Abstract Short‐term load forecasting is critical for power system planning and operations, and ensemble forecasting methods for electricity loads have been shown to be effective in obtaining accurate forecasts. However, the weights in ensemble prediction models are usually preset based on the overall performance after training, which prevents the model from adapting in the face of different scenarios, limiting the improvement of prediction performance. In order to improve the accurateness and validity of the ensemble prediction method further, this paper proposes an ensemble deep reinforcement learning approach using Q‐learning dynamic weight assignment to consider local behaviours caused by changes in the external environment. Firstly, the variational mode decomposition is used to reduce the non‐stationarity of the original data by decomposing the load sequence. Then, the recurrent neural network (RNN), long short‐term memory (LSTM), and gated recurrent unit (GRU) are selected as the basic power load predictors. Finally, the optimal weights are ensembled for the three sub‐predictors by the optimal weights generated using the Q‐learning algorithm, and the final results are obtained by combining their respective predictions. The results show that the forecasting capability of the proposed method outperforms all sub‐models and several baseline ensemble forecasting methods.

Key Clinical Message Ultrasound‐guided core needle biopsy combined with immunohistochemistry and molecular testing could improve the diagnostic accuracy of bone metastases from follicular thyroid carcinoma, help to predict distant metastasis and prognosis. Abstract Metastatic thyroid follicular carcinoma presenting initially with bone lesion is uncommon, its prime symptom is gradual onset, localized pain. Patient with bone metastasis who were diagnosed before thyroidectomy had a higher rate of mortality, clinician should be cautious in eliciting the clinical history and this insidious symptom in middle age group, carry out further examination. We are presenting two case reports of a follicular thyroid carcinoma with bone metastasis, ultrasound‐guided core needle biopsy combined with immunohistochemistry (IHC) were carried out by our clinical team to determine the source and nature of the tumor, relevant literature was reviewed, molecular testing was discussed, we believe core needle biopsy combined with IHC and molecular testing improve the diagnostic accuracy of bone metastases from follicular thyroid carcinoma.

Abstract The prevalence of infertility caused by endometrial defects is steadily increasing, posing a significant challenge to women's reproductive health. In this study, injectable “homing‐like” bioactive decellularized extracellular matrix short‐fibers (DEFs) of porcine skin origin are innovatively designed for endometrial and fertility restoration. The DEFs can effectively bind to endometrial cells through noncovalent dipole interactions and release bioactive growth factors in situ. In vitro, the DEFs effectively attracted endometrial cells through the “homing‐like” effect, enabling cell adhesion, spreading, and proliferation on their surface. Furthermore, the DEFs effectively facilitated the proliferation and angiogenesis of human primary endometrial stromal cells (HESCs) and human umbilical vein endothelial cells (HUVECs), and inhibited fibrosis of pretreated HESCs. In vivo, the DEFs significantly accelerated endometrial restoration, angiogenesis, and receptivity. Notably, the deposition of endometrial collagen decreased from 41.19 ± 2.16% to 14.15 ± 1.70% with DEFs treatment. Most importantly, in endometrium‐injured rats, the use of DEFs increased the live birth rate from 30% to an impressive 90%, and the number and development of live births close to normal rats. The injectable “homing‐like” bioactive DEFs system can achieve efficient live births and intrauterine injection of DEFs provides a new promising clinical strategy for endometrial factor infertility.

