The effectively early detection and determination of disease progression of gastric cancer (GC) are still required. An emerging demand for identifying the novel targets adherent to cancer cells has been still challenged since those valuable profilings not only could act as for early gastric tumor discovery but also being potential therapeutic views. We have retrospectively analyzed GC biopsies to identify those specific target peptides in association with disease progression. We have detected the polypeptide by liquid mass technology initiated BIO-HIGH innovational assay technology for tumor-specific target peptide identification. We have validated the accessibility and feasibility of multiple target cytotoxic T-lymphocyte for the assessment of potential molecular markers by equally comparing the frequencies of tumor peptides' loci identified in 138 GC patients. The aim was to separate peripheral blood lymphocytes by density gradient centrifugation and use specific target peptides in in vitro culture of lymphocytes. The Cell Counting Kit-8 assay was set up to prove the lymphocytes' proliferation stimulated by identified peptides. Both of GC-specific peptide and shared peptide were detected in the peripheral blood, and the frequencies and quantities were correlated with disease status and cancer differentiation, in which BHGa1510 (78%), BHGa1310 (66%), BHGa0910 (57%), BHGa0310 (54%), BHGa0210 (40%), BHGa0810 (35%), BHGa0110 (33%), and BHGa1410 (30%) were apparently scoped out as high-frequency (HF) peptides could be potentially specific tumor markers. Moreover, BHGa1410 was significantly associated with cancer progression, and BHGa0910 and BHGa0210 were significantly associated with TNM stage. The IHC data have shown that both the HF BHGa1510 and HF BHGa1310 were expressions by 100% in contrast with paracancerous tissues of 40% (p < 0.05) and 33%, respectively (p < 0.05). Those specific peptide pools could be valued in assessment of advanced tumor and differential status in GC patients.

Energy conversion plays an important role in the metabolism of photosynthetic organisms. Improving energy transformation by promoting a proton gradient has been a great challenge for a long time. In the present study, we realize a directional proton migration through the construction of oriented bacteriorhodopsin (BR) microcapsules coated by FoF1-ATPase molecular motors through layer-by-layer (LBL) assembly. The changes in the conformation of BR under illumination lead to proton transfer in a radial direction, which generates a higher proton gradient to drive the synthesis of adenosine triphosphate (ATP) by FoF1-ATPase. Furthermore, to promote the photosynthetic activity, optically matched quantum dots were introduced into the artificial coassembly system of BR and FoF1-ATPase. Such a design creates a new path for the use of light energy.

BACKGROUND: The Apolygus lucorum is one of the most destructive insect pests in China with a wide range of host plants. Interaction of A. lucorum with surrounding environment heavily relies on chemical communication. Deorphanization of receptors involved in odors detection elevates our understanding of the olfactory system of this pest and may help to develop a chemical ecology‐based control strategy. RESULTS: AlucOR80, an odorant receptor (OR) in A. lucorum was newly cloned. Gene expression analysis showed that this receptor was mainly expressed in the antennae and head of both sexes but with a male bias. The Xenopus oocytes heterologous expression system coupled with the two‐electrode voltage‐clamp (TEVC) recording revealed that AlucOR80 was tuned to 21 selected compounds. Furthermore, electroantennogram (EAG) tests confirmed that all 21 ligands of AlucOR80 were electrophysiologically active in antennae of both sexes. Behavioral trials in a three‐cage olfactometer indicated that 16 compounds were behaviorally active, amongst which, 12 components were attractants and four components were repellents for adults of both sexes. Butyl butyrate and Dimethyl disulfide (DMDS) were the strongest attractive and repellant compounds, respectively. Importantly, we found the repellency of 1, 8‐Cineole, S‐(−)‐cis‐Verbenol and (1S)–(1)‐beta‐Pinene against adults of A. lucorum. CONCLUSION: Although AlucOR80 is a general OR, may play important role in the olfactory perception of A. lucorum. Screening of AlucOR80 ligands by behavioral assay provided valuable insights by which olfactory‐based management approaches could be developed by utilizing the behaviorally active components as attractants or repellents. © 2019 Society of Chemical Industry

Macrophages hold great potential in cancer drug delivery because they can sense chemotactic cues and home to tumors with high efficiency. However, it remains a challenge to load large amounts of therapeutics into macrophages without compromising cell functions. This study reports a silica-based drug nanocapsule approach to solve this issue. The nanocapsule consists of a drug-silica complex filling and a solid silica sheath, and it is designed to minimally release drug molecules in the early hours of cell entry. While taken up by macrophages at high rates, the nanocapsules minimally affect cell migration in the first 6-12 h, buying time for macrophages to home to tumors and release drugs in situ. In particular, it is shown that doxorubicin (Dox) as a representative drug can be loaded into macrophages up to 16.6 pg per cell using this approach. When tested in a U87MG xenograft model, intravenously (i.v.) injected Dox-laden macrophages show comparable tumor accumulation as untreated macrophages. Therapy leads to efficient tumor growth suppression, while causing little systematic toxicity. This study suggests a new cell platform for selective drug delivery, which can be readily extended to the treatment of other types of diseases.

