Background: Strongylid and ascarid parasites are omnipresent in equine stud farms, and ever-increasing levels of anthelmintic resistance are challenging the industry with finding more sustainable and yet effective parasite control programs., Objectives: To evaluate egg count levels, bodyweight and equine health under defined parasite control protocols in foals and mares at two Standardbred and two Thoroughbred stud farms., Study Design: Longitudinal randomised field trial., Methods: A total of 93 foals were enrolled and split into two treatment groups, and 99 mares were enrolled and assigned to three treatment groups. All horses underwent a health examination, and episodes of colic or diarrhoea were recorded at each faecal collection date. Bodyweights were assessed using a weight tape, and mares were body condition scored. Group A foals (FA) were dewormed at 2 and 5 months of age with a fenbendazole/ivermectin/praziquantel product, while group B foals (FB) were dewormed on a monthly basis, alternating between the above-mentioned product and an oxfendazole/pyrantel embonate product. Group A mares (MA) were dewormed twice with fenbendazole/ivermectin/praziquantel, group B mares (MB) were dewormed with the same product, when egg counts exceeded 300 strongylid eggs per gram, and group C mares (MC) were dewormed every 2 months, alternating between the two products. Health data were collected monthly for 6 months (foals) and bimonthly for 13 months (mares). Data were analysed with mixed linear models and interpreted at the α = 0.05 significance level., Results: There were no significant bodyweight differences between foal groups, but MA mares were significantly lighter than the other two groups. Very few health incidents were recorded. Foals in group FA had significantly higher ascarid and strongylid egg counts, whereas no significant differences were observed between mare groups., Main Limitations: Study duration limited to one season., Conclusions: Anthelmintic treatment intensity was lowered from the traditional intensive regimes without measurable negative health consequences for mares and foals., (© 2020 EVJ Ltd.)

Aims: The role of the immune response to cyathostomin infections in horses remains unknown. Intestinal goblet cell hyperplasia has previously been noted as a component in cyathostomin infection; however, the function is unclear. The goal of this study was to evaluate the local and systemic gene expression to cyathostomin infections following larvicidal treatment and explore their relation to goblet cells., Methods and Results: Thirty-six ponies with naturally acquired cyathostomin infections were randomly allocated into three groups: fenbendazole-treated (10 mg/kg PO 5 days), moxidectin-treated (0.4 mg/kg PO once) and untreated control. Whole blood from all horses was collected weekly, and tissue samples from the large intestine collected during necropsy at 2 and 5 weeks post-treatment (WPT). Gene expression of interleukin (IL)-4, IL-5, IL-6, IL-10, IL-13, IL-17A, IL-22, IFN-γ, resistin-like molecule beta (RELM-β), Mucin 2 (MUC2) and tumour necrosis factor (TNF)-α was measured using qRT-PCR. There were statistically significant linear correlations between luminal worm burdens and MUC2 (r = -.2358) and RELM-β (r = -.2261)., Conclusion: This suggests an active role of immune system post-treatment in parasite expulsion, specifically in goblet cells, and that the organs respond differently to treatment and the larvae themselves. Introduction: This study aimed to assess Trichuris species infection and evaluate the anthelmintic efficacy of fenbendazole and ivermectin against natural trichurid infections in non-human primates (NHPs), kept at Mahendra Chaudhury (MC) Zoological Park, Chhatbir, India., Materials and Methods: Molecular confirmation of Trichuris infection was carried out using polymerase chain reaction targeting internal transcribed spacer sequences, and anthelmintic efficacy was assessed by fecal egg count reduction test, respectively., Results: A 710 base pair product confirmed Trichuris species infection in NHPs. Fenbendazole, 10 mg/kg body weight orally for 5 consecutive days and ivermectin, 100 μg/kg body weight orally for 3 alternate days proved effective and showed a maximum fecal egg reduction of 99.20% and 100% (P < .05) at day 7 post-treatment., Conclusions: This study highlighted the molecular confirmation of Trichuris species in non-human primates and its management using fenbendazole and ivermectin., (© 2018 John Wiley & Sons A/S. Mouse models have provided a significant insight into the role of T-helper (Th) 2 cytokines such as IL-5 and IL-13 in regulating eosinophilia and other key features of asthma. However, the validity of these models can be compromised by inadvertent infection of experimental mouse colonies with pathogens such as oxyurid parasites (pinworms). While the benzimidazole derivative, fenbendazole (FBZ), is commonly used to treat such outbreaks, the effects of FBZ on mouse models of Th2 disease are largely unknown. In this investigation, we show that mice fed FBZ-supplemented food during the in utero and post-weaning period developed attenuated lung eosinophilia, antigen-specific IgG1 and Th2 cytokine responses in a model of asthma. Treatment of the mediastinal lymph node cells from allergic mice with FBZ in vitro attenuated cell proliferation, IL-5 and IL-13 production and expression of the early lymphocyte activation marker, CD69 on CD4(+) T cells and CD19(+) B cells. In addition, eosinophilia and Th2 responses remained attenuated after a 4-week withholding period in allergic mice treated preweaning with FBZ. Thus, FBZ modulates the amplitude of Th2 responses both in vivo and in vitro.

