A convenient, commercial and simple process was developed for the synthesis of 1-[2,7-dichloro-9-(4-chlorobenzylidene)-9H-fluoren-4-yl]-2-(methylselanyl)ethanol. 2-(2,7-dichloro-9H-fluoren-4-yl)oxirane was converted to 1-(2,7-dichloro-9H-fluoren-4-yl)-2-(methylselanyl)ethanol in presence of dimethyl diselenide and sodium borohydride and consequently converted to 1-[2,7-dichloro-9-(4-chlorobenzylidene)-9H-fluoren-4-yl]-2- (methylselanyl)ethanol via Knoevenagel condensation with 4-chlorobenzaldehyde in presence of sodium hydroxide in reasonably good yield and purity. A series a novel of 1,2-disubstituted benzimidazole-5-carboxylates were prepared by the one-pot nitroreductive cyclization route using sodium dithionite, and its effect onN-debenzylation has been described. Different ethyl 4-(alkyl/arylamino)-3-nitrobenzoates were reacted with various aromatic/heteroaromatic aldehydes in dimethyl sulfoxide to yield the target benzimidazoles in good yield and purity. Wide varieties of aldehydes used for the cyclization were found to be susceptible under the reaction conditions. This method afforded the products within a very short reaction time and isolation was an easy workup procedure. Debenzylation resulting in the cleavage of theN-benzyl group was observed under experimental conditions with sodium dithionite without the aid of any cocatalyst and the presence of ethanol increased the extent of debenzylation. Several substituted thiazolo-[3,2-b]-1,2,4-triazole derivatives 4 were synthesized by a one-pot, three-component reaction of 3-(4-methylthiobenzyl)-1,2,4-triazole-5-thiol 3, substituted 5-aryl-furan-2-carboxaldehydes/substituted aromatic aldehydes, and monochloroacetic acid in acetic acid using acetic anhydride and anhydrous sodium acetate. Compound 3 was obtained from 4-methylthiophenyl acetic acid by esterification followed by hydrazinolysis. All structures of the newly synthesized compounds were elucidated by elemental analysis and spectral data. A series of 7-substituted arylidene-1,3-thiazolo[2,3-c]-1,2,4-triazine-4,6-diones were synthesized in an one-pot multicomponent reaction of 6-arylmethyl-3-mercapto-1,2,4-triazin-5-ones with substituted benzaldehydes and monochloroacetic acid in the presence of acetic anhydride, acetic acid, and sodium acetate. The structures of the new compounds were supported by IR, 1H NMR, MS, and analytical data. All the new compounds were tested for their antibacterial and antifungal activity. Arylidene thiazolo triazinediones with 1,3-benzodioxolo substituent at seventh and p-chlorophenyl or 2,4-dichlorophenyl substituents at third displayed good antibacterial and antifungal activity. A series of 7-substituted-3-t-butyl 4(H)-1,3,4-thiadiazolo(2,3-c)-1,2,4-triazine-4-one (3) have been synthesized by condensing 4-amino-6-t-butyl-3-mercapto-1,2,4-triazin-5(4H)-one (1) with substituted Arylfuroic acids (2) using POCl3 as a cyclizing agent. The newly synthesized compounds are characterized by elemental analysis, IR, 1H NMR, and mass spectral studies. The compounds were screened for their antibacterial and antifungal activities. A series of 4-arylidene-2-methyloxazol-5-ones 1 were condensed with thiosemi-carbazide to yield 6-arylmethyl-3-mercapto-1,2,4-triazin-5-ones 2. The resulting mercapto triazinones 2 were condensed with monochloroacetic acid and 5-aryl-furan-2-carboxaldehydes 3 in a one-pot three-component reaction in the presence of acetic anhydride, acetic acid, and sodium acetate to yield 4-substituted-5-oxo-7-(5-aryl-2-furfurylidene)-1,2,4-triazino[3,4-b]-thiazol-6-ones 4. Some of the newly synthesized compounds were tested for their antibacterial activity against Gram (+)ve and Gram (-)ve bacteria. The results of such studies are discussed in this paper. 3-Substituted-4-amino-5-mercapto-1,2,4-triazoles are versatile synthons for constructing various biologically active heterocycles. Their cyclization with carboxylic acids gives fused five-membered derivatives, whereas with α-bromoketones gives a six-membered heterocycle. In this article we report the synthesis of a series of 1,2,4-triazolo[3,4-b]thiadiazoles 5 starting from 4-amino-3-substituted-5-mercapto-1,2,4-triazoles 3 and fluorobenzoic acids 4 using phosphorous oxychloride as cyclizing agent.
