Abstract Protein aggregate species play a pivotal role in the pathology of various degenerative diseases. Their dynamic changes are closely correlated with disease progression, making them promising candidates as diagnostic biomarkers. Given the prevalence of degenerative diseases, growing attention is drawn to develop pragmatic and accessible protein aggregate species detection technology. However, the performance of current detection methods is far from satisfying the requirements of extensive clinical use. In this review, we focus on the design strategies, merits, and potential shortcomings of each class of detection methods. The review is organized into three major parts: native protein sensing, seed amplification, and intricate program, which embody three different but interconnected methodologies. To the best of our knowledge, no systematic review has encompassed the entire workflow, from the molecular level to the apparatus organization. This review emphasizes the feasibility of the methods instead of theoretical detection limitations. We conclude that high selectivity does play a pivotal role, while signal compilation, multilateral profiling, and other patient‐oriented strategies (i.e. less invasiveness and assay speed) are also important.

Abstract Although a high titre of malic acid is achieved by filamentous fungi, by‐product succinic acid accumulation leads to a low yield of malic acid and is unfavourable for downstream processing. Herein, we conducted a series of metabolic rewiring strategies in a previously constructed Myceliophthora thermophila to successfully improve malate production and abolish succinic acid accumulation. First, a pyruvate carboxylase CgPYC variant with increased activity was obtained using a high‐throughput system and introduced to improve malic acid synthesis. Subsequently, shifting metabolic flux to malate synthesis from mitochondrial metabolism by deleing mitochondrial carriers of pyruvate and malate, led to a 53.7% reduction in succinic acid accumulation. The acceleration of importing cytosolic succinic acid into the mitochondria for consumption further decreased succinic acid formation by 53.3%, to 2.12 g/L. Finally, the importer of succinic acid was discovered and used to eliminate by‐product accumulation. In total, malic acid production was increased by 26.5%, relative to the start strain JG424, to 85.23 g/L and 89.02 g/L on glucose and Avicel, respectively, in the flasks. In a 5‐L fermenter, the titre of malic acid reached 182.7 g/L using glucose and 115.8 g/L using raw corncob, without any by‐product accumulation. This study would accelerate the industrial production of biobased malic acid from renewable plant biomass.

Abstract The vehicle suspension system is considered to have tremendous potential in energy harvesting and to be applicable for improving the efficiency of energy use of the vehicle. A novel piezoelectric energy harvester (PEH) with noncontact magnetic force has the feature of low friction loss and is designed based on the vibration characteristic of the suspension system. The research comprises two main components: the motion conversion component and the energy conversion component. A screw‐nut mechanism that can convert linear vibration between the vehicle body and the wheel into rotational motion is used as the motion conversion component. The energy conversion component is a core component of the PEH; its function is to convert rotational energy into electrical energy through noncontact magnetic force and piezoelectric effect. The magnetic excitation model's credibility and the modeling method's feasibility are verified by experiment and simulation. Through vehicle field tests, the influence of the driving speed, road surface roughness, and cargo state on the harvested power is obtained. Driving on a random road, the maximum of the generated power is 24.28 W, at 60 km/h, under the laden state. Driving on a pulse road, the harvested power is 3346 W, at 30 km/h, under the unladen state. It is indicated that the PEH is a promising and practical option for harvesting energy for the vehicle's suspension system.

Abstract Biologically inspired neuromorphic sensory memory systems based on memristor have received a lot of attention in the booming artificial intelligence industry due to significant potential to effectively process multi‐sensory signals from complex external environments. However, many memristors have significant switching parameters disperse, which is a great challenge for using memristors in bionic neuromorphic sensory memory systems. Herein, a stable ferroelectric memristor based on the Pd/BaTiO3:Eu2O3/La0.67Sr0.33MnO3 grown on Silicon structure with SrTiO3 as buffer layer is presented. The device possesses low coercive field voltage (−1.3–2.1 V) and robust endurance characteristic (~1010 cycles) through optimizing the growth temperature. More importantly, an ultra‐stable artificial multimodal sensory memory system with visual and tactile functions was reported for the first time by combining a pressure sensor, a photosensitive sensor, and a robotic arm. Utilizing the above system, the sensitivity value of the system is expressed by the conductance of the memristor to realize the gradual change of external stimulus, and multi signals inputs at the same time to this system have faithfully achieved sensory adaptation to multimodal sensors. This work paves the way for future development of memristor‐based perception systems in efficient multisensory neural robots.

