Aims/Introduction: To investigate the association between specific bacterial pathogens and treatment outcome in patients with limb‐threatening diabetic foot infection (LT‐DFI). Materials and Methods: Consecutive patients treated for LT‐DFI in a major diabetic foot center in Taiwan were analyzed between the years 2014 and 2017. Patients with positive wound culture results at first aid were enrolled. Clinical factors, laboratory data, and wound culture results were compared. Lower‐extremity amputations and in‐hospital mortality were defined as a poor outcome. Results: Among the 558 patients, 272 (48.7%) patients had lower extremity amputation and 22 (3.9%) patients had in‐hospital mortality. Gram‐negative bacterial (GNB) infection was the independent factor following factors adjustment. When all the 31 microorganisms were analyzed, only E. coli (adjusted odds ratio [aOR], 3.01; 95% CI, 1.60–5.65), Proteus spp. (aOR, 2.99; 95% CI, 1.69–5.29), and Pseudomonas aeruginosa (aOR, 2.00; 95% CI 1.20–3.32) were associated with poor outcome. The analysis of specific GNB species in association with major‐ or minor‐ amputation have been reported. No specific pathogen was associated with cause of death in patients with mortality within 30 days. The antimicrobial‐resistant strains were not associated with a poor treatment outcome. Conclusions: The presence of GNB was associated with limb amputations. This study provides insight into more timely and appropriate management of the diabetic foot infection.

Ultrafine CoO particles immobilized into the mesopores of three-dimensional cubic bimodal ordered mesoporous carbon CMK-9 is successfully prepared by using a combination of nanocasting and wet-impregnation methods. It is found that the cubic bimodal interconnected mesoporous framework of CMK-9 plays a crucial role in achieving the excellent electrochemical performances by assisting the rapid mass and charge transfer. Among the prepared nanocomposites, CoO(10)@CMK-9 delivers a discharge capacity of 830 mAh g-1 after 200 cycles at a current density of 100 mA g-1 in lithium-ion batteries. At a higher current density of 1000 mA g-1 , the anode presents an outstanding discharge capacity of 636 mAh g-1 after 200 cycles. In sodium-ion batteries, the anode provides a discharge capacity of 296 mAh g-1 after 250 cycles at a current density of 100 mA g-1 . The remarkable performances of CoO(10)@CMK-9 demonstrate the promising potentials of the nanocomposite as the anode for rechargeable batteries.

Lung cancer is the most commonly diagnosed cancer worldwide, and metastasis in lung cancer is the leading cause of cancer‐related deaths. Thus, understanding the mechanism of lung cancer metastasis will improve the diagnosis and treatment of lung cancer patients. Herein, we found that expression of cluster of differentiation 109 (CD109) was correlated with the invasive and metastatic capacities of lung adenocarcinoma cells. CD109 is upregulated in tumorous tissues, and CD109 overexpression was associated with tumor progression, distant metastasis, and a poor prognosis in patient with lung adenocarcinoma. Mechanistically, expression of CD109 regulates protein kinase B (AKT)/mammalian target of rapamycin (mTOR) signaling via its association with the epidermal growth factor receptor (EGFR). Inhibition of CD109 decreases EGFR phosphorylation, diminishes EGF‐elicited activation of AKT/mTOR, and sensitizes tumor cells to an EGFR inhibitor. Taken together, our results show that CD109 is a potential diagnostic and therapeutic target in lung cancer patients., CD109 promotes lung cancer metastasis through promoting EGFR‐AKT‐mTOR signaling and CD109 is an independent prognostic marker for lung adenocarcinoma.

