A phylogenetic study of Uebelmannia investigates the relationships of a newly discovered taxon in this enigmatic, early‐diverging genus that is sister to all the other c. 460 species of tribe Cereeae. Here we present a coalescent‐based phylogenetic tree inferred with nuclear genes captured by the Cactaceae591 probe set encompassing all Uebelmannia species. The new taxon U. nuda is sister to U. pectinifera, while U. gummifera and U. buiningii form another pair of closely related species. The unusual characteristics of the new cactus (semi‐subterranean/geophytic habit, turbinate stem, naked areoles) distinguish it from all other Uebelmannia species. The definition of the relationships within this early‐derived genus endemic to the campo rupestre of Minas Gerais State became more evident, and this new find adds to the importance of this biogeographic province for the evolution of tribe Cereeae. Background: Dysregulated T lymphocyte response is thought to play a key role in chronic intestinal inflammation (CIE). Objectives: To evaluate the presence of changes in peripheral and intestinal T lymphocyte subsets and to describe potential immune and inflammatory biomarkers in dogs with CIE. Animals: Sixteen healthy dogs and 26 dogs were diagnosed with CIE. Methods: Prospective case‐control study evaluating peripheral and intestinal T lymphocytes using flow cytometry and inflammatory markers obtained from complete blood cell counts. Results: Dogs with CIE had higher peripheral activated T helper (Th) lymphocytes (87/μL [18‐273] CIE, 44/μL [16‐162] healthy control (HC, P =.013) and regulatory T cells (Treg; 108/μL [2‐257] CIE, 34/μL [1‐114] HC, P =.004). In the intestinal epithelium, CIE dogs presented lower percentages of Th (4.55% [1.75‐18.67] CIE, 8.77% [3.79‐25.03] HC, P =.002), activated Th cells (0.16% [0.02‐0.83] CIE, 0.33% [0.05‐0.57] HC, P =.03) and CD4/CD8 ratio (0.08 [0.02‐0.39] CIE, 0.21 [0.07‐0.85] HC, P =.003). Conversely, higher percentage of activated T cytotoxic cells (20.24% [3.12‐77.12] CIE, 12.32% [1.21‐39.22] HC, P =.04) and interferon‐gamma (IFN‐γ) producing T lymphocytes (7.36% [0.63‐55.83] CIE, 1.44% [0.00‐10.56] HC, P =.01) within the epithelium was observed. In the lamina propria the percentage of Treg lymphocytes was higher (6.02% [1.00‐21.48] CIE, 3.52% [0.18‐10.52] HC, P =.02). Conclusions and Clinical Importance: Systemic and intestinal immune alterations occur in dogs with CIE suggesting that blood IFN‐γ producing T lymphocytes and the systemic immune‐inflamation index (SII) could potentially serve as biomarkers for the disease. A new study of the phylogeny of Cereus investigates generic monophyly and previous subgeneric classification. A coalescent approach was used to assess phylogenetic relationships among Cereus species based on nuclear orthologous genes captured by the Cactaceae591 probe set. The analysis included two unusual new taxa from eastern and northern Brazil to ascertain their position within the genus. Amongst the additional taxa, one was found to be early derived to the Atlantic Forest and coastal Cereus species and is described as C. ingens. Its imposing habit, considerably broader flower, yellow fruits and large seeds differentiate it from other Cereus species, and its inland agreste forest habitat in Minas Gerais and Bahia leads it to be categorized as Endangered, as little remains of this vegetation type. A second new species, C. gerardi, is described from the state of Tocantins (northern Brazil). Vegetatively this taxon is remarkable for resembling members of Pilosocereus, but has typical Cereus flowers, matching those of the type of the genus, C. hexagonus. Premise: Pilosocereus (Cactaceae) is an important dry forest element in all subregions and transitional zones of the neotropics, with the highest diversity in eastern Brazil. The genus is subdivided into informal taxonomic groups; however, most of these are not supported by recent molecular phylogenetic inferences. This lack of confidence is probably due to the use of an insufficient number of loci and the complexity of cactus diversification. Here, we explored the species relationships in Pilosocereus in more detail, integrating multilocus phylogenetic approaches with the assessment of the ancestral range and the effect of geography on diversification shifts. Methods: We used 28 nuclear, plastid, and mitochondrial loci from 54 plant samples of 31 Pilosocereus species for phylogenetic analyses. We used concatenated and coalescent phylogenetic trees and Bayesian models to estimate the most likely ancestral range and diversification shifts. Results: All Pilosocereus species were clustered in the same branch, except P. bohlei. The phylogenetic relationships were more associated with the geographic distribution than taxonomic affinities among taxa. The genus began diversifying during the Plio‐Pleistocene transition in the Caatinga domain and experienced an increased diversification rate during the Calabrian age. Conclusions: We recovered a well‐supported multispecies coalescent phylogeny. Our results refine the pattern of rapid diversification of Pilosocereus species across neotropical drylands during the Pleistocene and highlight the need for taxonomic rearrangements in the genus. We recovered a pulse of diversification during the Pleistocene that was likely driven by multiple dispersal and vicariance events within