Due to a change in capital funding formula, many school boards across the Province of Ontario engaged in Accommodation Reviews to rationalize the supply of school capacity. This process led to numerous school closures and raised important policy questions regarding the economic value of a school in terms of its capitalization into property values and, by extension, how the closure of a school might affect local neighborhoods. To explore these questions, this research uses spatial hedonic methods to estimate the implicit value of accessibility to schools in the City of Hamilton, Ontario. Spatial Durbin model results provide evidence of a significant negative correlation between distance to schools and housing prices in the Canadian context. This suggests that accessibility to schools is capitalized into property values and that the closure of a neighborhood school may result in potentially significant losses of economic value in communities. Per‐ and polyfluoroalkyl substances (PFAS) are a broad group of persistent organic compounds with vastly differing physicochemical and toxicological properties. Some jurisdictions have proposed to regulate PFAS as a single class to overcome the limitations of regulating such a diverse group on a chemical‐by‐chemical basis. Implications of regulating PFAS as a single class have been discussed for PFAS production and use, but equivalent discussion of implications for managing contaminated sites is largely lacking. This opinion piece summarizes the views of a group of environmental consultants, environmental regulators, land managers, and academics with significant experience in researching or managing PFAS. The group considered that neither a single PFAS class approach nor a chemical‐by‐chemical approach is well suited to managing risks from PFAS in a contaminated site setting, and defining PFAS subgroups would have value. Second, some but not all in the group, hypothesize that PFAS properties that drive fate and transport are those that influence toxicity and bioaccumulation in animals. This may be a valuable observation for future discussions on dividing PFAS into subclasses for contaminated site regulation based on physicochemical properties rather than purely structural definitions. Only a fraction of the total number of per‐ and polyfluoroalkyl substances (PFAS) are monitored on a routine basis using targeted chemical analyses. We report on an approach toward identifying bioactive substances in environmental samples using effect‐directed analysis by combining toxicity testing, targeted chemical analyses, and suspect screening. PFAS compete with the thyroid hormone thyroxin (T4) for binding to its distributor protein transthyretin (TTR). Therefore, a TTR‐binding bioassay was used to prioritize unknown features for chemical identification in a PFAS‐contaminated sediment sample collected downstream of a factory producing PFAS‐coated paper. First, the TTR‐binding potencies of 31 analytical PFAS standards were determined. Potencies varied between PFAS depending on carbon chain length, functional group, and, for precursors to perfluoroalkyl sulfonic acids (PFSA), the size or number of atoms in the group(s) attached to the nitrogen. The most potent PFAS were the seven‐ and eight‐carbon PFSA, perfluoroheptane sulfonic acid (PFHpS) and perfluorooctane sulfonic acid (PFOS), and the eight‐carbon perfluoroalkyl carboxylic acid (PFCA), perfluorooctanoic acid (PFOA), which showed approximately four‐ and five‐times weaker potencies, respectively, compared with the native ligand T4. For some of the other PFAS tested, TTR‐binding potencies were weak or not observed at all. For the environmental sediment sample, not all of the bioactivity observed in the TTR‐binding assay could be assigned to the PFAS quantified using targeted chemical analyses. Therefore, suspect screening was applied to the retention times corresponding to observed TTR binding, and five candidates were identified. Targeted analyses showed that the sediment was dominated by the di‐substituted phosphate ester of N‐ethyl perfluorooctane sulfonamido ethanol (SAmPAP diester), whereas it was not bioactive in the assay. SAmPAP diester has the potential for (bio)transformation into smaller PFAS, including PFOS. Therefore, when it comes to TTR binding, the hazard associated with this substance is likely through (bio)transformation into more potent transformation products. Around the globe, on‐site wastewater‐treatment systems (OWTSs) are critical for rural communities without access to a municipal sewer system. However, their treatment efficiency does not match that of modern wastewater‐treatment plants. The impact of OWTS discharge on nearby aquatic ecosystems and their resident fish species is poorly understood. In the present study, larval and adult fathead minnows (Pimephales promelas) and adult sunfish (Lepomis macrochirus) were exposed for 21 days to two trace organic contaminant (TOrC) mixtures replicating water chemistry derived from a previous environmental study. Larval fathead minnows were assessed for survival, growth, predator avoidance, and feeding efficiency. Adult fathead minnows and sunfish were assessed for a suite of physiological endpoints (condition indices, vitellogenin, glucose), histological changes, and fecundity. The only observed effect of TOrC mixture exposure on larval fathead minnows was a decrease in feeding efficiency. Effects were mixed in exposed adult fishes, except for male sunfish which realized a significant induction of vitellogenin (p < 0.05). The consequences of TOrC mixture exposure in the present controlled laboratory study match effects observed in wild‐caught sunfish in a corresponding field study. The present study begins to bridge the gap by connecting nonpoint OWTS pollution with biological effects observed in resident lake fish species. Given the effects observed despite the brevity of the laboratory mixture exposure, longer‐term studies are warranted to understand the full impacts of OWTS discharge to nearby aquatic ecosystems. Per‐ and polyfluoroalkyl substances (PFAS) are compounds manufactured for use in paints, cleaning agents, fire suppressants, nonstick cookware, food containers, and water‐resistant products. Concerns about PFAS stem from their ubiquitous presence in the environment, persistence, and variable/uncertain bioaccumulation and toxicity. In the present study, 5 perfluoroalkyl acids and one polyfluoroalkyl substance were administered to white‐footed mice (Peromyscus leucopus) to elucidate the kinetics of each chemical over 28 d of exposure. Perfluorooctanoate, perfluorohexane sulfonate (PFHxS), and perfluorobutane sulfonate were administered to male and female mice via drinking water. Perfluorooctane sulfonate, perfluorononanoate, 6:2 fluorotelomer sulfonate, and PFHxS were administered to male and female mice via oral gavage. Blood samples collected after 14 or 21 and 28 d of exposure were analyzed for individual PFAS concentrations via liquid chromatography‐tandem mass spectrometry. In general, a plateau in serum concentration in this toxicity test–relevant timeline depended on interactions between 1) the type of PFAS (i.e., perfluoroalkyl sulfonic acids [PFSAs] vs perfluoroalkyl carboxylic acids [PFCAs] vs polyfluorinated), 2) continuous versus bolus dosing, and 3) to a lesser extent, sex. Specifically, PFCAs were detected at higher concentration in females than males, whereas PFSAs were generally detected at similar levels across sex. An exception occurred when PFHxS yielded higher serum levels in males than females through bolus, but not continuous, dosing. Type of PFAS had the largest impact on serum concentrations, whereas sex had the lowest. Purpose: We present a novel perfusion phantom for validation of arterial spin labeled (ASL) perfusion MRI methods and protocols. Methods: Impinging jets, driven by a peristaltic