Atualmente, sabe-se que a aplicação regular de filtros solares constitui umas das principais medidas na prevenção dos danos induzidos pela radiação ultravioleta (UV). No entanto, o uso de associações fotoinstáveis pode reduzir a eficácia fotoprotetora e levar à formação de intermediários reativos, os quais podem apresentar potencial em causar fotoirritação, fotoalergia e fotogenotoxicidade na pele. Além disso, tem-se observado uma tendência na adição de substâncias antioxidantes aos filtros solares convencionais, a fim de prevenir os danos oxidativos mediados pelas espécies reativas do oxigênio (EROs), sendo que a quercetina (QT) é uma opção interessante. No entanto, sua limitada absorção cutânea pode levar à falta de eficácia como agente fotoprotetor, o que leva a necessidade da utilização de sistemas de liberação, a fim de melhorar sua penetração na pele. Portanto, este trabalho teve como objetivo avaliar a segurança e eficácia de formulações fotoprotetoras associadas a carreadores lipídicos nanoestruturados contendo quercetina (CLN-QT) em cultura celular, em modelo de pele humana reconstituída e em humanos. Primeiramente, quatro formulações fotoprotetoras (F1, F2, F3 e F4), contendo diferentes associações dos filtros solares dietilamino hidroxibenzoil hexil benzoato (DHHB), metoxicinamato de etilexila (MTX), avobenzona (AVO) e bemotrizinol (BMTZ), foram submetidas aos estudos de fotoestabilidade, fotorreatividade, geração de EROs (por meio da sonda DCFH2-DA), fototoxicidade em cultura de fibroblastos 3T3 e fotogenotoxicidade, por meio do ensaio do cometa. Uma vez que a associação A2 (contendo DHHB e MTX) apresentou melhores resultados quanto à fotoestabilidade e fotorreatividade em relação às associações A1 (contendo MTX, AVO) e A3 (contendo MTX, AVO e 3% BMTZ) ela foi associada aos CLN-QT e estudos posteriores foram realizados. Além disso, a associação A2 não aumentou os níveis de EROs intracelular após irradiação UVA e também não foi considerada fototóxica. No presente estudo foi observado que a inclusão de ensaios biológicos complementares ao estudo de fotoestabilidade forneceu informações mais relevantes e completas em relação ao ensaio de fotodegradação realizado de forma isolada para a seleção da associação mais adequada. Além disso, houve correlação entre a produção de EROs intracelular e fotorreatividade, portanto, sugerimos a inclusão desse ensaio na estratégia de avaliação da fotoestabilidade. Os resultados do estudo de avaliação da segurança e eficácia dos CLN-QT associados aos filtros solares demonstraram que os mesmos não apresentaram citotoxicidade em queratinócitos humanos primários e não foram considerados fototóxicos no modelo de pele humana reconstituída. Além disso, os CLN-QT associados aos filtros solares, apesar de não terem alterado o FPS (fator de proteção solar) em humanos (proteção contra eritema), reduziram significativamente a geração de EROs intracelular induzidos pela irradiação UVA (cerca de 30%) em relação aos filtros solares isolados e desta forma, complementaram a eficácia dos fotoprotetores. Nowadays it is well known that the regular application of UV filters is one of the main strategies in the prevention of damage induced by ultraviolet radiation (UV). However, the use of photounstable combinations can reduce photoprotective efficacy and lead to the formation of reactive intermediates, which may have potential to cause photoirritation, photoallergy and photogenotoxicity on the skin. In addition, there has been a tendency to combine antioxidants to conventional UV filters in order to prevent oxidative damage mediated by reactive oxygen species (ROS) and quercetin (QT) is an interesting option. However, its limited cutaneous absorption can lead to a lack of photoprotective efficacy, which leads to necessity of the use of delivery systems in order to improve its penetration into the skin. Therefore, this study aimed to evaluate safety and efficacy of photoprotective formulations containing quercetin in nanostructured lipid carriers (CLN-QT) in cell culture, reconstructed skin model and humans. Firstly, four photoprotective formulations (F1, F2, F3 and F4), containing different combinations of diethylamino hydroxybenzoyl hexyl benzoate (DHHB), ethylhexyl methoxycinnamate (MTX), avobenzone (AVO) and bemotrizinol (BMTZ) were submitted to the studies of photostability, photoreactivity, ROS generation (using DCFH2-DA probe), phototoxicity in 3T3 fibroblast culture and photogenotoxicity by comet assay. The combination C2 (containing DHHB and MTX) presented better results regarding photostability and photoreactivity when compared to combinations C1 (containing MTX and AVO) and C3 (containing MTX, AVO and 3% BMTZ), so it was combined to CLN-QT and subsequent studies were performed. Moreover, combination C2 did not increase intracellular ROS levels after UVA irradiation and it was not considered phototoxic. In the present study, it was observed that the inclusion of complementary biological assays to the photostability study provided more relevant results when compared to the photostability as a stand-alone assay for the selection of the best combination. In addition, it was demonstrated a good correlation between intracellular ROS production and photoreactivity, so we suggest the inclusion of intracellular ROS production in the photostability evaluation strategy. The safety and efficacy results demonstrated that UV filters + CLN-QT did not present cytotoxicity in primary human keratinocytes and were not considered phototoxic in reconstructed skin model. The CLN-QT + UV filters did not alter SPF (sun protection factor) in humans (protection against erythema), however they reduced intracellular ROS generation after UVA irradiation (about 30%), when compared to UV filters alone and thus complemented the efficacy of photoprotective formulations.

