Trophic niche partitioning is a mechanism that facilitates the coexistence of ecologically similar species by sharing their resource use. However, detailed information of the trophic niche in insectivorous birds is usually limited by the lack of accurate identification of consumed food resources. The use of DNA metabarcoding has proved useful for molecular identification of the taxa present in bird faecal samples. Here, we used this molecular technique to study the diets of six steppe passerine species distributed in two Special Protection Areas in central Spain, and to characterize the dietary niche overlap and the prey composition differences between bird species. In total, we distinguished 112 diet items, covering 39 arthropod families of 13 orders. Although significant dietary differences existed in prey species composition, our results indicated a 74% overlap in steppe bird dietary niche, mostly due to high consumption of abundant arthropod prey such as beetles, grasshoppers and spiders in the breeding season by all bird species. The lowest overlap was found for the dietary niches of the Greater Short-toed Lark Calandrella brachydactyla and Dupont's Lark Chersophilus duponti, a scarce and threatened species, which appeared to be the species with the most distinct dietary niche within the community. Our results make a significant contribution to the knowledge of shrub-steppe bird diets and their trophic interactions, indicating that some extent of interspecific resource partitioning occurs in the study area, notably between Dupont's Lark and the Greater Short-toed Lark. Spatial co-occurrence patterns are determined by environmental factors, such as food availability or habitat characteristics and by biotic associations. When resources are limited, competition which implies a dominant hierarchy can shape species assemblage. Here, we study space and time co-occurrence of steppe passerines during the breeding season in a natural steppe habitat, its modulation by environmental filtering, potential biotic interactions and random processes. We applied the joint species distribution model of hierarchical modelling of species community (HMSC) to data on species presence–absence and environmental, temporal and spatial covariates acquired from seven plots in a natural steppe in central Spain during two consecutive years. Our results reveal the patterns of bird species co-occurrence and suggest that this assemblage might be competitively structured. The assemblage appears to be configured around one dominant species, the Eurasian skylark, which establishes principally negative associations with many of the coexistent species. Arthropod biomass is a key element in ecosystem functionality and a basic food item for many species. It must be estimated through traditional costly field sampling, normally at just a few sampling points. Arthropod biomass and plant productivity should be narrowly related because a large majority of arthropods are herbivorous, and others depend on these. Quantifying plant productivity with satellite or aerial vehicle imagery is an easy and fast procedure already tested and implemented in agriculture and field ecology. However, the capability of satellite or aerial vehicle imagery for quantifying arthropod biomass and its relationship with plant productivity has been scarcely addressed. Here, we used unmanned aerial vehicle (UAV) and satellite Sentinel-2 (S2) imagery to establish a relationship between plant productivity and arthropod biomass estimated through ground-truth field sampling in shrub steppes. We UAV-sampled seven plots of 47.6–72.3 ha at a 4-cm pixel resolution, subsequently downscaling spatial resolution to 50 cm resolution. In parallel, we used S2 imagery from the same and other dates and locations at 10-m spatial resolution. We related several vegetation indices (VIs) with arthropod biomass (epigeous, coprophagous, and four functional consumer groups: predatory, detritivore, phytophagous, and diverse) estimated at 41–48 sampling stations for UAV flying plots and in 67–79 sampling stations for S2. VIs derived from UAV were consistently and positively related to all arthropod biomass groups. Three out of seven and six out of seven S2-derived VIs were positively related to epigeous and coprophagous arthropod biomass, respectively. The blue normalized difference VI (BNDVI) and enhanced normalized difference VI (ENDVI) showed consistent and positive relationships with arthropod biomass, regardless of the arthropod group or spatial resolution. Our results showed that UAV and S2-VI imagery data may be viable and cost-efficient alternatives for quantifying arthropod biomass at large scales in shrub steppes. Passive acoustic monitoring is a non-invasive tool for automated wildlife monitoring. This technique has several advantages and addresses many of the biases related to traditional field surveys. However, locating animal sounds using autonomous recording units (ARUs) can be technically challenging and therefore ARUs have traditionally been little employed to estimate animal density. Nonetheless, several approaches have been proposed in recent years to carry out acoustic-based bird density estimations. We conducted a literature review of studies that used ARUs for estimating bird densities or bird abundances in order to describe the applications and improve future monitoring programmes. We detected a growing interest in the use of ARUs for estimating bird density in the last 