pump, were used to achieve perfusion‐like mixing of magnetically labeled inflowing fluid within a perfusion compartment. The phantom was validated by varying pump rates and obtaining ASL‐MRI data at multiple postlabeling delays using a pseudo‐continuous ASL sequence with a 3D stack‐of‐spirals readout. An additional data set was acquired using a pseudo‐continuous ASL sequence with a 2D EPI readout. Phantom sensitivity to pseudo‐continuous ASL labeling efficiency was also tested. Results: Fluid dynamics simulations predicted that maximum mixing would occur near the central axis of the perfusion compartment. Experimentally observed signal changes within this region were reproducible and well fit by the standard Buxton general kinetic model. Simulations and experimental data showed no label outflow from the perfusion chamber and calculated perfusion rates, averaged over the entire phantom volume, agreed with the expected volumetric flow rates provided by the flow pump. Phantom sensitivity to pseudo‐continuous ASL labeling parameters was also demonstrated. Conclusion: Perfusion‐like signal can be simulated using impinging jets to create a well‐mixed compartment. Observed perfusion and transit time values were reproducible and within the physiological range for brain perfusion. The Atlantic Forest is the second most diverse forest system in South America, and only a fraction of its original distribution remains. In this study, we aim to use robust datasets of small mammals along the entire forest system to disentangle the main drivers for diversity along this gradient. More specifically, we aim to disentangle whether deforestation (recent), biogeographical variables, including 19 bioclimatic variables (historic), or historical trapping bias best describe patterns of taxonomic, functional, and phylogenetic diversities using small mammal assembles, from northeastern Brazil to eastern Paraguay. For that, we applied regression tree analyses to determine what environmental variables best describe each of the dimensions of diversity. Additionally, we implemented polynomial regression to test nonlinear relationships between biodiversity metrics and patch size. We found that patterns of overall taxonomic, functional, and phylogenetic diversities; rodent taxonomic diversity; and marsupial functional diversity were better explained by temperature variables. Meanwhile, marsupial taxonomic and phylogenetic diversities, and rodent functional and phylogenetic diversities were best explained by precipitation variables. Furthermore, patch area, trapping, and latitude were never the best descriptors for any of the diversity dimensions. Although all dimensions of biodiversity are correlated, they have unique information and should be considered individually to better understand biodiversity and inform conservation strategies. We found that fragmentation is impactful at a local scale and becomes less important at a biogeographical scale. Therefore, climatic variables drove biogeographical faunal patterns for all clades, probably reflecting important historical assembly process at large spatial scales. [ABSTRACT FROM AUTHOR]

This article cites a study on women employees who are in part-time employment. The characterization of work-family balance implies that, although part-time work by definition reduces time spent in the work role, the level of demands at home and at work ultimately will shape the psychological experience of balance and the meanings individuals attach to their participation in the work-family system. Individuals who have difficulty balancing work and family may experience problems in two major areas. First, they may experience time management difficulties and find it hard to work out the logistics of both work and family demands. Second, they may perceive high levels of work-family conflict as a form of interrole conflict in which pressures from the work and family domains are in some respects mutually incompatible. This research suggests that although part-time work may facilitate the logistics of time management for women in career and earner positions, the overall perception of work-family conflict may be a function of the specific job context. Women in earner positions appeared to derive far greater benefit through part-time schedules than did women in careers.

Produced water (PW) from oil and gas operations is considered a potential resource for food crop irrigation because of increasing water scarcity in dryland agriculture. However, efforts to employ PW for agriculture have been met with limited success. A greenhouse study was performed to evaluate the effects of PW on physiological and morphological traits of spring wheat (Triticum aestivum). Plants were irrigated with water treatments containing 10 and 50% PW (PW10 and PW50, respectively) and compared to a matching 50% salinity (NaCl50) and 100% tap water controls. Compared to controls, plants watered with PW10 and PW50 exhibited developmental arrest and reductions in aboveground and belowground biomass, photosynthetic efficiency, and reproductive growth. Decreases in grain yield ranged from 70 to 100% in plants irrigated with PW compared to the tap water control. Importantly, the PW10 and NaCl50 treatments were comparable for morphophysiological effects, even though NaCl50 contained 5 times the total dissolved solids, suggesting that constituents other than NaCl in PW contributed to plant stress. These findings indicate that despite discharge and reuse requirements focused on total dissolved solids, salinity stress may not be the primary factor affecting crop health. The results of the present study are informative for developing guidelines for the use of PW in agriculture to ensure minimal effects on crop morphology and physiology. Contaminants of emerging concern present in domestic waste streams include a highly diverse group of potentially biologically active compounds that can be detected at trace levels in wastewater. Concerns about potential uptake into crops arise when reclaimed water is used in food crop production. The present study investigated how 9 contaminants of emerging concern in reclaimed water are taken up into edible portions of two food crops. Two flame retardant chemicals, tris(1-chloro-2-propyl) phosphate (TCPP) and tris(2-chloroethyl) phosphate (TCEP) and several polar pharmaceuticals (carbamazepine, diphenhydramine, sulfamethoxazole, and trimethoprim) accumulated in a linear, concentration-dependent manner in lettuce ( Lactuca sativa) irrigated with reclaimed water, suggesting passive uptake of both neutral and ionizable chemical contaminants in lettuce. Furthermore, concentration-dependent accumulation of TCEP and TCPP from reclaimed water was also observed in strawberry fruits ( Fragaria ananassa). Collectively, these data suggest that highly polar or charged contaminants can be taken up by crops from water bearing contaminants of emerging concern and can be accumulated in the edible portions. Using these data, however, estimates of human exposure to these contaminants from reclaimed water food crop accumulation suggest that exposure to the contaminants of emerging concern examined in the present study is likely substantially lower than current exposure guidelines. Arid agricultural regions often turn to using treated wastewater (reclaimed water) to irrigate food crops. Concerns arise, however, when considering the potential for persistent contaminants of emerging concern to accumulate into plants intended for human consumption. The present study examined the accumulation of a suite of 9 contaminants of emerging concern into 2 representative food crops, lettuce and strawberry, following uptake via the roots and subsequent distribution to other plant tissues. Calculating accumulation metrics (concentration factors) allowed for comparison of the compartmental affinity of each chemical