Baseline blood chemistry data could be particularly valuable if reference values from free-ranging populations of rare or endangered species are not available. The Canada lynx (Lynx canadensis) is listed as threatened under the Endangered Species Act in the conterminous United States, even though the species is managed as a furbearer in Alaska and in most provinces of Canada. Body mass, blood chemistry, and hematologic data for free-ranging lynx were collected from 2003 to 2007 and for captive lynx from 1984 to 2007. Up to 2 yr of age, captive lynx were consistently heavier than free-ranging lynx. Body mass of adult free-ranging lynx was similar to body mass of captive adult lynx. Some differences in blood chemistry between free-ranging and captive lynx were statistically significant, but most measured values were within reference ranges for domestic cats. Free-ranging lynx had higher concentrations of aspartate aminotransferase, alanine aminotransferase, and blood urea nitrogen than did captive lynx, and these were outside the reference value ranges for domestic cats. Alkaline phosphatase and phosphorus were higher in juveniles (<12 mo when captured) as compared to adults. Free-ranging lynx maintained body mass between serial captures. Hematologic values, blood chemistry values, and body mass of free-ranging Canada lynx provide support for the hypothesis that Canada lynx in Minnesota, at the southern edge of their range, are in normal physical condition.

Plasma protein electrophoresis is recognized as a reliable diagnostic tool in avian medicine; however, the influence of circannual phenomena such as molt on protein electrophoregrams is poorly documented. The molt is a period of heavy hormonal and metabolic change in birds. The purpose of this study was to investigate the effects of molt on total protein concentration and electrophoresis patterns in birds. Blood samples were taken from 19 Bar-headed Geese (Anser indicus) from mid-May to mid-August, at 15-day intervals. At the same time, molting stage of each bird was recorded. Total protein concentrations were measured and plasma agarose gel electrophoresis was performed on these samples. The Bar-headed Goose was chosen as a model, because they molt over a very short period. The total protein concentration and albumin, alpha-2, beta, and gamma fractions were at their minimum values during molt, whereas the prealbumin and alpha-1 fractions rose to their maximum levels. This study provides baseline information relevant to changes occurring in avian proteinograms throughout the molt. The increase in prealbumin and alpha-1 fractions may be related to an increase in plasma thyroid hormones during molt. The decrease observed in albumin, alpha-2, beta, and gamma fractions may be related to protein and energy shifts toward feather growth, as well as to an expansion of the circulatory system located around the feather follicles with secondary dilutional effects on protein fractions. From a clinical point of view, the observed changes associated with molting were less significant than initially expected, and would not likely results in incorrect diagnoses based on interpretation of the protein electrophoretic patterns.

Protein electrophoresis is recognized as a reliable diagnostic tool for birds even though results are seldom pathognomonic. Unfortunately, this technique is underused in avian medicine because many factors interfere with electrophoresis patterns; hemolysis is one of these factors and is often associated with improper specimen handling. In human laboratory medicine, hemolysis is a known interference factor that can lead to erroneous results. Published data on the influence of hemolysis on protein electrophoresis in birds is currently restricted to a single study in Psittacidae. The aim of this study was to further investigate this effect and to analyze potential interspecific differences. Blood samples were drawn from 28 Black Kites (Milvus migrans) and 19 Bar-headed Geese (Anser indicus) and separated into two aliquots. One aliquot was dipped into liquid nitrogen for 5 sec in order to cause freeze-thawing hemolysis before centrifugation. Total plasma protein concentration, plasma hemoglobin concentration, and plasma protein electrophoresis patterns were determined for both hemolyzed and nonhemolyzed samples. In both species, hemolysis resulted in falsely elevated total plasma protein concentration. In Bar-headed Geese, hemolysis caused a rise in the gamma fraction. In Black Kites, this rise involved not only the gamma fraction but also the beta fraction, stressing the potential for species-related differences. In both species, the effects of hemolysis mimicked a chronic inflammatory condition with resulting antigenic stimulation.