Algumas macroalgas vermelhas produzem aminoácidos do tipo micosporina (MAAs, do inglês mycosporine like-aminoacids), que são capazes de absorver a radiação UV e atuar como antioxidantes. No entanto, o uso dessas substâncias ainda foi pouco explorado devido ao difícil processo para isolá-las, então o uso de um extrato contendo altas concentrações de MAAs pode ser mais vantajoso para fins comerciais. Portanto, no presente trabalho foram avaliados o potencial fotoprotetor e antioxidante de extratos de macroalgas vermelhas da Antártica, Curdiea racovitzae, Iridaea cordata e Myriogramme mangini, bem como a sua toxicidade para o uso em produtos cosméticos. A caracterização química foi realizada por análise qualitativa e quantitativa de MAAs, teor de polifenóis e flavonoides totais. As propriedades fotoprotetoras foram caracterizadas por espectrofotometria no UV/VIS e fotoestabilidade. A capacidade antioxidante foi avaliada por ensaios de captura de radicais DPPH e ânion superóxido. Os efeitos fotoprotetores UVB e UVA foram avaliados em queratinócitos da linhagem HaCaT, pela viabilidade celular após irradiação com UVB e quantificação intracelular de espécies reativas de oxigênio (EROs) induzido por UVA, respectivamente. A toxicidade foi estimada pela citotoxicidade e fototoxicidade em fibroblastos 3T3 BALB/c, e pela avaliação da irritação ocular pelo ensaio de HET-CAM. Além disso, foram avaliados os efeitos do CRHE na fotoestabilidade e fotoreativadade da combinação de filtros avobenzona (AVO) e metoxicinamato de etilhexila (MTX). A análise qualitativa dos MAAs mostrou que os extratos das algas C. racovitazae e I. cordata (CRHE e ICPE) possuem majoritariamente palitina seguido por asterina-330, enquanto os extratos da M. mangini (MMCE) são constituídos apenas por chinorina. O CRHE apresentou alta concentração de MAAs (150 ?g de MAAs/mg de extrato), sendo o extrato com maiores concentrações de MAAs dentre os estudados. No entanto, o MMCE foi o extrato com os maiores teores de polifenóis totais e flavonoides, sendo que 28% do seu teor de polifenóis correspondem a flavonoides, o que provavelmente ocorreu devido aos diferentes processos de extração. Os extratos das algas vermelhas absorveram na região do UVB e UVA-2 e foram considerados fotoestavéis após a irradiação com UVA. Nos ensaios antioxidantes, o MMCE apresentou a melhor atividade antioxidante entre os extratos avaliados, que pode estar correlacionada com suas concentrações de polifenóis totais. O CRHE (1 e 0,5 mg/mL), o ICPE (1 e 0,5 mg/mL) e o MMCE (1 mg/mL) aumentaram significativamente a viabilidade celular dos queratinocitos irradiados com UVB. O CRHE (1 e 0,5 mg/mL) conferiu proteção UVB similar a benzofenona-3, filtro UVB e UVA-2, e melhor do que o Helioguard (5% e 3%). O MMCE reduziu significativamente a geração de EROS induzidos por UVA em todas as concentrações testadas, ainda o MMCE (100 ?g/mL) provocou maior redução de EROS do que a quercetina (10 e 100 ?g/ mL) e o Helioguard (3%). O CREB foi capaz também de reduzir a geração de EROs, quando utilizado a 100 e 50 ?g/mL. Os extratos das algas vermelhas foram considerados não citotóxicos, não fototoxicos e não irritantes. A adição de CRHE à combinação AVO e MTX aumentou a sua fotoestabilidade e reduziu a sua fotoreatividade. Portanto, os extratos das algas vermelhas são bons candidatos para serem adicionados a formulações antienvelhecimento e fotoprotetoras para proteger contra o estresse oxidativo, melhorar o espectro de absorção dos filtros UV e fotoestabilizar filtros ou combinações instáveis. Some red macroalgae synthesize mycosporine-like amino acids (MAAs), which are able to absorb UV radiation and act as antioxidants. However, the use of these substances was few explored due to the difficult process to isolate them, thus the use of an extract containing high levels of MAAs might be most advantageous for commercial purposes. Therefore, the photoprotective and antioxidant potential antarctic red macroalgae extracts, Iridaea cordata (ICPE), Curdiea racovitzae (CRHE) and Myriogramme mangini (MMCE), as well as their toxicity for use in cosmetic products, were evaluated. The chemical characterization was performed by qualitative and quantitative analysis of MAAs, the total content of polyphenols and flavonoids. The photoprotective properties were characterized by spectrophotometry in the UV/VIS and photostability. The antioxidant capacity was evaluated by DPPH and superoxide anion scavenging assays. Then, their UVB and UVA photoprotective effects were evaluated in HaCaT keratinocytes by cell viability after UVB irradiation and quantification of intracellular reactive oxygen species (ROS) induced by UVA, respectively. The toxicity was estimated by the cytotoxicity and phototoxicity in fibroblasts 3T3 BALB/c, and ocular irritation using the HET-CAM test. Moreover, the effects of the extract of CRHE on the photostability and photoreactivity of the combination of avobenzone (AVO) and ethylhexylmethoxycinnamate (MTX) filters were evaluated. The qualitative analysis of the MAAs showed that the extracts from the algae C. racovitazae and I. cordata (CRHE and ICPE) had mostly palythine followed by asterina-330, whereas the M. mangini extracts (MMCE) is practically constituted just for shinorine. CRHE had a high concentration of MAAs (150 ?g MAAs/mg extract), being the extract with the highest MAAs concentration among the studied. However, the MMCE was the extract with the highest content of total polyphenols and flavonoids, which 28% of polyphenol content corresponding to flavonoids, which was probably due to the different extraction processes. The red algae extracts absorved in the UVB and UVA-2 region and were considered photostable after UVA irradiation. In the antioxidant assays, the MMCE presented the best antioxidant activity among the evaluated extracts, which can be correlated with its total content of polyphenols. CRHE (1 and 0.5 mg/mL), ICPE (1 and 0.5 mg/mL) and MMCE (1 mg/mL) increased significantly the cell viability of UVB-irradiated keratinocytes. CRHE (1 and 0.5 mg/mL) conferred UVB protection similar to benzophenone-3, UVB and UVA-2, filter and better than Helioguard (5% and 3%). The MMCE significantly reduced the generation of ROS induced by UVA at all tested concentrations, further MMCE (100 ?g/mL) lead to a greater EROS reduction than quercetin (10 and 100 ?g/mL) and Helioguard (3%). CREB was also able to reduce the generation of EROs, when used at 100 and 50 ?g/mL. The red algae extracts were considered non-cytotoxic, non-phototoxic and non-irritant. The adition of CRHE to the combination of AVO and MTX increased their photostability and reduced their photoreactivity. Therefore, the red algae extracts are good candidates to be added in anti-aging and photoprotective formulations to protect against oxidative stress, improve the absorption spectrum of UV filters and photostabilize unstable filters or combinations

Tendo em vista que a radiação ultravioleta A (UVA) e a luz visível (VIS) apresentam alto potencial para gerar oxigênio singleto (1O2) e provocar estresse oxidativo, é importante buscar substâncias capazes de prevenir ou minimizar o impacto desse evento sobre a pele, já que atualmente, há poucos filtros solares orgânicos no mercado que oferecem ampla proteção contra a UVA I e não há filtros orgânicos que ofereçam proteção contra a luz VIS. Estudos da literatura indicam que alguns antioxidantes, tais como a apigenina (API), a crisina (CRI) e o beta-caroteno (BTC), podem neutralizar o 1O2 e apresentam potencial de uso na fotoproteção. Deste modo, o objetivo do presente projeto foi avaliar a segurança da API, da CRI e do BTC, por meio dos ensaios de fotoestabilidade, fotorreatividade e fototoxicidade, e também sua eficácia, através de ensaios para determinar a proteção contra a morte celular induzida pela fotossensibilização da melanina, que gera 1O2, e a proteção de membrana contra danos fotooxidativos. Inicialmente, foi avaliada a fotoestabilidade da API, da CRI e do BTC quando submetidos à radiação UVA e luz VIS, por meio de CLAE. A fototoxicidade das substâncias em estudo frente à radiação UVA e luz VIS, empregando-se cultura de fibroblastos (Balb 3T3 clone A31), de melanócitos (Melan-a) e de melanoma (B16F10) também foi realizada. Para avaliar a eficácia, o efeito da API, da CRI e do BTC sobre a viabilidade das células B16F10 com níveis basais de melanina e após indução da melanogênese, bem como seu efeito sobre a proteção de membrana em modelo mimético de lipossomas, foram avaliados frente à luz VIS. Os resultados do estudo de fotoestabilidade mostraram que os antioxidantes, isolados ou em associação, foram considerados fotoestáveis em ambas as faixas de radiação avaliadas (UVA/VIS e VIS). No estudo de fotorreatividade, a API, a CRI e a associação (API+CRI+BTC) se mostraram fracamente fotorreativas após exposição ao UVA/VIS. No entanto, na presença de VIS, nenhum dos compostos apresentou fotorreatividade. No estudo de fototoxicidade, foi observado potencial fototóxico após irradiação por UVA/VIS para a CRI, BTC e a associação (API+CRI+BTC) nas três linhagens celulares avaliadas. Já a