Abstract Walking is fundamental to normal human life. However, many people suffer from walking impairments due to various diseases that may severely affect their daily activities. Early detection of an abnormal gait can aid subsequent treatment and rehabilitation. This paper proposes a novel abnormal gait recognition method based on a perceptual loss convolutional temporal autoencoder (PLCTAE) network. It comprises upstream and downstream tasks, both of which utilise radar micro‐Doppler spectrograms as inputs. The upstream task employs a convolutional autoencoder with the perceptual loss to encode and decode micro‐Doppler spectrograms, achieving unsupervised pretraining and obtaining the initial parameters for the convolutional part of the PLCTAE. The downstream task fine‐tunes the convolutional part of the PLCTAE through supervised training to extract spatial features from the micro‐Doppler spectrograms and incorporates a bidirectional long short‐term memory (BiLSTM) network to further extract temporal features, accomplishing the task of abnormal gait classification. The experimental results demonstrate that the proposed method achieves good classification performance on the self‐established dataset which is collected by Texas Instruments' IWR6843ISK millimetre‐wave radar and contains eight types of abnormal gaits. The generalisation performance is also validated on a public dataset from the University of Glasgow containing six types of human activities.

Abstract A strategy was proposed to predict accurately the dielectric permittivity and dielectric strength based on the Onsager local field theory. The molecular dynamic simulation was utilised to analyse the dipole moment fluctuation in polyimide (PI) to reflect the polarisation response of applied electric field. The simulation results revealed that the optical dielectric permittivity and static dielectric permittivity were 2.62 and 3.34, showing that the electronic displacement polarisation of the PI acted as a major contributor. The deviation between simulated and measured values of the frequency‐dependent relative dielectric permittivity was no more than 5%, which exhibited high accuracy. As the polarisation response of the polar groups in the PI was at infrared frequencies, the conductive loss may be the dominant role in 102–106 Hz at room temperature. Moreover, the effect of Joule heat on the structure, dielectric permittivity and Young modulus was considered to accurately predict the dielectric strength of the PI (359 kV/mm), which was in agreement with experimental values in the literature. These results establish a clear correlation between structural characteristics and dielectric properties of the PI, which would be the theoretical insights into the design and synthesis of the PI with tailored dielectric properties.

Abstract To enhance the accuracy of fine‐grained image classification and address challenges such as excessive interference factors within the dataset, inadequate extraction of local key features, and insufficient channel semantic association, a dual‐branch information interaction model that integrates convolutional neural networks (CNN) with Vision Transformers is proposed. This model leverages the Vision Transformer branch to extract global features, which are subsequently combined with the CNN branch to further augment the model's capability for local information extraction. In order to enhance the ability of the CNN branch to extract global information and reduce the loss of feature information, a feature enhancement module is added to the CNN branch. Since the Vision Transformer branch directly convolves with the convolution kernel will result in the inability to learn the underlying features of the image, a shallow feature extraction module is proposed, and the CNN and Vision Transformer branches interact with the information of the dual branches through the down‐sampling Down module and the up‐sampling UP module. The accuracy of the improved method on CUB‐200‐2011, Stanford Cars and FGVC‐Aircraft fine‐grained image classification datasets are 95.2%, 97.1% and 96.9%, respectively. The experimental results show that the method has good generalization on different datasets.

Abstract Blockchain trilemma is a considerable obstacle for today's decentralized systems. It is hard to achieve a perfect balance among decentralization, security, and scalability. Many popular blockchain platforms sacrifice scalability to preserve decentralization and security, resulting in low speed, reduced throughput, and poor real‐time performance (the time from transaction initiation to confirmation). Currently, there are several technologies, such as sharding, directed acyclic graph technology, sidechains, off‐chain state channels etc., that aim to improve throughput and real‐time performance. However, most of these solutions compromise the core feature of blockchain, which is decentralization, and introduce new security risks. In this paper, the authors propose a novel method, called MEchain, based on the Proof of Time Series Algorithm. MEchain consists of two models: the multi‐chain model and the elastic‐chain model. The authors’ experimental results show that these two models can enhance real‐time performance and throughput to a higher level in the industry.

Abstract Second‐life batteries (SLBs), which are batteries retired from electric vehicles (EVs), can be used as energy storage systems to enhance the performance of distribution networks. Two issues should be addressed particularly for the optimal sizing of SLBs. Compared with fresh batteries, the failure rate of SLBs is relatively high, and timely and preventive replacement is needed. In addition, the flexibility introduced by EVs and installed SLBs should be coordinated to achieve optimal economic benefits. This paper focuses on the efficient utilization of SLBs by highlighting reliability‐flexibility concerns in optimal sizing. The model is formulated as a bi‐level model. On the upper‐level, considering the operational reliability constraints of SLBs, decisions regarding the investment and replacement of SLBs are optimized. Distribution network operations are improved on the lowerlevel, with an effective spatiotemporal flexible dispatch strategy for EVs. Finally, a linearized process for the optimal sizing of SLBs is presented and efficiently implemented. The Sioux Falls network and IEEE 69‐node distribution network are coupled as the test system. According to the simulation results, when the state of health of the SLBs decreased to 70%, the conditions were unreliable. The differences in the optimal SLB size and costs considering reliability and flexibility are highlighted.

Abstract Glaucoma poses a significant threat to vision, capable of causing irreversible damage and, in severe instances, leading to permanent blindness. Accurate optic cup (OC) and optic disc (OD) segmentation are essential in glaucoma screening. In this study, a novel OC and OD segmentation approach is proposed. Based on U‐Net, it is optimized by introducing cardinality dimensions. Moreover, attention gates are implemented to reinforce salient features while suppressing irrelevant information. Additionally, a convolutional block attention module (CBAM) is integrated into the decoder segment. This fusion hones in on effective information in both channel and spatial dimensions. Meanwhile, an image processing procedure is proposed for image normalization and enhancement. All of these increase the accuracy of the model. This model is evaluated on the ORIGA and REFUGE datasets, demonstrating the model's superiority in OC and OD segmentation compared to the state‐of‐the‐art methods. Additionally, after the proposed image processing, cup‐to‐disc ratio (CDR) prediction on a batch of 155 in‐house fundus images yields an absolute CDR error of 0.099, which is reduced by 0.04 compared to the case where only conventional processing was performed.

Abstract Drug‐drug interaction (DDI) prediction is a crucial issue in molecular biology. Traditional methods of observing drug‐drug interactions through medical experiments require significant resources and labour. The authors present a Medical Knowledge Graph Question Answering (MedKGQA) model, dubbed MedKGQA, that predicts DDI by employing machine reading comprehension (MRC) from closed‐domain literature and constructing a knowledge graph of “drug‐protein” triplets from open‐domain documents. The model vectorises the drug‐protein target attributes in the graph using entity embeddings and establishes directed connections between drug and protein entities based on the metabolic interaction pathways of protein targets in the human body. This aligns multiple external knowledge and applies it to learn the graph neural network. Without bells and whistles, the proposed model achieved a 4.5% improvement in terms of DDI prediction accuracy compared to previous state‐of‐the‐art models on the QAngaroo MedHop dataset. Experimental results demonstrate the efficiency and effectiveness of the model and verify the feasibility of integrating external knowledge in MRC tasks.