for each plant tissue compartment. The root concentration factor was found to exhibit a positive linear correlation with the pH-adjusted octanol-water partition coefficient ( DOW) for the target contaminants of emerging concern. Coupled with the concentration-dependent accumulation observed in the roots, this result implies that accumulation of these contaminants of emerging concern into plant roots is driven by passive partitioning. Of the contaminants of emerging concern examined, nonionizable contaminants, such as triclocarban, carbamazepine, and organophosphate flame retardants displayed the greatest potential for translocation from the roots to above-ground plant compartments. In particular, the organophosphate flame retardants displayed increasing affinity for shoots and fruits with decreasing size/octanol-water partition coefficient ( KOW). Cationic diphenhydramine and anionic sulfamethoxazole, once transported to the shoots of the strawberry plant, demonstrated the greatest potential of the contaminants examined to be then carried to the edible fruit portion. Mexico has higher mammalian diversity than expected for its size and geographic position. High environmental hetero geneity throughout Mexico is hypothesized to promote high turnover rates (β-diversity), thus contributing more to observed species richness and composition than within-habitat (α) diversity. This is true if species are strongly associated with their environments, such that changes in environmental attributes will result in changes in species composition. Also, greater heterogeneity in an area will result in greater species richness. This hypothesis has been deemed false for bats, as their ability to fly would reduce opportunities for habitat specialization. If so, we would expect no significant relationships between 1) species composition and environmental variables, 2) species richness and environmental heterogeneity, 3) β-diversity and environmental heterogeneity. We tested these predictions using 31 bat assemblages distributed across Mexico. Using variance partitioning we evaluated the relative contribution of vegetation, climate, elevation, horizontal heterogeneity (a variate including vegetation, climate, and elevational heterogeneity), spatial variation (lat-long), and vertical hetero geneity (of vegetation strata) to variation in bat species composition and richness. Variation in vegetation explained 92% of the variation in species composition and was correlated with all other variables examined, indicating that bats respond directly to habitat composition and structure. Beta-diversity and vegetational heterogeneity were significantly correlated. Bat species richness was significantly correlated with vertical, but not horizontal, heterogeneity. Nonetheless, neither horizontal nor vertical heterogeneity were random; both were related to latitude and to elevation. Variation in bat community composition and richness in Mexico were primarily explained by local landscape heterogeneity and environmental factors. Significant relationships between β-diversity and environmental variation reveal differences in habitat specialization by bats, and explain their high diversity in Mexico. Understanding mechanisms acting along environmental or geographic gradients is as important for understanding spatial variation in community composition as studying mechanisms that operate at local scales. An extensive review of the literature reveals a lack of insight into why some employees and their families benefit from the adoption of mobile technology while others do not. The paper summarizes the authors' efforts to answer this question. The authors undertook a longitudinal case study of the adoption and use of a Black Berry Smartphone by 25 professional knowledge workers. Four theoretical lenses were used to help with the data analysis process: boundary theory, the social constructivist view of technology, sensemaking and attribution theory. Analysis of the Time 2 data identified three groups. Segmentors (n = 4) did not use their smartphones outside work hours. Integrators (n = 8), used their smartphones to connect to both work and family anywhere, but not any time (temporally separated work and family roles). Struggling segmentors (n = 13) felt pressured by their organization to use their device 24/7 and did so. The analysis indicates that the relationship between the use of mobile technology and successful boundary management depends on the development of a strategy to manage the device prior to adoption, the ability to change one's strategy to respond to concerns at home, and self-control. As the production and applications of silver nanoparticles (AgNPs) increase, it is essential to characterize fate and effects in environmental systems. Nanosilver materials may settle from suspension; therefore, the authors' objective was to utilize environmentally relevant bioassays and study the impact, bioaccumulation, tissue distribution, uptake, and depuration of AgNPs on a sediment-dwelling invertebrate, Lumbriculus variegatus. Hydrodynamic diameters of uncoated 30-nm, 80-nm, and 1500-nm AgNP powders and a polyvinyl pyrrolidone (PVP) AgNP suspension were measured utilizing dynamic light scattering in freshwater media (0-280 µS/cm). Aggregation for 30 nm, 80 nm, and 1500 nm silver increased with conductivity but was minimal for PVP silver. Lumbriculus variegatus were exposed to AgNPs or silver nitrate (AgNO3) spiked into sediment (nominally 100 mg/kg) and water (PVP 30 nm and 70 nm Ag, nominally 5 mg/L). Uptake was assessed through inductively coupled plasma mass spectroscopy (ICP-MS) and hyperspectral imaging. Particle sizes were examined through field flow fractionation-ICP-MS (FFF-ICP-MS) and ICP-MS in single particle mode (SP-ICP-MS). Lumbriculus variegatus were also depurated for 6 h, 8 h, 24 h, and 48 h to determine gut clearance. Bioaccumulation factors of sediment-exposed L. variegatus were similar regardless of particle size or coatings. The FFF-ICP-MS and SP-ICP-MS detected AgNPs for up to 48 h post depuration. Increasing manufacture and use of engineered nanoparticles is leading to a greater probability for release of engineered nanoparticles into the environment and exposure to organisms. In particular, zinc oxide (ZnO) is toxic, although it is unclear whether this toxicity is due to the zinc oxide nanoparticles, dissolution to Zn2+, or some combination thereof. The goal of this study was to determine the relative solubilities of both commercially available and in-house synthesized ZnO in matrices used for environmental fate and transport or biological toxicity studies. Dissolution of ZnO was observed in nanopure water (7.18-7.40 mg/L dissolved Zn, as measured by filtration) and Roswell Park Memorial Institute medium (RPMI-1640) (∼5 mg/L), but much more dissolution was observed in Dulbecco's modified Eagle's medium, in which the dissolved Zn concentration exceeded 34 mg/L. Moderately hard water exhibited low Zn solubility, likely because of precipitation of a Zn carbonate solid phase. Precipitation of a Zn-containing solid phase in RPMI also appeared to limit Zn solubility. Equilibrium conditions with respect to ZnO solubility were not apparent in these matrices, even after more than 1,000 h of dissolution. These results suggest that solution chemistry exerts a strong influence on ZnO dissolution and can result in limits on Zn solubility from precipitation of less soluble solid phases. Environ. Toxicol. The environmental prevalence of engineered nanomaterials, particularly nanoparticulate silver (AgNP), is expected to increase substantially. The ubiquitous use of commercial products containing AgNP may result in their release to the environment, and the potential for ecological effects is unknown. Detecting engineered nanomaterials is one of the greatest challenges in quantifying their risks. Thus, it