Ascidian embryos develop with a fixed cell lineage into simple tadpoles. Their lineage is almost perfectly conserved, even between the evolutionarily distant species Halocynthia roretzi and Ciona intestinalis,which show no detectable sequence conservation in the non-coding regions of studied orthologous genes. To address how a common developmental program can be maintained without detectable cis-regulatory sequence conservation, we compared in both species the regulation of Otx, a gene with a shared complex expression pattern. We found that in Halocynthia, the regulatory logic is based on the use of very simple cell line-specific regulatory modules, the activities of which are conserved,in most cases, in the Ciona embryo. The activity of each of these enhancer modules relies on the conservation of a few repeated crucial binding sites for transcriptional activators, without obvious constraints on their precise number, order or orientation, or on the surrounding sequences. We propose that a combination of simplicity and degeneracy allows the conservation of the regulatory logic, despite drastic sequence divergence. The regulation of Otx in the anterior endoderm by Lhx and Fox factors may even be conserved with vertebrates.

Two axial structures, a neural tube and a notochord, are key structures in the chordate body plan and in understanding the origin of chordates. To expand our knowledge on mechanisms of development of the neural tube in lower chordates, we have undertaken isolation and characterization ofHrzicN, a new member of the Zic family gene of the ascidian,Halocynthia roretzi. HrzicN expression was detected by whole-mount in situ hybridization in all neural tube precursors, all notochord precursors,anterior mesenchyme precursors and a part of the primary muscle precursors. Expression of HrzicN in a- and b-line neural tube precursors was detected from early gastrula stage to the neural plate stage, while expression in other lineages was observed between the 32-cell and the 110-cell stages.HrzicN function was investigated by disturbing translation using a morpholino antisense oligonucleotide. Embryos injected with HrzicNmorpholino (`HrzicN knockdown embryos') exhibited failure of neurulation and tail elongation, and developed into larvae without a neural tube and notochord. Analysis of neural marker gene expression inHrzicN knockdown embryos revealed that HrzicN plays critical roles in distinct steps of neural tube formation in the a-line- and A-line precursors. In particular HrzicN is required for early specification of the neural tube fate in A-line precursors. Involvement of HrzicNin the neural tube development was also suggested by an overexpression experiment. However, analysis of mesodermal marker gene expression inHrzicN knockdown embryos revealed unexpected roles of this gene in the development of mesodermal tissues. HrzicN knockdown led to loss of HrBra (Halocynthia roretzi Brachyury) expression in all of the notochord precursors, which may be the cause for notochord deficiency.Hrsna (Halocynthia roretzi snail) expression was also lost from all the notochord and anterior mesenchyme precurosrs. By contrast,expression of Hrsna and the actin gene was unchanged in the primary muscle precursors. These results suggest that HrzicN is responsible for specification of the notochord and anterior mesenchyme. Finally,regulation of HrzicN expression by FGF-like signaling was investigated, which has been shown to be involved in induction of the a- and b-line neural tube, the notochord and the mesenchyme cells inHalocynthia embryos. Using an inhibitor of FGF-like signaling, we showed that HrzicN expression in the a- and b-line neural tube, but not in the A-line lineage and mesodermal lineage, depends on FGF-like signaling. Based on these data, we discussed roles of HrzicN as a key gene in the development of the neural tube and the notochord.

Ascidian embryogenesis shares several developmental features with vertebrates. Thus, it is presumed that some molecular mechanisms that are critical for vertebrate development may also act in the early development of ascidians. Here, we investigated expression of the ascidian labial group Hox gene HrHox-1 in the development of Halo-cynthia roretzi. HrHox-1 showed a spatially restricted expression pattern along the anterior-posterior axis, which is remarkably similar to that of the vertebrate gene, Hoxb-1. The expression of HrHox-1, however, was exclusively in tissues of ectoderm origin unlike its vertebrate counterpart. Exposure of the embryos to 10−6 M all-trans retinoic acid induced a larval phenotype with elimination of the anteriormost structures, the papillae. In this phenotype, the level of HrHox-1 expression was enhanced and ectopic expression was observed at the anterior terminal epidermis where the papillae are otherwise formed. These observations suggest that there are some conserved mechanisms in the spatial regulation of expression of labial group genes in embryogenesis of ascidians and vertebrates.

We have isolated a novel ascidian homeobox gene, designated AHoxl, by screening the genomic DNA of Halocynthia roretzi with the Bombyx mori Antennapedia type homeobox as a probe. The AHoxl gene encodes a protein that consists of 741 amino acids. The homeobox of AHoxl is interrupted by 2 introns each of which is about 300 bp in length and it shows about 70 % similarity at a deduced amino acid level to that of Drosophila H2.0. This suggests that AHoxl is one of the most diverged homeobox genes so far characterized. Northern blot hybridization with an AHoxl probe showed the presence of single transcripts approximately 2.8 kb in length in larvae, juveniles and some adult tissues. The expression of AHoxl is scarcely detected during the course of early development but it increases to a moderate level at the larval stage. After metamorphosis, the level of AHoxl expression increases as development proceeds. In situ hybridization to the juvenile 7 days after metamorphosis showed that the site of AHoxl expression is the epithelium of digestive tract. Among the adult tissues examined, digestive tract, digestive gland and coelomic cells were the major sites of the expression of AHoxl. In gonad, body wall muscle and pharyngeal epithelium, the expression of AHoxl is relatively weak. These results suggest that AHoxl is primarily expressed in the tissues of endodermal origin and that the gene expression may be associated with differentiation of the endodermal tissues.