Abstract Background The role of Claudin‐1 in tongue squamous cell carcinoma (TSCC) metastasis needs further clarification, particularly its impact on cell migration. Herein, our study aims to investigate the role of Claudin‐1 in TSCC cell migration and its underlying mechanisms. Methods 36 TSCC tissue samples underwent immunohistochemical staining for Claudin‐1. Western blotting and immunofluorescence analyses were conducted to evaluate Claudin‐1 expression and distribution in TSCC cells. Claudin‐1 knockdown cell lines were established using short hairpin RNA transfection. Migration effects were assessed through wound healing assays. Furthermore, the expression of EMT‐associated molecules was measured via western blotting. Results Claudin‐1 expression decreased as TSCC malignancy increased. Adenosine monophosphate–activated protein kinase (AMPK) activation led to increased Claudin‐1 expression and membrane translocation, inhibiting TSCC cell migration and epithelial–mesenchymal transition (EMT). Conversely, Claudin‐1 knockdown reversed these inhibitory effects on migration and EMT caused by AMPK activation. Conclusions Our results indicated that AMPK activation suppresses TSCC cell migration by targeting Claudin‐1 and EMT pathways.

Abstract N6‐acetyl‐L‐lysine residue is abundant in dietary protein but little is known about its potential influences on the diet‐consumers. Herein, it is reported that Lysyl‐tRNA synthetase (KARS) mediates co‐translational deposition of diet‐derived N6‐acetyl‐L‐lysine (AcK) in nascent proteins to contribute to the acetylome in cells. Acetylated dietary protein is a direct source of AcK that can widely and substantially regulate the acetylome in multiple organs of mice. By analyzing the mechanisms underlying AcK contributing to the acetylome in mammalian cells, it is found that KARS can utilize AcK as an alternative substrate to produce N6‐acetyl‐l‐lysyl‐tRNA. The crystal structure of KARS in complex with AcK at 2.26 Å resolution shows that AcK shares the same substrate‐binding pocket as L‐lysine, allowed by a sidechain flip of Tyr499. The generated N6‐acetyl‐L‐lysyl‐tRNA introduces AcK into growing nascent polypeptide and results in protein acetylation, including the regions buried inside folded proteins that are post‐translational modification (PTM)‐inaccessible and functionally important. This undocumented protein modification mechanism is inherently different from PTM and termed as co‐translational modification (coTM). It is expected to extend the repertoire of acetylome and improve the understanding of protein modification mechanisms in cells.

ABSTRACT Background The 2021 WHO Classification of Central Nervous System Tumors introduces more molecular markers for glioma reclassification, including TERT promoter (TERTp) mutation as a key feature in glioblastoma diagnosis. Aims Given the changes in the entities included in each subtype under the new classification, this research investigated the distribution, prognostic value, and correlations with other molecular alterations of TERTp mutation in different subgroups under this latest classification. Methods All glioma patients admitted to Peking Union Medical College Hospital for surgical resection or biopsy from 2011 to 2022 were included. Samples were analyzed for TERTp mutation and 59 other gene alterations and chromosome copy number variations. Results A total of 207 patients were included. The occurrence of TERTp mutations varied with percentages of 4.55%, 100%, and 77.92% in astrocytoma, oligodendroglioma, and glioblastoma, respectively. 65% of all adult‐type glioma patients and 42.6% of IDH‐wildtype histology grade 2 or 3 patients were TERTp‐mutant. Survival analysis showed that TERTp mutation was a predictor of better prognosis in IDH‐mutant grade 2 gliomas (median OS (mOS): not reached (NA) (95% CI: NA–NA) vs. 75.9 (95% CI: 55.4–NA) months, HR = 0.077 (95% CI: 0.01–0.64), p = 0.003), while poor OS was associated with all Grade 4 gliomas (mOS: 17.5 (95% CI: 12.6–24.2) vs. 40.5 (95% CI: 24.4–83.8) months, HR = 2.014 (95% CI: 1.17–3.47), p = 0.01) and all IDH‐wildtype histology grade 2 or 3 gliomas (median OS: 12.6 (95% CI: 11–24.2) vs. 83.8 (95% CI: 35.2–NA) months, HR = 3.768 (95% CI: 1.83–7.78), p

Abstract The morphology of plated lithium (MPL) metal on graphite anodes, traditionally described as “moss‐like” and “dendrite‐like”, exert a substantial negative influence on the performance of lithium‐ion batteries (LIBs) by modulating the metal‐electrolyte interface and side reaction rates. However, a systematic and quantitative analysis of MPL is lacking, impeding effective evaluation and manipulation of this detrimental issue. In this study, we transition from a qualitative analysis to a quantitative one by conducting a detailed examination of the MPL. Our findings reveal that slender lithium dendrites reduces the lifespan and safety of LIB by increasing the side reaction rates and promoting the formation of dead lithium. To further evaluate the extent of the detrimental effect of MPL, we propose the specific surface area (SSA) as a critical metric, and develop an in situ method integrating expansion force and electrochemical impedance spectroscopy to estimate SSA. Finally, we introduce a pulse current protocol to manipulate hazardous MLP. Phase field model simulations and experiments demonstrate that this protocol significantly enhances the reversibility of plated lithium. This research offers a novel morphological perspective on lithium plating, providing a more detailed fundamental understanding that facilitates effective evaluation and manipulation of plated lithium, thereby enhancing the safety and extending the cycle life of LIBs.

Abstract Plants are equipped with multi‐layered immune systems that recognize pathogen‐derived elicitors to activate immunity. Verticillium dahliae is a soil‐borne fungus that infects a broad range of plants and causes devastating wilt disease. The mechanisms underlying immune recognition between plants and V. dahliae remain elusive. Here, a V. dahliae secretory protein, elicitor of plant defense gene (VdEPD1), acts as an elicitor that triggers defense responses in both Nicotiana benthamiana and cotton plants is identified. Targeted gene deletion of VdEPD1 enhances V. dahliae virulence in plants. Expression of VdEPD1 triggers the accumulation of reactive oxygen species (ROS) and the activation of cell death in cotton plants. Gossypium barbadense EPD1‐interacting receptor‐like cytoplasmic kinase (GbEIR5A) and GbEIR5D interact with VdEPD1. Silencing of GbEIR5A/D significantly impairs VdEPD1‐triggered cell death in cotton plants, indicating the contribution of GbEIR5A/D to VdEPD1‐activated effector‐triggered immunity (ETI). VdEPD1 stimulates the expression of GbEIR5A and GbEIR5D in cotton plants. Interestingly, cotton plants with silenced GbEIR5A/D genes exhibit compromised pathogen‐associated molecular patterns (PAMPs)‐triggered ROS accumulation, whereas overexpression of GbEIR5A or GbEIR5D enhances PAMP‐induced ROS. These findings indicate that recognition of VdEPD1 potentiates GbEIRs to enhance cotton PAMP‐triggered immunity (PTI), uncovering a cooperative interplay of PTI and ETI in cotton.

