Society increase in live expectancy will cause a rise in Alzheimer’s Disease (AD) diagnosis, thus, there is a need to further understand how it works. One of the main culprits of this disease amyloid-β peptide (Aβ), this protein has a physiological function as monomers. But, once starts to aggregate, Aβ becomes neurotoxic. In this thesis, I have demonstrated a novel physiological function of monomeric Aβ as inductor of insulin receptor signalling, while its oligomers block the same signalling. I have described a neurotoxic feedback loop between oligomeric Aβ and an increase in BACE1 presence. I have studied novel calcium regulators in the development of the disease. I showed a novel pathway that links Aβ as a blocker of LTP. Finally, I have characterised the region of interaction of albumin c-terminus with Aβ and its neuroprotective effect. L’increment en l’esperança de vida a la nostre societat generarà un augment en el nombre de persones diagnosticades amb la malaltia d’ Alzheimer. Per això, és necessari poder comprendre millor el seu funcionament. Un dels principals actors d’aquesta malaltia es la proteïna β- amiloide, aquesta proteïna en estat monomèric te funcions fisiològiques, però quan agrega es torna neurotòxica. En aquesta tesis he demostrat una nova funció fisiològica de l’Aβ com a inductor de la senyalització regulada pel receptor d’insulina, mentre que els oligòmers la bloquegen. He descrit un cercle viciós entre Aβ oligomerica i un increment en la presencia de BACE1. He estudiat el paper de nous reguladors de calci en el desenvolupament d’AD. He mostrat una nova relació entre Aβ i el bloqueig del LTP. Finalment he caracteritzat la regió d’interacció del c-terminus de l’albúmina i l’Aβ i el seu paper neuroprotector.

All living organisms respond to environmental stimuli by modulating the activity of different genes. This response can occur across many locations throughout the cell such as at the level of transcription, translation, and even post-translational modifications. Some of this variation between species is attributable to the presence or absence of lineage-specific genes. The birth of new genes via de novo emergence from previously non-genic DNA is predicted to contribute significantly to this diversity. To explore this question, we generated high-depth transcriptomic data for 11 species of yeast, as well as ribosome profiling and proteomics data for Saccharomyces cerevisiae in rich media and oxidative stress conditions. We identified 213 putative de novo genes in S. cerevisiae; over a third of them are translated, and approximately half of them overlap more ancient genes on the antisense strand. We found that a significant fraction of S. cerevisiae genes (~5%) have emerged de novo over the past 20 million years. Tots els organismes vius responen als estímuls ambientals modulant l’activitat dels seus gens. Aquesta resposta es produeix simultàniament en molts llocs a tota la cèl·lula i a través de diferents mecanismes, com el nivell de transcripció, el nivell de traducció i fins i tot modificacions posteriors a la traducció. Una part d’aquesta variació entre espècies és atribuïble a la presència o absència de gens específics del llinatge. Es preveu que el naixement de nous gens mitjançant l’emergència de novo a partir d’ADN anteriorment no gènic contribueixi significativament a aquesta diversitat. Per explorar aquesta questió, hem generat dades transcriptòmiques d’alta profunditat per a 11 espècies de llevats, així com dades de empremta ribosomal i dades de proteòmica per a Saccharomyces cerevisiae en condicions de medi de cultiu ric i sota estrès oxidatiu. Hem identificat 213 gens putatius de novo a S. cerevisiae; més d’un terç d’ells es tradueixen, i aproximadament la meitat d’ells es troben superposats a gens més antics a la cadena antisentit. Hem trobat que una part important dels gens de S. cerevisiae (~5%) han sorgit de novo en els darrers 20 milions d'anys.