Excessive extraction of groundwater resulting in serious land subsidence as well as intensified rainfall and storm surges due to climate change complicate the flooding problems in southwest Taiwan. A coupled set of different models was proposed to analyze the effect of inundation risk considering land subsidence and climate change. Three models, including the groundwater flow model, land subsidence model, and the physiographic drainage–inundation model, were used in this study, enabling simulations of different considerations. The results revealed that more severe flooding would result from land subsidence and climate change. The findings of a 21% increase in flood area with an inundation depth greater than 1.5 m for 200‐yr return period events clearly showed more severe flooding would result from land subsidence. The flooding in those severely subsiding areas would increase in a range from 3.4 to 21.5% when further considering climate change. While the simulation results revealed that the flood area could be decreased by as much as 50% taking into account the implemented policies, the coastal region would still be exposed to a high risk of being flooded. It thus suggests that policies focusing on infrastructure would be insufficient, and river basin management as well as spatial planning should be investigated further. The tumor microenvironment plays an important role in tumor initiation and progression. It is well documented that nicotine participates in cigarette smoking-related malignancies. Previous studies focused on the effects of nicotine on tumor cells; however, the role of the microenvironment in nicotine-mediated tumorigenesis is poorly understood. Herein, we investigated the effect and molecular mechanism of nicotine on fibroblasts and its contribution to breast cancer. We found that nicotine induced the epithelial-mesenchymal transition (EMT) of breast cancer cells and promoted activation of fibroblasts. Interestingly, conditioned medium from nicotine-activated fibroblasts (Nic-CM) had a greater impact on promoting the EMT and migratory capability toward cancer cells than did treatment with nicotine alone. Production of connective tissue growth factor (CTGF) and transforming growth factor (TGF)-β by nicotine-treated fibroblasts was demonstrated to be crucial for promoting the EMT and cancer cell migration, and blocking of CTGF and TGF-β in Nic-CM-suppressed tumor motility. Moreover, nicotine induced expressions of CTGF, and TGF-β in fibroblasts as identified through α7 nicotinic acetylcholine receptor (nAChR)-dependent activation of the AKT/TAZ signaling mechanism. Together, our data showed for the first time that activation of fibroblasts is largely responsible for accelerating smoking-mediated breast cancer progression.

Mutation of KRAS in non-small-cell lung cancer (NSCLC) shows a poor response to epidermal growth factor receptor (EGFR) inhibitors and chemotherapy. Currently, there are no direct anti-KRAS therapies available. Thus, new strategies have emerged for targeting KRAS downstream signaling. Panobinostat is a clinically available histone deacetylase inhibitor for treating myelomas and also shows potentiality in NSCLC. However, the therapeutic efficacy of panobinostat against gefitinib-resistant NSCLC is unclear. In this study, we demonstrated that panobinostat overcame resistance to gefitinib in KRAS-mutant/EGFR-wild-type NSCLC. Combined panobinostat and gefitinib synergistically reduced tumor growth in vitro and in vivo. Mechanistically, we identified that panobinostat-but not gefitinib-inhibited TAZ transcription, and the combination of panobinostat and gefitinib synergistically downregulated TAZ and TAZ downstream targets, including EGFR and EGFR ligand. Inhibition of TAZ by panobinostat or short hairpin RNA sensitized KRAS-mutant/EGFR-wild-type NSCLC to gefitinib through abrogating AKT/mammalian target of rapamycin (mTOR) signaling. Clinically, TAZ was positively correlated with EGFR signaling, and coexpression of TAZ/EGFR conferred a poorer prognosis in lung cancer patients. Our findings identify that targeting TAZ-mediated compensatory mechanism is a novel therapeutic approach to overcome gefitinib resistance in KRAS-mutant/EGFR-wild-type NSCLC.

All-trans retinoic acid (ATRA), a derivative of retinoid, is involved in the onset of differentiation and apoptosis in a wide variety of normal and cancer cells. MicroRNAs (miRNAs) are small non-coding RNAs that control gene expression. Several miRNAs were identified to participate in ATRA-mediated cell differentiation. However, no studies have demonstrated whether miRNA can enhance ATRA cytotoxicity, thereby resulting in cell apoptosis. This study investigated the effects of ATRA-mediated miRNA expression in activating apoptotic pathways in glioblastoma. First, we found that high-dose ATRA treatment significantly reduced cell viability, caspase-dependent apoptosis, endoplasmic reticular (ER) stress activation, and intracellular reactive oxygen species accumulation. From microarray data, miR-302b was analyzed as a putative downstream regulator upon ATRA treatment. Furthermore, we found that ATRA up-regulated miR-302b expression in a dose- and time-dependent manner through retinoic acid receptor α-mediated pathway. Overexpression and knockdown of miR-302b significantly influenced ATRA-mediated cytotoxicity. E2F3, an important transcriptional regulator of glioma proliferation, was validated to be a direct target gene of miR-302b. The miR-302b-reduced E2F3 levels were also identified to be associated with ATRA-mediated glioma cell death. These results emphasize that an ATRA-mediated miR-302b network may provide novel therapeutic strategies for glioblastoma therapy. We propose that high-dose all-trans retinoic acid (ATRA) treatment, a derivative of retinoid, significantly induces glioblastoma cell apoptosis via caspase-dependent apoptosis, endoplasmic reticular (ER) stress, and intracellular reactive oxygen species (ROS) accumulation. The miR-302b overexpression enhanced by ATRA-mediated retinoic acid receptor (RAR)α pathway was also identified. The E2F3 repression, a novel target gene of miR-302b, was involved in ATRA-induced glioblastoma cell cytotoxicity.