Abstract Biomimicry is the enduring pursuit in the field of bone implants, wherein bio‐materials with adjustable elastic modulus and porosity, the same as natural bone, offer a novel strategy for developing and applying new bone repair materials. Conventional biomaterials are often used to repair bone defects without complete consideration of structural and functional osseointegration, leading to interface repair failure. In this study, organic‐inorganic interpenetrating network technology was employed using varying amounts of nano‐hydroxyapatite (nHAP) and methacrylated gelatin (GelMA) and osteogenic growth peptide (OGP) to construct biomimetic bones with low, medium, and high nano‐hydroxyapatite content (GelMA‐c‐OGP/nHAP). As the concentration of nano‐hydroxyapatite increases, comprehensive evaluations of the biomimetic materials were conducted using osteogenic ability tests, Micro‐CT scans, nanoindentation tests, and mechanical tests. The developed biomimetic structural material exhibits well‐controlled mechanical properties. Compared to natural bone trabeculae, this biomimetic material not only maintains the organic and inorganic ratio of natural bone but also demonstrates exceptional mechanical load‐bearing capabilities. Additionaly，this scaffold exhibits good porosity and mechanical properties. It enhances cell adhesion, integrates perfectly with bone tissue, and demonstrates excellent osteogenic ability both in vitro and in vivo. This study lays the foundation for constructing biomimetic scaffolds with adjustable mechanical properties, presenting high prospects for applications in the field of tissue engineering.

Event cameras demonstrate substantial potential in handling challenging situations, such as motion blur and high dynamic range. Herein, event–visual–inertial state estimation and 3D dense mapping (EVI‐SAM) are introduced to tackle the problem of pose tracking and 3D dense reconstruction using the monocular event camera. A novel event‐based hybrid tracking framework is designed to estimate the pose, leveraging the robustness of feature matching and the precision of direct alignment. Specifically, an event‐based 2D–2D alignment is developed to construct the photometric constraint and tightly integrated with the event‐based reprojection constraint. The mapping module recovers the dense and colorful depth of the scene through the image‐guided event‐based mapping method. Subsequently, the appearance, texture, and surface mesh of the 3D scene can be reconstructed by fusing the dense depth map from multiple viewpoints using truncated signed distance function fusion. To the best of knowledge, this is the first nonlearning work to realize event‐based dense mapping. Numerical evaluations are performed on both publicly available datasets, which qualitatively and quantitatively demonstrate the superior performance of our method. EVI‐SAM effectively balances accuracy and robustness while maintaining computational efficiency, showcasing superior pose tracking and dense mapping performance in challenging scenarios.

Abstract Background Lumbar disc degeneration (LDD) is a ubiquitous finding in low back pain. Many different etiology factors may explain the LDD process, such as bone morphogenetic proteins (BMPs), DNA methylation, and gut microbiota. Until recently the mechanisms underlying the LDD process have been elusive. Methods BMP‐related genes were extracted from the GeneCards database. The LDD transcriptome dataset was obtained from the Gene Expression Omnibus. We used linear regression and meta‐analysis to screen and integrate the differentially expressed genes associated with BMPs in LDD. Genome‐wide association studies (GWASs) of LDD were from FinnGen and UKBB. The expression quantitative trait loci (eQTLs) and DNA methylation quantitative trait loci from the blood were identified via the summary data‐based Mendelian randomization (SMR) method, and the possible blood BMP genes and their regulatory elements associated with the risk of LDD were prioritized. Intestinal eQTLs and fecal microbial QTLs (mbQTLs) were integrated, and the potential interactions between BMP gene expression in host intestinal tissue and the gut microbiota were revealed through SMR and colocalization analysis. The GWAS catalog (GCST90246169) was used to validate SMR results. Results A meta‐analysis of five datasets revealed that 113 BMP genes were differentially expressed between LDD and control tissues. Seven genes were selected as candidate pathogenic genes of LDD via the three‐step SMR method: CREB1, BMP6, PTCH1, GLI1, MEG3, GALNS, and NF1. SMR analysis also revealed five possible gut genes: HFE, MET, MAPK3, NPC1, and GDF5. The correlation between the gut microbiota and BMP gene expression in intestinal tissues was verified by eQTL‐mbQTL colocalization. Conclusion This multi‐omics study revealed that the BMP genes associated with LDD are regulated by DNA methylation. There are genetic differences between gut gene expression and the gut microbiota. These findings provide evidence for new therapeutic targets in the future.

Abstract Astrocytic processes minutely regulate neuronal activity via tripartite synaptic structures. The precision‐tuning of the function of astrocytic processes is garnering increasing attention because of its significance in promoting brain repair following ischemic stroke. Microdomain calcium (Ca2+) transients in astrocytic processes are pivotal for the functional regulation of these processes. However, the understanding of the alterations and regulatory mechanism of microdomain Ca2+ transients during stroke remains limited. In the present study, a fast high‐resolution, miniaturized two‐photon microscopy is used to show that the levels of astrocytic microdomain Ca2+ transients are significantly reduced in the peri‐infarct area of awake ischemic stroke mice. This finding correlated with the behavioral deficits shown by these mice under freely‐moving conditions. Mitochondrial Ca2+ activity is an important factor driving the microdomain Ca2+ transients. DEAD Box 1 (DDX1) bound to circSCMH1 (a circular RNA involved in vascular post‐stroke repair) facilitates the formation of membrane‐associated RNA‐containing vesicles (MARVs) and enhances the activity of astrocytic microdomain Ca2+ transients, thereby promoting behavioral recovery. These results show that targeting astrocytic microdomain Ca2+ transients is a potential therapeutic approach in stroke intervention.

Abstract Enriched environment (EE), characterized by multi‐sensory stimulation, represents a non‐invasive alternative for alleviating epileptic seizures. However, the mechanism by which EE exerts its therapeutic impact remains incompletely understood. Here, it is elucidated that EE mitigates seizure susceptibility through the augmentation of adult neurogenesis within the entorhinal cortex (EC) circuit. A substantial upregulation of adult hippocampal neurogenesis concomitant with a notable reduction in seizure susceptibility has been found following exposure to EE. EE‐enhanced adult‐born dentate granule cells (abDGCs) are functionally activated during seizure events. Importantly, the selective activation of abDGCs mimics the anti‐seizure effects observed with EE, while their inhibition negates these effects. Further, whole‐brain c‐Fos mapping demonstrates increased activity in DG‐projecting EC CaMKIIα+ neurons in response to EE. Crucially, EC CaMKIIα+ neurons exert bidirectional modulation over the proliferation and maturation of abDGCs that can activate local GABAergic interneurons; thus, they are essential components for the anti‐seizure effects mediated by EE. Collectively, this study provides compelling evidence regarding the circuit mechanisms underlying the effects of EE treatment on epileptic seizures, shedding light on the involvement of the EC‐DG circuit in augmenting the functionality of abDGCs. This may help for the translational application of EE for epilepsy management.

Abstract The presence of Griffiths phase to cluster spin glass transition has theoretically been predicted in both classical and quantum systems. However, its detection in a classical system has been lacking for decades, which hinders a complete understanding of the relationship between the Griffiths phase and cluster spin glass. Here, the experimental discovery of the Griffiths phase to cluster spin glass transition is reported in a classical magnetic system, diluted ferromagnets La2/3Sr1/3(Mn1‐3xAl2xTix)O3 (0 ≤ x ≤ 0.12). The phase diagram of the system shows a transition from the Griffiths phase into a ferromagnetic state in the low disorder concentration range (0.01 < x ≤ 0.09). In the high disorder concentration range (0.09 < x ≤ 0.12), a Griffiths phase to cluster spin glass transition is identified, which nicely matches that of disordered quantum systems. Moreover, the Griffiths phase is essentially an unfrozen cluster spin glass with partially broken ergodicity is demonstrated experimentally. These findings serve as crucial experimental references for understanding the glassy phenomena in disordered magnets, facilitating future exploration of their unique properties and functionalities.