Williams-Beuren syndrome (WBS) is a rare neurodevelopmental disorder caused by a heterozygous deletion of ~1.5 Mb in chromosome region 7q11.23. The use of murine models of the disease has helped to broaden the knowledge about the syndrome and the role of the affected genes and pathways, but a deep insight of the molecular basis of the disease would be of great interest for the development of possible treatments. We have used a WBS mouse model carrying a heterozygous deletion that mimics the most common deletion found in patients. We have characterized the vascular phenotype of these animals and identified molecular alterations relevant for both the neurocognitive and cardiovascular phenotypes of the disease. In addition, we have performed preclinical tests of two novel therapeutic approaches. The results highlight the importance of this model for the study of molecular mechanisms underlying the disease and the evaluation of the effect of new therapeutic strategies. La síndrome de Williams-Beuren (WBS) és una malaltia rara del neurodesenvolupament causada per una deleció heterozigota de ~1.5 Mb a la regió cromosòmica 7q11.23. L’ús de models murins de la malaltia ha contribuït a ampliar el coneixement sobre la síndrome i la funció dels gens i vies afectats, però conèixer les bases moleculars de la malaltia seria de gran interès per al desenvolupament de possibles tractaments. Hem utilitzat un model de ratolí de la WBS portador d’una deleció heterozigota que mimetitza la deleció més comú trobada en pacients. Hem caracteritzat el fenotip vascular d’aquests animals i identificat alteracions moleculars rellevants tant per al fenotip neurocognitiu com cardiovascular associats a la malaltia. A més, hem dut a terme assajos preclínics amb dues aproximacions terapèutiques innovadores. Els resultats remarquen la importància d’aquest model per a l’estudi dels mecanismes moleculars subjacents a la malaltia i per a l’avaluació de l’efecte de noves estratègies terapèutiques.

Reactive oxygen species (ROS) are a variety of molecules derived from molecular oxygen during different metabolic processes. ROS can react with different cellular components resulting in lipid peroxidation, protein oxidation and DNA damage. Single-cell organisms have to cope with a wide range of environmental changes and exposition to toxic compounds. In response to hydrogen peroxide (H2O2), S. pombe activates different signalling pathways depending on the severity of the stress exerted, mediate at least by two transcriptional factors (TFs), the AP-1 like Pap1 and the bZIP Atf1 controlled by the mitogen-activated protein kinase. In fission yeast, the Pap1 and Sty1 pathways constitute the key protective responses to oxidative stress. The MAPK Sty1 is activated upon several stress situations, like osmotic and oxidative stress. Since the modulation of gene expression is one of the main outputs of this response, we focused this work on the characterization of Sty1 pathway as a sensor of oxidative stress and the different molecular events leading to its transcriptional program activation. We have also, studied the role of translation by Elongator in the induction of the stress response, Els radicals lliures d’oxigen (ROS) són unes molècules derivades de l’oxigen provinents de diferents processos metabòlics. ROS poden reaccionar amb diferent components cel•lulars, resultant en peroxidació d’àcids grassos, oxidació de proteïnes i danys al DNA. Els organismes unicel•lulars estan sotmesos a una àmplia varietat de canvis ambientals i a l’exposició de compostos tòxics. En resposta a l’estrés oxidatiu provocat pel peròxid d’hidrogen, S. pombe activa diferent vies de senyalització segons la severitat de l’estrés sofert, dut a terme per dos factors de transcripció, Pap1 i el bZIP Atf1 sota el control aquest últim de la MAPK. En el llevat S. pombe, Pap1 i Sty1 constitueixen un paper clau per lat resposta a l’estrés oxidatiu. La MAPK Sty1 s’activa sota l’efecte de diferent estressos, aixi com l’estrés oxidatiu o l’osmòtic. La modulació de la resposta gènica és un dels principals punts en aquesta resposta, hem centrat el treball d’aquesta tesis en la caracterització de la ruta de Sty1 com a sensor de l’estrés oxidatiu així com els diferents events moleculars que activen el programa transcripcional. També hem estudiat el paper de la traducció per l’Elongato en la inducció de la resposta a estrés.

