In order to enhance wear resistance of cold work molds, WC−10Co4Cr coating was fabricated on Cr12MoV steel by laser cladding. The morphologies, chemical compositions, and phases of obtained coatings were analyzed using a scanning electron microscopy (SEM), energy disperse spectroscopy, and X−ray diffraction, respectively. The effect of laser power on the tribological performance was analyzed using a ball−on−plate friction machine, and the wear mechanism was also discussed. The results show that the WC−10Co4Cr coating is composed of WC and Co6W6C phases, and the average hardness of coating cross−sections fabricated at the laser power of 1200, 1500, and 1800 W was 1296, 1375, and 1262 HV0.5, respectively, in which that fabricated at the laser power of 1500 W is the highest among the three kinds of coatings. The average coefficients of friction of coatings fabricated at the laser power of 1200, 1500, and 1800 W are 0.61, 0.52, and 0.59, respectively; and the corresponding wear rates are 64.38, 35.38, and 123.92 μm3•N−1•mm−1, respectively, showing that the coating fabricated at the laser power of 1500 W has best friction reduction and wear resistance. Zanubrutinib is a highly selective, potent, orally available, targeted covalent inhibitor (TCI) of Bruton's tyrosine kinase (BTK). This work investigated the in vitro drug metabolism and transport of zanubrutinib, and its potential for clinical drug–drug interactions (DDIs). Phenotyping studies indicated cytochrome P450 (CYP) 3A are the major CYP isoform responsible for zanubrutinib metabolism, which was confirmed by a clinical DDI study with itraconazole and rifampin. Zanubrutinib showed mild reversible inhibition with half maximal inhibitory concentration (IC50) of 4.03, 5.69, and 7.80 μM for CYP2C8, CYP2C9, and CYP2C19, respectively. Data in human hepatocytes disclosed induction potential for CYP3A4, CYP2B6, and CYP2C enzymes. Transport assays demonstrated that zanubrutinib is not a substrate of human breast cancer resistance protein (BCRP), organic anion transporting polypeptide (OATP)1B1/1B3, organic cation transporter (OCT)2, or organic anion transporter (OAT)1/3 but is a potential substrate of the efflux transporter P-glycoprotein (P-gp). Additionally, zanubrutinib is neither an inhibitor of P-gp at concentrations up to 10.0 μM nor an inhibitor of BCRP, OATP1B1, OATP1B3, OAT1, and OAT3 at concentrations up to 5.0 μM. The in vitro results with CYPs and transporters were correlated with the available clinical DDIs using basic models and mechanistic static models. Zanubrutinib is not likely to be involved in transporter-mediated DDIs. CYP3A inhibitors and inducers may impact systemic exposure of zanubrutinib. In natural images, it remains challenging to estimate dissimilarities between image elements for scene representation due to gradual variations of illuminations, textures or clutters. To tackle this problem, we utilise a path-based bottleneck analysis method that captures the semantic information between image elements to measure the dissimilarity. By integrating both the spatial continuity and feature consistency into the understanding of the semantic information, we detect the bottlenecks on the proposed double-S path to define the bottleneck distance, which demonstrates a favourable capability of grouping image elements that follow a similar pattern and separating different ones. In the experiments, the method is proved to be robust to noises and invariant to changing illumination and arbitrary scales in natural images. Achieving reliable and energy-saving broadcast is the main issue discussed in this study. Firstly, a multi-hop broadcast model is introduced, and the energy consumption and reliability of the network are defined in this study. Then five broadcast-based automatic repeat request (ARQ) protocols (automatic retransmission N times, stop-and-wait end-to-end-ARQ, stop-and-wait hop-by-hop-ARQ, stop-and-wait hop-by-hop oriented implicit ARQ, stop-and-wait hop-by-hop implicit and explicit ARQ) are proposed to realise the reliable transmission. Secondly, based on five protocols, the relationship between the system reliability and the number of retransmissions of the root node is established analytically, and the maximum number of retransmissions is solved under the required system reliability. Thirdly, the energy consumptions of the root node are calculated, and then the analytical expressions of the minimum system energy consumption are obtained separately according to five broadcast-based retransmission mechanisms. Finally, the relationships between the maximum number of retransmissions, energy consumption and reliability, the broadcast probability and feedback probability are discussed in the numerical analysis. The measurable electric field of the partial discharge ultra-high-frequency (PD UHF) detection system calibrated by a gigahertz transverse electromagnetic cell (GTEM) has been used as a key indicator to evaluate the performance of the detection system and regarded as the electric power standard. In this study, the sweep frequency and pulse signal were injected to the GTEM cell, and then the waveform and amplitude of the calibrated field strength were calculated by using the transfer function and voltage response of the monopole antennas. In the case of sweep frequency, comparing the measured result of the Narda field strength metre and the calculated result of monopole antennas, the error was found to be <8%, which verifies the correctness of the calibration of the monopole antennas to the steady-state electric field in the GTEM. In the case of pulse injection, comparing the simulation result of computer simulation technology and the calculated result of monopole antennas, the error of the peak of the field strength was found to be 1.7%, which verifies the correctness of the calibration of the