is imperative to develop techniques capable of measuring and characterizing exposures, while dealing with the innate difficulties of nanomaterial detection in environmental samples, such as low-engineered nanomaterial concentrations, aggregation, and complex matrices. Here the authors demonstrate the use of inductively coupled plasma-mass spectrometry, operated in a single-particle counting mode (SP-ICP-MS), to detect and quantify AgNP. In the present study, two AgNP products were measured by SP-ICP-MS, including one of precisely manufactured size and shape, as well as a commercial AgNP-containing health food product. Serial dilutions, filtration, and acidification were applied to confirm that the method detected particles. Differentiation of dissolved and particulate silver (Ag) is a feature of the technique. Analysis of two wastewater samples demonstrated the applicability of SP-ICP-MS at nanograms per liter Ag concentrations. In this pilot study, AgNP was found at 100 to 200 ng/L in the presence of 50 to 500 ng/L dissolved Ag. The method provides the analytical capability to monitor Ag and other metal and metal oxide nanoparticles in fate, transport, stability, and toxicity studies using a commonly available laboratory instrument. Aim We tested the hypothesis that distributions of Mexican bats are defined by shared responses to environmental gradients for the entire Mexican bat metacommunity and for each of four metaensembles (frugivores, nectarivores, gleaning insectivores, and aerial insectivores). Further, we identified the main environmental factors to which bats respond for multiple spatial extents. Location Mexico. Methods Using bat presence-absence data, as well as vegetation composition for each of 31 sites, we analysed metacommunity structure via a comprehensive, hierarchical approach that uses reciprocal averaging (RA) to detect latent environmental gradients corresponding to each metacommunity structure (e.g. Clementsian, Gleasonian, nested, random). Canonical correspondence analysis (CCA) was used to relate such gradients to variation in vegetation composition. Results For all bat species and for each ensemble, the primary gradient of ordination from RA, which is based on species data only, recovered an axis of humidity that matched that obtained for the first axis of the CCA ordination, which is based both on vegetation attributes and on species composition of sites. For the complete assemblage as well as for aerial and gleaning insectivores, analyses revealed Clementsian or quasi-Clementsian structures with discrete compartments (distinctive groups of species along portions of an environmental gradient) coincident with the humidity gradient and with the Nearctic-Neotropical divide. Within-compartment analysis further revealed Clementsian or quasi-Clementsian structures corresponding to a gradient of elevational complexity that matched the second ordination axis in CCA. Frugivores had quasi-nested structure, whereas nectarivores had Gleasonian structure. Main conclusions Our hierarchical approach to metacommunity analysis detected complex metacommunity structures associated with multiple environmental gradients at different spatial extents. More importantly, the resulting structures and their extent along environmental gradients are determined by ensemble-specific characteristics and not by arbitrarily circumscribed study areas. This property renders compartment-level analyses particularly useful for large-scale ecological analyses in areas where more than one gradient may exist and species sorting may occur at multiple scales. Elevational gradients provide a natural experiment for assessing the extent to which the structure of animal metacommunities is molded by biotic and abiotic characteristics that change gradually, or is molded by aspects of plant community composition and physiognomy that change in a more discrete fashion. We used a metacommunity framework to integrate species-specific responses to environmental gradients as an approach to detect emergent patterns at the mesoscale in the Luquillo Mountains of Puerto Rico. Elements of metacommunity structure (coherence, species turnover and range boundary clumping) formed the basis for distinguishing among random, checkerboard, Gleasonian, Clementsian, evenly spaced and nested patterns. Paired elevational transects (300-1000 m a.s.l.) were sampled at 50 m intervals to decouple underlying environmental mechanisms: a mixed forest transect reflected changes in abiotic and biotic conditions, including forest type (i.e. tabonuco, palo colorado and elfin forests), whereas another transect reflected changes in environmental conditions but not forest type, as its constituent plots were located within palm forest. Based on distributional data (presence versus absence of species), the mixed forest transect exhibited Clementsian structure, whereas the palm forest transect exhibited quasi-Gleasonian structure. In contrast, the distribution of modes in species abundance was random with respect to the latent environmental gradient in the mixed forest transect and clumped with respect to the latent environmental gradient in the palm forest transect. Such contrasts suggest that the environmental factors affecting abundance differed in form or type from those affecting distributional boundaries. Variation among elevational strata with respect to the first axis of correspondence from reciprocal averaging was highly correlated with elevation along each transect, even though axis scores were not correlated between mixed forest and palm forest transects. This suggests that the identity of the environmental characteristics, or the form of response by the fauna to those characteristics, differed between the two elevational transects. Despite the proximity of the transects, the patchy configuration of palm forest, and the pervasive distribution of the dominant palm species, the relative importance of abiotic variables and habitat in structuring gastropod metacommunities differed between transects, which is remarkable and attests to the sensitivity of metacommunity structure to environmental variation. The presence of the antimicrobial chemicals triclocarban (TCC) and triclosan (TCS) in municipal biosolids has raised concerns about the potential impacts of these chemicals on soil ecosystems following land application of municipal biosolids. The relative persistence of TCC and TCS in agricultural fields receiving yearly applications of biosolids at six different loading rates over a three-year period was investigated. Soil and biosolids samples were collected, extracted, and analyzed for TCC and TCS using liquid chromatography-tandem mass spectrometry. In addition, the potential for bioaccumulation of TCC and TCS from the biosolids-amended soils was assessed over 28 d in the earthworm Eisenia foetida. Standard 28-d bioaccumulation tests were conducted for three biosolids loading rates from two sites, representing agronomic and twice the agronomic rates of biosolids application plots as well as control plots receiving no applications of biosolids. Additional bioaccumulation kinetic data were collected for the soils receiving the high biosolids loadings to ensure attainment of quasi steady-state conditions. The results indicate that TCC is relatively more persistent in biosolids-amended soil than TCS. In addition, TCC bioaccumulated in E. foetida, reaching body burdens of 25 ± 4 and 133 ± 17 ng/g in worms exposed for 28 d to the two soils amended with biosolids at agronomic rates. The 28-d organic carbon and lipid-normalized biota soil accumulation factors (BSAFs) were calculated for TCC and ranged from 0.22 ± 0.12 to 0.71 ± 0.13. These findings suggest that TCC bioaccumulation is somewhat consistent with the traditional hydrophobic organic contaminant (HOC) partitioning paradigm. The primary objective of this study