High-fat diet (HFD) feeding for mice is commonly used to model obesity. However, conflicting results have been reported on the relationship between HFD and bone mass. In this systematic review and meta-analysis, we synthesized data from 80 articles to determine the alterations in cortical and trabecular bone mass of femur, tibia, and vertebrae in C57BL/6 mice after HFD. Overall, we detected decreased trabecular bone mass as well as deteriorated architecture, in femur and tibia of HFD treated mice. The vertebral trabecula was also impaired, possibly due to its reshaping into a more fragmentized pattern. In addition, pooled cortical thickness declined in femur, tibia, and vertebrae. Combined with changes in other cortical parameters, HFD could lead to a larger femoral bone marrow cavity, and a thinner and more fragile cortex. Moreover, we conducted subgroup analyses to explore the influence of mice's sex and age as well as HFD's ingredients and intervention period. Based on our data, male mice or mice aged 6-12 weeks old are relatively susceptible to HFD. HFD with50% of energy from fats and intervention time of 10 weeks to 5 months are more likely to induce skeletal alterations. Altogether, these findings supported HFD as an appropriate model for obesity-associated bone loss and can guide future studies.

Background Concerns about breast cancer had become the most dangerous cancer to women over the world, more and more anti‐cancer agents are developed to treat this malignancy. Pharmorubicin is a cytotoxic drug, widely used in the treatment of breast cancer, but its role is limited because of chemoresistance produced by cells. This study focused on exploring the influence of autophagy on the resistance of pharmorubicin in breast cancer cells. Methods The cell survival of breast cancer cells was detected by MTT. The mRNA expression of heme oxygenase‐1 (HO‐1) was tested by qRT‐PCR. The protein expression of HO‐1, autophagic proteins (LC3‐I,LC3‐II and Beclin‐1), PI3K and Akt was detected by Western blot. Cell autophagy was examined by Cyto‐ID Autophagy Detection Kit. Results After being treated with pharmorubicin, the expression of HO‐1 and autophagy related proteins was significantly enhanced, but the cell survival ratio in the two cell lines decreased. After autophagy was inhibited, HO‐1 expression in two cells was down‐regulated. When pharmorubicin‐resistant cells were transfected with si‐HO‐1, the cell survival decreased and the protein expression of HO‐1, autophagic proteins (LC3‐II/LC3‐I and Beclin‐1) as well as autophagy were all down‐regulated, while in pharmorubicin‐resistant cells transfected with pcDNA3.1‐HO‐1, the results were reverse. When the PI3K or Akt was inhibited, PI3K, p‐Akt, HO‐1, autophagic proteins and autophagy were decreased remarkably. Conclusion It was proved that HO‐1 induction mediated chemoresistance of pharmorubicin in breast cancer cells by promoting autophagy via PI3K/Akt pathway.

Citations in scientific literature are important both for tracking the historical development of scientific ideas and for forecasting research trends. However, the diffusion mechanisms underlying the citation process remain poorly understood, despite the frequent and longstanding use of citation counts for assessment purposes within the scientific community. Here, we extend the study of citation dynamics to a more general diffusion process to understand how citation growth associates with different diffusion patterns. Using a classic diffusion model, we quantify and illustrate specific diffusion mechanisms which have been proven to exert a significant impact on the growth and decay of citation counts. Experiments reveal a positive relation between the “low p and low q” pattern and high scientific impact. A sharp citation peak produced by rapid change of citation counts, however, has a negative effect on future impact. In addition, we have suggested a simple indicator, saturation level, to roughly estimate an individual article's current stage in the life cycle and its potential to attract future attention. The proposed approach can also be extended to higher levels of aggregation (e.g., individual scientists, journals, institutions), providing further insights into the practice of scientific evaluation.

