The changes in the East Asian polar‐front jet (EAPJ) and the East Asian subtropical jet (EASJ) profoundly impact the weather and climate in East Asia. However, the link between aerosols and the jet streams is still unclear. Here, we investigated the decadal co‐variation of aerosols and the East Asian jet streams in the boreal winter during the period of 1980–2019. In synch with a positive change in aerosol optical depth over East Asia, 300‐hPa winds show an equatorward shift of the land branch of the EASJ, and weakened EAPJ and oceanic branch of the EASJ. This can be linked to the enhanced meridional temperature gradient along 30°–50° N but weakened in northern regions and the decreasing synoptic‐scale transient eddy kinetic energy over subtropical Pacific. Relative importance estimation of aerosols and ocean signals emphasized the contributions of aerosols in jet variations. In turn, meteorological conditions related to jet streams also contribute to variations in aerosols, the decadal co‐variations are a result of their interactions, particularly for the oceanic branch of EASJ. Transport properties of silicate melts control magma ocean dynamics on the early terrestrial planets and rocky exoplanets. Here we calculate the viscosity (transport of momentum) of peridotite liquid at potential magma ocean conditions (0–159 GPa, 2,200–6,000 K) using ab initio molecular dynamics simulations. We find that, unlike MgSiO3 or basaltic melts, the viscosity of the highly depolymerized peridotite liquid (a) increases monotonically with pressure without an anomalous drop and (b) is lower than those of other melts over the entire mantle pressure range. Low viscosity would promote fractional crystallization in a less polymerized magma ocean and thus contribute to mantle heterogeneity from its earliest stage. Given the compositional dependence of magma ocean properties, emphasis on multicomponent bulk silicate Earth‐like composition, instead of simple end‐members, are rendered necessary, in order to better understand high‐energy planetary accretion processes and their aftermaths. Seismic measurements made on Mars indicate that the liquid iron‐nickel core is rich in light elements; however, the effects of these light components on the elasticity of Mars' core remain poorly constrained. Here, we calculate elastic properties of various liquid Fe‐X (X = Ni, S, C, O and H) mixtures using ab initio molecular dynamics simulations. We find that, at martian core conditions, the addition of S and O most effectively decreases the density of liquid iron, followed by C and H, while Ni has a minimal effect. As for compressional sound velocity (Vp), C increases Vp of liquid Fe throughout Mars' core, while both S and O reduce Vp, the intensity of which diminishes with increasing pressure. Assuming a martian core made of a binary mixture, the seismically‐inferred density would require the presence of at least 30 wt% S. The amounts of these LEs can be inferred from geophysical measurements such as seismology, an approach used to deduce the composition of Earth's core. Similar seismic measurements have been obtained for Mars with the InSight mission, which have allowed us to place limits on the mean density of Mars' core and its chemical make‐up. Relative to Earth's core, Mars' core is significantly less dense, requiring the presence of considerable amounts of LEs. We performed simulations based on first principles to determine the elastic properties of various Fe‐Ni‐rich‐LE mixtures that can be compared with those obtained from for example, InSight to place further constraints on its core composition, but can also be used for other planets such as Mercury and mid‐sized exoplanets. Nitrogen (N) is a major ingredient of the atmosphere, but a trace component in the silicate Earth. Its initial inventory in these reservoirs during Earth's early differentiation requires knowledge of N speciation in magmas, for example, whether it outgasses as N2 or is sequestered in silicate melts as N3−, which remains largely unconstrained over the entire mantle regime. Here we examine N species in anhydrous and hydrous pyrolitic melts at varying P‐T‐redox conditions by ab‐initio calculations, and find N‐N bonding under oxidizing conditions from ambient to lower mantle pressures. Under reducing conditions, N interacts with the silicate network or forms N‐H bonds, depending on the availability of hydrogen. Redox control of N speciation is demonstrated valid over a P‐T space encompassing probable magma ocean depths. Finally, if the Earth accreted from increasingly oxidized materials toward the end of its accretion, an N‐enriched secondary atmosphere might be produced and persist until later impacts. Our results suggest that, if Earth's building blocks were sufficiently oxidized at later times, magma ocean outgassing might produce an N‐enriched atmosphere. Given the violent impact history on the early Earth, as inferred from that recorded on the surface of the Moon, atmospheric loss of N by impact erosion may provide an explanation for Earth's "missing" nitrogen. Aim: To explore the mechanism of secreted frizzled‐related protein I (sFRP1) involvement in the osteogenic differentiation of human periodontal ligament cells (hPDLCs) under inflammatory conditions. Methodology: hPDLCs were cultured in an osteogenic differentiation‐inducing medium (odi) and subjected to the stimulation of Porphyromonas gingivalis lipopolysaccharide (P. gingivalis LPS) with or without the inhibition of sFRP1. Quantitative real‐time polymerase chain reaction, Western blot and enzyme‐linked immunosorbent assay were carried out to evaluate the expression of osteogenic markers as well as the classic Wnt signalling pathway. Periapical periodontitis was induced in Wistar rats to further confirm the effect of sFRP1 inhibitor on bone loss in vivo. After the Shapiro–Wilk normality test, data were analysed by Student's paired t‐test or one‐way Anova test with a P value less than 0.05 as the level of statistical significance. Results: Significantly decreased mRNA and protein expression of osteogenic markers were detected in hPDLCs treated with P. gingivalis LPS during osteogenic induction (P < 0.001). Increased expression of sFRP1 was observed (P < 0.01), whilst Wnt/β‐catenin signalling pathway was inhibited by the addition of P. gingivalis LPS (P < 0.01). After the addition of the sFRP1 inhibitor, the decrease of osteogenic markers (P < 0.05) and the inhibition of Wnt/β‐catenin signalling pathway (P < 0.05) were reversed