Abstract

Aim: Transrectal ultrasound (TRUS) guided prostate biopsy is a typical early prostate examination. However, the ultrasound imaging suffers from blurred contour, intensity inhomogeneity and small surrounding soft tissue differentiation. To take advantage of clear magnetic resonance imaging (MRI) into robotic prostate biopsy navigation, the prostate regions in the MRI and TRUS images need to be segmented separately. This paper proposes an improved level set segmentation model based on prior shape, which aims to provide a better solution to the prostate segmentation problems in TRUS and MRI., Methods: In our segmentation model, the Gaussian probability model is used to establish the statistical learning of the prior shape, and the cosine function is used to represent the energy term fitting of the traditional prior shape and the local intensity information., Results: The experiment results show that the model can adapt to different forms of prostate in MRI and TRUS more accurately, and the prostate biopsy accuracy in our biopsy system can reach 2.24 ± 1.44 mm., Conclusion: This segmentation model has high accuracy, meets the clinical needs in robotic prostate biopsy navigation., (© 2020 John Wiley & Sons Ltd.)

Abstract

In Saccharomyces cerevisiae under conditions of nutrient stress, meiosis precedes the formation of spores. Although the molecular mechanisms that regulate meiosis, such as meiotic recombination and nuclear divisions, have been extensively studied, the metabolic factors that determine the efficiency of sporulation are less understood. Here, we have directly assessed the relationship between metabolic stores and sporulation in S. cerevisiae by genetically disrupting the synthetic pathways for the carbohydrate stores, glycogen (gsy1/2Δ cells), trehalose (tps1Δ cells), or both (gsy1/2Δ and tps1Δ cells). We show that storage carbohydrate-deficient strains are highly inefficient in sporulation. Although glycogen and trehalose stores can partially compensate for each other, they have differential effects on sporulation rate and spore number. Interestingly, deletion of the G 1 cyclin, CLN3, which resulted in an increase in cell size, mitochondria and lipid stores, partially rescued meiosis progression and spore ascus formation but not spore number in storage carbohydrate-deficient strains. Sporulation efficiency in the carbohydrate-deficient strain exhibited a greater dependency on mitochondrial activity and lipid stores than wild-type yeast. Abstract

Inflammation plays a role in scar formation; therefore, decreasing inflammation benefits scar removal. Jiashitang scar removal ointment (JST) is a commercially available traditional Chinese medicinal formulation. It is composed of extracts from Carthamus tinctorius L. (Car), Rheum officinale Baill. (Rhe), Salvia miltiorrhiza Beg. (Sal), and Panax notoginseng (Burk.) F. H. Chen (Pan), which are all herbs with potent antiinflammatory activities. Our aims are to optimize the formula of JST and to elucidate its antiinflammatory active components. Response surface methodology was applied to optimize proportions of the four herb extracts. The antiinflammatory effects were evaluated using in vitro and in vivo models. To screen for active components in this formula, a bioactivity-based ultra-performance liquid chromatography/quadrupole time-of-flight mass spectrometry analysis was performed. After optimization, the antiinflammatory effects of the new formula were significantly superior to the original one. Screening identified 13 active ingredients: a series of saffiomin, emodin, salvianolic acid, tanshinone, and triterpenoid saponin derivatives. These active ingredients were predicted to exert nuclear factor-κB inhibiting effects through MAPK, PI3K/AKT, and NIK-IKK pathways. In conclusion, the original formula was successfully optimized with more potent antiinflammatory activity.
