Natural products, especially fungal secondary metabolites, have been served as valuable sources of drug leads in pharmaceutical industry. Medicinal plants-associated endophytic fungi possess a well-developed secondary metabolism. In this study, chemical investigation on Penicillium ochrochloron YT2022-65, an endophytic fungus associated with Lonicera Japonica, led to the isolation of six structurally diversified indole alkaloids, including a new one, namely peniochroloid A (1), as well as five previously reported alkaloids, flavonoid B (2), brocaeloid C (3), isoroquefortine C (4), roquefortine C (5), and dihydrocarneamide A (6). Their structures, including the absolute configuration of 1, were determined by a combined analysis of HRESIMS, NMR spectroscopic data, and calculation of the optical rotation. Their cytotoxicity against A549, HepG2, MCF-7, and THP-1 cell lines were evaluated in vitro. The new compound 1 was found to possess considerable cytotoxicity against MCF-7 and THP-1 cell lines with IC50 values of 10.2 and 11.0 μM, respectively. [ABSTRACT FROM AUTHOR]

Marine fungi are promising sources of bioactive natural products. The harsh marine conditions favour the production of natural products with unique structures and functions. The different classes of bioactive metabolites produced by these marine fungi can exhibit cytotoxic, apoptotic, anti-proliferative, antiangiogenic, and autophagy inducing effects on a plethora of cancer cell lines. This review, based on research articles that have been published from 2002 to 2023, provides a concise overview of the anticancer properties of metabolites from marine Aspergillus fungal species. A total of 204 papers are reviewed and 208 most active cytotoxic molecules are reported from Aspergillus. The source as well as the growth medium utilised for the production of cytotoxic metabolites are listed. The mechanism of action of some compounds, which could be used as potential drugs, is also reported. These fungi, under optimal growth conditions, have immense potential as anticancer agents, produce novel metabolites with specific structures that can kill a panel of human cancer cells. However, there is a dire need for more clinical trials and understanding of the mechanisms of action of pharmacologically active constituent. Maize grain samples collected from 129 small-scale farmers' stores in southern and southwestern Ethiopia were analysed by LC-MS/MS for a total of 218 mycotoxins and other fungal metabolites of which 15% were regulated mycotoxins. Mycotoxins produced by Penicillium, Aspergillus, and Fusarium accounted for 31%, 17%, and 12% of the metabolites, respectively. Most of the current samples were contaminated by masked and/or emerging mycotoxins with moniliformin being the most prevalent one, contaminating 93% of the samples. Each sample was co-contaminated by 3 to 114 mycotoxins/fungal metabolites. Zearalenone, fumonisin B1, and deoxynivalenol were the dominant mycotoxins, occurring in 78%, 61%, and 55% of the samples with mean concentrations of 243, 429, and 530 µg/kg, respectively. Dothiorella species are fungal plant pathogens associated with Botryosphaeria dieback of grapevine. Symptoms caused by these fungi on grapevines suggest possible implication of phytotoxic metabolites in the infection mechanisms. However, few studies were conducted to investigate the secondary metabolism of these fungi. Mycotoxins are secondary fungal metabolites harmful to humans and animals. Patulin (PAT) is a toxin found in different food products but especially in apples and their derivative products. The most common fungi producers of this compound are Aspergillus clavatus and Penicillium expansum. The production of patulin, as other mycotoxins, can be impacted by diverse phenomena such as water and nutrient availability, UV exposure, and the presence of antagonistic organisms. Consequently, gaining a comprehensive understanding of climate and environmental conditions is a crucial step in combating patulin contamination. In this study, moulds were isolated from 40 apple samples collected from seven locations across Hungary: Csenger, Damak, Pallag, Lövőpetri, Nagykálló, and Újfehértó. A total of 183 moulds were morphologically identified, with 67 isolates belonging to the Alternaria, 45 to the Aspergillus, and 13 to the Penicillium groups. The location possessed a higher influence than farming method on the distribution of mould genera. Despite the requirement of higher temperature, Aspergillus species dominated only for the region of Újfehértó with approximately 50% of the isolates belonging to the genus. Four of the seven locations assessed: Csenger, Debrecen-Pallag, Nyírtass and Nagykálló, were dominated by Alternaria species. All isolates belonging to the genera Aspergillus and Penicillium were tested for the presence of the isoepoxidone dehydrogenase (idh) gene, a key player in the patulin metabolic pathway. To guarantee patulin production, this ability was confirmed with TLC assays. The only Aspergillus strain that presented a positive result was the strain Aspergillus clavatus B9/6, originated from the apple cultivar Golden Reinders grown in Debrecen-Pallag by integrated farming. Of the Penicillium isolates only one strain, B10/6, presented a band of the right size (500-600 bp) for the idh gene. Further sequencing of the ITS gene showed that this strain should be classified as Talaromyces pinophilus. Challenges exist worldwide for the storage of animal feed, though year-long feeding of livestock is especially difficult in northern climates, including Canada. Usage of silage allows for crops to be harvested at the optimal nutritional value and stored for extended periods in customized storage systems. This case study investigated various feed management practices typical of small beef cattle feedlot farms in Southwestern Ontario, Canada. Crop samples were collected monthly and analyzed for mycotoxins and fungal secondary metabolites by liquid chromatography mass spectrometry (LC-MS/MS). Stored barley contained modest concentrations of deoxynivalenol and related compounds, as well as near trace amounts of fumonisin. Stored hay contained low concentrations of deoxynivalenol and beauvericin from the field. Hot spots of the Penicillium metabolite citrinin were detected after 7–8 months of storage in barley (39.2 ng g−1). Citrinin was also detected after 7–8 months in hay (16.3–17.5 ng g−1) along with the appearance of trace amounts of other storage toxins, including penitrem A (8.84 ng g−1), griseofulvin (34.8 ng g−1) and sterigmatocystin (14.1–261 ng g−1). This case study was a unique opportunity to use 'citizen science' to observe the onset of postharvest fungi and associated mycotoxins in various storage conditions allowing for the farmer to make necessary modifications before major problems begin. An improved understanding of the storage conditions that foster fungal growth and mycotoxin production in this working farm led to better agronomic practices, ultimately improving feed quality, livestock health, and profitability. Since the first years of history, microbial fermentation products such as bread, wine, yogurt and vinegar have always been noteworthy regarding their nutritional