ABSTRACT Aims/Introduction Type 2 diabetes mellitus is a major metabolic disease that seriously endangers life and health, but women with gestational diabetes mellitus are at increased risk for developing type 2 diabetes mellitus. This study aimed to evaluate the effectiveness of postpartum lifestyle intervention on the prevention of type 2 diabetes mellitus, and the effect of lifestyle intervention on glycemic outcomes and anthropometric measures. Materials and Methods We searched PubMed and other databases to retrieve articles published before May 21, 2023, on randomized controlled trials of postnatal lifestyle interventions (diet and/or physical activity) in women with gestational diabetes mellitus. We estimated the pooled odds ratios using fixed or random effects models and conducted a subgroup analysis of the different intervention methods to explore differences in the different lifestyle interventions. Results The review included 17 randomized controlled trials. Overall, lifestyle changes started after a pregnancy complicated by gestational diabetes mellitus an 11% (RR = 0.89; 95% CI: 0.74–1.07) reduction in diabetes risk; significant differences were found for weight (MD = −1.33; 95% CI: [−1.76; −0.89], P

Abstract Objective Deep brain stimulation (DBS) is a promising approach for the treatment of epilepsy. However, the optimal target for DBS and underlying mechanisms are still not clear. Here, we compared the therapeutic effects of DBS on distinct septal subregions, aimed to find the precise targets of septal DBS and related mechanisms for the clinical treatment. Methods Assisted by behavioral test, electroencephalography (EEG) recording and analyzing, selectively neuronal manipulation and immunohistochemistry, we assessed the effects of DBS on the three septal subregions in kainic acid (KA)‐induced mouse seizure model. Results DBS in the medial septum (MS) not only delayed generalized seizure (GS) development, but reduced the severity; DBS in the vertical diagonal band of Broca (VDB) only reduced the severity of GS, while DBS in the horizontal diagonal band of Broca (HDB) subregion showed no anti‐seizure effect. Notably, DBS in the MS much more efficiently decreased abnormal activation of hippocampal neurons. EEG spectrum analysis indicated that DBS in the MS and VDB subregions mainly increased the basal hippocampal low‐frequency (delta and theta) rhythm. Furthermore, ablation of cholinergic neurons in the MS and VDB subregions blocked the anti‐seizure and EEG‐modulating effects of septal DBS, suggesting the seizure‐alleviating effect of DBS was dependent on local cholinergic neurons. Significance DBS in the MS and VDB, rather than HDB, attenuates hippocampal seizure by activation of cholinergic neurons‐augmented hippocampal delta/theta rhythm. This may be of great therapeutic significance for the clinical treatment of epilepsy with septal DBS. Plain Language Summary The optical target of deep brain stimulation in the septum is still not clear. This study demonstrated that stimulation in the medial septum and vertical diagonal band of Broca subregions, but not the horizontal diagonal band of Broca, could alleviate hippocampal seizure through cholinergic neurons‐augmented hippocampal delta/theta rhythm. This study may shed light on the importance of precise regulation of deep brain stimulation therapy in treating epileptic seizures.

Abstract Current transformers (CTs) are widely used for energy metering, relay protection, condition monitoring and control circuits. However, CT saturation may lead to non‐negligible measurement errors and relay malfunctions, posing a threat to the stability and security of the power grid. In order to address the problem of CT saturation, a novel measurement‐protection‐integrated current transformer (MPICT) is proposed. First, the working states of the MPICT are summarised and the approximate expressions for the steady‐state measuring characteristics, the transient response characteristics, and the measurement errors are derived from the equivalent circuit model. Then, the feasibility of the MPICT and the theoretical analyses are verified by the 3D FEM simulation imitating the presented MPICT excited by diverse currents. Finally, a down‐scale prototype is fabricated and a series of tests are conducted in the laboratory to validate the effectiveness of the equipment. The simulation and experimental results suggest that the output of the MPICT can accurately reconstruct the primary current waveform, even if the primary current contains decaying or constant DC component.

Abstract Despite progress significant advances in immunotherapy for some solid tumors, pancreatic ductal adenocarcinoma (PDAC) remains unresponsive poorly responsive to such interventions, largely due to its highly immunosuppressive tumor microenvironment (TME) with limited CD8+ T cell infiltration. This study explores the role of the epigenetic factor Sin3B in the PDAC TME. Using murine PDAC models, we found that tumor cell‐intrinsic Sin3B loss reshapes the TME, increasing CD8+ T cell infiltration and cytotoxicity, thus impeding tumor progression and enhancing sensitivity to anti‐PD1 treatment. Sin3B‐deficient tumor cells exhibited amplified CXCL9/10 secretion in response to Interferon‐gamma (IFNγ), creating a positive feedback loop via the CXCL9/10‐CXCR3 axis, thereby intensifying the anti‐tumor immune response against PDAC. Mechanistically, extensive epigenetic regulation is uncovered by Sin3B loss, particularly enhanced H3K27Ac distribution on genes related to immune responses in PDAC cells. Consistent with the murine model findings, analysis of human PDAC samples revealed a significant inverse correlation between SIN3B levels and both CD8+ T cell infiltration and CXCL9/10 expression. Notebly, PDAC patients with lower SIN3B expression showed a more favorable response to anti‐PD1 therapy. The findings suggest that targeting SIN3B can enhance cytotoxic T cell infiltration into the tumor site and improve immunotherapy efficacy in PDAC, offering potential avenues for therapeutic biomarker or target in this challenging disease.

Abstract High‐time‐resolved mapping results of two rare reactivation processes following a negative return stroke are discovered and analyzed. The reactivation processes can be roughly divided into two different stages. At first, positive discharges occurred for tens of milliseconds in a limited space near a decayed negative leader channel. The discharges produced no detectable electric field change. Then, negative discharges started and propagated along the decayed downward negative leader channels at a speed exceeding 106 m/s for a few microseconds and produced negative electric field changes. The analysis reveals that the reactivation processes are physically distinct from recoil leaders and side discharges regarding propagation behaviors, electromagnetic characteristics, and time scales. The possible mechanisms of the processes are discussed. The observation suggests that the reactivation processes of the negative leaders may lead to subsequent return strokes.

Abstract Carbonized polymer dots (CPDs) have shown exceptional potential across a wide range of applications. However, their practical utilization is significantly greatly impeded by the lack of precise control over their structures and functionalities. Consequently, the development of controlled synthesis strategies for CPDs with well‐defined structures and tailored functionalities remains a critical challenge in the field. Here, the controlled synthesis of functional CPDs with reversible assembly properties via airflow‐assisted melt polymerization, followed by a one‐step post‐synthetic doping strategy, is reported. This synthetic approach achieves high product yield, uniform and tunable structures, as well as customized functionalities including solid‐state emission, enhanced catalytic performance (3.5–45 times higher than conventional methods), and selective gas storage in the resulting CPDs. The ability to tailor the properties of CPDs through controlled synthesis opens up new opportunities for their practical application in photocatalysis and gas storage.

Abstract Progress in the semiconductor industry relies on the development of increasingly compact devices consisting of complex geometries made from diverse materials. Precise, label‐free, and real‐time metrology is needed for the characterization and quality control of such structures in both scientific research and industry. However, optical metrology of 2D sub‐wavelength structures with nanometer resolution remains a major challenge. Here, a single‐shot and label‐free optical metrology approach that determines 2D features of nanostructures, is introduced. Accurate experimental measurements with a random statistical error of 18 nm (λ/27) are demonstrated, while simulations suggest that 6 nm (λ/81) may be possible. This is far beyond the diffraction limit that affects conventional metrology. This metrology employs neural network processing of images of the 2D nano‐objects interacting with a phase singularity of the incident topologically structured superoscillatory light. A comparison between conventional and topologically structured illuminations shows that the presence of a singularity with a giant phase gradient substantially improves the retrieval of object information in such an optical metrology. This non‐invasive nano‐metrology opens a range of application opportunities for smart manufacturing processes, quality control, and advanced materials characterization.