The Buqingshan Tectonic Melange Belt in the south margin of the East Kunlun Orogen, located in west section of Buqingshan–A'nyemaqen Suture Zone, is one of the key areas to understand continental tectonics and continental dynamics of China. This paper reports zircon U–Pb dating results and geochemistry of the Manite granodiorite (rock mass) in the Buqingshan Tectonic Melange Belt. Zircons from the granodiorite show oscillatory zoning structures and relatively high Th/U ratios, indicating that they are magmatic zircons. Zircon LA–ICP–MS U–Pb dating for the Manite granodiorite yields ages of 487 ± 11 Ma (MSWD = 2.3) and 479 ± 2 Ma (MSWD = 0.56), implying that the Manite granodiorite were formed in the Late Cambrian to Early Ordovician. Geochemical analyses show that the rocks have high contents of SiO2 (66.06 wt.%–68.60 wt.%) and Al2O3 (14.84 wt.%–16.54 wt.%), and low alkaline (6.17 wt.%–7.43 wt.%), belonging to the middle-K calc-alkaline series. The A/CNK (Al2O3/(CaO + Na2O + K2O)) ratios are 1.02–1.15, indicating that the granodiorite is weakly peraluminous. The contents of rare earth elements (REEs) are lower (89.64–130.41 ppm), with weakly negative to weakly positive Eu anomalies (δEu = 0.83–1.12). The primitive mantle-normalized trace elements are characterized by evidently negative anomalies of Nb, Ta, P, Hf, Ti, etc., and positive anomalies of Th, La, Nd, Zr, Eu, etc. Moreover, the rocks show features of typical I-type granite. This leads us to conclude that the Manite granodiorites is derived from the partial melting of crust and formed in an island-arc environment. Combined with previous studies, we believe that the subduction of the Proto-Tethyan Ocean in the Buqingshan area was ongoing during 487 to 479 Ma and formed island-arc-type granites represented by the Late Cambrian to Early Ordovician Manite granodiorite. This study focuses on the zircon U–Pb geochronology and geochemistry of the Bairiqiete granodiorite intrusion (rock mass) from the Buqingshan tectonic melange belt in the southern margin of East Kunlun. The results show that the zircons are characterized by internal oscillatory zoning and high Th/U (0.14–0.80), indicative of an igneous origin. LA–ICP–MS U–Pb dating of zircons from the Bairiqiete granodiorite yielded an age of 439.0 ± 1.9 Ma (MSWD = 0.34), implying that the Bairiqiete granodiorite formed in the early Silurian. Geochemical analyses show that the rocks are medium-K calc-alkaline, relatively high in Al2O3 (14.57–18.34 wt%) and metaluminous to weakly peraluminous. Rare-earth elements have low concentrations (45.49–168.31 ppm) and incline rightward with weak negative to weak positive Eu anomalies (δ5Eu = 0.64–1.34). Trace-element geochemistry is characterized by negative anomalies of Nb, Ta, Zr, Hf and Ti and positive anomalies of Rb, Th and Ba. Moreover, the rocks have similar geochemical features with adakites. The Bairiqiete granodiorite appears to have a continental crust source and formed in a subduction-related island-arc setting. The Bairiqiete granodiorite was formed due to partial melting of the lower crust and suggests subduction in the Buqingshan area of the Proto-Tethys Ocean.

Annexin 1, a glucocorticoid (GC)-inducible protein, can play an important role via formyl peptide receptor like 1 (FPR2/ALX, also known as FPRL1) in inflammatory pain modulation. The aim of this review is to analyze different lines of evidence for the role of ANXA1 with different mechanisms on inflammatory pain and describe the profile of ANXA1 as a potential analgesic. A Medline (PUBMED) search using the terms 'Annexin 1 distribution OR expression, FPR2/ALX distribution OR expression, Annexin 1 AND pain, Annexin 1 AND FPR2/ALX AND pain' was performed. Articles with a publication date up to Nov. 1st, 2012 were included. The antinociception of ANXA1 has been evaluated in diverse pain models. It has been suggested that ANXA1 may exerts its action via: (1) inhibiting vital cytokines involved in pain transmission, (2) inhibiting neutrophil accumulation through preventing transendothelial migration via an interaction with formyl peptide receptors, (3) facilitating tonic opioid release from neutrophil in inflammatory site, (4) interrupting the peripheral nociceptive transmission by suppressing neuronal excitability. In general, ANXA1 is a potential mediator for anti-nociception and the role with its receptor constitute attractive targets for developing anesthesia and analgesic drugs, and their interaction may prove to be a useful strategy to treat inflammatory pain.