Aquesta tesi demostra que el peroxinitrit produït com a conseqüència del pèptid beta-amiloide (A) contribueix l'augment de la relació A42/A40 que ocorre a la malaltia d'Alzheimer. L'A42 contribueix a l'aparició de la malaltia degut a la seva major toxicitat (quan es compara amb l'A40) que resulta d'una gran estabilitat i capacitat agregativa. A més el peroxinitrit incrementa la toxicitat d'aquest degut a què potencia la seva agregació en forma d'oligomers altament tòxics. De fet els oligomers formats de nitro-A42 presenten una major toxicitat que aquells formats de A42 . En conjunt aquest resultats senyalen l'important paper que l'A42 té en la malaltia d'Alzheimer. Per altra banda, des de la identificació dels agregats d'A i la subseqüent formació dels cabdells neurofibrilars (NFT) com a els dos trets distintius de la malaltia, un gran esforç s'ha dedicat a establir els mecanismes moleculars que uneixen ambdós processos. Aquesta tesi demostra que el peroxinitrit format a partir de l'agregació de d'Ai la conseqüent nitrotirosinació de proteïnes, potencia l'agregació de la proteïna tau en forma de fibres. D'aquesta forma, la nitrotirosinació de la proteïna triosafosfat isomerasa (TPI) podria ser el vincle entre la toxicitat derivada del agregats d'Ai la patologia derivada de la proteïna tau. Per tant, la nitrotirosinació de la TPI podria explicar la progressió temporal que ocorre als cervells de pacients amb la malaltia d'Alzheimer des de la toxicitat induïda per l'Ai l'aparició dels NFT. Els resultats presentats en aquesta tesi podrien obrir nous aspectes en la recerca de la malaltia d'Alzheimer així com en altres malalties que cursin amb estrès oxidatiu i plegament erroni de proteïnes., This thesis demonstrates that amyloid ß-peptide (Aß)-induced peroxynitrite contributes to the switch of the Aβ42/Aβ40 ratio that occurs in Alzheimer's disease (AD). Since Aβ42 is more toxic due to its higher aggregation and stability, it contributes to the trigger of the disease. In addition the aggregation of Aβ42 in form of the highly toxic oligomers is incremented by the presence of peroxynitrite. Moreover, these nitro-Aß42 oligomers are more toxic than those non-nitrated. All these results support the important role of peroxynitrite in AD etiology. Furthermore, since the identification of Aß accumulation and the subsequent formation of neurofibrillary tangles (NFT) as the two defining pathological hallmarks of AD, a fair amount of research on AD has been driven by the need to find the molecular mechanism linking Aß and NFT. This thesis shows the Aß-induced peroxynitrite, and the consequent nitrotyrosination of proteins, promotes tau fibrillization. Thus triosephosphate isomerase (TPI) nitrotyrosination could be the link between Aß-induced toxicity and tau pathology. Therefore, TPI nitrotyrosination may explain the temporal progression from Aß toxicity to NFT formation in AD brain. The work presented in this thesis could open a novel angle in the research of the pathophysiology of AD and could also have an impact to the research in other neurodegenerative diseases involving oxidative stress and protein misfolding.

