Lane width is crucial for traffic safety management. Previous studies have shown that inappropriate width will lead to an increase in accident probability and unsafe driving behaviors. However, there have been limited studies on determination method of special lane width for different models, especially for passenger cars. To tackle this problem, the lateral driving behavior characteristics of the expressway need to be clarified. This work aims to obtain the appropriate lane width according to the vehicles' width and lateral position characteristics. In this study, the lane lateral residual width (the distance between the vehicle body's contour and the lane marking on the same side) and the lateral safety margin are extracted as characterization indices of vehicle lateral characteristics from natural driving trajectory data of expressways. The effect of vehicle type, driving speed, and lane position on the expressway's trajectory behavior is investigated. The results show that the utilization rate of lane width is higher in the outer lane compared to the inner lane. As driving speed increases, vehicles in the inner and middle lanes exhibit shy away effect, moving away from obstacles. Substantial variations exist in the lateral width of lanes among different vehicle types. When driving in the same lane, passenger cars' lane lateral residual width that is 0.4–0.5 m wider than that of heavy vehicles. The recommended lane width for the safe operation of trucks on expressways is 3.75 m. After a comprehensive analysis of vehicle width, trajectory oscillation, and lateral safety margin, this study proposes a minimum lane width of 3.25 m and a recommended general width of 3.5 m for a passenger car‐only expressway. Chaiqin Qingning Capsule (CQQNC), a traditional Chinese patent medicine, can effectively shorten the duration of fever and significantly improve fever symptoms. However, the mechanism of its antipyretic effect needs to be further elucidated. Therefore, we aimed to investigate the molecular mechanism of CQQNC in the treatment of fever. We used the network pharmacology method to analyze the mechanism of action of CQQNC in the treatment of fever and validated our study primarily by molecular docking. Finally, the predictive results were verified by IL‐1β‐induced bEnd.3 cells. The results showed that quercetin, kaempferol, cubebin, chenodeoxycholic acid, isorhamnetin, bilirubin, cholic acid, and baicalin were the major components of CQQNC against fever. A total of 381 common targets have been crossed by CQQNC for the treatment of fever. Furthermore, we found that CQQNC targets several deregulated genes in fever such as AKT1, COX2, AVP, cAMP, IL6, IL1B, TNF, mPGES1, and PI3K, biological functions such as endopeptidase, cytokine receptor binding, and phosphatase activity, and signaling pathways such as the PI3K‐Akt pathway and the AGE‐RAGE signaling pathway in diabetic complications. The docking study revealed that the core components of CQQNC both had high affinity for hub targets, especially the targets of COX‐2, cAMP, mPGES1, and PI3K proteins. To further investigate the mechanism of CQQNC, an Elisa assay and Western blot detection were performed as part of an in vitro study. Elisa's result showed that CQQNC can significantly decrease the expression levels of cPLA2, sPLA2, PGE2, cAMP, and 15‐PGDH after stimulating IL‐1β to bEnd.3 cells in a dose‐dependent manner (p <.01, p <.001, p <.0001). In addition, detection of the PGE2/ COX /cMAP pathway via immunoblotting showed that CQQNC can significantly downregulate the protein expression of COX‐1, COX‐2, EP3, cAMP, and mPGES1 in bEnd.3 (p <.0001, p <.001, p <.01). In conclusion, our study confirmed that the antipyretic mechanism of CQQNC affects the synthesis and secretion processes of PGE2 via the PGE2/ COX/cAMP pathway, providing insight into the antipyretic mechanism of CQQNC in clinical application. Aims: To investigate changes in functional connectivity (FC) focusing on parietal operculum cortex 2 (OP2) in benign paroxysmal positional vertigo (BPPV) patients with residual dizziness (RD) after successful canalith repositioning procedure (CRP). Methods: High‐resolution three‐dimensional T1 and resting‐state functional magnetic resonance imaging (fMRI) were performed on 55 healthy controls (HCs), 55 BPPV patients with RD, and 55 patients without RD after successful CRP. Seed‐based (bilateral OP2) FC was calculated to investigate the changes in FC among the three groups. Additionally, we further explored the associations between abnormal FC and clinical