Abstract Background Immune checkpoint inhibitors (ICIs) are widely used in recurrent or metastatic head and neck squamous cell carcinoma (R/M HNSCC); however, the toxicity profiles are inconclusive. Methods Clinical trials evaluating ICIs for R/M HNSCC were searched from online databases. The characteristics of the studies and the results of incidences of any grade treatment‐related adverse events (trAEs), grade three or more trAEs, treatment‐related deaths, trAEs leading to discontinuation of treatment, and specific trAEs were extracted. Results Twenty studies with 3756 patients were included. The pooled incidences of any grade trAEs, grade three or more trAEs, treatment‐related deaths, trAEs leading to discontinuation of treatment for overall population were 62.07% (95% CI, 59.07%–65.02%), 13.82% (95% CI, 11.23%–16.62%), 0.39% (95% CI, 0.15%–0.71%), 3.99% (95% CI, 2.36%–5.95%), respectively. Programmed cell death protein 1 (PD‐1) inhibitors monotherapy and ICIs combination therapy had significantly higher incidences of any grade trAEs (odds ratio [OR], 1.25, 95% CI, 1.05–1.49 and 1.36, 95% CI, 1.15–1.60, respectively), grade three or more trAEs (OR, 1.41, 95% CI, 1.08–1.84 and 1.79, 95% CI, 1.39–2.30, respectively), trAEs leading to discontinuation of treatment (OR, 3.98, 95% CI, 2.06–7.70 and 10.14, 95% CI, 5.49–18.70, respectively) compared with programmed death‐ligand 1 (PD‐L1) inhibitors monotherapy. ICIs combination therapy had a significantly higher incidence of grade three or more trAEs compared with PD‐1 inhibitors monotherapy (OR, 1.27, 95% CI, 1.03–1.55); however, the incidences of any grade trAEs and trAEs leading to discontinuation of treatment were not significant different. Conclusions Our study suggests that the incidences of grade three or more trAEs, treatment‐related deaths, and trAEs leading to discontinuation of treatment are low in R/M HNSCC patients treated with ICIs. PD‐L1 inhibitors monotherapy may be safer compared with PD‐1 inhibitors monotherapy and ICIs combination therapy.

Objectives The reconstruction of bone defects in tibial revision knee arthroplasty is challenging. In this study, we evaluated the primary stability of a novel three‐dimensional (3D)‐printed custom‐made metaphyseal cone for Anderson Orthopedic Research Institute (AORI) IIb or III bone defect reconstruction in tibial revision knee arthroplasty using the combination of finite‐element analysis and biomechanical experiments. Methods In the finite‐element analysis, AORI II b and III medial tibial bone defects were designed at varying depths. A novel 3D‐printed custom‐made metaphyseal cone was designed and used to reconstruct the bone defect with or without a stem in simulated revision total knee arthroplasty (RTKA). A no‐stem group and a stem group were established (based on whether a stem was used or not). Von Mises stress and micromotion were calculated with varying depths of bone defects, ranging from 5 mm to 35 mm, and then micromotions at the bone–implant interface were calculated and compared with the critical value of 150 μm. In the biomechanical experiment, the no‐stem group was used, and the same bone defects were made in four synthetic tibias using patient‐specific instruments. Abstract Background Immune checkpoint inhibitors (ICIs) showed antitumor activity for recurrent or metastatic head and neck squamous cell carcinoma (R/M HNSCC). However, the results from different studies were controversial. Methods Online databases were searched for randomized clinical trials (RCTs) evaluating ICIs for R/M HNSCC. The characteristics of the studies and the results of overall survival (OS), progression‐free survival (PFS), objective response rate (ORR), treatment‐related adverse events (TRAEs) were extracted. Results A total of 4936 patients from eight studies were included. Anti‐PD1/PDL1 monotherapy significantly improved OS in total population (hazard ratio, HR, 0.87, 95% CI, 0.79–0.95, p = 0.003) and PD‐L1 high expression patients (HR, 0.71, 95% CI, 0.55–0.90, p = 0.006) with significant lower incidence of any grade TRAEs (odds ratio, OR, 0.16, 95% CI, 0.07–0.37, p

Abstract Marine heatwaves (MHWs) are extremely warm ocean temperature events that significantly affect marine environments, but their effects on the coastal carbonate system are still uncertain. In this study, we systematically quantify MHWs' impacts on air‐sea carbon dioxide (CO2) flux anomalies (FCO2′) in the Mid‐Atlantic Bight (MAB) and South Atlantic Bight (SAB) from 1992 to 2020. During the longest MHW in both regions, oceanic CO2 uptake capabilities substantially decreased, primarily due to significant increases in the seawater partial pressure of CO2 (pCO2sea). For all cases, MHWs played a more significant role in driving pCO2sea changes in the MAB than the SAB, where non‐thermal drivers dominated pCO2sea variability. In the MAB, weakened wind speeds related to wintertime atmospheric perturbations increase ocean temperatures and pCO2sea, further reducing CO2 uptake during winter MHWs. This work is the first to connect extreme temperatures to coastal air‐sea CO2 fluxes. The reduction in CO2 absorption noted during MHWs in this study has important implications for coastal regions to act as continued sinks for excess CO2 emissions in the atmosphere.