API, não apresentou potencial fototóxico apenas no ensaio realizado com fibroblastos. Na faixa da luz VIS, o BTC apresentou potencial fototóxico nas três linhagens celulares avaliadas; entretanto, para os demais antioxidantes (API e CRI) e sua associação (API+CRI+BTC) houve variação na resposta entre as diferentes linhagens. Os resultados dos estudos para avaliação da eficácia mostraram que os antioxidantes foram capazes de proteger as células contra morte induzida pela fotossensibilização da melanina, sendo que o efeito da API foi superior ao da CRI e do BTC. No estudo de proteção de membrana apenas a API foi capaz de conferir proteção contra os danos fotooxidativos induzidos pela luz VIS. A investigação do potencial de uso da API, da CRI e do BTC na proteção contra a radiação UVA e luz VIS mostrou que o uso de tais antioxidantes e da sua combinação visando a fotoproteção da pele é mais seguro na faixa da luz VIS. Além disso, concluiu-se que API é o mais promissor entre os antioxidantes estudados em relação à segurança e à eficácia, e apresenta elevado potencial de uso na proteção contra a luz VIS. Considering that ultraviolet A (UVA) radiation and visible light (VIS) present high potential to generate singlet oxygen (1O2) and cause oxidative stress, it is important to search for substances that are able to prevent or minimize the impact of such event on the skin, since currently, there are few organic UV-filters on the market that offer broad protection against UVA I and there is no organic UV-filter which provide VIS light protection. Previous studies showed that some antioxidants, such as apigenin (API), chrysin (CRI) and beta-carotene (BTC) can neutralize 1O2 and exhibit potential for use in photoprotection. Therefore, the aim of this study was to evaluate the safety of API, CRI and BTC, by performing photostability, photoreactivity and phototoxicity assays, and also its efficacy, through the determination of the protection against cell death induced by melanin photosensitization, which generates 1O2, and also the assessment of membrane protection against photooxidative damage. Firstly, API, CRI and BTC photostability was evaluated after exposure to UVA and VIS light by using HPLC. The phototoxicity of the antioxidants subjected to UVA and VIS light, using cultured fibroblasts (Balb 3T3 clone A31), melanocytes (melan-a) and melanoma (B16F10) was also performed. In order to assess efficacy, API, CRI and BTC effect on the viability of B16F10 cells with melanin basal levels and after melanogenesis induction as well as their effect on membrane protection using mimetic liposome model were evaluated after VIS light exposure. Results of photostability studies showed that the antioxidants, isolated or combined, were considered photostable in both ranges (UVA/VIS and VIS). In the photoreactivity study API, CRI and the combination (API + CRI + BTC) were considered weakly photoreactive after UVA/VIS exposure. However, in the presence of VIS light, none of the compounds exhibited photoreactivity. In the phototoxicity study, phototoxic potential was observed for CRI, BTC and the combination (API + CRI + BTC) after UV/VIS irradiation in the three studied cell lines. API showed no phototoxic potential only in the assay performed with fibroblasts. In the VIS range, BTC showed phototoxic potential in the three cell lines; however, for the other antioxidants (API and CRI) and their combination (API + CRI + BTC) there was variance in response among different cell lines. Efficacy studies showed that the studied antioxidants were able to protect cells from death induced by melanin photosensitization, and API effect was superior to that of CRI and BTC. In membrane protection study, only API provided protection against VIS photooxidative induced damage. The investigation of the potential use of API, CRI and BTC for protection against UVA and VIS light showed that the use of such antioxidants and their combination aiming skin photoprotection is safer in the VIS range. Furthermore, it was concluded that API is the most promising antioxidant regarding safety and efficacy and presents high potential for use in protection against VIS light.

Melanina é um polímero constituído por uma grande heterogeneidade de monômeros tendo como característica comum a presença de grupos indóis. Por outro lado, a eumelanina produzida pela oxidação enzimática da tirosina é um polímero mais simples constituído principalmente de monômeros 5,6-dihidroxindol (DHI) e de indol-5,6-quinona (IQ). Tirosinase é a enzima chave na produção de melanina, sendo que a sua atividade cinética é medida em função da formação do intermediário dopacroma. Nanopartículas (NPs) de sílica são partículas nanométricas compostas de oxido de silício e são obtidas pelo processo sol-gel desenvolvido por Stöber de hidrólise e condensação de tetraetilortosilicato (TEOS), usando etanol como solvente em meio alcalino. As NPs foram funcionalizadas com 3-Aminopropiltrietoxissilano (ATPES) e depois com glutaraldeído. Este último permitiu a imobilização da tirosinase na superfície da sílica. Caracterizamos as NPs antes e após a reação da enzima, a atividade catalítica da enzima ligada à NP e o mecanismos de formação de melanina na superfície da sílica. As NPs foram caracterizadas por espectrofotometria de absorção e de reflectância, termogravimetria e microscopia eletrônica. A síntese da NP de sílica retornou partículas esféricas com 55nm de diâmetro e a funcionalização da partícula mostrou modificar eficientemente a sua superfície. A imobilização da tirosinase por ligação covalente foi de 99,5% contra 0,5% da adsorção física. A atividade da tirosinase foi caracterizada pela formação de dopacroma. O Km da enzima imobilizada não sofreu alteração em comparação com a tirosinase livre, mas a eficiência catalítica - que considera a eficiência recuperada - foi de apenas 1/3 para a enzima ligada covalentemente, significando que 2/3 das enzimas ligadas não estão ativas. Obtivemos NPs revestidas com melanina a partir de oxidação de tirosina solubilizada em duas preparações: NP com tirosinase ligada covalentemente na superfície e NP funcionalizada com glutaraldeido dispersa em solução de DHI e IQ. O revestimento de melanina foi na forma de um filme fino com espessura ~1,9nm, conferindo perfil de absorção luminosa equivalente ao da própria melanina. Mostramos que o mecanismo de polimerização passa pela oxidação da tirosina pela tirosinase, que gera intermediários oxidados (principalmente DHI e IQ) que vão para solução (mesmo quando a tirosinase está ligada covalentemente na sílica). Estes intermediários ligam-se ao glutaraldeido e a superfície da sílica passa a funcionar como ambiente de polimerização da melanina. Melanin is a polymer consisting of a large heterogeneity of monomers having as a common feature the presence of indole groups. Contrarily, eumelanin produced by enzymatic oxidation of tyrosine is a simpler polymer consisting mainly of 5,6-dihidroxindol (DHI) and indole-5,6-quinone (IQ) monomers. Tyrosinase is the key enzyme in melanin production, and its kinetic activity is measured by the formation of the intermediate dopacroma. Nanoparticles (NPs) are made of silica nanoparticles of silicon oxide and are obtained by sol-gel method developed by Stöber of hydrolysis and condensation of tetraethylorthosilicate (TEOS), using ethanol as solvent in an alkaline medium. NPs were functionalized with 3-Aminopropyltriethoxysilane (ATPES) and then with glutaraldehyde. The latter allows the immobilization of tyrosinase on the silica surface. We characterized NPs before and after the reaction of the enzyme, the catalytic activity of the enzyme bound to the NP and melanin-forming mechanisms on the silica surface. NPs were characterized by absorption spectrophotometry and reflectance, electron microscopy and thermogravimetric analysis. The synthesis of silica NP returned spherical particles of 55nm diameter and particle functionalization showed efficiently modify its surface. The immobilization of tyrosinase by covalent bond was 99.5% versus 0.5% by physical adsorption. The activity of tyrosinase was characterized by the formation of dopacroma. The Km of the immobilized enzyme did not change compared to the free tyrosinase, but the catalytic efficiency - considering the recovered efficiently - was only 1/3 for the enzyme covalently bound, meaning that 2/3 of the enzymes are not connected active. We obtained melanin coated NPs from tyrosine oxidation in two preparations: NP with covalently bound tyrosinase in the NP surface and NP functionalized with glutaraldehyde dispersed in DHI and IQ solution. The melanin coating was in the form of a thin film with the thickness of ~ 1,9 nm, giving light absorption profile equivalent to that of melanin itself. We showed that the polymerization mechanism involves the oxidation of tyrosine by tyrosinase, which generates oxidized intermediates (especially DHI and lQ) that go into solution (even when tyrosinase is covalently bound to the silica). These intermediates bind the glutaraldehyde and the surface of the silica begins to function as an environment for melanin polymerization.