Osteosarcoma (OS) is a prototypical sarcoma, predominantly affecting adolescents. The hallmark of cancer stem cell (CSC) behavior pervades OS, invariably signifying an unfavorable prognosis. However, the intricacies underlying OS metastasis remain incompletely comprehended. As the frontiers of the scientific investigation push forward, encompassing both bulk sequencing and single-cell sequencing (scRNA), the domain of bioinformatics finds multifaceted utility. In this study, we combined scRNA and bulk sequencing with the clinical metadata to excavate the latent molecular substrates governing metastatic propensities within OS. Our scRNA analysis indicated that cell-stemness-related pathways might play vital roles in OS metastasis. Subsequently, an autonomous reservoir of bulk sequencing data set was subjected to weighted gene co-expression network analysis (WGCNA) to identify 10 gene clusters. After analyzing the clinical data, we were able to identify two hub genes, HDAC2 and HSPA4, which are strongly linked to the cancer cell stemness. Moreover, we performed transwell assays to validate the regulation of metastatic behaviors by miR-1-3p, HDAC2, and HSPA4 in OS cells. Significantly, our study was fortified by immunohistochemistry (IHC) analyses performed on tumor tissues acquired from OS patients, thereby accentuating the clinical import of our experimental endeavor. Notably, we unveiled the suppressive influence of miR-1-3p on both HDAC2 and HSPA4, with miRTarBase substantiating lower expression in tumor tissue relative to the normative cohort. Insights gleaned from the Therapeutically Applicable Research to Generate Effective Treatments (TARGET) program (https://ocg.cancer.gov/programs/target) further augmented the clinical significance of the identified hub genes. In conclusion, our findings highlight the significant role of cell junctions in governing OS stemness and metastasis, underscored by the integration of single-cell sequencing and bulk-sequencing analyses. Moreover, our foundational experiments identified three hub molecules closely associated with the metastatic behaviors. Our findings provide novel insights into the clinical treatment and fundamental understanding of OS.

Background. In-stent restenosis (ISR) is a common complication after percutaneous coronary intervention (PCI) surgery for patients with coronary atherosclerotic heart disease (CHD). Reports indicate alprostadil may reduce ISR, and this study aimed at reviewing and summarizing the effect of nanoliposome alprostadil on ISR by meta-analysis. Methods. Articles were searched from databases, and meta-analysis was performed in Review Manager software. Funnel plots were performed to evaluate the publication bias, and sensitivity analysis was performed to determine the robustness of the overall treatment effects. Results. Initially, 113 articles were identified, and 5 studies of 463 subjects were included for analysis eventually. The primary endpoint, i.e., the occurrence of ISR after PCI, occurred in 11.91% of the alprostadil treatment group (28 from 235 patients) vs. 21.49% of the conventional treatment group (49 from 228 patients) and showed a statistical significance in our pooled data (χ2 = 7.654, P=0.006), while there was no statistically significant difference in all of the separate studies. We observed no statistical methodological heterogeneity among the studies (P=0.64, I2 ≈ 0%). The pooled odds ratio (OR) of the occurrence of ISR was 49% in a fixed-effect model, and the 95% confidence boundary (95% CI) was 29% to 81%. The funnel plot did not show serious publication bias, and sensitivity analysis showed well robustness of the overall treatment effect. Discussion. In conclusion, the early application of nanoliposome alprostadil after PCI could effectively reduce the occurrence of ISR, and the overall effect of alprostadil treatment in reducing ISR after PCI was relatively stable.

Abstract High power‐generation efficiency can be achieved by optimising the shape of the claw pole used as automobile generators. The working process of the stator and rotor in the claw pole generator was simulated using the finite‐element method. First, one of the eight typical claw pole types commonly used in industry was selected. Then, orthogonal optimisation was performed based on the simulation results, yielding an optimal claw shape design. To examine the adaptability of the designed claw pole to shape change, the non‐destructive test for evaluating the magnetic properties of claw pole (NDTEM) was employed. Finally, a claw pole with an optimal shape was forged, and its magnetic induction intensity was assessed using the NDTEM. The results indicated that compared with the unoptimised claw pole, the magnetic induction intensity of the optimised claw pole increased by a maximum of 8.11% and its weight reduced by 4.86%. This optimisation effectively improved the efficiency of power generation and achieved the lightweight of the claw pole.