Abstract Kafirins are the primary storage proteins in sorghum kernels that provide amino acids for seed germination. The highly proteolytic resistant γ‐ and β‐kafirins form the cross‐linked outer layers that encapsulate α‐kafirins to generate protein bodies, resulting in poor digestibility of sorghum grains. The sorghum kafirins thus contribute to the poor quality of the kernels. The nutritional quality and digestibility of sorghum grains can be improved by reducing the contents of kafirin. γ‐Kafirin is encoded by the K2G gene, which is located on sorghum chromosome 2. In this study, we used CRISPR/Cas9 gene editing to target the K2G gene to create new sorghum lines with reduced levels of kafirin. A guide RNA (sgRNA) was designed to introduce mutations in the CDS region that encodes the endoplasmic reticulum signal peptide of γ‐kafirin. The pK2GsgRNA/Cas9 vector was transformed into sorghum using the pollen‐mediated transformation method. Sequencing of the transformants showed that three out of 24 transgenic plants contain genetic mutations in the targeted region. Compared with the wildtype, the γ‐kafirin contents of the mutant plants decreased by 12.75%–19.22%, and the protein digestibility of the mutant kernels increased by 26.91%–74.31% in raw flour. Although the grain weights remained comparable to those of the wildtype, the growth of the mutant plants was more vigorous as the mutant shoots grew taller and thicker compared with those of the wildtype. Our work advances the ability to improve the digestibility of an important crop. The resulting quality improvements can also be rapidly deployed for breeding and generation of transgene‐free, improved cultivars of sorghum, a major crop worldwide.

Abstract Background In recent years, light therapy has been tried for the treatment of depression and sleep in pregnancy or postnatal period women, but the results have been inconclusive. This meta‐analysis is the first to systematically review the effects of light therapy on depression and sleep disturbances in women during pregnancy and the postnatal period. Methods We searched for randomized controlled studies in PubMed, Embase, Cochrane Library, Web of Science, Chinese National Knowledge Infrastructure, and Chinese Biomedical Database up to January 2023. The standardized mean difference (SMD) was used to assess the efficacy of the outcome indicators. Results Eight studies were eventually included in the analysis. The results showed that light therapy was more effective than the placebo group in terms of depression (SMD = .34, CI = .08–.61) and sleep (SMD = .64,95%CI = .28–1.00). Subgroup analysis could not explain the significant heterogeneity. There were no serious adverse effects in either the light therapy or placebo groups. Conclusions Light therapy could be considered an effective treatment for depression and sleep disturbances in women during pregnancy and the postnatal period. However, future high‐quality trials with larger sample sizes are still needed.

Abstract We aimed to identify a novel prognostic biomarker for head and neck squamous cell carcinoma (HNSCC) based on tumor immunology‐related alternative splicing (AS). Data for 502 HNSCC and 44 normal samples were obtained from the TCGA database and used to establish an AS‐related risk model through univariate, least absolute shrinkage, and selection operator Cox regression analyses. Fresh HNSCC and normal oral tissues were surgically obtained from 44 HNSCC patients. Western blotting and quantitative reverse transcription‐PCR were used to assess gene expression levels. Kaplan–Meier was performed to evaluate patients' overall survival (OS) rate. The CIBERSORT algorithm, single‐sample gene set enrichment analysis, and immune checkpoint analyses were performed to compare immune activities between subgroups. Abstract The sCO2 Brayton cycle has gained interest because of its flexibility and ability to provide higher thermomechanical conversion efficiency. Studies on failure conditions and corresponding emergency measures are of great significance to ensure the safety of the system, while there are limited studies related to the sCO2 Brayton cycle because the test circuit has not been applied in practice at present. In this study, we have developed a dynamic model of the CO2 Brayton cycle, and carefully validated its components against experimental data. Preliminary safety assessment and dynamic response under loss of cooling water (LOCW), loss of heat source (LOHS), and pipeline leakage have been analyzed. Emergency strategies are proposed to maintain the safe operation under failure conditions. The primary objective of this study is to reveal the dynamic performance and emergency measures under failure conditions of the sCO2 Brayton cycle. The results demonstrate that the system net power decreases at rates close to 10%/s and less than 0.1%/s under LOCW and LOHS, respectively. The system responds more dramatically under LOCW compared with LOHS. The system needs emergency measures under LOCW, while it has sufficient time to realize a normal slow shutdown under LOHS. The leakage rate is at least five times higher in the high‐pressure pipeline compared with the low‐pressure pipeline for the same area of the leakage hole. Under pipeline leakage, the heat source should be cut‐off slowly, and the rotation speeds of the main compressor and recompressor should be reduced simultaneously to protect the system.