Atualmente, devido ao conhecimento dos danos causados pela exposição da pele à radiação UV, existe uma tendência em utilizar, além dos filtros solares convencionais, substâncias naturais com potencial antioxidante. Entretanto, o uso de associações fotoinstáveis, pode, além de comprometer a capacidade fotoprotetora, levar à formação de intermediários reativos que podem ocasionar dermatites de contato e reações fototóxicas na pele. Dessa forma, o objetivo deste trabalho foi avaliar a fotoestabilidade e fototoxicidade de formulações fotoprotetoras contendo diferentes associações de filtros solares acrescidas ou não dos polifenóis mangiferina e naringenina. Para o estudo de fotoestabilidade, amostras das formulações foram aplicadas em lâminas de vidro e expostas à radiação UVA e, a seguir, foram feitas análises por cromatografia líquida de alta eficiência (CLAE), para dosagem do teor de filtros solares e antioxidantes, e por espectrofotometria, para determinação da razão UVA/UVB. A fototoxicidade foi avaliada por meio do uso de cultura de fibroblastos 3T3, submetida ou não à radiação UVA, para determinação da viabilidade celular. Os resultados de fotoestabilidade por CLAE demonstraram que a naringenina, o metoxicinamato de etilexila e avobenzona foram considerados fotoinstáveis. Além disso, foi demonstrado que a adição de mangiferina à associação contendo avobenzona e naringenina, proporcionou um aumento da fotoestabilidade das mesmas. A presença do dietilamino hidroxibenzoil hexil benzoato (DHHB) na formulação contendo avobenzona e naringenina promoveu melhora da fotoestabilidade da naringenina, no entanto não houve melhora da fotoestabilidade da avobenzona. As análises espectrofotométricas demonstraram que as formulações contendo a associação de avobenzona e DHHB (associação 3) apresentaram uma proteção no UVA superior às demais associações, ou seja, que continham avobenzona e DHHB isoladamente. O estudo de fototoxicidade mostrou que a avobenzona e o DHHB apresentaram potencial fototóxico. Quando as associações de filtros solares e antioxidantes foram analisadas, foi observado que a associação de avobenzona e naringenina, combinada ou não ao DHHB, apresentou potencial fototóxico. Dessa forma, esta não é recomendada para o desenvolvimento de formulações fotoprotetoras. A associação DHHB / avobenzona, por apresentar elevada proteção UVA, pode ser vantajosa para o desenvolvimento de fotoprotetores, entretanto a sua associação com antioxidantes deve ser utilizada com cautela e analisada caso a caso. A naringenina não é indicada para fotoprotetores contendo avobenzona, uma vez que esta associação foi considerada fotoinstável e fototóxica. Já a utilização da mangiferina pode ser considerada mais segura, e, além disso, a sua elevada atividade antioxidante pode complementar a eficácia de fotoprotetores. Nowadays, due to the knowledge of UV-induced skin damage, there is a tendency to use natural substances with antioxidant potential, beyond conventional UV-filters. However, the use of photounstable combinations can compromise the photoprotective capacity and lead to formation of reactive intermediates that can cause contact dermatitis and phototoxic skin reactions. Thus, the objective of this study was to evaluate the photostability and phototoxicity of sunscreen formulations containing different UV-filter combinations supplemented or not with the polyphenols mangiferin and naringenin. For photostability studies, samples of the formulations were spread onto glass plates, exposed to UVA radiation and then analyzed by HPLC to determine the UV-filters and antioxidants concentrations, and also by spectrophotometry to determine the UVA/UVB ratio. Phototoxicity was evaluated by using the 3T3 fibroblast cultures, which was exposed to UVA radiation for the determination of cell viability. The results of photostability by HPLC analysis showed that naringenin, ethylhexyl methoxycinnamate and avobenzone were considered photounstable. Furthermore, it was demonstrated that the addition of mangiferin combined with avobenzone and naringenin, provided an increase in their photostability. The presence of DHHB in the formulation containing avobenzone and naringenin promoted an improvement in naringenin photostability, what was not observed for avobenzone. The spectrophotometric analysis showed that the formulations containing the combination of avobenzone and DHHB (combination 3) presented higher protection against UVA than the other combinations, i.e. the combinations containing avobenzone and DHHB, separately. The phototoxicity study showed that avobenzone and DHHB presented phototoxic potential. When UV-filters and antioxidants combinations were analyzed, it was observed that the combination of avobenzone and naringenin, combined or not with DHHB, presented phototoxic potential. Therefore, this is not recommended for the development of sunscreens. The combination DHHB / avobenzone, due to its high UVA protection, may be suitable on the development of sunscreens, however its combination with antioxidants should be used with caution and analyzed case-by-case. Naringenin is not indicated for sunscreens containing avobenzone, since this combination was considered photounstable and phototoxic. Moreover, the use of mangiferin can be considered safer than naringenin and, besides that, its high antioxidant activity can improve the photoprotective effects of sunscreens.

