Chrysin (CHR) is a flavonoid with extensive pharmacological activity. The molecular formula of CHR is C15H10O4. CHR is reported to have antioxidative, antitumour and antiviral functions. To evaluate its potential function as a vaccine adjuvant, we prepared a melanoma vaccine using a soluble protein extract of B16F10 melanoma cells as antigen and CHR as an adjuvant. The melanoma model was developed after two immunizations, and it was discovered that combining B16F10 soluble protein antigen‐mixed CHR vaccine could inhibit tumour growth in the mouse model, and the overall survival rate was higher than that of the B16F10 antigen vaccine alone. In vivo and in vitro experiments were conducted to determine whether CHR functioned as an adjuvant by activating antigen‐presenting cells (APCs). We discovered that CHR activated APCs both in vivo and in vitro and may enhance Th1 cell function by activating the IL12‐STAT4 signal pathway, thereby enhancing the antitumour response of cytotoxic T lymphocytes (CTLs) in vivo. Next, to verify the critical role of CD8+ T cells in suppressing melanoma development, we transplanted CD8+ T cells from immunized mice to B16F10 tumour‐bearing mice and discovered that the survival rate of tumour‐bearing mice was significantly prolonged. In summary, our experimental results indicate that CHR can be used as a potential adjuvant to enhance antigen immunogenicity, inhibit B16F10 tumour growth in mice and improve tumour immune response. Hesperetin (HES) is a dihydroflavone with the molecular formula of C16H14O6. It has been reported that Hesperetin has antioxidant and anticancer effects. Recent studies showed that it can also regulate immune responses. To assess its potential function as a vaccine adjuvant, we formulated HES with inactivated B16F10 melanoma cells and determined whether it would enhance the activation of antigen‐presenting cells by experiments in vivo and in vitro. We found that HES activated the PI3K‐Akt signalling pathway in antigen‐presenting cells (APCs), enhanced cytotoxic T lymphocyte (CTL) responses and deactivated tolerogenic T cells. We also observed that inactivated B16F10 cells in combination with HES vaccine inhibited the growth of mice tumours, resulting in improved overall survival compared to the effects of inactivated B16F10 cell vaccine. To verify that CD8+T cells play a key role in inhibiting the development of melanoma, we transferred the sorted CD8+T cells from immunized mice to B16F10 challenged models and found that the survival rate of tumour‐bearing mice was significantly prolonged. MicroRNAs (miRNAs) participate in the development and progression of melanoma. However, while dysregulation of microRNA‐378 (miR‐378) has been seen in various cancer types, its clinical importance and function in melanoma are poorly elucidated. In this work, miR‐378 expression in melanoma and in adjacent non‐cancerous tissue was evaluated with a quantitative real‐time polymerase chain reaction. A series of assays (wound healing, Transwell, and nude mouse subcutaneous tumor model) were used to investigate the implications of abnormal miR‐378 regulation on melanoma cell migration and invasion in vitro, and on tumorigenicity in vivo. Prediction and conformation of the miR‐378 target gene was undertaken using bioinformatic analysis and luciferase reporter system. Expression of miR‐378 was often increased in melanoma, and shown to potentiate its migration, invasion, and tumorigenicity. miR‐378 acted, at least partially, through inhibition of the potential target FOXN3 and via Wnt/β‐catenin pathway activation. The findings indicate that miR‐378 triggers melanoma development and progression. MicroRNAs (miRNAs) play an increasingly important role in cancer growth by coordinately suppressing genes that control cell migration, proliferation, and invasion. The above results can be achieved through the regulation of gene expression by miRNAs by suppressing translation or the direct sequence-specific degradation of the targeted mRNA. In the present study, we indicate that the expression of miR-216b could be effectively repressed both in human melanoma tissues through a comparison with primary melanoma and in human melanoma cell lines through a comparison with a normal human keratinocyte line. Moreover, miR-216b induced a clear decrease in melanoma cell proliferation and migration in vitro. Forkhead box M1 (FOXM1) was confirmed as a target gene of miR-216b, and the overexpression of miR-216b markedly repressed the luciferase activity of reporter plasmids containing the FOXM1 3′-UTR (untranslated region). Furthermore, miR-216b suppressed melanoma cell growth in nude mice in vivo, with the effects of miR-216b overexpression on melanoma cell growth and proliferation reversed by FOXM1 overexpression. The results demonstrated that miR-216b is a tumor suppressor in melanoma, identified the FOXM1 signaling pathway as a target of miR-216b action, and suggested a potential therapeutic role for miR-216b in melanoma. Background/purposes Fluorescence emission from in vivo cutaneous melanin was recently detected under near-infrared ( NIR) excitation by our group. We then built a prototype NIR autofluorescence imaging system to observe and characterize the melanin distribution in human skin. In this article, we reported a new setup of NIR fluorescence imaging system and calibration methods to optimize the system for better clinical feasibility and clearer image. Methods The imaging system was designed to perform both fluorescence and reflectance imaging with a 785-nm fiber-coupled laser source. The illumination light was purified by a 785-nm bandpass filter for fluorescence excitation; while the spontaneous components were selected by a longpass filter for NIR reflectance imaging. A hand-controlled filter wheel was used to switch these two filters for different imaging modes. A dichroic filter was used to guide the illuminating light onto the skin surface for excitation. Reflectance and fluorescence signals were collected sequentially by a NIR optimized CCD camera. The captured images were calibrated by the reflectance images of a standard reflectance disk for non-uniform illuminations and light collection efficiencies. Results The clinical results demonstrated that NIR fluorescence intensities and distribution patterns vary among lesion types. It was also confirmed that pigmented skin lesions emitted higher NIR fluorescence than the surrounding normal skin due to the presentation of higher concentrations of cutaneous melanin within the lesions. Conclusion NIR autofluorescence imaging system could be utilized as a powerful tool for visualizing melanin distribution in pigmented skin lesions and as a potential method for aiding melanoma detection.