Abstract Job shop scheduling problem is a classical scheduling problem, and it is very difficult to work out. To solve it well, the meta‐heuristic algorithm is a good choice, and the evaluation method of neighbourhood solutions, which affects the efficiency of the algorithm and the quality of the solution, is one of the keys in the algorithm. We propose an approximate evaluation method by exploring domain knowledge in neighbourhood solutions. Firstly, we reduce the computational time of the evaluation by analysing the unnecessary computational operations. Secondly, according to the domain knowledge, we prove that the evaluated value of the neighbourhood solution is the exact value under certain conditions. At the same time, a set of critical parameters are calculated to correct the estimated value of the neighbourhood solutions that do not meet the conditions to improve the evaluation accuracy. With all of these, an approximate evaluation method for neighbourhood solutions in job shop scheduling problems is proposed. The experiments on different numerical instances show the superiority of the method proposed.

Abstract Rosmarinus officinalis (rosemary) is widely used as a food ingredient. Rosemary extract (containing 40% carnosic acid) exhibited potent antiobesity activity. However, the relationship between carnosic acid (CA) and changes in the gut microbiota of high‐fat diet (HFD)‐induced obese mice has not been fully investigated. C57BL/6 mice were fed a normal diet, an HFD, or an HFD containing 0.1% or 0.2% CA for 10 weeks. CA exhibited promising antiobesity effects and caused marked alterations in the gut microbiota of HFD‐induced obese mice. CA caused the prevalence of probiotics and functional bacteria, including Akkermansia muciniphila, Muribaculaceae unclassified, and Clostridium innocuum group, and inhibited diabetes‐sensitive bacteria, including Proteobacteria and Firmicutes. The ratio of Firmicutes to Bacteroidetes was regulated by CA in a dose‐dependent manner, decreasing it from 13.22% to 2.42%. Additionally, CA reduced bile acid‐metabolizing bacteria, such as Bilophila, Clostridium, Lactobacillus, and Leuconostoc. The results of the linear discriminant analysis and effect size analysis indicated that CA attenuated the microbial changes caused by HFD. The high CA (HCA) group (HFD containing 0.2% CA) exhibited a greater abundance of Verrucomicrobiae (including Akkermansia muciniphila, genus Akkermansia, family Akkermansiaceae, and order Verrucomicrobiales), Eubacterium, and Erysipelatoclostridium, and the low CA (LCA) group (HFD containing 0.1% CA) exhibited a greater abundance of Eisenbergiella, Intestinimonas, and Ruminococcaceae. Our results demonstrate that the antiobesity effects of CA might be strongly related to its prebiotic effects.

Abstract Background Lupus nephritis (LN) is among the most common complication of systemic lupus erythematosus (SLE) with high mortality and morbidity. The analysis of LN kidney's local immune response through single‐cell and spatial transcriptome enables the study of potential therapeutic targets. Methods By single cell sequencing and spatial transcriptome, we profile cells from LN kidney and normal kidney tissues to characterize cellular composition and elucidate the potential upstream monocyte/macrophage (Mono/MΦ) initiating the auto‐immune response. After the high‐throughput synergy screening, we performed the immunofluorescence to identify the specific cells in LN patients. The function experiments were finished by flow cytometry and Elisa. Results By immunofluorescence and spatial transcriptome, we identified differential subsets of Mono/MΦ and demonstrated that they exhibit temporal expression of TIMP1, IL1B, SPP1 and APOE. With the function experiments, we found that the APOE+ Mono may be compensatorily increased in LN, and the capacity of antigen presenting was decreased with the overexpression of APOE. Furthermore, how do the LN‐specific Mono/MΦ transport in and out the glomerulus to active the local immune response remains unclear. Our results showed that lymphangiogenesis occurred in LN kidneys but not in normal kidneys, suggesting the presence of a new lymphatic vessel may serve as a ‘green channel’ for LN‐specific Mono/MΦ. Conclusions In LN, APOE+ Mono are compensatorily elevated, with decreased antigen presenting ability and reduced secretion of interferons. The lymphangiogenesis in LN prompts the trafficking of Mono/MΦ in LN kidney.