Ferroelectric Hf0.5Zr0.5O2 (HZO) thin films have attracted wide attention in terms of potential applications of nonvolatile ferroelectric memories. However, the effect of strain on the resistance switching characteristics of the ferroelectric HZO thin‐film memristors has not been fully studied so far. In this work, the strain effects on the HZO thin‐film memristors are investigated. HZO films with different resistance properties are also prepared by controlling the value of oxygen pressure. Based on the testing results, it is proposed that the resistance switching behavior of HZO films may be caused by the joint participation between ferroelectricity and oxygen vacancy migration. The study also found that HZO films can successfully simulate learning behavior similar to the human brain. The applied pulses with a width of tens of nanoseconds timescale are beneficial to realize fast learning and computing. These results provide a fundamental and deep insight on HZO‐based ferroelectric semiconductor oxide thin‐film memristors and their potential applications in next‐generation artificial electronic synaptic devices.

Abstract Energy storage system can be used to increase the fuel economy of fuel cell system (FCS). In this study, a new method was introduced for optimizing the energy management strategy (EMS) for fuel cell vehicle (FCV) to reduce fuel consumption. The membership function (MF) in fuzzy control is subjective; thus, 12 design variables in the input‐output MFs were selected using sensitivity analysis, and elliptical basis function neural network method was used to establish a high‐precision approximate model of FCV. Multi‐island genetic algorithm was used to optimize the MFs. The effectiveness of the optimized fuzzy control EMS and the proposed optimization method were demonstrated in simulations of two EMSs under four driving cycles. The simulation results confirmed that the optimized fuzzy control EMS provided smoother and more stable output power from FCS reducing hydrogen consumption by 8.4%, 1.1%, 5.1%, and 7.6%, respectively, compared to the original fuzzy control EMS; and hydrogen saved by the optimized EMS provided extra range of 9.15, 1.10, 5.37, and 8.25 km per 100 km in the four driving cycles, respectively. The optimized EMS can reduce hydrogen consumption to increase fuel economy and extend the life span of the fuel cell.

As well known, the crack is a typical fault of turbine blades, which is difficult to detect due to the weak fault features contained in the rotor vibration. Although some fault diagnosis methods based on blade tip timing (BTT) are proposed to detect blade cracks, the limited fault features can be extracted because the BTT signals contain only one-dimensional fault information. Based on the dynamic analysis, the cracked blade undergoes three-dimensional (3D) deformation so that it would be reflected in the changes of the three-dimensional blade tip clearance (3D-BTC). Therefore, the dynamic characteristics of the 3D-BTC of turbine blades with typical cracks should be investigated to obtain its change rule and achieve a more comprehensive diagnosis of blade cracks. In this paper, the 3D finite element model of the turbine blisk is established, and the fluid-thermal-solid coupling analysis is implemented to calculate the blade deformation. Then, a novel dynamic computation method is proposed to calculate the 3D-BTC based on the blade deformation. The typical blade cracks are taken into consideration, and the dynamic characteristics of the 3D-BTC regarding the typical cracks are analyzed to obtain its change rule. The results show that abundant crack fault information can be obtained based on the 3D-BTC, which can facilitate the comprehensive diagnosis of turbine blade cracks.

To understand common but distinctive systems to drive oncogenic stages in human tumors is critical for understanding disease programme and developing novel therapeutic strategies. PBRM1 is a critical gene in oncogenesis. We found that PBRM1 is upregulated in multiple cancer genes. Prognostic analyses indicated that higher PBRM1 showed better disease outcomes of head and neck squamous cell carcinoma (HNSC), KIRC, and UCEC, while poorer outcomes in KICH, skin cutaneous melanoma (SKCM), and esophageal carcinoma (ESCA). PBRM1 mutation was most frequent in renal cell carcinoma and showed better disease outcomes of pan-cancer. We also discovered that PBRM1 performance was associated with endothelial cell invasion status in COAD, HNSC, KIRC, LUAD, LUSC, OV, and PAAD, and cancer-related fibroblast invasion was observed in COAD, HNSC, KIRC, LUSC, MESO, OV, and PAAD. We also make the comparison of PBRM1’s phosphorylation between normal and basic tumor systems as well as explore potential systems with distinctive functions in PBRM1-mediated oncogenesis. The analysis of pan-cancer offers us an outline of PBRM1’s functions in various human cancers, which could promote a comprehensive understanding of PBRM1 in tumorigenesis.