A previously healthy Scottish terrier developed verminous pneumonia due to Filaroides hirthi. The disease was diagnosed on the basis of cytology of a transtracheal wash, which contained 17 per cent eosinophils and a large number of parasite larvae. Treatment with 50 mg/kg bodyweight fenbendazole daily for three weeks eliminated the parasites and improved the clinical condition of the dog.

The efficacy of in-feed fenbendazole at a dose rate of 7.5 mg/kg bodyweight for three consecutive days was assessed in five Arabian oryx (Oryx leucoryx), six scimitar-horned oryx (Oryx dammah), 14 slender-horned gazelles (Gazella leptoceros), eight Soay sheep (Ovis aries aries soay), 13 alpine ibex (Capra ibex ibex), six red deer (Cervus elaphus hippelaphus) and 11 Nelson's elk (Cervus elaphus nelsoni) kept in five herds in a zoo. The efficacy was assessed by means of repeated faecal egg count reduction (fecr) tests and in vitro egg hatch assays. Fenbendazole was highly effective against nematodes in five of the seven species, consistently reducing egg shedding by more than 90 per cent. In the egg hatch assays of the five herds, 50 per cent inhibition of hatching (ld50) was observed at a concentration of thiabendazole below 0.1 microg/ml. In the Arabian oryx and alpine ibex the efficacy of fenbendazole was less than 90 per cent, and the ld50 in the egg hatch assays was between 0.1 and 0.2 microg/ml thiabendazole.

Reasons for Performing Study: Possible anthelmintic resistance on a breeding farm where a rapid rotation anthelmintic programme had been implemented for 9 years was investigated. Cyathostomins resistant to fenbendazole and pyrantel were documented by faecal worm egg count reduction test (FWECRT)., Objectives: To 1) manage small strongyle transmission in a herd of horses in which resistance to both pyrantel pamoate and fenbendazole was identified and thereby reduce the risk of clinical disease in the individual animal, 2) monitor the change in resistance patterns over time and 3) monitor the efficacy of ivermectin over the study period., Methods: Targeted ivermectin treatment of horses on the farm was instituted for mature horses with faecal worm egg counts (FWEC) > 200 eggs/g (epg) and for horses < age 2 years with FWEC > 100 epg., Results: Over a 30 month period, targeted ivermectin treatment achieved acceptable control in mares, as judged by FWEC, and improved control of patent cyathostome infection in consecutive foal crops. Egg reappearance time (ERT) after treatment with ivermectin was < 8 weeks in mares and foals more frequently in the second year of the study than in the first year. Numbers of anthelmintic treatments were reduced by 77.6 and 533% in the mare and foal group, respectively., Conclusions: Targeted ivermectin treatment may be an economically viable method of managing multiple drug resistant cyathostominosis., Potential Relevance: Use of ivermectin should be monitored closely for development of resistance.

The results of a serological test for Encephalitozoon cuniculi in 125 pet rabbits are reviewed, together with follow-up studies of clinical cases. Blood samples were taken from 38 asymptomatic rabbits and 87 rabbits showing neurological, renal or ocular signs suggestive of encephalitozoonosis. In the asymptomatic group, six of 26 (23 per cent) apparently healthy rabbits, sampled as part of a health screen, were seropositive; of the remaining 12 asymptomatic rabbits, sampled because they lived with seropositive companions, eight (66 per cent) were seropositive. Fifty-eight of the rabbits with clinical disease showed neurological signs, including head tilt, seizures, ataxia and swaying; three of them also showed renal signs and two showed ocular signs, and these five rabbits were all seropositive. Head tilt was the most common neurological sign in 21 of 23 (91 per cent) of the seropositive cases. All nine rabbits with ocular lesions were seropositive. In follow-up studies of clinical cases, the rabbits showed variable responses to treatment with albendazole, fenbendazole, antibiotics or corticosteroids, and some cases recovered without treatment.