significantly. The animal experiment further confirmed that the sFRP1 inhibitor significantly reduced bone loss of periapical lesions in vivo (P < 0.0001). Conclusions: Wnt antagonist sFRP1 was involved in the osteogenic differentiation of hPDLCs under inflammation. We report on the development of a high‐resolution and highly efficient beamline for soft X‐ray resonant inelastic X‐ray scattering (RIXS) located at the Taiwan Photon Source. This beamline adopts an optical design that uses an active grating monochromator (AGM) and an active grating spectrometer (AGS) to implement the energy compensation principle of grating dispersion. Active gratings are utilized to diminish defocus, coma and higher‐order aberrations, as well as to decrease the slope errors caused by thermal deformation and optical polishing. The AGS is mounted on a rotatable granite platform to enable momentum‐resolved RIXS measurements with scattering angles over a wide range. Several high‐precision instruments developed in‐house for this beamline are described briefly. The best energy resolution obtained from this AGM–AGS beamline was 12.4 meV at 530 eV, achieving a resolving power of 4.2 × 104, while the bandwidth of the incident soft X‐rays was kept at 0.5 eV. To demonstrate the scientific impact of high‐resolution RIXS, we present an example of momentum‐resolved RIXS measurements on a high‐temperature superconducting cuprate, i.e. La2–xSrxCuO4. The measurements reveal the A1g buckling phonons in superconducting cuprates, opening a new opportunity to investigate the coupling between these phonons and charge‐density waves. This paper presents a computationally efficient numerical model for predicting seismic responses of post‐tensioned cross‐laminated timber (CLT) rocking wall systems. The rocking wall is modeled as a simple linear beam element with a nonlinear rotational spring at the base. The model is primarily intended for preliminary design and assessment of multistory buildings using this particular lateral system. A method was developed to determine the nonlinear rotational spring parameters by considering the dimension of the CLT wall panel and post‐tensioned steel rods and energy dissipating devices' contributions. The proposed model was validated by comparing the simulated results with the responses from a series of shake table tests of a full‐scale two‐story building with CLT rocking walls. Isoprene (2‐methyl‐1, 3‐butadiene) is a nonmethane volatile organic compound (VOC) with the largest global emission and high reactivity. The oxidation of isoprene is crucial to atmospheric photochemistry and contributes significantly to the global formation of secondary organic aerosol. Here, we conducted comprehensive observations in polluted megacities of Nanjing and Shanghai during summer of 2018. We identified multiple functionalized isoprene oxidation products, of which 72% and 88% of the total mole concentration were nitrogen‐containing species with the dominant compound being C5 dihydroxyl dinitrate (C5H10N2O8). We calculated the volatility using the group‐contribution method and estimated the particle‐phase concentration by equilibrium gas/particle partitioning. The results showed that the multifunctional products derived from isoprene oxidation can contribute to 2.6% of the total organic aerosol mass (0.28 ± 0.27 μg/m3), highlighting the potential importance of isoprene oxidation in polluted regions. In polluted urban areas, where isoprene would interact significantly with anthropogenic emissions, for example, NOx, the understanding of SOA formation potential of isoprene is rather controversial. Here, we used state‐of‐the‐art instruments to measure isoprene, highly functionalized intermediate compounds and organic aerosol simultaneously at two suburban sites in eastern China region and identify several tens of isoprene oxidation products with most of which identified as nitrogen‐containing species, confirming the significant interaction between the isoprene derived peroxy radicals and NOx. These multifunctional products are mostly semi‐volatile species and can contribute considerably to the observed SOA, highlighting an important role of biogenic volatile organic compounds oxidation on SOA formation under high‐NOx conditions, for example, anthropogenic emission dominant regions. Aim: To measure the coronal root canal morphology of permanent mandibular first molars using 3D coordinates for more precise conservative endodontic cavity preparation. Methodology: In total, 57 cone beam computed tomography (CBCT) images of sound mandibular molars with fully formed apices without previous endodontic treatment were taken from 33 patients aged 16 to 75 years. The CBCT machine (MCT‐1[EX‐2F], J. Morita Manufacturing Corp, Kyoto, Japan) provided 14‐bit greyscale images with the voxel size of 0.125 mm. All images were taken at 80 kV and 5.0 mA, with a 17‐s exposure time, which were then reconstructed in 3D models and viewed by an endodontist. The gender of the patients and the tooth position, and number of root canals, were recorded. The landmarks of coronal root canals were determined, then the distribution of landmarks, maximum curvature in the axial direction and curvature directions in the horizontal direction of coronal root canals were measured. Distributions of landmarks were analysed using a spatial statistics method. Data about curvature were compared using a t test. Results: Overall, the distribution of root canal orifices and the centre of the canal primary curve were more centralized than other landmarks. The landmarks were located more mesiobuccally to the centre of the occlusal plane of mandibular first molars. Specifically, the measurements of the maximum curvature of coronal root canals in the axial direction were as follows: in 3‐canal teeth, the average angles of curvatures were 22°,23°,15° for mesiobuccal (MB), mesiolingual (ML) and distobuccal (DB) canals, respectively; in 4‐canal teeth, the average angles of curvatures were 24°,26°,15°,22°for MB, ML, DB and distolingual (DL) canals, respectively. The degrees of coronal root canal curvatures in the horizontal direction were as follows: in 3‐canal teeth, the average angles of curvatures were −8°,47°,−2° for MB, ML and DB canals, respectively; in 4‐canals teeth, the average angles were −11°,50°,−28°,45° for MB, ML, DB and DL canals, respectively. Statistically, there was no reliable side or sex difference in any of the measurements (P > 0.05). Aim: To explore the function and mechanisms of NLRP6 (NOD‐, LRR‐ and pyrin domain‐containing 6) in the inflammatory response of human periodontal ligament cells (HPDLCs). Methodology: Tissues associated with apical periodontitis were obtained from three patients who underwent endodontic microsurgery. The expression of NLRP6 in 3 human apical periodontitis tissues and HPDLCs was examined by immunohistochemistry and immunofluorescence, respectively. The expressions of NLRP6, Phospho(p)‐ p65, p65, IκB‐α, p‐ IκB‐α, ERK, p‐ ERK, NLRP3, Pro interleukin (IL)‐1β, Pro caspase‐1 and apoptosis‐associated speck‐like protein containing a CARD (ASC) were examined by western blot. The gene expression and secretion of proinflammatory cytokines were detected using quantitative real‐time polymerase chain reaction and enzyme‐linked immunosorbent assay. Data were analysed statistically with independent sample t‐tests. Results: NLRP6 was expressed in inflammatory periapical tissues and HPDLCs. Lipopolysaccharide (LPS) from Escherichia coli induced NLRP6 in HPDLCs (P < 0.05). After silencing NLRP6, E. coli LPS‐induced activation of NF‐κB and ERK signalling was enhanced, which was also accompanied by elevated levels of IL‐6 and tumour necrosis factor‐α (TNF‐α) (P < 0.05). Moreover, knockdown of NLRP6 led to up‐regulation of NLRP3, Pro IL‐1β and Pro caspase‐1 (P < 0.05), whereas down‐regulation of ASC (P < 0.05), which may contribute to unchanged levels of IL‐1β in HPDLCs inflammation. Conclusion: NLRP6 was functionally expressed in inflamed periapical tissues and HPDLCs. Objective: The purpose of this study was to investigate the clinical and histopathological characteristics of GLUT1 in human tongue squamous cell carcinoma (TSCC) and the role of plumbagin (PLB)‐mediating GLUT1 in the growth of TSCC. Subjects and methods: Forty‐five cases of TSCC samples were collected and the expression and location of GLUT1 was analyzed. The role and mechanism of PLB meditating GLUT1 in the inhibitory growth of human TSCC cell line CAL27 were investigated in vitro and vivo. Results: The expression of GLUT1 was observed in all samples of human TSCC by immunohistochemical staining. GLUT1 expression was significantly correlated with lymph node metastasis and clinical stage in TSCC. PLB treatment decreased cell viability and colony formation, and increased cell apoptosis in association with the downregulation of GLUT1 via inhibiting PI3K/Akt pathway in vitro and PLB suppressed tumor growth in correlation with downregulation of GLUT1, compared with control group in vivo. Conclusions: The findings demonstrated a novel anti‐cancer mechanism of PLB, inhibitory TSCC growth via suppressing PI3K/Akt/GLUT1 pathway, which will supply a theoretical basis for PLB to treat TSCC. Idiopathic Multicentric Castleman Disease (iMCD) is a rare IL-6-driven hematological disorder characterized by systemic lymphadenopathy, elevated immunoglobulin levels, and prominent plasmacytosis in the bone marrow and lymph nodes. Germline whole genome sequencing of the affected twins identified pathogenic homozygous mutations of I NCOA4 i c.G1322A and monoallelic mutations of I TRAF3 i c.G1504A - both genes recently implicated in IL-6 signaling and B-cell regulation. An "inflammatory" peripheral monocytosis enriched for the expression of S100A family genes was evident in both twins, as well as a group of monocytes expressing cytotoxic gene signatures in the affected twin with milder clinical manifestations. Summary: Background: Tong‐Xie‐Yao‐Fang (TXYF) is a Chinese herbal formula for treating chronic diarrhoea accompanied by abdominal pain. The results were inconsistent in previous trials examining its effect. Aim: To study the efficacy of TXYF granules for treating diarrhoea‐predominant irritable bowel syndrome (IBS‐D). Methods: We performed a double‐blind, placebo‐controlled randomised trial and enrolled 160 participants with IBS‐D. The participants had VAS scores ≥3 cm in IBS‐D global symptoms and ≥2 days in a week with abdominal pain and loose stools (Bristol score 5, 6 or 7). They were randomly assigned to received TXYF or placebo during a treatment period of 4 weeks, and they were followed up for 8 weeks after treatment. The primary outcome was adequate relief of IBS‐D global symptoms for at least 2 of 4 weeks during weeks 1‐4. Secondary outcomes included mean weekly VAS scores of IBS‐D major symptoms, mean weekly stool frequency, mean weekly Bristol score, and adverse events. Results: 155 of 160 patients completed the trial. We found a significantly higher rate of adequate relief of global symptoms in TXFY group during weeks 1 to 4 (57.5% vs 37.5%, χ2 = 5.6391, P = 0.017); logistic regression analysis showed a similar result (OR 2.2, 95% CI 1.2‐4.4, P = 0.016). Most of the secondary outcomes showed superiority of TXYF over placebo in weekly assessment from week 3 to week 7. The adverse event rate was low in both groups (3.8% vs 3.8%, P = 1.000). Summary: It is of great significance to develop and evaluate noninvasive indexes predicting the level of liver fibrosis. The aim of this study was to comparatively evaluate gamma‐glutamyl transpeptidase‐to‐platelet ratio (GPR) versus aspartate aminotransferase‐to‐platelet ratio index (APRI) and fibrosis index based on 4 factors (FIB‐4) in predicting different levels of liver fibrosis of chronic hepatitis B (CHB) within the framework of HBeAg‐positive and HBeAg‐negative patients. A total of 1157 HBeAg‐positive and 859 HBeAg‐negative CHB patients were enrolled, among whom the pathological stage ≥S2, ≥S3, ≥S4 were defined as significant fibrosis, extensive fibrosis and cirrhosis, respectively. Receiver operating characteristic (ROC) curves were used to evaluate the performance of GPR, APRI and FIB‐4 in predicting different levels of liver fibrosis. In HBeAg‐positive patients, the area under ROC curves (AUROCs) of GPR in predicting extensive fibrosis and cirrhosis were both significantly larger than those of APRI (P = .0001 and P < .0001). In HBeAg‐negative patients, the AUROCs of GPR in predicting significant fibrosis and cirrhosis were significantly larger than those of FIB‐4 (P = .0006 and P = .0041). The AUROC of GPR in predicting extensive fibrosis was significantly larger than that of APRI and FIB‐4 (P = .0320 and P = .0018). Using a cut‐off of GPR > 0.500 as standard, the sensitivities and specificities of GPR in predicting significant fibrosis in HBeAg‐positive patients were 59.6% and 81.2%, and for cirrhosis 80.9% and 63.8%, respectively; and those of HBeAg‐negative patients were 60.3% and 78.3%, 84.5% and 