Abstract Aims This study explores the current occupational well‐being status of male nurses in Chengdu, China, and identifies the concomitant protective and risk factors. Design This study has a cross‐sectional survey design. Abstract This study presents a 4.6–12.8 GHz injection‐locked frequency divider (ILFD) based on modified varactor‐less Colpitts oscillator topology. A modified Colpitts ILFD is proposed to improve the LR and reduces power consumption, simultaneously. Meanwhile, the forward body bias technique is utilized to further decrease power consumption. Based on the 55 nm CMOS technology, a modified varactor‐less Colpitts ILFD with a free‐running frequency of 4.05 GHz is implemented. The proposed modified varactor‐less Colpitts ILFD exhibits 94.3% LR from 4.6 to 12.8 GHz with an injection power of 0 dBm. It consumes 2.7 mW power from 0.6 V supply voltage and the core area is 0.38 mm2.

Abstract Ion exchange membranes (IEMs) play important roles in energy generation and storage field, such as fuel cell, flow battery, however, a major barrier in the way of large‐scale application is the high cost of membranes (e.g., Nafion membranes price generally exceeds USD$ 200 m−2). The membrane‐less technology is one of the promising approaches to solve the problem and thus has attracted much attention and been explored in a variety of research paths. This review introduces one of the representative membrane‐less battery types, Biphasic membrane‐less redox batteries that eliminate the IEMs according to the principle of solvent immiscibility and realizes the phase splitting in a thermodynamically stable state. It is systematically classified and summarizes their performances as well as the problems they are suffering from, and then several effective solutions are proposed based on the modification of electrodes and electrolytes. Finally, special attention is given to the challenges and prospects of Biphasic membrane‐less redox batteries, which could contribute to the development of membrane‐less batteries.

Background High expression of monoamine oxidase A (MAOA) in non‐small cell lung cancer (NSCLC) is related to epithelial‐mesenchymal transition (EMT) and the development of clinicopathological features of NSCLC. Nevertheless, the role of MAOA in drug resistance still remains unclear. Hence, the aim of this article was to evaluate a previously synthesized MAOA inhibitor (G11) on inhibiting paclitaxel‐resistant NSCLC metastasis and growth. Methods First, MAOA expression level was evaluated in several NSCLC cell lines. An MTT assay was used to validate the inhibitory effect of G11 on NSCLC cells in vitro. Second, gene expression in G11‐treated H460/PTX cells was analyzed by microarray gene expression. Third, transwell assay was performed to assess the invasion and metastasis of G11‐treated A549/PTX and H460/PTX cells and western blot assay used to analyze vital protein expression level in G11‐treated H460/PTX cells. Finally, the antimetastatic effect of G11 was tested in an NSCLC in vivo model. Results Our data revealed that G11 significantly inhibited the viability of paclitaxel (PTX)‐resistant NSCLC cell lines (A549/PTX and H460/PTX). G11 dramatically reduced the expression of MAOA in A549/PTX and H460/PTX cells, which exhibited relatively high MAOA expression levels. Additionally, G11 was found to hinder A549/PTX and H460/PTX cell migration and invasion. Furthermore, the in vivo study indicated that the coadministration of G11 and paclitaxel significantly suppressed tumor metastasis in H460/PTX lung metastasis models. Conclusions These findings indicated G11 showed a moderate inhibitory effect on paclitaxel‐resistant NSCLC metastasis and growth, and support further investigation on MAOA potentially as a promising therapeutic target for paclitaxel‐resistant NSCLC treatment. Key points Significant findings of the study Inhibition of MAOA might contribute to the suppression of metastasis and growth in PTX‐resistant NSCLC cells. What this study adds This study explored the potential function of MAOA in drug‐resistant NSCLC and might consider MAOA as a promising target for the treatment of drug‐resistant NSCLC.

Abstract Tachycardia and atrial fibrillation, early symptoms of hyperthyroidism indicate significant hemodynamic variation in cardiovascular system, if left untreated and further deterioration in hemodynamics can result in chronic heart failure and liver dysfunction even a fatal event. We describe a female patient of Graves' hyperthyroidism to present the continuum of the pathophysiology development of the disease, to highlight the hemodynamic variation is a dominant contributing factor of Graves' hyperthyroidism complication, we wish to emphasize cardiac manifestations in the setting of thyrotoxicosis should be treated promptly and aggressively.