O aumento do conhecimento em relação aos danos provocados pela radiação solar, tanto na faixa do UVB quanto na faixa do UVA, o avanço nas tecnologias relacionadas ao produto e às formas de avaliação bem como a disponibilização de novas moléculas no mercado levaram ao aumento da qualidade da proteção proporcionada pelos fotoprotetores. Entretanto, ainda há preocupações em relação à segurança de alguns filtros solares devido a sua fotoinstabilidade e penetração cutânea. Dessa forma, torna-se necessário o desenvolvimento de novos filtros solares mais efetivos e seguros, que apresentem relevância científica e potencial de inovação. Assim, a presente pesquisa teve como objetivo desenvolver novos análogos ao filtro solar benzofenona-3 e determinar o seu potencial fotoprotetor por meio da avaliação da absorção UV e avaliação da fotoestabilidade, fototoxicidade e atividade antioxidante. Para tal, inicialmente seis análogos ao filtro solar químico benzofenona-3 foram sintetizados visando aumentar o tamanho da molécula e aumentar a sua absorção no UVA longo (340-400nm). Foram avaliados os espectros de absorção no UV e a fotodegradação dessas substâncias. A fototoxicidade das substâncias selecionadas foi avaliada por meio do uso de cultura de fibroblastos 3T3, para a determinação da viabilidade celular na presença e ausência da radiação. A atividade antioxidante das substâncias foi avaliada por meio da quimioluminescência gerada pela reação HRP-H2O2-luminol. Os resultados permitiram demonstrar a importância da relação entre a estrutura molecular dos compostos e sua absorção no UV. As moléculas benzofenona-3 fenil amino (B5) e o carbazol da benzofenona-3 fenil amino (B6) apresentaram maior absorção no UVA, longo e curto, e no UVB, quando comparadas com a molécula de partida (benzofenona-3). Apenas as substâncias que apresentavam ponte de hidrogênio intramolecular (B5 e B6) foram consideradas fotoestáveis. Somente a molécula B5 não apresentou potencial para fototoxicidade aguda. Além disso, essa molécula apresentou atividade antioxidante, o que sugere o seu grande potencial para utilização com filtro solar. The increase of our knowledge of not only UVB but also UVA-induced damages, the advances on product and testing technologies as well as new sunscreen molecules leaded to and enhancement of quality of UV protection provided by topical sunscreens. However there are some safety concerns involving some UV-filters due to their photoinstability and skin penetration. Therefore it is necessary to develop new safer and more effective UV-filters, which also presents scientific relevance and innovation potential. Thus, the aim of the present research was to develop new analogues based on benzophenone-3, and to evaluate their photoprotective potential through their photostability, phototoxicity and antioxidant activity. For this purpose, firstly six new synthetic analogues based on benzophenone-3 were prepared in order to promote molecular weight enhancement as well as improve long-wave UVA absorption (340-400 nm). The UV absorption spectra and photodegradation of these compounds were also analyzed. Phototoxicity of selected compounds was evaluated by using 3T3 monolayer fibroblast culture to determine cell viability in the presence and absence of UVA radiation. The antioxidant activity was evaluated by HRP-H2O2-luminol induced chemiluminescence. The results showed that relationship between the molecular structure and UV absorption. Me molecules phenylamine benzophenone-3 (B5) and phenylamine benzophenone-3 carbazol (B6) showed higher short and long-wave UVA and UVB absorption, when compared to benzophenone-3. Only B5 and B6, which presented intermolecular hydrogen bond, were considered photostable. B5 did not present any acute phototoxicity potential; in addition it has antioxidant activity, which suggests its high UVfilter potential.

A exposição excessiva às radiações UV é capaz de limitar a capacidade dos sistemas de defesa antioxidante em nosso organismo, provocando o estresse oxidativo. Assim, extratos vegetais ricos em compostos antioxidantes são fortes candidatos a serem veiculados em formulações tópicas para a prevenção ou tratamento dos danos causados pela RUV na pele. Dentre a gama de extratos vegetais com atividade biológica, o extrato de açaí, fruto da espécie Euterpe olerecea Mart., tem se destacado por apresentar grande quantidade de antioxidantes na sua composição. Geralmente, a quantidade de ativos que consegue penetrar na pele é pequena, exigindo métodos analíticos muito sensíveis. Assim, o presente trabalho teve como objetivo avaliar o emprego do método de quimioluminescência para a quantificação da penetração dos componentes do extrato de açaí na pele por medida da atividade antioxidante. Os resultados mostraram que o IC50 do extrato de açaí para o ensaio de inibição da quimioluminescência gerada no sistema xantina/luminol/xantina oxidase foi de 0,63 ?g/mL. Além disso, a quantificação do extrato de açaí utilizando esse método foi possível para porcentagens de inibição obtidas até o valor de 60%. Ainda, esse método mostrou-se preciso e exato na determinação da porcentagem de inibição da quimioluminescência do extrato de açaí na concentração próxima a IC50, sendo essa inibição não influenciada pelos componentes das diferentes formulações estudadas. No entanto, o uso da quimioluminescência como um método de quantificação da penetração de componentes antioxidantes de extratos hidrossolúveis torna o processo de extração desses componentes da pele um fator importante, no qual a escolha do solvente extrator é um ponto crítico. Assim, o solvente extrator escolhido para a realização dos estudos de penetração/retenção cutânea foi metanol:água (80:20), visto que esse solvente foi capaz de extrair os componentes antioxidantes do extrato de açaí sem retirar grande quantidade dos compostos inerentes da pele com atividade antioxidante. Por fim, a determinação da inibição da quimioluminescência gerada no sistema xantina/luminol/xantina oxidase mostrou-se um método importante na medida da atividade antioxidante de extratos na pele. Ainda, esse método foi eficaz na quantificação do extrato de açaí nos estudos de penetração/retenção com célula de difusão vertical, apesar de todos os erros inerentes desse método. The excessive exposure to UV radiation is able to decrease the antioxidant defense systems in the skin leading to the oxidative stress. Thus, plant extracts, rich in antioxidant compounds, are strong candidates to be added in topical formulations for the prevention or treatment of UV radiation induced damages. Among the range of plant extracts with biological activity, the extract of açai, fruit of the Euterpe olerecea Mart. species has become known due to the considerable amount of antioxidants in its composition. Generally, the amount of active compounds that can penetrate in the skin is low requiring sensitive analytical methods. Thus, the present study aimed to evaluate the use of chemiluminescence assay for the quantification of the penetration of açai extract components in the skin by measuring the antioxidant activity. The results showed an IC50 value of 0.63 ?g/mL for the antioxidant activity determined by chemiluminescence assay using the xanthine/luminol/xanthine oxidase system. Moreover, the quantification of acai extract using this method was possible for percentages of inhibition obtained until the 60% of inhibition value. Additionally, this method proved to be precise and accurate in determining the percentage of chemiluminescence inhibition of acai extract at concentrations values close to the IC50 value. Additionally, it was observed that the inhibition was not influenced by the different components of the formulations studied. However, the use of chemiluminescence assay to quantify the penetration of water-soluble extract antioxidant components makes the extraction process of these components from the skin an important step in which the selection of the extractor solvent is a critical point. In the present study it was selected methanol:water (80%) as an organic extractor solvent which was able to extract the antioxidant components of the acai extract without removing large amounts of inherent compounds of the skin with antioxidant activity. Finally, the determination of the chemiluminescence generated in the xanthine/luminol/xanthine oxidase system inhibition showed to be an important method for the measurement of the antioxidant activity of plant extracts in the skin. Still, this method was effective in quantifying the acai extract in cutaneous penetration/retention studies using vertical diffusion cells despite all the intrinsic errors of this used method.