The efficacy of fenbendazole for preventing an experimental infection of Encephalitozoon cuniculi and for eliminating the spores from the central nervous system of naturally infected rabbits was investigated. Fenbendazole (20 mg/kg bodyweight daily) was administered from seven days before until two or 21 days after rabbits had been infected orally with 10(6) spores of E. cuniculi. Both regimens were effective in preventing the establishment of the parasites, as demonstrated by negative parasitic-specific serology and by the failure to isolate the parasite from brain tissue. In naturally infected, seropositive rabbits, parasites were successfully isolated from seven of nine untreated animals, but not from the brain tissue of eight animals treated with fenbendazole-medicated pellets for four weeks.

The protective immune response to larval migration in pigs, with or without adult intestinal worm populations, 10 weeks after 3 weekly Ascaris suum inoculations, was studied in 45 pigs. Controlled adult worm populations were achieved by oral transfer of 10 adult worms to previously immunized pigs after anthelmintic drenching. A significant reduction in larval recovery from lungs on day 7, and small intestine on day 14, was observed in immunized pigs compared with previously uninfected control pigs after challenge inoculation. The strong anamnestic response to larval migration was characterized by blood eosinophilia and specific immune responses measured by peripheral blood enzyme-linked immunospot and immunosorbent assays using larval excretory-secretory products and adult body fluid as well as Western blotting with a panel of stage-specific A. suum antigens. Immune detection of a previously unreported 10 kDa band, specific to the L2 larval stage and egg hatch fluid, emerged in all pigs after challenge, while the major adult body fluid constituent, ABA-1, remained unrecognized. No significant effect of an intestinal adult worm burden on the larval recovery after a challenge inoculation or on the immune response before or after challenge inoculation could be detected. These results indicate that a significant protective memory immune response to A. suum challenge inoculation can be induced in pigs, and that this protective immunity is not significantly modulated by the presence of adult parasites in the gut.

This study reports on the prevalence of anthelmintic resistance in strongyles of horses in Denmark. Of 5 methods used for the calculation of faecal egg count reduction (FECR) the method recommended by the World Association for the Advancement of Veterinary Parasitology, for the detection of resistance in sheep was the most sensitive procedure for detecting resistance. Using this method benzimidazole resistance was detected on 33 of 42 farms (79%) examined. Pyrantel was tested on 15 farms and FECR tests indicate resistance on 3 (30%) farms. On 2 farms on which resistance to pyrantel was detected resistance to benzimidazoles was also detected. On one of 16 farms examined ivermectin resistance was indicated at Day 14 but not at Day 19. On the 15 remaining farms ivermectin was effective. Due to the high prevalence of anthelmintic resistance in Danish horse herds it is recommended that tests of anthelmintic efficacy be conducted routinely to monitor the effectiveness of the strongyle control programmes.

The effect of three intraruminal sustained-release devices (SRD) against Dictyocaulus viviparus infection was tested in five groups of six calves. Group 1 served as untreated controls, and groups 2, 3 and 4 were dosed with a levamisole SRD, a fenbendazole SRD, and an ivermectin SRD, respectively. Group 5 was vaccinated against lungworm and received a levamisole SRD. The calves were turned out on May 28 and the devices given seven days later. All the calves received trickle infections with a total of 200 lungworm larvae between 9 and 34 days after turnout. They were housed on October 28, challenged with 5000 lungworm larvae and slaughtered three weeks later. No clinical signs of parasitic bronchitis were observed during the study. The treated groups gained significantly more weight (P < 0.05) than the controls, but did not differ among themselves. Larvae were first detected in the faeces of the control group between 25 and 32 days after the first infection, and had a group mean of 21 larvae per gram (lpg) after 60 to 80 days, after which the lpg gradually decreased. In group 2, larvae were detected near the end of the grazing season and never exceeded a group mean of 1.5 lpg. In group 3, a very low larval output was observed after housing (group mean 0.1 lpg). Groups 4 and 5 never became patent. The results of an ELISA followed the pattern of larval output; optical densities above the cut-off value were recorded in groups 1, 2 and 3. On the basis of worm recoveries after challenge, group 1 was immune. Group 4 had significantly more lungworms than group 2. There were no significant differences in worm numbers between groups 2, 3 and 5, but the worms in group 5 were retarded in growth (P < 0.05).

