Abstract In the era of data‐driven transportation development, traffic forecasting is crucial. Established studies either ignore the inherent spatial structure of the traffic network or ignore the global spatial correlation and may not capture the spatial relationships adequately. In this work, a Dynamic Spatial‐Temporal Network (DSTN) based on Joint Latent Space Representation (JLSR) is proposed for traffic forecasting. Specifically, in the spatial dimension, a JLSR network is developed by integrating graph convolution and spatial attention operations to model complex spatial dependencies. Since it can adaptively fuse the representation information of local topological space and global dynamic space, a more comprehensive spatial dependency can be captured. In the temporal dimension, a Stacked Bidirectional Unidirectional Gated Recurrent Unit (SBUGRU) network is developed, which captures long‐term temporal dependencies through both forward and backward computations and superimposed recurrent layers. On these bases, DSTN is developed in an encoder‐decoder framework and periodicity is flexibly modeled by embedding branches. The performance of DSTN is validated on two types of real‐world traffic flow datasets, and it improves over baselines.

Abstract Rapid and accurate diagnostic tests are fundamental for improving patient outcomes and combating infectious diseases. The Clustered Regularly Interspaced Short Palindromic Repeats (CRISPR) Cas12a‐based detection system has emerged as a promising solution for on‐site nucleic acid testing. Nonetheless, the effective design of CRISPR RNA (crRNA) for Cas12a‐based detection remains challenging and time‐consuming. In this study, we propose an enhanced crRNA design system with deep learning for Cas12a‐mediated diagnostics, referred to as EasyDesign. This system employs an optimized convolutional neural network (CNN) prediction model, trained on a comprehensive data set comprising 11,496 experimentally validated Cas12a‐based detection cases, encompassing a wide spectrum of prevalent pathogens, achieving Spearman's ρ = 0.812. We further assessed the model performance in crRNA design for four pathogens not included in the training data: Monkeypox Virus, Enterovirus 71, Coxsackievirus A16, and Listeria monocytogenes. The results demonstrated superior prediction performance compared to the traditional experiment screening. Furthermore, we have developed an interactive web server (https://crispr.zhejianglab.com/) that integrates EasyDesign with recombinase polymerase amplification (RPA) primer design, enhancing user accessibility. Through this web‐based platform, we successfully designed optimal Cas12a crRNAs for six human papillomavirus (HPV) subtypes. Remarkably, all the top five predicted crRNAs for each HPV subtype exhibited robust fluorescent signals in CRISPR assays, thereby suggesting that the platform could effectively facilitate clinical sample testing. In conclusion, EasyDesign offers a rapid and reliable solution for crRNA design in Cas12a‐based detection, which could serve as a valuable tool for clinical diagnostics and research applications.

Abstract Timely and precise short‐term passenger flow forecasting (STPFF) is a key to intelligent urban rail transit (URT) operation. The uncertainty of passenger travel behaviors leads to passenger flow data showing high nonlinearities and complex spatio‐temporal correlation. Therefore, the ability to capture such correlations determines the prediction performance of the model. However, current methods either consider only the physical topology of URT network or lack the ability to model global spatial dynamics, making it difficult to obtain satisfactory predictions. Therefore, a Multiple Spatio‐Temporal Features Fusion (MSTFF) network is proposed for STPFF in URT networks. Specifically, the connectivity and passenger flow similarity relationships in URT networks are encoded as physical adjacency graph and virtual functional similarity graph, respectively, and then these two complementary graphs are input into two designed Residual Graph Convolutional Gated Recurrent Unit (RGCGRU) modules to extract their spatio‐temporal features. Second, an Attention‐based Gated Recurrent Unit (AGRU) module is designed to capture global dynamic evolutionary trends in both spatial and temporal dimensions. Finally, a fully connected neural network is used to fuse these features. Experiments on two real passenger flow datasets of URT show that MSTFF performs well on various prediction tasks (15, 30, 45, and 60 min).