Em virtude da necessidade de proteger a pele contra as espécies reativas de oxigênio, geradas excessivamente após exposição ao raios UV, substâncias antioxidantes têm sido adicionadas aos fotoprotetores. Entretanto associações fotoinstáveis podem levar à formação de intermediários reativos, que são prejudiciais ao organismo, e à redução da atividade fotoprotetorados filtros solares e antioxidantes. Além disso, as características de penetração dos filtros solares e antioxidantes influenciam diretamente a segurança e a eficácia dos fotoprotetores. Dessa forma, o objetivo deste trabalho foi desenvolver e avaliar a fotoestabilidade e a penetração cutânea de formulações fotoprotetoras contendo diferentes associações de filtros solares acrescidas ou não de trans-resveratrol e beta-caroteno, por meio do uso de CLAE e espectrofotometria e de células de Franz, respectivamente. Para tal, foram desenvolvidas formulações contendo diferentes associações de filtros solares (associações 1, 2 e 3), que apresentavam em comum avobenzona, metoxicinamato de etil-hexila e octocrileno, acrescidas de trans-resveratrol e beta-caroteno, isoladamente ou em combinação. Para avaliar a fotoestabilidade, amostras das formulações foram aplicadas em lâminas de vidro e expostas à radiação UVA e, a seguir, submetidas a análises por CLAE, para determinar o teor dos filtros solares e antioxidantes em estudo, e por espectrofotometria, para determinação da razão UVA/UVB. Para avaliar a penetração cutânea, as formulações contendo a associação de filtros solares mais fotoestável foram aplicadas sobre a pele de orelha de porco, e foi realizada a quantificação das substâncias em estudo, presentes no estrato córneo (EC), na epiderme/derme (E+D) e na solução receptora, por CLAE. O estudo de fotoestabilidade por CLAE dos filtros solares e antioxidantes presentes nas formulações avaliadas mostrou que, apesar de terem apresentado boa fotoestabilidade, todas as substâncias foram fotoinstáveis, com exceção do bemotrizinol, octiltriazona e octocrileno. Além disso, foi observado que o uso dos antioxidantes em associação foi melhor do que utilizá-los separadamente em fotoprotetores. As análises espectrofotométricas mostraram que, após irradiação, houve redução significativa da razão UVA/UVB de todas as formulações e que as formulações contendo bemotrizinol (associação 2) e octiltriazona (associação 3) apresentaram, respectivamente, aumento e redução significativos da razão UVA/UVB, com relação às formulações que não apresentavam esses filtros. No estudo de penetração cutânea, foi observado que, após aplicação das formulações contendo bemotrizinol (associação 2), os filtros solares e antioxidantes avaliados penetraram a pele, mas não permearam até a solução receptora, ou seja, ficaram retidos no EC, majoritariamente, e na E+D. Além disso, foi observada redução significativa da retenção no EC dos filtros solares e antioxidantes avaliados na presença do beta-caroteno. Os resultados desse estudo contribuíram para mostrar os benefícios da utilização de uma combinação de antioxidantes em fotoprotetores, uma vez que a combinação do trans-resveratrol e do beta-caroteno, resultou em melhor fotoestabilidade das formulações e também apresentou vantagens no estudo de penetração cutânea, pois reduziu a penetração dos filtros solares. Due to the need to protect the skin against reactive oxygen species, which are excessively generated after exposure to UV rays, antioxidants compounds have been added to sunscreens. However, photounstable combinations can lead to generation of reactive intermediates, that are harmful to the body, and decrease the photoprotective activity of UV-filters and antioxidants. Furthermore, the penetration characteristics of UV-filters and antioxidants influence directly sunscreens safety and efficacy. Thus, the aim of this study was to develop and to evaluate the photostability and cutaneous penetration of sunscreen formulations containing different combinations of UV-filters supplemented or not with trans-resveratrol and beta-carotene, by using HPLC and spectrophotometry and Franz cells, respectively. For this, formulations containing different UV-filters combinations (combinations 1, 2 and 3), which had in common avobenzone, ethyl-hexyl methoxycinnamate and octocrylene, supplemented with trans-resveratrol and beta-carotene, alone or in combination, were prepared. For photostability evaluation, samples of the formulations were spread onto glass plates, exposed to UVA radiation and then analyzed by HPLC to determine the concentration of UV-filters and antioxidants under study, and by spectrophotometry to determine the UVA/UVB ratio. To assess cutaneous penetration, formulations containing the most photostable combination of UV-filters were applied on pig ear skin, and the quantification of substances under study present into stratum corneum (SC), viable epidermis plus dermis (E+D) and receptor fluid was performed by HPLC. HPLC photostability study, which analyzed the UV-filters and antioxidants present in formulations, showed that all substances, despite having good photostability, were photounstable, except bemotrizinole, octyltriazone and octocrylene. Furthermore, it was observed that the use of antioxidants in combination was better than using them separately in sunscreens. The spectrophotometric analysis showed that after irradiation, all formulations had a significant decrease of UVA/UVB ratio, and that formulations containing bemotrizinole (combination 2) and octyltriazone (combination 3) presented, respectively, significant increase and decrease of the UVA/UVB ratio, in comparison with formulations that did not contained these UV-filters. In the cutaneous penetration study it was observed that after application of formulations containing bemotrizinole (combination 2), the UV-filters and antioxidants under study penetrated the skin, but did not reach the receptor fluid, which means that they were retained in SC, predominantly, and in E+D. Furthermore, a significant decrease on SC retention of UV-filters and antioxidants under study in the presence of beta-carotene was observed. The results of this study contributed to demonstrate the benefits of using a combination of antioxidants in sunscreens, since the combination of trans-resveratrol with beta-carotene, resulted in improved photostability of the formulations and also showed advantages in the skin penetration study, because it reduced UV-filters penetration.