and health effects. Similarly, mushrooms have been a valuable food product in point of both nutrition and medicine due to their rich chemical components. Alternatively, filamentous fungi, which can be easier to produce, play an active role in the synthesis of some bioactive compounds, which are also important for health, as well as being rich in protein content. Therefore, this review presents some important bioactive compounds (bioactive peptides, chitin/chitosan, β-glucan, gamma-aminobutyric acid, L-carnitine, ergosterol and fructooligosaccharides) synthesized by fungal strains and their health benefits. In addition, potential probiotic- and prebiotic fungi were researched to determine their effects on gut microbiota. The current uses of fungal based bioactive compounds for cancer treatment were also discussed. The use of fungal strains in the food industry, especially to develop innovative food production, has been seen as promising microorganisms in obtaining healthy and nutritious food. Fungal-based bioactive compounds have various health benefits. Prebiotic fungi play an active role in the regulation of gut microbiota. Fungi-mediated synthesis of Gold nanoparticles (AuNPs) has advantages in: high efficiency, low energy consumption, no need for extra capping and stabilizing agents, simple operation, and easy isolation and purification. Many fungi have been found to synthesize AuNPs inside cells or outside cells, providing different composition and properties of particles when different fungi species or reaction conditions are used. This is good to produce AuNPs with different properties, but may cause challenges to precisely control the particle shape, size, and activities. Besides, low concentrations of substrate and fungal biomass are needed to synthesize small-size particles, limiting the yield of AuNPs in a large scale. To find clues for the development methods to solve these challenges, the reported mechanisms of the fungi-mediated synthesis of AuNPs were summarized. The mechanisms of intracellular AuNPs synthesis are dependent on gold ions absorption by the fungal cell wall via proteins, polysaccharides, or electric absorption, and the reduction of gold ions via enzymes, proteins, and other cytoplasmic redox mediators in the cytoplasm or cell wall. The extracellular synthesis of AuNPs is mainly due to the metabolites outside fungal cells, including proteins, peptides, enzymes, and phenolic metabolites. These mechanisms cause the great diversity of the produced AuNPs in functional groups, element composition, shapes, sizes, and properties. Many methods have been developed to improve the synthesis efficiency by changing: chloroauric acid concentrations, reaction temperature, pH, fungal mass, and reaction time. The malting industry faces a dilemma in which barley, initially containing low deoxynivalenol levels, occasionally exhibits an unexpected increase in deoxynivalenol levels during malting. In the current study, we investigated mycotoxin accumulation and hyphal localization in single kernels from Fusarium head blight infected barley and malt samples which showed an aberrant behavior of deoxynivalenol increases during malting. While deoxynivalenol levels in these bulk barley samples already ranged from <0.20 to 1.27 µg/g, they further increased on average by approximately 500% after malting, along with Fusarium growth. In addition, deoxynivalenol-3-glucoside and 3-acetyl deoxynivalenol levels increased from <0.20 µg/g to the mean levels of 2.47 µg/g and 1.03 µg/g in the bulk malt, respectively. In the investigation of single kernels, results showed that only 7% of barley single kernels (n = 385) had deoxynivalenol >1.00 μg/g, with the maximum level of 35.97 μg/g. However, 31% of malt single kernels contained deoxynivalenol >1.00 μg/g and up to 255.43 μg/g. Deoxynivalenol-3-glucoside and 3-acetyl deoxynivalenol were found in 40% of malt single kernels. Fungal hyphae were observed in the aleurone layer and embryo of barley kernels with deoxynivalenol >10.00 μg/g, in addition to the husk and vascular bundles of kernels with deoxynivalenol <1.00 μg/g. Hyphae spread largely in these tissues following malting, and even into the endosperm and pericarp cavities of extremely high deoxynivalenol kernels (i.e., >100.00 μg/g). [ABSTRACT FROM AUTHOR]

Mycotoxins are produced primarily as secondary fungal metabolites. Mycotoxins are toxic in nature and naturally produced by various species of fungi, which usually contaminate food and feed ingredients. The growth of these harmful fungi depends on several environmental factors, such as pH, humidity, and temperature; therefore, the mycotoxin distribution also varies among global geographical areas. Various rules and regulations regarding mycotoxins are imposed by the government bodies of each country, which are responsible for addressing global food and health security concerns. Despite this legislation, the incidence of mycotoxin contamination is continuously increasing. In this review, we discuss the geographical regulatory guidelines and recommendations that are implemented around the world to control mycotoxin contamination of food and feed products. Researchers and inventors from various parts of the world have reported several innovations for controlling mycotoxin-associated health consequences. Unfortunately, most of these techniques are restricted to laboratory scales and cannot reach users. Consequently, to date, no single device has been commercialized that can detect all mycotoxins that are naturally available in the environment. Therefore, in this study, we describe severe health hazards that are associated with mycotoxin exposure, their molecular signaling pathways and processes of toxicity, and their genotoxic and cytotoxic effects toward humans and animals. We also discuss recent developments in the construction of a sensitive and specific device that effectively implements mycotoxin identification and detection methods. In addition, our study comprehensively examines the recent advancements in the field for mitigating the health consequences and links them with the molecular and signaling pathways that are activated upon mycotoxin exposure. Methicillin-resistant Staphylococcus aureus (MRSA) increases its antibiotic resistance by forming biofilms. Natural products (NP) or specialized metabolites have demonstrated their ability to decrease the virulence and pathogenesis of MRSA infections by inhibiting biofilm formation. The present study evaluated the antimicrobial and antibiofilm potential against MRSA of a small library of fungal NP isolated from Mexican biodiversity. The most potent antibacterial activity was observed for myrotecisin B, epiequisetin, equisetin, stachybotrolide acetate, monorden A, zearalenone, fuscin, and fusarubin. On the other hand, epifiscalin C, fiscalin C, dimethylglyotoxin, aspernolide B, and butyrolactones I and IV inhibited the biofilm formation without decreasing bacterial growth. To determine the putative mechanism of action of these compounds, docking analyses were performed against SarA and AgrA proteins, targets known to regulate biofilm production in MRSA. Residual amounts of pharmaceutical compounds (PhCs) and by-products are