-flight spectrometer. After preparing the first overtone of the antisymmetric stretch (?3?=?2) with direct IR excitation, the reaction is initiated by photolysing Cl2 with 355?nm light to produce mono-energetic Cl atoms with a translational energy of 0.18?eV. The CD3 and DCl products are state-selectively detected via resonance enhanced multiphoton ionization (REMPI) and analysed with the core-extraction technique. Unusual structure in the 3p z CD3 REMPI spectrum suggests the presence of a previously unobserved transition, which we assign to the stretch–bend combination () band. The product correlated energy disposal and the scattering distributions are compared with the same quantities for the Cl?+?CH4(?3?=?2) reaction and found to be similar, although subtle differences are observed. The results for the Cl?+?CD4(?3?=?2) reaction support a localized chemistry model in which the Cl atom interacts with a single C–D oscillator and leaves the CD3 methyl radical as a spectator. Benefit-cost analysis is of growing importance in developing policies to reduce exposures to environmental contaminants. To quantify health benefits of reduced exposures, economists generally rely on dose-response relationships estimated by risk assessors. Further, to be useful for benefits analysis, the endpoints that are quantified must be expressed as changes in incidence of illnesses or symptoms that are readily understood by and perceptible to the layperson. For most noncancer health effects and for nonlinear carcinogens, risk assessments generally do not provide the dose-response functions necessary for economic benefits analysis. This article presents the framework for a case study that addresses these issues through a combination of toxicology, epidemiology, statistics, and economics. The case study assesses a chemical that disrupts proper functioning of the thyroid gland, and considers the benefits of reducing exposures in terms of both noncancer health effects (hypothyroidism) and thyroid cancers. The effects are presumed to be due to a mode of action involving interference with thyroid–pituitary functioning that would lead to nonlinear dose response. The framework integrates data from animal testing, statistical modeling, human data from the medical and epidemiological literature, and economic methodologies and valuation studies. This interdisciplinary collaboration differs from the more typical approach in which risk assessments and economic analyses are prepared independently of one another. This framework illustrates particular approaches that may be useful for expanded quantification of adverse health effects, and demonstrates the potential of such interdisciplinary approaches. ABSTRACT: The human relevance framework (HRF) outlines a four-pan process, beginning with data on the mode of action (MOA) in laboratory animals, for evaluating the human relevance of animal tumors. Drawing on U.S. EPA and IPCS proposals for animal MOA analysis, the HRF expands those analyses to include a systematic evaluation of comparability, or lack of comparability, between the postulated animal MOA and related information from human data sources. The HRF evolved through a series of case studies representing several different MOAs. HRF analyses produced divergent outcomes, some leading to complete risk assessment and others discontinuing the process, according to the data available from animal and human sources. Two case examples call for complete risk assessments. One is the default: When data are insufficient to confidently postulate a MOA for test animals, the animal tumor data are presumed to be relevant for risk assessment and a complete risk assessment is necessary. The other is the product of a data-based finding that the animal MOA is relevant to humans. For the specific MOA and endpoint combinations studied for this article, full risk assessments are necessary for potentially relevant MOAs involving cytotoxicity and cell proliferation in animals and humans (Case Study 6, chloroform) and formation of urinary-tract calculi (Case Study 7, melamine). In other circumstances, when data-based findings for the chemical and endpoint combination studied indicate that the tumor-related animal MOA is unlikely to have a human counterpart, there is little reason to continue the risk assessment for that combination. Similarly, when qualitative considerations identify MOAs specific to the test species or quantitative considerations indicate that the animal MOA is unlikely to occur in humans, such hazard findings are generally conclusive and further risk assessment is not necessary for the endpoint—MOA combination under study. Case examples include a tumor-related protein specific to test animals (Case Study 3, d-limonene), the tumor consequences of hormone suppression typical of laboratory animals but not humans (Case Study 4, atrazine), and chemical-related enhanced hormone clearance rates in animals relative to humans (Case Study 5, phenobarbital). The human relevance analysis is highly specific for the chemical-MOA-tissue-endpoint combination under analysis in any particular case: different tissues, different endpoints, or alternative MOAs for a given chemical may result in different human relevance findings. By providing a systematic approach to using MOA data, the HRF offers a new tool for the scientific community's overall effort to enhance the predictive power, reliability and transparency of cancer risk assessment. The attack on the World Trade Center occurred less than a quarter of a mile from Pace University in Lower Manhattan. This paper represents a compilation of the recollection of the events of September 11, 2001, and its aftermath from the four senior staff members of the Counseling Center. Our purpose is to share our experiences and perspectives of these events from the point of view of the closest university to the attack. We will also share some of the ways in which we coped as a staff, how we helped the larger community to cope, and some of the lessons we learned from both a professional and personal point of view. There is increasing evidence that the pathogenesis and progression