was to determine whether total biodiversity (γ) is partitioned into within-community (α) and among-community (β) components differently for taxonomic and functional organization. I hypothesized that α diversity will contribute more to the functional organization of γ diversity and that β diversity will contribute more to the taxonomic organization of γ diversity. A secondary objective was to determine whether the relationship between taxonomic and functional diversity is scale dependent. Species abundance data was obtained from fisheries surveys conducted by the Texas Parks and Wildlife Dept that focused on least disturbed streams from 11 different ecoregions of Texas, including 62 localities from 18 drainages. Functional and taxonomic organization of assemblages was quantified with two different measures of biodiversity, including richness and the numbers equivalent of Shannon diversity. Scale-dependent effects on these indices were assessed by multiplicatively partitioning γ into α and β components. The contribution of α and β components to γ diversity differed between functional and taxonomic organization and among different measures of biodiversity. Among-community components were more influential in structuring the taxonomic organization of stream-fish assemblages, whereas within-community components were more important in structuring the functional organization of assemblages. The relationship between taxonomic and functional diversity differed between α and β components and between spatial scales. The metacommunity framework is a powerful platform for evaluating patterns of species distribution in geographic or environmental space. Idealized patterns (checkerboard, Clementsian, evenly spaced, Gleasonian and nested distributions) give the framework shape. Each pattern represents an area in a multidimensional continuum of metacommunity structures; however, the current approach to analysis of spatial structure of metacommunities is incomplete. To address this, we describe additional non-random structures and illustrate how they may be discerned via objective criteria. First, we distinguish three distinct forms of species loss in nested structures, which should improve identification of structuring mechanisms for nested patterns. Second, we define six quasi-structures that are consistent with the conceptual underpinnings of Clementsian, Gleasonian, evenly spaced and nested distributions. Finally, we demonstrate how combinations of structures at smaller spatial extents may aggregate to form Clementsian structure at larger extents. These refinements should facilitate the identification of best-fit patterns, associated structuring mechanisms, and informative scales of analysis and interpretation. This conceptual and analytical framework may be applied to network properties within communities (i.e. structure of interspecific interactions) and has broad application in ecology and biogeography. The antimicrobial triclocarban (TCC) has been detected in streams and municipal biosolids throughout the United States. In addition, TCC and potential TCC transformation products have been detected at high levels (ppm range) in sediments near major cities in the United States. Previous work has suggested that TCC is relatively stable in these environments, thereby raising concerns about the potential for bioaccumulation in sediment-dwelling organisms. Bioaccumulation of TCC from sediments was assessed using the freshwater oligochaete Lumbriculus variegatus. Worms were exposed to TCC in sediment spiked to 22.4 ppm to simulate the upper bound of environmental concentrations. Uptake from laboratory-spiked sediment was examined over 56 d for TCC and 4,4′- dichlorocarbanilide (DCC), a chemical impurity in and potential transformation product of TCC. The clearance of TCC from worms placed in clean sediment was also examined over 21 d after an initial 35-d exposure to TCC in laboratory-spiked sediment. Concentrations of TCC and DCC were monitored in the worms, sediment, and the overlying water using liquid chromatography/tandem mass spectrometry. Experimental data were fitted using a standard biodynamic model to generate uptake and elimination rate constants for TCC in L. variegatus. These rate constants were used to estimate steady-state lipid (lip)- and organic carbon (OC)- normalized biota-sediment accumulation factors (BSAFs) for TCC and DCC of 2.2 ± 0.2 and 0.3 ± 0.1 g OC/g lip (goc/glip), respectively. Alternatively, directly measured BSAFs for TCC and DCC after 56 d of exposure were 1.6 ± 0.6 and 0.5 ± 0.2 goc/glip, respectively. Loss of TCC from pre-exposed worms followed first-order kinetics, and the fitted elimination rate constant was identical to that determined from the uptake portion of the present study. Understanding patterns in terms of the mechanistic processes that produce them is the essence of ecology. However, many studies merely document nonrandom patterns of species coexistence without providing insight into the structuring mechanisms and their scale dependence. Moreover, few studies address the mechanisms driving functional diversity. The objectives of this study were to document empirical patterns of species coexistence within stream fish assemblages across a broad geographic region, addressing both the functional and taxonomic organization of those assemblages, and to determine whether the structuring mechanisms are scale dependent. We gathered data from a report published by the Texas Parks and Wildlife Department that provided empirical species abundance distributions for 62 assemblages from 7 ecoregions and 18 river basins. For each assemblage, we simulated expected species abundances using five previously published models of niche apportionment. These models generate relative abundances in proportion to niche breadth and are constrained to maintain observed species richness, but they do not include information on species identity. These models included four stochastic variants of a geometric series (dominance preemption, random fraction, broken stick, and dominance decay) in which species abundances relate directly to niche breadth and one model (random assortment) in which abundances are independent of niche partitioning altogether. To assess scale dependence, we repeated the analyses after pooling assemblages by ecoregion, river basin, and the entire state. The patterns of niche apportionment for assemblages pooled according to river basin are more consistent with local assemblages than with assemblages pooled according to ecoregions or the entire state. These results suggest that niche apportionment plays a crucial role in structuring stream fish assemblages and that functional groups divide niche space more evenly than do species. Multidrug resistance, the phenomenon by which cells treated with a drug become resistant to the cytotoxic effect of a variety of other structurally and functionally unrelated drugs, is often associated with the expression of P-glycoprotein, an efflux membrane pump coded by the MDR1 (ABCB1) gene. Transcription from MDR1 can start at 2 promoters: a well-characterized downstream promoter and an as yet uncharacterized upstream promoter (USP). We have previously determined that the USP is activated in some drug-resistant cell lines, in primary breast tumors and in metastatic epithelial cells isolated from the lymph nodes of breast cancer patients. In this study, we report the cloning and characterization of the MDR1 USP and studied its association with chemotherapy response in breast cancer patients. Deletion analysis indicated that a nearby endogenous retroviral long terminal repeat is not responsible for promoter activation, and that the region within the first 400 nucleotides upstream from the transcription start point contained all the elements necessary for promoter activity in drug-resistant cells. We identified an element