The current study examined the effects of different types of instructor comments on student engagement in an online discussion. In particular, this study examined three comment types: (1) praise-oriented comments, agreeing with students' initial messages and recapping their ideas, (2) elaboration-encouraging comments, requesting elaboration on the initial messages, and (3) perspective-widening comments, suggesting different or thought-provoking opinions responding to the initial messages. Seventeen graduate students enrolled in an online course were recruited. A content analysis was employed to evaluate the levels of knowledge construction. Interactivity was measured by the number of posts and idea units. The findings revealed that perspective-widening comments facilitated students' evaluation on the peer postings and brought up diverse perspectives, contributing to knowledge construction. While elaboration-encouraging comments led to high interactivity among students, praise-oriented comments had no significant effect on the frequency of interactivity. [ABSTRACT FROM AUTHOR]

Recently identified highly pathogenic avian influenza (HPAI) H5N8 viruses (clade 2.3.4.4) are relatively low to moderately pathogenic in mammalian hosts compared with HPAI H5N1 viruses. In this study, we generated reassortant viruses comprised of A/MD/Korea/W452/2014(H5N8) with substitution of individual genes from A/EM/Korea/W149/2006(H5N1) to understand the contribution of each viral gene to virulence in mammals. Substituting the PB2 gene segment or the NA gene segment of the H5N8 virus by that from the H5N1 virus resulted in significantly enhanced pathogenicity compared with the parental H5N8 virus in mice. Of note, substitution of the PB2 gene segment of the H5N8 virus by that from the H5N1 virus resulted in a 1000-fold increase in virulence for mice compared with the parental virus (MLD50 decreased from 105.8 to 102.5 EID50). Further, the W452W149PB2 virus also induced the highest virus titers in lungs at all time points and the highest levels of inflammatory cytokine responses among all viruses tested. This high virulence phenotype was also confirmed by high viral titers in the respiratory tracts of infected ferrets. Further, a mini-genome assay revealed that W452W149PB2 has significantly increased polymerase activity (p < 0.001). Taken together, our study demonstrates that a single gene substitution from other avian influenza viruses can alter the pathogenicity of recent H5N8 viruses, and therefore emphasizes the need for intensive monitoring of reassortment events among co-circulating avian and mammalian viruses. Phenylbutazone was crystallized from solutions by the liquid antisolvent recrystallization technique. Acetone was used as a solvent, and distilled water was selected as an antisolvent. The influence of processing parameters, such as drug concentration, temperature, injection rate of drug solution, and mixing method of drug solution with antisolvent, on the particle size distribution were investigated. Furthermore, to examine the variation of resulting particle size in the presence of the ultrasound, the ultrasonic wave was applied to all experiments. Larger crystals were obtained when crystallization took place at higher temperatures. The enhancement of drug concentration favored decreased particle size. Regarding the mixing method of the drug solution and antisolvent, smaller particles were produced when the drug solution was injected into antisolvent, and larger crystals were obtained when the antisolvent was injected into drug solutions. As the injection rate of the drug solution increased, the average particle size decreased. The processed particles consistently exhibited an acicular crystal habit. The modal coordinate formulation (MCF) has been used to describe small deformations, and the absolute nodal coordinate Formulation (ANCF) has usually been employed to describe large deformations in flexible multibody dynamics. In this article, a hybrid coordinate formulation (HCF) was suggested to combine the modal coordinates for small deformations with the absolute nodal coordinates for large deformations. The kinematic constraint equations for a spherical joint and the equation for two perpendicular vectors were developed with both coordinates and applied to other types of kinematic joints including a fixed joint. A stepped cantilever beam was tested to show the validity of the suggested hybrid formulation. Physical experiments with a high-speed camera were carried out to capture the large deformation of the beam and to verify the results of computer simulations. The numerical results obtained from the hybrid coordinates were compared with the results from the simulation using the absolute nodal coordinates only and physical experiments. Supercritical extraction has been performed in a fixed column to desorb phenol and 4-nitrophenol from organically modified smectite. The experiments were carried out in the sequence of adsorption of hexadecyltrimethylammonium (HDTMA) to montmorillonite, adsorption of phenols to organoclay in aqueous solutions, desorption of phenols from loaded organoclay using supercritical carbon dioxide, and adsorption of phenols to regenerated organoclay. The desorption characteristics of phenols were investigated at various pressures up to 420 bar; at temperatures