6 years (2014–19), with a total of 31 articles assessing the topic. The most common approach was to estimate the relationship between the number of vocalizations per recording time with bird density or bird abundance estimated in the field (61%). In 26 studies (79%), bird estimates obtained by human surveyors agreed with those obtained using ARUs. Some approaches have proven able to reduce biases in acoustic surveys, such as considering imperfect detection (spatially explicit capture–recapture, using microphone arrays), applying paired acoustic sampling to control for different sampling radius between humans and ARUs, or including relative sound level measurements that allow researchers to estimate bird distance to recorder. However, several studies did not include any covariates to reduce existing biases and some did not estimate the sampling radius of the recorder, which may hamper future comparisons between human and ARU surveys. Future studies should include a measurement of the sampling radius of the recorder employed to be able to obtain density estimations using ARUs. Finally, we provide some guidelines to improve the applicability of ARUs to infer bird population estimates in future studies. Over 3 years, we studied grouping behavior of guanaco (Lama guanicoe) families in Chilean Patagonia during the birthing season and determined their response to variation in predator occurrence and perceived predation risk (habitat structure, calf/adult rate, and density of conspecifics). We considered the effect of two predators, puma (Puma concolor) and culpeo fox (Lycalopex culpaeus). We measured two common (family group size and vigilance) and one novel (family group cohesion) behavioral responses of guanaco. Our results show that guanaco family groups adapted their grouping behavior to both predator occurrence and perceived predation risk. Larger family groups were found in open habitats and areas with high puma occurrence, while guanacos stayed in small family groups in areas with high shrub cover or low visibility. Group cohesion increased in areas with higher occurrence of pumas and culpeo foxes, and also increased in smaller family groups and in areas with low guanaco density. Vigilance (number of vigilant adults) was mainly related to group size and visibility, increasing in areas with low visibility, while residual vigilance (vigilance after removing the group-size effect) did not vary with the explanatory variables examined. Habitat partitioning is a common ecological mechanism to avoid competition among coexisting species, and the introduction of new species into existing assemblages can increase competitive pressures. However, situations of species in allopatry and sympatry only differing in species presence but not in environmental conditions are scarce. Thus, discerning whether niche segregation arises from competition or from different habitat preferences is usually unfeasible. Here, we analyse species’ habitat niches in an assemblage of native and introduced herbivores in southern Patagonia. We test if niche overlap is higher between native and domestic herbivores than among natives as expected from the relatively short time of coexistence, and we evaluate the effect of intra- and interspecific competition on niche breadth. We use a probabilistic multidimensional approach and null models to evaluate overlap and changes in niche dimensions. Overlap among native species is low as expected for species coexisting in evolutionary time. In native-domestic species pairs, niche overlap was higher than among natives, although showing some niche segregation indicating niche differentiation in ecological time. Moreover, the presence of domestic species was associated with niche narrowing of both native and introduced species, revealing interspecific density-dependent effects on their habitat niche during resource shortage periods.

Sal1p, a novel Ca2+-dependent ATP-Mg/Pi carrier, is essential in yeast lacking all adenine nucleotide translocases. By targeting luciferase to the mitochondrial matrix to monitor mitochondrial ATP levels, we show in isolated mitochondria that both ATP-Mg and free ADP are taken up by Sal1p with a K(m) of 0.20 +/- 0.03 mM and 0.28 +/- 0.06 mM respectively. Nucleotide transport along Sal1p is strictly Ca2+ dependent. Ca2+ increases the V(max) with a S(0.5) of 15 muM, and no changes in the K(m) for ATP-Mg. Glucose sensing in yeast generates Ca2+ transients involving Ca2+ influx from the external medium. We find that carbon-deprived cells respond to glucose with an immediate increase in mitochondrial ATP levels which is not observed in the presence of EGTA or in Sal1p-deficient cells. Moreover, we now report that during normal aerobic growth on glucose, yeast mitochondria import ATP from the cytosol and hydrolyse it through H+-ATP synthase. We identify two pathways for ATP uptake in mitochondria, the ADP/ATP carriers and Sal1p. Thus, during exponential growth on glucose, mitochondria are ATP consumers, as those from cells growing in anaerobic conditions or deprived of mitochondrial DNA which depend on cytosolic ATP and mitochondrial ATPase working in reverse to generate a mitochondrial membrane potential. In conclusion, the results show that growth on glucose requires ATP hydrolysis in mitochondria and recruits Sal1p as a Ca2+-dependent mechanism to import ATP-Mg from the cytosol. Whether this mechanism is used under similar settings in higher eukaryotes is an open question.