continually released into surface water with effluents from conventional wastewater treatment plants (WWTPs). This study evaluated the ability of fungal isolate to remove selected PhCs [carbamazepine (CBZ), diclofenac (DCF) and ibuprofen (IBP)] from wastewater. The fungus used was Rhizopus sp. which was isolated from tuberous roots of cassava (Manihot esculenta). The isolate exhibited an important removal efficiency up to 100% and this was linked to ligninolytic enzymatic activity for lignin peroxidase (15.29 ± 2.69U/L) and manganese peroxidase (85.22 ± 4.26U/L), except laccase. This activity was optimum on day 9 of treatment. PhC metabolites were identified during the experiment revealing the existence of a biotransformation process catalysed by the isolated fungus. The disappearance of PhCs was attributed to their biosorption and biotransformation. However, it was not possible to establish a relationship between the ligninolytic enzymatic activity and the removal efficiency, which leads to the conclusion that there are other fungal metabolites which also play an important role in the biotransformation and biodegradation of the selected PhCs. Food and feed contamination by nonlegislated mycotoxins beauvericin (BEA) and enniatin B (ENB) is a worldwide health concern in the present. The principal objective of this work is to assess some of the existing protocols to discover the single nucleotide variants (SNVs) in transcriptomic data obtained by RNA-seq from Jurkat cells in vitro samples individually exposed to BEA and ENB at three concentration levels (1.5, 3 and 5 µM). Moreover, previous transcriptomic results will be compared with new findings obtained using a different protocol. SNVs rs201003509 in BEA exposed cells and the rs36045790 in ENB were found in the differentially expressed genes in all doses compared to controls by means of the Genome Analysis Toolkit (GATK) Best Practices workflow. SNV-RNA-seq complementary pipeline did not show any SNV. Concerning gene expression, discrepant results were found for 1.5 µM BEA exposed cells compared with previous findings. However, 354 overlapped differentially expressed genes (DEGs) were identified in the three ENB concentrations used, with 147 matches with respect to the 245 DEGs found in the previous results. In conclusion, the two discovery SNVs protocols based on variant calling from RNA-seq used in this work displayed very different results and there were SNVs found manually not identified by any pipeline. Additionally, the new gene expression analysis reported comparable but non identical DEGs to the previous transcriptomic results obtained from these RNA-seq data. Mitidjospirone, a new spiridioxynaphthalene, was isolated from the mycelial extract of a strain of Lasiodiplodia mitidjana, a recently described species belonging to the family Botryosphaeriaceae. Its structure was elucidated by extensive spectroscopic analysis and the absolute configuration was determined by electronic circular dichroism (ECD) experiment. An alternative to controlling weeds resistant to conventional herbicides is the isolation of new active principles. Fungi can produce phytotoxic metabolites that may be used in the development of new herbicides. The objectives of this study were: (1) isolate, select, and identify a fungus producer of phytotoxic metabolites and (2) optimize the culture conditions of this fungus in a low-cost culture medium, with the aim of increasing the phytotoxic effects of their metabolites in weeds and commercial plants. Fungi were isolated from the leaves of Conyza sp. with disease symptoms and selected according to the production of phytotoxic metabolites in solid and submerged fermentation in a low-cost culture medium. A Plackett–Burman Design and Central Composite Rotational Design were used to optimize the conditions of temperature, agitation, pH, and concentrations of glucose and yeast extract in submerged fermentation. The phytotoxic metabolites produced under optimal conditions were tested on 10 commercial plants and weeds that are difficult to control. Of the nine fungi isolated, Mycoleptodiscus indicus UFSM54 produced higher leaf lesions. The production of phytotoxic metabolites was optimized when the fungus was cultivated at 35°C, 50 rpm, and 1.5 g L−1 of glucose in submerged fermentation. The metabolites of M. indicus caused severe phytotoxic effects on germination and seedling growth, and enhanced lesion development on detached plant leaves. Luminescent nanoparticles synthesized via bio-based protocols that generate nanoparticles having different chemical compositions along with other functionalities (size and morphology) have received huge attention. We have focused our research on gold sulfide nanoparticles (Au2S NPs) and have biosynthesized these NPs using the fungus Humicola sp. The nanoparticles were characterized by Transmission Electron Microscopy, which showed spherical morphology of Au2S. UV–Visible-NIR spectrophotometry, luminescence spectrophotometry, Selected Area Electron Diffraction, Energy Dispersive Analysis of X-rays, and X-ray diffraction were performed. FTIR confirmed that the fungal metabolites including biomolecules secreted in the reaction medium are primarily responsible for nanoparticle synthesis and stabilization. The fungus reduced the precursor solution (HAuCl4 and Na2SO3) and at the same time capped them with secreted biomolecules. The anti-leishmanial activity of Au2S NPs was determined against L. donovani promastigote (Ag83 strain). Au2S NPs displayed less cytotoxicity towards both normal and cancer (Daudi, ZR-75-1) cell lines. Hemocompatibility was determined via hemolysis assays. This novel fungal-based system demonstrates an economical and environmentally benign process for biosynthesis of Au2S nanoparticles which may find application in bioimaging and labeling studies. Mycotoxins are health-threatening fungal metabolites that have been found in several foods around the world. Although agricultural, transportation, and storage management strategies have been employed to reduce the production of mycotoxins, they are not effective in eliminating mycotoxins. In this context, the application of ozone has emerged for the degradation of mycotoxins. Ozone has a strong oxidation rate and generates more free radicals, which can counter the functional groups of the mycotoxin by changing their molecular structures and forming products having lower molecular weight reduced number of double bonds, and reduced toxicity. Research indicates that ozone could be able to destroy mycotoxins without leaving any residues in the commodities. The ozone processing parameters, surface and nutritional properties of food, and fungal species are the primary determinants affecting the processing efficacy. The occurrence of secondary metabolites and co-contaminants in dairy cow feed samples (n = 115), concentrate, roughage, and mixed feed, collected from Ratchaburi and Kanjanaburi provinces, Thailand, between August 2018 and March 2019 were investigated using LC-MS/MS based multi-toxin method. A total of 113 metabolites were found in the samples. Fungal metabolites were the predominant compounds, followed by plant metabolites. Among major mycotoxins, zearalenone and