Abstract The ruminal microbiota generates biogenic methane in ruminants. However, the role of host genetics in modifying ruminal microbiota‐mediated methane emissions remains mysterious, which has severely hindered the emission control of this notorious greenhouse gas. Here, we uncover the host genetic basis of rumen microorganisms by genome‐ and transcriptome‐wide association studies with matched genome, rumen transcriptome, and microbiome data from a cohort of 574 Holstein cattle. Heritability estimation revealed that approximately 70% of microbial taxa had significant heritability, but only 43 genetic variants with significant association with 22 microbial taxa were identified through a genome‐wide association study (GWAS). In contrast, the transcriptome‐wide association study (TWAS) of rumen microbiota detected 28,260 significant gene–microbe associations, involving 210 taxa and 4652 unique genes. On average, host genetic factors explained approximately 28% of the microbial abundance variance, while rumen gene expression explained 43%. In addition, we highlighted that TWAS exhibits a strong advantage in detecting gene expression and phenotypic trait associations in direct effector organs. For methanogenic archaea, only one significant signal was detected by GWAS, whereas the TWAS obtained 1703 significant associated host genes. By combining multiple correlation analyses based on these host TWAS genes, rumen microbiota, and volatile fatty acids, we observed that substrate hydrogen metabolism is an essential factor linking host–microbe interactions in methanogenesis. Overall, these findings provide valuable guidelines for mitigating methane emissions through genetic regulation and microbial management strategies in ruminants.

Abstract The development of tumor drug microcarriers has attracted considerable interest due to their distinctive therapeutic performances. Current attempts tend to elaborate on the micro/nano‐structure design of the microcarriers to achieve multiple drug delivery and spatiotemporal responsive features. Here, the desired hydrogel microspheres are presented with spatiotemporal responsiveness for the treatment of gastric cancer. The microspheres are generated based on inverse opals, their skeleton is fabricated by biofriendly hyaluronic acid methacrylate (HAMA) and gelatin methacrylate (GelMA), and is then filled with a phase‐changing hydrogel composed of fish gelatin and agarose. Besides, the incorporated black phosphorus quantum dots (BPQDs) within the filling hydrogel endow the microspheres with outstanding photothermal responsiveness. Two antitumor drugs, sorafenib (SOR) and doxorubicin (DOX), are loaded in the skeleton and filling hydrogel, respectively. It is found that the drugs show different release profiles upon near‐infrared (NIR) irradiation, which exerts distinct performances in a controlled manner. Through both in vitro and in vivo experiments, it is demonstrated that such microspheres can significantly reduce tumor cell viability and enhance the efficiency in treating gastric cancer, indicating a promising stratagem in the field of drug delivery and tumor therapy.

Abstract Due to the weak radiation generated by positive leaders, our understanding of how positive leaders are initiated and reach the ground remains limited. This study investigated positive cloud‐to‐ground (CG) lightning induced after the long intracloud lightning based on high‐speed video and fast antenna mapping results. Below the cloud base in the field of view, re‐breakdowns of decayed negative branches occurred consecutively beneath the horizontal negative channel in the form of bidirectional leaders. These bidirectional leaders advanced along the same channel, and eventually, the third reached the ground, generating a positive return stroke (RS) with a peak current of 157 kA. Following the RS, new negative discharges emerge adjacent to the vertical return stroke channel, persisting and propagating to form a continuing current (CC) lasting over 200 milliseconds.

Abstract 5‐Fluorouracil (5‐FU) has been used for chemotherapy for colorectal and other cancers for over 50 years. The prevailing view of its mechanism of action is inhibition of thymidine synthase leading to defects in DNA replication and repair. However, 5‐FU is also incorporated into RNA causing defects in RNA metabolism, inhibition of pseudouridine modification, and altered ribosome function. We examined the impact of 5‐FU on post‐transcriptional small RNA modifications (PTxMs) and the expression and export of RNA into small extracellular vesicles (sEVs). EVs are secreted by all cells and contain a variety of proteins and RNAs that can function in cell‐cell communication. We found that treatment of colorectal cancer (CRC) cells with 5‐FU represses sEV export of miRNA and snRNA‐derived RNAs, but promotes export of snoRNA‐derived RNAs. Strikingly, 5‐FU treatment significantly decreased the levels of pseudouridine on both cellular and sEV small RNA profiles. In contrast, 5‐FU exposure led to increased levels of cellular small RNAs containing a variety of methyl‐modified bases. These unexpected findings show that 5‐FU exposure leads to altered RNA expression, base modification, and aberrant trafficking and localization of small RNAs.

Abstract RNA splicing is a dynamic molecular process in response to environmental stimuli and is strictly regulated by the spliceosome. Sm proteins, constituents of the spliceosome, are key components that mediate splicing reactions; however, their potential role in hepatocellular carcinoma (HCC) is poorly understood. In the study, SNRPD2 (PD2) is found to be the most highly upregulated Sm protein in HCC and to act as an oncogene. PD2 modulates DDX39A intron retention together with HNRNPL to sustain the DDX39A short variant (39A_S) expression. Mechanistically, 39A_S can mediate MYC mRNA nuclear export to maintain high MYC protein expression, while MYC in turn potentiates PD2 transcription. Importantly, digitoxin can directly interact with PD2 and has a notable cancer‐suppressive effect on HCC. The study reveals a novel mechanism by which DDX39A senses oncogenic MYC signaling and undergoes splicing via PD2 to form a positive feedback loop in HCC, which can be targeted by digitoxin.

Abstract The Ice Sheet Model Intercomparison Project for CMIP6 (ISMIP6) is the primary effort of CMIP6 (Coupled Model Intercomparison Project–Phase 6) focusing on ice sheets, designed to provide an ensemble of process‐based projections of the ice‐sheet contribution to sea‐level rise over the twenty‐first century. However, the behavior of the Antarctic Ice Sheet beyond 2100 remains largely unknown: several instability mechanisms can develop on longer time scales, potentially destabilizing large parts of Antarctica. Projections of Antarctic Ice Sheet evolution until 2300 are presented here, using an ensemble of 16 ice‐flow models and forcing from global climate models. Under high‐emission scenarios, the Antarctic sea‐level contribution is limited to less than 30 cm sea‐level equivalent (SLE) by 2100, but increases rapidly thereafter to reach up to 4.4 m SLE by 2300. Simulations including ice‐shelf collapse lead to an additional 1.1 m SLE on average by 2300, and can reach 6.9 m SLE. Widespread retreat is observed on that timescale in most West Antarctic basins, leading to a collapse of large sectors of West Antarctica by 2300 in 30%–40% of the ensemble. While the onset date of retreat varies among ice models, the rate of upstream propagation is highly consistent once retreat begins. Calculations of sea‐level contribution including water density corrections lead to an additional ∼10% sea level and up to 50% for contributions accounting for bedrock uplift in response to ice loading. Overall, these results highlight large sea‐level contributions from Antarctica and suggest that the choice of ice sheet model remains the leading source of uncertainty in multi‐century projections.

