It is frequently recounted that shortly before Michelangelo died, the eighty-eight-year-old artist sat before a fire and burned many drawings. Critical examination reveals various motivations for such a destructive act and elicits suggestions for what Michelangelo might have burned. Mainly because of old age and significant changes in his artistic practice, Michelangelo made far fewer drawings in his final two decades than earlier in his career. Therefore, he may have destroyed less than is usually imagined and nothing that would have impaired the completion of his current projects, most important of which was New Saint Peter's. With the development of nanotechnology, there is a tremendous growth of the application of nanomaterials, which increases the risk of human exposure to these nanomaterials through inhalation, ingestion, and dermal penetration. Among different types of nanoparticles, single-walled carbon nanotubes (SWCNT) with extremely small size (1 nm in diameter) exhibit extraordinary properties and offer possibilities to create materials with astounding features. Since the release of nanoparticles in an enclosed environment is of great concern, a study of possible genotoxic effects is important. Our previous data showed that pharyngeal aspiration of SWCNT elicited pulmonary effects in C57BL/6 mice that was promoted by a robust, acute inflammatory reaction with early onset resulting in progressive interstitial fibrogenic response and the formation of granulomas. In the present study, the genotoxic potential of SWCNT was evaluated in vitro. The genotoxic effects of nanoparticles were examined using three different test systems: the comet assay and micronucleus (MN) test in a lung fibroblast (V79) cell line, and the Salmonella gene mutation assay in strains YG1024/YG1029. Cytotoxicity tests showed loss of viability in a concentration- and time-dependent manner after exposure of cells to SWCNT. Results from the comet assay demonstrated the induction of DNA damage after only 3 h of incubation with 96 μg/cm2 of SWCNT. The MN test indicated some but not significant micronucleus induction by SWCNT in the V79 cell line at the highest concentrations tested. Respirable quartz and kaolin particles were treated with fluorescent-labeled phospholipids to model contact of fibrogenic and nonfibrogenic particles with pulmonary surfactant in the alveolar regions of the lung. Particles were used to challenge rat pulmonary macrophages in vitro at times from 1 d to 10 d. The objective was to develop a quantitative method to track surfactant components that adsorb to respirable particles in the lung or inside cells. Confocal laser scanning microscopy was used to image and quantify surfactant remaining on particles internalized by cells. Results indicate that the fluorescent label is removed from quartz particles quickly, with the fluorescence intensity less than 15% of initial value at 3 d, and about 5% at 10 d. In contrast, the kaolin particle-associated fluorescence was still ~39% of initial intensity at 3 d, and 10–15% at 10 d. Unchallenged cells showed a background of ~5%, and noninternalized particles did not exhibit any loss of fluorescence over the 10-d exposure. The results indicate the method may be useful in label-removal rate studies of respirable particles in vitro, with some cautions and limitations. Diesel exhaust particulate material (DPM) was assayed for induction of chromosomal aberrations (CA), micronucleus (MN) formation, and 6-thioguanine-resistant (TG r ) gene mutation in V79 cells as a dispersion in dipalmitoyl phosphatidylcholine (DPPC) in physiological saline, a simulated pulmonary surfactant. Filter-collected automobile DPM provided for the study was not organic solvent extracted, but was directly mixed into DPPC in saline dispersion as a model of pulmonary surfactant conditioning of a soot particle depositing in a lung alveolus. A statistically significant difference was found between treated and control groups at all concentrations tested in a CA assay. Assay for MN induction also gave a positive response: Above 50 μg/ml, the frequencies of micronucleated cells (MNC) were about 2 times higher than those in the control group. The forward gene mutation assay did not show a positive response when cells were treated with up to 136 μg DPM/ml for 24 h, as dispersion in DPPC in saline. Some comparison assays were run on direct dispersions of the DPM into dimethyl sulfoxide, with results equivalent to those seen with a DPPC–saline preparation: DPM in dimethyl sulfoxide (DMSO) was positive for MN induction but was negative for forward gene mutation in V79 cells. The positive clastogenicity results are consistent with other studies of DPM dispersed into DPPC–saline surfactant that have shown activity in mammalian cells for sister chromatid exchange, unscheduled DNA synthesis, and MN induction. In this report, we elucidate the structure of a small molecule liquid crystal/polymer interface using specular neutron reflectivity. More specifically, we have examined the interfacial transition zone width of a small molecule liquid crystal/polymer sample as a function of increasing temperature. The interface between a thin film (≈ 1000 Å thick) of the liquid crystal 4′-n-octyl-4-cyanobiphenyl (8CB) and a thin film (≈ 800 Å thick) of deuterated poly(methyl methacrylate) (d-PMMA) is broad and broadens with increasing temperature; it was also observed that the thin film geometry influences the mixing behaviour of the 8CB/d-PMMA system. These results may have implications for current theories of liquid crystal display devices formed by the phase separation of liquid crystal polymer mixtures. [ABSTRACT FROM AUTHOR]