fumonisins were most frequently detected in concentrate and mixed feed samples, while deoxynivalenol and aflatoxin B1 were found at the frequency lower than 50%. Other metabolites, produced by Aspergillus, Fusarium, Penicillium, and Alternaria species, occurred in the samples. Flavoglaucin, 3-nitropropionic acid, averufin, and sterigmatocystin were the most prevalent Aspergillus metabolites. Common Fusarium metabolites occurring in the samples included moniliformin, beauvericin, and enniatins. For Penicillium metabolites, mycophenolic acid, questiomycin A, quinolactacin A, oxaline, citrinin, and dihydrocitrinone were frequently detected. The toxic Alternaria metabolites, alternariol, and alternariol monomethyl ether showed the high incidence in the samples. Plant metabolites were commonly found, mainly cyanogenic compounds and isoflavones, from cassava and soybean meal used as feed ingredients. Overall, 96.6% of feed samples contained at least two metabolites, in a range from 2 to 69. According to co-contamination of mycotoxins found in feed samples, zearalenone were mostly found in combination with fumonisin B1, deoxynivalenol, and aflatoxin B1. Fumonisin B1 co-occurred with aflatoxin B1 and deoxynivalenol. The mixtures of deoxynivalenol and aflatoxin B1, and of zearalenone, fumonisin B1 and deoxynivalenol were also found. Due to known individual toxicity of fungal and plant metabolites and possible additive or synergistic toxic effects of multi-mycotoxins, the occurrence of these metabolites and co-contaminants should be monitored continuously to ensure food safety through the dairy supply chain. A total of 117 fungal metabolites were detected in grains collected in Gunja-G (flooded village) and Gornji Stupnik-GS (control village), located in the Zagreb County, Croatia. Major mycotoxins and derivatives (17), ergot alkaloids (14), Fusarium (23), Aspergillus (18), Penicillium (18), Alternaria (7) and other fungal and unspecific metabolites (20) were found. A higher number of metabolites co-occurred per sample in grains from G (115) than in GS (91). Regulated mycotoxins were below maximum limits except fumonisins B1,2 in 15–20% of grains and aflatoxin B1.Fusarium metabolites contaminated more than 50% of grains at both locations. Besides FB1,2, bikaverin, aurofusarin, culmorin and 15-hidroxyculmorin were detected at relatively high concentrations. Ergot alkaloids were detected at 2–18 times higher concentrations in grains from G as compared to GS. Majority of Aspergillus mycotoxins were present at a low frequency (5–15%). A polyphasic approach consisting of physical, chemical and molecular characterization was applied to three Alternaria species, A. alternata, A. chlamydospora and A. solani, to characterize and identify isolates producing and non-producing silver nanoparticles (AgNPs). Based on physical characters, the synthesized AgNPs were characterized by transmission electron microscopy and formation of spherical AgNPs sized 5 nm to 60 nm. Scanning electron microscopy (SEM) of the AgNPs did not show a uniform surface; the shape was irregular, like a coral reef. Energy Dispersive Spectroscopy (EDS) showed the presence of Ag element. Fourier transform-infrared spectroscopy (FTIR) confirmed the conversion of Ag ions to AgNPs by fungal metabolites. A. alternata PNU75 non-producing AgNPs showed negative results when characterized by physical devices. Chemical characterization involved the screening of three Alternaria species for fatty acids (FAs) with similar FAs composition while different in terms of relative concentration. The principal FAs were palmitic, stearic, oleic acid and linoleic acid, which comprised 90% or more of the total FAs composition of these species. Molecularly, the fungal protein pattern of the three Alternaria species showed changes against the control by finding 6 proteins associated with AgNPs-producing species. Inter simple sequence repeats (ISSR) analysis using ten primers indicated a high level of genetic variability of three Alternaria species producing and non-producing AgNPs. The results also indicated that the AgNPs synthesized by A. alternata at the concentration 100 mg/L gave the highest inhibition zone against Aspegillus flavus and Aaltenaria alternata, where the percentages of reduction were 77.1% and 87.6% respectively. The strains of actinobacteria are unique as they lie between true fungi and bacteria and several of them were reported as endophytic actinobacteria as they were isolated from the inner regions of various plant parts and will enhance uptake of nutrients and improve defense against pathogens. Literature and scientific communications reported the relationship between the endophytes and plants, most of them concluded the association as commensalism. Remarkably, bioactive compounds from endophytic Streptomyces sp. were confirmed with various applications. A retrospective consolidation on the endophytic Streptomyces sp. and their metabolite application in day to day life is presented here. It was deduced that this group of the organism are a source for a wide range of bioactive compounds including anticancer agents, immune suppressor, plant growth promoters, anti-inflammatory agents, anti-tumor agents, enzymes and antimicrobial substances. These antimicrobial metabolites show broad-spectrum activity and are effective against bacteria and fungi. There is scarce data on the mycotoxin profile in retailed fruit juices in Nigeria. Thirty-five industrially-processed fruit juice samples randomly purchased from retailers in Ogun state, Nigeria, were analysed for the presence of > 650 toxic fungal and plant metabolites using a liquid chromatography tandem mass spectrometric method. Only 18 metabolites, including 3-nitropropionic acid, alternariol methylether and emodin, but excluding citrinin, fumonisin B2, ochratoxin A and patulin, were detected in trace levels in at least one juice sample. Amygdalin, a plant cyanogen, was quantified (2.05–359 µg/L) in 40% of the samples. Although the levels of mycotoxins and toxic plant metabolites found in the juice may be relatively low, daily consumption of juices containing such low levels may contribute to dietary exposures to these natural chemical contaminants in consumers. Isoshamixanthone (1), a new stereoisomeric pyrano xanthone together with the previously known fungal metabolites, epiisoshamixanthone (2), sterigmatocystin (3), arugosin C (4), norlichexanthone (5), diorcinol (6), ergosterol and methyllinoleate, were obtained from the endophytic fungal strain Aspergillus sp. ASCLA isolated from leaf tissues of the medicinal plant Callistemon subulatus. The chemical structure of the new xanthone (1) was elucidated by extensive 1D, 2D NMR, and ESI HR mass measurements, and by comparison with literature data. The constitutions and absolute configurations of 1 and epiisoshamixanthone (2) were additionally confirmed by X-ray crystallography. Compounds 1,2 were evaluated for their potential anticancer activity using the human cervix carcinoma cell line (KB-3-1). In our ongoing search for new secondary metabolites from fungal strains, one novel compound (1) and nine known compounds (2-10) were isolated from the EtOAc-soluble layer of the culture broth