Abstract Hippocampal CA1 neurons show intense firing at specific spatial locations, modulated by isolated landmarks. However, the impact of real‐world scene transitions on neuronal activity remains unclear. Moreover, long‐term neural recording during movement challenges device stability. Conventional rigid‐based electrodes cause inflammatory responses, restricting recording durations. Inspired by the jellyfish tentacles, the multi‐conductive layer ultra‐flexible microelectrode arrays (MEAs) are developed. The tentacle MEAs ensure stable recordings during movement, thereby enabling the discovery of soft boundary neurons. The soft boundary neurons demonstrate high‐frequency firing that aligns with the boundaries of scene transitions. Furthermore, the localization ability of soft boundary neurons improves with more scene transition boundaries, and their activity decreases when these boundaries are removed. The innovation of ultra‐flexible, high‐biocompatible tentacle MEAs improves the understanding of neural encoding in spatial cognition. They offer the potential for long‐term in vivo recording of neural information, facilitating breakthroughs in the understanding and application of brain spatial navigation mehanisms.

Abstract Our current knowledge regarding the distribution patterns of ancient and recent recycled materials, as well as the origin of intraplate basalts in the eastern Asia Big Mantle Wedge (BMW), is limited. To address this, we conducted a detailed geochemical analysis of olivine melt inclusions (OLMI) in nephelinite samples. The normal OLMIs detected in our investigation exhibit geochemical features that closely resemble those of the hosting nephelinite, indicating a consistent association with a carbonated mantle source. Additionally, we identified a distinct group of anomalous OLMIs that displayed different geochemical characteristics from the hosting nephelinite but showed similarities to regional alkali basalts sourced from pyroxenite. The observed geochemical diversity in the nephelinite OLMIs suggests an interaction between deeply derived carbonated melts originating from the flattened Pacific slab in the mantle transition zone and pyroxenite in the lithospheric mantle. Carbonated fluid‐fluxed melting is key to basalt formation in the BMW.

Abstract In this letter, a Quasi‐Helmholtz Projector method including multibranch Rao‐Wilton‐Glisson (MB‐RWG) basis function to calculate the scattering problem of multiscale targets at low frequencies is proposed. The loop basis function and the star basis function including MB‐RWG basis function are constructed, and the loop basis function and the star basis function including MB‐RWG basis function are used to construct the Quasi‐Helmholtz Projector. The proposed method is coined MB‐Quasi‐Helmholtz Projector (MB‐QHP) method. The MB‐QHP method can effectively solve the low frequency breakdown (LFB) problem in electric field integral equation (EFIE). Compared with the MB‐loop‐star method, the condition number is lower, the convergence is faster, and the search for global loop is avoided. Meanwhile, due to the existence of the MB‐RWG basis function, the proposed method can divide the calculation region into several regions with different mesh sizes. Numerical example shows the advantages of the proposed method.

Abstract The evolution of charge structure involved in lightning discharge of a thunderstorm over the central Tibetan Plateau is investigated for the first time, based on the data from very high frequency interferometer, radar and sounding. During the developing‐mature stage, the TP thunderstorm exhibited a tripolar charge structure evolved from an initial inverted dipole. Abstract Creating photonic crystals that can integrate and switch between multiple structural color images will greatly advance their utility in dynamic information transformation, high‐capacity storage, and advanced encryption, but has proven to be highly challenging. Here, it is reported that by programmably integrating newly developed 1D quasi‐periodic folding structures into a 3D photonic crystal, the generated photonic superstructure exhibits distinctive optical effects that combine independently manipulatable specular and anisotropic diffuse reflections within a versatile protein‐based platform, thus creating different optical channels for structural color imaging. The polymorphic transition of the protein format allows for the facile modulation of both folding patterns and photonic lattices and, therefore, the superstructure's spectral response within each channel. The capacity to manipulate the structural assembly of the superstructure enables the programmable encoding of multiple independent patterns into a single system, which can be decoded by the simple adjustment of lighting directions. The multifunctional utility of the photonic platform is demonstrated in information processing, showcasing its ability to achieve multimode transformation of information codes, multi‐code high‐capacity storage, and high‐level numerical information encryption. The present strategy opens new pathways for achieving multichannel transformable imaging, thereby facilitating the development of emerging information conversion, storage, and encryption media using photonic crystals.

Abstract Background The diagnosis of glioma has advanced since the release of the WHO 2021 classification with more molecular alterations involved in the integrated diagnostic pathways. Our study aimed to present our experience with the clinical features and management of astrocytoma, IDH mutant based on the latest WHO classification. Methods Patients diagnosed with astrocytoma, IDH‐mutant based on the WHO 5th edition classification of CNS tumors at our center from January 2009 to January 2022 were included. Patients were divided into WHO 2–3 grade group and WHO 4 grade group. Integrate diagnoses were retrospectively confirmed according to WHO 2016 and 2021 classification. Clinical and MRI characteristics were reviewed, and survival analysis was performed. Results A total of 60 patients were enrolled. 21.67% (13/60) of all patients changed tumor grade from WHO 4th edition classification to WHO 5th edition. Of these, 21.43% (6/28) of grade II astrocytoma and 58.33% (7/12) of grade III astrocytoma according to WHO 4th edition classification changed to grade 4 according to WHO 5th edition classification. Abstract Objectives Patients with advanced colorectal cancer (CRC) have multiple concurrent physical and psychological symptoms. This study aimed to explore the relationship between anxiety, depression, and symptom burden in advanced CRC. Methods A multicenter cross‐sectional study was conducted in 10 cancer centers from geographically and economically diverse sites in China. A total of 454 patients with advanced CRC completed the Hospital Anxiety and Depression Scale and the MD Anderson Symptom Inventory. Abstract The management of infected wounds is always of great significance and urgency in clinical and biomedical fields. Recent efforts in this area are focusing on the development of functional wound patches with effective antibacterial, drug delivery, and sensor properties. Here, we present novel hyaluronic acid (HA) microneedle patches with these features by encapsulating aminobenzeneboronic acid‐modified gold nanoclusters (A‐GNCs) for infected wound management. The A‐GNCs loaded microneedle patches were derived from negative‐mold replication and showed high mechanical strength to penetrate the skin. The release of the A‐GNCs was realized by the degradation of HA, and the self‐monitor of the released actives was based on the dynamic bright orange fluorescence emitted from A‐GNCs under ultraviolet radiation. As the A‐GNCs could destroy bacteria membranes, the microneedle patches were with excellent in vitro antibiosis ability. Based on these features, we have demonstrated the bacteria inhibition, residual drug self‐monitoring, and wound healing promotion abilities of the microneedle patches in Escherichia coli‐ or Staphylococcus aureus‐infected wound management. These results indicated the great potential of such A‐GNCs loaded microneedle patches for clinical applications.

Abstract Mycobacterium tuberculosis (MTB) is the causal pathogen of tuberculosis (TB). Rapid and accurate detection of live MTB is important for transmission control and patient treatment. Here, we described a clustered regularly interspaced short palindromic repeat (CRISPR)‐Cas13a‐based molecular diagnosis approach for rapid and specific detection of live MTB. This detection method, which we termed CRISPR‐Live‐MTB, contained two consecutive reactions including nuclear acid sequence‐based amplification (NASBA) and CRISPR‐Cas13a collateral cleavage reaction. CRISPR‐Live‐MTB could efficiently detect MTB single‐stranded RNA (ssRNA) in 2 hours with high specificity over double‐stranded DNA (dsDNA). Importantly, CRISPR‐Live‐MTB exhibited a limit of detection of 2.4 copies for MTB ssRNA, which was 1000 times lower than that of the clinically used NASBA method. Moreover, lateral flow was integrated into the CRISPR‐Live‐MTB method to enable point‐of‐care testing application with a sensitivity of 95% and a specificity of 100%. Overall, our study demonstrated the feasibility of CRISPR‐Live‐MTB as a rapid, sensitive, and specific approach for live MTB detection.