and among the Caatinga, Cerrado, and Atlantic Forest domains. Bovine vector‐borne diseases have a considerable economic impact worldwide and affect health of humans and animals. However, different aspects of their epidemiology and their pathogenesis remain unclear. Despite the frequent description of clinical cases reported by practitioners attending cattle from Madrid, Central Spain, molecular prevalence of Anaplasma spp. and Babesia spp. has not been described. The aim of this study was to assess the positivity rate of A. phagocytophilum, A. marginale, A. centrale, B. bigemina and B. divergens in livestock of this area and to evaluate the existence of associations between these pathogens and haematological, biochemical and epidemiological data. Babesia divergens and A. phagocytophilum were detected for the first time in cattle from Madrid. Their positivity percentages were low (2.2% ± 1.4% and 1.8% ± 1.2%, respectively), but this description is of special interest, as these agents are potentially zoonotic. Both agents were found in areas of higher altitude and relative humidity and lower temperature. The detection of ticks in livestock during the moment of sampling was confirmed as a risk factor for these infections. Anaplasma marginale showed the highest molecular infection rate (30% ± 4.1%) in this study, followed by B. bigemina (21.9% ± 3.7%). Higher positivity rates of A. marginale and B. bigemina were found in the areas of mountain climate and warm‐summer continental Mediterranean climate. The use of ectoparasiticide treatment was found as a risk factor for infection by A. marginale and B. bigemina. This finding could lead to reconsider the ectoparasiticide protocols that are used nowadays. Grazing on pastures with domestic or wild ruminants and the presence of wild carnivores were associated with higher rates of infection by these four agents and coinfections were frequently found. Delimiting species boundaries is a major goal in evolutionary biology. An increasing volume of literature has focused on the challenges of investigating cryptic diversity within complex evolutionary scenarios of speciation, including gene flow and demographic fluctuations. New methods based on model selection, such as approximate Bayesian computation, approximate likelihoods, and machine learning are promising tools arising in this field. Here, we introduce a framework for species delimitation using the multispecies coalescent model coupled with a deep learning algorithm based on convolutional neural networks (CNNs). We compared this strategy with a similar ABC approach. We applied both methods to test species boundary hypotheses based on current and previous taxonomic delimitations as well as genetic data (sequences from 41 loci) in Pilosocereus aurisetus, a cactus species complex with a sky‐island distribution and taxonomic uncertainty. To validate our method, we also applied the same strategy on data from widely accepted species from the genus Drosophila. The results show that our CNN approach has a high capacity to distinguish among the simulated species delimitation scenarios, with higher accuracy than ABC. For the cactus data set, a splitter hypothesis without gene flow showed the highest probability in both CNN and ABC approaches, a result agreeing with previous taxonomic classifications and in line with the sky‐island distribution and low dispersal of P. aurisetus. Our results highlight the cryptic diversity within the P. aurisetus complex and show that CNNs are a promising approach for distinguishing complex evolutionary histories, even outperforming the accuracy of other model‐based approaches such as ABC. Aim: The interconnectedness and biotic interchange among Neotropical biomes are thought to play an important role in driving adaptation and diversification. However, how these processes are in synteny to trait evolution in species of open and xeric areas is poorly studied. Here, we investigate the spatial and temporal dimensions of evolution and candidate traits associated with biome shifts in xeric vegetation, focusing on the family Cactaceae. Location: Xeric and open areas of South America. Taxon: Genus Cereus Mill. (Cactaceae, Cereeae). Methods: We applied biogeographical reconstructions on a time-calibrated phylog- eny inferred from multilocus data (ddRAD-Seq) using Bayesian analyses on BEAST2, species distribution modelling in Maxent, the reconstruction of biome affinities and niche shift analyses based on abiotic traits (climate and soil) using Mk-model in BioGeoBEARS, and phenotypic trait-based analysis in Mesquite. Results: The Cerrado domain is the ancestral area of Cereus, with most diversification events occurring in a time of intense orogenesis, climatic changes, and marine regressions within the last 5 Mya. Events of biome transition from the seasonally dry tropical forest (SDTF) were also associated with trait and niche shifts. Main conclusions: The diversification of the xerophyte genus Cereus is associated with the climatic and geomorphological instabilities of the Pliocene and Pleistocene epochs. The Cerrado domain states an important region of dispersal for the genus. Some geographical range movements involved biome shifts associated with niche evolution while others were restricted to a simple biogeographical transition without niche change. Particular clades that experienced biome shifts