Two new cembrane diterpenes, sinulaparvalides A and B 1and 2, resp., which contain an intriguing sevenmembered lactone moiety, along with the four known related cembranoids 3–6, were isolated from the Hainan soft coral Sinularia parva.The structures of compounds 1and 2were established by spectroscopic methods, mainly on the basis of 2DNMR techniques, and were confirmed by singlecrystal Xray diffraction analysis. The in vitrocytotoxic activities of these compounds were tested on human colon carcinoma HCT116, human promyelocytic leukemia HL60, and human lung carcinoma A549 tumor cell lines. Two new biscembranoids, ximaolides F 1 and G 2, were isolated from the Hainan soft coral Sarcophyton tortuosum.The structures and relative configurations of the two new compounds were elucidated by the combination of spectroscopic methods, chemical conversion of ximaolide F 1 into ximaolide G 2, and comparison with related model compounds. The biomimetic synthesis of sacculatane-type epimeric compounds 14a and 14b is reported. The key synthetic step is the low-temperature superacidic cyclization of (all-E)-ω-acetoxygeranylgeraniol 12 obtained in nine steps from geranyllinalool (13). The 19-acetoxysacculata-7,17-dien-11-ol (14a) could be an important and convenient starting compound for the synthesis of other sacculatane diterpenoids. [ABSTRACT FROM AUTHOR]
