Purpose: Recent observations of several preferred orientations of diffusion in deep white matter may indicate either (a) that axons in different directions are independently bundled in thick sheets and function noninteractively, or more interestingly, (b) that the axons are closely interwoven and would exhibit branching and sharp turns. This study aims to investigate whether the dependence of dMRI Q-ball signal on the interpulse time Δ can decode the smaller-than-voxel-size brain structure, in particular, to distinguish scenarios (a) and (b)., Methods: High-resolution Q-ball images of a healthy brain taken with b = 8000  s/mm 2 for 3 different values of Δ were analyzed. The exchange of water molecules between crossing fibers was characterized by the fourth Fourier coefficient f 4 ( Δ ) of the signal profile in the plane of crossing. To interpret the empirical results, a model consisting of differently oriented parallel sheets of cylinders was developed. Diffusion of water molecules inside and outside cylinders was simulated by the Monte Carlo method., Results: Simulations predict that f 4 ( Δ ) , agreeing with the empirical results, must increase with Δ for large b-values, but may peak at a typical Δ that depends on the thickness of the cylinder sheets for intermediate b-values. Thus, the thickness of axon layers in voxels with 2 predominant orientations can be detected from empirical f 4 ( Δ ) taken at smaller b-values., Conclusion: Based on the simulation results, recommendations are made on how to design a dMRI experiment with optimal b-value and range of Δ in order to measure the thickness of axon sheets in the white matter, hence to distinguish (a) and (b)., (© 2021 International Society for Magnetic Resonance in Medicine.)

A diffusion-weighted (DW) template in a standard coordinate system is often necessary for the analysis of white matter (WM) structures using DW images. Although several DW templates have been constructed in the ICBM-152 space, a template for diffusion spectrum imaging (DSI) is still lacking. In this study, we developed a DSI template in the ICBM-152 space from 122 healthy adults. This high quality template, NTU-DSI-122, was built through incorporating the macroscopic anatomical information using high-resolution T1 -weighted images and the microscopic structural information obtained from DSI datasets. Two evaluations were conducted to examine the quality of NTU-DSI-122. The first evaluation examined the anatomical consistency of NTU-DSI-122 in matching to the ICBM-152 coordinate system. The results showed that this template matched to the ICBM-152 templates very well across the whole brain, not only in the deep white matter regions as other DW templates but also in the superficial white matter regions. In the second evaluation, a large number of independent diffusion tensor imaging (DTI) datasets were registered to the DTI template derived from NTU-DSI-122. The examination was performed by quantifying the anatomical consistency among the registered DTI datasets. The results showed that using NTU-DSI-122 as the registration template the registered DTI datasets can achieve high anatomical alignment. Both evaluations demonstrate that NTU-DSI-122 is a useful high quality DW template. Therefore, NTU-DSI-122 can serve as a representative DSI dataset for a healthy adult population, and will be of potential value for brain research and clinical applications. Quantitative