Introduction: The exact mechanisms underlying the development and maintenance of phantom limb pain (PLP) are still unclear. This study aimed to identify the factors affecting pain intensity in patients with chronic, lower limb, traumatic PLP. Methods: This is a cross‐sectional analysis of patients with PLP. We assessed amputation‐related and pain‐related clinical and demographic variables. We used univariate and multivariate models to evaluate the associated factors modulating PLP and residual limb pain (RLP) intensity. Results: We included 71 unilateral traumatic lower limb amputees. Results showed that (1) amputation‐related perceptions were experienced by a large majority of the patients with chronic PLP (sensations: 90.1%, n = 64; residual pain: 81.7%, n = 58); (2) PLP intensity has 2 significant protective factors (phantom limb movement and having effective treatment for PLP previously) and 2 significant risk factors (phantom limb sensation intensity and age); and (3) on the other hand, for RLP, risk factors are different: presence of pain before amputation and level of amputation (in addition to the same protective factors). Conclusion: These results suggest different neurobiological mechanisms to explain PLP and RLP intensity. While PLP risk factors seem to be related to maladaptive plasticity, since phantom sensation and older age are associated with more pain, RLP risk factors seem to have components leading to neuropathic pain, such as the amount of neural lesion and previous history of chronic pain. In Switzerland, fresh herbage is a favoured feed for dairy cows due to its high quality and availability and low production costs. However, transition and early lactation are periods characterized by an increased nutrient demand that may not be covered by herbage alone. To compare the effects of concentrate supplementation in early lactation on nutrient efficiency and ruminal fermentation, 24 multiparous Holstein cows were assigned to two performance groups according to their previous lactation milk yield: high‐ (8,959 ± 984 kg) and low‐ (6,204 ± 1,000 kg) potential cows. Within this group, cows were allocated to two treatment groups receiving either herbage ad libitum (n = 11) or herbage supplemented with concentrate (n = 13). The experiment started for each cow 2 weeks before the predicted calving date (LW‐2) and lasted until lactation week (LW) 8. Milk yield and dry matter intake (DMI) were recorded daily. The reticular pH was measured continuously using a telemetric pH bolus. Milk components and ruminal fermentation traits were analysed in LW‐2, LW2, LW4, LW6 and LW8. Supplemented cows (p < 0.001) and high‐potential cows (p = 0.015) produced more milk than unsupplemented cows and low‐potential cows, respectively. Milk acetone was affected by supplementation (p < 0.001) and milk potential (p = 0.002) and was especially high in unsupplemented, high‐potential cows until LW6. Supplementation caused a decrease in herbage DMI (p < 0.001) but resulted in an increased total DMI (p < 0.001), whereas milk potential had no effect on DMI. Associated with an increasing DMI (p < 0.001), ruminal volatile fatty acid concentration (p = 0.024) increased and reticular pH (p < 0.001) decreased from LW2 until LW6. Apart from that, effects on ruminal fermentation and reticular pH were minor. In conclusion, even though apparent nutrient efficiency was high, high‐potential cows without supplementation seem to struggle more with reduced nutrient availability than other cows; therefore, they appear to be more prone to metabolic stress and consequently to production diseases. Abstract: Due to seasonal changes in the quality and quantity of herbage, the nutrient supply to grazing dairy cows is not always sufficient, which may increase their metabolic load. To investigate the temporal pattern of behavioural changes in relation to concomitant metabolic alterations, we subjected 15 multiparous early lactating Holstein dairy cows (24 (SD 7.4) days in milk) to a short‐term metabolic challenge, which we provoked by abruptly withdrawing concentrate for 1 week. Cows grazed full‐time and were supplemented with concentrate in experimental week (EW) 1 and EW 3, whereas concentrate was withdrawn in EW 2. We analysed milk and blood samples to characterise the metabolic changes and found that the total yield of milk and protein decreased (p < 0.05) and fat yield, fat‐to‐protein ratio and acetone content increased (p < 0.05) from EW 1 to EW 2. Plasma glucose and insulin concentrations were lower (p < 0.05), and