As part of a large-scale disease screening program, blood samples were collected from 534 African buffalo (Syncerus caffer) in South Africa's Hluhluwe-iMfolozi Park in October 2005 and May 2006 to establish age- and sex-specific reference intervals for erythrogram and leukogram values. Sixty-seven of the animals were positive for bovine tuberculosis (TB), allowing for comparisons between TB-positive and TB-negative groups. Positive animals had basopenia and slight lymphopenia compared to TB-negative animals. Blood values were compared to those reported for captive African buffalo, American bison (Bos bison), and cattle (Bos taurus). The free-ranging buffalo sampled in this study had higher white blood cell counts than captive buffalo, and this difference was driven by lymphocytes. Free-ranging buffalo also had higher red blood cell counts, mean corpuscular hemoglobin concentration (MCHC), white blood cell counts, neutrophils and lymphocytes, and lower mean corpuscular volume (MCV) than cattle. Demographic and environmental factors strongly affected hematologic values in the study population. Older animals had significantly higher hemoglobin, hematocrit, MCV, and mean corpuscular hemoglobin (MCH), while younger animals had a higher red blood cell count, red cell distribution width (RDW), and white blood cell count, which was due to lymphocytes and basophils. Females had a higher hemoglobin concentration, hematocrit, MCV, MCH, and basophils than males. At the end of the wet season, hemoglobin, red blood cell count, hematocrit, MCHC, RDW, white blood cell count, and neutrophils were all significantly higher, while basophils and MCV were lower, than at the end of the dry season. Our results emphasize the need to use species-specific data when interpreting hematologic values and point to important differences in hematology between captive and free-ranging animals of the same species. Strong variability in hematologic values with animal age and sex, season, and herd affiliation indicates that ''normal'' hematologic values in wild animals vary throughout their lives and subject to fluctuating environmental conditions.

Haptoglobin (Hp) is an acute-phase protein synthesized in the liver that circulates at elevated concentrations in response to tissue damage caused by inflammation, infection, and trauma. As part of a larger study, sera Hp concentrations were measured in temporarily captive (n = 21) and free-range (n = 38) western stock juvenile Steller sea lions (Eumetopias jubatus) sampled from 2003 to 2006. Baseline Hp concentration at time of capture was 133.3 +/- 17.4 mg/dl. Temporarily captive animals exhibited a 3.2-fold increase in Hp concentrations during the first 4 wk of captivity, followed by a return to entry levels by week 5. Haptoglobin levels were not influenced by age, season, or parasite load. There was a significant positive correlation between Hp concentrations and white blood cell count (P < 0.001) and globulin levels (P < 0.001) and a negative correlation to red blood cell count and hematocrit (P < 0.001 for both). There was no correlation between Hp levels and platelet count (P = 0.095) or hemoglobin (P = 0.457). Routine blubber biopsies collected under gas anesthesia did not produce a measurable Hp response. One animal with a large abscess had an Hp spike of 1,006.0 mg/dl that returned to entry levels after treatment. In conclusion, serum Hp levels correlate to the stable clinical health status observed during captivity, with moderate Hp response during capture and initial acclimation to captivity and acute response to inflammation and infection.

The objectives of this study were to compare the hematology and serum chemistry values between free-ranging and stranded harbor seal (Phoca vitulina richardsi) pups and to ascertain how blood values of stranded pups changed during the rehabilitation process. Coincident with these comparisons, reference values were obtained for free-ranging pups. Stranded harbor seal pups (n = 28) recovered from areas between Pebble Beach and Moss Landing, California (USA) were admitted to The Marine Mammal Center, Sausalito, from March to May 1995, 1996, and 1998. Blood samples were collected from harbor seal pups before and after rehabilitation. As a control group, wild harbor seal pups were captured at Pebble Beach and Elkhorn Slough (n = 42) during the 1995, 1996, and 1998 pupping seasons. Mean eosinophil and calcium values of wild pups were significantly greater than those of newly admitted pups, whereas mean bands, aspartate aminotransferase, alanine aminotransferase, total bilirubin, and chloride values were significantly lower (P < or = 0.05). Mean neutrophil, band, lymphocyte, eosinophil, basophil, calcium, phosphorus, blood urea nitrogen, potassium, total protein, and globulin values of rehabilitated pups increased significantly after 2-3 mo in captivity, whereas, mean red blood cell, hemoglobin, hematocrit, cholesterol, and total bilirubin values decreased significantly (P < or = 0.05).