The influence of arthropod availability on the formation of male little bustard breeding territories in central Spain was studied in two consecutive years. Arthropods (68,217 individuals) were sampled using pitfall trapping and male territories estimated by means of kernel functions. Hymenopterans (ants), beetles, mites and spiders were the main taxa found in the samples, while the highest contributions in terms of biomass came from hymenopterans, beetles and orthopterans. The only group that exhibited significant biomass differences between territory and non-territory locations was that of the beetles; this pattern was consistent between years. Carabidae was the main beetle family in terms of biomass, followed by other large beetles such as Tenebrionidae, Cleridae, Scarabeidae and Elateridae. The biomass of Carabidae that were larger than 15 mm was significantly higher within territories than outside them. The biomass of Cleridae was greater in fallows, while Elateridae were more abundant in arable fields. The total biomass of beetles that were larger than 15 mm was higher in arable fields than in fallows. Family richness differed between years, while diversity showed differences between agricultural fields (it was highest in fallows), although these variables did not differ between territory and non-territory locations. Results indicate that large beetles (particularly carabids) seem to be a valuable food resource that is defended by little bustard males during the highly energy-demanding breeding season, which is discussed here in relation to habitat selection, the mating system and its implications for the conservation of the species.

Incorporation of a human bone allograft requires osteoclast activity and growth of recipient osteoblasts. The aim of this work was to study the effects produced by autoclavated and −80°C frozen bone allografts on osteoblast proliferation and synthesis of interleukin 6 (IL6), activator of bone resorption, aminoterminal propeptide of procollagen I (PINP), marker of bone matrix formation, and osteoprotegerin (OPG), inhibitor of osteoclast activity and differentiation. Allografts were obtained from human femoral heads. Human osteoblasts were cultured in the presence (problem group) or in the absence (control group) of allografts during 15 days. Allografts produced a decrease in osteoblast proliferation in the first week of the experiment, and an increase in IL6 mRNA, both at 3 h and 2 days, and an increase in the IL6 released to the culture medium the second day of the experiment. We found a decrease in OPG released to the culture on the 2nd and fourth days. These results suggest an increase in bone resorption and a decrease in bone formation in the first week of the experiment. In the second week, allografts produced an increase in osteoblast proliferation and PINP release to the culture medium, indicating an increase in bone formation; an increase in OPG released to the culture medium, which would indicate a decrease in bone resorption; and a decrease in IL6, indicating a decrease in bone resorption stimulation. These results demonstrate that autoclavated and −80°C frozen bone allografts produce in bone environmemt changes that regulate their own incorporation to the recipient bone. © 2007 Orthopaedic Research Society. BACKGROUND The aim of this work was to evaluate the effect produced by conditioned medium from human prostatic carcinoma cell (PC-3) culture on human osteoblast (HOB) interleukin 6 (IL-6) synthesis. METHODS PC-3 cells were cultured in Ham's F12K medium with 10% fetal calf serum (FCS) up to confluence. Medium was changed by Dulbecco modified Eagle medium (DMEM)/F12K (1:1) with 0.1% bovine serum albumin. Cells were cultured for 24 hr, and medium (PC-3-CM) was collected. HOBs were cultured up to confluence, and after 48 hr without FCS, medium was removed and PC-3-CM was added to the wells. After 24 hr, supernatant was collected for the determination of IL-6. In another experiment, HOBs were cultured up to confluence in Petri dishes, and after 48 hr without FCS, PC-3-CM or DMEM/F12K (1:1) was added. After different periods of time, medium was removed, and total RNA was extracted. IL-6 mRNA was quantified using reverse transcription polymerase chain reaction. RESULTS PC-3-CM significantly enhanced IL-6 secretion into HOB culture supernatants (between 1,812% and 372%, depending on the osteoblastic line) with respect to HOBs cultured in DMEM/F12K. PC-3-CM also produced an increase in IL-6 mRNA levels in HOBs. CONCLUSIONS Prostate carcinoma cells (PC-3) produce a factor or factors that enhance the synthesis and release of IL-6, a known activator of bone resorption.
