OBJECTIVE: This work aimed to analyze the relative expression level of long intergenic non-protein coding ribonucleic acid 1503 (LINC01503) in cholangiocarcinoma tissues and cells, and to explore the effects of LINC01503 on cell proliferation, migration and invasion. PATIENTS AND METHODS: Logarithmic growth phase cholangiocarcinoma cells were selected and transfected with Lipofectamine 2000 (si-LINC01503, si-NC). The expression of LINC01503 was detected by quantitative Real Time-Polymerase Chain Reaction (qRT-PCR). The proliferation of cells was observed by 3-(4, 5-dimethyl-2-thiazolyl)-2, 5-diphenyl-2-H-tetra-zolium bromide (MTT) assay. Terminal deoxynucleotidyl transferase dUTP nick end labeling (TUNEL) assay was used to detect cell apoptosis. Transwell assay was used to observe the cell migration and invasion ability. RESULTS: The expression of LINC01503 was significantly increased in cholangiocarcinoma tissues compared with adjacent tissues (p<0.05), and the up-regulated expression of LINC01503 was associated with lymph node metastasis. Compared with normal bile duct cells (HIBEC), cholangiocarcinoma cells (RBE, QBC939) have higher expression of LINC01503, and si-LINC01503 transfection can effectively reduce the proliferation, migration, invasion and epithelial-mesenchymal transition (EMT) of cholangiocarcinoma cells. CONCLUSIONS: LINC01503 is highly expressed in cholangiocarcinoma and can effectively promote the proliferation, migration, invasion and EMT process of cancer cells, and LINC01503 is expected to be a potential biomarker for cholangiocarcinoma. OBJECTIVE: The aim of this study was to investigate the effects of retinoid X receptor α (RXRα) on the proliferation and apoptosis of pancreatic cancer cells through the transforming growth factor-β (TGF-β)/Smad signaling pathway. PATIENTS AND METHODS: The expression of RXRα in pancreatic cancer tissues and para-carcinoma tissues was detected via immunohistochemistry. Human pancreatic cancer PANC-1 cells were cultured and treated with RXRα in vitro. The apoptosis rate of cells was detected via flow cytometry. Furthermore, changes in the protein expression level of TGF-β/Smad signaling pathway were detected via Western blotting. RESULTS: The protein expression level of RXRα in pancreatic cancer tissues was significantly higher than that of para-carcinoma tissues. RXRα significantly promoted the proliferation and inhibited the apoptosis of pancreatic cancer cells. Moreover, RXRα could also activate the TGF-β/Smad signaling pathway. CONCLUSIONS: RXRα promotes the proliferation and inhibits the apoptosis of pancreatic cancer cells through the TGF-β/Smad signaling pathway. [ABSTRACT FROM AUTHOR]

OBJECTIVE: Previous research indicated that miR-144-3p was associated with the regulation of the carcinogenic processes, but the role of miR-144-3p in glioblastoma (GBM) remains unclear. In this study, we aimed to analyze the role of miR-144-3p in GBM. PATIENTS AND METHODS: The expression of miR-144-3p was measured in GBM tissues and adjacent non-malignant tissues using qRT-PCR. The correlation of miR-144-3p with clinicopathological features and prognosis was also analyzed. Cell proliferation, invasions, and migration assay were applied to assess the function of miR-144- 3p in vitro. Bioinformatics prediction and luciferase assays were employed to identify the predicted microRNA (miRNA) which regulates Frizzled-7 (FZD7). The levels of FZD7 and FZD7 mRNA were determined by Western blot and RT-PCR. RESULTS: The results showed that the miR-144-3p expression was significantly downregulated in tumor tissues and GBM cell lines compared with that in normal brain tissues and the normal human astrocytes. The levels of miR-144-3p were negatively correlated with the status of WHO grade and recurrence. Furthermore, patients with low serum levels of miR-144-3p had a significantly shorter median overall survival rate. Multivariate Cox regression analysis confirmed that low level of miR-144-3p expression predicted poor prognosis independently. Further function assays showed that miR-144-3p inhibited proliferation invasion and migration of GBM cells. Finally, miR-144-3p was demonstrated to bind to the wild-type 3' untranslated region of FZD7 but not with its mutant. CONCLUSIONS: The results of the present study indicate that miR-144-3p suppresses tumor metastasis by targeting FZD7. OBJECTIVE: Hepatitis C virus (HCV) infection is a high morbidity disease in China. The aim of this study was to explore the latest distribution of HCV genotypes in China, mainly focusing on epidemiological characteristics and risk factors of HCV genotype 1 in Chinese Han population. PATIENTS AND METHODS: 793 HCV-positive patients were enrolled from 21 hospitals across China. The association of epidemiological data with HCV genotype 1 and the potential risk factors was analyzed using univariate and multivariate logistic regression analysis. RESULTS: The HCV genotype 1b was detected in 66.9% of 793 patients. The way of infection, disease course, HCV RNA level, history of blood transfusion and smoking were found to be significantly different between patients infected with HCV genotype 1 and non-genotype 1. The result of univariate logistic regression analysis showed that HCV genotype 1 infection has a close relationship with age, gender, smoking history, the way of infection, allergy, disease course, and HCV RNA level. Moreover, the HCV RNA level, disease course, and blood transfusion history were the related risk factors for infection of HCV genotype 1 as demonstrated by multivariate logistic regression analysis. CONCLUSIONS: The HCV genotype 1 was still the main genotype of HCV infection in China. We should intensify HCV screening for those people who aged over 50 years and had a history of blood transfusion in China.