diffusion imaging is a powerful technique for the characterization of complex tissue microarchitecture. However, long acquisition times and limited signal-to-noise ratio represent significant hurdles for many in vivo applications. This article presents a new approach to reduce noise while largely maintaining resolution in diffusion weighted images, using a statistical reconstruction method that takes advantage of the high level of structural correlation observed in typical datasets. Compared to existing denoising methods, the proposed method performs reconstruction directly from the measured complex k-space data, allowing for gaussian noise modeling and theoretical characterizations of the resolution and signal-to-noise ratio of the reconstructed images. In addition, the proposed method is compatible with many different models of the diffusion signal (e.g., diffusion tensor modeling and q-space modeling). The joint reconstruction method can provide significant improvements in signal-to-noise ratio relative to conventional reconstruction techniques, with a relatively minor corresponding loss in image resolution. Simultaneous multislice Echo Planar Imaging (EPI) acquisition using parallel imaging can decrease the acquisition time for diffusion imaging and allow full-brain, high-resolution functional MRI (fMRI) acquisitions at a reduced repetition time (TR). However, the unaliasing of simultaneously acquired, closely spaced slices can be difficult, leading to a high g-factor penalty. We introduce a method to create interslice image shifts in the phase encoding direction to increase the distance between aliasing pixels. The shift between the slices is induced using sign- and amplitude-modulated slice-select gradient blips simultaneous with the EPI phase encoding blips. This achieves the desired shifts but avoids an undesired "tilted voxel" blurring artifact associated with previous methods. We validate the method in 3× slice-accelerated spin-echo and gradient-echo EPI at 3 T and 7 T using 32-channel radio frequency (RF) coil brain arrays. The Monte-Carlo simulated average g-factor penalty of the 3-fold slice-accelerated acquisition with interslice shifts is <1% at 3 T (compared with 32% without slice shift). Combining 3× slice acceleration with 2× inplane acceleration, the g-factor penalty becomes 19% at 3 T and 10% at 7 T (compared with 41% and 23% without slice shift). Methods are presented to map complex fiber architectures in tissues by imaging the 3D spectra of tissue water diffusion with MR. First, theoretical considerations show why and under what conditions diffusion contrast is positive. Using this result, spin displacement spectra that are conventionally phase-encoded can be accurately reconstructed by a Fourier transform of the measured signal's modulus. Second, studies of in vitro and in vivo samples demonstrate correspondence between the orientational maxima of the diffusion spectrum and those of the fiber orientation density at each location. In specimens with complex muscular tissue, such as the tongue, diffusion spectrum images show characteristic local heterogeneities of fiber architectures, including angular dispersion and intersection. Cerebral diffusion spectra acquired in normal human subjects resolve known white matter tracts and tract intersections. Finally, the relation between the presented model-free imaging technique and other available diffusion MRI schemes is discussed.

