Accurately evaluating particle surface micro-texture levels and distribution at different wavelength scales of granular material is critical to optimise service stability of granular material under long-term dynamic loads. Firstly, generalised regression neural network (GRNN) and empirical mode decomposition (EMD) were adopted to impute missing data points and remove the arc-shaped tendency of granular materials' particle wear raw surface profile. Then, granular materials' particle surface micro-texture levels and their distribution within constant bandwidth narrow band spectrum and octave/fractional octave band spectrum were obtained with spectral analysis method, which could characterise granular materials' particle surface properties at different wavelength scales. Fourteen types of parent rocks of granular materials were tested with a modified micro tribological experiment simulating tribological behaviour among particle contact interfaces under dynamic loads. A contrastive analysis with traditionally used surface mean roughness was performed. The results indicate that the micro-texture levels and their distribution obtained in this study can detect the level of the exact texture scales (i.e. 32 and 2 μm wavelengths) significantly influencing the kinetic friction coefficient of granular materials, while surface mean roughness can only represent the global surface property. This paper discusses the implementation considerations of Balanced Mix Design (BMD) in both design and production phases according to the recent experience from the agency and a contractor in Vermont since BMD was implemented by Vermont Agency of Transportation (VTrans) in 2019. In the design, the sensitivity of selected performance tests to mix design factors and the impact of asphalt binder source are two important aspects to consider as both agency and contractor need to understand the implications on the potential change of mix compositions. There are also several critical things to consider if BMD is implemented in production, including but not limited to the impact of production variation, the difference between design and production, and so forth. Objective Preeclampsia (PE) is a disorder that occurs during the pregnancy and could affect the maternal and perinatal mortality as well as morbidity. The aim of our study is to investigate the associations between the hypertension susceptibility genes ITGA9, MOV10 and CACNB2 with PE in Chinese Han population. Methods A case–control study including 178 PE patients and 202 healthy controls was conducted to assess the associations between three loci (ITGA9 rs155524, MOV10 rs2932538 and CACNB2 rs4373814) and PE. The TaqMan probe assay was applied for genotyping in our study. Quantitative real-time PCR was performed to detect the mRNA expression levels of ITGA9, MOV10 and CACNB2. ELISA was carried out to detect the concentration of serum sFlt-1 or PLGF. Results Our study detected no significant differences in allelic frequencies of three SNPs between PE patients and healthy controls. In the genetic model, the results showed that the patients with ITGA9 rs155524 GA or AA genotypes had a higher risk of PE development compared to those with GG genotype in codominant model. And PE patients had a higher frequency of GA + AA genotypes based on the dominant model. Subgroup analysis showed ITGA9 rs155524 was associated with early-onset PE but not with late-onset PE. No association was observed between MOV10 and CACNB2 with PE in any genetic model and subgroup analysis. Quantitative real-time PCR results showed that ITGA9 mRNA expression level was apparently increased in the placental tissues of PE patients. In addition, ITGA9 expression levels of GA + AA subjects were apparently higher than that in the genotype GG of placental tissues. sFlt-1/PLGF ratio was higher in GA + AA subjects than that in GG subjects. Regression analysis revealed that ratio of sFlt-1/PLGF was positively correlated with ITGA9 mRNA expression level. Conclusion This study has identified ITGA9 is a promising candidate susceptibility gene for early-onset PE. Pavement skid resistance plays a key role in traffic safety. Meanwhile, tire-pavement noise is a major source of traffic noise in urban areas. Current asphalt mixture design methods, however, mostly focus on volumetric and mechanical properties and pay little attention to the skid resistance and noise reduction performance of asphalt mixtures, which are significantly affected by the surface textures of asphalt mixtures. Incorporating the evaluation of surface texture into the mixture design would aid in a more rational selection of materials considering both mechanical and functional properties of asphalt mixtures. In this paper, the surface texture properties of several types of asphalt mixtures are measured using a recently developed 2-Dimensional Image Texture Analysis Method. A prediction model correlating the mixture