Abstract Combination therapy has emerged as an efficacy strategy for treating complex diseases. Its potential to overcome drug resistance and minimize toxicity makes it highly desirable. However, the vast number of potential drug pairs presents a significant challenge, rendering exhaustive clinical testing impractical. In recent years, deep learning‐based methods have emerged as promising tools for predicting synergistic drug combinations. This review aims to provide a comprehensive overview of applying diverse deep‐learning architectures for drug combination prediction. This review commences by elucidating the quantitative measures employed to assess drug combination synergy. Subsequently, we delve into the various deep‐learning methods currently employed for drug combination prediction. Finally, the review concludes by outlining the key challenges facing deep learning approaches and proposes potential challenges for future research.

Abstract Background and Objective While earlier studies have focused on the relationship between stress and obesity, there was a gap in understanding the potential impact of positive psychological factors, such as resilience, on obesity. By investigating the role of psychological resilience with obesity, this study aimed to address this gap and tackle obesity through a positive psychological framework. Methods Participants consisted of 2445 community residents from Shenzhen, China, with a mean age of 41.09 ± 13.72 years, comprising 846 males and 1599 females. Psychological resilience was measured using the Brief Resilience Scale; gender, age, marital status, education level, smoking status, alcohol consumption, frequency of physical exercise, and perceived stress were considered potential confounding factors. The relationship between psychological resilience and body mass index (BMI) was examined through multiple linear regression and logistic regression analyses. Results The participants had an average psychological resilience score of 3.46 (standard deviation [SD] = 0.62) and an average BMI of 22.59 (SD = 3.35), with 104 individuals (4.3%) identified with obesity. In the fully adjusted multiple linear regression model, a higher psychological resilience score was associated with a higher BMI (β = 0.507, 95% CI:0.283, 0.731). In the logistic regression model, higher psychological resilience scores were linked to increased obesity risk, with a more significant association observed among males (odds ratio [OR] = 2.169, 95% CI:1.155, 4.073), while psychological resilience acted as a protective factor against underweight among females (OR = 0.528, 95% CI:0.376, 0.816). Conclusion The study demonstrated a significant link between higher psychological resilience and elevated BMI, emphasizing the complex relationship between psychological fortitude and weight management. Interventions targeting socioeconomic status, education, lifestyle habits, and physiological well‐being might offer a promising strategy for enhancing psychological resilience and promoting healthier weight. Emphasizing self‐efficacy and coping skills at the individual level could contribute to balanced weight and comprehensive health outcomes, addressing the global challenge of obesity.

Abstract Background Patients diagnosed with advanced stage cancer face an elevated risk of suicide. We aimed to develop a suicidal ideation (SI) risk prediction model in patients with advanced cancer for early warning of their SI and facilitate suicide prevention in this population. Patients and Methods We consecutively enrolled patients with multiple types of advanced cancers from 10 cancer institutes in China from August 2019 to December 2020. Demographic characteristics, clinicopathological data, and clinical treatment history were extracted from medical records. Symptom burden, psychological status, and SI were assessed using the MD Anderson Symptom Inventory (MDASI), Hospital Anxiety and Depression Scale (HADS), and Patient Health Questionnaire‐9 (PHQ‐9), respectively. A multivariable logistic regression model was employed to establish the model structure. Results In total, 2814 participants were included in the final analysis. Nine predictors including age, sex, number of household members, history of previous chemotherapy, history of previous surgery, MDASI score, HADS‐A score, HADS‐D score, and life satisfaction were retained in the final SI prediction model. The model achieved an area under the curve (AUC) of 0.85 (95% confidential interval: 0.82–0.87), with AUCs ranging from 0.75 to 0.95 across 10 hospitals and higher than 0.83 for all cancer types. Conclusion This study built an easy‐to‐use, good‐performance predictive model for SI. Implementation of this model could facilitate the incorporation of psychosocial support for suicide prevention into the standard care of patients with advanced cancer.

Abstract License plate detection is an important task in Intelligent Transportation Systems (ITS) and has a wide range of applications in vehicle management, traffic control, and public safety. In order to improve the accuracy and speed of mobile recognition, an improved lightweight YOLOv5s model is proposed for license plate detection. First, an improved Stemblock network is used to replace the original Focus layer in the network, which ensures strong feature expression capability and reduces a large number of parameters to lower the computational complexity; then, an improved lightweight network, ShuffleNetv2, is used to replace the backbone network of the YOLOv5s, which makes the model lighter and ensures the detection accuracy at the same time. Then, a feature enhancement module is designed to reduce the information loss caused by the rearrangement of the backbone network channels, which facilitates the information interaction in the feature fusion process; finally, the low‐, medium‐ and high‐level features in the Shufflenetv2 network structure are fused to form the final high‐level output features. Experimental results on the CCPD dataset show that compared to other methods this paper obtains better performance and faster speed in the license plate detection task, in which the average precision mean value reaches 96.6%, and can achieve a detection speed of 43.86 frame/s, and the parameter volume is reduced to 5.07 M.

Objective The surgical treatment of patients with ankylosing spondylitis and severe thoracolumbar kyphosis combined with hip flexion contracture is very difficult for all the surgeons. The femoral neck osteotomy (FNO) is the first step to break the ice. The evaluation of a new modified FNO method is very important to improve the curative effect. Methods Five male patients with nine bone‐fused hips who underwent the new femoral neck osteotomy were included from October 2021 to March 2022. The FNO was designed that the saw blade was manipulated from the lateral femoral neck base to the inferior part of the femoral head, keeping Pauwels' angle less than 30° on the coronal plane. On the transverse plane, the angle between the saw blade and the coronal plane was more than 15°. On the sagittal plane, the saw blade cut through the femoral neck. They accepted pedicle subtraction osteotomy (PSO) after FNO according to the patient’ recovery. Then, 2 weeks later, the patients underwent total hip arthroplasty (THA). The visual analogue scale (VAS), Harris hip score (HHS) and passive hip flexion‐extension range of motion (ROM) were used to evaluate hip function. The data were analyzed by paired t‐test. Results The average operation time and blood loss of FNO, the average interval between FNO and THA were collected. The average angle of the trunk and lower limb (ATL) was 36.33° ± 16.36° pre‐FNO, 82.89° ± 13.51° post‐FNO and 175.22° ± 3.42° post‐THA. The average VAS scores were 0 pre‐FNO, 5 ± 1.58 post‐FNO and 2.6 ± 0.55 post‐THA. The average HHS was 43.56 ± 1.59 preoperatively and 83.89 ± 2.21 postoperatively. Objective Intertrochanteric fracture is a very common but serious type of hip fracture in nonagenarians. The surgical treatment remains a significant challenge for orthopedists. The objective of this study was to investigate postoperative complications and survival outcomes compared between bipolar hemiarthroplasty (HA) and proximal femoral nail anti‐rotation (PFNA) in nonagenarians with intertrochanteric fractures, and to evaluate the efficacy and safety of the two surgical procedures in this patient population. Methods A total of 113 consecutive nonagenarians who underwent bipolar HA or PFNA for the treatment of intertrochanteric fractures from January 2006 to August 2021 were retrospectively studied in the current paper. There were 34 males and 79 females, with a mean age of 92.2 years (range 90–101 years) at the time of operation. The average duration of follow‐up was 29.7 months (range 1–120 months). The full cohort was divided into bipolar HA (77 cases) and PFNA (36 cases) groups. Damage control orthopedics was used to determine the optimal surgery time and assist in perioperative management. A restrictive blood transfusion strategy was employed, along with appropriate adjustments under multidisciplinary assessment, throughout the perioperative period. Perioperative clinical information and prognostic data were analyzed. Kaplan–Meier survival curves were used for survival analysis, and landmark analysis divided the entire follow‐up period into 1–12 months (short‐term), 13–42 months (medium‐term) and 43–120 months (long‐term) according to the configurations of Kaplan–Meier survival curves. Results Both groups had similar general variables except for the proportion of high adjusted Charlson comorbidity index (aCCI) (≥6 points) (6.5% in bipolar HA group and 22.2% in PFNA group, p = 0.024). Intraoperative blood loss and transfusion requirements were greater, and the intraoperative transfusion rates were higher in the bipolar HA group compared to the PFNA group (all p 0.05). Both groups had similar overall survival curves (p = 0.37). However, landmark analysis revealed that bipolar HA group exhibited higher survival rates in medium‐term (p = 0.01), while similar survival rates were observed in the short‐ and long‐term post‐operation periods (both p > 0.05). Cox regression with survival‐time‐dependent covariate calculated the hazard ratio (HR) of bipolar HA was 0.41 in medium‐term (p = 0.039). Conclusion Bipolar HA is equally effective and reliable as PFNA for treating intertrochanteric fractures in nonagenarians. Despite resulting in more intraoperative blood loss and transfusions, bipolar HA therapy is associated with a higher medium‐term survival rate compared to PFNA treatment. The application of damage control orthopedics and precise perioperative patient blood management could contribute to the positive clinical outcomes observed in this patient population.