recognized by the transcription factor NF-IL6 (activated upon interleukin-6 exposure) which is necessary for promoter activity in drug-resistant cells and plays a role in the activation of the promoter in response to interleukin-6 in breast cancer MCF-7 cells. Although transcripts from this promoter are associated with translating polyribosomes, their low abundance makes the amount of synthesized P-glycoprotein insufficient to affect the response to first-line chemotherapy in patients with advanced breast cancer. © 2007 Wiley-Liss, Inc. Distributions of species often exhibit nested structure, such that assemblages at species-poor sites are proper subsets of taxa at more species-rich sites. Traditionally, this has been viewed as a large-scale biogeographic pattern and treated implicitly as static from a temporal perspective. Nonetheless, recent work suggests that nestedness may arise at multiple spatio-temporal scales. A 13-year data set encompassing the effects of two large-scale natural disturbances (hurricanes Hugo and Georges) on terrestrial gastropod assemblages was used to test the hypothesis that changes in species composition resulting from disturbance alter the degree of nestedness exhibited over time at two spatial scales. Gastropod assemblages were least nested immediately following disturbance, and nestedness increased thereafter. Although land-use history influenced the degree of nestedness, trajectories of nestedness following disturbance were similar irrespective of disturbance history or hurricane identity. The effects of hurricanes with respect to nestedness of terrestrial gastropods may be general and predictable, even though species respond to hurricanes in disparate fashions. By damaging some localities within the forest more severely than others, a hurricane dismantles extant patterns of species composition and severs connections among sites, as inhospitable microclimatic conditions limit dispersal of gastropods. As time passes and the forest canopy regenerates, conditions ameliorate, and movement among sites becomes more frequent. Thus, a conclusion based on a single time period may not characterize the study system in general. Consequently, explanations for nested structure that incorporate variability in ecological as well as evolutionary time will improve the applicability and comparability of nested subsets analysis across study systems. Nestedness has received considerable attention in community ecology and conservation biology from both theoretical and empirical perspectives. This has lead to the creation of various metrics and null models to analyze nested subsets, all of which rely on the random placement of species to assess significance. However, if immigration and extinction are the processes that underlie species distributions on island systems, then null models might be better determined on the basis of randomly placed individuals. Consequently, we examined the effects of species–abundance distributions (uniform, dominance–decay, random–assortment, and dominance–preemption), island–size distributions (uniform and linear decrease), and total abundances (128, 256, 512, 1024, 2048, 4096 and 8192) on the degree of nestedness and its significance. Generally, matrices of species presence and absence created from the random placement of individuals were nested significantly according to null models based on the random placement of species. Island size and abundance had less of an effect on nestedness in systems dominated by only a few species than in systems in which abundances were distributed more evenly. Stochastic processes, such as the random placement of individuals, predispose systems to evince patterns of nestedness at the species level, which may account, in part, for the ubiquity of nestedness in nature. This study investigated differences in general values, work values and organizational commitment among 549 private sector, public sector, and parapublic sector knowledge workers. No differences in general values were observed across sectors, although five significant work value differences were revealed: parapublic employees value work that contributes to society more than public servants, who value it more than private sector employees; parapublic employees value opportunities for advancement less than both public and private sector employees; public servants value intellectually stimulating and challenging work more than parapublic employees; and private sector employees value prestigious work more than public servants. Private sector employees displayed greater organizational commitment than the employees in the other two sectors. Overall, the findings suggest only limited value differences among employees of the various sectors. The finding of some work value differences between employees in the public and parapublic sectors suggests that these two groups merit separate consideration in comparative studies such as this one. We describe the morphological changes associated with starvation in larval largemouth bass and develop bivariate and multivariate morphological indices of nutritional status. We obtained hatchery-reared largemouth bass, raised them until completion of fin development, and divided them into two randomized experimental groups of fed and unfed fishes. Fed fishes were provided with newly hatched brine shrimp twice daily. We quantified morphological changes in body shape using 23 morphometric characters. After only 3 d of food deprivation, we were able to detect statistically significant differences in morphology between the fed and unfed fish using multivariate analysis. The magnitude of the mean difference increased over time. An unexpected result suggested that a simple, bivariate ratio of standard length to body depth at the anus was almost as efficient and robust at classifying fed and unfed largemouth bass from an independent data set as a multivariate index based on all 23 morphometric characters. The relative ease of use, combined with a small level of misclassification error rate and a firm foundation in whole-body changes, makes these ratio indices effective tools for detecting food deprivation in larval largemouth bass. Multidrug transporters play a dual role in haematopoietic cells, mediating the efflux of xenobiotics and regulating cell migration. For several reasons including the lack of specific antibodies, reports of multidrug transporter distribution on lymphocytes conflict. Murine B cells have been reported to completely lack transporter activity. Through analysis of parental and 'knockout' mice we show that, contrary to previous studies, murine B and T lymphocytes possess at least three active multidrug transporters and also a hitherto unrecognised drug-specific import activity. Surprisingly, the drug specificity of P-glycoprotein appears cell type dependent. The dominant theory of what people can learn implicitly is that they learn chunks of adjacent elements in sequences. A type of musical grammar that goes beyond specifying allowable chunks is provided by serialist or 12-tone music. The rules constitute operations over variables and could not be appreciated as such by a system that can only chunk elements together. A series of studies investigated the extent to which people could implicitly (or explicitly) learn the structures of serialist music. We found that people who had no background in atonal music did not learn the structures, but highly selected participants with an interest in atonal music could implicitly learn to detect melodies instantiating the structures. The results have implications for both theorists of implicit learning and composers who may wish to know which structures they put into a piece of music can be appreciated. [Copyright &y& Elsevier]