66.1%, respectively. Summary: To gain an understanding of the mechanisms by which Leydig cell steroidogenic function degenerates with ageing, we explored steroidogenic gene expression in relation to antioxidation status and endoplasmic reticulum (ER) stress during the ageing of mice. Expression of StAR, P450scc and other steroidogenic enzymes decreased starting at middle age (12‐month‐old) compared to that of the young control (3‐month‐old) mice. The immunohistochemical staining intensity of 3β‐HSD for Leydig cells was significantly weaker in the aged (24‐month‐old) group than that in the young control group. The number of Leydig cells showed no significant difference between the groups. A progressive reduction in antioxidants MnSOD and GPx4 was observed in the testicular tissue with down‐regulated SIRT1 protein level in the middle‐aged and aged (24‐month‐old) mice. The number of testicular macrophages was significantly higher in the aged group than that in the middle‐aged and young mice. Age‐associated up‐regulation of ER stress markers such as GRP78 and Chop was observed. Chronic hepatitis B ( CHB) is one of the major public health challenges in the world. Due to a strong interplay between specific T-cell immunity and elimination of hepatitis B virus ( HBV), efforts to develop novel immunotherapeutics are gaining attention. TG1050, a novel immunotherapy, has shown efficacy in an animal study. To support the clinical development of TG1050 in China, specific immunity to the fusion antigens of TG1050 was assessed in Chinese patients. One hundred and thirty subjects were divided into three groups as CHB patients, HBV spontaneous resolvers, and CHB patients with HBsAg loss after antiviral treatment. HBV-specific T-cell responses to pools of HBV Core or Polymerase genotype D peptides included in TG1050 were evaluated. HBV Core- or Polymerase-specific cells were detected in peripheral blood mononuclear cells ( PBMCs) from the different cohorts. The frequencies and intensities of HBV Core-specific immune responses were significantly lower in CHB patients than in HBsAg loss subjects. In CHB patients, a dominant pool derived from Polymerase (Pol1) was the most immunogenic. CHB patients with low viral loads (<106 IU/ mL) were more likely to have a positive response specific to the Core peptide pool. Overall, genotype D-derived peptides included in TG1050 could raise broad and functional T-cell responses in PBMCs from Chinese CHB patients infected with genotype B/C isolates. Core-specific immunogenic domains appeared as 'hot spots' with the capacity to differentiate between CHB vs HBsAg loss subjects. Aim To explore the role of NLRP3 ( NACHT [nucleotide-binding oligomerization], LRR [leucine-rich repeat] and PYD [pyrin domain] domains-containing protein 3) inflammasome in the inflammatory response of human periodontal ligament fibroblasts ( HPDLFs). Methodology The expression of NLRP3 and apoptosis-associated speck-like protein containing a CARD ( ASC) in inflammatory periapical tissues and HPDLFs was examined by immunohistochemical and immunofluorescent staining. HPDLFs were stimulated with muramyl dipeptide ( MDP) and lipopolysaccharide ( LPS) from E. coli with or without the silencing of ASC. The expression of NLRP3, ASC and caspase-1 was examined using quantitative real-time polymerase chain reaction. The secretion of proinflammatory cytokines, including interleukin-1β ( IL-1β), interleukin-6 ( IL-6) and tumour necrosis factor-α ( TNF-α) was measured in the cell supernatant with an enzyme-linked immunosorbent assay. Data were statistically analysed using independent sample t-tests. Results Immunohistochemistry and immunocytochemistry staining revealed that NLRP3 and ASC were expressed in HPDLFs and inflammatory periapical tissues. MDP and LPS promoted the expression of NLRP3, ASC and caspase-1 in HPDLFs ( P < 0.05). The secretion of proinflammatory cytokines was also increased with MDP and LPS stimulation ( P < 0.05). After silencing ASC, the secretion of IL-1β induced by MDP and LPS was significantly attenuated ( P < 0.05). Several noninvasive blood biomarkers have been established for the assessment of liver fibrosis in patients with chronic hepatitis B virus ( HBV) infection, but their clinical performance remains inconclusive. Here, we compared the diagnostic performance of these biomarkers and developed a novel algorithm for assessing liver fibrosis. Six hundred and sixteen chronically HBV-infected and treatment-naïve patients who underwent liver biopsy were enrolled and randomly divided into training (N=410) and internal validation cohorts (N=206). One hundred and fifty-nine patients from another centre were recruited as an external validation cohort. Receiver operating characteristic ( ROC) curves were used to analyse the performance of the gamma-glutamyltransferase-to-platelet ratio ( GPR), red cell volume distribution width-to-platelet ratio ( RPR), FIB-4 index, aspartate aminotransferase-to-platelet ratio index ( APRI) and HBV DNA level against liver histology, and a novel algorithm was developed using the recursive partitioning and regression tree ( RPART) method. In the training cohort, the area under the ROC curve of FIB-4 was significantly higher than that of APRI ( P=.038) but was comparable to those of GPR, RPR and HBV DNA; however, the performance of the biomarkers was similar among the validation cohort. The established RPR- HBV DNA algorithm performed better in the training cohort than any individual blood biomarker, and the corresponding sensitivity, specificity, positive predictive value and negative predictive value were 63%, 90%, 72% and 80%, respectively. In the internal and external validation cohorts, the performance of the algorithm in assessing liver fibrosis was also superior to that of other biomarkers. These results suggest that the established RPR- HBV DNA algorithm might improve the diagnostic accuracy of liver fibrosis in treatment-naïve patients with chronic HBV infection, although additional studies are warranted to confirm these findings. A new ignition method of the spark gap based on plasma ejection is proposed in this paper, as the conventional trigatron spark gap performs poorly under the low working coefficient (the ratio of the charging voltage to the self-breakdown voltage) in air. The plasma is generated by the capillary discharge, which has high pressure, high temperature and high velocity. The capillary discharge device is placed inside the low voltage electrode. As long as the triggering