of Panus rudis. The culture broth of P. rudis was extracted in acetone and fractionated by solvent partition; column chromatography using silica gel, Sephadex LH-20, and Sephadex G-10; MPLC; and HPLC. The structures of isolated compounds were elucidated by one-and two-dimensional NMR and LC-ESI-mass measurements. One new compound, panepoxydiol (1), and nine known compounds, (E)-3-(3-hydroxy-3-methylbut-1-en-1-yl)-7-oxabicyclo[ 4.1.0]hept-3-ene-2,5-diol (2), isopanepoxydone (3), neopanepoxydone (4), panepoxydone (5), panepophenanthrin (6), 4-hydroxy-2,2-dimethyl-6-methoxychromane (7), 6-hydroxy-2,2-dimethyl-3-chromen (8), 2,2-dimethyl-6-methoxychroman-4-one (9), 3,4-dihydroxy-2,2-dimethyl-6-methoxychromane (10), were isolated from the culture broth of P. rudis. We investigated the antifungal activities of an endophytic fungus identified as Acaromyces ingoldii, found on a loblolly (Pinus taeda L.) pine bolt in Louisiana during routine laboratory microbial isolations. The specific objectives were to determine the inhibitory properties of A. ingoldii secondary metabolites (crude extract) on the mycelial growth of a brown-rot fungus Gloeophyllum trabeum and a white-rot fungus Trametes versicolor, and to determine the effective concentration of A. ingoldii crude preparation against the two decay fungi in vitro. Results show the crude preparation of A. ingoldii from liquid culture possesses significant mycelial growth inhibitory properties that are concentration dependent against the brown-rot and white-rot fungi evaluated. An increase in the concentration of A. ingoldii secondary metabolites significantly decreased the mycelial growth of both wood decay fungi. G. trabeum was more sensitive to the inhibitory effect of the secondary metabolites than T. versicolor. Identification of specific A. ingoldii secondary metabolites, and analysis of their efficacy/specificity warrants further study. Findings from this work may provide the first indication of useful roles for Acaromyces species in a forest environment, and perhaps a future potential in the development of biocontrol-based wood preservation systems. Infestation of food crops by fungi has led to serious economic losses in the international market. There is a paucity of information on the multi-mycotoxin profile of cashew nuts from different geographical locations in Nigeria and South Africa. This work aims at comparing the fungal metabolite profile of cashew nut from two African countries using HPLC-FLD and LC-MS/MS. The differences in distribution of the fungal metabolites across the two countries were also examined. A total of 75 (39; Nigeria, 36; South Africa) roasted cashew nuts were purchased from 14 different locations in Nigeria and 10 major supermarkets in South Africa. The samples were subjected to HPLC analysis for aflatoxin and zearalenone and LC-MS/MS analysis for multiple metabolite profiling. Total aflatoxins (0.03-0.77 µg/kg and 0.01-0.28 µg/kg) and zearalenone (123.2 and 788.6 µg/kg) were quantified in nut samples from South Africa and Nigeria, respectively. In contrast, LC-MS/MS analysis showed that none of the major fungi metabolites except for one (citrinin) was present in detectable amount in Nigerian cashew nuts (31.3 μg/kg). Other microbial metabolites present in the nuts were; Aspergillus metabolites (< LOD to 398 × 104 μg/kg and < LOD to 1760 μg/kg); flavoglaucin (5.4-177.4 μg/kg and 45.5-18,368 μg/kg), 7-hydroxykaurenolide (5.5-45.8 μg/kg and 3.5-33.7 μg/kg), chloramphenicol (0.3-4.5 μg/kg) for South African and Nigerian cashew nuts, respectively. The incidence rate of the fusarium toxins beauvericin and bikaverin were higher in Nigerian cashew nut (100% and 17%) than the South African nuts (8% and 8%), 7-Hydroxykaurenolide and chloramphenicol were found in all the cashew nut samples. A total of 15 unspecific metabolites (17-100%) were recorded in the nuts from both countries. Sixty traditional leafy vegetables, comprising of mutete (Hibiscus sabdariffa) (n = 20) and omboga (Cleome gynandra) (n = 40) were analysed for fungal, plant and bacterial metabolites using liquid-chromatography-tandem mass spectrometry. No European Union legislated mycotoxins were quantified and no vegetables contained levels above the FAO/WHO limit of 10 mg/kg for cyanogenic potential, suggesting comparative safety regarding regulated mycotoxins and cyanogenic glycosides. Quantified fungal metabolites included averufin and 3-Nitropropionic acid from Aspergillus flavus, beauvericin and equisetin from Fusarium, citrinin and curvularin from Penicillium and altertoxin −1 and tentoxin from Alternaria. Of the plant cyanogenic glycosides, linamarin was quantifiable in 65% of mutete at a maximum of 398 µg/kg but not in omboga, while lotaustralin was quantifiable in both omboga and mutete. Mycotoxins are secondary fungal metabolites that cause both acute and chronic disease in humans and animals. Grains are a common substrate for molds and the production of mycotoxins. This study compared mycotoxin concentrations between grain and grain-free commercial dog foods. In total, 60 samples of dry and wet dog foods produced by five major manufacturers within the US were purchased from pet food retailers in southern California. A standard mycotoxin panel was performed by a reference laboratory in Seattle using industry standard methodology for pet foods. Results of the study demonstrated measurable mycotoxin concentrations in dry dog foods containing grains but not in grain-free dry dog foods, or in wet foods either containing grains or grain-free. This study suggests that the risk of mycotoxin exposure is higher in dry dog foods containing grains. Context:Salvia miltiorrhiza Bunge (Labiatae) is a traditional Chinese herb. Endophytic fungi, which are biotic elicitors, can induce accumulation of secondary metabolites in their host plants. Objective: To analyze the interaction mechanism between S. miltiorrhiza and endophytic fungi. Materials and methods: Endophytic fungi U104 producing tanshinone IIA were isolated from the healthy disease-free tissue of root of S. miltiorrhiza by conventional methods. The endophytic fungus U104 of S. miltiorrhiza was co-cultured with the sterile seedlings of S. miltiorrhiza for 20 d (temp:day/night = 26 °C/18 °C, photoperiod:12/12 h, illuminance:2000 Lx). Transcriptome sequencing of S. miltiorrhiza seedlings after 20 d of co-cultivation was performed using the Illumina platform. Results: A total of 3713 differentially expressed genes (DEGs) were obtained. These different expression genes, such as STPII, LTP2, MYB transcription factors, CNGC, CDPK, Rboh, CaM, MAP2K1/MEK1, WRKY33, SGT1/SGT and Hsp90/htpG, showed that host S. miltiorrhiza had biological defence response in the initial stage of interaction. Under the induction of endophytic fungi, 14 key enzyme genes were up-regulated in the tanshinone biosynthesis pathway: DXS, DXS2, DXR, HMGR3, AACT, MK, PMK, GGPPS2, GPPS, KSL, IDI, IPII, FDPS and CPS. Discussion and conclusions: A total of 14 key genes were obtained from the tanshinone component synthesis and metabolic pathways, providing a reasonable explanation for the accumulation of tanshinone components, an accumulation induced by endophytic fungi, in the host