surface texture levels at different central texture wavelength in octave band with the important mix design parameters is established using a multivariate non-linear regression analysis. The model is validated through laboratory test and imaging measurement indicating its capability of predicting the level and distributions of mixture surface texture. Based on the multiplicative update rule of nonnegative matrix factorization (NMF), we add the nonnegative constraint of endmember matrix to the objective function of iterative constrained endmembers (ICE), and propose a simple multiplicative update rule for ICE, named NMF-ICE. Our method avoids the use of quadratic programming method in ICE, therefore can greatly improve the computational efficiency. Bright/Arid3a has been characterized both as an activator of immunoglobulin heavy-chain transcription and as a proto-oncogene. Although Bright expression is highly B lineage stage restricted in adult mice, its expression in the earliest identifiable hematopoietic stem cell (HSC) population suggests that Bright might have additional functions. We showed that >99% of Bright-/- embryos die at midgestation from failed hematopoiesis. Bright-/- embryonic day 12.5 (E12.5) fetal livers showed an increase in the expression of immature markers. Colony-forming assays indicated that the hematopoietic potential of Bright-/-mice is markedly reduced. Rare survivors of lethality, which were not compensated by the closely related paralogue Bright-derived protein (Bdp)/Arid3b, suffered HSC deficits in their bone marrow as well as B lineage-intrinsic developmental and functional deficiencies in their peripheries. These include a reduction in a natural antibody, B-1 responses to phosphocholine, and selective T-dependent impairment of IgG1 class switching. Coagulation is a commonly adopted process as a pretreatment step for minimizing membrane fouling. Three coagulants, polyaluminum chloride (PACl), alum, and FeCl3 were tested under four mixing-settling schemes for turbidity and natural organic matters (NOM) removal. The organic matters in the raw waters were fractionated by high performance size exclusion chromatography (HP-SEC) system and were characterized using the excitation-emission matrix (EEM) fluorescence spectra. Sufficient rapid mixing and slow mixing or applying two-stage coagulation benefit turbidity removal using PACl or FeCl3 under “electrostatic patch coagulation” (EPC) mechanism. The EPC mechanism is not efficient for alum coagulation. At higher coagulant doses, the NOM removal is not affected by mixing condition. Intensive rapid mixing alone benefits NOM removal using PACl or FeCl3. Alum is a poor coagulant for NOM removal under EPC mechanism. [ABSTRACT FROM AUTHOR]

Glucose-6-phosphate dehydrogenase (G6PD), the first and rate-limiting enzyme of the pentose phosphate pathway, is indispensable to maintenance of the cytosolic pool of NADPH and thus the cellular redox balance. The role of G6PD as an antioxidant enzyme has been recognized in erythrocytes for a long time, as its deficiency is associated with neonatal jaundice, drug- or infection-mediated hemolytic crisis, favism and, less commonly, chronic non-spherocytic hemolytic anemia. To a large extent, advances in the field were made on the pathophysiology of G6PD-deficient erythrocytes, and the molecular characterization of different G6PD variants. Not until recently did numerous studies cast light on the importance of G6PD in other aspects of the physiology of both cells and organisms. Deficiency in G6PD activity, and hence a disturbance in redox homeostasis, can lead to dysregulation of cell growth and signaling, anomalous embryonic development, altered susceptibility to viral infection as well as increased susceptibility to degenerative diseases. The present review covers recent developments in this field. A new anaerobic rumen fungus was isolated from the rumen fluid of a yellow cow (Bos indicus). This fungus appears to be a previously undescribed species of the genus Caecomyces, it possessing uniflagellate zoospores, a spherical holdfast, tubular sporangiophores and bulbous rhizoids. This new fungus also features distinctive multisporangiate thallus sympodially distributed on sporangiophores. The fungus resembles Caecomyces communis and C. equi in that it characterizes bulbous rhizoids and uniflagellate zoospores but differs from C. communis and C. equi in that it possesses multisporangiate and sympodial sporangia. This new fungus and Cyllamyces aberensis both reveal similar morphology during early thallus development in having a spherical holdfast, but they vary from unbranched sporangiophores and additional bulbous rhizoids. In addition, the molecular phylogenetic analyses ITS1 (internal transcribed spacer 1) also conform to the results of the morphological examinations of Caecomyces. For the mentioned reasons, this new species of fungus is described