O objetivo desta pesquisa foi desenvolver e aperfeiçoar uma formulação com fase gel lamelar contendo óleo de Calendula officinalis e avaliar seu potencial na atividade cicatrizante de úlceras em ratos. A formulação estudada possui valor de EHL 6,0, constituída por óleo de calêndula e sistema tensoativo formado por derivados etoxilados dos alcoóis cetílico e estearílico (Ceteth 2/Steareth 20). Primeiramente, determinou-se a região do diagrama ternário de fases em que se encontravam as emulsões com fase gel lamelares (EFGL), macroscopicamente estáveis. Em seguida foi avaliada a estabilidade do sistema e parâmetros que poderiam influenciar na formação das estruturas anisotrópicas. Avaliou-se o comportamento das emulsões utilizando o teste de perda de massa por evaporação. Na avaliação in vitro, foram realizados testes de citotoxidade do óleo de calêndula frente às células de fibroblastos da linhagem L929 através do ensaio de apoptose e necrose. O teste in vivo foi realizado através do Índice de Cicatrização de Úlceras (ICU) no modelo em dorso de ratos (úlcera excisional contrátil) a fim de avaliar o potencial cicatrizante da emulsão proposta comparando com o sham (grupo controle). As úlceras foram avaliadas mediante análise de imagem nos tempos de 0, 2, 7, 14 e 21 dias após o procedimento cirúrgico. A EFGL demonstrou maior estabilidade frente aos testes de estabilidade preliminar e acelerada em relação as demais formulações. Além disso, esta formulação demonstrou menor área de histerese (tixotropia), portanto menor grau de espalhabilidade e com maior tempo de contato com a úlcera. Durante a evaporação das emulsões houve manutenção das estruturas anisotrópicas O óleo bruto da Calendula officinalis não apresentou interferência na via da apoptose e necrose na concentração de até 1000 ?g/mL em fibroblastos L929. A formulação proposta promoveu melhor cicatrização no modelo de úlcera cutânea (ICU) na região dorsal de ratos, supondo- se modular a fase inflamatória do processo de cicatrização, pois o maior recrutamento de células inflamatórias bem como a colagênese, diminuída no grupo EFGL, foram fatores essenciais que permitiram a total reepitelização das úlceras cutâneas. Portanto, pode-se concluir que a metodologia utilizada nesta pesquisa foi útil para a obtenção de emulsões com fase gel lamelar, sendo que a formação dessa estrutura é importante para a estabilidade do sistema, podendo ser utilizada como uma formulação viável e eficaz no processo cicatricial de feridas. The aim of this research was to improve a lamellar gel phase emulsion containing Calendula officinalis oil and to investigate its potential as a modern wound dressing. First, we determined the region of the ternary phase diagram in which the emulsions were stable and we also evaluated the intrinsic stability of the system and the action of some parameters which may influence the formation of lamellar gel phase. In addition, we analyzed the samples\' behavior during the evaporation process. Moreover, an in vitro cytotoxicity assay of the calendula oil was performed in order to evaluate apoptosis and necrosis in fibroblast cell line L929. Then, an in vivo test was carried out by the wound healing rate (WHR), using a specific model of ulcer in rats (excisional contractile ulcer), in order to assess potential healing of the proposed emulsion (LGP) by comparing with the sham group. The ulcers were evaluated by image analysis at 0, 2, 7, 14 and 21 days after surgery. The emulsion 6 \'showed greater stability in the preliminary and accelerated stability tests in relation to the previous studied formulation. Moreover, this formulation presented more compatible characteristics, because it showed smaller hysteresis area of ? (thixotropy), therefore a lower degree of spreadability and, accordingly, the formulation may increase contact span with ulcer, which is a desirable characteristic. During evaporation of the emulsions, the anisotropic structures were maintained, but their type varied depending on the decreased amount of water in the system. The crude oil of Calendula officinalis showed no interference with the pathway of apoptosis and necrosis in the concentration of 1000 mg / mL in L929 fibroblasts. The proposed model has promoted better wound healing rate in the ulcers in the dorsal region of rats. It seemed to modulate the inflammatory phase of the healing process, because of the increased recruitment of inflammatory cells as well as collagenesis, once LGP emulsion decreased in the group, were factors essential that allowed total reepithelialization of skin ulcers in rats treated with LGP emulsion. In conclusion, this study produced an enhanced and useful LGP which can be used as a new approach to stimulate the healing process and treat wounds efficiently.

A pele é exposta a inúmeros agentes, dentre os quais destaca-se a radiação ultravioleta (RUV), que pode energizar os cromóforos celulares da pele e estes reagindo com o oxigênio molecular podem resultar na geração das espécies reativas de oxigênio (EROs). Nestas circunstâncias, mesmo possuindo um sistema de defesa antioxidante, a concentração de radicais livres pode aumentar, rompendo o equilíbrio pró oxidante/antioxidante, levando ao estresse oxidativo na pele. Neste contexto, o uso tópico de formulações adicionadas de substâncias naturais tem sido um eficiente modo para enriquecer o sistema protetor cutâneo endógeno, reduzindo os danos oxidativos causados pela RUV na pele. Sendo assim, o presente trabalho teve como objetivo estudar o potencial fotoquimioprotetor do extrato e da fração de média polaridade de Inga edulis e desenvolver formulações estáveis física e quimicamente a fim de selecionar a que prover melhor liberação, penetração e retenção dos componentes do extrato e/ou da fração na epiderme viável. A partir do perfil cromatográfico foi possível observar que o extrato possui compostos comuns à fração e alguns deles foram identificados, sendo o composto vicenina-2 o escolhido para ser monitorado nos estudos de penetração e retenção cutânea. Dentre as formulações estudadas a formulação a base de Hostacerin SAF® adicionada da fração foi a que proveu a maior penetração/retenção do composto vicenina-2 e de compostos antioxidantes na epiderme viável e também foi considerada estável por todos os parâmetros avaliados no estudo de estabilidade preliminar. A fração de Inga edulis, veiculada por esta formulação, mostrou alta atividade antioxidante frente aos métodos estudados, apresentou baixa citotoxidade em cultura celular de fibroblastos, sendo a quantidade citotóxica estabelecida muito acima da quantidade de vicenina-2 retida na epiderme viável. A fração também apresentou fotoestabilidade frente a altas doses de radiação UVA e UVB quanto à citotoxidade, capacidade de reduzir o radical sintético DPPH? e de sequestrar o ânion superóxido in vitro. Assim, a formulação gel-creme com Hostacerin SAF® adicionada de 1% da fração de média polaridade do Inga edulis mostrou ser muito promissora e deve ser melhor estudada, visando futura aplicação como formulação fotoquimiopreventiva. The skin is exposed to numerous agents, among which stands out the ultraviolet radiation (UVR) that can energize the chromophores of cell skin and these react with molecular oxygen resulting in the generation of reactive oxygen species (ROS). In these circumstances, the endogenous antioxidant defense system would be impaired leading to a pro oxidant state named oxidative stress. In this context, the topical formulations added with natural substances has been an efficient way to aid endogenous skin protective system, minimizing the oxidative damage caused by UV rays. Thus, the aim of the present work was to study the potential photochemopreventive activity of the extract and medium polarity fraction of Inga edulis and develop formulations physically and chemically stable in order to select the penetration and retention of the components of the extract and / or fraction oin the viable epidermis. The chromatographic profiles showed that the some compounds of the extract are also present in the fraction and some of them were identified. The compound vicenin-2 was chosen to be monitored in studies of cutaneous penetration and retention. Among the formulations studied, the formulation Hostacerin SAF® added to the fraction provided the highest penetration / retention of the compound vicenin-2 and antioxidant compounds in viable epidermis and also was considered stable at all parameters evaluated in the study of preliminary stability. The fraction of Inga edulis, conveyed by this formulation, showed strong antioxidant activity in the methods studied and showed low cytotoxicity in cell culture of fibroblasts. The cytotoxicity concentration was greater than the amount of vicenin-2 retained in viable epidermis. The fraction also showed photostability when it was exposed to high doses of UVA and UVB. The irradiated fraction showed no change in cytotoxicity and antioxidant activity. Thus, the formulation Hostacerin SAF® added with 1% of medium polarity fraction of Inga edulis proved very promising and should be further studied for a future application in photochemoprevention.