L'asma és una malaltia d'etiologia complexa, formada per factors genètics i ambientals, on la interrelació de ambdós factors mitjançant interaccions gen-ambient juga un paper clau. L'objectiu d'aquesta tesi ha sigut aprofundir en el coneixement del paper dels polimorfismes genètics, i la seva interacció amb factors ambientals, en la ocurrència d'asma, atòpia i hiperreactivitat bronquial. Aquest objectiu ha estat desenvolupat a través de la replicació de variants genètiques prèviament identificades, l'avaluació d'interaccions gen-ambient i la identificació de nous gens de susceptibilitat mitjançant un disseny basat en el genotipatge de variants genètiques all llarg del genoma en pools d'ADN. La tesi ha estat majoritàriament duta a terme dins l'estudi European Community Respiratory Health Survey (ECRHS) que està comprès per 5.000 individus seguits durant 9 anys, pels quals es disposa d'un qüestionari complet sobre símptomes respiratoris, avaluacions clíniques, informació sobre exposicions ambientals i mostres de ADN. Aquesta tesi a replicat l'associació del polimorfismes dels gens TNFA i NPSR1 amb asma. A més s'han establert les interaccions entre TNFA i obesitat, NQO1 i contaminació atmosfèrica, i NPSR1 i edat d'inici d'asma. L'anàlisi de pools d' ADN ha permès associar la regió on es situa el gen SGK493 amb atòpia. Aquesta tesi contribueix al coneixement de l'etiologia d'asma amb la identificació i replicació d'associacions genètiques i interaccions gen-ambient., Asthma is a disease with a complex etiology, involving multiple genetic and environmental factors, and with an important role of the interplay of these factors through gene-environment interactions. In this thesis I aimed to advance our knowledge on the importance of genetic polymorphisms and their interaction with environmental data for the occurrence of asthma and related phenotypes (atopy and bronchial hyperreactivity). This objective was developed through the replication of genetic associations previously reported, the assessment of gene-environment interactions and the identification of new susceptibility genes using genome-wide analysis based on a pooling DNA strategy. The thesis was, mostly, performed within the European Community Respiratory Health Survey (ECRHS). This cohort has information and DNA samples from approximately 5,000 adult subjects followed-up for 9 years, with extensive questionnaires on respiratory symptoms, clinical evaluations and information on environmental exposures. This thesis replicates previous effects on asthma of polymorphisms in TNFA and NPSR1 genes. In addition, interactions have been established between TNFA and obesity, NQO1 and air-pollution, and NPSR1 and age at onset of asthma. The approach based on genome-wide analysis of DNA pools identified the SGK493 region being associated with atopy. This thesis contributes to the understanding of the etiology of asthma through the identification and replication of genetic associations and gene-environment interactions.

En aquest treball de tesis doctoral s'estudia el paper de l'estrès oxidatiu tant a la etiologia com a la patofisiologia del dany vascular associat a la malaltia d'Alzheimer. Es demostra la capacitat de les cèl·lules musculars llises vasculars de produir pèptid β-amiloide (Aβ). L'estrès oxidatiu associat a edats avançades, estimula el processament amiloidogènic de l' APP a cèl·lules musculars llises vasculars portant a un augment de producció d' Aβ1-40 i Aβ1-42 contribuint així, a la formació dels depòsits amiloides vasculars. Alhora, els depòsits amiloides vasculars indueixen dany vascular a través d'estrès oxidatiu, a on la nitrotirosinació de proteïnes juga un paper clau en la inactivació d'enzimes essencials per la viabilitat cel·lular. La vitamina E i els estrogens tenen un efecte protector a neurones i cèl·lules musculars llises vasculars mentre que la vitamina E, però no els estrogens, és capaç de protegir les cèl·lules endotelials davant la toxicitat induïda pel pèptid Aβ., In this thesis we study the effect of oxidative stress in the etiology and pathophysiology of the vascular damage associated to Alzheimer's disease. We demonstrate the production of amyloid-β-peptide (Aβ) by vascular smooth muscle cells. Oxidative stress related to advance ages, induces the amyloidogenic APP cleavage producing an increase of Aβ1-40 and Aβ1-42 by vascular smooth muscle cells, which in tum contribute to vascular amyloid deposits generation. Vascular amyloid deposits induce oxidative stress, in which protein nitrotyrosination plays a key role in essential enzymes inactivation. Vitamin E and estrogens are able to protect neurons and vascular smooth muscle cells while vitamin E, but not estrogens, is able to protect endothelial cells against Aβ-mediated toxicity.

