The aim of the present work was to simulate water treatment practices with hydrogen peroxide (HP) in recirculating aquaculture systems (RAS). Six identical 1,700-L pilot-scale RAS were divided into two experimental groups based on daily feed allocation and operated under constant conditions for a period of 3 months. The organic and nitrogenous loadings of the systems differed fourfold between the two groups and were achieved by predefined constant daily feed loads and constant additions of water. The fixed cumulative feed burden was 1.6 × 103 mg feed/L in the low-intensity RAS and 6.3 × 103 mg/L in the high-intensity RAS. The decay of HP in rearing tanks and disconnected biofilter units was investigated by means of HP spiking experiments. The decay in high-intensity RAS rearing units and biofilters was orders of magnitude faster than that in low-intensity units. Ultra-thin free-standing nanoporous gold leaf made by dealloying exhibits excellent electrocatalytic activities toward nitrite oxidation. The electrochemical responses of nitrite ions on this novel nano-electrode is found not dependent on pH over a wide range from 4.5 to 8.0, which is markedly different from that of gold oxidation, a process known to be highly pH-sensitive. Amperometric study shows a linear relationship for nitrite determination in a concentration range from 1 µM to 1 mM. This nanostructured gold electrode displays good stability, repeatability and selectivity which suggests its potential for the development of new electrochemical sensors.

Nitrospina bacteria are among the most important nitrite oxidizers in coastal and open‐ocean environments, but the relevance of the genus contrasts with the scarceness of information on their ecophysiology and habitat range. Thus far, Nitrospina bacteria have been the only nitrite oxidizers detected at high abundance in Chilean coastal waters. These levels are often higher than at other latitudes. In this study, the abundance of 16S‐rRNA gene transcripts of Nitrospina (hereafter just transcripts) was measured by reverse transcription quantitative PCR in a rocky intertidal gradient and compared with the nearshore counterpart off central Chile (~33°S). Rocky pond transcripts were also compared with the taxonomic composition of the macrobiota and bacterioplankton (by 16S‐rRNA gene‐based T‐RFLP) in the intertidal gradient. Transcripts increased from warmer, saltier, and low‐nitrite ponds in the upper intertidal zone (19.5 ± 1.6°C, 39.0 ± 1.0 psu, 0.98 ± 0.17 μmol/L) toward cooler, less salty, and high‐nitrite ponds (17.8 ± 2.6°C, 37.7 ± 0.82 psu, 1.23 ± 0.21 μmol/L) from middle and low zones. These varied from ~1,000 up to 62,800 transcripts. Nitrification was investigated in a model freshwater sediment by the combined use of microsensors and fluorescence in situ hybridization with rRNA-targeted oligonucleotide probes. In situ nitrification activity was restricted mainly to the upper 2 mm of the sediment and coincided with the maximum abundance of nitrifying bacteria, i.e. 1.5 x 107 cells cm-3 for ammonia-oxidizing Beta-proteobacteria (AOB) and 8.6 x 107 cells cm-3 for Nitrospira-like nitrite-oxidizing bacteria (NOB). Cell numbers of AOB decreased more rapidly with depth than numbers of NOB. For the first time, Nitrospira-like bacteria could be quantified and correlated with in situ nitrite oxidation rates in a sediment. Estimated cell-specific nitrite oxidation rates were 1.2-2.7 fmol NO 2 - cell-1 h-1.