Abstract Vehicles are subject to a variety of road unevenness and random road excitations that potentially cause the vehicle to undergo a significant amount of energy dissipation, while compromising energy efficiency. This paper focuses on designing a novel piezoelectric energy harvester and aims to assess the energy harvesting potential, from the vehicle suspension, under random and pulse road excitations. To describe the energy harvesting process, a dual‐mass suspension system vibration model of a light electric logistics vehicle, equipped with a piezoelectric energy harvester, is developed. Various parameters, such as driving speed, ratio of the moment arms of the lever, and piezoelectric material cross‐sectional area, are included in the model, for basic harvesting energy. The root mean square (RMS) value of harvested power, in the case of random road, is up to 18.83 W, while the maximum respective value, in the case of pulse road, is 102.24 W and is obtained at 30 km/h. The harvested electricity is very valuable and useful, as it can be used to power automotive electrical equipment. The results of this paper provide an important reference frame for future research, related to energy harvesting from vehicle suspension.

Abstract Kidney damage initiates the deteriorating metabolic states in tubule cells that lead to the development of end‐stage renal disease (ESTD). Interleukin‐22 (IL‐22) is an effective therapeutic antidote for kidney injury via promoting kidney recovery, but little is known about the underlying molecular mechanisms. Here, we first provide evidence that IL‐22 attenuates kidney injury via metabolic reprogramming of renal tubular epithelial cells (TECs). Specifically, our data suggest that IL‐22 regulates mitochondrial function and glycolysis in damaged TECs. Further observations indicate that IL‐22 alleviates the accumulation of mitochondrial reactive oxygen species (ROS) and dysfunctional mitochondria via the induction of AMPK/AKT signaling and PFBFK3 activities. In mice, amelioration of kidney injury and necrosis and improvement of kidney functions via regulation of these metabolism relevant signaling and mitochondrial fitness of recombinant IL‐22 are certificated in cisplatin‐induced kidney damage and diabetic nephropathy (DN) animal models. Taken together, our findings unravel new mechanistic insights into protective effects of IL‐22 on kidneys and highlight the therapeutic opportunities of IL‐22 and the involved metabolic regulators in various kidney diseases.

The standard serologic markers used to diagnose hepatitis B infection include hepatitis B surface antigen (HBsAg), hepatitis B surface antibody (anti-HBs), total hepatitis B core antibody (anti-HBc), and IgM antibody to hepatitis B core antigen (IgM anti-HBc). Different markers or combinations of markers are used to identify different phases of HBV infection and determine whether a patient has acute or chronic infection or immunity due to prior infection or vaccination or is seronegative and susceptible to future infection. Isolated HBsAg seropositivity is a peculiar serological pattern that requires investigation. Herein, we present a case of an asymptomatic female without a history of liver disease or evident risk factors for hepatitis, who underwent screening for infectious disease prior to resection of basal cell carcinoma involving her eyelid. The patient’s laboratory testing showed positivity for HBsAg and the HIV 1/2 screen. To investigate, we performed serial dilutions, utilized heterophilicantibody blocking tubes, and repeated analysis using a different commercial assay (Abbott Architect i2000), all in support of a false-positive result attributed to a heterophilic antibody. Hence, we demonstrate that heterophilic antibody interference can result in isolated HBsAg positivity and recommend considering this form of interference in the differential where there is low clinical suspicion for viral infection.

Background. Papillary thyroid carcinoma (PTC) accounts for most of the proportion of thyroid cancer (TC). The objective of this study was to identify diagnostic, differentially expressed long noncoding RNAs (lncRNAs) and microRNAs (miRNAs), contributing to understanding the epigenetics mechanism of PTC. Methods. The data of lncRNA, miRNA, and mRNA were downloaded from the Cancer Genome Atlas (TCGA) dataset, followed by functional analysis of differentially expressed mRNAs. Optimal diagnostic lncRNA and miRNA biomarkers were identified via random forest. The regulatory network between optimal diagnostic lncRNA and mRNAs and optimal diagnostic miRNA and mRNAs was identified, followed by the construction of ceRNA network of lncRNA-mRNA-miRNA. Expression validation and diagnostic analysis of lncRNAs, miRNAs, and mRNAs were performed. Overexpression of ADD3-AS1 was performed in PTC-UC3 cell lines, and cell proliferation and invasion assay were used for investigating the role of ADD3-AS1 in PTC. Results. A total of 107 differentially expressed lncRNAs, 81 differentially expressed miRNAs, and 515 differentially expressed mRNAs were identified. 11 lncRNAs and 6 miRNAs were regarded as the optimal diagnostic biomarkers for PTC. The epigenetic modifications via the above diagnostic lncRNAs and miRNAs were identified, including MIR181A2HG-FOXP2-hsa-miR-146b-3p, BLACAT1/ST7-AS1-RPS6KA5-hsa-miR-34a-5p, LBX2-AS1/MIR100HG-CDHR3-hsa-miR-34a-5p, ADD3-AS1-PTPRE-hsa-miR-9-5p, ADD3-AS1-TGFBR1-hsa-miR-214-3p, LINC00506-MMRN1-hsa-miR-4709-3p, and LOC339059-STK32A-hsa-miR-199b-5p. In the functional analysis, MMRN1 and TGFBR1 were involved in cell adhesion and endothelial cell migration, respectively. Overexpression of ADD3-AS1 inhibited cell growth and invasion in PTC cell lines. Conclusion. The identified lncRNAs/miRNAs/mRNA were differentially expressed between normal and cancerous tissues. In addition, identified altered lncRNAs and miRNAs may be potential diagnostic biomarkers for PTC. Additionally, epigenetic modifications via the above lncRNAs and miRNAs may be involved in tumorigenesis of PTC.