signal is sent to the device, a column of the plasma flow is ejected in axial direction and develops rapidly towards the high voltage electrode. Subsequently, the gap is broken down and a high resistive channel is formed, where the thermal ionization takes place and the arc across the whole gap is generated and develops into a well conductive channel. The process of the thermal ionization of the high resistive channel varies with the change of the spark gap distance. The breakdown delay and the delay jitter of the spark gap increase with the spark gap distance, as both parameters are mainly determined by the developing process of the plasma ejection. The characteristics of the plasma flow determine the possibility of the breakdown of the spark gap under the low working coefficient. The ignition method based on capillary plasma ejection has been proved by the preliminary experiments, which indicate that under the gap length of 8 cm and the working coefficient of less than 3%, the effective ignition is still achievable. Dental caries, trauma, and other possible factors could lead to injury of the dental pulp. Dental infection could result in immune and inflammatory responses mediated by molecular and cellular events and tissue breakdown. The inflammatory response of dental pulp could be regulated by genetic and epigenetic events. Epigenetic modifications play a fundamental role in gene expression. The epigenetic events might play critical roles in the inflammatory process of dental pulp injury. Major epigenetic events include methylation and acetylation of histones and regulatory factors, DNA methylation, and small non-coding RNAs. Infections and other environmental factors have profound effects on epigenetic modifications and trigger diseases. Despite growing evidences of literatures addressing the role of epigenetics in the field of medicine and biology, very little is known about the epigenetic pathways involved in dental pulp inflammation. This review summarized the current knowledge about epigenetic mechanisms during dental pulp inflammation. Progress in studies of epigenetic alterations during inflammatory response would provide opportunities for the development of efficient medications of epigenetic therapy for pulpitis. This case-control study was designed to investigate the safety of the AID technology. The health status of the offspring conceived by 1620 couples who underwent 7272 AID cycles in our Center for Reproductive Medicine between June 2006 and December 2012 was retrospectively analysed. The control group included 1018 women who naturally conceived and delivered in the same period. Twin birth rate was significantly higher in the AID group (no triplet birth) than in the control group (2.01% versus 0.39%, P < 0.01). In the AID group, Caesarean delivery was used in 1299 cases (81.65%), spontaneous vaginal delivery (18.04%) and forceps-assisted vaginal delivery (0.31%).There was no significant difference in male/female ratio of the offspring between AID and control groups (113.55 : 100 versus 113.36 : 100, P > 0.05). Compared to natural pregnancy, a pregnancy through AID resulted in higher multiple birth rate, premature delivery rate and neonatal congenital malformation rate. Increased multiple birth rate was attributable to ovulation induction, and increased rate of low-birthweight infants was related to multiplets and premature delivery. Pharmacokinetics and elimination of florfenicol and florfenicol amine in grouper held in sea water at 23.3 ± 0.8 °C were studied using HPLC method after they were given a single peroral dose of florfenicol at 24 mg kg−1 body weight. Florfenicol was rapidly absorbed from intestine and distributed extensively to all the tissues examined. The maximum concentrations ( Cmax, μg g−1 or μg mL−1) in plasma and tissues were observed at 2-6 h (the time to reach maximum concentration, Tmax) except for bile ( Tmax = 24 h) and were in the order of intestine (52.02 ± 25.07) > bile (49.41 ± 28.16) > gill (45.12 ± 11.10) > plasma (28.28 ± 5.43) > liver (21.97 ± 12.08) > muscle (21.63 ± 6.12) > kidney (20.88 ± 11.28) > skin (19.10 ± 5.88). The drug distribution level was higher in plasma than in extravascular tissues except for bile, based on the ratios of the area under concentration-time curve between tissue and plasma (AUCtissue/plasma). The elimination of florfenicol was rapid in fish, and the corresponding half-lives ( T1/2β) in the order of magnitude were bile (13.92 h) > muscle or liver (12.31 h) > skin (11.77 h) > plasma (11.57) > gill (11.04 h) > intestine (10.55 h) > kidney (10.05 h). Transformer ( tra) is the key gene that turns on the sex-determination cascade in Drosophila melanogaster and in some other insects. The honeybee Apis mellifera has two duplicates of tra, one of which (complementary sex determiner, csd) is the primary signal for complementary sex-determination (CSD), regulating the other duplicate (feminizer). Two tra duplicates have been found in some other hymenopteran species, resulting in the assumption that a single ancestral duplication of tra took place in the Hymenoptera. Here, we searched for tra homologues and pseudogenes in the Hymenoptera, focusing on five newly published hymenopteran genomes. We found three tra copies in the fig wasp Ceratosolen solmsi. Further evolutionary and expression analyses also showed that the two duplicates ( Csoltra-B and Csoltra-C) are under positive selection, and have female-specific expression, suggesting possible sex-related functions. Moreover, Aculeata species exhibit many pseudogenes generated by lineage-specific duplications. We conclude that phylogenetic reconstruction and pseudogene screening provide novel evidence supporting the hypothesis of independent duplications rather an ancestral origin of multiple tra paralogues in the Hymenoptera. Background Hydrogen sulfide (H2S) is endogenously generated from L-cysteine (L-Cys) by the enzymes cystathionine- β-synthase ( CBS) and cystathionine- γ-Lyase ( CSE). Hydrogen sulfide is also produced from D-cysteine (D-Cys) by D-Amino acid oxidase ( DAO). Methods The H2S production was measured by the methylene blue assay. The expression of DAO was investigated by Western blotting and immunohistochemistry. The short-circuit current ( Isc) was recorded using the Ussing chamber technique. Key Results The epithelium in rat jejunum possesses DAO, and generates H2S. D-cysteine, originally used as a negative control for L-Cys, significantly increases the H2S release, which is inhibited by I2 