In order to improve the performance of magnetic resonance imaging and spectroscopy of atherosclerotic plaque the potential use of novel radiofrequency coil structures with sensitive detection volumes tailored to the geometry of the arterial wall was investigated. It was found that a cylindrical meanderline (zig-zag) coil design provides a sensitive volume that is restricted to a cylindrical shell, thereby maximizing the filling factor and signal-to-noise ratio for plaques while reducing the intense blood signal. The cylindrical meanderline coil has the added advantages of an open interior, which allows for unimpeded blood flow during scanning, and the potential to be expanded against the walls of the artery, thereby stabilizing the coil against the pulsatile blood flow and minimizing motion artifacts. The performance of cylindrical meanderline coils with theoretical simulations of the electromagnetic fields as well as with experimental images of test objects (phantoms) and human endarterectomy surgical specimens is demonstrated. This radically new RF coil geometry offers the potential to improve the efficiency of MR data acquisition in medical applications in which curved surfaces or slabs contain the material of interest., (Copyright 2004 Wiley-Liss, Inc.)

The mechanism of ventricular thickening in normal humans was investigated using in vivo MRI. The hypothesis that myocardial laminar sheets contribute to ventricular thickening predominantly via sheet shear and sheet extension, as previously found invasively in canine studies at particular ventricular sites, was tested. In normal human subjects, registered images of myocardial sheet architecture and strain at the mid-left ventricle (mid-LV) at mid-systole were acquired with diffusion and strain MRI. Sheet function was analyzed by computing myocardial strain in the local fiber-sheet coordinates. In general, myocardial sheets contribute to ventricular thickening through all three cross-fiber strain components: sheet shear, sheet extension, and sheet-normal thickening (previously undocumented). Each of these components demonstrated substantial spatial heterogeneity, with sheet shear and sheet extension usually predominant in the anterior free wall, and sheet-normal thickening predominant near the right ventricular (RV) insertions. However, considerable intersubject variability was also found. In all cases, the contributions to thickening of fiber strains were small. Sheet function in normal humans was found to be heterogeneous and variable, contrasting with the uniform and symmetric ventricular patterns of fiber shortening and wall thickening. Image distortion due to field gradient eddy currents can create image artifacts in diffusion-weighted MR images. These images, acquired by measuring the attenuation of NMR signal due to directionally dependent diffusion, have recently been shown to be useful in the diagnosis and assessment of acute stroke and in mapping of tissue structure. This work presents an improvement on the spin-echo (SE) diffusion sequence that displays less distortion and consequently improves image quality. Adding a second refocusing pulse provides better image quality with less distortion at no cost in scanning efficiency or effectiveness, and allows more flexible diffusion gradient timing. By adjusting the timing of the diffusion gradients, eddy currents with a single exponential decay constant can be nulled, and eddy currents with similar decay constants can be greatly reduced. Magnetic resonance (MR) diffusion tensor imaging (DTI) can resolve the white matter fiber orientation within a voxel provided that the fibers are strongly aligned. However, a given voxel may contain a distribution of fiber orientations due to, for example, intravoxel fiber crossing. The present study sought to test whether a geodesic, high b-value diffusion gradient sampling scheme could resolve multiple fiber orientations within a single voxel. In regions of fiber crossing the diffusion signal exhibited multiple local maxima/minima as a function of diffusion gradient orientation, indicating the presence of multiple intravoxel fiber orientations. The multimodality of the observed diffusion signal precluded the standard tensor reconstruction, so instead the diffusion signal was modeled as arising from a discrete mixture of Gaussian diffusion processes in slow exchange, and the underlying mixture of tensors was solved for using a gradient descent scheme. We present a method for diffusion tensor MRI in the beating heart that is insensitive to cardiac motion and strain. Using a stimulated echo pulse sequence with two electrocardiogram (ECG) triggers, diffusion-encoding bipolar gradient pulses are applied at identical phases in consecutive cardiac cycles. In this experiment, diffusion is encoded at a single phase in the cardiac cycle of less than 30 ms in duration. This encoding produces no phase shifts for periodic motion and is independent of intervening strains. Studies in a gel phantom with cyclic deformation confirm that by using this sequence we can map the diffusion tensor free of effects of cyclic motion. In normal human subjects, myocardial diffusion eigenvalues measured with the present method showed no significant change between acquisitions encoded at maximum contractile velocity (peak) vs. at myocardial standstill (end-systole), demonstrating motion independence of in vivo diffusion measurements. Diffusion tensor images acquired with the present method agree with registered data acquired with a previous cardiac diffusion MRI method that was shown to be valid in the normal heart, strongly supporting the validity of MRI diffusion measurement in the beating heart. Myocardial sheet and fiber dynamics measured during systole showed that normal human myocardial sheet orientations tilt toward the radial during systole, and fiber orientations tilt toward the longitudinal, in qualitative agreement with previous invasive studies in canines. A method to encode multiple two-dimensional Fourier transform (2D FT) images within a single echo train is presented. This new method, simultaneous echo refocusing (SER), is a departure from prior echo planar image (EPI) sequences which use repeated single-shot echo trains for multislice imaging. SER simultaneously acquires multiple slices in a single-shot echo train utilizing a shared refocusing process. The SER technique acquires data faster than conventional multislice EPI since it uses fewer gradient switchings and fewer preparation pulses such as diffusion gradients. SER introduces a new capability to simultaneously record multiple spatially separated sources of physiologic information in subsecond image acquisitions, which enables several applications that are dependent on temporal coherence in MRI data including velocity vector field mapping and brain activation mapping., (Copyright 2002 Wiley-Liss, Inc.)