Lung surfactant serves as a protective coating when adsorbed on particle surfaces, so its removal or rate of removal in vivo may affect expression of mineral cytotoxicity. Removal of phospholipid surfactant components from the surface of mineral particles ingested by alveolar macrophages (AM) was measured using fluorescence microscopy. Dipalmitoylphosphatidylcholine with a fluorescent label (BODIPY ) substituted for C1-C4 on the second acyl chain (DPPC*), was mixed with dioleoylphosphatidylcholine (DOPC) to coat respirable quartz and kaolin particles. Fluorescence from quartz or kaolin particles of 3-4, 5-6 and 8-9 µm size decreased in intensity with increasing ratios of DOPC/DPPC* for the same DOPC concentration of 0.4 mg/ml. There was a direct correlation between fluorescence and residual phospholipid surfactant remaining on particles using phospholipase A2 (PLA2) digestion in a cell-free system, indicating that the presence of the fluorophore on DPPC did not hinder enzymatic recognition. Lavaged primary AM obtained from male Fischer rats were challenged in vitro with DOPC/DPPC* (10:1 mol:mol) coated particles at 50 µg particles/10[sup 6] cells. In contrast to the biexponential response seen in cell-free experiments, the rate of fluorescence decay from ingested coated quartz or kaolin particles over 7 days was monoexponential, with the same t[sub 1/2] (41 h) for each dust. This study suggests that the rate of phagolysosomal digestion and removal of the adsorbed surfactant is not a determinant of the different mineral-specific pathogenicities or toxicities of quartz and kaolin, although residual fluorescence remained on particles even after 7-8 days. Chrysotile fibers (NIEHS intermediate length) were treated with ultrapure HCl to alter the fiber surface chemistry without substantially changing fiber morphology or dimensions. The objective of the study was to determine whether fiber surface chemistry is an important variable in fiber genotoxicity in vitro. The modified fibers, along with native chrysotile fibers, were used to challenge Chinese hamster lung fibroblasts (V79) in vitro using the micronucleus induction genotoxicity assay. Fiber dimensions were assessed using scanning electron microscopy by measuring the distribution of fiber lengths in 3 length ranges: less than 3 mum, 3-10 mum, and greater than 10 mum. For both treated and native fiber samples, 500 fibers were examined. Results indicate that acid-treated fibers were about 20% shorter than untreated chrysotile. Surface chemistry alterations were verified by zeta-potential reversal, x-ray photoelectron spectroscopy (XPS), and scanning electron microscopy/energy-dispersive x-ray spectroscopy (SEM-EDS) elemental analysis. Scanning Auger spectrometry indicated the presence of Mg, O, and Si in both treated and native chrysotile samples, which confirmed the surface purity of both fiber samples. Both XPS and SEM-EDS analysis demonstrated substantial depletion of Mg from fiber surfaces. Results of the micronucleus assay showed a positive concentration-related response for both samples, with toxicity evident only at the highest concentration. No significant difference was found for the treated and untreated chrysotile samples. These results indicate that the surface chemistry is not an important variable in the in vitro genotoxicity of chrysotile asbestos in V79 cells as detected by the micronucleus assay under the conditions used in this study, and support a model of chemically nonspecific chromosomal and spindle damage effects. Results from a study directed towards identifying and measuring the mutagenicity of diesel exhaust particulate matter involved in mining exposures, especially those which may synergistically affect genotoxic hazard, are presented in this paper. Particulate matter emissions from a diesel engine, representative of the ones found in underground mines, were sampled and assayed to determine the genotoxic potential as a function of engine operating conditions. Diesel exhaust was diluted in a multi‐tube mini‐dilution tunnel and the particulate matter was collected on 70 mm fluorocarbon coated glass fiber filters as well as on 8” × 10” hi‐volume filters. A six node steady state duty cycle was used to relate engine operating conditions to the mutagenicity potential. Protocols developed for using a primary component of pulmonary surfactant as a medium for collecting and applying respirable particulate material to cells for mutagenicity testing offer a true representation of in‐vivo delivery of such respired aerosols to pulmonary cells and the genetic material within. The surfactant dispersion technique is discussed in this paper. Methods have been developed to sample and assay the diesel soot and diesel soot/mineral dust complexes which would not compromise the surface and physical properties of the particulate material.