CA, an inhibitor of DAO. In vitro study by Ussing chamber technique reveals that D-Cys decreases the Isc across the epithelium of the rat jejunum and enhances the Na+-coupled L-alanine transport. Background and Objective Although surveys in the USA have shown that male subjects are more prone to develop periodontitis, sex as a risk factor in periodontitis, and its mechanism, remain controversial. Animal models are ideal for investigating immunological mechanisms of sex dimorphism in periodontitis because in these models it is possible to exclude the interference of gender-related risk factors, such as smoking and oral hygiene habits. Based on surveys in humans and reports on sex dimorphism in other diseases, our hypothesis is that sex is a risk factor in periodontitis. Material and Methods Different murine models (oral gavage model and ligature model) for periodontitis have been utilized to determine susceptibility to periodontitis in female and male mice. Periodontal bone levels were measured as the distance from the cemento-enamel junction to the alveolar bone crest (CEJ-ABC) in young female or male mice (8-10 wk of age). Differential expression of inflammatory mediators in the gingivae of female and male mice was determined by quantitative real-time PCR. Results In comparison with male mice, female mice displayed significantly ( p < 0.05) increased periodontal bone loss, accompanied by elevated expression of proinflammatory cytokines (interleukin-1β, interleukin-6 and interleukin-17A) and higher numbers of oral bacteria. Conclusion In contrast to the results in humans, in which periodontitis susceptibility is also influenced by confounding gender-related behaviors, in the murine oral gavage model and ligature model, female mice appear to be more susceptible to periodontal bone loss than male mice. In the ligature model, we observed significantly ( p < 0.05) higher CEJ- ABC distance, gingival proinflammatory cytokine production and number of oral bacteria in female mice. Furthermore, our results imply that female mice develop periodontitis with a higher progression rate. Our study has therefore established that animal models can be used to dissect the mechanisms underlying genuine gender-based differences in periodontal disease susceptibility and/or progression. The histone chaperone FACT plays an important role in facilitating nucleosome assembly and disassembly during transcription. FACT is a heterodimeric complex consisting of Spt16 and SSRP1. The N-terminal domain of Spt16 resembles an inactive aminopeptidase. How this domain contributes to the histone chaperone activity of FACT remains elusive. Here, the crystal structure of the N-terminal domain (NTD) of human Spt16 is reported at a resolution of 1.84 Å. The structure adopts an aminopeptidase-like fold similar to those of the Saccharomyces cerevisiae and Schizosaccharomyces pombe Spt16 NTDs. Isothermal titration calorimetry analyses show that human Spt16 NTD binds histones H3/H4 with low-micromolar affinity, suggesting that Spt16 NTD may contribute to histone binding in the FACT complex. Anti-inflammation strategy is one of the proposed therapeutic approaches to hepatic fibrosis. IL-17 is critical in inflammation, but the role of IL-17 in liver fibrosis has not yet been elucidated. In this study, we investigate the role of IL-17 on bile duct ligation-induced liver injury and fibrosis in C57BL/6 mice. Animals were sacrificed at designated times, and serum and liver tissues were collected for analysis of liver function and serum IL-6, IL-1β, tumour necrosis factor-alpha ( TNF-α) and transforming growth factor-β ( TGF-β) levels. IL-17 blockade with anti- IL-17A mAb significantly improved liver function and decreased hepatocellular necrosis, pro-inflammatory cytokines, neutrophils and macrophages influx. Furthermore, CD3 + and CD8 + lymphocytes, neutrophils and macrophages were found to express IL-17, and neutrophils are the principal IL-17-producing cells after BDL-induced liver injury. These data indicated that IL-17 signal contributes to the pathogenesis of cholestatic liver injury and blocked of IL-17 could potentially benefit patients with cholestatic liver disease. Aim Recent studies have shown that sarcopenia is associated with negative postoperative outcomes. However, none of these studies analysed muscle strength or physical performance, which are also important components of sarcopenia. The present study aimed to investigate whether sarcopenia itself, as defined by low muscle mass, strength and physical performance, would predict complications after surgery for colorectal cancer. Method We conducted a prospective study of patients who underwent surgery for colorectal cancer at our department between August 2014 and February 2015. Sarcopenia was diagnosed by a combination of third lumbar vertebra muscle index (L3 MI), handgrip strength and 6-m usual gait speed. Univariate and multivariate analyses evaluating the risk factors for postoperative complications were performed. Only complications classified as Grade II or above according to the Clavien-Dindo classification were analysed in this study. Results A total of 142 patients were included in the study, and 17 patients were diagnosed as having sarcopenia. Postoperative complications of Grade II or above occurred in 40 patients, including 10 with sarcopenia and 30 without sarcopenia. Multivariate analysis showed that sarcopenia and previous abdominal surgery were independent risk factors for postoperative complications. Patients with sarcopenia also had an obvious tendency to a higher incidence of infectious complications. By comparing two logistic regression models, sarcopenia showed a better predictive power for postoperative complications than did low muscle mass. Conclusion Sarcopenia and previous abdominal surgery are independent risk factors for complications after surgery for colorectal cancer. Ty1 -copia retrotransposons are widespread and diverse in insects. Some features of their hosts, such as mating and genetic systems, are predicted to influence the spread of selfish genetic elements like Ty1 -copia. Using part of the reverse transcriptase gene as a reference, we experimentally surveyed Ty1 -copia elements in eight species of fig wasps (Hymenoptera: Chalcidoidea), and performed an in silico analysis of six available genomes of chalcid wasps. Contrary to initial expectations that selfish elements such as Ty1-copia would be purged from the genomes of these species because of inbreeding and haplodiploidy, almost