Abstract Introduction Insects use their antennae to detect food, mates, and predators, mainly via olfactory recognition of specific volatile compounds. Honeybees also communicate, learn complex tasks, and show adaptable behavior by recognizing and responding to specific odors. However, the relationship between the electroantennogram and the passion of honeybee has not been determined. Methods We established a four‐channel maze system to detect the degree of sensitivity of the honeybee's antenna to different odors. In addition, electroantennography (EAG) signal was recorded from the right antennae of the honeybees in our experiments to explore electrophysiological responses to different volatiles. Results The olfactory sensilla on the antennae of honeybees engender distinct electrophysiological responses to different volatiles. The bees were exposed to honey, 1‐hexanol and formic acid, and EAG parameters like depolarization time, falling slope, and amplitude were measured. The EAG indicators varied significantly between honey and formic acid, indicating either “happy” or “anxious” moods. Conclusions Honeybee can express its passion by the characteristic changes of EAG parameters. We defined a preference factor (F) to quantify the preference of bees to varying concentrations of different compounds, where greater positive values indicate an increased passion. Our findings provide novel insights into the understanding of odor recognition in insects.

This paper investigates the hybrid-driven mechanism problem for Markov jump system, where both channel quantization (BCQ) and network-induced delay based on uncertain network are considered. Firstly, comparing with the traditional event-triggered scheme, a hybrid-driven mechanism is employed in networked control systems (NCSs) for the finite capacity of communication bandwidth resources and system performance in equilibrium. Then, the quantization technology is applied in the communication channel from sensor-to-controller and controller-to-actuator. The application of BCQ is for further investigation that mitigate data packet transmission rate. Thirdly, Markov jump system is modeled for the hybrid-driven mechanism and network-induced delay. By constructing the Lyapunov–Krasovskii function, a sufficient condition is derived as the stability criterion, and the controller is designed in which the nonlinear term is rewritten for simplifying the calculation. Finally, two simulation examples are provided to demonstrate the effectiveness of the proposed approach.

Background. High-risk human papillomavirus (hr-HPV) infection is a necessary cause of cervical cancer. However, other common lower genital tract microbes may increase hr-HPV infection and their related cervical cytopathy. Methods. To confirm this hypothesis, cervical brush and vaginal swab specimens were collected from 826 adult patients who were divided into the hr-HPV-positive group (254) and the negative group (572) by real-time PCR assay. Cervical specimens were tested for Ureaplasma parvum (UP), Ureaplasma urealyticum (UU), and Chlamydia trachomatis (CT) using PCR analysis. Vaginal secretion was detected for Trichomonas vaginalis (TV), Candida spp., and bacterial vaginosis (BV) with conventional assay. Results. Among hr-HPV-positive women, UP was found in 51.6%, UU in 15.4%, CT in 15.7%, Candida spp. in 11.0%, TV in 3.1%, and BV in 20.5%. In the hr-HPV-negative group, UP was positive in 36.2%, UU in 8.6%, CT in 4.0%, Candida spp. in 12.4%, TV in 0.2%, and BV in 7.0%. Multivariate logistic regression analysis with age-adjusted showed that UU (OR, 1.757), UP (OR, 1.804), CT (OR, 3.538), BV (OR, 3.020), and TV (OR, 14.109) were risk factors on hr-HPV infection (P

This paper is devoted to the co-design strategy of event-triggered scheme and dynamic output feedback controller for a class of discrete-time networked control systems (NCSs) with random time delay. An event-triggered mechanism is given to ease the information transmission. Both the sensor and controller are set with mode-dependent quantizers in the system. A Markov process is used to model the time delay which is used to describe the quantization density. By employing the Lyapunov-Krasovskii functional and linear matrix inequality (LMI), sufficient conditions are obtained for the system. A specific example is given to demonstrate the proposed approach.