Quantitative measurements of inherently three-dimensional (3D) cardiac strain and strain rate require 3D data; MRI provides uniquely high sensitivity to material strain by combining phase contrast with single-shot acquisition methods, such as echo-planar imaging (EPI). Previous MRI methods applied to 3D strain used multiple two-dimensional (2D) acquisitions and suffered loss of sensitivity due to magnification within the strain calculation of physiologic noise related to cardiac beat-to-beat variability. In the present work, each single-shot acquisition generates 3D image data by acquiring two contiguous 2D Fourier transform (FT) images in a single echo train of an EPI readout. Although strain encoding divides across multiple EPI shots, each strain component is computed only within single-shot data, avoiding noise magnification. Strain tensor maps are displayed using iconic 3D graphics or a simple color code of tensor shape. In a deforming gel phantom, gradient-recalled echo (GRE) MRI movies of 3D strain rates match expected strain fields. In normal human subjects, 3D strain rate tensor movies of heart and brain comprising seven slices in each of seven cardiac phases were completed in 56 heartbeats. Stimulated echo (STE) MRI of net systolic 3D strain was also demonstrated. Cardiac diffusion MRI with diffusion encoding that spans a cardiac cycle is complicated by myocardial strains. This paper presents a method to obtain accurate diffusion data without strain correction. Owing to the synchrony of normal cardiac motion, there are time points in the cardiac cycle, "sweet spots," when the cardiac configuration approximates its temporal mean. If the diffusion is encoded then, the net effect of strain on the observed diffusion approximates zero. To test this, MRI diffusion and strain-rate movies are performed on cyclically deformed gel phantoms and in five normal subjects. In phantoms, the sweet spots predicted from the strain time curves agree with the times when the observed diffusion equals the true diffusion. In humans, the strain prediction of the sweet spots and the locations determined by the diffusion trace show a high correlation, r = 0.99. Methods are presented to image the fiber architecture of the human myocardium in vitro and in vivo. NMR images are obtained of the diffusion anisotropy tensor, indicative of local myofiber orientation. Studies of cardiac necropsy specimens demonstrate classic features of ventricular myoarchitecture including the continuous endocardial to epicardial variation of fiber helix angles (angles to the ventricular circumferential direction) of approximately +1.3 to -1.3 radians. Cross-fiber anisotropy is also observed. In the beating heart, NMR diffusion data must be corrected for the effects of myocardial deformation during the cardiac cycle. This correction can be performed using an independent MRI method to map the strain-rate tensor field of the myocardium through time. Combining fiber orientation with local myocardial strain rate, local rates of myocardial fiber shortening may be computed.

We present methods to acquire and analyze NMR movies of myocardial strain rates in which cardiac motion is suppressed and the histories of strain rates are accurately defined for each voxel of myocardial tissue. By means of stimulated echoes, the myocardial strain-rate tensor is phase-encoded at progressive delays in the cardiac cycle while the slice-select and spatial encoding of the image acquisition are performed at a constant cardiac delay. In these data, every image shows the identical myocardial tissue, and the anatomic configuration of the heart appears motionless. The myocardial strain-rate data, however, indicate the state of motion which existed in this slice at the time of the velocity phase-encoding, and these data evolve with the progressive delay as a movie. Using echo-planar MRI, motionless movies of myocardial strain rate of four to eight cardiac delays are obtained in a breath-hold. As an application, a quantitative characterization of cardiac mechanical synchrony is accomplished by principal component analysis (PCA) of the time series of strain rates.

