Abstract

Abstract EMT dysfunction is a dominant mechanisms of hypospadias. Thus, identification of EMT‐related lncRNAs based on transcriptome sequencing data of hypospadias might provide novel molecular markers and therapeutic targets for hypospadias. First, the microarray data related to hypospadias were downloaded from Gene Expression Omnibus (GEO). Besides, the differentially expressed lncRNAs and messenger RNAs (mRNAs) related to EMT were screened to construct lncRNA‐mRNA co‐expression interaction pairs. In addition, the microRNA (miRNA) prediction analysis was performed through bioinformatics methods to construct a ceRNA network. Moreover, function prediction and function enrichment and pathway analyses were also performed. Finally, the core EMT‐related lncRNAs were verified based on mRNA expression changes and cell functions. A total of 6 EMT‐related lncRNAs were identified and 123 mRNA‐lncRNA co‐expression interaction pairs were screened in this study. Additionally, a ceRNA regulatory network comprising 17 mRNAs, 4 lncRNAs, and 28 miRNAs was constructed based on the prediction of hypospadias‐related miRNAs. The validation results of the dataset GSE121712 revealed that only BEX1 was positively correlated with the expression of the lncRNA GNAS‐AS1 (r = 0.874, P < 0.01), both of which had high expression. The cell experiment results demonstrated that interfering with the expression of GNAS‐AS1 significantly promoted the proliferation, migration, and EMT of cells. Importantly, it was confirmed that GNAS‐AS1 can serve as a ceRNA and play an important role in the EMT of hypospadias. Hence, it may be considered as a potential target in the treatment of this disease.

Abstract

Abstract Familial hemophagocytic lymphohistiocytosis (FHL) is a fatal autosomal recessive disorder that often occurs in infants and young children, and rarely reported in adults. In this paper, we retrospectively reported an elderly woman with recurrent fever, this patient was diagnosed with familial hemophagocytic syndrome by perfecting relevant examinations after admission, improvement was observed after standard chemotherapy. In order to further determine the possible underlying genetic causes, we performed gene mutation analysis and found that there were compound heterozygous missense mutations c.133G > A (p.Gly45Arg) and c.147C > A (p.Asp49Glu) on the exon2 of PRF1 gene in this patient. According to the clinical manifestations and test results, the patient was further confirmed as late‐onset FHL‐2 type. Without a suitable donor, the patient did not perform hematopoietic stem cell transplantation. Therefore, the relevant genetic examination should be performed as early as possible in young patients with a family history of hemophagocytic lymphohistiocytosis, and it can provide a basis for etiological diagnosis and radical treatment by hematopoietic stem cell transplantation. It is essential to further study the molecular mechanism and clinical application value for late‐onset elderly FHL patients without appropriate donors.

Abstract

Abstract The exponentially increasing heat generation in electronic devices, induced by high power density and miniaturization, has become a dominant issue that affects carbon footprint, cost, performance, reliability, and lifespan. Liquid metals (LMs) with high thermal conductivity are promising candidates for effective thermal management yet are facing pump‐out and surface‐spreading issues. Confinement in the form of metallic particles can address these problems, but apparent alloying processes elevate the LM melting point, leading to severely deteriorated stability. Here, we propose a facile and sustainable approach to address these challenges by using a biogenic supramolecular network as an effective diffusion barrier at copper particle‐LM (EGaIn/Cu@TA) interfaces to achieve superior thermal conduction. The supramolecular network promotes LM stability by reducing unfavorable alloying and fluidity transition. The EGaIn/Cu@TA exhibits a record‐high metallic‐mediated thermal conductivity (66.1 W m−1 K−1) and fluidic stability. Moreover, mechanistic studies suggest the enhanced heat flow path after the incorporation of copper particles, generating heat dissipation suitable for computer central processing units, exceeding that of commercial silicone. Our results highlight the prospects of renewable macromolecules isolated from biomass for the rational design of nanointerfaces based on metallic particles and LM, paving a new and sustainable avenue for high‐performance thermal management.

Abstract

Abstract The measurement noise covariance R plays a vital role in the Kalman filter (KF) algorithm. Traditionally, a constant R is obtained by experience or experiments. However, the KF cannot achieve optimal estimation using constant R under non‐Gaussian conditions. A robust strategy for adaptive estimation of R is proposed to suppress the influence of non‐Gaussian noise on the in‐motion alignment performance of the Doppler velocity log (DVL) velocity‐aided strapdown inertial navigation system (SINS). Furthermore, an improved Sage–Husa robust adaptive KF algorithm (SHRAKF) based on the Mahalanobis distance (MD) algorithm is proposed to handle the outliers that frequently occur within the complicated underwater environment. The contributions of this work are twofold—the SHRAKF (1) designs a robust strategy to adaptively estimate R in real time and (2) further improves filtering robustness and adaptability with the MD algorithm, conditional on the DVL outputs being contaminated by non‐Gaussian noise. A semi‐physical simulation experiment for SINS/DVL in‐motion alignment based on the test data is carried out, and the experimental results show that the SHRAKF adaptively estimates R in real time and effectively suppresses observational outliers. For non‐Gaussian noise pollution, including outliers and heavy‐tailed noise, the SHRAKF performs better than traditional methods.