concentrations of nonesterified fatty acids and beta‐hydroxybutyrate were higher (p < 0.05) in EW 2 compared with EW 1. Apart from ingestive and rumination behaviour and activity, we also monitored the use of an automated brush on pasture. While time spent eating and ruminating increased (p < 0.05) in EW 2 compared with EW 1, time spent idling decreased (p < 0.05). Concomitantly, while time standing and moving increased (p < 0.05) from EW 1 to EW 2, walking time decreased (p < 0.05). The daily proportion of cows using the automated brush decreased (p < 0.05) in EW 2 compared with EW 1, as did the duration of brushing per day. In conclusion, grazing cows experiencing a metabolic challenge try to compensate for the nutrient deficiency by increasing eating time, a behavioural element important for short‐term survival. Due to the strong impact of weather conditions, we cannot currently recommend observation of outdoor brushing activity to address short‐term alterations in the metabolic state of grazing cows. This study's objective was to compare two options of pasture supplementation: corn silage (CS) alone or corn silage mixed with protein concentrate. The experiment was conducted with 18 lactating Holstein cows in mid‐lactation in a crossover design that included three treatments and three data collection periods. All cows had access to pasture for 17 h/day with an average herbage allowance of 16 kg dry matter (DM)/cow/day and were offered in‐barn corn silage, corn silage mixed with protein concentrate, or no supplementation. Cows were equipped with pH sensors residing in the reticulum and, during the 7‐day data collection periods, with a jaw movement recorder. Nonsupplemented cows produced 21.3 kg energy‐corrected milk (ECM) and ate 13.3 kg DM herbage at pasture. Cows supplemented with corn silage and corn silage plus protein produced 2.5 and 4.5 kg/day more ECM, respectively, consumed 3.4 and 3.3 kg/day more DM in total, respectively, ate for a shorter period of time, and ruminated longer than their nonsupplemented peers. Supplemented cows were almost able to cover their energy requirements and mobilised less body mass in contrast to the nonsupplemented cows. Cows offered corn silage plus protein showed increased ECM production, increased milk urea content and lower nitrogen use efficiency (NUE) compared to cows supplemented with corn silage only. Nonsupplemented dairy cows had the highest milk urea content and performed worst in terms of NUE. The best feed conversion efficiency resulted from the nonsupplemented dairy cows and those supplemented with corn silage plus protein. In nonsupplemented cows, the high feed conversion efficiency seemed to be due to the increased mobilisation of body mass. As a result of the starch‐rich supplementations, the ruminal acetic:propionic acid ratio became smaller, and the proportions of n‐butyric acid increased. The mean reticular pH values did not substantially vary across the three feeding treatments. For the choice of a supplementation option, herbage allowance and cost of supplement will have to be considered, but aspects of feed–food competition as well as animal welfare should not be ignored. Herbage feeding with only little input of concentrates plays an important role in milk production in grassland dominated countries like Switzerland. The objective was to investigate the effects of a solely herbage-based diet and level of milk production on performance, and variables related to the metabolic, endocrine and inflammatory status to estimate the stress imposed on dairy cows. Twenty-five multiparous Holstein cows were divided into a control (C+, n = 13) and a treatment group (C−, n = 12), according to their previous lactation yield (4679-10 808 kg) from week 3 ante partum until week 8 post-partum (p.p.). While C+ received fresh herbage plus additional concentrate, no concentrate was fed to C− throughout the experiment. Within C+ and C−, the median of the preceding lactation yields (7752 kg) was used to split cows into a high ( HYC+, HYC−)- and low-yielding ( LYC+, LYC−) groups. Throughout the study, HYC+ had a higher milk yield (35.9 kg/d) compared to the other subgroups (27.2-31.7 kg/d, p < 0.05). Plasma glucose (3.51 vs. 3.72 mmol/l) and IGF-1 (66.0 vs. 78.9ng/ mL) concentrations were lower in HYC−/ LYC− compared to HYC+/ LYC+ cows (p < 0.05). Plasma FFA and BHBA concentrations were dramatically elevated in HYC− (1.1 and 1.6 mmol/l) compared to all other subgroups (mean values: 0.5 and 0.6 mmol/l, p < 0.05). Saliva cortisol, plasma concentrations of