symptoms. Results: One‐way analysis of covariance showed significant FC differences among the three groups. Post‐hoc analysis showed that patients with RD exhibited decreased FC between left OP2 and regions of left angular gyrus (AG), thalamus, precuneus, middle frontal gyrus (MFG), and right cerebellum posterior lobe (CPL) in comparison with HCs. In addition, compared with patients without RD, patients with RD showed decreased FC between left OP2 and regions of left MFG, AG, middle temporal gyrus, and right CPL. Moreover, in patients with RD, the FC between left thalamus and OP2 was negatively correlated with duration of RD, and the FC between left AG and OP2 was negatively correlated with duration of BPPV. Conclusion: BPPV patients with RD showed reduced FC between brain regions involved in vestibular processing and spatial cognition; These results suggested that BPPV patients with RD might have diminished central processing of vestibular information and impaired spatial cognition. A large‐scale physics‐based simulation is conducted to investigate ground motion distribution in the Mw 7.0 2016 Kumamoto earthquake, Japan. The model simulates the earthquake scenario from fault rupture to wave propagation, and localized site response with consideration of the combined effect of soil nonlinearity and topographic amplification. The simulation domain is 51 km × 43 km × 25 km, and the obtained ground motion time histories are compared satisfactorily with recordings from KiK‐net and K‐NET. Ground motion distribution considering nonlinear soil response and topographic amplification is presented. A 3D equivalent linear model is developed to mimic the soil nonlinearity, and it is demonstrated that neglecting soil nonlinearity could over‐predict peak ground acceleration (PGA) and underestimate peak ground velocity (PGV) near the fault. The topographic amplification factors (TAFs) of PGA and PGV are found between 0.5 and 2.0, with a correlation coefficient of 0.7 between them. Predictive equations are proposed to correlate TAFs of PGA and PGV with topographic features, which are represented by relative heights obtained at different length scales. Due to their involvement in numerous biochemical pathways, neuropeptides have been the focus of many recent research studies. Unfortunately, classic analytical methods, such as western blots and enzyme‐linked immunosorbent assays, are extremely limited in terms of global investigations, leading researchers to search for more advanced techniques capable of probing the entire neuropeptidome of an organism. With recent technological advances, mass spectrometry (MS) has provided methodology to gain global knowledge of a neuropeptidome on a spatial, temporal, and quantitative level. This review will cover key considerations for the analysis of neuropeptides by MS, including sample preparation strategies, instrumental advances for identification, structural characterization, and imaging; insightful functional studies; and newly developed absolute and relative quantitation strategies. While many discoveries have been made with MS, the methodology is still in its infancy. Many of the current challenges and areas that need development will also be highlighted in this review. Post‐synaptic density 93 (PSD‐93) mediates glutamate excitotoxicity induced by ischemic brain injury, which then induces microglial inflammatory response. However, the underlying mechanisms of how PSD‐93 mediates the crosstalk between neurons and microglia in the post‐synaptic dense region remain elusive. CX3 chemokine ligand 1 (CX3CL1) is a chemokine specifically expressed in neurons while its receptor CX3CR1 is highly expressed in microglia. In this study, we examined the interaction of PSD‐93 and CX3CL1 in the crosstalk between neurons and microglia in acute ischemic stroke. We utilized male C57BL/6 mice to establish the middle cerebral artery occlusion model (MCAO) and designed a fusion small peptide Tat‐CX3CL1 (357‐395aa) to inhibit PSD‐93 and CX3CL1 interaction. The combination peaks of PSD‐93 and CX3CL1 at 6 hr after I/R were observed. The binding sites were located at the 420–535 amino acid sequence of PSD‐93 and 357–395 amino acid sequence of CX3CL1. Tat‐CX3CL1 (357‐395aa) could inhibit the interaction of PSD‐93 and CX3CL1 and inhibited the pro‐inflammatory cytokine IL‐1β and TNF‐α expression and provided neuroprotection following reperfusion. Together, these data suggest that PSD‐93 binds CX3CL1 to activate microglia and initiate neuroinflammation. Specific blockade of PSD‐93‐CX3CL1 interaction reduces I/R induced neuronal cell death, and provides a new therapeutic target for ischemic stroke. [ABSTRACT FROM AUTHOR]