Abstract

Abstract Hunting for the wild meat trade, medicines and other human uses has decimated Indo‐Burma's vertebrate biota and has led to widespread defaunation. Yet, there is surprisingly little data on how hunting impacts wild bird assemblages in different landscapes here. Based on concurrent snapshot surveys of bird hunting, food markets and hunting attitudes across six Indo‐Burma countries, we found that hunting threatens species not only in forested landscapes but also wetlands and farmlands such as orchards and paddy fields—ecosystems overlooked by past studies, with at least 47 species associated with wetlands and agricultural lands identified from market surveys across the region. High rates of mortality are suffered when hunting tools such as nets are used to exclude perceived bird pests in both aquaculture and agricultural landscapes, with over 300 individual carcasses of at least 29 identifiable species detected in one aquaculture landscape sampled in Thailand. We warn that the potentially unsustainable trapping of species for consumption and trade in Indo‐Burma, coupled with high incidental mortalities, could decimate the populations of erstwhile common and/or legally unprotected species. There is an urgent need for stronger regulatory oversight on the hunting take of wild birds and the use of hunting tools such as nets. Alongside this, conservation practitioners need to better engage with rural communities to address unsustainable hunting practices, especially outside of protected areas.

24. <scp>ABO</scp> blood groups and expression of blood group antigens of epithelial ovarian cancer in Chinese women

Abstract

ABO blood groups has been associated with risk of several cancers; however, the results for an association with ovarian cancer are inconsistent and little is known about the expression of histo-blood group (ABH) antigens and ABO gene in ovarian tumor tissues.To assess the impact of genotype-derived ABO blood types on the risk of EOC, we conducted a case-control study in 1,870 EOC and 4,829 controls. Expression of A and B antigen in 70 pairs of ovarian tumor tissues and adjacent normal tissues were detected by immunohistochemistry. Gene expression and DNA methylation profiling was conducted in ovarian tumor tissues.We identified that blood group A was associated with increased risk for EOC compared to blood group O (OR = 1.18, 95% CI = 1.03-1.36, p = 0.019). Increased frequency of aberrant expression of histo-blood group antigens was observed in patients with blood group A (76.5%) compared to patients with blood group O (21.1%) and B (5.0%) by immunohistochemistry (p 0.001). ABO gene expression was down-regulated in ovarian tumor tissues compared with paired adjacent normal tissues (p = 0.027). In addition, ABO gene expression was positively correlated with NFYB (r = 0.38, p 0.001) and inversely correlated with DNA methylation level of four CpG sites on ABO gene (cg11879188, r = - 0.3, p = 0.002; cg22535403, r = - 0.30, p = 0.002; cg13506600, r = - 0.22, p = 0.025; cg07241568, r = - 0.21, p = 0.049) in ovarian tumor tissues.We identified blood group A was associated with increased EOC risk in Chinese women and provided the clues of the possible molecular mechanisms of blood group A related to ovarian cancer risk.

Abstract

Dysregulation of G6PD involved in the pentose phosphate pathway (PPP) is known to promote tumorigenesis. The PPP plays a pivotal role in meeting the anabolic demands of cancer cells. However, the detailed underlying molecular mechanisms of targeting the G6PD-regulated PPP in breast cancer remain unclear. In this study, we aimed to elucidate the molecular pathways mediating the effects of G6PD on cancer progression. Clinical sample analysis found that the expression of G6PD in breast cancer patients was higher than that in normal controls, and patients with higher G6PD expression had poor survival. Gene knockdown or inhibition of G6PD by 6-AN in MCF-7 and MDA-MB-231 cells significantly decreased cell viability, migration, and colony formation ability. G6PD enzyme activity was inhibited by 6-AN treatment, which caused a transient upregulation of ROS. The elevated ROS was independent of cell apoptosis and thus associated with abnormal activated autophagy. Accumulated ROS levels induced autophagic cell death in breast cancer. Inhibition of G6PD suppresses tumour growth in preclinical models of breast cancer. Our results indicate that targeting the G6PD-regulated PPP could restrain tumours in vitro and in vivo, inhibiting G6PD caused cell death by over-activating autophagy, therefore leading to inhibited proliferation and tumour formation.

44. Multi‐Sequence and Multi‐Regional <scp>MRI</scp> ‐Based Radiomics Nomogram for the Preoperative Assessment of Muscle Invasion in Bladder Cancer

Whether bladder cancer (BCa) invades muscle is a determinant of management. However, the accuracy of preoperative diagnosis of muscle invasion is not satisfactory.To investigate the value of multi-sequence and multi-regional magnetic resonance imaging (MRI)-based radiomics nomogram for assessing muscle invasion of BCa.Retrospective.342 BCa patients, divided into a training set (239 cases), a validation set (68 cases), and a test set (35 cases).3.0 T/TPatients were divided into muscle-invasive (79 cases) and non-muscle-invasive (263 cases). Two radiologists delineated the whole tumor, tumor body, and muscle layer of BCa, respectively, and extracted radiomic features.Recursive feature elimination, Pearson correlation coefficient, logistic regression, least absolute shrinkage and selection operator (Lasso) regression analysis, and 5-fold cross-validation were used to screen features and build a radiomics model. The clinical data were collected to construct a clinical model and a radiomics-clinical nomogram.23,688 features were extracted. After screening, the radiomics scoring model was constructed using nine radiomics features with area under curve (AUC) values of 0.933, 0.913, and 0.931 in the training, validation, and test sets, respectively. The clinical model was constructed using five clinical independent risk factors; the AUC values in the training, validation, and test set were 0.876, 0.859, and 0.824, respectively. After logistic regression analysis, the AUC values of the radiomics-clinical nomogram were made up of four clinical independent risk factors and radiomics scores were 0.955, 0.922, and 0.935 for the training, validation, and test sets, respectively. The DeLong test between clinical model and radiomics-clinical nomogram shows P 0.001.Multi-sequence and multi-regional MRI-based radiomics models could effectively assess the state of BCa muscular invasion. The radiomics-clinical nomogram is superior to clinical model for assessing BCa muscular invasion.4 TECHNICAL EFFICACY: Stage 2.