plants. Aphids are noxious insect pests of major crops including cereals and legumes. Particularly, pea aphid (Acyrthosiphon pisum) causes significant yield and quality loses in pea. Crop protection is largely based on noxious chemical pesticides which have prompted a renewed interest in the discovery of natural products as alternatives to synthetic insecticides. In this study different classes of natural compounds were tested in dual choice bioassays to evaluate their feeding deterrence and mortality effect on pea aphid. High feeding deterrence was produced by some of the compounds, particularly1-hexadecanol, gliotoxin, cyclopaldic acid and seiridin. On the contrary, aphid mortality was low although significant for 1-heptadecanol, cytochalasin A, 1-nonadecanol and gliotoxin. Phytotoxicity assessment showed low or imperceptible plant damaged for cytochalasin A, seiridin and 1-nonadecanol. This study aimed to assess aflatoxin and fumonisin intake through corn tortilla consumption in Veracruz city. Between October 2013 and February 2015, a total of 120 corn tortilla samples (2 kg samples, 40 samples per year) were randomly collected. Aflatoxins and fumonisins were quantified by high performance liquid chromatography coupled with a fluorescence detector. A probability density function (PDF) was used for describing corn tortilla intake, body weight of the Veracruz city population, mycotoxin content of corn tortilla samples and estimated mycotoxin daily intake. The Monte Carlo method with 10,000 iterations was employed to assess the population exposure risk. The highest level of total aflatoxins (AFT) was 22.17 μg kg−1, and 526.6 μg kg−1 for fumonisins B1 plus B2, with 85% and 90% of contaminated samples respectively. Up to 69.7 % of the population was estimated to consume a higher aflatoxin dose than that recommended by the JECFA (1 ng kg−1 of body weight per day); it was found that the recommended dose was exceeded to a greater extent in the male population, due to higher consumption of corn. The risk of fumonisin intake was less than 5 % due to the low presence and levels of these toxins in corn tortillas. The results suggest that corn tortilla consumers are at dietary risk caused by AFT contamination; this information should be considered when taking action to protect public health. Edible and medicinal mushrooms have usually been considered as a sustainable source of unique bioactive metabolites, which are valued as promising provisions for human health. Antrodia cinnamomea is a unique edible and medicinal fungus widespread in Taiwan, which has attracted much attention in recent years for its high value in both scientific research and commercial applications owing to its potent therapeutic effects, especially for its hepatic protection and anticancer activity. Due to the scarcity of the fruiting bodies, the cultivation of A. cinnamomea by submerged fermentation appears to be a promising substitute which possesses some unique advantages, such as short culture time period and its high feasibility for scale-up production. However, the amount of fungal bioactive metabolites derived from the cultured mycelia of A. cinnamomea grown by submerged fermentation is much less than those obtained from the wild fruiting bodies. Hence, there is an urgent need to bridge such a discrepancy on bioactive metabolites between the wild fruiting bodies and the cultured mycelia. The objective of this article is to review recent advances and the future development of the mycelial submerged fermentation of A. cinnamomea in terms of enhancement for the production of fungal bioactive components by the optimization of culture conditions and the regulation of fungal metabolism. Consumers demand "fresh-like" and convenient products with preserved nutritional and organoleptic characteristics that can be kept for extended periods of time without compromising safety. This has posed greater challenges to food industries. High-pressure processing (HPP) can improve food safety by destroying the microorganisms that cause food-borne illness and spoilage that cause diseases. It also leads to the reduction of fungal toxic metabolites in food products when combined with moderate temperature. HPP holds promise as an emerging food treatment to process premium value food products while retaining food quality, maintaining natural freshness, and extends microbiological shelf life. Forty-four endophytes were isolated from Zingiber cassumunar and identified morphologically. The ethyl acetate extracts of all endophytes were obtained. The ethyl acetate extracts were subjected to study antibacterial and antioxidant activities. The ethyl acetate extract of the Arthrinium sp. MFLUCC16-1053 showed activity against both gram-positive and gram-negative bacteria. Specifically, the minimum inhibition concentration against Staphylococcus aureus and Escherichia coli was 31.25 and 7.81 µg/mL, respectively. In addition, the crude extract showed highest antioxidant activity in 2,2-diphenyl-1-picrylhydrazyl scavenging at IC50 value of 28.47 µg/mL. Gas chromatography-mass spectrometry analysis revealed that the extract of the Arthrinium sp. MFLUCC16-1053 contains various antibacterial and antioxidant compounds which are β-cyclocitral, 3E-cembrene A, laurenan-2-one, sclareol, 2Z,6E-farnesol, cembrene, β-isocomene and γ-curcumene. The present study was conducted to explore the occurrence of mycotoxins in commercial baby foods in Doha-Qatar. LCMS/MS- and HPLC-based analysis of baby food (n = 67) for 12 mycotoxins confirmed the presence of aflatoxin M1 (AFM1, 33%), ochratoxin A (OTA, 31%), deoxynivalenol (DON, 27%), aflatoxin B1 (AFB1, 22%), fumonisin B2 (FB2, 10%), zearalenone (ZEN, 4%) and T-2 toxin (2%). Noodles exhibited the maximum contamination percentage, with 33% of the samples being contaminated above the EU maximum limits, for at least one mycotoxin. Among the multi-grain flake samples, up to 28% and for the milk and milk-based-cereal samples, 14% contained at least one mycotoxin above the EU maximum limits. From all cereal-based food samples, 22%, 5%, 2% and 2% were concurrently contaminated with 2, 3, 4 and 5 mycotoxins, respectively. Consumption of mycotoxin contaminated foodstuffs is common in regions where foods are not adequately controlled and routinely monitored, and this could have adverse effects on the health of consumers. In this study, 100 samples of roasted nuts (50 cashew nuts and 50 peanuts) vended within two cities of Sierra Leone were analysed for mycotoxins and other microbial metabolites by a LC-MS/MS method. The peanut samples contained detectable levels of 17 microbial metabolites, including aflatoxins B1, B2, G1 and G2 and alternariol, while none of these metabolites were found in the cashew samples. Aflatoxins (max: 5,729 μg/kg; mean: 487.8 μg/kg) and alternariol (3 μg/kg) were found in 24% and 2% of the peanut samples, respectively. One-third of the aflatoxin-contaminated peanut samples contained aflatoxins at levels exceeding the total aflatoxin limit of 4 μg/kg set by the European Union. Lasiodiplodia theobromae is a phytopathogenic fungus that causes diseases not only in a broad number of plant hosts but also occasionally in humans. The capacity of L. theobromae