Emulsões são sistemas dispersos constituídos de pelo menos duas fases líquidas imiscíveis e um agente emulsificante para estabilização das mesmas. a fase dispersa destesw sistemas é conhecida como fase interna e o meio dispersante como fase externa, porém, são sistemas termodinamicamente instáveis. A presença de cirstais líquidos aumenta a estabilidade destas dispersões e além de alterar a viscosidade. São capazes de promover o encapsulamento de ativos, permitindo liberação sustentada, bem como a proteção dos fármacos foto- e termosensíveis. O óleo de girassol é uma interessante fase oleosa em uma emulsão, pois possui proprie3dades cicatrizantes devido à alta concentração de ácido linoléico e propriedades antioxidantes devido à presença de polifenóis e vitamina E Possuemaplicações em diferentes áreas como: cosméticos, alimentos, veículos de medicamentos e outros. Assim, os objetivos deste estudo foram desenvolver emulsões estáveis contendo cristais líquidos a partir da mistura de óleo de girassol (Helianthus annus L.) e mineral (vaselina líquida), caracterizá-las e avaliar a influência de diferentes ésteres graxos na formação dos cristais líquidos. As formulações foram desenvolvidas utilizando o método de diagrama e pseudo-diagrama ternário, a partir dos quais foram selecionadas duas formulações que demonstraram se estáveis e apresentaram cristais líquidos durante todo o teste de estabilidade acelerada. As mesmas foram aditivadas com extrato glicólico de polifenois e ésteres graxos e permaneceram estáveis com cristal líquido. A caracterização reológica observou-se fluxo pseudo-plástico e tixotrópico, ideal para produtos farmacêuticos e cosméticos. O óleo de girassol apresentou atividade antioxidante mesmo quando aquecidos para o processo de emulsificação pelo método do radical DPPH·. As cadeias carbônicas dos ésteres graxos utilizados em produtos cosméticos podem influenciar na formação e manutenção dos cristais líquidos. Por meio dos testes in vivo observou-se que as formulações desenvolvidas promoveram aumento prolongado da hidratação da pele e não foram observados sinais de irritação cutânea. Emulsions are dispersed systems consisting of at least two immiscible liquid phases and an emulsifying agent to stabilize them. The dispersed phase of these systems is known as the internal phase and the dispersing environ is the external phase, which are, however, thermodynamically unstable systems. The presence of liquid crystals increases the stability of these dispersions and also change the viscosity. They are able to promote the encapsulation of active, allowing sustained release as well as the protection of photo-and thermo-sensitive drugs. Sunflower oil is an interesting phase in an oily emulsion, because it has healing properties due to the high concentration of linoleic acid, and antioxidant properties due to the presence of polyphenols and vitamin E. They have applications in different areas such as cosmetics, food, medicines and other vehicles. The objectives of this study were to develop stable emulsions containing liquid crystals from a mixture of sunflower oil (Helianthus annus L.) and mineral (mineral oil), characterize them and evaluate the influence of different fatty esters in the formation of liquid crystals. The formulations were developed using the method of diagram and ternary pseudo-diagram, from which we selected two formulations that have been demonstrated stable and presented as liquid crystals throughout the accelerated stability test. They were doped with polyphenols glycolic extracts and acid esters, and have been kept stable with liquid crystal. At the rheological characterization was observed pseudo-plastic flow and thixotropic, ideal for pharmaceuticals and cosmetics. Sunflower oil showed antioxidant activity even when heated to the emulsification process by the radical DPPHo method. The carbon chains of fatty esters used in cosmetics can influence the formation and maintenance of liquid crystals. In vivo tests showed that the formulations developed promoted prolonged increase in skin hydration and were not seen signs of skin irritation.

Os atributos físicos, físico-químicos e químicos dos sistemas emulsionados são influenciados pelas características das gotículas da fase interna destes, como: concentração, tamanho e morfologia. Dessa forma, os objetivos envolveram a caracterização física e físico-química de sistemas emulsionados obtidos a partir dos tensoativos álcool estearílico 21 OE (óxidos de etileno) (steareth-21) e álcool oleílico 20 OE (oleth-20), bem como, a introdução da tomografia por coerência óptica (OCT) como técnica analítica para determinação do tamanho de partículas e caracterização morfológica das emulsões. Prepararam-se três formulações, diferindo-se no tensoativo utilizado, sendo o sistema emulsionado SE-1 composto por 8,0% (p/p) de oleth-20; o SE-2 por 4,0% (p/p) de oleth-20 e 4,0% (p/p) steareth-21; e o SE-3 por 8,0% (p/p) steareth-21. Avaliaram-se a estabilidade preliminar, o valor de pH, os perfis reológico e termogravimétrico. A presença de fase gel cristalina foi determinada por meio da microscopia com luz polarizada e, o tamanho das gotículas, por meio da microscopia óptica comum, difração a laser e OCT. As amostras não apresentaram sinais de instabilidade por meio dos testes da centrifugação e do estresse térmico. O valor de pH foi considerado compatível com o da pele humana (4,6 a 5,8). Os sistemas apresentaram comportamento reológico não-Newtoniano pseudoplástico. SE-1 apresentou viscosidade aparente inferior (223,53 mPa.s) e área de histerese (2238,38 Pa/s) estatisticamente igual ao SE-2 (2911,19 Pa/s); SE-2 apresentou valor intermediário de viscosidade aparente (332,20 mPa.s) e, SE-3, valores superiores para ambos os parâmetros (636,40 mPa.s e 4248,97 Pa/s). Os testes oscilatórios constataram a predominância do caráter elástico para os três sistemas. Por meio da termogravimetria, o perfil das três amostras foi semelhante, independentemente do tipo e concentração do tensoativo não-iônico. Observou-se a presença de fase gel cristalina para os três sistemas, sendo aparentemente mais pronunciado para o SE-2. Tanto na análise microscópica quanto na difração a laser foram obtidos tamanho médio de partículas menor que 6,0 µm. De acordo com os resultados da OCT, as partículas menores que 6,0 µm não foram possíveis de serem conclusivamente observadas e aquelas maiores sugeriram ser a fase interna dos sistemas. The physical, physicochemical and chemical attributes of the emulsified systems are influenced by the characteristics of their internal phase droplets, such as: concentration, size and morphology. Thus, the aims involved the physical and physicochemical characterization of emulsions obtained from the stearyl alcohol condensed with 21 mols ethylene oxide (EO) (steareth-21) and oleyl alcohol with 20 mols EO (oleth-20), as well as, the introduction of the optical coherence tomography (OCT) as the analytical technique tool to the determination of the particle size and morphological characterization of the emulsified systems. Three formulations were prepared, differing at the surfactant used, being the emulsified system SE-1 composed by 8.0% (p/p) of oleth-20, the SE-2 by 4.0% (p/p) of oleth-20 and 4.0% (p/p) steareth-21; and the SE-3 by 8.0% (p/p) steareth-21. The preliminary stability was evaluated, such as the ph value and the rheological and thermogravimetric profiles. The presence of the crystalline gel phase was determined from microscopy with polarized light and the droplet size through regular optical microscopy, laser diffraction and OCT. The samples did not present signs of instability throughout the centrifugation and thermal stress tests. The ph value was considered compatible to human skin (4.6 to 5.8). Systems represent the non-Newtonian pseudoplastic rheological behaviour. The SE-1 presented inferior apparent viscosity (223.53 mPa.s) and the hysteresis area (2238.38 Pa/s) statistically equal to SE-2 (2911.19 Pa/s); the SE-2 presented an average value of apparent viscosity (332.20 mPa.s) and, the SE-3, superior values to both parameters (636.40 mPa.s and 4248.97 Pa/s). The oscillating tests verified the elastic character predomination to the three systems. Throughout the thermogravimetry, the profile of the three samples was similar, independent from the kind and concentration of the non-ionic surfactant. The presence of the crystalline gel phase was identified at the three systems, being apparently more pronounced to the SE-2. Either at the microscopic analysis as well as at the laser diffraction were obtained an average size of the particles minor than 6.0 µm. According to OCT results, the particles below 6.0 µm were not possible to be identified and the major particles were suggested be the internal phase of the systems, however new studies should be performed.