BackgroundReconstruction of the aortic major branches during thoracic endovascular aortic repair is complicated because of the complex anatomic configuration and variation of the aortic arch. In situ laser fenestration has shown great potential for the revascularization of aortic branches. This study aims to evaluate the feasibility, effectiveness, and safety of in situ laser fenestration on the three branches of the aortic arch during thoracic endovascular aortic repair. Methods and ResultsBefore clinical application, the polytetrafluoroethylene and Dacron grafts were fenestrated by an 810‐nm laser system ex vivo, which did not damage the bare metal portion of the endografts and created a clean fenestration while maintaining the integrity of the endografts. In vivo, 6 anesthetized female swine survived after this operation, including stent‐graft implantation in the aortic arches, laser fenestration, and conduit implantation through the innominate arteries and the left carotid arteries. Based on the animal experiments, in situ laser fenestration during thoracic endovascular aortic repair was successively performed on 24 patients (aged 33–86 years) with aortic artery diseases (dissection type A: n=4, type B: n=7, aneurysm: n=2, mural thrombus: n=7). Fenestration of 3 aortic branches was performed in 2 (8.3%) patients. Both the left carotid artery and the left subclavian artery were fenestrated in 6 (25%) patients. Only left subclavian artery fenestration surgery was done in 16 (66.7%) patients. Among these patients, 1 fenestration was abandoned secondary to an acute takeoff of the innominate artery in a type III aortic arch. The average operative time was 137±15 minutes. The technical success rate was 95.8% (n=23). No fenestration‐related complications or neurological morbidity occurred after this operation. During a mean postoperative 10‐month follow‐up (range: 2–17 months), 1 patient died of severe pneumonia, and all the left subclavian artery and carotid artery stents were patent with no fenestration‐related endoleaks upon computed tomography angiography images. ConclusionsIn situ laser fenestration is a feasible, effective, rapid, repeatable, and safe option for the reconstruction of aortic arch during thoracic endovascular aortic repair, which might be available to revascularize the 3 branches. However, follow‐up periods should be extended to evaluate the robustness of this technique.

In order to diagnose sensor fault of aeroengine more quickly and accurately, a double redundancy diagnosis approach based on Weighted Online Sequential Extreme Learning Machine (WOS-ELM) is proposed in this paper. WOS-ELM, which assigns different weights to old and new data, implements weighted dealing with the input data to get more precise training models. The proposed approach contains two series of diagnosis models, that is, spatial model and time model. The application of double redundancy based on spatial and time redundancy can in real time detect the hard fault and soft fault much earlier. The trouble-free or reconstructed time redundancy model can be utilized to update the training model and make it be consistent with the practical operation mode of the aeroengine. Simulation results illustrate the effectiveness and feasibility of the proposed method.

Aims. Stroke is a leading cause of morbidity and mortality. This study aimed to determine whether nicotinamide administration could improve remyelination after stroke and reveal the underlying mechanism. Methods. Adult male C57BL/6J mice were intraperitoneally (i.p.) administered with nicotinamide (200 mg/kg, daily) or saline after stroke induced by photothrombotic occlusion of the middle cerebral artery. FK866 (3 mg/kg, daily, bis in die), an inhibitor of NAMPT, and ANA-12 (0.5 mg/kg, daily), an antagonist of tropomyosin-related kinase B (TrkB), were administered intraperitoneally 1 h before nicotinamide administration. Functional recovery, MRI, and histological assessment were performed after stroke at different time points. Results. The nicotinamide-treated mice showed significantly lower infarct area 7 d after stroke induction and significantly higher fractional anisotropy (FA) in the ipsilesional internal capsule (IC) 14 d after stroke induction than the other groups. Higher levels of NAD+, BDNF, and remyelination markers were observed in the nicotinamide-treated group. FK866 administration reduced NAD+ and BDNF levels in the nicotinamide-treated group. ANA-12 administration impaired the recovery from stroke with no effect on NAD+ and BDNF levels. Furthermore, lesser functional deficits were observed in the nicotinamide-treated group than in the control group. Conclusions. Nicotinamide administration improves remyelination after stroke via the NAD+/BDNF/TrkB pathway.