all of these wasps harbour an abundance of diverse Ty1 -copia elements. Phylogenetic analyses suggest that the families of Ty1 -copia elements found in these species have had a long association with their chalcid hosts. These results suggest an evolutionary scenario in which there was ancestral polymorphism followed by some taxa-specific events including stochastic loss and further diversification. Furthermore, estimating natural selection within the internal and terminal portions of the Ty1 -copia phylogenies demonstrated that the elements are under strong evolutionary constraints for their long-term survival, but evolve like pseudogenes in the short term, accompanied by the rise and fall of parasitic elements in the history of wasp lineage. Background Controversy exists regarding glucocorticoids therapy and the risk of peptic ulcer bleeding ( PUB). Aim The present study was undertaken to determine whether short-term use of glucocorticoids is associated with the occurrence of peptic ulcer bleeding. Methods The records of adult patients hospitalised for newly diagnosed peptic ulcer bleeding from 2000 to 2012 were retrieved from the Taiwan National Health Insurance Research Database, a nationwide population-based registry system. The association between systemic glucocorticoids usage and peptic ulcer bleeding was determined with a conditional logistic regression model comparing cases and controls during time windows of 7, 14 and 28 days using a case-crossover design. Results Of the 8894 enrolled patients, the adjusted self-matched odds ratios for peptic ulcer bleeding after exposure to the glucocorticoids were 1.37 (95% CI: 1.12-1.68, P = 0.003) for the 7-day window, 1.66 (95% CI: 1.38-2.00, P < 0.001) for the 14-day window and 1.84 (95% CI: 1.57-2.16, P < 0.001) for the 28-day window. Moderate to high, but not low dose glucocorticoids (methylprednisolone <4 mg/day or its equivalence) were associated with an increased risk of peptic ulcer bleeding. Concomitant use of a nonselective nonsteroidal anti-inflammatory drug ( NSAID) or aspirin further elevated the risk. However, it does not eliminate the effect of underlying diseases flare-up that may have placed the patients at risk for peptic ulcer bleeding in this kind of study design. Conclusions Short-term (7-28 days) exposure to glucocorticoids is significantly associated with peptic ulcer bleeding; this risk seems dose-dependent and is higher when nonselective NSAIDs or aspirin are used concurrently. Given the current extinction crisis coupled with the shortfall in funding, there is a pressing need to establish species conservation priorities. The prioritization of phylogenetic diversity and evolutionary distinctiveness is one approach; however, taking such an approach requires more phylogenetic data than are currently available for most taxa. Here, we investigate the effects of increased phylogenetic knowledge on the accuracy of evolutionary distinctiveness ( ED) scores over time using scleractinian corals as a case study. ED scores were calculated from four molecular-based phylogenies from 2008 to 2013, each one representing a chronological step of increased phylogenetic knowledge for scleractinian corals, finally resulting in a full species-level phylogeny which is used here as the reference dataset. As expected, the most complete and up-to-date phylogenies performed well at predicting scores taken from a recent, full-coverage species-level phylogeny of scleractinian corals. Surprisingly, however, older phylogenies and scores derived from expert opinion also performed well. More unexpectedly, the expert opinion-led scores, when used as a basis for imputing scores for missing species, achieved a close second in terms of prediction accuracy compared with the most recent and largest tree, which had nearly 10 times more taxonomic coverage. The prevalence of obesity among child-bearing women has increased significantly. The adverse consequences of maternal obesity on the descendants have been well accepted, although few studies have examined the underlying mechanisms. We investigated whether neonatal overfeeding in female mice alters metabolic phenotypes in the offspring and whether hypothalamic leptin signalling is involved. Neonatal overfeeding was induced by reducing the litter size to three pups per litter, in contrast to normal litter size of 10 pups per litter. Normal and neonatally overfed female mice were bred with normal male mice, and offspring of overfeeding mothers ( OOM) and control mothers ( OCM) were generated. We examined body weight, daily food intake, leptin responsiveness and the number of positive neurones for phosphorylated-signal transducer and activator of transcription 3 ( pSTAT3) along with neuropeptide Y ( NPY) in the arcuate nucleus of the hypothalamus ( ARH) and NPY in the nucleus tractus solitarius ( NTS) of the brain stem. The body weight and daily food intake of OOM were significantly higher than those of OCM. Leptin significantly reduced food intake and increased the number of pSTAT3 positive neurones in the ARH of OCM mice, whereas no significant changes in food intake and pSTAT3 neurones were found in leptin-treated OOM mice. The number of NPY neurones in the ARH and NTS of the OOM mice was significantly higher than that of OCM mice. The results of the present study indicate that the obese phenotype from mothers can be passed onto the subsequent generation, which is possibly associated with hypothalamic leptin resistance. Although double-skin façade (DSF) is an environmental-friendly architectural feature, its fire behaviour is a deep concern. The interior glass system including the glass pane, metal frame and associated accessories will be hotter than the exterior glass system as demonstrated by earlier studies. The glass pane above the fire room will be broken to spread flame into the upper compartment. Aprons are proposed to protect the air cavity of DSF in a way similar to those outside a single-skin façade. In this paper, the effect of aprons in protecting against fire spread from an underlying compartment to the compartments above by preventing glass breakage of the inner glass pane was studied. Fire and smoke from a post-flashover room fire adjacent to the DSF would be trapped in the air cavity between the two glass panes. Spreading of hot gases with different apron widths was studied by numerical simulations with CFD first. Fire environment with and without breaking the apron immediately above the fire room was studied. Full-scale