Existing magnetic resonance methods for diffusion imaging, including echo planar, are ineffective in the beating heart due to motion-induced signal attenuation. To overcome this problem, we used a diffusion-weighted stimulated echo-echo planar magnetic resonance imaging sequence. The two lobes of the diffusion-sensitizing gradient were synchronized to the same point in successive cardiac cycles in order to fix the cardiac position and avoid bulk motion effects. The apparent diffusion coefficients (ADCs) of the interventricular septum in 12 healthy subjects for diffusion gradients along the x-, y-, and z-directions were 1.40 +/- 0.27, 1.48 +/- 0.35, and 1.78 +/- 0.27 x 10(-3) mm2/s. The ADCs of the interventricular septum in a second group of 15 healthy subjects for diffusion gradients along the short axis, horizontal and vertical long axes were 0.92 +/- 0.15, 1.50 +/- 0.15, and 1.10 +/- 0.24 x 10(-3) mm2/s. Because the ADCs were less than the measured values for skeletal muscle and their standard deviations were low, it seems unlikely that bulk motion effects made the dominant contribution to the measured myocardial ADC for the interventricular septum, although motion and/or susceptibility artifacts frequently degraded measurements in the free wall of the left ventricle. Additional evidence that ADC was not predominantly determined by wall motion was obtained in a third group of patients with various cardiac abnormalities, in whom there was only a weak correlation between ADC and ejection fraction.(ABSTRACT TRUNCATED AT 250 WORDS)

The process of MRI signal attenuation due to in-plane intravoxel velocity inhomogeneity is described. Given rigid rotation or linear shear, velocity phase-sensitivity will induce a phase distribution that varies linearly with position, which is exactly equivalent to the effect of a spatial phase encoding gradient pulse. It follows that the effect of such motion on the raw MRI signal is to displace it a fixed distance in kappa-space. Attenuation becomes marked when the center of the spin-echo reaches an edge of kappa-space, which happens when intravoxel phase shifts reach pi radian/voxel. Because spin echoes are typically peaked sharply at center, this attenuation usually is abrupt. Analytic and numerical simulations of linear and nonlinear velocity fields confirm abrupt MRI attenuation where phase dispersion exceeds pi radian/voxel. Examples of this phenomenon include the abrupt loss of blood signal adjacent the vessel wall in laminar flow, abrupt loss of subendocardial signal in early diastole, and sudden disappearance due to rotation of a kidney during a measurement of diffusion.

Detection and quantification of flow of the left anterior descending (LAD) coronary artery in healthy volunteers are demonstrated using echo-planar imaging (EPI). A time-of-flight (TOF) model was used to derive coronary flow velocities from wash-in curves, free of cardiac wall motion contamination. Short-axis cardiac studies were performed using a gated, gradient echo EPI technique to limit the effect of cardiac wall motion on coronary vessel imaging. A series of 10 to 20 single or multislice images were acquired within a single breath-hold. Real-time cine series showed the LAD coronary artery with a detectability of 91% (n = 23) and revealed beat-to-beat variability in vessel position of a magnitude equal to or greater than its diameter. Flow velocity was measured in the proximal portion of the artery at rest and during exercise. The data demonstrated the known phasic pattern of LAD flow: Vsystole < or = 5 cm/s and peak Vdiastole = 14 +/- 3 cm/s (n = 11, V = mean laminar flow velocity). During isometric exercise, a LAD flow velocity increase (52 +/- 24%) was detected in eight of nine subjects. The capacity of the EPI TOF method to detect flow velocity changes should prove clinically useful for future assessment of coronary flow reserve.

A magnetic resonance imaging (MRI) method is presented to detect, localize, and quantify myocardial kinematics by measuring the material rate-of-strain tensor at each pixel in gated NMR images of the heart. The immediate, local effect of muscular activity is self-deformation, and the strain tensor is the basic mathematical device by which such deformation may be quantified. The present method, called "strain-phase" MRI (SP-MRI), entails four steps: (1) the velocity of the myocardium is encoded by means of a set of motion-sensitive NMR image acquisitions, one image per velocity component; (2) the spatial derivatives of the velocity are computed at each pixel; (3) the velocity-derivative data are combined to compute an approximation of the strain-rate tensor of the myocardium at each pixel; and (4) the strain-rate tensor data are simplified to produce a color-coded functional image which represents strain-rate components which are of particular biomedical interest in the myocardium. We present a quantitative SP-MRI methodology suited to conventional MRI, and in addition present an "echo-planar" methodology, able to produce qualitative functional images of myocardial kinematics at almost real-time speeds. Two-dimensional strain-phase MRI data acquired in normal human subjects are presented. These data demonstrate the practicability of SP-MRI in vivo, that SP-MRI resolves myocardial kinematics at the single-pixel scale, having resolution comparable to that of conventional MRI, and that SP-MRI data may have a signal-to-noise ratio up to 50% as great as that of the conventional MRI data from which they are produced. SP-MRI measurements of the local instantaneous strain rates in the human left ventricular myocardium are quantitatively consistent with known transmural average values of myocardial strain.