displayed some phenotypic state changes, suggesting a role of biotic traits for environment transition. The results observed in Cereus may be a biogeographical pattern that should be tested with other cactus species, such as Pilosocereus spp., or species of xeric habitats, such as Annonaceae and Vochysiaceae. The aim of this study was to determine the pharmacokinetic parameters of doxycycline in dogs and assess the efficacy of an oral drug dosage regimen of 10 mg/kg daily for 28 days through Pharmacokinetic/Pharmacodynamic (PK/PD) target analysis based on Monte Carlo simulation, using previously published data for the zoonotic pathogen Staphylococcus pseudintermedius. After a multiple‐dosage regimen, the accumulation index was 1.88 ± 0.82. The Cmaxss and Cminss values were 5.18 ± 1.81 µg/ml and 1.91 ± 1.35 µg/ml, respectively. There were statistically significant differences for Cmax, Cmin at 24 hr, MRTt, AUCt and AUC∞ between days 1 and 28. The Cminss value was over the MIC of the principal pathogens, and Cmaxss was higher than the resistance values (>2 μg/ml). For AUC/MIC indices of 12, 25 and 40, the cumulative fraction responses (CFR) were 94.01%, 69.55% and 60.86%, respectively; for an MIC value of 2 µg/ml, the corresponding probability of target attainment (PTA) was 99.94%, 84.78% and 45.16%, respectively. Doxycycline was used against numerous localized infections in different organs and tissues. For the strains with MIC < 1 μg/mL, PTA was close to 100%, even for the most demanding ones, specifically 94.98% for an index of 40% and 99.9% for an index of 25. [ABSTRACT FROM AUTHOR]

Aim: To investigate how the potential bias from isolation by distance (IBD) in inferences of population structure has been addressed, in studies aiming biodiversity conservation. Location: Global. Methods: We reviewed the literature on the impact of IBD on the performance of the widely used software structure. We also performed a literature survey in the Web of Knowledge to assess how data have been analysed in biodiversity conservation studies when IBD is detected. Results: By reviewing the literature on structure performance in IBD data sets, we found a high number of studies showing that structure outputs are extremely affected by IBD. This misleading inference results principally in the detection of artificial genetic clusters. The literature survey showed that IBD was present in most data sets (60.56%) and that a substantial number of the articles only tested IBD by Mantel tests (82.21%). The most concerning result is that several articles have been using structure even after detecting IBD (57.99%), and a substantial number of them are drawing formal conservation strategies, notwithstanding the potentially biased results (51.49%). Main conclusions: Our results are of great concern, as conservation strategies may be distinct under different population structure, and its success can be affected by the incorrect identification of populations. We recommend possible stages to be considered in the presence of IBD, which can help conservation investigators before the proposition of explicit conservation strategies. [ABSTRACT FROM AUTHOR]

In order to investigate biogeographic influences on xeric biota in the Brazilian Atlantic Forest ( BAF), a biodiversity hotspot, we used a monophyletic group including three cactus taxa as a model to perform a phylogeographic study: Cereus fernambucensis subsp. fernambucensis, C. fernambucensis subsp. sericifer, and C. insularis. These cacti are allopatric and grow in xeric habitats along BAF, including isolated granite and gneiss rock outcrops (Inselbergs), sand dune vegetation (Restinga forest), and the rocky shore of an oceanic archipelago (islands of Fernando de Noronha). The nucleotide information from nuclear gene phytochrome C and plastid intergenic spacer trnS-trnG was used to perform different approaches and statistical analyses, comprising population structure, demographic changes, phylogenetic relationships, and biogeographic reconstruction in both spatial and temporal scales. We recovered four allopatric population groups with highly supported branches in the phylogenetic tree with divergence initiated in the middle Pleistocene: southern distribution of C. fernambucensis subsp. fernambucensis, northern distribution of C. fernambucensis subsp. fernambucensis together with C. insularis, southern distribution of C. fernambucensis subsp. sericifer, and northern distribution of C. fernambucensis subsp. sericifer. Further, the results suggest that genetic diversity of population groups was strongly shaped by an initial colonization event from south to north followed by fragmentation. The phylogenetic pattern found for C. insularis is plausible with peripatric speciation in the archipelago of Fernando de Noronha. To explain the phylogeographic patterns, the putative effects of both climatic and sea level changes as well as neotectonic activity during the Pleistocene are discussed. [ABSTRACT FROM AUTHOR]

