Objectives: Disruption of the oxidative stress defense system is involved in developing various diseases. Sulfur compounds such as glutathione (GSH) and cysteine (CysSH) are representative antioxidants in the body. Recently, supersulfides, including reactive persulfide and polysulfide species, have gained attention as potent antioxidants regulating oxidative stress and redox signaling. However, their involvement in the pathogenesis of chronic rhinosinusitis (CRS) remains unclear., Methods: To clarify the changes in sulfur compounds within the sinus mucosa of each CRS subtype, we measured sulfur compound levels in the sinus mucosa of control individuals ( n  = 9), patients with eosinophilic CRS (ECRS) ( n  = 13), and those with non-ECRS (nECRS) ( n  = 11) who underwent sinus surgery using mass spectrometry., Results: GSH and CysSH levels were significantly reduced, and the glutathione disulfide (GSSG)/GSH ratio, an oxidative stress indicator, was increased in patients with ECRS. Despite the absence of notable variations in supersulfides, patients with ECRS and nECRS exhibited a significant reduction in glutathione trisulfide (GSSSG), which serves as the precursor for supersulfides., Conclusions: This study is the first quantitative assessment of supersulfides in normal and inflamed sinus mucosa, suggesting that sulfur compounds contribute to the pathogenesis of CRS., Level of Evidence: N/A., Competing Interests: The authors declare no conflicts of interest., (© 2024 The Author(s). Objective/hypothesis: Middle ear cholesteatoma is characterized by abnormal growth of the keratinizing squamous epithelium of the temporal bone. d- β -aspartic acid is the major isomer of d-aspartic acid found in elderly tissue. We assessed the immunoreactivity to k- β -aspartic acid of congenital and acquired middle ear cholesteatomas., Study Design: Case-control studies., Material and Methods: Tissue samples were collected from 21 patients comprising 21 ears with congenital middle ear cholesteatoma and 26 patients comprising 29 ears with acquired type. Their clinical and histopathological features were investigated. We divided the middle ear cholesteatoma samples into three layers: the perimatrix, matrix, and cystic contents. The patterns of immunoreactivity to d- β -aspartic acid expression were then assessed immunohistochemically., Results: Two patterns of immunoreactivity to d- β -aspartic acid were detected in middle ear cholesteatoma: infiltrative and diffuse. In congenital middle ear cholesteatoma, d- β -aspartic acid expression was observed throughout all the layers (perimatrix, matrix, and cystic contents), and immunoreactivity to d- β -aspartic acid was dramatically strong in all layers. The expression levels of d- β -aspartic acid to the cystic content and perimatrix were significantly higher in congenital middle ear cholesteatoma than in the acquired type., Conclusions: This study showed the expression levels of d- β -aspartic acid in middle ear cholesteatoma to differ significantly between congenital and acquired middle ear cholesteatoma. Our results indicate that overexpression of d- β -aspartic acid is likely to be involved in the pathogenesis of cholesteatoma, and we speculate that d- β -aspartic acid could be a novel biomarker for, and a therapeutic target in, congenital and acquired middle ear cholesteatoma., Level of Evidence: 4., Competing Interests: The authors declare no conflict of interest., (© 2022 The Authors. Trypanosomiasis, leishmaniasis, and malaria are protozoan infections of public health importance with thousands of new cases recorded annually. Control of these infection(s) with existing chemotherapy is limited by drug toxicity, lengthy parenteral treatment, affordability, and/or the emergence of resistant strains. Medicinal plants on the other hand are used in the treatment of various infectious diseases although their chemical properties are not fully evaluated. In this study, we screened 112 crude extracts from 72 selected Ghanaian medicinal plants for anti-Trypanosoma, anti-Leishmania, and anti-Plasmodium activities in vitro and investigated their mechanisms of action. Twenty-three extracts from 20 plants showed significant antiprotozoan activity against at least 1 of 3 protozoan parasites screened with IC 50 values less than 20 μg/ml. Eleven extracts showed high anti-Trypanosoma activity with Bidens pilosa whole plant and Morinda lucida leaf extracts recording the highest activities. Their IC 50 (selectivity index [SI]) values were 5.51 μg/ml (35.00) and 5.96 μg/ml (13.09), respectively. Nine extracts had high anti-Leishmania activity with Annona senegalensis and Cassia alata leaf extracts as the most active. Their IC 50 (SI) values were 10.8 μg/ml (1.50) and 10.1 μg/ml (0.37), respectively. Six extracts had high anti-Plasmodium activity with the leaf and stem-bark extracts of Terminalia ivorensis recording the highest activity. Their IC 50 (SI) values were 7.26 μg/ml (129.36) and 17.45 μg/ml (17.17), respectively. Only M. lucida at 25 μg/ml induced significant apoptosis-like cell death in Trypanosoma parasites. Anti-Leishmania active extracts induced varying morphological changes in Leishmania parasites such as multiple nuclei and/or kinetoplast, incomplete flagella division, or nuclear fragmentation. Background: Gonadotropin-releasing hormones (GnRHs) are neuropeptides that play central roles in the reproduction of vertebrates. In the ascidian Ciona intestinalis, GnRHs and their receptors are expressed in the nervous systems at the larval stage, when animals are not yet capable of reproduction, suggesting that the hormones have non-reproductive roles., Results: We showed that GnRHs in Ciona are involved in the animal's metamorphosis by regulating tail absorption and adult organ growth. Absorption of the larval tail and growth of the adult organs are two major events in the metamorphosis of ascidians. When larvae were treated with GnRHs, they completed tail absorption more frequently than control larvae. cAMP was suggested to be a second messenger for the induction of tail absorption by GnRHs. tGnRH-3 and tGnRH-5 (the "t" indicates "tunicate") inhibited the growth of adult organs by arresting cell cycle progression in parallel with the promotion of tail absorption., Conclusions: This study provides new insights into the molecular mechanisms of ascidian metamorphosis conducted by non-reproductive GnRHs., (© 2014 Wiley Periodicals, Inc.)

The effect of D-mannitol as an indigestible sugar on apparent digestibility of nutrients, nitrogen (N) utilization and growth performance, have been investigated in growing rabbits fed a urea-containing diet. In experiment 1, 40-day-old male rabbits were fed a commercial diet with urea 10 g/kg with added glucose or D-mannitol 60 g/kg each. Thereafter, in experiment 2 the rabbits aged 55 days were fed a commercial diet with or without urea 10 g/kg. Feed restriction at 55 g/kg(0.75)/day was carried out in both experiments. Addition of D-mannitol to the urea-containing diet decreased urinary N excretion, which consequently increased N retention and ratios of retained N to consumed N or apparently absorbed N. Addition of urea alone to the diet increased daily N intake and urinary N excretion but did not affect N retention. Weight gain and feed efficiency were improved by D-mannitol feeding, whereas feeding with urea alone did not affect these parameters. Utilization of dietary urea was enhanced in growing rabbits fed the urea-containing diet with D-mannitol, resulting in improvement of N retention and growth performance., (© 2012 The Authors.