serum amyloid A ( SAA), haptoglobin (Hp), beta-endorphin ( BE) and activity of alkaline phosphatase ( AP) were not different between C+ and C−. In conclusion, herbage-fed high-yielding cows without supplementary concentrate experienced a high metabolic load resulting in a reduced performance compared to cows of similar potential fed accordingly. Low-yielding cows performed well without concentrate supplementation. Interestingly, the selected markers for inflammation and stress such as cortisol, Hp, SAA, BE and AP gave no indication for the metabolic load being translated into compromised well-being. Calcar‐guided short stems in total hip arthroplasty (THA) permit surgeons to successfully reconstruct postoperative femoroacetabular offset, accurately restore leg length, and adequately re‐establish a wide range of caput‐collum‐diaphyseal angles. However, their effect on femoral antetorsion is less known. Indeed, controlling antetorsion of the femoral stem can be challenging because of the differences in individual femoral geometry and curvature. Therefore, we investigated if calcar‐guided short‐stem THA alters femoral antetorsion and compared it with conventional‐stem THA. Using 12 Thiel‐fixed, full‐body cadaver specimens from donors without known hip disorders, we compared an uncemented calcar‐guided femoral short‐stem prosthesis with an uncemented conventional straight‐stem prosthesis. In a paired study setup, each specimen received a calcar‐guided short stem on one side and a conventional stem on the other. On the acetabular side, all specimens received a press‐fit, monobloc acetabular cup. Femoral antetorsion angles were measured using the Waidelich method, and pre‐ and post‐operative angles of both sides were recorded. The mean preoperative femoral antetorsion angles were similar in both groups (24.8° ± 7.5° vs. 23.8° ± 6.1°, p = 0.313). Mean postoperative femoral antetorsion angles were 23.0° ± 5.5° in short‐stem and 13.5° ± 7.1° in conventional‐stem hips. Short‐stem hips had a small but nonsignificant difference in femoral antetorsion angles pre‐ and post‐operatively (1.8° ± 3.2°, p = 0.109), while the difference for conventional‐stem hips was much larger and highly significant (10.3° ± 5.8°, p < 0.001). Calcar‐guided short‐stem THA effectively restores femoral antetorsion. However, how this affects long‐term clinical outcomes and complications warrants further exploration. [ABSTRACT FROM AUTHOR]

Population bottlenecks can have dramatic consequences for the health and long‐term survival of a species. Understanding of historic population size and standing genetic variation prior to a contraction allows estimating the impact of a bottleneck on the species' genetic diversity. Although historic population sizes can be modelled based on extant genomics, uncertainty is high for the last 10–20 millenia. Hence, integrating ancient genomes provides a powerful complement to retrace the evolution of genetic diversity through population fluctuations. Here, we recover 15 high‐quality mitogenomes of the once nearly extinct Alpine ibex spanning 8601 BP to 1919 CE and combine these with 60 published modern whole genomes. Coalescent demography simulations based on modern whole genomes indicate population fluctuations coinciding with the last major glaciation period. Using our ancient and historic mitogenomes, we investigate the more recent demographic history of the species and show that mitochondrial haplotype diversity was reduced to a fifth of the prebottleneck diversity with several highly differentiated mitochondrial lineages having coexisted historically. The main collapse of mitochondrial diversity coincides with elevated human population growth during the last 1–2 kya. After recovery, one lineage was spread and nearly fixed across the Alps due to recolonization efforts. Our study highlights that a combined approach integrating genomic data of ancient, historic and extant populations unravels major long‐term population fluctuations from the emergence of a species through its near extinction up to the recent past. Low plasma total cholesterol (TC) concentrations are characteristic during the negative energy balance in early lactating dairy cows. The objective was to investigate short‐term effects of different TC concentrations during an aggravated energy deficiency through a 1‐week concentrate withdrawal on adaptations of metabolism and milk production. Multiparous Holstein cows (n = 15) were investigated during 3 week beginning at 24 ± 7 DIM (mean ± SD). Cows were kept on pasture and received