Aim The aim of this paper is to investigate the influence and extent of past climatic changes on South American biota. To this end, we establish phylogeographical hypotheses for a monophyletic group of four cactophilic species of Drosophila (the Drosophila serido haplogroup) found in xeromorphic vegetation in Brazil. The effects of Quaternary palaeoclimatic oscillations on the demographic fluctuations of our biological model and its sister group Drosophila antonietae are investigated. Location Areas of eastern Brazil with open vegetation, including seasonally dry tropical forest ( SDTF) areas and montane savannas, which are disjunctly distributed in eastern Brazil. We also analysed populations of sand dune vegetation from littoral areas in Brazil. Methods Nucleotide information from 630 bp of the cytochrome c oxidase subunit I gene from 441 individuals was used to perform a series of statistical analyses to infer a phylogeographical hypothesis for each species and to date the divergence time between the D. serido and D. antonietae haplogroups. Results All of the analysed species experienced population expansion during the Pleistocene, probably following the historical migratory routes that have been proposed for the expansion of SDTFs in Brazil. A bottleneck event in the Holocene was inferred for Drosophila seriema, probably due to ecological factors related to the geographical distribution of the host plant. The divergence time for the D. serido and D. antonietae haplogroups was calculated to have occurred during the early Pleistocene. Main conclusions Geographical and chronological evidence suggests that the major vicariant events between the D. serido and D. antonietae haplogroups, as well as the demographic fluctuations in each of the species within these lineages, could be related to the causal effects of Quaternary palaeoclimatic changes on the spatial dynamics of the SDTFs in Brazil. Infection by different Leishmania spp. in cats has been reported in many countries. In Spain, since the first Leishmania infection described in 1933, sporadic clinical cases in cats have been reported. Various serologic studies performed in other areas of Spain have shown seroprevalences ranging between 1.7 and 60%. The aim of the present study was to determine the prevalence of leishmaniasis in cats from Central Spain (Madrid), and to assess the existence of associations between Leishmania infantum infection and relevant data obtained from each cat. Two-hundred thirty-three cats attended at the Veterinary Teaching Hospital in Madrid between September 2005 and June 2006 were tested for L. infantum using the indirect immunofluorescent antibody (IFA) test (cutoff: 1:100) and PCR. PCR testing was performed on the samples to detect Leishmania infection, targeting the kinetoplast DNA (kDNA). Our results showed a seroprevalence of 1.29% (3/233) using IFA test. Another seven cats were also seroreactive to L. infantum one dilution under the cutoff (1:50). Considering all the seroreactive samples, the percentage of positive animals to L. infantum was 4.29%. Only one of the cats (0.43%) included in the study was PCR-positive. Relative lymphocytosis and an increase in alanine aminotransferase (ALT) value were statistically associated with seroreactivity to L. infantum. Our results demonstrate the presence of cats seroreactive to L. infantum in Central Spain, an endemic area for this disease in dogs. [ABSTRACT FROM AUTHOR]

The aim of this study was to compare different polymerase chain reaction (PCR) methods for the detection of Ehrlichia canis in blood samples and to relate these results to clinical findings and serology to E. canis using the indirect fluorescent antibody (IFA) test. Nine seropositive and nine seronegative dogs were included in this study. DNA was extracted once and used in one simple PCR and five nested PCR protocols previously described. In selected dogs (three seropositive and one seronegative) blood samples were aseptically collected in order to attempt the isolation of E. canis in the DH82 cell line. Results show that nested PCR protocols seem to be more sensitive than the simple PCR. Considering only nested PCR protocols, 33% of the IFA-positive samples were PCR positive using the five different protocols. The rest of the IFA-positive samples were PCR positive or negative depending on the protocol used. Clinical signs and laboratory findings compatible with canine monocytic ehrlichiosis (CME) were found in 67% of dogs positive by the IFA test. All samples positive by both techniques (IFA test and PCR) were from dogs suffering from clinical CME. IFA-negative samples were PCR negative, except 22% that were PCR positive when using only one of the nested PCR protocols. Antibodies to Ehrlichia spp. and inclusion bodies compatible with Ehrlichia spp. in feline blood cells have been previously detected in Spain. The aim of this study was to assess the presence of antibodies to E. canis, N. risticii, and A. phagocytophilum in 122 feline serum samples from Madrid (central Spain). In addition, Ehrlichia genus-specific, one-tube, nested polymerase chain reaction (PCR) was performed from blood samples from these cats. Of the cats, 10.6% were seropositive for E. canis, 2.4% were positive for N. risticii, and 4.9% were seropositive for A. phagocytophilum. Two N. risticii-positive cats and one animal seropositive to A. phagocytophilum were also seropositive for E. canis. Despite these seropositive results, all the blood samples analyzed by PCR were negative. Our results demonstrate reactivity against agents implicated in feline ehrlichiosis in Spain. Further studies should be performed in order to clarify the significance of serology and PCR in the diagnosis of feline ehrlichiosis. [ABSTRACT FROM AUTHOR]