En aquest treball es presenta l'evolució molecular i estudi funcional de gens localitzats a les duplicacions segmentàries de la regió 7q11.23, implicada en la Síndrome de Williams-Beuren (SWB). S'ha datat l'aparició d'aquestes duplicacions en els últims 25 milions d'anys d'evolució i s'ha proposat un model evolutiu amb reordenaments específics i mecanismes de generació. Correlacions clínico-moleculars en els pacients amb la SWB han permès determinar que l'haploinsuficiència per NCF1, un gen localitzat a les duplicacions, és un factor protector per hipertensió. S'ha proposat un model patogènic per la hipertensió, implicant l'oxidasa NAD(P)H i estrès oxidatiu, suggerint que noves estratègies terapèutiques podrien ser utilitzades. A més, s'ha caracteritzat parcialment la funció de GTF2IRD2, un altre gen de les duplicacions. GTF2IRD2 interacciona amb altres factors de transcripció relacionats, té una localització subcel·lular variable i no s'uneix a ADN. Aquests resultats contribueixen a conèixer millor els mecanismes mutacionals i patogènics de la SWB., This work presents the molecular evolution along with the functional analysis of the genes located in the segmental duplications flanking the 7q11.23 region, involved in Williams-Beuren syndrome (WBS). The generation of the segmental duplications has been dated to the last 25 million years of evolution and an evolutionary model with specific rearrangements and mechanisms has been proposed. Clinical-molecular correlations in WBS patients have allowed to determine that haploinsufficiency at NCF1, a gene located in the duplications, is a protective factor for hypertension. A pathogenic model for hypertension has been proposed, implicating NAD(P)H oxidase and oxidative stress, and suggesting that novel therapeutic strategies could be used. In addition, the functional characterization of another gene of the duplications, GTF2IRD2, has been partially achieved. GTF2IRD2 has been shown to interact with other related transcription factors, to display variable subcellular localization and to lack DNA binding properties. These results contribute to a better knowledge of the mutational and pathogenic mechanisms of the WBS.

Las especies reactivas del oxígeno (ROS), superóxido (O2o-), peróxido de hidrógeno (H2O2), y radical hidroxilo (OHo), se generan a partir de la reducción parcial del oxígeno molecular durante procesos metabólicos como la respiración o tras la exposición a ciertos agentes ambientales como las radiaciones UV. Estas ROS pueden reaccionar con biomoléculas como lípidos, proteínas y DNA e inactivar su función, por lo que las células han desarrollado actividades enzimáticas que se encargan de mantener niveles no-tóxicos de estos oxidantes. Se llama estrés oxidativo a la situación en la cual se produce un incremento en la concentración intracelular de ROS como consecuencia de un aumento en la generación o una disminución en la degradación de las mismas. En respuesta a estrés oxidativo, la célula activa rutas de señalización y factores de transcripción específicos que activan la expresión de proteínas antioxidantes encargadas de reestablecer los niveles redox intracelulares y de reparar los desperfectos causados por estos oxidantes. La levadura Schizosaccharomyces pombe es un organismo modelo ideal para el estudio de las respuestas a estrés oxidativo en las células eucariotas ya que posee sensores específicos a estrés oxidativo como el factor de transcripción Pap1 (pombe AP-1-like) y rutas de respuesta global a estrés, como las descritas en las células de mamífero, que son activadas por diferentes tipos de estrés. En el centro de esta ruta de respuesta global a estrés se encuentra la MAPK (Mitogen-activated protein kinase) Sty1. El factor de transcripción Pap1, de localización citoplasmática basal, se acumula en el núcleo en respuesta a estrés oxidativo. Este cambio de localización subcelular es debido a la inhibición del exporte nuclear dependiente de Crm1, aunque se desconocía el mecanismo molecular utilizado por este factor de transcripción para sensar y responder a oxidantes como H2O2 y dietilmaleto (DEM). Los