The selection of the optimal metal milling parameters greatly impacts final product quality and production efficiency in modern manufacturing systems. The profit rate is also sensitive to the selected parameters. This research focuses on determining the optimal parameters of a multipass milling process using an improved particle swarm optimization (PSO) method. The objective is to minimize the production time. The proper number of passes, the optimal cut speed, and feed rate are considered as the parameters (the decision variables in the model) which are needed to be optimized. Furthermore, the permissive arbor strength, arbor deflection, and motor power are the constraints of the model. The penalty function method is used as the constraints handling technique to address the constraints efficiently in the proposed method. A case is adopted and solved to evaluate the performance of the proposed method. The experimental part is analyzed and compared with advanced methods. Experimental results show that the proposed method is very effective for parameters optimization of a multipass milling process and outperforms other methods.

Abstract Background Accumulating works show that lncRNAs play critical roles in the development of gastric cancer (GC). LncRNA HLA complex group 18 (HCG18) was implicated in the progression of bladder cancer and glioma, but its role in GC is unknown. Methods RT‐PCR was used to detect HCG18 and miR‐141‐3p expression in GC specimen. GC cell lines (AGS and MKN‐28) were exploited as cell model. The biological effect of HCG18 on cancer cells was probed by CCK‐8, colony formation, flow cytometry, Transwell and wound‐healing experiments in vitro, and subcutaneous xenotransplanted tumor model and tail vein injection model in vivo. Interaction between HCG18 and miR‐141‐3p was determined by bioinformatics analysis, RT‐PCR, and luciferase reporter experiments. Downstream gene expression of miR‐141‐3p, including Wiskott–Aldrich syndrome protein interacting protein family member 1 (WIPF1), Yes associated protein 1 (YAP), and tafazzin (TAZ) were detected using Western blot. Results HCG18 was markedly up‐regulated in GC specimens, while miR‐141‐3p was markedly down‐regulated. Down‐regulation of HCG18 inhibited viability, migration, and invasion of GC cells, while miR‐141‐3p transfection led to opposite effect. HCG18 could down‐regulate miR‐141‐3p through adsorbing it, and a negative association between HCG18 and miR‐141‐3p was found in GC specimens. HCG18 promoted WIPF1, YAP and TAZ expression, nonetheless, such influence was reversed by co‐transfecting with miR‐141‐3p. Conclusion HCG18 was aberrantly up‐regulated in GC tissues, and it indirectly regulated the activity of Hippo signaling through counteracting miR‐141‐3p expression.

Abstract Background Breast cancer is a heterogeneous malignant disease, which has variation in clinical behaviors. High‐throughput technologies have added important genetic alternative and biological change information for breast cancer. CARNS1 is an important ATPases. It can catalyze the synthesis of carnosine, which has antiproliferative activity in cancer. Here, we hypothesize that CARNS1 plays an essential role in the development of breast cancer. Methods The expressions of CARNS1 in breast cancer were data‐mined and analyzed from TCGA (the Cancer Genome Atlas) and GEO (the Gene Expression Omnibus) databases. The correlation of CARNS1 expression with clinical characteristics and the diagnostic capability of CARNS1 were assessed. Experimental studies were conducted in two cohorts (n = 60) of breast cancer patients by qRT‐PCR and immunohistochemical analysis. Results CARNS1 was significantly downregulated in breast cancer. The expression was correlated with tumor molecular and histological types, T and M stages, and vital status. Kaplan–Meier survival analysis showed that the downregulation of CARNS1 was significantly related to poor overall survival and relapse‐free survival. Moreover, these scenarios have been extended to ER, PR, and HER2 positive patients. Univariate and multivariate analysis showed that CARNS1 can be considered as an independent prognostic predictor for patients with breast cancer. Experimental data supported that the protein and mRNA levels of CARNS1 in breast cancer are indeed significantly downregulated. Conclusion Our findings have demonstrated that CARNS1 acts as a tumor suppressor gene and may be an independent prognostic indicator for breast cancer patients.