of 40, 60, and 70°C, and at low concentrations of a cosolvent. The extraction percentages of phenols reached up to 90% in 3 hours of extraction. The results showed that under the experimental conditions investigated, the activity of HDTMA was intact during the supercritical extraction of phenols, and hence HDTMA-modified montmorillonite exhibited undiminished adsorption power toward phenols after several regeneration cycles. Purpose: To investigate whether a signal alteration (SA) in the optic nerve head (ONH) on 3D T2-weighted magnetic resonance imaging (MRI) is associated with glaucomatous optic neuropathy Materials and Methods: A total of 35 patients with bilateral open-angle glaucoma and 31 age-matched controls underwent 3D high-resolution (0.98 × 0.98 × 1 mm3) T2-weighted MRI and detailed ophthalmologic examinations including spectral-domain optical coherence tomography (OCT). Two independent reviewers blinded to subject data determined mild or prominent SA on 3D T2-weighted MRI for the incomplete or complete loss of ocular hypointense continuity in the ONH, respectively. The prevalence of SAs was compared between the two groups with the chi-square test. The OCT measurements were compared among the eyes with a mild or prominent SA and those without an SA using the Kruskal–Wallis test. Results: Of the 35 eyes with glaucoma, 26 eyes (74.3%) exhibited an SA in the ONH, whereas it was observed in 5 (16.1%) of the 31 controls (P < 0.001). The eyes with a prominent SA had a significantly different average retinal nerve fiber layer thickness (P = 0.002) and the ONH parameters except for the disk area (allP < 0.001) than those without an SA. The eyes with a mild SA had a significantly narrower neural rim area, larger cup volume, and larger average and vertical cup-to-disk ratios compared with those without an SA (P = 0.011, 0.003, 0.004, and 0.004, respectively) Conclusions: The SA in the ONH on 3D T2-weighted MRI was significantly more frequent in eyes with open-angle glaucoma than in the controls. A new finite element of a thin spatial beam is proposed using the absolute nodal coordinate formulation (ANCF). In contrast to other large deformation formulations, the equations of motion contain constant mass matrices and generalized gravity forces as well as zero centrifugal and Coriolis inertia forces. The only nonlinear term is a vector of elastic forces. The elements of equation of motion are developed in details including air resistance forces and constraint equations to satisfy boundary conditions. Sialidases are key virulence factors that remove sialic acid from the host cell surface glycan, unmasking receptors that facilitate bacterial adherence and colonisation. In this study, we developed potential agents for treating bacterial infections caused by Streptococcus pneumoniae Nan A that inhibit bacterial sialidase using Turmeric and curcumin analogues. Design, synthesis, and structure analysis relationship (SAR) studies have been also described. Evaluation of the synthesised derivatives demonstrated that compound 5e was the most potent inhibitor of S. pneumoniae sialidase (IC50 = 0.2 ± 0.1 µM). This compound exhibited a 3.0-fold improvement in inhibitory activity over that of curcumin and displayed competitive inhibition. These results warrant further studies confirming the antipneumococcal activity 5e and indicated that curcumin derivatives could be potentially used to treat sepsis by bacterial infections. This study examined the types of learning behaviors students demonstrated while performing inquiry tasks. It also explored the relationship between the learning behaviors and students’ domain knowledge. We observed fourteen students in five groups during a ninth-grade biology course. Three types of learning behaviors (inquiry, collaborative, and minimally productive behaviors) were identified and time on each type was measured. The results suggest that students demonstrated different patterns according to the types of learning behaviors. Correlation analysis revealed that learning outcomes had a strong positive correlation with the inquiry behaviors but a strong negative correlation with the minimally productive behaviors. The results suggest that attention should be paid to facilitate the more meaningful inquiry behaviors while reducing minimally productive behaviors. [ABSTRACT FROM AUTHOR]

Pandemic H1N1 2009 [A(H1N1)pdm09] variants associated with oseltamivir resistance have emerged with a histidine-to-tyrosine substitution in the neuraminidase(NA) at position 274 (H274Y). To determine whether the H274Y variant has increased virulence potential, A(H1N1)pdm09 virus, with or without the H274Y mutation, was adapted by serial lung-to-lung passages in mice. The mouse-adapted H274Y (maCA04H274Y) variants showed increased growth properties and virulence in vitro and in vivo while maintaining high NA inhibitor resistance. Interestingly, most maCA04H274Y and maCA04 viruses acquired common mutations in HA (S183P and D222G) and NP (D101G), while only maCA04H274Y viruses had consensus additional K153E mutation in the HA gene, suggesting a potential association with the H274Y substitution. Collectively, our findings highlight the potential emergence of A(H1N1)pdm09 drug-resistant variants with increased virulence and the need for rapid development of novel antiviral drugs.