Projective MR images of vascular anatomy and flow are performed at 0.14 T by using phase contrast to suppress the signal contribution of the stationary background. The source of the contrast is the distinctive phase evolution of moving protons under the influence of the read-out gradient of a conventional two-dimensional Fourier transform (2D FT) spin-echo pulse sequence. By using short echo times, small phase shifts may be obtained. When phase shifts are less than about 45 degrees, the phase contrast assumes a simple and useful form. The flow image intensity at any pixel becomes proportional to the net flux or flow volume of protons which cross the corresponding voxel. This proportionality is demonstrated in images of flow phantoms as is the reproducibility of measured flow volume under a variety of transformations of imaging conditions and of the subject. Projective images gated in vivo produce angiographic views of arteries and veins, in systole and diastole, in the neck of a dog and in the lower extremities of a human subject.

Using a one-dimensional rapid imaging technique, we have found that injection of lanthanide chelates such as Gd(DTPA)2- leads to a significant decrease (50%) in rat brain signal intensity at 1.45 T using T2-weighted pulse sequences; however, no effect of comparable size is observed with T1-weighted pulse sequences. The transient effect and its kinetics were followed with a temporal resolution of between 1 and 8 s. Experiments with different lanthanide chelates show that the observed decrease in signal intensity correlates with the magnetic moment of each agent but not with their longitudinal relaxivity. Three-dimensional chemical-shift resolved experiments demonstrate significant line broadening in brain during infusion with Dy(DTPA)2-. Our results show that the cause of this effect is the difference in susceptibility between the capillaries, containing the contrast agent, and the surrounding tissue. As a result of these susceptibility differences, field gradients are produced in the tissue and diffusion of water through these gradients leads to a loss of spin phase coherence and thus a decrease in signal intensity. We propose this as a new type of contrast agent mechanism in NMR. The effect and its kinetics are likely to be related to important physiological parameters such as cerebral blood volume and cerebral blood flow, and do not depend on a breakdown of the blood-brain barrier as do conventional contrast agent techniques.

Most magnetic resonance imaging (MRI) techniques are subject to a "motional blurring" arising from the acquisition of data in the presence of a frequency-encoding gradient. The Fourier transform of the signal from a spin moving along a magnetic field gradient obeys an equation analogous to the free space Schrödinger equation. Computer simulations of the Bloch equations illustrate the implications of this motional blurring in MRI.

In current magnetic resonance imaging (MRI), valuable information must often be discarded because the NMR signal has greater dynamic range than the analog-to-digital converter (ADC) hardware. Typically, a small set of high-intensity data points near the center of the spin echo is responsible for most of the MRI data dynamic range. We predict that it is possible to reduce the dynamic range of the MRI spin echo by incorporating an identical nonlinear gradient pulse into each repetition of the imaging pulse sequence, prior to data sampling. This pulse converts the phase distribution of the subject, ordinarily a linear function of image coordinates, into a nonlinear function. A nonlinear phase distribution can have a negligible impact on image magnitude and yet a profound impact on spin-echo magnitude. Given a nonlinear phase distribution, there will no longer be a single data point at which all of the protons have an identical phase (the echo center). Instead, the protons become phase coherent on a piecemeal basis, the echo peak is smoothed out, and its maximum amplitude and dynamic range are greatly diminished. Using gradient pulses of quadratic spatial variation, we estimate that maximum echo amplitude and dynamic range can be reduced in most cases by an order of magnitude.
