Abstract Studies in evolution, ecology and conservation are increasingly based on genetic and genomic data. With increased focus on molecular approaches, ethical concerns about destructive or more invasive techniques need to be considered, with a push for minimally invasive sampling to be optimised. Buccal swabs have been increasingly used to collect DNA in a number of taxa, including amphibians. However, DNA yield and purity from swabs are often low, limiting its use. In this study, we compare different types of swabs, preservation method and storage, and DNA extraction techniques in three case studies to assess the optimal approach for recovering DNA in anurans. Out of the five different types of swabs that we tested, Isohelix MS‐02 and Rapidry swabs generated higher DNA yields than other swabs. When comparing storage buffers, ethanol is a better preservative than a non‐alcoholic alternative. Dried samples resulted in similar or better final DNA yields compared to ethanol‐fixed samples if kept cool. DNA extraction via a Qiagen™ DNeasy Blood and Tissue Kit and McHale's salting‐out extraction method resulted in similar DNA yields but the Qiagen™ kit extracts contained less contamination. We also found that samples have better DNA recovery if they are frozen as soon as possible after collection. We provide recommendations for sample collection and extraction under different conditions, including budgetary considerations, size of individual animal sampled, access to cold storage facilities and DNA extraction methodology. Maximising efficacy of all of these factors for better DNA recovery will allow buccal swabs to be used for genetic and genomic studies in a range of vertebrates.

Abstract The Southern Ocean regulates atmospheric CO2 and Earth's climate as a critical region for air‐sea gas exchange, delicately poised between being a CO2 source and sink. Here, we estimate how long a water mass has remained isolated from the atmosphere and utilize 14C/12C ratios (Δ14C) to trace the pathway and escape route of carbon sequestered in the deep ocean through the mixed layer to the atmosphere. The position of our core at the northern margin of the Southern Indian Ocean, tracks latitudinal shifts of the Southern Ocean frontal zones across the deglaciation. Our results suggest an expanded glacial Antarctic region trapped CO2, whereas deglacial expansion of the subantarctic permitted ventilation of the trapped CO2, contributing to a rapid atmospheric CO2 rise. We identify frontal positions as a key factor balancing CO2 outgassing versus sequestration in a region currently responsible for nearly half of global ocean CO2 uptake.

The sustainable plant-derived biomass are now understood to be important sources for the prevention of corrosion in metallic alloys. This study examined the effect of a potential novel low-cost, low-toxicity inhibitor utilizing pumpkin peel extract (PPE) on aluminum corrosion hydrochloric acid 1M HCl medium. Several experimental methods were used to investigate the inhibitor effect of PPE which include, AFM-XPS surface analysis, electrochemical studies (electrochemical impedance spectroscopy-electrochemical frequency modulation-potentiodynamics), and weight loss procedures. According to the weight loss results, when PPE is introduced to a 1M HCl solution, the inhibitory performance rises up to 95.42% with an increase in PPE concentration in the range of 0–300 ppm at 25°C. Potentiodynamic results showed the investigated PPE inhibitor is a mixed-type inhibitor. Corrosion inhibition at different temperatures was also examined, and the thermodynamic activation parameters were identified. It was found that the adsorption of the inhibitor on the aluminum followed the Langmuir and kinetic isotherm models. Quantum chemical indices contributed to a greater comprehension of the inhibitory mechanism. To investigate the configurational adsorption performance of the investigated PPE on the aluminum surface, molecular dynamics (MD) simulations were carried out.

