Targeted drug delivery has been extensively studied in the last decade, whereas both passive and active targeting strategies still face many challenges, such as off-target drug release. Light-responsive drug delivery systems have been developed with high controllability and spatio-temporal resolution to improve drug efficacy and reduce off-target drug release. Photoremovable protecting groups are light-responsive moieties that undergo irreversible photocleavage reactions upon light irradiation. They can be covalently linked to the molecule of interest to control its structure and function with light. In this review, we will summarize recent applications of photocleavage technologies in nanoparticle-based drug delivery for precise targeting and controlled drug release, with a highlight of strategies to achieve long-wavelength light excitation. A greater understanding of these mechanisms and emerging studies will help design more efficient photocleavage-based nanosystems to advance photoresponsive drug delivery.

Globalized drug development studies, such as multiregional clinical trials (MRCTs), have attracted much attention due to their ability to expedite drug development and shorten the time lag of drug release. While observing the overall effect of a new drug, the region-specific effects to support drug registration in constituent regions can also be evaluated. Several challenges arise in conducting MRCTs, such as the heterogeneity in the variability of the primary endpoint across regions. However, most of the existing statistical methods assume a common variability, which may not be valid in practice due to differences across regions (eg, diversities in ethnicity or disparities in medical culture/practice). We present a statistical method for the design and evaluation of MRCTs to consider the heterogeneous variability across regions. We assessed the overall sample size requirement and addressed the region-specific sample size determination to establish the consistency of treatment effects between the specific region and the entire group. We demonstrate the proposed approach with numerical examples.

Because some of the potentially most useful boronic acids are inherently unstable in blood plasma and exhibit poor selective retention in tumours, 2-heterocyclic N-methyliminodiacetic acid (MIDA) boronates provide a stable, spacious and highly effective harbor for prodrug conjugates. Herein we report MIDA boronates in conjunction with naphthalene-based fluorophores as suitable compounds for tumour diagnosis by virtue of their remarkable specificity and uniform benchtop stability. The shielding group was found to be effective at imparting stability under physiological conditions (pH 7.4), with rapid release of the drug upon exposure to the acidic microenvironment of the tumor. This approach significantly enhanced the efficiency of drug release and was found to exhibit fewer side effects, thus indicating its great potential for precision therapeutics.

Supramolecular coordination cages show a wide range of useful properties including, but not limited to, complex molecular machine‐like operations, confined space catalysis, and rich host–guest chemistries. Here we report the uptake and release of non‐covalently encapsulated, pharmaceutically‐active cargo from an octahedral Pd cage bearing polymer chains on each vertex. Six poly(ethylene glycol)‐decorated bipyridine ligands are used to assemble an octahedral PdII 6(TPT)4 cage. The supramolecular container encapsulates progesterone and ibuprofen within its hydrophobic nanocavity and is activated by shear force produced by ultrasonication in aqueous solution entailing complete cargo release upon rupture, as shown by NMR and GPC analyses., An octahedral PdII 6(TPT)4 cage with polymer chains on each vertex is activated by shear force from ultrasonication in aqueous solution. The metal–organic cage encapsulates progesterone or ibuprofen within its hydrophobic nanocavity and shows complete cargo release upon rupture.

Molecularly imprinted polymers (MIPs) are chemically synthesized affinity materials with tailor‐made binding cavities complementary to the template molecules in shape, size, and functionality. Recently, engineering MIP‐based nanomedicines to improve cancer therapy has become a rapidly growing field and future research direction. Because of the unique properties and functions of MIPs, MIP‐based nanoparticles (nanoMIPs) are not only alternatives to current nanomaterials for cancer therapy, but also hold the potential to fill gaps associated with biological ligand‐based nanomedicines, such as immunogenicity, stability, applicability, and economic viability. Here, we survey recent advances in the design and fabrication of nanoMIPs for cancer therapy and highlight their distinct features. In addition, how to use these features to achieve desired performance, including extended circulation, active targeting, controlled drug release and anti‐tumor efficacy, is discussed and summarized. We expect that this minireview will inspire more advanced studies in MIP‐based nanomedicines for cancer therapy., This minireview surveys recent advances in research on molecularly imprinted polymer nanoparticles (nanoMIPs) for cancer therapy and highlights the distinct features of nanoMIPs for the rational design and construction of cancer nanomedicines with desired performances, including extended circulation, active targeting, controlled drug release, and antitumor efficacy.

Nanocarrier-mediated drug delivery is a promising strategy to maximize the power of chemotherapy and minimize side effects. However, current approaches show insufficient drug-loading capacity and inefficient drug release, and require complex modification processes. Attempts to enhance one of these features often compromise other merits. We describe here a block copolymer assembly system that combines desirable characteristics. The design of self-immolative and crosslinkable hydrophobic moieties offer stable and high encapsulation. Redox-triggerable polymer self-immolation promotes drug release by switching the hydrophobic core into completely hydrophilic chains. The reactive amine handles, presented on their surface, allow "plug to direct" modification with targeting ligands. Functionalized nanoassemblies have been programmed to target specific subcellular compartments. The simplicity, versatility, and efficacy of the system open up possibilities for an all-in-one delivery system.