The efficacy of five daily treatments with 7.5 mg fenbendazole/kg bodyweight against mucosal cyathostome larvae was evaluated in 20 12- and 24-month-old ponies with naturally acquired cyathostome infections. After three weeks communal grazing on infected pasture and six weeks indoors, one group of 10 ponies were treated. Six weeks later, both groups of ponies were humanely destroyed and their burdens of large intestinal cyathostome worms, including luminal parasites and mucosal larvae, were assessed. In the control animals approximately 7 per cent of the total worm burden was present in the gut lumen and 93 per cent was present as larvae in the large intestinal mucosa. The efficacy of fenbendazole against the luminal cyathostomes was 90.7 per cent (P < 0.01). The total numbers of mucosal larvae, recovered after digestion, were reduced by 95.3 per cent (P < 0.0005); mucosal late third and fourth stage larvae were reduced by 99.4 per cent (P < 0.0001), and early inhibited third stage larvae by 91.5 per cent (P < 0.005).

The efficacy of a fenbendazole slow release bolus in controlling nematode infections of first-season cattle was evaluated in a field study in northern Germany. Two groups, each of 11 male calves, were set-stocked on separate pastures from May until October 1989 (157 days). The animals of one group were given the bolus at turnout and the animals of the control group were treated with fenbendazole (7.5 mg/kg bodyweight) eight weeks after turnout. Clinical inspections and measurements of faecal egg and larval counts, herbage trichostrongyle larval counts, plasma pepsinogen concentrations and bodyweight were made throughout the study. All the animals were slaughtered for worm counts and the evaluation of carcase quality two weeks after housing. The pasture grazed by the control group showed a marked increase in trichostrongyle larvae from late August onwards and, as a result, the control calves had increasing faecal egg counts and increased plasma pepsinogen concentrations in the latter part of the grazing season, although no clinical signs of parasitic gastroenteritis were apparent. The fenbendazole slow release bolus suppressed the trichostrongyle infections during the grazing season, and larval counts on the pasture grazed by the bolus-treated group remained low throughout the study. Postmortem examination showed that the bolus-treated calves harboured significantly (P < 0.01) fewer trichostrongyle worms, including inhibited stages, than the controls. Because of an inadequate lungworm challenge during the grazing season it was not possible to evaluate the efficacy of the fenbendazole slow release bolus in preventing parasitic bronchitis. At slaughter, the bolus-treated animals weighed more than the controls and tended to have a better carcase quality.

An abattoir survey was carried out during the 1991 to 1992 slaughter season to examine the structure of the ostertagid populations in red deer. Most of the animals were 15- to 22-month-old stags, but there were also hinds on two occasions and calves on one. Ostertagids of mixed species were present in all of the animals, and 43 of 120 (35.8 per cent) stags and 10 of 17 (58.8 per cent) hinds, but no calves, also carried small numbers of Trichostrongylus axei. Most animals carried adult and developing ostertagids and after September they all contained hypobiotic larvae (EL4). EL4 were present in three of nine stags in September and they increased in number through the autumn. In stags killed between November and February the group mean numbers of EL4 represented 62 to 94 per cent of the total burdens present, compared with 37 and 55 per cent on April 1 and 7, respectively. In the hinds, the number and the structure of the ostertagid populations was similar to that in the stags. Of 14 calves killed on October 9, only two contained a few EL4. Subcutaneous ivermectin at 400 micrograms/kg removed 100 per cent of the adult and developing worms, but although it also removed 95 per cent of EL4, some larvae remained in every treated animal. The efficacy of fenbendazole at 15 mg/kg given over five days was 75 per cent against EL4 and although the long interval after treatment obscured the result, it was probably much higher against later stages.

In a series of controlled trials involving 59 naturally infected greyhounds, fenbendazole at a dose rate of 50 mg/kg/day for three consecutive days reduced the overall numbers of third and fourth stage Toxocara canis by 94.0 per cent and third stage, fourth stage and immature adult stages of Toxascaris leonina by 92.4 per cent. In contrast, piperazine at 100 mg/kg had little or no useful effect against the larval stages of T canis and T leonina and variable efficacy against immature adult T leonina. Fenbendazole was also 100 per cent effective against immature Trichuris vulpis. In a separate controlled experiment, puppies in three litters exposed to reinfection with T canis were treated with fenbendazole at two weeks old and again only after their mean faecal egg counts exceeded 200 epg. Between one and three doses were required to suppress the output of eggs during the puppies' first 12 weeks of life.