Solid solutions of mixed metal dodecaborides of ZrB12, YB12, and ScB12 were prepared by arc-melting and studied for their mechanical properties. Zr1–xYxB12 formed an essentially perfect solid solution, closely following Vegard’s law. Zr1–xScxB12 and Y1–xScxB12 undergo a face centered-cubic to body-centered tetragonal transition at 90–95 at. % Sc as determined by powder X-ray diffraction and transmission electron microscopy. The compounds Zr0.5Y0.5B12, Zr0.5Sc0.5B12, and Y0.5Sc0.5B12 are superhard (Vickers hardness ≥ 40 GPa) and demonstrate an increase in hardness to 45.8 ± 1.3, 48.0 ± 2.1, and 45.2 ± 2.1 GPa under a load of 0.49 N, respectively, compared to 40.4 ± 1.8, 40.9 ± 1.6, and 41.7 ± 2.2 GPa for pure ZrB12, YB12, and ScB12, respectively. In addition, Zr0.5Y0.5B12, Zr0.5Sc0.5B12, and Y0.5Sc0.5B12 solid solutions show a substantial increase in oxidation resistance to approximately 630, 685, and 695 °C, respectively, when compared to other superhard metal borides (e.g., ∼400 °C for WB4) and their all...

The rapid progress of information techniques enables people to have a relatively convenient environment and life. This study designs and develops the Web2.0-based Home Appliances controL service platform (WHALE) for users to easily control home appliances. WHALE is a three-tier architecture, which consists of Device Detection and Registration Service Platform (DDRSP), Web2.0 Service Platform (WSP), and Common Service Platform (CSP). WHALE is based on the standards of Devices Profile for Web Services (DPWS), Open Services Gateway initiative (OSGi), and Web2.0 techniques. Users can use many kinds of handheld devices to get services from WSE When DDRSP finds a new home appliance, this home appliance will be registered into CSE CSP records the related information and services which are provided by this home appliance. WSP not only integrates and shows the related services of all home appliances but also provides controlling interfaces to control the actions of all home appliances. All delivered messages are based on Simple Object Access Protocol (SOAP) and Web Services Description Language (WSDL). Furthermore, for the Web QoS (Quality of Service) strategy, this study designs and proposes an Intelligent Web quality of Service-aware maTching (iWIST) model to satisfy different characteristics of the user's requirements, iWIST consists of Components Classifier, Admission Controller, and Dynamic Content Adaptor. Component Classifier can classify users' requesting components with different QoS constraints. Admission Controller determines the maximum available components for the user. Dynamic Content Adaptor dynamically delivers component information to users. Experiment results illustrate that WHALE can provide appropriate contents with low response time to users. Finally, WHALE has integrity, scalability, operability, adaptivity, and extensibility functions. It can be a reference model for developing home appliance controllers in the future.

Environmental protection has taken serious notice of global warming. The Intergovernmental Panel on Climate Change (IPCC) has proved that CO2 is one of the factors for global warming. Furthermore, the majority of this emission comes from the vehicles using petroleum fuels. For this reason, alternative fuels become a very popular issue. There are many automakers and researchers endeavoring to develop alternative fuel vehicles (AFVs). There already have been many types of alternative fuels used in vehicles, such as electric vehicles (EVs), hybrid electric vehicles (HEVs), fuel cell (hydrogen) vehicles, natural gas vehicles (CNG), methanol vehicles, ethanol vehicles, and bio-diesel vehicles, etc. Different types of the alternative fuels have different characteristics to which attention should be paid. It is difficult for users to identify which one is the best choice for them to use. This problem concerns the characteristics of each alternative fuel. A lot of criteria and aspects related to the alternative fuels should be considered at the same time. In this study, we try to apply the Decision-Making Trial and Evaluation Laboratory (DEMATEL) method to deal with the importance and causal relationships among the evaluation factors of AFVs. Results show the top three critical evaluation criteria are ‘new car cost’, ‘cruising range’, and ‘fuel efficiency’; ‘air pollution’ is the key cause factor influencing others; and ‘cruising range’ is the key effect factor influenced by others. It is no accident that cost is the most critical factor for users. Consequently, it is suggested to automakers and researchers to carefully analyze the characteristics of each alternative fuel. Fuel efficiency is not only focused on by users, but also it will affect the cruising range. Air pollution is what users care about, so even zero-emission vehicles (ZEVs), which cost more but have lower emission and higher fuel efficiency, could be accepted. There seems to be no perfect AFV, but to endeavor on ZEVs will reduce some disadvantages. Copyright © 2010 John Wiley & Sons, Ltd.