Abstract Background Lung adenocarcinoma (LUAD) is a common subtype of non‐small cell lung cancer with high morbidity and mortality rates and is usually detected at advanced stages because of the early onset of metastasis. Adenosine deaminase RNA‐specific 1 (ADAR1) is an RNA editing enzyme that catalyzes the important physiological process of adenosine‐to‐inosine editing and has been shown to participate in the progression of LUAD. Increasing evidence has suggested that immune infiltration of the tumor immune microenvironment has prognostic value for most human solid organ malignancies; however, much is unknown about the functions of ADAR1. Methods The expression of ADAR1 was analyzed in The Cancer Genome Atlas ‐LUAD database and validated in our LUAD cohort. To assess the prognostic value of ADAR1, Kaplan‐Meier survival analyses and Cox regression analyses were carried out in LUAD cohorts. The association between ADAR1 and LUAD immune infiltrates via analyses of cell‐type identification by estimating relative subsets of known RNA transcripts. Abstract Subarachnoid hemorrhage (SAH) is a devastating subtype of stroke with high mortality and disability rate. Meningeal lymphatic vessels (mLVs) are a newly discovered intracranial fluid transport system and are proven to drain extravasated erythrocytes from cerebrospinal fluid into deep cervical lymph nodes after SAH. However, many studies have reported that the structure and function of mLVs are injured in several central nervous system diseases. Whether SAH can cause mLVs injury and the underlying mechanism remain unclear. Herein, single‐cell RNA sequencing and spatial transcriptomics are applied, along with in vivo/vitro experiments, to investigate the alteration of the cellular, molecular, and spatial pattern of mLVs after SAH. First, it is demonstrated that SAH induces mLVs impairment. Then, through bioinformatic analysis of sequencing data, it is discovered that thrombospondin 1 (THBS1) and S100A6 are strongly associated with SAH outcome. Furthermore, the THBS1‐CD47 ligand‐receptor pair is found to function as a key role in meningeal lymphatic endothelial cell apoptosis via regulating STAT3/Bcl‐2 signaling. The results illustrate a landscape of injured mLVs after SAH for the first time and provide a potential therapeutic strategy for SAH based on mLVs protection by disrupting THBS1 and CD47 interaction.

Abstract Coreopsis tinctoria Nutt. has been widely cultivated in Xinjiang, Yunnan as well as other western regions of China. In folk medicine, the flower tea C. tinctoria is often used as an adjunctive therapy for diabetes and Alzheimer's disease. To explore the chemicals responsible for the health benefits of C. tinctoria, a water‐soluble polysaccharide (CT70‐1) was isolated from C. tinctoria. The structure feature analyses revealed that CT70‐1 was a heteropolysaccharide with a relative molecular weight of 2.1×103 Da. It consisted of α‐D‐Glcp‐(1→, →4)‐α‐D‐Glcp‐(1→, →5)‐α‐L‐Araf‐(1→, →6)‐2‐OAc‐β‐D‐Galp‐(1→, →3)‐α‐L‐Rhap‐(1→, →3,6)‐β‐D‐Manp‐(1→, →6)‐β‐D‐Manp‐(1→, →1,6)‐β‐D‐Fruf‐(2→, α‐L‐Araf‐(1→, and →3)‐β‐D‐Manp‐(1→. Further, CT70‐1 exhibited α‐amylase and α‐glucosidase inhibitory activity with IC50 values of 2.79 ± 0.17 mM and 3.00 ± 0.02 mM, respectively. Moreover, CT70‐1 significantly inhibited lipopolysaccharide‐stimulated nitric oxide production of RAW264.7 and BV2 cells with IC50 values of 0.07 mM and 0.12, respectively. In conclusion, the above findings provide a foundation for promising natural ingredient CT70‐1 in the food and pharmaceutical industries.