resultados obtenidos indican que H2O2 oxida de forma reversible dos residuos de cisteína de Pap1 induciendo, seguramente, la formación de un puente disulfuro intramolecular, mientras que, DEM actúa como un agente alquilante que modifica de forma irreversible los residuos de cisteína del dominio C-terminal de Pap1. El gen que codifica para el factor de transcripción Pap1 fue aislado inicialmente como un gen que, en elevado número de copias, confería a las células un fenotipo de resistencia a ciertas drogas como estaurosporina. Esto es debido a que, tras acumularse en el núcleo en respuesta a estrés oxidativo, Pap1 activa la transcripción de genes implicados tanto en la respuesta antioxidante como en la resistencia a multidrogas. Todos aquellos genes que, al igual que pap1 fueron identificados por su implicación en la resistencia a multidrogas, codifican para proteínas que regulan la actividad del factor de transcripción Pap1. hba1 fue el único gen relacionado con resistencia a multidrogas, cuyo producto génico, una proteína con un dominio de unión a Ran (Ran-binding domain), Hba1, no había sido relacionado con la actividad de Pap1. Uno de los objetivos de mi trabajo experimental era el de determinar si Hba1 tenía un papel en la regulación de la actividad de Pap1. Nuestros resultados indican que la proteína Hba1, localizada en el nucleoplasma de la célula, participa en el exporte nuclear mediado por Crm1 de ciertas proteínas como el factor de transcripción Pap1 y la MAPK Sty1, aunque no de otras como la proteína PKI. Por ello, la pérdida de función de Hba1, por sobreexpresión o deleción del gen hba1, induce la localización nuclear constitutiva de Pap1 y Sty1 en ausencia de estrés. Esta localización nuclear de Pap1 es suficiente para la activación transcripcional de sus genes diana. Por lo tanto, el fenotipo de resistencia aumentada a multidrogas de las cepas en las que se ha perdido la actividad de la proteína Hba1, es debido a la acumulación de Pap1 en el núcleo en condiciones de no-estrés.

Protein nitration is considered as a marker of reactive nitrogen species formation. Heme oxygenases (HOs) are important for the defence against oxidative stress. We evaluated the involvement of the neuronal (nNOS), the endothelial (eNOS), and the inducible (iNOS) in nitrotyrosine formation and localitzation, and both the expression and funcional significance (HO inhibition and contractility studies) of HOs in sepsis-induced muscle contractile dysfunction. Sepsis was elicited by injecting rats and transgenic mice deficient in either nNOS, eNOS, or iNOS isoforms with E.Coli lipolysaccharide (LPS). Nitrotyrosine formation and HO expressions were assessed by immunoblotting. Oxidative stress was assessed measuring protein oxidation, lipid peroxidation, and muscle glutathione. We conclude that protein tyrosine nitration occurs in normal muscles, and sepsis-mediated increase in nitrotyrosine formation is limited to the mitochondria and membrane muscle fractions. The iNOS isoform is mostly involved in nitrotyrosine formation. HOs protect normal and septic muscles from the deleterious effects of oxidants., En un model de sepsi de disfunció diafragmàtica, s´ha avaluat el paper de les sintetases de l'òxid nítric (NOS) en la formació i localitzacio de 3-nitrotirosina, i l´expressió i significat biològic de les hemo oxigenases (HOs) (inhibidor de les HOs i estudis de contractilitat) davant l' estrès oxidatiu. La sepsi s'induí mitjançant injecció de 20 mg/kg del lipolisacàrid (LPS) d´Escherichia Coli a rates, i a ratolins deficients en les NOS induïble (iNOS), neuronal (nNOS) i endotelial (eNOS). Les proteïnes nitrificades i les HOs es van detectar amb anticossos específics. L' estrès oxidatiu s' avaluà mitjançant l' oxidació proteica, la peroxidació lipídica i el glutation muscular. Concloem que hi han proteïnes nitrificades en el múscul normal i aquestes s'incrementen durant la sepsi en les fraccions mitocondrial i membranar. L'isoforma iNOS és majorment responsable de la formació de nitrotirosina. Les HOs protegirien el múscul normal i sèptic dels efectes deleteris dels oxidants.
