A multiphase microwave drying (MMD) was proposed to produce dried Chinese yam cubes with good quality comparable to freeze-dried products. The effects of different microwave power loading schemes on the conversion point (water phase in samples changes from solid-to-gas to liquid-to-gas) of MMD were assessed, and the macroscopic physical properties and microstructure characteristics of dried yams were investigated. Results showed that with an increase of microwave power, the time required to reach the conversion point was reduced. And the moisture content at the conversion point as affected by microwave power loading schemes had vital influence on sample quality. When it was 0.378 g·g−1, the properties of the yam cubes resembled those of freeze-dried samples in terms of rehydration, texture, and microstructure. Furthermore, a fractal theory-based microstructure characterization can accurately reflect texture characteristics. Gastrodin, one of the major components extracted from the Chinese herb Gastrodia elata Bl., has been widely used as an anticonvulsant, sedative, analgesic and hypotensive. In our study, we aimed to investigate the effects and possible mechanisms of gastrodin on vascular KATP channels. Tension experiments were used on rat mesenteric artery rings without an endothelium. Patch clamp experiments were executed to investigate the influences of gastrodin on the membrane current in mesenteric artery smooth muscle cells. Gastrodin induced vasorelaxation in a concentration dependent manner when rat mesenteric artery rings were pre-contracted with Phenylephrine. The vasorelaxation effect was partially diminished by pre-treating with a KATP channel inhibitor, or a PKA inhibitor. With whole-cell patch-clamp recording techniques, we found that gastrodin is a activator of KATP in rat mesenteric artery smooth muscle cells, and this effect was eliminate by pre-treating with H89or PKI, PKA inhibitor. In addition, when rat vascular smooth muscle cells were treated with 100 μM gastrodin for 24 h, maximum KATP current density increased by 28.1%. The results indicate that gastrodin exerts vasorelaxation effect through activation of PKA and subsequent opening of smooth muscle KATP channels. [ABSTRACT FROM AUTHOR]

Magnetic recyclable bactericidal nanocomposites (Fe3O4@PDMC) were well-designed and prepared by coating of Fe3O4nanoparticles with quaternarized N-halamine polymers via the free radical polymerization process. In which, 5,5-dimethylhydantoinyl-(3-ethyl-methacrylamine)propyl dimethylammonium bromide (DEMPA), a new monomeric N-halamine precursor, was used as a coating material as well as the dual-functional bactericidal agent. The developed Fe3O4@PDMC nanocomposites exhibited suitable size and super-paramagnetic responsibility. The antibacterial results showed that the Fe3O4@PDMC nanocomposites had excellent biocidal abilities againstStaphylococcus aureus(gram-positive) andEscherichia coli(gram-negative). Furthermore, the TTC (Triphenyl Tetrazolium Chloride)–dehydrogenase activity assay confirmed that the reductions of the bacteria were mainly attributed to powerful biocidal effects of the coating polymer instead of bacteria capture by the cationic surface. Interestingly, due to the magnetic responsive performance of Fe3O4, the as-prepared Fe3O4@PDMC nanocomposites can be recycled by a magnet and reused for antibacterium through the quenching/rechlorination procedure. All the results presented in this study show that the proposed Fe3O4@PDMC nanocomposites could be a competitive candidate for water purification systems and household sanitation. Angiogenesis plays a critical role in the progression of cardiovascular disease, retinal ischemia, or tumorigenesis. The imbalance of endothelial cell proliferation and apoptosis disturbs the establishment of the vasculogenesis, which is affected by several arachidonic acid metabolites. 15-Hydroxyeicosatetraenoic acid (15-HETE) is one of the metabolites. However, the underlying mechanisms of angiogenesis induced by 15-HETE in human umbilical vascular endothelial cells (HUVECs) are still poorly understood. Since extracellular signal-regulated kinase (ERK) is a critical regulator of cell proliferation, there may be a crosstalk between 15-HETE-regulating angiogenic process and ERK-proliferative effect in HUVECs. To test this hypothesis, we study the effect of 15-HETE on cell proliferation, angiogenesis, and apoptosis using cell viability measurement, cell cycle analysis, western blot, scratch–wound, tube formation assay, and nuclear morphology determination. We found that 15-HETE promoted HUVEC angiogenesis, which were mediated by ERK. Moreover, 15-HETE-induced proliferation and cell cycle transition from the G0/G1 phase to the G2/M + S phase. All these effects were reversed after blocking ERK with PD98059 (an ERK inhibitor). In addition, HUVEC apoptosis was relieved by 15-HETE through the ERK pathway. Thus, ERK is necessary for the effects of 15-HETE in the regulation of HUVEC angiogenesis, which may be a novel potential target for the treatment of angiogenesis-related diseases. Stromal cell–derived factor 1 (CXCL12) and its receptor CXC chemokine receptor 4 (CXCR4) are known to modulate hypoxia-induced pulmonary hypertension (PH) and vascular remodeling by mobilization and recruitment of progenitor cells to the pulmonary vasculature. However, little is known about CXCL12/CXCR4 regulating proliferation and cell cycle progression of pulmonary arterial smooth muscle cells (PASMCs). To determine whether CXCL12/CXCR4 regulates PASMC proliferation and the cell cycle, immunohistochemistry, Western blot, bromodeoxyuridine incorporation and cell cycle analysis were preformed in this study. Our results showed that CXCR4 was induced by hypoxia in pulmonary arteries and PASMCs of rats. Hypoxia-increased cell viability, DNA synthesis and proliferating cell nuclear antigen expression were blocked by administration of CXCR4 antagonist AMD3100, silencing CXCR4 or CXCL12. Furthermore, inhibition of CXCL12/CXCR4 suppressed cell cycle progression, decreased the number of cells in S+G2/M phase and attenuated the expression of proteins that regulate the cell cycle progression at these phases. In addition, PI3K/Akt signaling mediated CXCL12/CXCR4 regulating proliferation and cell cycle progression in PASMCs. Thus, these results indicate that blockade of CXCL12/CXCR4 inhibited PASMC proliferation and cell cycle progression in hypoxia-induced PH via PI3K/Akt signaling pathway. [ABSTRACT FROM PUBLISHER]

In China, one of the major problems in upgrading rural health services is the difficulty of communicating between the rural and urban areas. Enabling local agencies to access the Internet in resource-poor areas can provide an efficient means of diffusing current training and information and will have far-reaching policy implications. To test the feasibility of using the Internet to deliver needed health information to the countryside, the UCLA School of Public Health and the Institute of Health Studies of Kunming Medical College (IHS-KMC) collaborated in an experimental website project to improve the quality of reproductive health services to promote women's health in three rural counties of Yunnan. The project involved the county government and the Bureau of Public Health, the Bureau of Family Planning; the Bureau of Education, Women's Federation, and the Maternal and Child Health Station targeting village health workers and teachers; women's cadres. Three counties, matched on socioeconomic status, participated in the study and were randomized to receive three programs. Nanhua County received computer skill training and logistic support including a planning workshop for information diffusion. Mouding County received computers only. Dayao, the control county, did not receive the full program until the conclusion of the project. The study demonstrated that the use of a website to disseminate health information in remote rural areas is not only feasible but that it also will be enthusiastically adopted by local health workers and interested parties. Moreover, the knowledge was diffused from the primary population of village doctors, family planning workers, women's cadres, and teachers to the secondary population of villagers and students.