P-glycoprotein (P-gp), an adenosine triphosphate (ATP)-binding cassette transporter which acts as a drug efflux pump, is highly expressed at the blood–brain barrier (BBB) where it plays an important role in brain protection. Recently, P-gp has been reported to be located in the caveolae of multidrug-resistant cells. In this study, we investigated the localization and the activity of P-gp in the caveolae of endothelial cells of the BBB. We used an in vitro model of the BBB which is formed by co-culture of bovine brain capillary endothelial cells (BBCEC) with astrocytes. Caveolar microdomains isolated from BBCEC are enriched in P-gp, cholesterol, caveolin-1, and caveolin-2. Moreover, P-gp interacts with caveolin-1 and caveolin-2; together, they form a high molecular mass complex. P-gp in isolated caveolae is able to bind its substrates, and the caveolae-disrupting agents filipin III and nystatin decrease P-gp transport activity. In addition, mutations in the caveolin-binding motif present in P-gp reduced the interaction of P-gp with caveolin-1 and increased the transport activity of P-gp. Apoptotic cell volume decrease (AVD) and exposure of phosphatidylserine (Ptdser) at the cell surface are early events in apoptosis. However, the ion channels responsible for AVD, and their relationship to Ptdser translocation and cell death are poorly understood. Real-time analysis of calcium-induced apoptosis in lymphocytes and thymocytes showed that AVD occurs rapidly, and precedes Ptdser translocation. Blockers of the K+ channel IKCa1 completely inhibited AVD. Blockade of IKCa1, and hence AVD, also completely prevented Ptdser translocation and cell death. P-glycoprotein (P-gp) is an ABC (ATP-binding cassette) transporter, which hydrolyses ATP and extrudes cytotoxic drugs from mammalian cells. P-gp consists of two transmembrane domains (TMDs) that span the membrane multiple times, and two cytoplasmic nucleotide-binding domains (NBDs). We have determined projection structures of P-gp trapped at different steps of the transport cycle and correlated these structures with function. In the absence of nucleotide, an ∼10 Å resolution structure was determined by electron cryo-microscopy of two-dimensional crystals. The TMDs form a chamber within the membrane that appears to be open to the extracellular milieu, and may also be accessible from the lipid phase at the interfaces between the two TMDs. Nucleotide binding causes a repacking of the TMDs and reduction in drug binding affinity. Thus, ATP binding, not hydrolysis, drives the major conformational change associated with solute translocation. A third distinct conformation of the protein was observed in the post-hydrolytic transition state prior to release of ADP/Pi. The bacterial LmrA protein and the mammalian multidrug resistance P-glycoprotein are closely related ATP-binding cassette (ABC) transporters that confer multidrug resistance on cells by mediating the extrusion of drugs at the expense of ATP hydrolysis. The mechanisms by which transport is mediated, and by which ATP hydrolysis is coupled to drug transport, are not known. Based on equilibrium binding experiments, photoaffinity labeling and drug transport assays, we conclude that homodimeric LmrA mediates drug transport by an alternating two-site transport (two-cylinder engine) mechanism. The transporter possesses two drug-binding sites: a transport-competent site on the inner membrane surface and a drug-release site on the outer membrane surface. The interconversion of these two sites, driven by the hydrolysis of ATP, occurs via a catalytic transition state intermediate in which the drug transport site is occluded. H-NS is a major component of the bacterial nucleoid, involved in condensing and packaging DNA and modulating gene expression. The mechanism by which this is achieved remains unclear. Genetic data show that the biological properties of H-NS are influenced by its oligomerization properties. We have applied a variety of biophysical techniques to study the structural basis of oligomerization of the H-NS protein from Salmonella typhimurium. The N-terminal 89 amino acids are responsible for oligomerization. The first 64 residues form a trimer dominated by an α-helix, likely to be in coiled–coil conformation. Extending this polypeptide to 89 amino acids generated higher order, heterodisperse oligomers. Similarly, in the full-length protein no single, defined oligomeric state is adopted. The C-terminal 48 residues do not participate in oligomerization and form a monomeric, DNA-binding domain. These N- and C-terminal domains are joined via a flexible linker which enables them to function independently within the context of the full-length protein. Multidrug resistance of cancer cells is, at least in part, conferred by overexpression of P-glycoprotein (P-gp), a memeber of the ATP-binding cassette (ABC) superfamily of active transporters. P-gp actively extrudes chemotherapuetic drugs from cells, thus reducing their efficacy. As a typical ABC transporter, P-gp has four domains: two transmembrane domains, which form a pathways through the membrane through which substrates are transpoerted, and two hydrophilic nucleotide-binding domains (NBDs), located on the cytoplasmic side of the membrane, which couple the energy of ATP hydrolysis to substrate translocation. It has been proposed that the NBDs of ABC transporters, including the histidine permease of Salmonellla tyhimuruim and the cystic fibrosis trransbrane conductance regulator, are accesible from the extracelular surface of the cell, spanning the membrane directly or potenially contributing to the transmembrane pore. Such organization would have significant implications for the transport mechanism. We determined to establish whether the NBss of P-gp are exposed extracellularly and which amino acids are accessible, using cysteine-scanning mutagenesis and limited proteolysis. The recent completion of the Escherichia coli genome sequence (Blattner et al., 1997) has permitted an analysis of the complement of genomically encoded ATP-binding cassette (ABC) proteins. A total of 79 ABC proteins makes this the largest paralogous family of proteins in E. coli. These 79 proteins include 97 ABC domains (as some proteins include more than one ABC domain) and are components of 69 independent functional systems (as many systems involve more than one ABC domain). The ABC domains are often, but not exclusively, the energy-generating domains of multicomponent membrane-bound transporters. Thus, 57 of the 69 systems are ABC transporters, of which 44 are periplasmic-binding protein-dependent uptake systems and 13 are presumed exporters. The genes encoding these ABC transporters occupy almost 5% of the genome. Of the 12 systems that are not obviously transport related, the function of only one, the excision repair protein UvrA, is known. A phylogenetic analysis suggests that the majority of ABC proteins can be assigned to 10 subfamilies. The article presents a study focusing on the understanding of work-family conflict by the examination of life-cycle stage and gender differences of work-family conflict. The three components examined are role overload, interference from work to family, and interference from family to work. The life-cycle focus recognizes the dynamic nature of family roles and circumstances as families and individuals move through their lives and that these changes in roles, relationships and responsibilities over time produce corresponding changes in family needs, resources and vulnerabilities. The focus on gender acknowledges that despite the many changes in gender roles in the past 20 years, tradition prescribes a different emphasis between work and family for men and women. The data presented in this paper suggest that this strategy may, in fact, stall the career progression of many professional women rather than advance it The time period where these women will have dependent children at home will coincide with a period of maximum career visibility.