to produce bioactive metabolites at 25 C (environmental mean temperature) and at 37 C (body mean temperature) was investigated. Two strains, CAA019 and CBS339.90, isolated respectively from a coconut tree and a human patient were characterized. The phytotoxicity and cytotoxicity (on mammalian cells) of the secretomes of both strains of L. theobromae were investigated. Also, phytotoxicity and cytotoxicity of pure compounds were evaluated. The phytotoxicity of the secretome of strain CAA019 was higher than the phytotoxicity of the secretome of strain CBS339.90 at 25 C. However, the phytotoxicity for both strains decreased when they were grown at 37 C. Only the secretome of strain CBS339.90 grown at 37 C induced up to 90% Vero and 3T3 cell mortality. This supports the presence of different metabolites in the secretome of strains CAA019 and CBS339.90. Metabolites typical of L. theobromae were isolated and identified from organic extracts of the secretome of both strains (e.g., 3-indolecarboxylic acid, jasmonic acid, lasiodiplodin, four substituted 2-dihydrofuranones, two melleins, and cyclo-(Trp-Ala)). Also, metabolites such as scytalone, not previously reported for this species, were isolated and identified. Metabolite production is affected by strain and temperature. In fact, some metabolites are strain specific (e.g., lasiodiplodin) and some metabolites are temperature specific (e.g., jasmonic acid). Although more strains should be characterized, it may be anticipated that temperature tuning of secondary-metabolite production emerges as a putative contributing factor in the modulation of L. theobromae pathogenicity towards plants, and also towards mammalian cells. An ultrahigh-performance liquid chromatography–tandem mass spectrometry method was developed and validated for the simultaneous determination of sterigmatocystin, verruculogen, enniatin A, fusarenon-X, fumonisins B1, B2, B3, aflatoxins B1, B2, G1, G2, ochratoxin A, deoxynivalenol, 3-acetyldeoxynivalenol, 5-acetyldeoxynivalenol, and zearalenone. The mycotoxins were extracted and cleaned up using a multitoxin column, separated on a C18 column, and then detected on a triple-quadrupole mass spectrometer. The limits of detection and quantification ranged within 0.2–2 µg/kg and 1–10 µg/kg, respectively. The recoveries ranged from 70.8 to 118.4%, with relative standard deviations below 15%. The method was used to analyze 80 samples obtained from Shandong Province in China. Fifty-eight samples were contaminated with 10 mycotoxins at concentrations ranging from 1.4 to 6566.1 µg/kg. Mycotoxins are a serious food safety concern for human and animal health. Much attention should be paid to the dietary exposure to mycotoxins in order to minimise the risk of mycotoxin contamination in the food chain. Among the reported strategies to manage the mycotoxin contamination into food and feed, biological control seems a promising approach, depending on their biological origins, and on the use of living organisms or their derivatives. Marine microorganisms have developed unique metabolic and physiological capabilities to thrive in extreme habitats and produce novel metabolites which are not often present in microbes of terrestrial origin. Some marine bacteria and fungi have a good potential for the control of fungal phytopathogens and mycotoxins. Isaria cicadaeis an entomogenous fungus that has been used as a traditional Chinese medicinal materials to treat different diseases, including cancer. However,Isaria cicadaeconidia for inhibitory activity against breast cancer cells growth are still not systematically studied. A hybrid linear ion trap-quadrupole-Orbitrap high-resolution mass spectrometry (LTQ-Orbitrap-HRMS) was used to qualitatively and quantitatively analyse the myriocin inIsaria cicadaeand its allies. The rates of 1,1-diphenyl-2-picryhydrazyl (DPPH)-free radical scavenging and hydroxyl radical eliminating were employed as indexes to qualitatively and quantitatively analyse the radical scavenging activity of the extracts with different solvents from fermented mycelia ofHirsutella huangshanensisRCEF0868, the anamorph ofOphiocordyceps formosana(FMOF). The results showed that both the aqueous extract and the methanol extract had significant radical scavenging activity. The half maximal inhibitory concentrations (IC50) of the aqueous extract on DPPH-free radical and hydroxyl radical are 0.85 and 1.37 mg/mL, respectively, while the IC50of methanol extract is 1.23 and 2.91 mg/mL, respectively. The combined liquid chromatography-diode array detector-high resolution mass spectrometer(HRMS) analysis and activity determination revealed that the molecular formulas of three radical scavengers in aqueous extract were C6H14O6(1), C14H17N5O8(2) and C20H33N5O9(3). In addition to the compound 1, the methanol extract had another three radical scavengers as C30H22O11(4), C30H22O10(5) and C30H18O10(6). All the six kinds of components exhibited the DPPH radical scavenging activity, and compounds 1, 4 and 5 also showed the hydroxyl radical eliminating activity at the same time. By querying the database of natural products and reference to the relevant reports, compound 3 may be a new entity. The others were respectively cordycepic acid, succinoadenosine, 4α-oxyrugulosin, rugulosin and skyrin and, among them, succinoadenosine and 4α-oxyrugulosin were found in entomogenous fungi for the first time. Microbial spoilage of bread and the consequent waste problem causes large economic losses for both the bakery industry and the consumer. Furthermore the presence of mycotoxins due to fungal contamination in cereals and cereal products remains a significant issue. The use of conventional chemical preservatives has several drawbacks, necessitating the development of clean-label alternatives. In this review, we describe current research aiming to extend the shelf life of bread through the use of more consumer friendly and ecologically sustainable preservation techniques as alternatives to chemical additives. Studies on the in situ-production/-expression of antifungal compounds are presented, with special attention given to recent developments over the past decade. Sourdough fermented with antifungal strains of lactic acid bacteria (LAB) is an area of increasing focus and serves as a high-potential biological ingredient to produce gluten-containing and gluten-free breads with improved nutritional value, quality and safety due to shelf-life extension, and is in-line with consumer's demands for more products containing less additives. Other alternative biopreservation techniques include the utilization of antifungal peptides, ethanol and plant extracts. These can be added to bread formulations or incorporated in antimicrobial films for active packaging (AP) of bread. Mycotoxins are secondary fungal metabolites produced mainly byAspergillus, Penicillium, andFusarium. As evidenced by large-scale surveys, humans and animals are simultaneously exposed to several mycotoxins. Simultaneous exposure could result in synergistic, additive or antagonistic effects. However, most toxicity studies addressed the effects of mycotoxins separately. We present the experimental designs and we discuss the conclusions drawn from in