Abstract Pure ZnO particles, doped and co‐doped with Cu2+ and Mg2+ ions, were obtained by the simple sonochemical method. The wurtzite hexagonal structure was confirmed by X‐ray diffraction (XRD) patterns. The images obtained by field emission scanning electron microscopy (FE‐SEM) showed irregular and more agglomerated particles with the increase of Cu2+ and Mg2+ ions concentration in the ZnO lattice, with the average diameter of particles in the range of 57.23–170.77 nm. The photocatalytic activity was investigated by decolorizing the methylene blue (MB) dye under ultraviolet C (UVC) light irradiation which indicates that 1% Mg‐doped ZnO particles have better the photocatalytic activity than the other samples, and presented the highest kinetic constant value. The co‐doped samples showed a reduced global surface area which did not favor the good photocatalytic performance. The sacrificial agents showed that OH• radicals are the main species involved in the photocatalytic activity of this system and defects generated in the ZnO lattice promoted photoluminescent emission in the red and green regions.

Abstract Objective In the diagnosis of HTLV‐1‐associated myelopathy (HAM), while magnetic resonance imaging (MRI) is essential to exclude other diseases, its power is limited regarding HAM diagnosis, as only 30% of affected patients present with spinal cord atrophy. Diffusion tensor imaging (DTI) may enable the detection of damage in the white matter microstructure. Here, we quantitatively assess spinal cord damage using DTI and evaluate conventional MRI parameters of the spinal cord in HTLV‐1‐infected individuals. Methods This cross‐sectional study involved 33 HTLV‐1 carriers, 28 patients with definite‐HAM, and 11 seronegative healthy subjects (HS). Region‐of‐interest (ROI)‐based fractional anisotropy (FA) and mean diffusivity (MD) measurements were performed in the upper thoracic and lumbar regions of the spinal cord. Thoracic index was defined as 1/ (anteroposterior diameter × transverse diameter) measured at the fifth 5th vertebral level. Receiver operating characteristic (ROC) curve analysis was used to determine optimal cutoff FA, MD, and thoracic index values. Results Spinal cord atrophy was observed in 15 (53.6%) patients with definite‐HAM. The area under the ROC curve in the thoracic spinal cord was 0.824 (95% CI, 0.716–0.932), 0.839 (95% CI: 0.736–0.942), and 0.838 (95% CI: 0.728–0.949) for FA, MD, and the thoracic index, respectively. Abstract This paper proposes new analytical and optimal design procedures of the proportional‐resonant (PR) controller and its harmonic compensators (HCs) for three‐phase grid‐connected voltage source inverters (VSIs) powered by renewable energy resources. The modeling and analysis based on stationary reference frame are performed for VSIs collaborated with an L‐type filter. The theoretical verification and simulation validation of the proposed design guidelines are done to approve its effectiveness and robustness. Particle swarm, grey wolf and Harris hawks' optimization techniques are applied and compared for a proper selection of the parameters of the proposed PR controller and its HCs. To accomplish this study, multi‐objective error functions are employed and compared to minimize the total harmonic distortion of the grid output current. The proposed PR controller and its HCs are tested, using MATLAB/Simulink, along with the allowable changes of inverter output active and reactive powers, and also under the grid voltage distortion. Moreover, their performance is evaluated according to IEEE and IEC harmonics standards, and compared with the conventional PI controller based on reference frame. Furthermore, the experimental validation for the proposed controllers is done based on the hardware‐in‐the‐loop real‐time simulator using C2000TM‐microcontroller‐LaunchPadXL‐TMS320F28377S kit.