The Escherichia coli degradosome is a multienzyme complex with four major protein components: the endoribonuclease RNase E, the exoribonuclease PNPase, the RNA helicase RhlB and enolase. The first three of these proteins are known to have important functions in mRNA processing and degradation. In this work, we identify an additional component of the degradosome, polyphosphate kinase (PPK), which catalyses the reversible polymerization of the 7-phosphate of ATP into polyphosphate {poly(P)). An E. coli strain deleted for the ppk gene showed increased stability of the ompA mRNA. Purified His-tagged PPK was shown to bind RNA, and RNA binding was prevented by hydrotysable ATP. Chemical modification of RNA by PPK, for example the addition or removal of 3′ or 5′ terminal phosphates, could not be detected. However, polyphosphate was found to inhibit RNA degradation by the degradosome in vitro. This inhibition was overcome by the addition of ADP, required for the degradation of polyphosphate and for the regeneration of ATP by PPK in the degradosome. Thus, PPK in the degradosome appears to maintain an appropriate microenvironment, removing inhibitory polyphosphate and NDPs and regenerating ATP. [ABSTRACT FROM AUTHOR]

According to the twJn-supercoiled domain model, a local domain with a high level of DNA supercoiling can be generated between two divergently transcribed promoters. We have tested this model directly using the rate of DNA photo-cross-linking by 4,5′,8-trimethylpsoralen to measure local regions of unconstrained supercoiling in vivo. We demonstrate that, in plasmids, a localized domain of highly supercoiled DNA is generated upstream of the tetA promoter. The level of supercoiling of this domain depends on the activity of the tetA promoter and on the extent of translation of the tetA gene product. The highest level of supercoiling within this localized domain was observed in a topA strain, although the generation of a localized supercoiling domain could also be detected in a topA+ background, consistent with potential physiological significance. The level of supercoiling in this localized domain correlated with activation of the supercoiling-sensitive leu-500 promoter located upstream of tetA. These data provide a direct demonstration that localized domains of increased negative supercoiling can be generated upstream of an actively transcribed promoter, and that this can result in supercoiling-mediated transcriptional coupling between two promoters. mRNA degradation in Escherichia coli is mediated by a combination of exo- and endoribonucleases. We present evidence for a multiprotein complex which includes at least two enzymes that play important roles in mRNA degradation: the exoribonuclease polynucleotide phosphorylase (PNPase) and the endoribo-nuclease RNase E. An activity which impedes the processive activity of PNPase at stem-loop structures also appears to be associated with the complex. This complex is estimated to have a molecular mass of about 500 kDa and includes several additional polypeptides whose functions are unknown. The identification of a complex which includes several activities associated with mRNA degradation has implications for the mechanisms and coordinated control of mRNA degradation. Stem-loop structures can protect upstream mRNA from degradation by impeding the processive activities of 3′-5′ exoribonucleases. The ability of such structures to impede exonuclease activity in vitro is insufficient to account for the stability they can confer on mRNA in vivo. In this study we identify a factor from Escherichia coli which specifically impedes the processive activity of the 3′-5′ exonuclease PNPase at stem-loop structures in vitro. This factor can, potentially, reconcile the apparent discrepancy between the ability of 3′ stem-loop structures to stabilize upstream mRNA in vitro and in vivo. A TnphoA-generated mutant C5060, attenuated for virulence, was derived from the mouse-virulent Salmonella typhimurium strain C5. This mutation, designated hns-112::TnphoA, harbours the transposon to the 3′ end of hns, with the alkaline phosphatase open reading frame in the opposite orientation to that of hns. Bacterial strains harbouring hns-112::TnphoA were mucoid and had altered levels of DNA supercoiling, as monitored using pUC18 as a reporter plasmid. Transduction of hns-112::TnphoA into mouse virulent strains, including S. typhimurium SL1344 and Salmonella enteritidis Se795, resulted in attenuation. When an independent hns mutation, harbouring e kanamycin-resistance cassette inserted into the Kpnl site at base pair 237 of the hns gene, was introduced into S. typhimurium C5, the isolates were also attenuated. S. typhimurium C5 isolates harbouring the multicopy plasmid pGB651, which encodes the Escherichia coli hns gene, were partially attenuated in mice. Transductional analysis, using Tn10 insertions located close to the hns gene, showed that virulence could be restored in genetic crosses that eliminated the resident hns mutations. However, some hns+ transductants were still attenuated, suggesting that secondary attenuating lesions can accumulate in hns-deficient strains. The chromosomes of enteric bacteria are divided into about 50 independently supercoiled domains. It is not known whether the net level of DNA supercoiling is similar in each domain, or whether the domains are differentially supercoiled. We have addressed this question genetically, using a supercoiling-sensitive promoter to probe the relative levels of supercoiling at defined points around the Salmonella typhimurium chromosome. We conclude that, within the limits of resolution of this approach, the level of supercoiling does not differ significantly between chromosomal domains, and that each domain responds in a similar fashion to factors that perturb supercoiling.