monopole antennas to the pulse electric field in the GTEM. Advances in metering and two-way communication technologies foster the studies of Home Energy Management System (HEMS). This study proposes a new HEMS, which optimally schedules the distributed residential energy resources (DRERs) in a smart home environment with varying electricity tariff and high solar penetrations. The uncertainties of solar power output are captured by using Monte Carlo sampling technique to generate multiple solar output scenarios based on the probabilistic solar radiation model. The homeowner's rigid and elastic restrictions on the operations of the automatically controlled household appliances are modelled. Based on this, an optimal DRER scheduling model is proposed to minimise the home operation cost while taking into account the homeowner's requirements. A new heuristic optimisation algorithm recently proposed by the authors, i.e. natural aggregation algorithm, is used to solve the proposed model. This investigation aims to improve the ultradrawing and ultimate tensile properties of ultrahigh molecular weight polyethylene (UHMWPE) fibers by incorporating small amounts of functionalized activated nanocarbon particles with a wide range of specific surface areas (ca. 100–1,400 m2/g) during gel spinning processes of UHMWPE fibers. The ultradrawing, ultimate tensile, orientation properties, and “microfibril” characteristics of UHMWPE/functionalized activated nanocarbon fibers was discovered to improve considerably with the increase in specific surface areas of functionalized activated nanocarbon. An extraordinary high ultimate tensile strength at 95.8 g/d was obtained for the best prepared UHMWPE/functionalized activated nanocarbon drawn fiber. This value is the highest value ever reported for one‐stage drawn UHMWPE fibers and is about 2.9 times that of the UHMWPE drawn fiber prepared in this study. In addition to thermal, ultimate tensile, and orientation factor properties of as‐prepared and/or drawn UHMWPE/functionalized activated nanocarbon fibers, specific surface area, Fourier transform infrared, and morphological analyses of original and functionalized activated nanocarbons were performed to comprehend the considerably improved ultradrawing, ultimate tensile properties, and microfibril characteristics of the UHMWPE/functionalized activated nanocarbon fibers. A sensorless current-sharing strategy for two-phase and multiphase input-parallel output-parallel (IPOP) DC-DC converters is proposed here. A dual-active-bridge (DAB) DC-DC converter is chosen as the basic DC-DC converter. With this strategy in two-phase IPOP DAB DC-DC converters, the parameter mismatches between phases are estimated by perturbing the duty cycle in one phase and measuring the changes of duty cycles in both phases, then the duty cycles are adjusted to compensate the mismatches, thus achieving current sharing without current sensor. With this strategy in multiphase IPOP DAB DC-DC converters, by perturbing the duty cycles in (N - 1) out of N phases in turn and measuring the changes of duty cycles, respectively, the parameter mismatches among phases are estimated. According to the mismatches, a set of variables, which are proportional to the per-phase output currents, are calculated. Then with a current-sharing regulator, parameter mismatches are compensated, thus achieving current sharing without current sensor. Floral nectaries are closely associated with biotic pollination, and the nectar produced by corolla nectaries is generally enclosed in floral structures. Although some Swertia spp. (Gentianaceae), including S. bimaculata, evolved a peculiar form of corolla nectaries (known as "gland patches") arranged in a conspicuous ring on the rotate corolla and that completely expose their nectar, little is known about the pollination of these plants. Two hypotheses were made concerning the possible effects of gland patches: visual attraction and visitor manipulation. The floral traits, mating system, and insect pollination of S. bimaculata were examined, and the pollination effects of gland patches were evaluated. A comparative study was made using Swertia kouitchensis, a species with fimbriate nectaries. Swertia bimaculata flowers were protandrous, with obvious stamen movement leading to herkogamy in the female phase and to a significant reduction in nectary-anther distance. The species is strongly entomophilous and facultatively xenogamous. The daily reward provided per flower decreased significantly after the male phase. The most effective pollinators were large dipterans, and the visiting proportion of Diptera was significantly higher in S. bimaculata than in S. kouitchensis. Most visitors performed "circling behavior" in S. bimaculata flowers. Removing or blocking the nectaries caused no reduction in visiting frequency but a significant reduction in visit duration, interrupting the circling behavior. The circling behavior was encouraged by nectar abundance and promoted pollen dispersal. Visitor species with small body size had little chance to contact the anthers or stigma, revealing a filtration effect exerted by the floral design. These results rejected the "visual attraction" hypothesis and supported the "visitor manipulation" hypothesis. The nectary whorl within a flower acted like a ring-shaped track that urged nectar foragers to circle on the corolla, making pollination in S. bimaculata flowers more orderly and selective than that in classically generalist flowers. Premise of the study: Apocarpous plants possess carpels that are separated in the gynoecium. Extragynoecial compita, commonly occurring in basal angiosperms, have been proposed to have the potential to increase offspring quantity in apocarpous species through the intercarpellary growth of pollen tubes. To date, the impact of an extragynoecial