Two different hydrogel-coated streptavidin (SAv) piezoelectric chips were investigated. One was directly prepared by immobilizing SAv molecules covalently onto a dextran-modified crystal, and the other one was indirectly prepared by physically adsorbing SAv onto a biotin-linked dextran surface. The covalent preparation yielded 80% more SAv-binding and better subsequent adsorption of biotinylated bovine serum albumin (bBSA). Both chips displayed the best binding affinity with bBSA at pH 5.0 in a flow injection analysis and exhibited reproductive real-time response during layer-by-layer assembly of a bBSA and SAv multilayer film. In the multilayer assembly, approximately 3-7 SAv molecules were captured by each immobilized bBSA, and the estimated apparent KD values of the binding of flowing bBSA with surface SAv were 0.24 and 0.11 microM in the first two cycles of the covalently prepared chip, respectively. Two Escherichia coli cells, each flagellum-displaying Strep-tag I and Strep-tag II, respectively, were selectively detected by both kinds of SAv chips. These studies suggest the potential application of both chips in real-time screening SAv affinity ligands from a cell-display random peptide library.

Among 13 different cell lines, gossypol (GOS) showed the most potent cytotoxic effect against human colorectal carcinoma cells including HT29, COLO205, COLO320HSR and COLO320DM cells according to an MTT assay. The cytotoxic effect of GOS was mediated by its induction of apoptosis as characterized by the occurrence of DNA ladders, apoptotic bodies and chromosome condensation in both COLO205 and HT29 cells. Activation of caspase 3, 6, 8 and 9, but not caspase 1, accompanied by the appearance of cleaved fragments of PARP (85 kDa), and caspase 3 (p17/p15), was identified in GOS-treated cells. Decreases in Bcl-xL and phosphorylated Bad proteins were found in GOS-treated cells. GOS induction of ROS production was detected by in vitro plasmid digestion, and an increase in the intracellular peroxide level was observed in GOS-treated COLO205 cells by the DCHF-DA assay. Antioxidants including N-acetyl-L-cysteine (NAC), catalase (CAT), tempol (TEM) and melatonin (MEL), but not allopurinol (ALL), pyrrolidine dithiocarbamate (PDTC) or diphenylene iodonium (DPI), significantly inhibited GOS-induced Reactive oxygen species (ROS) production through blocking the occurrence of apoptosis. GOS induced mitochondrial dysfunction characterized by a loss of the mitochondria membrane potential via DiOC6 staining, and the release of cytochrome c (Cyt c) and apoptosis-inducing factor (AIF) from mitochondria to the cytoplasm was observed. Removing mitochondria by ethidium bromide (EtBr) treatment significantly reduced the apoptotic effect of GOS in COLO205 cells. Furthermore, an intraperitoneal injection of GOS or gossypol acetic acid (GAA) significantly reduced the growth of colorectal carcinoma induced by a subcutaneous injection of COLO205 cells in nude mice. Results of the present study provide the first evidences demonstrating the in vitro and in vivo antitumor effects of GOS via an ROS-dependent mitochondrial apoptosis in colorectal carcinoma.

A high-efficiency white organic light emitting diode with a power- efficiency of 34.9 lm/W (EQE =22.8 %) at 100 cd/m2, and 24.5 lm/W (EQE =18.8 %) at 1,000 cd/m2 will be presented. The high efficiency may be attributed to the use of electroluminescence efficient materials and effective host-to- guest energy transfer.
