Four novel cell lines from tissues of eye, gill, kidney and brain of Etroplus suratensis were developed and characterized. The cell lines of eye, gill, kidney and brain were sub-cultured for 245, 185, 170 and 90 passages, respectively, since 2008. These cell lines showed predominantly epithelial-like cells. Effects of temperature and foetal bovine serum concentration on the growth of these cell lines were examined and optimum growth was found at the temperature of 28° C with 20% foetal bovine serum. All the four cell lines were successfully cryopreserved and revived at different passage levels. Cell-cycle analysis of these cell lines was carried out by fluorescence-activated cell sorting. Polymerase chain reaction (PCR) products obtained from the cells and tissues of E. suratensis with primers specific to the conserved region of 16S ribosomal RNA and cytochrome oxidase I genes of E. suratensis revealed the origin of cell lines from E. suratensis. Antibodies raised against the tissues and cells of eye, kidney and gill were highly cross reacted to their specific tissue and cells of E. suratensis. Chromosomal analysis revealed that E. suratensis cells have a normal diploid karyotype with 2n = 48. The cells of these cell lines were successfully transfected with pEGFP vector DNA. The eye (IEE), gill (IEG) and kidney (IEK) cell lines were found to be susceptible to nodavirus but resistant to infectious pancreatic necrosis virus (IPNV). The cells of gill, kidney and eye were applied to test the cytotoxicity of tannery effluents.

A new myxosporean, Myxobolus etropli sp. nov., was found to infect the bulbus arteriosus of Etroplus suratensis (Bloch) from brackishwater lagoons of Muttukkadu, Chennai coast, India. A survey from May 1993 to October 1994 revealed a prevalence rate of 33.7% of this parasite. Macroscopic discoloured foci/cysts were seen in the bulbus arteriosus of the fish. The parasite showed strict host and site specificity. Histopathology showed that the infection was restricted to the bulbus. This is the first report of a myxosporean from E. suratensis.

The morphometry, feeding habits and reproductive biology of Etroplus suratensis, an indigenous cichlid food fish, were studied for a period of one year on specimens from three different habitats in Sri Lanka. The morphometric studies implied that the length weight relationships are curvilinear. Macrophytic fragments formed the most important component of food, although detritus, diatoms, and animal matter are also ingested. First sexual maturity is attained in the length ranges 8.0-9.0 cm in females from brackish water habitats. Fecundity ranged from 700-3900 eggs in mature specimens. The wide range of the gonadosomatic index suggests that spawning occurs several times during the year.

The digestibility of the aquatic macrophyte Hydrilla verticellata by the Asian cichlid, Etroplus suratensis was estimated using three indigenous markers viz. hydrolysis resistant organic matter (HROM), crude fibre (CF) and hydrolysis resistant ash (HRA). In the fodder HROM showed the least variation and HRA the highest. The recovery of HRA in the faeces was always higher than 100% whereas HROM was the nearest to 100%. The total digestibility varied between 34·6% and 51·9%, and the digestibility estimates were higher when HRA was considered as the marker. The overall protein and lipid digestibility varied between 59·2%–70·9% and 65·0%–70·1 % respectively. It is suggested that HROM is a better reference marker in comparison CF and HRA. Daily variation in protein digestibility ranged from 51·2% to 81·8%.