compitum on fruit or seed set has not been studied in any species. This study investigated the pollen tube pathway between adjacent carpels and its contribution to fruit set in Schisandra sphenanthera. Methods: We investigated the fruit set ratio in the field and collected hundreds of gynoecia at their full flowering stage. Pollinated carpel ratio and pollen tube pathway observations were performed using fluorescence optics. Key results: Pollen grains germinated and tubes extended along the pseudostyle surface. Some of them turned and entered the ovules at the end of the stigmatic crest, whereas others subsequently grew into neighboring carpels through promontory connections located at the base of the unfused carpels. No tubes were found growing on the surface of the receptacle. More than 24 carpels could be fertilized by pollen tubes from one carpel through hand pollination. The pollinated carpel ratio was significantly lower than the fruit set ratio under natural conditions. Conclusions: Pollen tubes from one carpel can easily cross in the extragynoecial compitum between the adjacent carpels of S. sphenanthera, and this intercarpellary growth of pollen tubes can significantly increase the fruit set of apocarpous species, at least in S. sphenanthera. Recent studies have shown a critical function for the ubiquitin-proteasome system (UPS) in regulating the signalling network for DNA damage responses and DNA repair. To search for new UPS targets in the DNA damage signalling pathway, we have carried out a non-biased assay to identify fast-turnover proteins induced by various types of genotoxic stress. This endeavour led to the identification of Rad17 as a protein exhibiting a distinctive pattern of upregulation followed by subsequent degradation after exposure to UV radiation in human primary cells. Our characterization showed that UV-induced Rad17 oscillation is mediated by Cdh1/APC, a ubiquitin-protein ligase. Studies using a degradation-resistant Rad17 mutant demonstrated that Rad17 stabilization prevents the termination of checkpoint signalling, which in turn attenuates the cellular re-entry into cell-cycle progression. The findings provide an insight into how the proteolysis of Rad17 by Cdh1/APC regulates the termination of checkpoint signalling and the recovery from genotoxic stress. [ABSTRACT FROM AUTHOR]

In response to DNA damage or replication block, cells activate a battery of checkpoint signaling cascades to control cell cycle progression and elicit DNA repair in order to maintain genomic stability and integrity. Identified as a homolog of its fission yeast counterpart, human Rad9 was proposed to form a Rad9-Hus1-Rad1 protein complex to mediate checkpoint signals. However, the precise function of Rad9 in the process of checkpoint activation is not fully understood. Using the RNA interference technique, we investigated the role of Rad9 in the genotoxic stress-induced activation of S-phase checkpoint and the maintenance of chromosomal stability. We found that Rad9 knockdown reduced the phosphorylation of Rad17, Chk1 and Smc1 in response to DNA replication block and certain types of DNA damage. Immunofluorescence studies showed that the removal of Rad9 disrupted the foci formation of phosphorylated Chk1, but not ATR. Moreover, Rad9 knockdown resulted in radioresistant DNA synthesis and reduced cell viability under replication stress. Finally, removal of Rad9 by RNAi led to increased accumulation of spontaneous chromosomal aberrations. Taken together, these results suggest a critical and specific role of Rad9 in the activation of S-phase checkpoint and the maintenance of chromosome stability. [ABSTRACT FROM AUTHOR]

The ataxia-telangiectasia mutated and rad3-related (ATR) kinase orchestrates cellular responses to DNA damage and replication stress. Complete loss of ATR function leads to chromosomal instability and cell death. However, heterozygous ATR mutations are found in human cancers with microsatellite instability, suggesting that ATR haploinsufficiency contributes to tumorigenesis. To test this possibility, we generated human cell line and mouse model systems in which a single ATR allele was inactivated on a mismatch repair (MMR)-deficient background. Monoallelic ATR gene targeting in MLH1-deficient HCT 116 colon carcinoma cells resulted in hypersensitivity to genotoxic stress accompanied by dramatic increases in fragile site instability, and chromosomal amplifications and rearrangements. The ATR+/- HCT 116 cells also displayed compromised activation of Chk1, an important downstream target for ATR. In complementary studies, we demonstrated that mice bearing the same Atr+/-/Mlh1-/- genotype were highly prone to both embryonic lethality and early tumor development. These results demonstrate that MMR proteins and ATR functionally interact during the cellular response to genotoxic stress, and that ATR serves as a haploinsufficient tumor suppressor in MMR-deficient cells. We have cloned from a cDNA library of neuronal derivatives of retinoic-acid-induced embryonic carcinoma ceils a nuclear receptor that may be involved in the control of late neurogenesis and early neuronal differentiation. The receptor which is practically identical in sequence with germ cell nuclear factor, has been designated neuronal cell nuclear factor (NCNF). NCNF is exclusively expressed in the neuronal derivatives of PCC7-Mz1 cells, with the expression beginning within hours of exposure to retinoic acid. In the developing mouse brain, NCNF is expressed in the marginal zones of the neuroepithelium which are known to contain young postmitotic neurons. NCNF binds to the DR0 sequence thereby silencing transcription. Because NCNF does not recognize hormone response elements of other nuclear receptors tested and does not heterodirnerize with these, it probably binds exclusively as a homodimer.