Recently, we demonstrated the potential of a [(18) F]-trimethylammonium BODIPY dye for cardiac imaging. This is the first example of the use of the [(18) F]-ammonium BODIPY dye for positron emission tomography (PET) myocardial perfusion imaging (MPI). In this report, we extend our study to other ammonium BODIPY dyes with different nitrogen substituents. These novel ammonium BODIPY dyes were successfully prepared and radiolabeled by the SnCl4 -assisted (18) F-(19) F isotopic exchange method. The microPET results and the biodistribution data reveal that nitrogen substituent changes have a significant effect on the in vivo and pharmacological properties of the tracers. Of the novel [(18) F]-ammonium BODIPY dyes prepared in this work, the [(18) F]-dimethylethylammonium BODIPY is superior in terms of myocardium uptake and PET imaging contrast. These results support our hypothesis that the ammonium BODIPY dyes have a great potential for use as PET/optical dual-modality MPI probes.

Fibroblast activation protein (FAP) is highly expressed in the tumor-associated fibroblasts (TAFs) of most human epithelial cancers. FAP plays a critical role in tumorigenesis and cancer progression, which makes it a promising target for novel anticancer therapy. However, mere abrogation of FAP enzymatic activity by small molecules is not very effective in inhibiting tumor growth. In this study, we have evaluated a novel immune-based approach to specifically deplete FAP-expressing TAFs in a mouse 4T1 metastatic breast cancer model. Depletion of FAP-positive stromal cells by FAP-targeting immunotoxin αFAP-PE38 altered levels of various growth factors, cytokines, chemokines and matrix metalloproteinases, decreased the recruitment of tumor-infiltrating immune cells in the tumor microenvironment and suppressed tumor growth. In addition, combined treatment with αFAP-PE38 and paclitaxel potently inhibited tumor growth in vivo. Our findings highlight the potential use of immunotoxin αFAP-PE38 to deplete FAP-expressing TAFs and thus provide a rationale for the use of this immunotoxin in cancer therapy.

A high-performance plasmonic biosensor based on two-dimensional (2D) Ag nanowell crystals is rationally designed and fabricated by colloidal litho­graphy. The crystals act as 2D metallic gratings that directly couple incident photons to surface plasmon polaritons (SPPs). The nanowell sizes are properly tuned to enable coupling between the SPPs and Raleigh anomalies, which proves to greatly contribute to sharp reflectance dips. Other geometric parameters including the nanowell depth and the lattice constant are respectively adjusted to optimize the lineshape and sensitivity. These inferences and experimental results are verified by finite-difference time-domain (FDTD) calculations. In this study, the fabricated crystal with 670 nm lattice constant, 483 nm nanowell size, and 40 nm nanowell depth possesses the highest refractive index sensitivity (RIS) (623.7 nm RIU−1), the maximum figure of merit (FOM) (55.2 RIU−1), and a very high value of full height/full width at half-maximum (FH/FWHM) (28.3 ×10−3 nm−1). As a proof-of-concept, we adopt the well-studied antigen-antibody couple as a model system to illustrate the potential of the nanowell crystal in a quantitative analytical bioassay. The excellent sensing performance, together with the compact structure and the simple read-out apparatus, suggest that the 2D Ag nanowell crystal is an excellent candidate for a label-free biosensing platform.

A radiolabeling method for bioconjugation based on the Diels-Alder reaction between 2,6-diaryl-tetrazines, such as (III), and a 18F-labeled trans-cyclooctene analogue of (II) is described.

Bisbinaphthyl-based macrocycles are found to carry out enantioselective fluorescent recognition of α-amino acid derivatives. It is observed that one enantiomer of a N -protected phenyl glycine can increase the fluorescence intensity of the binaphthyl fluorophores by over 4-fold but the other enantiomer does not cause much fluorescence enhancement. This highly enantioselective fluorescent response makes the binaphthyl macrocycles practically useful for the enantioselective fluorescent recognition of the amino acid substrate.