of many forms of pulmonary vasculopathy are related to abnormalities in endothelial mediators, including endothelin-1 (ET-1) and nitric oxide (NO). Using a rat model of chronic unilateral pulmonary artery ligation, we investigated the role of ET-1 and NO in postobstructive pulmonary vasculopathy (POPV). Eight months after a left thoracotomy with either left main pulmonary artery ligation (ligated group) or no ligation (sham group), rat lungs, including those contralateral to the ligation (hyperperfused group), were fixed and mounted for histologic sectioning. Morphometric measurements were carried out by computer-assisted image analysis and immunohistochemical staining was performed using specific antibodies against ET-1, ET[sub A], and EB[sub B] receptors, and endothelial NO synthase (eNOS). Compared to sham lungs, the ligated lungs showed (1) an increase in muscular, adventitial, and intimal thickness of pulmonary artery; (2) increase in external diameter of the bronchial artery (39.8+/-2.2 mum vs.16.8+/-0.9 mum in sham group; P < .005) and number of bronchial arteries per bronchiole (3.21+/-mu0.26vs.1.86+/-mu0.21 in sham group; P < .001); and (3) increase in the intensity of eNOS and ET[sub A, B] receptor immunoreactivity. No morphometric or immunohistochemical differences were observed between the hyperperfused and sham lungs. These ndings suggest that increased synthesis of endothelial NO may serve as a compensatory mediator in ET-1 mediated vascular remodeling. [ABSTRACT FROM AUTHOR]

We present a geographical analysis of the first 22 months of the Department of Defense's human immunodeficiency virus (HIV) screening program for military applicants. This analysis concerns four HIV epidemic loci in the United States: New York City; Miami, Florida: Houston, Texas; and San Francisco, California. The cartographic analysis, using a novel smoothing algorithm to stabilize county-based rates, highlights the diffusion of the HIV epidemic from areas with relatively high prevalence to areas with relatively low prevalence of antibody to HIV. While there are particularities specific to each region, the diffusion patterns are remarkably consistent: areas closer to an endemic focus experience a slight decline over time, while more distant areas increase. The diffusion process depicted in map sequences demonstrates expansion diffusion in all four areas, while some provide a clear indication of a mixture of expansion and hierarchical processes. A previous analysis of these data for a fifteen-month period had concluded there was no evidence of a geographic trend in the data. Those conclusions were based on all ages pooled and used multi-state geographic units, making it unlikely that trends in prevalence, which varied by proximity to HIV epicenters, would have been detected. In view of the temporal crossover between the endemic and nonendemic areas' prevalence rates observed in Florida, Texas and California, any spatial analysis of these data using statewide summary rates will obscure the spatial-temporal trends. These data provide compelling evidence for the existence of sub-state level epidemics growing spatially and temporally into areas some miles from the original epicenters. We hope these results will encourage other public health personnel to undertake detailed surveillance of the AIDS and HIV infection epidemics at a unit of analysis small enough to permit quantifying spatial diffusion of this epidemic. Purpose: To describe the location of the zygomatico-orbital foramen on the inferolateral orbital wall. Methods: This anatomic study examined 28 orbits of 14 dry human adult skulls. The zygomatico-orbital foramen was identified by passing a thin wire from the zygomatico-facial foramen to its orbital aspect and a thin flexible ruler was used to measure 1) the distance perpendicular to the closest point on the inferior orbital rim, 2) the distance from the inferior orbital fissure, and 3) the distance from the area used for retrobulbar injections. Results: The mean distance from the zygomatico-orbital foramen to the closest point on the inferior orbital rim was 4.7 mm (range from 1 to 7 mm). The mean distance from the inferior orbital fissure was 14.9 mm (range from 10 to 18 mm). The mean distance from the area of retrobulbar injection was 6.0 mm (range from 3 to 10 mm). Conclusions: The location of the zygomatico-orbital foramen within the inferolateral orbit is quite variable. This is the first study to attempt to quantify its proximity to the site of retrobulbar injection. We conclude that it is an important anatomical structure to consider when giving retrobulbar anesthesia, especially given the variability in technique among ophthalmologists. [ABSTRACT FROM AUTHOR]

The homeodomain (HD) transcription factors are a structurally conserved family of proteins that, through networks of interactions with other nuclear proteins, control patterns of gene expression during development. For example, the network interactions of the pituitary-specific HD protein Pit-1 control the development of anterior pituitary cells and regulate the expression of the hormone products in the adult cells. Inactivating mutations in Pit-1 disrupt these processes, giving rise to the syndrome of combined pituitary hormone deficiency. Pit-1 interacts with CCAAT/enhancer-binding protein alpha (C/EBPα) to regulate prolactin transcription. Here, we used the combination of biochemical analysis and live-cell microscopy to show that two different point mutations in Pit-1, which disrupted distinct activities, affected the dynamic interactions between Pit-1 and C/EBPα in different ways. The results showed that the first α-helix of the POU-S domain is critical for the assembly of Pit-1 with C/EBPα, and they showed that DNA-binding activity conferred by the HD is critical for the final intranuclear positioning of the metastable complex. This likely reflects more general mechanisms that govern cell-type-specific transcriptional control, and the results from the analysis of the point mutations could indicate an important link between the mislocalization of transcriptional complexes and disease processes.