Enzymatic reactions are often terminated by severe aqueous environments. Inspired by bio‐protection of silk cocoons, a system of silk fibroin (SF) nanococoons is developed, which is capable of providing a bio‐protective environment for enzymatic activities on one hand, and facilitating the process of cascade enzymatic reactions on the other hand. This gives rise to the significant enhancement in enzymatic stability and efficacy. For instance, the protected glucose oxidase and horseradish peroxidase enzymes exhibit a 17.5‐ and 7.0‐fold increase when stored at 60 °C (>50% activity; ≈28 h) and 25 °C (>67.5% activity; ≈14 days), respectively, compared with free enzymes. It follows that this protective effect is subject to the mutual templated crystallization between enzymes and SF molecules, which prevents enzyme molecules from unfolding. In animal experiments, it shows that alcohol oxidase and catalase protected by silk nanococoons are able to reduce the alcohol levels of intoxicated mice with >5.0‐time efficacy at the alcohol decomposition level of >80% in 4.0 h compared with the unprotected. To examine the mechanism of the network formation of silk fibroin (SF), monodispersed colloidal particles (MDCPs) are used as well defined foreign substrates to quantify their effect on the primary nucleation of β-crystallites in molecular networks (silk nanofibrils) and the hierarchical network formation of SF. It follows that MDCPs are capable of accelerating the SF gelation by reducing the multistep nucleation barrier, which gives rise to a high density of silk fibril domain networks due to the increase of primary nucleation sites. Consequently, through governing the change in the hierarchical mesoscopic structure, the macroscopic performance of silk materials (e.g., the rheological properties of SF hydrogels and the tensile stress of fibers) can be controlled directly. As SF hydrogels represent a typical example of weak fibril domain-domain network interactions, the increase of fibril domain density leads to weaker gels. On the other hand, SF fibers correspond to strong fibril domain-domain network interactions, the increase of fibril domain density ends up with much tougher fibers. The knowledge obtained provides a facile strategy in controlling the complex hierarchical structure and macroscopic performance of SF materials, and offers useful routes for general design and functionalization of soft materials. New detrital low-temperature thermochronometry provides estimates of long-term erosion rates and the timing of initiation of river incision from across the interior of the Tibetan Plateau. We use the erosion history of this region to evaluate proposed models of orogenic development as well as regional climatic events. Erosion histories of the externally drained portion of the east-central Tibetan Plateau are recorded in modern river sands from major rivers across a transect that spans >750 km and covers a region with no published thermochronometric ages. Individual grains from eight catchments were analyzed for apatite (U-Th)/He and fission track thermochronometry. A wide distribution in ages that, in most cases, spans the entire Cenozoic and Late Mesozoic eras requires a long period of slow or no erosion with a relative increase in erosion rate toward the present. We apply a recently developed methodology for inversion of detrital thermochronometric data for three specified erosion scenarios: constant erosion rate, two-stage erosion history, and three-stage erosion history. Modeling results suggest that rates increase by at least an order of magnitude between 11 and 4 Ma following a period of slow erosion across the studied catchments. Synchroneity in accelerated erosion across the whole of the Tibetan Plateau rather than a spatial or temporal progression challenges the widely held notion that the plateau evolved as a steep, northward-propagating topographic front, or that south to north precipitation gradients exert a primary control on erosion rates. Instead, we suggest that accelerated river incision late in the orogen's history relates to regional-scale uplift that occurred in concert with eastern expansion of the plateau. On page 8978, Y. H. Lin, Y. Y. Liu, and co‐workers demonstrate a precise control over the nucleation process of silk materials with the aid of mono‐disperse colloidal particles. Manipulation of the mesoscopic silk structure toward a desired macroscopic performance is achieved.