Abstract Nitrogen isotope analysis of chloropigments provides information on the sources of nitrogenous nutrients assimilated by phytoplankton. The abundant, ubiquitous chlorophyll a records nitrogen isotopic compositions (δ15N) of eukaryotic phytoplankton and cyanobacteria, whereas more source‐specific chloropigments, such as the divinylchlorophylls exclusively possessed by the marine picocyanobacterium Prochlorococcus, can potentially resolve isotopic variability within the phytoplankton community. In this study, we analyzed the δ15N of both chlorophyll a and divinylchlorophyll a isolated from suspended particulate material collected at the subsurface chlorophyll maximum (SCM) along a meridional transect at 88°E in the oligotrophic eastern Indian Ocean. The nitrogen isotopic compositions of Prochlorococcus (δ15NPRO) and the combined biomass of eukaryotic phytoplankton and Synechococcus (δ15NEU+SYN) estimated from the δ15N of divinylchlorophyll a and chlorophyll a, respectively, revealed systematic variations that were not apparent from bulk isotope measurements. Whereas consistently low δ15NPRO indicated reliance of Prochlorococcus on regenerated nitrogen throughout the transect, elevation in δ15NEU+SYN values at several stations was interpreted to reflect assimilation of subsurface NO3− by eukaryotic phytoplankton. The δ15N distributions revealed subtle differences in NO3− availability at the SCM along the transect, which were consistently explained by the occurrence of mesoscale eddies in the Bay of Bengal, deepening of the mixed layer induced by a seasonal Wyrtki Jet in the equatorial region, and substantial deepening of the nutricline in the South Indian Ocean gyre. Our results highlight the utility of compound‐specific isotopic measurements of multiple species of chlorophylls in obtaining essential non‐incubation‐based biogeochemical constraints on the primary production in the ocean.

Abstract Direct and semidirect estimations of leaf area (LA) and leaf area index (LAI) are scarce in dense tropical forests despite their importance in calibrating remote sensing products, forest dynamics, and biogeochemical models. We destructively sampled 61 trees belonging to 13 most abundant species in a semideciduous forest in southeastern Cameroon. For each tree, all leaves were weighed, and for a subsample of branches, leaves were counted and the LA measured. Allometric models were calibrated to allow semidirect estimation of LAI at tree and stand levels based on forest inventory data (R2 = 0.7, bias = 21.2%, error = 39.5%) and on predictors that could be extracted from very high resolution remote sensing data (R2 = 0.63, bias = 35.1%, error = 58.73). Using twenty‐one 1‐ha forest plots, stand level estimations of LAI ranged from 4.42–13.99. These values are higher than previous estimates generally obtained using indirect methods. These results may have important consequences on ecosystem exchanges and the role of tropical forest in global cycles.

Abstract Integration of inland waters into regional and global carbon (C) budgets requires a comprehensive understanding of factors regulating organic carbon (OC) delivery and in situ processing. This study reviews advances in optical, molecular, and isotopic approaches to resolve the sources, ages, and transformations of OC in aquatic systems. OC characterization using excitation emission matrix spectra, Fourier transform ion cyclotron mass spectrometry, and nuclear magnetic resonance provides detailed molecular level insight. Radiocarbon isotopic approaches and compound‐specific techniques resolve the input, metabolic fate, and turnover time of OC in ecosystems ranging in size from streams to the open ocean. Accumulating evidence suggests that aquatic OC is composed of diverse biogeochemical components. We conclude with enduring and emerging questions that underscore the role of inland systems in the global C cycle and propose unique combinations of approaches to better discern their role in the delivery and transformation of OC from soils to seas.

The anomalies of X chromosome are classified as numerical or structural. Concomitant structural anomalies in this chromosome that associate partial loss of its long arm with duplications in its short arm are uncommon. Only a few cases have been published and in most of them the reported patients present ovarian dysfunction, tall stature, and overdosage of the SHOX gene with locus Xp22.33. Considering these reports, we evaluated the case of a woman with a deletion in the long arm of the X chromosome, premature ovarian failure, tall stature, and multiple arterial vascular disease. With the aim to find a relationship between karyotype and phenotype, we explored associated anomalies in Xp and certified the overdosage of the SHOX gene in this case by MLPA. Also, taking into account the fact that the gene locus of the angiotensin-converting enzyme type 2 (ACE2) is located in Xp, our goal was to investigate the influence of this gene in the development of cardiovascular disease. The detection of the gene product of ACE2 by ELISA was undetectable. We have proposed that cytogenetic anomalies in X chromosome could contribute to decrease this protein synthesis in this gender.