additional concentrate in experimental week 1 and 3, while in week 2, concentrate was withdrawn. Blood was sampled once and milk twice daily. Based on their average TC concentration during week 1 (prior to concentrate withdrawal), cows were retrospectively assigned into a high (H‐Chol; n = 8, TC ≥ 3.36 mmol/L) and a low TC groups (L‐Chol; n = 7, TC < 3.36 mmol/L). Concentrations of phospholipids and lipoproteins were higher in H‐Chol compared to L‐Chol throughout the study (p < 0.05). During concentrate withdrawal, milk yield, glucose and insulin concentrations decreased similarly in both groups, while milk fat, milk acetone and plasma BHB were higher in H‐Chol compared to L‐Chol (p < 0.05). Compared to initial values, plasma NEFA, TAG and VLDL increased in both groups within 2 days after concentrate withdrawal (p < 0.05). Concentrations of NEFA during week 2 were greater in L‐Chol compared to H‐Chol (p < 0.05). Despite reintroduction of concentrate, milk yield in H‐Chol remained lower for two more days compared to week 1 (p <.05), whereas milk yield recovered immediately in L‐Chol. Activity of aspartate aminotransferase was higher in H‐Chol compared to L‐Chol in week 2 (p < 0.05). Greater plasma TC concentrations were associated with a reduced increase of NEFA. Further research is warranted if TC concentrations are related to adipose tissue mobilization and fatty acid turnover. [ABSTRACT FROM AUTHOR]

Summary Grass may have a differential impact on the metabolism of the dairy cow, depending on the grazing system applied. In this study, the hypothesis was tested that metabolism of grazed vs. zero-grazed dairy cows is differently regulated throughout the vegetation period. The study included three experimental periods (p1, p2 and p3) of 14 days each, and two treatments [grazing from pasture, PASTURE, n = 9; or zero-grazing in a free-stall barn (BARN, n = 9)]. Blood and liver samples were collected at the end of each period when the cows were on average 64, 120 and 197 DIM. Concentrations of metabolites and hormones, and activities of various enzymes were determined in plasma. Liver samples were measured for mRNA abundance of genes encoding enzymes and nuclear receptors involved in metabolic pathways. PASTURE cows had higher plasma concentrations of T3, BHB, and total protein than BARN cows across periods (p < 0.05). BARN cows had higher concentrations of NEFA (in p1) and urea (in p1 and p2) in relation PASTURE cows (p < 0.05), and had higher mRNA abundance of liver-X-receptor-α and glycerol-3-phosphate-acyltransferase across periods (p < 0.05). The results confirm the hypothesis that metabolism is different between BARN and PASTURE cows throughout the vegetation period, and show that the observed differences are mainly reflected in parameters of lipid metabolism. Summary This study compared productivity of dairy cows with different body weight (BW), but a constant ratio of maintenance to production requirements in their first lactation, in a pasture-based production system with spring calving. Two herds, Herd L (13 and 14 large cows in 2003 and 2004 respectively; average BW after calving, 721 kg) and Herd S (16 small cows in both years; 606 kg) [Correction added after online publication 14 January 2011: 16 small cows in both years; 621 kg was changed to 16 small cows in both years; 606 kg], all in their second or following lactations, were each allocated 6 ha of pasture and rotationally grazed on 10 parallel paddocks with equal herbage offer and nutritional values. Winter hay, harvested from the same pastures, was offered ad libitum in the indoor periods in a tied stall barn. Each herd received, per lactation and year, approximately 2000 kg dry matter (DM) of concentrates and of fodder beets, equally distributed to every individual. Indoors, the L-cows ingested more DM than the S-cows (18.7 vs. 16.3 kg DM/cow per day; p < 0.01), but DM intake per 100 kg of metabolic BW was similar (13.0 vs. 13.1 kg DM/cow per day). Estimates based on the n-alkane technique gave similar results on pasture (17.9 vs. 15.5 kg DM/cow per day; p < 0.001). Roughage intakes per 100 kg of metabolic BW, at 13.5 kg DM/cow per day, were similar. Mean annual yield of energy-corrected milk (ECM)/ha was slightly higher for the S-herd than the L-herd (13 026 vs. 12 284 kg) but was associated with a higher stocking rate (on average +20%) for the S-herd. Feed conversion efficiency (1.2 vs. 1.3 kg ECM/kg DM intake) and overall milk production efficiency (45.3 vs. 47.3 kg ECM/kg metabolic BW) were similar in L- and S-cows.