Background. Post-renal transplant patients have a high likelihood of developing renal cancer. However, the underlying biological mechanisms behind the development of renal cancer in post-kidney transplant patients remain to be elucidated. Therefore, this study aimed to investigate the underlying biological mechanism behind the development of renal cell carcinoma in post-renal transplant patients. Methods. Next-generation sequencing data and corresponding clinical information of patients with clear cell renal cell carcinoma (ccRCC) were obtained from The Cancer Genome Atlas Program (TCGA) database. The microarray data of kidney transplant patients with or without rejection response was obtained from the Gene Expression Omnibus (GEO) database. In addition, statistical analysis was conducted in R software. Results. We identified 55 upregulated genes in the transplant patients with rejection from the GEO datasets (GSE48581, GSE36059, and GSE98320). Furthermore, we conducted bioinformatics analyses, which showed that all of these genes were upregulated in ccRCC tissue. Moreover, a prognosis model was constructed based on four rejection-related genes, including PLAC8, CSTA, AIM2, and LYZ. The prognosis model showed excellent performance in prognosis prediction in a ccRCC cohort. In addition, the machine learning algorithms identified 19 rejection-related genes, including PLAC8, involved in ccRCC occurrence. Finally, the PLAC8 was selected for further research, including its clinical and biological role. Conclusion. In all, our study provides novel insight into the transition from the rejection of renal transplant to renal cancer. Meanwhile, PLAC8 could be a potential biomarker for ccRCC diagnosis and prognosis in post-kidney transplant patients.

Abstract Background Both prepregnancy obesity and gestational diabetes mellitus (GDM) have been linked to adverse neonatal birth weight. However, the mediating role of GDM between prepregnancy obesity and neonatal birth weight is unclear. Method The cohort study included 17 260 singleton pregnant women and their newborns. Participants' demographic characteristics, disease history, family history of the disease, and the perinatal outcomes were recorded. The association between maternal prepregnancy body mass index (BMI) status and small for gestational age (SGA) or large for gestational age (LGA) neonates was analyzed using logistic regressions, before and after adjusting for covariates and GDM. The potential mediation of GDM on the association between prepregnancy BMI and adverse birth weight was examined. Result Multivariate logistic regression demonstrated that prepregnancy underweight women were more likely to deliver SGA neonates compared to those who had normal weights, whereas prepregnancy obese pregnant women were more likely to have LGA neonates. The RMediation analyses illustrated that the mediation effect of GDM on the maternal prepregnancy BMI (continuous variable) and the risk of SGA was not significant, whereas the association between prepregnancy BMI and LGA was statistically mediated by GDM (95%CI of a*b: 0.009‐0.051). The Iacobacci (2012) method indicated that the impact of maternal prepregnancy overweight (Zmediation = 2.418, P = .015) and obesity (Zmediation = 2.165, P = .030) on LGA was partially mediated by GDM, with an indirect effect of 16.3% and 13.1%, respectively. Conclusion Prepregnancy BMI was observed to be associated with SGA and LGA. The association of prepregnancy overweight and obesity with LGA was found to be partially mediated by GDM.

Abstract Aim This study aims to evaluate the feasibility and advantages of immediate urinary catheter removal compared with prolonged indwelling catheterization in lung cancer lobectomy. Design This study was designed as a prospective, single‐centre, randomized and open‐label clinical study. Methods People with lung cancer undergoing lobectomy/pneumonectomy were recruited and randomly allocated to two groups. One group had their urinary catheter removed immediately while the other group had it removed 48 hr after surgery. Results No significant difference in the incidence of postoperative urinary retention (POUR) was observed between the two groups. However, the incidence of postoperative catheter‐associated urinary tract infection (CAUTI) in the immediate removal group (6.7%) was lower than the control group (17.2%) (p = .030). Furthermore, the incidence of catheter‐associated emergence agitation (CAEA) in the control group (25.3%) was higher than the immediate removal group (8.9%) (p = .007). The average length of hospital stay of the immediate removal group [6.51(4–11) days] was shorter than the control group [7.20(5–12) days] (p = .002). Immediate removal of urinary catheter appeared to have fewer complications and shorter hospital stay than delayed removal.