This Review presents polysaccharides, oligosaccharides, nucleic acids, peptides, and proteins as functional stimuli-responsive polymer scaffolds that yield hydrogels with controlled stiffness. Different physical or chemical triggers can be used to structurally reconfigure the crosslinking units and control the stiffness of the hydrogels. The integration of stimuli-responsive supramolecular complexes and stimuli-responsive biomolecular units as crosslinkers leads to hybrid hydrogels undergoing reversible triggered transitions across different stiffness states. Different applications of stimuli-responsive biomolecule-based hydrogels are discussed. The assembly of stimuli-responsive biomolecule-based hydrogel films on surfaces and their applications are discussed. The coating of drug-loaded nanoparticles with stimuli-responsive hydrogels for controlled drug release is also presented.

Limited tumor permeability of therapeutic agents is a great challenge faced by current cancer therapy methods. Herein, a kind of near infrared light (NIR)-driven nanomotor with autonomous movement, targeted ability, hierarchical porous structure, multi-drugs for cancer chemo/photothermal therapy is designed, prepared and characterized. Further, we establish a method to study the interaction between nanomotors and cells, along with their tumor permeability mechanism, including 2D cellular models, 3D multicellular tumor spheroids and in vivo models. In vivo tumor elimination results verify that the movement behaviour of the nanomotors can greatly facilitate them to eliminate tumor through multiple therapeutic methods. This work tries to establish systematic research and evaluation models, providing strategies to understand the relationship between motion behaviour and tumor permeation efficiency of nanomotors in depth.

PURPOSE High-intensity focused ultrasound (HIFU)-mediated drug release becomes a promising therapeutic technique for treatment of cancer, which has merits of deep penetration, noninvasive approach and nonionizing radiation. However, conventional thermocouple-based approach for treatment monitoring would encounter big challenges such as the viscous heating artifact and difficulty in monitoring in the deep region. In this study, we develop an effective method based on thermal strain imaging (TSI) for the evaluation of HIFU-mediated drug release. METHODS Both phantom experiments and preliminary animal experiments were performed to investigate the feasibility of the proposed approach. Doxorubicin (DOX)-loaded cerasomes (HIFU and temperature-sensitive cerasomes, HTSCs) were prepared. In the phantom experiments, the HTSC solution is contained inside a cylindrical chamber within a tissue-mimicking phantom. In the animal experiments, the HTSCs are intravenously injected into tumor-bearing mice. An HIFU transducer is used to trigger DOX release from the HTSCs within the phantom or mice, and TSI is performed during HIFU heating. In the phantom experiments, the accuracy of temperature estimation using TSI is validated by measuring with a thermocouple. In animal experiments, the spatial consistency between the distribution of DOX released within the tumor and the location of the heating region estimated by TSI is validated using a spectrofluorophotometer. RESULTS In the phantom experiments, the HTSCs show a burst release of DOX when the temperature of the HTSC solution estimated by TSI reaches about 42°C, which is in agreement with the condition for drug release from the HTSCs. The temperature estimation using TSI has high accuracy with error below 2.5%. In animal experiments, fluorescence imaging of the tumor validates that the heating region of HIFU could be localized by the low-strain region of TSI. CONCLUSION The present framework demonstrates a reliable and effective solution to the evaluation of HIFU-mediated local drug delivery.

Films that can form bioadhesive hydrogels on wet biotissues absorbing blood or body fluids are useful for medical devices such as hemostats, adhesion barriers, wound dressings, and drug release devices. We focused on a hydrogen-bonding polymer complex consisting of poly(acrylic acid) (PAA) and poly(vinylpyrrolidone) (PVP). PAA is known as a tissue-adhesive polymer. However, simple mixing of aqueous PAA and PVP solutions resulted in the formation of an insoluble nonadhesive precipitate. We developed a novel solid/solution interface complexation method to afford a PAA/PVP complex that forms a strongly bioadhesive hydrogel with low cytotoxicity. The complex hydrogel can be slowly dissociated and dissolved in the body. The formation of the complexes as well as their swelling and degradation behavior depended strongly on the molecular weights and cross-linking densities of the component polymers. When the complex film was applied to a clipped incised jugular vein of a rat, it immediately formed a hydrogel and closed the incision. After removal of the clip, blood flowed through the vessel without any leakage. Application of the complex film to the surface of an incised mouse liver resulted in firm adhesion and the hemorrhage was effectively stopped. Such bioadhesive and biodissolvable materials consisting of low-toxicity synthetic polymers have high potential for implantable medical devices.

Current rational design of advanced coating materials has paved the way for controlled release technologies in biomedical applications. In this context, there is an increasing clinical need for designing and developing biomaterials and medicated dressings, to enhance the regeneration of injured or diseased tissues and organs, and to prevent microbial contamination. The development of novel biomaterialbased coatings with drug release capability is highly desirable. A variety of materials such as synthetic and natural polymers, ceramics, hybrid and composite materials have been engineered for the loading and releasing of drugs in a controlable and/or sustainable manner, thus improving active efficiency. In addition, nanoscale materials offer remarkable advantages in drug delivery applications owing to their high surface area and unique physicochemical properties, which depend on their size and shape. However, the physical and chemical properties of drugs, release routes and application are the driving forces of choosing the type of coating material and of controlled release system. The compatibility with active ingredients also plays a key role in the selection of materials. This chapter examines advanced coating nanomaterials describing the nuances of drug reléase systems and their proposed mechanisms. The advantages of different material combinations are described with special emphasis in their application to meet the current challenges for tissue regeneration, antibacterial activities and prevention of infections.