Abstract The emergence and rapid spread of antimicrobial resistance is of global public health concern. The gut microbiota harboring diverse commensal and opportunistic bacteria that can acquire resistance via horizontal and vertical gene transfers is considered an important reservoir and sink of antibiotic resistance genes (ARGs). In this review, we describe the reservoirs of gut ARGs and their dynamics in both animals and humans, use the One Health perspective to track the transmission of ARG‐containing bacteria between humans, animals, and the environment, and assess the impact of antimicrobial resistance on human health and socioeconomic development. The gut resistome can evolve in an environment subject to various selective pressures, including antibiotic administration and environmental and lifestyle factors (e.g., diet, age, gender, and living conditions), and interventions through probiotics. Strategies to reduce the abundance of clinically relevant antibiotic‐resistant bacteria and their resistance determinants in various environmental niches are needed to ensure the mitigation of acquired antibiotic resistance. With the help of effective measures taken at the national, local, personal, and intestinal management, it will also result in preventing or minimizing the spread of infectious diseases. This review aims to improve our understanding of the correlations between intestinal microbiota and antimicrobial resistance and provide a basis for the development of management strategies to mitigate the antimicrobial resistance crisis.

Abstract Background This real‐world study investigated the outcome of COVID‐19 in lymphoma patients participating in registered clinical trials and explored potential risk factors with the outcome of COVID‐19 during the first wave of the Omicron outbreak in China. Methods One hundred and ten patients participating in registered clinical trials and diagnosed with COVID‐19 in our center between December 1, 2022, and January 31, 2023, were included. Results Four (3.6%) patients were identified as severe COVID‐19 and 2 (1.8%) as critical COVID‐19, respectively. The mortality rate observed was 2.73% for the entire cohort, 33.3% for the severe/critical COVID‐19 group, and 18.8% for the hospitalized group. The 90‐day OS was 98.2% for the entire cohort, 66.7% for the severe/critical COVID‐19 group, and 87.5% for the hospitalized group. Advanced age (≥70 years), comorbidities, and PI3K inhibitor‐containing regimen were significantly associated with the severity of COVID‐19. Patients with indolent B‐cell non‐Hodgkin lymphomas were less likely to be hospitalized for COVID‐19. Conclusion This study reported similar clinical features of COVID‐19 in our cohort with that of non‐hematological malignancy (HM) patients, while the proportion of severe/critical COVID‐19 and the mortality rate were relatively higher than non‐HM patients. Our findings provided valuable experience to aid clinical researchers with managing lymphoma patients participating in registered clinical trials during the ongoing pandemic of the Omicron variant.

Abstract Background Lenvatinib is a multitargeted tyrosine kinase inhibitor used in the treatment of a variety of solid tumors. This study aims to investigate the potential pharmacokinetic interactions between lenvatinib and various azoles (ketoconazole, voriconazole, isavuconazole and posaconazole) when orally administered to rats. Methods A total of 30 Sprague–Dawley rats were randomly allocated into five groups and administered 20 mg/kg of ketoconazole, voriconazole, isavuconazole and 30 mg/kg of posaconazole and 0.5% CMC‐Na, through gavage for a duration of 7 days prior to the commencement of the experiment. On the final day, the rats were given 10 mg/kg of lenvatinib. The blood concentration of lenvatinib was determined using UPLC‐MS–MS. In vitro lenvatinib were incubated with azoles and rat liver microsomes (RLMs) or human liver microsomes (HLMs). Objective Surgical strategy for spinal kyphosis in patients with ankylosing spondylitis (AS) has been challenging. Pedicle subtraction osteotomy (PSO) through a minimally invasive (MI) approach has been developed with promising clinical outcomes. We aimed to compare the effectiveness and safety of PSO via an MI approach and a standard posterior approach (SPA) for treating AS‐related spinal kyphosis. Methods A total of 41 patients with AS‐related spinal kyphosis who underwent PSO through an MI approach (MI surgery [MIS] group: n = 25) or SPA (SPA group: n = 16) between January 2015 and July 2020 were retrospectively included. Spinopelvic parameters were evaluated before the surgery, immediately after the surgery, and at the 2‐year follow‐up. Clinical data including operative time, estimated blood loss, blood transfusion, level of fusion, incision length, bed rest period, length of hospitalization, and surgical complications were compared between the two groups. The Scoliosis Research Society outcomes instrument‐22 (SRS‐22) was administered to assess patients' quality of life at the latest follow‐up. Comparisons between the two groups were performed using independent sample t‐test or Chi‐square test. Results Characteristics and baseline kyphosis of the two groups were matched. At the 2‐year follow‐up, in the MIS group, the average correction values of the sagittal vertical axis and global kyphosis (GK) were 9.5 cm and 44.3°, respectively. Compared with the SPA group, the MIS group had similar correction values and correction losses after surgery. No obvious differences were observed in any radiographic parameters, except for GK, immediately after surgery and at the 2‐year follow‐up between the two groups (p > 0.05). The MIS group had a significantly shorter operative time, lesser blood loss, lesser transfusion volume, shorter fusion level, and lesser time to mobilization than did the SPA group. Higher average functional activity scores of SRS‐22 were obtained in the MIS group than in the SPA group. Conclusion Mini‐open PSO may be an effective alternative to the SPA for treating AS‐related spinal kyphosis, with comparable correction effect, lesser surgical trauma and faster recovery. This comparative study may provide valuable guidance for surgical decision‐making and patient counseling.