The higher contribution of traffic delay to environmental impacts is urging the highway agencies to take work zone management into the maintenance schemes decision-making. Aiming to understand the role of work zone management in user CO2 emissions reduction, this paper firstly developed a practical methodological framework of traffic delay-related CO2 emissions caused by highway maintenance based on a popular life cycle user cost analysis approach in regard of the microscopic vehicle operation analysis. The method was applied in an actual freeway flexible pavement with 15-year design life in Shaanxi Province, China, covering three types of preventive maintenance, correction maintenance, and rehabilitation. In addition, the impacts of key inputs of proposed method on work zone user CO2 emissions results were checked. The results show that traffic delay attributes to 29.4% of total CO2 emissions of the life cycle of highway pavement maintenance, and 51.8% of work zone user CO2 emissions result from preventive maintenance, especially from micro vehicle operations including speed change and queue near work zone (62% of total work zone user CO2 emissions). The work zone management alternative strategies related to less traffic volume or higher highway capacity including vehicle type limitation and the limited work zone speed have an advantage in reducing the work zone CO2 emissions over changing work zone length or work zone timing. The findings in this paper may present a useful tool and reference for robustly supporting the decision-making on highway maintenance carbon mitigation in work zone traffic.

Background Both drug‐coated balloon (DCB) angioplasty and conventional plain balloon angioplasty (PBA) can be implemented to treat hemodialysis dysfunction. The present study aims to compare the safety and efficacy of these 2 approaches by conducting a meta‐analysis of available randomized controlled trials. Methods and Results PubMed, Cochrane Library, and Embase databases were queried from establishment to January 2021. A total of 18 randomized controlled trials including 877 and 875 patients in the DCB and PBA groups, respectively, were included in the present meta‐analysis. Target lesion primary patency, circuit patency, target lesion revascularization, and mortality were pooled. Odds ratios (ORs) were reported with 95% CIs. To study the theoretical calculation method of the HEL for hard rock, the major factors determining the strength of rock under dynamic loadings are firstly discussed. Secondly, the calculation method of the HEL of hard rock is suggested based on the Lundborg strength criterion, and the parameters influencing the HEL are analyzed and discussed. Thirdly, the HEL obtained by theoretical calculation, numerical modeling, and experiments are compared. Fourthly, the abnormal decreasing or increasing of the HEL in plate impact experiments of hard rock is explained. This research shows that the HEL increases with Poisson’s ratio, the shear strength at zero pressure, the coefficient of internal friction and the plastic limit, and Poisson’s ratio and the plastic limit could be the most important factors. The simplified model of this work can give a rational and practical prediction of the HEL of hard rock in theory, and the complicated interaction between the “damage front” and the diffuse elastic precursor is assumed to explain two special effects of the HEL in plate impact experiments of hard rock, where Poisson’s ratio related to damage levels seems to be the dominant factor.

Abstract Programmable metasurfaces allow dynamic and real‐time control of electromagnetic (EM) waves in subwavelength resolution, holding extraordinary potentials to establish meta‐systems. Achieving independent and real‐time controls of orthogonally‐polarized EM waves via the programmable metasurface is attractive for many applications, but remains considerably challenging. Here, a polarization‐controlled dual‐programmable metasurface (PDPM) with modular control circuits is proposed, which enables a dibit encoding capability in modifying the phase profiles of x‐ and y‐polarized waves individually. The constructed extended interface circuit is able to extend the number of control interfaces from a field programmable gate array by orders of magnitude and also possesses memory function, which enhance hugely the rewritability, scalability, reliability, and stability of PDPM. As a proof‐of‐concept, a wave‐based exclusive‐OR logic gate platform for spin control of circularly‐polarized waves, a fixed‐frequency wide‐angle dual‐beam scanning system, and a dual‐polarized shared‐aperture antenna are demonstrated using a single PDPM. The proposed PDPM opens up avenues for realizing more advanced and integrated multifunctional devices and systems that have two independent polarization‐controlled signal channels, which may find many applications in future‐oriented intelligent communication, imaging, and computing technologies.