This study aimed to compare three most widely used surgeries for experimental lumbar spinal drug delivery in rats. The comparison consisted of assessing the effects of the surgeries and evaluating the deficits produced by these three methods. Sprague Dawley rats underwent acute needle puncture, chronic catheterization via laminectomy or nonlaminectomized catheterization. Body weight changes were measured, animals' general and neurological conditions were observed after surgeries, and motor function was examined by Rota Rod test both prior to and post surgery. Furthermore, nociceptive tests were performed to assess the animals' nociception; hematoxylin, and eosin staining of lumbar spinal cord tissue was performed to evaluate local inflammation caused by surgeries; and both lidocaine paralysis detection; and toluidine blue dye assay were used to confirm the exact location of the catheter. Both needle puncture and catheterization via laminectomy had relatively low success rate of surgery and induced various neurological signs; more severe motor dysfunction, hyperalgesia, allodynia, and local inflammation. Nonlaminectomized catheterization had a higher success rate of surgery, and induced only mild agitation, slight cerebral spinal fluid leakage, mild sensory and motor abnormalities, and minimum pathology in the lumbar spinal cord. The nonlaminectomized catheterization used in this study induces a phenotype of less detectable effects on the animal's behavior and is well-tolerated compared to the acute needle puncture and laminectomized catheterization that are widely used in the literature. Nonlaminectomized catheterization is a safe, accurate and effective way for lumbar drug delivery in rats.

The molecular mechanisms of signal transduction of plants in response to infection by Verticillium dahliae (VD) are not well understood. We previously showed that NO may act as an upstream signalling molecule to trigger the depolymerization of cortical microtubules in Arabidopsis. In the present study, we used the wild-type, and atrbohD and atrbohF mutants of Arabidopsis to explore the mechanisms of action of H(2)O(2) signals and the dynamic microtubule cytoskeleton in defence responses. We demonstrated that H(2)O(2) may also act as an upstream signalling molecule to regulate cortical microtubule depolymerization. The depolymerization of the cortical microtubules played a functional role in the signalling pathway to mediate the expression of defence genes. The results indicate that H(2)O(2) modulates the dynamic microtubule cytoskeleton to trigger the expression of defence genes against V. dahliae toxins (VD-toxins) in Arabidopsis.

The molecular mechanisms of signal transduction of plants in response to Verticillium dahliae (VD) are not known. Here, we show that Arabidopsis reacts to VD-toxins with a rapid burst of nitric oxide (NO) and cortical microtubule destabilization. VD-toxins treatment triggered a disruption of cortical microtubules network. This disruption can be influenced by NO production. However, cortical microtubule disruptions were not involved in regulating the NO production. The results indicated that NO may act as an upstream signalling molecule to trigger the depolymerization of cortical microtubule. Cortical microtubules may act as a target of NO signal and as a sensor to mediate the activation of PR-1 gene expression. These results suggested that NO production and cortical microtubule dynamics appeared to be parts of the important signalling system and are involved in the defence mechanisms to VD-toxins in Arabidopsis.

Here we show direct polymer growth on hydrogen-terminated silicon (Si-H) as a one-step process. Si-H is immersed in a heated solution of an initiator, a monomer, and a crosslinker. The resulting polymer films are 2-15 nm thick, the thickness can be tuned by varying reaction conditions, and the polymer films are robust. Thinner films are obtained in polymer growth without crosslinker. Radicals produced by the initiator and in growing polymer chains may pluck H atoms from Si-H to make reactive dangling bonds. These in turn appear to react with monomers and crosslinkers. Crosslinkers tethered through one double bond may be grafted into polymer chains growing in solution. Surface reactivity is demonstrated by optical ellipsometry, X-ray photoelectron spectroscopy, atomic force microscopy, and wetting.