vitro experiments exploring toxicological interactions of mycotoxins. We report more than 80 publications related to mycotoxin interactions. The studies explored combinations involving the regulated groups of mycotoxins, especially aflatoxins, ochratoxins, fumonisins, zearalenone and trichothecenes, but also the “emerging” mycotoxins beauvericin and enniatins. Over 50 publications are based on the arithmetic model of additivity. Few studies used the factorial designs or the theoretical biology-based models of additivity. The latter approaches are gaining increased attention. These analyses allow determination of the type of interaction and, optionally, its magnitude. The type of interaction reported for mycotoxin combinations depended on several factors, in particular cell models and the tested dose ranges. However, synergy amongFusariumtoxins was highlighted in several studies. In recent years climate changes recorded in temperate regions of Europe have led to aflatoxin (AF) contamination of maize. Thus, the aim of this study was to investigate the influence of weather conditions on levels of aflatoxin B1 (AFB1), aflatoxin B2 (AFB2), aflatoxin G1 (AFG1) and aflatoxin G2 (AFG2) in 180 maize samples collected from the main maize-growing regions (Western Bačka, North Banat, South Banat and Central Serbia) in Serbia after harvest in 2015. The concentrations of AFs were determined by a validated HPLC method with post-column derivatisation and fluorescence detection (HPLC-FLD). The presence of AFB1, AFB2, AFG1 and AFG2 was detected in 57.2%, 13.9%, 5.6% and 2.8% of maize samples in the concentration ranges of 1.3–88.8 µg kg–1, 0.60–2.8 µg kg–1, 1.8–28.5 µg kg–1 and 2.1–7.5 µg kg–1 respectively. The recorded smaller amount of precipitation and especially higher air temperatures during the summer of 2015 were favourable for AF production, which resulted in 32.2% and 21.1% of samples being unsuitable for human consumption, since AFB1 and the sum of AFs concentrations were above 5.0 and 10.0 µg kg–1 respectively. Furthermore, the findings in this study indicate that the microclimate conditions in the investigated regions had a great influence on the contamination frequency of maize with AFs. The highest percentage of samples unsuitable for human consumption, considering both AFB1 and total AFs content were 72.5% and 51.5% respectively from Central Serbia, whilst the lowest percentages of 15.6% and 6.2% respectively were found in Western Bačka. In this study a strain ofStreptomycessp. was isolated from soil and identified by 16S rRNA gene sequencing technology. The strain was screened for antibiotics production effective against biocontrol ofRhizoctonia solani AG-3to cure the target spot disease in tobacco. For enhance production of secondary metabolites, central composite design of response surface methodology (RSM) was applied in submerged fermentation. The maximum metabolite production was using medium volume of 55 mL in 250 mL flask, agitation speed of 165 rpm, incubation temperature 30 °C, initial medium pH of 6.8 and inoculum size of 7%. Solvent extraction method was used to extract the secondary metabolites and active compounds were purified by silica gel column chromatography. The purified fractions were further investigated by gas chromatography-mass spectrophotometer (GC-MS). GC-MS analysis showed 48 compounds, among them 12 were active against pathogen. This study reports the distribution of fungal isolates and fungal metabolites that naturally contaminate locally processed rice from five agro-ecological zones of Nigeria. The fungal species were isolated by the dilution plate technique and identified by appropriate diagnostics, while metabolites were determined by a liquid chromatographic tandem mass spectrometric method. AspergillusandPenicilliumspecies were the predominant isolates found in the rice samples whileFusariumspp. were not isolated. The mean fungal count differed significantly (p < 0.05) across the zones and ranged from 9.98 × 102 cfu g−1in the Southern Guinea Savannah to 96.97 × 102 cfu g−1in the Derived Savannah. For 16 fungal metabolites, selected from 63 positively identified fungal metabolites based on their concentration and spread across the zones, an occurrence map was constructed. Metarhiziumspp. are known to produce destruxin A (dtx A) and can act as endophytes. Data regarding the fate and behaviour of secondary metabolites in the environment are necessary for registration. Endophytic colonisation and dtx A production on potato plants were monitored at 24, 48, 72, 96 and 120 h after inoculation withMetarhizium brunneumstrains (BIPESCO5 and EAMa 01/58-Su). Both strains were recovered from leaves, stem, tuber and root fragments of fungal-challenged potato plants. Although a similar colonisation was observed for both strains, there were differences in percentages in different parts of the plants, with the higher values occurring in the leaves at 96 h for EAMa 01/58-Su (83.3%) and BIPESCO5 (81.6%), and the lower ones, 10–13.3%, observed in tubers and roots at 72, 96 and 120 h post-inoculation for both strains. For strain EAMa 01/58-Su, dtx A was quantified at 24 h (2.49 ± 1.7 and 2.0 ± 1.4 µg/kg, respectively), and the same concentration was found in both tuber and root at 96 h (2.5 ± 1.7 µg/kg); for BIPESCO5, the concentrations differed in tuber at 24 h and in root at 48 h (6.8 ± 4.8 and 2.1 ± 1.4 µg/kg, respectively). The concentration of dtx A in plant tissues was very low compared to the colonisation levels, suggesting that dtx A production by the fungus may be temporary and that the compound might degrade rapidly. Context:Endophytic fungi, being a prolific source of bioactive secondary metabolites, are of great interest for natural product discovery. Objective:Isolation and partial characterization of endophytic fungi inhabiting the leaves and woody parts ofTaxus fuanaNan Li & R.R. Mill. (Taxaceae) and evaluation of biological activity. Materials and methods:Endophytic fungal isolates were identified by molecular analysis of internal transcribed spacer (ITS) regions of 18S rDNA. Extracts of the endophytic fungi cultured on potato dextrose agar and modified medium were evaluated using cancer chemoprevention bioassays [inhibition of TNF-α-induced NFκB, aromatase and inducible nitric oxide synthase (iNOS); induction of quinone reductase 1 (QR1)] and growth inhibition with MCF-7 cells. Results:Nine of 15 fungal isolates were identified as belonging toEpicoccum,Mucor,Penicillium,Chaetomium, Paraconiothriym, PlectaniaorTrichoderma. Five of the 15 extracts inhibited NFκB activity (IC50values ranging between 0.18 and 17 μg/mL) and five inhibited iNOS (IC50values ranging between 0.32 and 12.9 μg/mL). In the aromatase assay, only two isolates mediated inhibition (IC50values 12.2 and 10.5 μg/mL). With QR1 induction, three extracts exhibited significant activity (concentrations to double activity values ranging between 0.20 and 5.5 μg/mL), and five extracts inhibited the growth of MCF-7 cells (IC50values ranging from 0.56 to 17.5 μg/mL). Six active cultures were derived from woody parts of the plant material. Conclusion:The endophytic fungi studied are capable of producing pharmacologically active natural compounds.