Several disease syndromes in captive rhinoceroses have been linked to low vitamin status. Blood samples from captive and free-ranging black (Diceros bicornis) and white rhinoceros (Ceratotherium simum) and tissue samples of captive individuals from four rhinoceros species were analysed for vitamins A and E. Circulating vitamin A levels measured as retinol for free-ranging versus captive black and white rhinoceros were 0.04 (+/- 0.03 SD) vs. 0.08 (+/- 0.08) and 0.07 (+/- 0.04) vs. 0.06 (+/- 0.02) microgram/ml, respectively. Circulating vitamin E levels measured as alpha-tocopherol were 0.58 (+/- 0.30) vs. 0.84 (+/- 0.96) and 0.62 (+/- 0.48) vs. 0.77 (+/- 0.32) microgram/ml, respectively. In contrast to earlier findings, there was no significant difference in vitamin E concentration between captive and free-ranging black rhinoceros. When the samples of captive black rhinoceros were grouped into those taken before 1990 and after 1990, however, those collected before 1990 had significantly lower (P < 0.001) vitamin E levels (0.46 +/- 0.83 microgram/ml) and those collected in 1990 or later significantly higher (P < 0.001) vitamin E levels (1.03 +/- 1.04 micrograms/ml) than the captive population as a whole. This is probably due to increased dietary supplementation. There were significant differences in circulating vitamin concentrations in black rhinoceroses from different regions in the wild. Serum 25-hydroxy (OH) vitamin D3 averaged 55.7 ng/ml in free-ranging rhinoceroses; no carotenoids were detected in any blood samples. Captive black and white rhinoceroses appear to be adequately supplemented in vitamin A and E. Captive Indian rhinoceroses (Rhinoceros unicornis) had significantly lower vitamin A concentrations in blood (P < 0.001) and higher vitamin A concentrations in liver tissue samples (P < 0.001) than other rhinoceros species. Equine requirements are not recommended as a model for rhinoceros vitamin requirements.

Hematologic and serum chemistry values were determined for 20 adult captive European wildcats (Felis silvestris) in Lleida (Catalonia, Spain). Seven wildcats (4 females and 3 males) were captured in the wild and 13 (4 females and 9 males) were born and raised in captivity. Samples were collected between September and December from 1993 to 1998. Blood was obtained by jugular venipuncture after administration of either ketamine and xylazine or ketamine and medetomidine. Females had significantly higher mean eosinophil counts, albumin concentration, (and A/G ratio) and lower mature neutrophil counts, although these differences were not clinically relevant. Results for many of the blood parameters fall within the reference range for domestic cats. Mean values for alanine aminotransferase, aspartate aminotransferase, creatine phosphokinase, and lactate dehydrogenase activities as well as blood urea nitrogen, glucose, and sodium concentrations were higher than the upper limit of the reference range for domestic cats.

We present baseline values for 12 hematologic and 17 serum chemistry parameters collected from 22 captive lynx (Felis lynx canadensis) in December 1992, at Ronan, Montana (USA). There were no significant differences in hematologic parameters between yearlings and adults or between sexes. Lynx originally captured in the wild had significantly higher mean (+/- SE) counts of neutrophils (7.7 +/- 0.37 x 10(3) versus 7.2 +/- 0.35 x 10(3)) and lower counts of lymphocytes (1.1 +/- 0.05 x 10(3) versus 1.6 +/- 0.08 x 10(3)) compared to lynx born and raised in captivity. Yearling lynx had significantly higher values for alkaline phosphatase than adults (51.0 +/- 6.0 IU/l versus 17.5 +/- 0.8 IU/l.

Eight free-range koalas (Phascolarctos cinereus) were captured on Kangaroo Island (South Australia) and later transported to the Adelaide Zoological Gardens. Blood samples were collected at the time of capture and at 6 hr, 24 hr and 7 days later. Routine hematological analyses were performed and plasma cortisol levels determined. Significant differences were observed between sampling times for erythrocyte numbers, hemoglobin concentrations, packed cell volumes, mean cell volumes, mean cell hemoglobin concentrations, numbers of leukocytes, neutrophils, lymphocytes and lymphocyte:neutrophil ratio (P less than 0.05). No significant differences were detected between sampling periods in the numbers of monocytes (P = 0.707), eosinophils (P = 0.174) or plasma cortisol levels (P = 0.192).

Normal values and ranges for 31 clinical hematology and serum chemistry tests are reported for the beluga or white whale (Delphinapterus leucas). The values were collected over a 6-yr period from eight belugas maintained for display at Sea World (San Diego, California, USA) facilities and represent long-term evaluations for each animal in a controlled environment. They represent the first report for a number of serum chemistry values for the beluga. Normal values such as these provide an important data base from which to detect diagnostically important changes in health status for belugas in a zoological setting. They also establish a baseline from which to evaluate differences in normal values in free-ranging belugas and from which to diagnose disease problems in wild populations.

Fifty-eight immobilizations were conducted using 21 lions (Panthera leo) and 4 drug combinations. The combinations used were ketamine-phencyclidine-promazine, xylazine-phencyclidine-promazine, xylazine-ketamine-phencyclidine-promazine, and tiletamine-zolazepam.

The mean plasma level of alpha-tocopherol (vitamin E) measured in 31 free-ranging black rhinoceros (Diceros bicornis) was significantly higher (P less than 0.001) than that in 11 captive animals (means +/- SE = 0.77 +/- 0.05 and 0.18 +/- 0.03 micrograms/ml, respectively). Vitamin E status may influence the health of captive black rhinoceros; in particular, it may be linked to hemolytic anemia commonly observed in these animals in captivity.
