Tricin [5,7-dihydroxy-2-(4-hydroxy-3,5-dimethoxyphenyl)-4H-chromen-4-one], a flavone, was recently established as an authentic monomer in grass lignification that likely functions as a nucleation site. It is linked onto lignin as an aryl alkyl ether by radical coupling with monolignols or their acylated analogs. However, the level of tricin that incorporates into lignin remains unclear. Herein, three lignin characterization methods: acidolysis; thioacidolysis; and derivatization followed by reductive cleavage; were applied to quantitatively assess the amount of lignin-integrated tricin. Their efficiencies at cleaving the tricin-(4′- O-β)-ether bonds and the degradation of tricin under the corresponding reaction conditions were evaluated. A hexadeuterated tricin analog was synthesized as an internal standard for accurate quantitation purposes. Thioacidolysis proved to be the most efficient method, liberating more than 91% of the tricin with little degradation. A survey of different seed-plant species for the occurrence and content of tricin showed that it is widely distributed in the lignin from species in the family Poaceae (order Poales). Tricin occurs at low levels in some commelinid monocotyledon families outside the Poaceae, such as the Arecaceae (the palms, order Arecales) and Bromeliaceae (Poales), and the non-commelinid monocotyledon family Orchidaceae (Orchidales). One eudicotyledon was found to have tricin ( Medicago sativa, Fabaceae). The content of lignin-integrated tricin is much higher than the extractable tricin level in all cases. Lignins, including waste lignin streams from biomass processing, could therefore provide a large and alternative source of this valuable flavone, reducing the costs, and encouraging studies into its application beyond its current roles. Using a functional genomics approach, four candidate genes ( PtGT34A, PtGT34B, PtGT34C and PtGT34D) were identified in Pinus taeda. These genes encode CAZy family GT34 glycosyltransferases that are involved in the synthesis of cell-wall xyloglucans and heteromannans. The full-length coding sequences of three orthologs ( PrGT34A, B and C) were isolated from a xylem-specific cDNA library from the closely related Pinus radiata. PrGT34B is the ortholog of XXT1 and XXT2, the two main xyloglucan (1→6)-α-xylosyltransferases in Arabidopsis thaliana. PrGT34C is the ortholog of XXT5 in A. thaliana, which is also involved in the xylosylation of xyloglucans. PrGT34A is an ortholog of a galactosyltransferase from fenugreek ( Trigonella foenum-graecum) that is involved in galactomannan synthesis. Truncated coding sequences of the genes were cloned into plasmid vectors and expressed in a Sf9 insect cell-culture system. The heterologous proteins were purified, and in vitro assays showed that, when incubated with UDP-xylose and cellotetraose, cellopentaose or cellohexaose, PrGT34B showed xylosyltransferase activity, and, when incubated with UDP-galactose and the same cello-oligosaccharides, PrGT34B showed some galactosyltransferase activity. The ratio of xylosyltransferase to galactosyltransferase activity was 434:1. Hydrolysis of the galactosyltransferase reaction products using galactosidases showed the linkages formed were α-linkages. Analysis of the products of PrGT34B by MALDI-TOF MS showed that up to three xylosyl residues were transferred from UDP-xylose to cellohexaose. We describe preparations of plant cell walls and polysaccharides obtained from plant cell walls that are added to food products for two purposes: as modifiers of food texture and/or as dietary fibres with potential health benefits. Although a number of different types of plant cell walls occur, only some are presently exploited. Commercial ‘fibre preparations’ range from those containing mostly primary walls to those containing mostly lignified secondary walls from which much of the lignin and non-cellulosic polysaccharides have been removed. Preparations of cell-wall polysaccharides are obtained from the following sources: cellulose mostly from secondary walls of cotton and wood, pectin from primary walls of dicotyledons, and (1→3),(1→4)- β-glucans and arabinoxylans from primary walls of cereal grains. Preparations of galactomannans, xyloglucans and the pectic polysaccharide rhamnogalacturonan I are obtained from non-lignified secondary walls of certain leguminous seeds. (1↵3),(1↵4)-β-Glucans had previously been detected in nonlignified cell wall preparations of only the Poaceae and five other families in the graminoid clade of the Poales (s.l.). Cell walls of vegetative organs of 12 species in nine families of the Poales (s.l.) were examined by immunogold labeling using a monoclonal antibody to (1↵3),(1↵4)-β-glucans. Three types of wall-labeling patterns were identified depending on the density of labeling of the nonlignified walls of epidermal and parenchyma cells and the lignified walls of sclerenchyma fibers and xylem tracheary elements: type 1 in Poaceae and Flagellariaceae, type 2 in Restionaceae and Xyridaceae, and type 3 in Cyperaceae and Juncaceae. Type 1 had the heaviest labeling of nonlignified walls and type 2 the heaviest labeling of lignified walls. Type 3 had the least wall labeling, with only very light labeling of nonlignified and lignified walls. No labeling was found over walls of Typhaceae, Sparganiaceae, or Bromeliaceae. Summary • (1 → 3),(1 → 4)-β-Glucans occur only in the cell walls of the family Poaceae (grasses and cereals) and related families, but little is known about their distribution among walls of different cell types or within walls. • The locations of (1 → 3)- and (1 → 3),(1 → 4)-β-glucans in the walls of the coleoptile, first leaf and root tip of barley (Hordeum vulgare ) seedlings were determined using immunogold labelling. • All the walls were labelled with the (1 → 3),(1 → 4)-β-glucan antibody, except those of the outer root cap cells. Labelling of the primary walls was heavy in the coleoptile and leaf, but light or very light in the root tip. Two types of primary wall labelling occurred: in the coleoptiles (except the walls of the epidermis and two layers of parenchyma under this) and in the leaf, labelling was throughout the walls; in the root tips, labelling was only adjacent to the plasma membrane. Small amounts of labelling occurred with the (1 → 3)-β-glucan antibody, mostly over plasmodesmata. The molecular mobility of the non-cellulosic polysaccharides in hydrated primary cell walls of three monocotyledons (Italian ryegrass, pineapple and onion) and one dicotyledon (cabbage) was studied using solid-state 13C NMR spectroscopy. These cell walls were chosen as they have different non-cellulosic polysaccharide compositions. By exploiting proton rotating-frame and spin-spin relaxation time constants three different cell wall domains which responded to cross-polarization experiments were identified. Most of the non-cellulosic polysaccharides occupied a mobile domain (C), but some occupied a partly rigid domain (B). Crystalline cellulose occupied a highly rigid domain (A). In the cell walls of Italian ryegrass and pineapple, domain C contained mainly glucuronoarabinoxylans and small amounts of rhamnogalacturonans; domain B contained small amounts of xyloglucans and galacturonans. However, in the cell walls of onion and cabbage, domain C contained mainly rhamnogalacturonans with galactans (in onion) or arabinans (in cabbage) as side chains; domain B contained galacturonans and xyloglucans. Single-pulse excitation was used on Italian ryegrass and cabbage cell walls to reveal signals from a highly mobile fourth domain (D). In Italian ryegrass cell walls domain D contained glucuronoarabinoxylans and small amounts of rhamnogalacturonan, whereas in cabbage cell walls it contained arabinan side chains of rhamnogalacturonans. A novel feature of the research was the use of solid-state 13C NMR spectroscopy to examine the molecular mobilities of the polysaccharides in monocotyledon cell walls that contain glucuronoarabinoxylans.