Aims. Endoscopic self-expandable metallic stent (SEMS) insertion for acute colonic obstruction caused by colorectal cancer (CRC) is always performed under fluoroscopic guidance. This study evaluated the feasibility and safety of an endoscopic stenting procedure without fluoroscopic guidance. Methods. A total of 36 patients with an acute colonic obstruction caused by CRC underwent endoscopic SEMS insertion using a colonoscope without fluoroscopic guidance, followed by analyses of the technical and clinical success and short-term complications. Results. Total technical success rate and clinical success rate were 91.7% and 86.1%, respectively. The mean procedure time was 21.2±10.3 minutes. There was no stent dislodgement. One case of hematochezia and two cases of tenesmus occurred in patients with left-sided complete obstructions. No other short-term complications occurred. Procedure time, technical success, and clinical success rate were 16.3±9.4 minutes, 93.1%, and 89.6% for left-sided obstructions, respectively, and were 26.8±10.7 minutes, 85.7%, and 71.4% for right-sided obstructions, respectively. For complete obstructions, procedure time, technical success, and clinical success rate were 22.5±8.9 minutes, 90%, and 83.3%, respectively. In the incomplete cases, procedure time, technical success, and clinical success were 13.5±6.7 minutes, 100%, and 100%, respectively. Technical success, clinical success, and short-term complications were not differed between lesion locations and degrees. Conclusions. This simple technique is feasible and safe for palliation of acute colonic obstruction caused by CRC.

Background. Renal anemia is a common complication of hemodialysis patients. Erythropoietin (EPO) hyporesponsiveness has been recognized as an important factor to poor efficacy of recombinant human erythropoietin in the treatment of renal anemia. More importantly, increased erythropoiesis resistance index (ERI) may be associated with inflammation and increased mortality. Objective. The objective of this research was to investigate correlated factors of EPO responsiveness and to clarify the relationships between EPO hyporesponsiveness and cardiovascular mortality and all-cause mortality among maintenance hemodialysis patients. Methods. This prospective cohort study enrolled 276 maintenance hemodialysis patients for a 55-month follow-up to investigate the factors related to ERI and its relationship to all-cause mortality and cardiovascular mortality. Abstract Introduction Patients under chronic obstructive pulmonary disease (COPD) has been reported to be associated with a higher prevalence of cognitive impairment (CI). However, it is still largely unknown whether the aberrant resting‐state spontaneous neuronal activity pattern reflected by the amplitude of low‐frequency fluctuation (ALFF) analysis will be associated with the CI in COPD patients. Materials A total of 28 COPD patients and 26 healthy controls were enrolled in this study. Of all the subjects, structural and functional MRI data, spirometry tests performance and neuropsychological assessments of different cognitive domains were collected. Voxel‐based two‐sample t tests were used to detect brain regions showing differences in the ALFF value between COPD patients and healthy controls. An additional fMRI runs with supplementary oxygen delivery were employed to explore the impact of elevated partial pressure of oxygen (PaO2) or moderate hyperoxia on ALFF in COPD patients and healthy controls respectively. Results More extensive white matter lesion was detected in COPD patients. COPD patients exhibit decreased ALFF value in bilateral basal ganglia areas and right thalamus, and aberrant ALFF value is correlated with PaO2 and pulmonary ventilation function (FEV1%pred). COPD patients performed worse in the Digit Span Test (reverse), Digit Symbol Substitution Test, Trail‐making test (A and B) than controls. After supplementary oxygen inhalation, the ALFF value of basal ganglia and right thalamus significantly increased in the controls, but not in the COPD patients. Conclusions COPD patients mainly exhibit impaired executive function but not long‐term memory in cognitive function assessment. Aberrant ALFF alteration in the deep brain may be directly related to lower PaO2 in COPD patients.

This paper proposes an improved predictor-corrector interior-point algorithm for the linear complementarity problem (LCP) based on the Mizuno-Todd-Ye algorithm. The modified corrector steps in our algorithm cannot only draw the iteration point back to a narrower neighborhood of the center path but also reduce the duality gap. It implies that the improved algorithm can converge faster than the MTY algorithm. The iteration complexity of the improved algorithm is proved to obtain √𝑂(𝑛𝐿) which is similar to the classical Mizuno-Todd-Ye algorithm. Finally, the numerical experiments show that our algorithm improved the performance of the classical MTY algorithm.