burning tests on part of an experimental DSF rig were then carried out to demonstrate the performance of horizontal apron in the DSF rig of 6 m tall and air cavity depth of 2 m with different apron widths. Background There is debate regarding the predictive value of interventricular septum ( IVS) wall thickness for adverse events. Aims The study investigated the association between the severity of thickened IVS and all-cause death in Chinese patients with coronary artery disease ( CAD). Methods A total of 2297 CAD patients verified by angiography was consecutively included. Patients were grouped according to the severity of thickened IVS. Cox regression analysis was conducted to determine the independent prognostic value of thickened IVS for all-cause death. Results During a median follow up of 25 months, 149 patients died. A gradient increase in the risk of death was observed across thickened IVS groups. Compared to patients with normal IVS thickness, the adjusted hazard ratio ( HR) was 1.49 (95% confidence interval ( CI) 1.00-2.23, P = 0.05) and 2.13 (95% CI 1.29-3.54, P = 0.003) for all-cause death in those with mildly and moderately/severely thickened IVS respectively. For one unit increase in IVS thickness, the risk of all-cause death was elevated by 14% (adjusted HR 1.14, 95% CI 1.05-1.24, P = 0.003). In patients with normal indexed left ventricular mass, thickened IVS was also demonstrated as an independent risk factor for all-cause death. The interaction between figs and their pollinating wasps is regarded as a model system for studying specialized co-evolved mutualism. Chemoreception of fig wasps plays an important role in this interaction, and odorant-binding proteins ( OBP) function in the first step of odorant detection. The OBP repertoire of the fig wasp Ceratosolen solmsi is reported to be one of the smallest among insects; however, it is unknown how these OBPs are related to the complicated mating process occurring within the fig cavity and the extreme host specificity of the species. In the present study, we combined a structural analysis of the conserved cysteine pattern and motif order, a phylogenetic analysis, and previous studies on ligand-binding assays to deduce the function of OBPs. We also quantified the expression of OBP genes in different life stages of female and male fig wasps by using real-time quantitative PCR, which can help to predict the function of these genes. The results indicated that CsolOBP1 and CsolOBP2 (or CsolOBP5) in males may bind to pheromones and play important roles in mate choice, whereas CsolOBP4 and CsolOBP5 may primarily function in host localization by females through binding of volatile compounds emitted by receptive figs. The design, construction and commissioning of a beamline and spectrometer for inelastic soft X-ray scattering at high resolution in a highly efficient system are presented. Based on the energy-compensation principle of grating dispersion, the design of the monochromator-spectrometer system greatly enhances the efficiency of measurement of inelastic soft X-rays scattering. Comprising two bendable gratings, the set-up effectively diminishes the defocus and coma aberrations. At commissioning, this system showed results of spin-flip, d-d and charge-transfer excitations of NiO. These results are consistent with published results but exhibit improved spectral resolution and increased efficiency of measurement. The best energy resolution of the set-up in terms of full width at half-maximum is 108 meV at an incident photon energy tuned about the Ni L3-edge. [ABSTRACT FROM AUTHOR]

Aim To compare the amount of apically extruded debris and irrigant produced by two Ni- Ti instruments and hand files when removing root fillings, and to compare two experimental models. Methodology Sixty single straight root canals in human mandibular premolars were prepared with K-files and filled with gutta-percha and AH Plus sealer. The teeth were randomly divided into three groups of 20 for removal of the root filling material with Reciproc files (Group 1, RP), Mtwo retreatment files (Group 2, MR) or hand files (Group 3, H). Each group was then equally divided into experimental subgroups: A, with 1.5% agar gel model ( AG); B, with empty tube model ( ET). Apically extruded debris and irrigant was quantified by subtracting the initial weight of the test apparatus without a tooth from its weight after the root canal retreatment. Comparative analysis of the amount of apically extruded debris and irrigant for each of the instruments and the experimental models was performed. Time for gutta-percha removal was recorded. Data were statistically analysed using one-way analysis of variance. Results Removal of root fillings with two Ni- Ti instruments produced less apically extruded debris and irrigant than hand files in both experimental models ( P < 0.05). More apically extruded debris and irrigant was produced with Reciproc files than Mtwo retreatment files using the 1.5% agar gel model ( P > 0.05). Significantly more apically extruded debris and irrigant was produced with Reciproc files than Mtwo retreatment files using the empty tube model ( P < 0.05). Eukaryotic horizontal gene transfer ( HGT) events are increasingly being discovered yet few reports have summarized multiple occurrences in a wide range of species. We systematically investigated HGT events in the order Lepidoptera by employing a series of filters. Bombyx mori, Danaus plexippus and Heliconius melpomene had 13, 12 and 12 HGTs , respectively, from bacteria and fungi. These HGTs contributed a total of 64 predicted genes: 22 to B. mori, 22 to D. plexippus and 20 to H. melpomene. Several new genes were generated by post-transfer duplications. Post-transfer duplication of a suite of functional HGTs has rarely been reported in higher organisms. The distributional patterns of paralogues for certain genes differed in the three species, indicating potential independent duplication or loss events. All of these HGTs had homologues expressed in some other lepidopterans, indicating ancient transfer events. Most HGTs were involved in the metabolism of sugar and amino acids. These HGTs appeared to have experienced amelioration, purifying selection and accelerated evolution to adapt to the background genome of the recipient. The discovery of ancient, massive HGTs and duplications in lepidopterans and their adaptive evolution provides further insights into the evolutionary significance of the events from donors to multicellular host recipients.