as Caecomyces sympodialis sp. nov. The genera of Neocallimasticaceae and species of Caecomyces are also keyed out. Consumers generally concern taste and odor in drinking water. In the Southern Taiwan, more than 5,000,000 people are suffered from earth/musty odor in drinking water, especially in the summer. Thus, ozonation of geosmin (GSM), 2-methylisoborneol (2-MIB), and 2-furfural (2-FF) in eutrophic surface waters has been studied in the present work. Experimentally, it was found that the water contained high dissolved organic carbon (DOC), humic substances, and specific ultraviolet absorbance (SUVA) resulting the highly ozone (O 3 ) demand. The natural organic matters (NOM) in the waters had a significant effect on the ozonation of GSM, 2-MIB and 2-FF. Their destruction rates were increased with high contents of aromatics, phenolics, and SUVAs. In addition, during ozonation of raw waters, O 3 and OH· played an important role in destruction of algae cells and caused excretion of extracellular organic matter (EOM) to the bulk phase. The article informs that in the autumn of 1971, Little Yü, the daughter of Commander Yü of the Lin-ts'ang Military Region, graduated from lower middle school. She was now faced with the kind of question she had not encountered in school: whether she should continue her studies or go down to the countryside. She had grown up in the military camps, and had always wanted to become a Liberation Army soldier to defend the Motherland. Soon after, the hsien revolutionary committee called upon those educated youths who had decided against continuing their studies to go down and settle in the countryside. When she first arrived at the Ta-chai Production Team, she felt that the mountains were immense and inaccessible. But now she felt that the scenery was beautiful and the weather pleasant. The poor and lower-middle peasants were close to her, and she loved every single blade of grass and tree in the place. She was now beginning to have an understanding of the ideas and life of the laboring people, and she now knew how she should serve them and help to build up the mountain area. Whenever any of the women, after a day' s work in the fields, had difficulty in carrying her Inby up the mountainside, Little Yü would help her carry the child.

Posttranslational modifications play important roles in regulating protein structure and function. Histone deacetylase 6 (HDAC6) is a mostly cytoplasmic class II HDAC, which has a unique structure with two catalytic domains and a domain binding ubiquitin with high affinity. This enzyme was recently identified as a multisubstrate protein deacetylase that can act on acetylated histone tails, α-tubulin and Hsp90. To investigate the in vivo functions of HDAC6 and the relevance of tubulin acetylation/deacetylation, we targeted the HDAC6 gene by homologous recombination in embryonic stem cells and generated knockout mice. HDAC6-deficient mice are viable and fertile and show hyperacetylated tubulin in most tissues. The highest level of expression of HDAC6 is seen in the testis, yet development and function of this organ are normal in the absence of HDAC6. Likewise, lymphoid development is normal, but the immune response is moderately affected. Furthermore, the lack of HDAC6 results in a small increase in cancellous bone mineral density, indicating that this deacetylase plays a minor role in bone biology. HDAC6-deficient mouse embryonic fibroblasts show apparently normal microtubule organization and stability and also show increased Hsp90 acetylation correlating with impaired Hsp90 function. Collectively, these data demonstrate that mice survive well without HDAC6 and that tubulin hyperacetylation is not detrimental to normal mammalian development. [ABSTRACT FROM AUTHOR]

Homologs of the Saccharomyces cerevisiae Sir2 protein, sirtuins, promote longevity in many organisms. Studies of the sirtuin SIRT3 have so far been limited to cell culture systems. Here, we investigate the localization and function of SIRT3 in vivo. We show that endogenous mouse SIRT3 is a soluble mitochondrial protein. To address the function and relevance of SIRT3 in the regulation of energy metabolism, we generated and phenotypically characterized SIRT3 knockout mice. SIRT3-deficient animals exhibit striking mitochondrial protein hyperacetylation, suggesting that SIRT3 is a major mitochondrial deacetylase. In contrast, no mitochondrial hyperacetylation was detectable in mice lacking the two other mitochondrial sirtuins, SIRT4 and SIRT5. Surprisingly, despite this biochemical phenotype, SIRT3-deficient mice are metabolically unremarkable under basal conditions and show normal adaptive thermogenesis, a process previously suggested to involve SIRT3. Overall, our results extend the recent finding of lysine acetylation of mitochondrial proteins and demonstrate that SIRT3 has evolved to control reversible lysine acetylation in this organelle.