Abstract Objective The IL‐33/ST2 axis has been extensively investigated in type 2 eosinophilic inflammation. Here, we aimed to investigate the role of the IL‐33/ST2 axis in neutrophil‐dominant allergic airway inflammation. Methods House‐dust mite (HDM) extract and lipopolysaccharide (LPS) were administered to establish a murine model of neutrophil‐dominant allergic airway inflammation. The formation of neutrophilic extracellular traps (NETs) in the lung tissues was demonstrated by immunofluorescence imaging. Mature IL‐33 in bronchoalveolar lavage fluid (BALF) was detected by Western blotting. The neutrophilic chemokine KC produced by bone marrow‐derived macrophages (BMDMs) or primary alveolar epithelial cells was measured with a commercial ELISA kit. Results In the present study, we observed neutrophilic inflammation and tight junction damage in the lungs of mice sensitised with HDM and LPS. Furthermore, sensitisation with HDM and LPS resulted in the formation of NETs, accompanied by increased levels of mature IL‐33 in the BALF. Moreover, LPS damaged the epithelial tight junction protein occludin directly or indirectly by inducing NET formation. Surprisingly, IL‐33 deficiency augmented neutrophilia and epithelial barrier injury in the lungs of mice after sensitisation with HDM and LPS. Similarly, the absence of ST2 exacerbated the neutrophilic inflammatory response, decreased the expression of occludin and exacerbated the severity of neutrophil‐dominant allergic airway inflammation in an HDM/LPS‐induced mouse model. Mechanistically, BMDMs and alveolar epithelial cells from IL‐33‐ or ST2‐deficient mice tended to produce higher levels of the neutrophilic chemokine KC. Conclusions These results demonstrated that the IL‐33/ST2 axis may play a protective role in neutrophil‐dominant allergic airway inflammation.

In accordance with “the response to injury” theory, the entry of monocytes into the intima guided by inflammation signals, taking up cholesterol and transforming into foam cells, and egress from plaques determines the progression of atherosclerosis. Multiple cytokines and receptors have been reported to be involved in monocyte recruitment such as CCL2/CCR2, CCL5/CCR5, and CX3CL1/CX3CR1, and the egress of macrophages from the plaque like CCR7/CCL19/CCL21. Interestingly, some neural guidance molecules such as Netrin-1 and Semaphorin 3E have been demonstrated to show an inhibitory effect on monocyte migration. During the processes of monocytes recruitment and migration, factors affecting the biomechanical properties (e.g., the membrane fluidity, the deformability, and stiffness) of the monocytes, like cholesterol, amyloid-β peptide (Aβ), and lipopolysaccharides (LPS), as well as the biomechanical environment that the monocytes are exposed, like the extracellular matrix stiffness, mechanical stretch, blood flow, and hypertension, were discussed in the latter section. Till now, several small interfering RNAs (siRNAs), monoclonal antibodies, and antagonists for CCR2 have been designed and shown promising efficiency on atherosclerosis therapy. Seeking more possible biochemical factors that are chemotactic or can affect the biomechanical properties of monocytes, and uncovering the underlying mechanism, will be helpful in future studies.

Carbonate reservoirs are the main reservoir types in China, which occupy the large ratio of reserves and production at present. The high-efficiency development of carbonate reservoirs is of great significance to assure the stability of national energy supply. The Lower Paleozoic reservoir in Jingbian gas field and the Sinian reservoir in Anyue gas field are two typical carbonate gas reservoirs, and their successful development experiences can provide significant references for other similar carbonate gas reservoirs. For Jingbian gas field, it is a lithological-stratigraphic reservoir developed in a westward monocline and multiple rows of nose-fold structures, and is a stable craton basin with simple palaeognomy distribution and stable connectivity, which has complex gas-water distribution. However, for Anyue gas field, it is a lithological-structural reservoir with multiple tectonic high points and multiple fault systems, and is biological dune beach facies under extensional setting with highly differentiated inside of the block in palaeognomy characteristics, which has limited connectivity and tectonic side water is in a local area. The difference of gas reservoir characteristics leads to the diverse development strategies. For these two gas reservoirs, although there are some similar aspects, such as the screen of enrichment areas, the application of irregular well pattern and reservoir stimulation techniques, the criteria of enrichment areas, the well types, and the means of reservoir stimulation are absolutely different. In addition, due to the differences of control reserves and production capacity for these two kinds of reservoirs, the mode of stable production is also different. The effective development of Jingbian gas field can give some references to the future exploitation on the Sinian gas reservoir. Firstly, the sedimentary characteristics should be studied comprehensively. Secondly, the distribution pattern and distribution characteristics of the palaeognomy should be found and determined. Thirdly, the distribution of fracture system in the reservoir should be depicted finely. Finally, dynamic monitoring on the production performance should be strengthened, and the management for this gas field should be improved further. The findings of this study can help for better understanding of the Karst weathering-crust carbonate gas reservoir formation characteristics and the optimal development technologies that should be taken in practice.