Six cashmere goat herds in Scotland were surveyed to assess the prevalence of anthelmintic resistance and the species of nematode involved. Resistance was determined in vivo by a faecal egg count reduction test and in vitro using an egg hatch assay and specific faecal egg identification techniques. Benzimidazole resistance was present in five of the herds, insufficient eggs were obtained from the other herd to draw firm conclusions. Teladorsagia (Ostertagia) species predominated in four of the five herds in which resistance was detected and Haemonchus contorus was the dominant species in the other.

In order to study the prevalence of benzimidazole-resistance in equine cyathostomes, 30 stables in south east England were selected according to strict criteria but with minimum bias to provide three matched groups of 100 horses. One group was treated with fenbendazole, one with pyrantel embonate and the third was left untreated. The overall efficacies of fenbendazole and pyrantel embonate, as judged by the faecal egg-count reduction test, were 56.3 and 95.8 per cent, respectively. The numbers of horses at individual stables were too small to draw conclusions at each stable, but whereas efficacies greater than 85 per cent were recorded for pyrantel at 26 of 27 stables, the corresponding figure for fenbendazole was five of 27. Cyathostomes accounted for more than 90 per cent of the larvae cultured from faeces before and after treatment. Trichostrongylus axei was detected on 20 sites, but Strongylus species were present on only five. In the area of the survey, benzimidazole-resistance was very widespread in stables which used benzimidazole anthelmintics regularly and had more than nine horses per stable, but the benzimidazole-resistant cyathostome strains were adequately controlled with the non-benzimidazole anthelmintic pyrantel embonate.

Studies carried out on nine farms in north-west England indicated that fenbendazole treatment of adult winter calving dairy cows in the dry period caused an overall increase of 173 kg in their subsequent lactation yield. There was some evidence to suggest that a correlation existed between pasture Ostertagia ostertagi larval burdens and subsequent serum pepsinogen and milk yield responses post treatment. Marked interherd and regional differences existed in the milk yield response to anthelmintic treatment and possible reasons for such results are discussed.

Two further trials are reported on the effect of fenbendazole on inhibited Ostertagia ostertagi larvae. In the first trial fenbendazole successfully eliminated inhibited burdens of O ostertagi, while in the second trial the effect was variable. As a result of these trials and previous reports it is concluded that a single oral dose of fenbendazole cannot be relied upon to prevent winter (type 2) ostertagiasis.

In a study of the comparative speed of action of anthelmintics against Dictyocaulus viviparus infection in cattle levamisole reduced the lungworm burdens by 87 per cent within three hours of administration whereas fenbendazole and febantel required 36 hours to achieve 80 and 87 per cent reduction, respectively. The more rapid action of levamisole may be advantageous in the treatment of bovine parasitic bronchitis since it allows virtually immediate elimination of the majority of the lungworms and limits undesirable sequelae dependent on the continued presence of the parasite.

Based on faecal larval counts, respiratory rates and live-weight gains, the anthelmintics levamisole and fenbendazole were equally efficacious against primary infection with Dictyocaulus viviparus in calves. Calves that were treated with levamisole or fenbendazole resisted the live-weight depressing impact of reinfection. Treatment somewhat impaired resistance to establishment of the secondary challenge as determined at necropsy. However, compared with previously uninfected controls, the treated animals harboured significantly fewer D viviparus, showing that they still strongly resisted reinfecting worms becoming established. The animal responses measured did not demonstrate any difference between levamisole and fenbendazole in relation to efficacy or to effect on resistance.

Fenbendazole was used to treat nematode infestations (Ascaridia species and Capillaria species) in 230 birds of six orders and 38 different species. Using a single dose of 100 mg/kg bodyweight initial treatment eliminated parasitic nematodes from 221 birds. A further course of treatment at a dose rate of 30 mg/kg daily for seven days eliminated the infestation from the remaining nine birds.

The efficacy of fenbendazole against immature stages of Trichonema spp., Strongylus vulgaris and Strongylus edentatus was evaluated. Naturally infected 6 to 12 month old ponies were given single, oral doses of 0, 15, 30 and 60 mg/kg of body weight. A dose response relationship was noted between increasing dose levels and efficiency against larval trichonemes and migrating stages of S. vulgaris and S. edentatus. Dose levels of 30 mg/kg and higher removed 93 per cent of mucosal stages of Trichonema spp., while doses of 60 mg/kg removed 83 per cent and 89 per cent of the migrating larvae of S. vulgaris and S. edentatus respectively.