Thermal properties are important for featuring the water-heat transfer capacity of soil. They are also key to many processes in earth sciences, such as the land surface processes and ecological and geoenvironmental dynamics and their changes in permafrost regions. With loose and porous structures, the organic matter layer in soil strata substantially influences soil thermal conductivity. So far, thermal conductivity of mineral soils has been explored extensively and in depth, but there are only limited studies on that of organic soils. In this study, influences of soil temperature, soil moisture saturation (SMS), and soil organic matter (SOM) content on soil thermal conductivity were analyzed on the basis of laboratory experiments on the silt-organic soil mixtures of varied mixing ratios. Results show that soil thermal conductivity declines slowly with the lowering temperatures from 10 to 0°C; however, it increases and finally stabilizes when temperature further lowers from 0 to -10°C. It is important to note that thermal conductivity peaks in the temperature range of -2~0°C (silty and organic-poor soil) and -5~0°C (organic-rich soil), possibly due to phase changes of ice/water in warm permafrost. Under both thawed and frozen states, soil thermal conductivity is positively related with SMS. However, with rising SOM content, the growth rate of soil thermal conductivity with SMS slows gradually. Given the same SMS, soil thermal conductivity declines exponentially with increasing SOM content. Based on the experimental and theoretical analyses, a new empirical computational formula of soil thermal conductivity is established by taking into account of the SOM content, SMS, and soil temperature. The results may help better parameterize in simulating and predicting land surface processes and for optimizing frozen soil engineering designs and provide theoretical bases for exploring the dynamic mechanisms of environmental changes in cold regions under a changing climate.

The prediction of flow and heat transfer characteristics of liquid sodium with CFD technology is of significant importance for the design and safety analysis of sodium-cooled fast reactor. The accuracies and uncertainties of the CFD models should be evaluated to improve the confidence of the numerical results. In this work, the uncertainties from the turbulent model, boundary conditions, and physical properties for the flow and heat transfer of liquid sodium were evaluated against the experimental data. The results of uncertainty quantization show that the maximum uncertainties of the Nusselt number and friction coefficient occurred in the transition zone from the inlet to the fully developed region in the circular tube, while they occurred near the reattachment point in the backward-facing step. Furthermore, in backward-facing step flow, the maximum uncertainty of temperature migrated from the heating wall to the geometric center of the channel, while the maximum uncertainty of velocity occurred near the vortex zone. The results of sensitivity analysis illustrate that the Nusselt number was negatively correlated with the thermal conductivity and turbulent Prandtl number, while the friction coefficient was positively correlated with the density and Von Karman constant. This work can be a reference to evaluate the accuracy of the standard k-ε model in predicting the flow and heat transfer characteristics of liquid sodium.

Abstract Amyloid beta (Aβ) aggregation is generally believed as the crucial and primary cause of Alzheimer's disease (AD). However, current Aβ‐targeted therapeutic strategies show limited disease‐modifying efficacy due to the irreversible damages in the late stage of AD, thus the treatment should be given before the formation of deposition and target primary Aβ species rather than advanced plaques. Herein, inspired by heat shock protein, a self‐assembly nanochaperone based on mixed‐shell polymeric micelle (MSPM) is devised to act as a novel strategy for AD prevention. With unique surface hydrophobic domains, this nanochaperone can selectively capture Aβ peptides, effectively suppress Aβ aggregation, and remarkably reduce Aβ‐mediated cytotoxicity. Moreover, the formed nanochaperone‐Aβ complex after Aβ adsorption can be easily phagocytosed by microglia and thereby facilitates Aβ clearance. As a result, the nanochaperone reduces Aβ burden, attenuates Aβ‐induced inflammation, and eventually rescues the cognitive deficits of APP/PS1 transgenic AD mice. These results indicate that this biomimetic nanochaperone can successfully prevent the onset of AD symptoms and serve as a promising candidate for prophylactic treatment of AD.
